Die Erfindung betrifft ein neues Verfahren zur Herstellung von 4-Sulfanilamido-5,6-dialkoxypyrimidinen
der allgemeinen Formel

worin R1 und R2 niedere Alkylgruppen, wie Methyl-,
Äthyl-, Propyl-, Isopropyl-, Butyl- oder Hexylgruppen, bedeuten.
Das Verfahren ist dadurch gekennzeichnet, daß man in an sich bekannter Weise aus einer Verbindung
der allgemeinen Formel

worin R1 und R2 die oben angegebene Bedeutung
haben und X für die Aminogruppe oder einen in diese überführbaren Substituenten, wie eine Acylaminogruppe,
bevorzugt die Acetylamino- oder eine Carbalkoxyaminogruppe, z. B. die Carbäthoxyamino-
oder Carbobenzyloxyaminogruppe, oder die Nitrogruppe steht, die SR2-Gruppe mittels Raney-Nickel
in sodaalkalischer oder ammoniakalischer Lösung durch Erwärmen hydrogenolytisch abspaltet
und einen im Reaktionsprodukt gegebenenfalls vorhandenen, in die Aminogruppe überführbaren Substituenten
in die Aminogruppe umwandelt.
Die vorstehend genannten, in die Aminogruppe überführbaren Substituenten X lassen sich nach an
sich bekannten hydrolytischen bzw. reduktiven Methoden in die Aminogruppe umwandeln. Die
Reduktion von reduzierbaren Gruppen, z. B. der Nitrogruppe, kann auch gleichzeitig mit der Hydrogenolyse
der SR2-Gruppe erfolgen. Zur Abspaltung von gegebenenfalls vorhandenen NrAcylgruppen
braucht das nach der Hydrogenolyse erhaltene, desulfurierte Reaktionsprodukt nicht isoliert zu werden.
Es kann direkt der Verseifung mit einem Alkalihydroxyd, z. B. mit Natronlauge, unterworfen werden.
Die freie Aminoverbindung der allgemeinen Formel I kann aus der alkalischen Lösung leicht durch Zugabe
von Säure, z. B. Essigsäure, bis zu einem pH-Wert von ungefähr 5 oder durch Einleiten von Kohlendioxyd
aus der alkalischen Lösung ausgefällt werden.

Die als Ausgangsstoffe verwendeten Verbindungen der allgemeinen Formel II können in nicht beanspruchter
Weise z. B. durch Kupplung eines Aminopyrimidins der allgemeinen Formel

mit einem Benzol sulfonylhalogenid, das in p-Stellung einen in die Aminogruppe überführbaren Substituenten
enthält, z. B. mit p-Nitro- oder mit p-Acetylamino-benzolsulfonylchlorid,
erhalten werden. Ausgangsstoffe mit freier Aminogruppe in p-Stellung können aus diesen Kupplungsprodukten durch an
sich bekannte Umwandlung des p-Substituenten in die Aminogruppe erhalten werden.
Die nach dem erfindungsgemäßen Verfahren erhältlichen Sulfonamide der Formel I zeichnen sich
durch hohe antibakterielle Wirksamkeit aus, z. B. gegen Infektionen von Staphylokokken, Pneumokokken
oder Colibazillen. Sie sind dem bekannten - Sulfanilamido - 2,6 - dimethoxypyrimidin in mehrfacher
Hinsicht pharmakologisch überlegen (vgl.
609 578/542
die nicht vorveröffentlichte deutsche Patentschrift
1 173 100).
Die Verfahrensprodukte sind unter Salzbildung löslich in starken Säuren, wie Schwefel- oder Salzsäure,
und in Alkalien, wie Alkalihydroxyd-, Alkalicarbonat- oder Alkalibicarbonatlösungen.
a) 1,3 g 2-Methylthio-4-(N4-acetylsulfanilamido> 5,6-dimethoxypyrimidin werden mit 25 ml Wasser
und 0,4 g wasserfreiem Natriumcarbonat in Lösung gebracht und mit etwa 6 bis 7 g Raney-Nickel
3x/2 Stunden unter Rückfluß erhitzt. Dann wird eine
Lösung von 1 g Natriumhydroxyd in 3 ml Wasser zugegeben und 1 Stunde weitererhitzt. Das Nickel
wird abfiltriert und das Filtrat mit Salzsäure auf kongosauer gestellt. Die salzsaure Lösung wird
dann mit Ammoniak auf pH 5 gestellt, wobei Kristallisation eintritt. Man erhält so 0,51 g 4-Sulfanilamido
- 5,6 - dimethoxypyrimidin vom Schmelzpunkt 190 bis 194° C (aus 50%igem Äthanol).
b) Das als Ausgangsstoff verwendete 2-Methylthio -A- (N4 - acetylsulfanilamido)- 5,6 - dimethoxypyrimidin
kann nicht beanspruchter Weise wie folgt erhalten werden:
Thioharnstoff wird mit a-Methoxy-cyanessigsäuremethylester in Gegenwart von Natriummethylat zum
2 - Thio - 4 - amino - 5 - methoxy - 6 - hydroxypyrimidin cyclisiert. Dieses Produkt wird in einer Natriummethylatlösung
mit Methyljodid zum 2-Methylthio-4-amino - 5 - methoxy- 6-hydroxypyrimidin methyliert
[Schmelzpunkt 2030C (aus Wasser)], und letzteres wird mit Phenyltrimethylammonium-toluolsulfonat
zum 2-Methylthio-4-amino-5,6-dimethoxypyrirnidin methyliert [Schmelzpunkt 112 bis 115°C (aus
20%igem Methanol)].
0,9 g des 2-Methylthio-4-amino-5,6-dimethoxypyrimidins
werden in 3 ml absolutem Pyridin gelöst, bei 00C mit 1,2 g p-Acetylaminobenzolsulfonylchlorid
versetzt und bis zur Lösung geschüttelt. Die Lösung wird 22 Stunden bei 0°C stehengelassen. Dann wird
das Pyridin im Vakuum (200C) entfernt. Das erhaltene
Produkt wird mit 20 ml Wasser und 3 ml Eisessig versetzt und kurz bis zum Sieden erhitzt,
wobei Kristallisation eintritt. Das erhaltene rohe Produkt wird in 40 ml 2,5%iger Sodalösung gelöst.
Diese Lösung wird nach Filtration mit Kohlendioxydgas übersättigt. Es werden 1,5 g (85%)
- Methylthio - 4 - (N4 - acetylsulfanilamido) - 5,6 - dimethoxypyrimidin vom Schmelzpunkt 220 bis 2210C
(aus 50%igem Äthanol) erhalten.
Claims (2)
1. Verfahren zur Herstellung von 4-Sulfanilamido-5,6-dialkoxypyrimidinen
der allgemeinen Formel

worin R1 und R2 niedere Alkylgruppen bedeuten,
dadurch gekennzeichnet, daß man in an sich bekannter Weise aus einer Verbindung der allgemeinen Formel

worin R1 und R2 die oben angegebene Bedeutung
haben und X für die Aminogruppe oder einen in die Aminogruppe überführbaren Substituenten
steht, die SR2-Gruppe mittels Raney-Nickel in sodaalkalischer oder ammoniakalischer Lösung
durch Erwärmen hydrogenolytisch abspaltet und einen im Reaktionsprodukt gegebenenfalls vorhandenen,
in die Aminogruppe überführbaren Substituenten in die Aminogruppe umwandelt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet,
