Abstract

技术领域
本发明是在National Institutes of Health拨款号P01-CA45548的政府资助下完成的。美国政府在本发明中有一定的权利。
本发明涉及新型内皮细胞增殖抑制剂。该抑制剂能抑制血管生成相关性疾病和调节血管生成过程。此外,本发明涉及测定生物流体样本中存在的抑制剂量的诊断分析和试剂盒,涉及定位该抑制剂的组织化学试剂盒,涉及编码该抑制剂的DNA序列和监测抑制剂生物合成与降解的分子探针,涉及该抑制剂的特异性抗体,涉及开发该抑制剂受体的肽兴奋剂和拮抗剂,涉及抗抑制剂受体的特异性抗体兴奋剂和拮抗剂,还涉及与该抑制剂连接的细胞毒性剂。
背景技术
本文中应用的术语“血管生成”指组织或器官内生长新血管,并包括内皮细胞增殖。在正常生理条件下,人或动物只是在相当特殊的局限境况下才经历血管生成。例如,血管生成一般见于创伤愈合,胎儿和胚胎的发育以及黄体、子宫内膜和胎盘的形成。术语“内皮”表示衬贴于浆膜腔、淋巴管和血管的平坦的上皮细胞薄层。
人们认为受控的和未受控的血管生成按相似方式进行。被基膜包围的内皮细胞和周皮细胞形成毛细血管。血管生成始于内皮细胞和白细胞释放的酶对基膜的侵蚀。然后衬于血管腔的内皮细胞穿过基膜伸出。血管生成的刺激物诱使内皮细胞迁移穿过被侵蚀的基膜。迁移的细胞形成脱离母体血管的“新芽”,此处内皮细胞经历有丝分裂和增殖。该内皮新芽相互结合形成毛细血管襻,生成新血管。
持续的、未受调节的血管生成出现于大量病态、肿瘤转移和内皮细胞反常生长中,并为见于这些疾病中的病理损伤提供证据。其中存在未受调节的血管生成的各种病理病态已被归为血管生成依赖性疾病或血管生成相关性疾病。
关于“肿瘤生长是血管生成依赖性的”这一假设最先提出于1971(Folkman J.,Tumor angiogenesis:Therapeutic implications.,N.Engl.Jour.Med.285:1182-1186,1971)。在其最简单的语句中阐述:“一旦出现肿瘤‘占领’,则每次肿瘤细胞数增加之前必有会聚在肿瘤上的新毛细血管的增加。”肿瘤‘占领’现在被理解为肿瘤生长的前血管期(prevascular phase),其中肿瘤细胞群占据数立方毫米体积且不多于数百万个细胞,它们能生存在现存的宿主微血管上。肿瘤体积扩大到该期以外需要新毛细血管的诱导。例如,小鼠早期前血管期中肺的微转移瘤,除非用高倍显微镜检术对组织切片检查,否则检查不出。
支持该概念的间接证据实例包括:
(1)植入小鼠皮下透明室内的肿瘤的生长速度在形成新血管之前缓慢且呈线性,但在形成新血管之后速度很快且接近指数形式。(AlgireGH等,Vascular reactions of normal and malignant tumors in vivo.I.Vascular reactions of mice to wounds and to normal and neoplastictransplants.J.Natl.Cancer Inst.6:73-85,1945)。
(2)生长于血管不增殖的隔离、灌注的器官中的肿瘤限于1-2mm3,但当它们被移植至小鼠时就会迅速扩大到该体积的1000倍以上并变成新血管化的肿瘤。(Folkman J,等,Tumor behavior in isolated perfusedorgans:In vitro growth and metastasis of biopsy material in rabbitthyroid and canine intestinal segments.Annals of Surgery 164:491-502,1966)
(3)生长于无血管角膜内的肿瘤生长速度缓慢且呈线性,但在新血管化后转为指数形式的生长。(Gimbrone,M.A.,Jr.等,Tumor growthand neovascularization:An experimental model using the rabbitcornea.J.Natl.Cancer Institute 52:41-427,1974)
(4)悬浮于兔眼前眼房的水性流体中的肿瘤能存活,无血管,大小限于<1mm3。但一旦被植入虹膜血管床,它们就会变成新血管化并迅速生长,在2周内就达到它们原先体积的16,000倍。(Gimbrone M.A.Jr.等,Tumor dormancy in vivo by prevention of neovascularization.J.Exp.Med.136:261-276)
(5)当将肿瘤植入雏鸡胚胎绒毛尿囊膜时,它们在>72小时的无血管期期间生长缓慢,平均直径不超过0.93+0.29mm。开始形成新血管后的24小时内肿瘤迅速扩大,到第7天这些血管化的肿瘤平均直径达8.0+2.5mm。(Knighton D.,Avascular and vascular phases of tumorgrowth in the chick embryo.British J.Cancer,35:347-356,1977)
(6)兔肝内转移瘤的血管管型表现出转移瘤大小不均一性,但在形成血管的大小处表现为相对均一的切断点。直径在1mm内的肿瘤通常无血管,但大于该直径就形成新血管。(Lien W.,等,The blood supply ofexperimental liver metastases.II.A microcirculatory study of normaland tumor vessels of the liver with the use of perfused silicone rubber。Surgery 68:334-340,1970)
(7)在胰岛的β细胞内生长癌的转基因小鼠中,血管前的增生岛大小限于<1mm。在6-7周龄时,4-10%的岛变成新血管化,从这些岛长出大的血管化的肿瘤,肿瘤体积比血管前岛的体积大1000倍以上。(Folkman J,等,Induction of angiogenesis during the transition fromhyperplasia to neoplasia.Nature 339:58-61,1989)
(8)抗VEGF(血管内皮生长因子)的特异性抗体减小微血管密度,并能“显著或强烈”抑制3种依赖于VEGF作为它们的血管生成(在裸鼠内)唯一介体的人肿瘤的生长。该抗体在体外不抑制肿瘤细胞的生长。(Kim K J,等,Inhibition of vascular endothelial growth factor-induced angiogenesis suppresses tumor growth in vivo.Nature362:841-844,1993)。
(9)抗bFGF单克隆抗体能70%抑制依赖于分泌bFGF作为其唯一血管形成介体的小鼠肿瘤的生长。该抗体在体外不抑制肿瘤细胞的生长。(Hori A,等,Suppression of solid tumor growth byimmunoneutralizing monoclonal antibody against human basicfibroblast growth factor.Cancer Research,51:6180-6184,1991)
(10)腹膜内注射bFGF能通过刺激肿瘤内毛细内皮细胞的生长而增强初生肿瘤及其转移瘤的生长。这些肿瘤细胞本身缺乏bFGF的受体,bFGF不是体外肿瘤细胞的促分裂原。(Gross JL,等,Modulationof solid tumor growth in vivo by bFGF.Proc.Amer.Assoc.Canc.Res.31:79,1990)
(11)特异性血管生成抑制剂(AGM-1470)在活体内抑制肿瘤生长和转移,但在体外抑制肿瘤细胞增殖中活性小得多。它抑制血管内皮细胞增殖半极大值时的浓度比它抑制肿瘤细胞增殖时小4个数量级。(Ingber D,等,Angioinhibins:Synthetic analogues of fumagillin whichinhibit angiogenesis and suppress tumor growth.Nature,48:555-557,1990).还有间接临床迹象表明肿瘤生长与血管生成相关。
(12)可转移至玻璃体的人成视网膜细胞瘤发展成无血管球状体,这些球形体尽管能存活并结合3H-胸苷(当从剜出的眼中脱除而体外分析时),但限于<1mm3。
(13)卵巢癌转移至腹膜时是极小的无血管白色籽(1-3mm3)。除非其中的一个或多个变成新血管化,否则这些移植物很少会长到更大。
(14)乳腺癌中(Weidner N,等,Tumor angiogenesis correlates withmetastasis in invasive breast carcinoma.N.Engl.J.Med.324:1-8,1991,以及Weidner N,等,Tumor angiogenesis:A new significant andindependent prognostic indicator in earlystage breast carcinoma,JNatl.Cancer Inst.84:1875-1887,1992)与前列腺癌中(WeidnerN,Carroll PR,Flax J,Blumenfeld W,Folkman J.Tumor angiogenesiscorrelates with metastasis in invasive prostate carcinoma.AmericanJournal of Pathology,143(2):401-409,1993)新血管化的强度同将来转移的危险性高度相关。
(15)在形成新血管之前从人皮肤的黑素瘤的转移较罕见。一开始形成新血管就导致损伤增厚,转移的危险性增大。(Srivastava A,等,Theprognostic significance of tumor vascularity in intermediate thickness(0.76-4.0mm thick)skin melanoma.Amer.J.Pathol.133:419-423,1988)
(16)在膀胱癌中,血管生成肽bFGF的尿含量,是比细胞学对疾病的状况和程度更灵敏的指示剂。(Nguyen M,等,Elevated levels ofan angiogenic peptide,basic fibroblast growth factor,inurine of bladder cancer patients.J.Natl.Cancer Inst.85:241-242,1993)
因此,血管生成显然在癌的转移中起主要作用。如果能抑制或消除或者控制和调节该血管生成活性,那么肿瘤即使存在也不会生长。在疾病状态中,防止血管生成能避免新微血管系统侵入而引起的损伤。针对控制血管生成过程的疗法会导致消除或减轻这类疾病。
因此,需要的是能抑制内皮细胞增殖如不希望的血管生长、特别是在肿瘤中生长的组合物和方法。还需要检验、测定和定位该组合物的方法。该组合物应能抑制转移前肿瘤中内源性生长因子的活性并防止肿瘤内毛细血管的形成,因而抑制肿瘤的生长。该组合物、组合物的部分和该组合物的特异性抗体,也应能调节其它血管生成过程如创伤愈合和再生中毛细血管的形成。抑制血管生成的该组合物和方法应优选是无毒的和产生很少的副作用。还需要检验、测定和定位该组合物的结合位点以及该组合物的生物合成位点的方法。该组合物和组合物的部分应能与用于放射性和非放射性标记的其它分子缀合。
发明内容
本发明包括应用分离的纤溶酶原Kringel 5区来抑制内皮增殖活性的方法。具有抑制活性的该分离的Kringle 5肽片段包括约80个氨基酸的序列:

本发明的内皮细胞增殖肽相当于从人纤溶酶原产生的肽片段,它始于人纤溶酶原的约氨基酸462再延伸约80个氨基酸。
本发明还包括诊断方法和治疗方法,用于检验体液中该抑制肽存在与否,并对于需要治疗上有效量的这类化合物以调节内皮细胞增殖的患者施用该肽或特异性地结合该肽的抗体。此外,该抑制肽可在体外与增殖内皮细胞培养物一起使用以测试减轻该肽的抑制效果的化合物,即筛选生长因子或能克服或改变内皮细胞增殖的抑制作用的其它化合物。
因此,本发明的一个目的是提供含内皮细胞增殖抑制剂的组合物,该抑制剂包括人纤溶酶原的约80个氨基酸的肽片段,该片断基本上相当于始于人纤溶酶原的氨基酸462的Kringle 5区。
本发明的另一个目的是提供治疗由内皮细胞增殖、特别是血管生成介导的疾病和过程的方法。
本发明的又一个目的是提供检测体液或组织中抑制剂的存在及其含量的诊断方法或预后方法和试剂盒。
本发明的又一个目的是提供治疗由血管生成介导的疾病和过程的方法和组合物,其中的疾病和过程包括但不限于:血管瘤、固体肿瘤、白血病、转移、毛细管扩张、牛皮癣、硬皮病、生脓性肉芽肿、心肌的血管生成、蚀斑新血管化、冠状侧突、大脑侧突、动静脉畸形、局部缺血性肢血管生成、角膜病、潮红、新血管性青光眼、糖尿病性视网膜病、晶状体后的纤维组织形成、关节炎、糖尿病性新血管化、黄斑变性、创伤愈合、胃溃疡、螺杆菌(Helicobacter)相关性疾病、骨折、瘢痕瘤、血管生成、血细胞生成、排卵、行经、胎盘形成和猫抓热。
本发明的另一个目的是提供用于治疗或抑制癌的生长的组合物。
本发明的一个目的是提供调节或模拟酶的生产或活性的化合物,其中的酶在活体内或活体外产生本发明的抑制剂。

本发明提供的组合物和方法可有效地抑制内皮细胞增殖,调节血管生成,并抑制不需要的血管生成、特别是与肿瘤生长相关的血管生成。本发明包括一种蛋白质内皮细胞增殖抑制剂,其特征在于可得自人纤溶酶原Kringle 5的约80个氨基酸的序列。抑制剂的该氨基酸序列可因种类不同而稍有变化。应懂得该活性抑制剂分子中的氨基酸数是可变的,所以具有内皮抑制活性的所有密切同源性氨基酸序列都应认为包括于本发明中。
本发明通过对患有不希望的内皮细胞增殖的人或动物施用包括能在体外分析中抑制内皮细胞增殖的人纤溶酶原约Kringle 5的组合物,而提供方法和组合物以治疗由不希望的和未受控的上皮细胞增殖如血管生成介导的疾病和过程。该分离的蛋白质优选至少约80%的纯度,更优选至少约90%的纯度,最优选至少约95%的纯度。本发明特别适于治疗或抑制肿瘤的生长。对患有前血管化转移瘤的人或动物施用该抑制剂有助于防止那些肿瘤的生长和扩增。
本发明还包括编码内皮细胞增殖抑制剂的DNA序列,含有编码内皮细胞增殖抑制剂的DNA序列的表达载体,以及含有一种或多种表达载体(该表达载体含编码该抑制剂的DNA序列)的细胞。本发明进一步包括基因疗法,借此将编码该内皮细胞增殖抑制剂的DNA序列引入患者以在活体内修饰抑制剂含量。
本发明还包括诊断方法和试剂盒,以检验和测定生物流体和组织中的内皮细胞增殖抑制剂,并定位组织和细胞中的该抑制剂。这种诊断方法和试剂盒可呈本领域普通技术人员熟知的任意结构。本发明还包括该内皮细胞增殖抑制剂及其部分的特异性抗体,以及抑制该内皮细胞增殖抑制剂的特异性抗体的结合的抗体。这些抗体可以是多克隆抗体或单克隆抗体。该内皮细胞增殖抑制剂的特异性抗体可用于诊断试剂盒中以检验该抑制剂的存在及其含量,它可诊断或预后由血管生成介导的癌或其它疾病的发作或复发。还可对人或动物施用内皮细胞增殖抑制剂的特异性抗体以被动地免疫该人或动物而抗该抑制剂,于是减小血管生成的抑制。

本发明还包括抗抑制剂受体特异性抗体,它与该抑制剂的受体结合并向细胞传导适当的信号,而且起兴奋剂或拮抗剂的作用。

该抑制剂可从纤溶酶原如人纤溶酶原分离,或者通过化学或生物方法(例如细胞培养,重组基因表达,肽合成,以及纤溶酶原或纤溶酶的体外酶促催化以生成活性抑制剂)合成。重组技术包括应用聚合酶链反应(PCR)从DNA源进行基因扩增,以及应用逆转录酶/PCR从RNA源进行基因扩增。

术语“基本上相似的”或“基本上同源的”用于表示该抑制剂的氨基酸和核酸序列时,指这种氨基酸序列,即它具有内皮细胞增殖抑制活性和大约14kD的分子量,它还具有与某蛋白质的高度序列同源性,所述蛋白质具有本文公开的特定的N端氨基酸序列;或者指编码内皮细胞增殖抑制剂的核酸序列,所述抑制剂具有大约14kD的分子量和与某氨基酸的高度同源性,该氨基酸具有本文公开的特定的N端氨基酸序列。

本发明还包括体液和组织中抑制剂的检验,旨在诊断或预后例如癌的疾病。本发明还包括细胞和组织中抑制剂结合位点和受体的检验。本发明还包括治疗或预防血管生成的疾病和过程的方法,这类疾病包括但不限于关节炎和肿瘤,是运用下列方法进行治疗或预防的,即通过刺激该抑制剂的产生,和/或通过对患者施用分离的抑制剂,或所需纯化了的抑制剂,或抑制剂兴奋剂或拮抗剂,和/或抑制剂特异性抗血清或针对抑制剂特异性抗血清的抗血清。另外的治疗方法包括施用该抑制剂,其生物活性片段,抑制剂类似物,抑制剂特异性抗血清,或与细胞毒性剂连接的抑制剂受体兴奋剂和拮抗剂。
应用特异性地结合该抑制剂的抗体的被动抗体疗法可被用于调节血管生成依赖性过程例如生殖,发育,以及创伤愈合和组织修复。此外,针对抑制剂特异性抗体的Fab区的抗血清可被施用以阻止内源性抑制剂特异性抗血清结合抑制剂的能力。
本发明还包括基因疗法,借此在患者体内调节编码该抑制剂的基因。转移或递送DNA至细胞以表达基因产物蛋白质的各种方法,或称为基因疗法,都公开于Gene Transfer into Mammalian Somatic Cellsin vivo,N.Yang,Crit.Rev.Biotechn.12(4):335-356(1992),该文献并入本文作参考。基因疗法包括将DNA序列掺入体细胞或种系细胞用于来自体内的或活体内的治疗。基因疗法起这样的作用,即置换基因、增大正常的或异常的基因功能,以及对抗传染病和其它病理。
用基因疗法处理这些医疗问题的对策包括:治疗策略,例如鉴定缺损基因,然后加入功能性基因以置换缺损基因的功能或增强弱小功能的基因;或者预防策略,例如加入产物蛋白质的基因,该蛋白质可治疗病况或将会使组织或器官对于治疗方案更敏感。预防策略的一个实例是,可将编码该抑制剂的核酸序列置于患者中,于是防止血管生成的发生;或者可插入使肿瘤细胞对辐射更敏感的基因,然后辐射该肿瘤就会增大对肿瘤细胞的杀伤力。
本发明设想了转移抑制剂DNA或抑制剂调节序列的许多方案。转染启动子序列,而不是常见的与该抑制剂特异性地结合的序列,或其它能增大该抑制剂蛋白质产量的序列,也设想为基因治疗的方法。该技术的一个实例见于Transkaryotic Therapies,Inc.of Cambridge,Massachusetts,应用同源重组将可开启促红细胞生成素基因的“基因开关”插入细胞。参见Genetic Engineering News,April 15,1994。这种“基因开关”可被用于激活不能正常表达该抑制剂(或该抑制剂的受体)的细胞中的该抑制剂(或抑制剂受体)。
用于基因疗法的基因转移方法归为三大类-物理法(例如电穿孔法,直接基因转移和粒子轰击),化学法(基于脂质的载体,或其它非病毒性载体)和生物法(得自病毒的载体和受体摄取)。例如,可应用非病毒性载体,它们包括DNA涂覆的脂质体。这种脂质体/DNA复合体可被直接静脉内注入患者。人们认为,该脂质体/DNA复合体在肝内被浓缩,它们在肝内将DNA递送至巨噬细胞和枯氏细胞。这些细胞能长久生存因而能长期表达该递送的DNA。此外,可将载体或基因的“裸”DNA直接注入所希望的器官、组织或肿瘤以定向递送治疗性DNA。
基因治疗方法学也可通过递送位点来描述。递送基因的基本方法包括:来自体内基因转移,活体内基因转移,和体外基因转移。在来自体内的基因转移中,细胞取自患者再在细胞培养物中生长。将DNA转染入细胞,该转染的细胞数量扩大后再重植入患者。在体外基因转移中,转化的细胞是生长于培养物中的细胞,如组织培养细胞,而不是得自特定的患者的特定细胞。转染这些“实验细胞”,选择转染的细胞并扩充后将它们植入患者或另作它用。
活体内的基因转移包括当这些细胞在患者体内时将DNA引入患者的细胞。方法包括应用病毒介导的基因转移,运用非感染性病毒将基因递送入患者或者将裸DNA注入患者的位点,于是该DNA以一定细胞百分数被吸收,在这些细胞中基因产物蛋白质被表达。此外,本文描述的其它方法(如“基因枪”的应用)可被用于体外插入内皮细胞增殖抑制剂DNA或抑制剂调节序列。
基因疗法的化学方法可涉及一种基于脂质的化合物,不一定是脂质体,来运送DNA穿过细胞膜。脂质转染剂或细胞转染剂,与荷负电的DNA结合的基于脂质的正离子,形成一种复合体能穿过细胞膜而将DNA送入细胞内部。另一种化学方法应用基于受体的胞吞作用,它包括将特异性配体与细胞表面受体结合并包膜,再将它转运穿过细胞膜。配体与DNA结合后再将该整个复合体转运入细胞。将该配体基因复合体注入血流,然后具有该受体的靶细胞将特异性地结合该配体并将该配体-DNA复合体转运入该细胞。
许多基因治疗方法应用病毒载体将基因插入细胞。例如,在来自体内的方法中业已应用改变的逆转录病毒载体来将基因引入外周的和肿瘤浸润的淋巴细胞、肝细胞、表皮细胞、肌细胞或其它体细胞。然后将这些改变的细胞引入患者以提供得自插入的DNA的基因产物。
病毒载体也已被用于通过活体内方案将基因插入细胞。为了指导组织特异性的外源基因表达,可应用已知是组织特异性的顺式作用调节元件或启动子。也可通过现场递送DNA或病毒载体至活体内特定的解剖位点而达此目的。例如,活体内基因转移至血管是通过在体外将转导的内皮细胞植入动脉壁上选定的位点而实现的。病毒感染也表达该基因产物的周围细胞。病毒载体例如通过导管可被直接递送至活体内的位点,于是只让一定的区被该病毒感染,从而提供长期、位点特异性的基因表达。应用逆转录病毒载体的活体内基因转移也已通过将改变了的病毒注入通向器官的血管而在乳腺组织和肝组织中得到了证实。
业已被用于基因疗法的病毒载体包括但不限于:逆转录病毒,其它的RNA病毒如脊髓灰质炎病毒或新培斯病毒,腺病毒,腺相关性病毒,疤疹病毒,SV40,疫苗和其它DNA病毒。复制缺损型鼠逆转录病毒载体是最广泛应用的基因转移载体。鼠白血病逆转录病毒包括与核心蛋白质和聚合酶(pol)复合的单链RNA,被包入蛋白质心(gag)并被确定宿主范围的糖蛋白包膜(env)包围。逆转录病毒的基因组结构包括这些gag、pol和env基因,它们被5′和3′长末端重复序列(LTR)围住。逆转录病毒系统利用下列事实,即假设供给的病毒结构蛋白在包装细胞系中呈反式结构,则含该5′和3′LTRs的最小载体和包装信号足以允许载体包装、感染和整合入靶细胞。用于基因转移的逆转录病毒载体的基本优点包括:在多数种类的细胞中有效的感染和基因表达,整合入靶细胞染色体DNA的准确的单拷贝载体,以及该逆转录病毒基因组的易操作性。
腺病毒包括与核心蛋白复合且被衣壳蛋白包裹的线型、双链DNA。分子病毒学的进展已导致能利用这些有机体的生物学以产生能在活体内将新型基因序列转导入靶细胞的载体。基于腺病毒的载体将高水平地表达基因产物肽。腺病毒载体甚至在低滴度的病毒下也具有高效感染性。此外,该病毒作为无细胞病毒颗粒是完全感染性的,所以不必注射生产者细胞系。腺病毒载体的另一个潜在优点是实现活体内长期表达异源基因的能力。
DNA递送的机械方法包括融合的脂质载体如适于膜融合术的脂质体或其它载体,掺入阳离子脂质的DNA的脂质粒子如脂质转染剂,聚赖氨酸介导的DNA的转移,DNA的直接注射,例如将DNA显微注入生殖细胞或体细胞,气动递送DNA涂覆的粒子,例如用于“基因枪”的金粒子,以及无机化学方法如磷酸钙转染。另一种方法,即配体介导的基因疗法,包括将DNA与特异性配体复合而形成配体-DNA缀合物,以指导该DNA至特定的细胞或组织。
已发现将质粒DNA注入肌细胞可得到被转染百分率高的细胞,这些细胞能持续表达标记基因。质粒的DNA能或不能整合入细胞的基因组。已转染的DNA的非整合作用会允许在终末分化的、非增殖性组织中长时间转染和表达基因产物蛋白质,而不用担心细胞基因组或线粒体基因组中的突变插入、缺失或改变。长期但不必永久的将治疗基因转移入特定的细胞可提供遗传病的疗法或用于预防。可定期地再注射DNA以维持基因产物水平而不会在受体细胞的基因组中出现突变。外源DNAs的非整合作用可允许一个细胞内存在数种不同的外源DNA构建物,所有这些构建物表达各不相同的基因产物。
粒子介导的基因转移方法首先被用于转化植物组织。应用粒子轰击装置或“基因枪”,产生的动力将DNA涂覆的高密度粒子(例如金或钨)加速至高速度因而能穿透靶器官、组织或细胞。粒子轰击可被用于体外系统,或者应用来自体内的或活体内的方法将DNA引入细胞、组织或器官。
基因转移的电穿孔法应用电流使细胞或组织对于电穿孔介导的基因转移敏感。应用一定场强的短暂电脉冲来增大膜的通透性使得DNA分子能穿透入细胞。该技术可用于体外系统中,或者应用来自体内的或活体内的方法将DNA引入细胞、组织或器官。
活体内载体介导的基因转移可被用于将外源DNA转染入细胞。该载体-DNA复合体可被方便地引入体液或血流然后被位点特异性地指导至体内靶器官或组织。可应用脂质体和聚阳离子,如聚赖氨酸、脂质转染剂或细胞转染剂。可开发细胞特异性的或器官特异性的脂质体,于是由脂质体携带的外源DNA将被靶细胞吸收。注射被导向某些细胞上特定的受体的免疫脂质体可被用作将DNA插入携带该受体的细胞的便利方法。已被应用的另一个载体系统是脱唾液酸糖蛋白/聚赖氨酸缀合物系统,它适于在活体内基因转移中携带DNA至肝细胞。
已转染的DNA也可与其它类型载体复合,于是该DNA被携带至受体细胞,然后存留于胞质或核质中。在特异性地工程化的小泡复合体中DNA可与载体核蛋白偶合而被直接携带入细胞核。
本发明的抑制剂的基因调节可这样实现,即通过施用与该抑制剂的基因结合的化合物,或与该基因相关的控制区,或其相应的RNA转录物以修饰转录或翻译的速度。此外,用编码该抑制剂的DNA序列转染的细胞可被施用给患者以提供活体内的抑制剂源。例如,细胞可用含编码该抑制剂的核酸序列的载体转染。
本文应用的术语“载体”表示可含有特异性的核酸序列或与之相关的载体,它的作用是将该特异性的核酸序列转运入细胞。载体的实例包括质粒和感染性微生物如病毒,或非病毒载体如配体-DNA缀合物、脂质体、脂质-DNA复合体。可能希望把包括内皮细胞增殖抑制剂DNA序列的重组DNA分子有效地与表达控制序列连接以形成能表达该抑制剂的表达载体。转染的细胞可以是得自患者的正常组织、患者的患病组织的细胞,或者可以是非患者细胞。
例如,从患者摘除的肿瘤细胞可用能表达本发明的抑制剂蛋白质的载体转染,再回引入患者。转染后的肿瘤细胞在患者中产生抑制剂的量能抑制肿瘤的生长。患者可以是人或者非人的动物。此外,抑制剂DNA可以不需借助于载体而被直接注入患者。具体地说,抑制剂DNA可被注入皮肤、肌肉或血液。
抑制剂表达可延续很长一段时间或者可定期施用抑制剂DNA以维持细胞、组织或器官或生物流体内所需的抑制剂蛋白质水平。
虽然不想受下列假设的束缚,但认为当肿瘤变成血管源性的时候,它释放一种或多种生成血管的肽(例如aFGF、bFGF、VEGF、IL-8、GM-CSF等),它们从血管外的方向局部地作用于初生肿瘤附近的靶内皮,且不循环(或者循环较短的半衰期)。这些生成血管的肽必需产生的量应足以克服内皮细胞抑制剂(血管生成的抑制剂)的作用,以便初生肿瘤继续扩增它的数量。一旦这种初生肿瘤生长良好,它就继续向循环系统释放内皮细胞抑制剂。根据这一假设,这些抑制剂在距离初生肿瘤遥远的地方从血管内的方向作用于转移瘤的靶毛细内皮,并继续循环。因此,当遥远的转移瘤恰好可能开始激活血管生成时,它附近的毛细内皮被进入的抑制剂抑制。
应用类似的技术实现本发明的内皮细胞增殖抑制剂的生产可运用包括下列步骤的重组DNA技术来完成,即(1)鉴定和纯化如本文所述和由附图例证的抑制剂,(2)确定纯化了的抑制剂的N端氨基酸序列,(3)通过合成产生该抑制剂序列的5′和3′DNA寡核苷酸引物,(4)应用聚合酶扩增该抑制剂基因序列,(5)将扩增了的序列插入适当的载体如表达载体,(6)将含基因的载体插入能表达该抑制剂基因的微生物或其它表达系统,以及(7)分离该重组生产的抑制剂。适当的载体包括病毒的、细菌的和真核的(例如酵母)表达载体。实验手册中更完整地描述了如上技术,这种手册例如“Molecular Cloning:A LaboratoryManual”Second Edition by Sambrook等,Cold Spring Harbor Press,1989,它被并入本文作参考。
然而生产该抑制剂或其生物活性片段的另一种方法是肽合成法。该抑制剂的氨基酸序列例如可通过自动化肽测序方法来测定。另外,一旦编码抑制剂的基因或DNA序列例如通过上述方法而得以分离,该DNA序列就可应用本领域熟知的人工或自动测序方法来测定。核酸序列反过来提供有关氨基酸序列的信息。
一旦知道该肽的氨基酸序列,就能通过本领域熟知的技术合成肽片段,例如通过“Solid Phase Peptide Synthesis:A PracticalApproach”E.Atherton和R.C.Sheppard,IRL Press,Oxford,England。可合成很多片段然后将它们连在一起而形成更大的片段。还可用在特定位置处氨基酸取代制备这些合成肽片段以便在体外或者在活体内测试兴奋活性和拮抗活性。可应用与组织高亲和力结合的肽片段在亲和柱上分离受体结合物抑制剂。
该抑制剂在治疗例如血管生成的疾病或过程中有疗效,该血管生成是由内皮细胞增殖介导的或者涉及内皮细胞增殖。本发明包括治疗血管生成介导的疾病的方法,它应用有效量的抑制剂,或其生物活性片段,或总体地具有抗血管生成活性的抑制剂片段的组合体,或抑制剂兴奋剂和拮抗剂。血管生成介导的疾病包括但不限于:固体肿瘤;血生肿瘤如白血病;肿瘤转移;良性肿瘤如血管瘤,听觉神经瘤,神经纤维瘤,沙眼,以及生脓性肉芽肿;类风湿性关节炎;牛皮癣;眼血管生成病,例如糖尿病性视网膜病,早熟性视网膜病,黄斑变性,角膜移植排异反应,新血管性青光眼,晶状体后的纤维组织形成,湖红;Osler-Webber综合征;心肌血管生成;蚀斑新血管化;毛细管扩张;血友病患者关节;血管纤维瘤;以及创伤粒化。
该抑制剂适用于治疗内皮细胞的过量刺激或异常刺激的疾病。这类疾病包括但不限于:肠粘连、动脉粥样硬化、硬皮病、以及肥大性疤痕即疤痕疙瘩。该抑制剂可通过防止胚胎植入所需的血管化而用作出节育剂。该抑制剂适用于治疗以血管生成为病理结果的疾病如猫抓病(Rochele minalia quintosa)和溃疡(ulcers)(幽门螺杆菌(Helicobacterpylori))。
该抑制剂的合成肽片段具有多种用途。与受体结合的该肽被放射性标记并用于采用放射自显影技术和膜结合技术的显示和定量分析结合位点,其中的受体能高度特异性和高亲合力地结合该抑制剂。
此外,短寿命同位素标记的抑制剂或其肽片段能在活体内显示受体结合位点,它应用正电子成象术或其它现代放射显影术以便用抑制剂结合位点确定肿瘤的位置。
将细胞毒性剂(如蓖麻毒蛋白)与该抑制剂及其高亲和性肽片段连接,从而提供一种破坏结合有该抑制剂的细胞的工具。这些细胞可见于很多部位,包括但不限于微转移瘤和初生肿瘤。与细胞毒性剂连接的肽以旨在最大程度地递送至所需部位的方式被灌注。例如,可通过插入脉管的插管供给靶位点或者直接插入靶的插管来完成递送。这类试剂还能以受控制的方式通过与灌注插管偶联的渗透泵而被递送。抑制剂拮抗剂的组合体可与血管生成的刺激物一起应用以增加组织的血管化。这种治疗方案提供消灭转移癌的有效方法。
该抑制剂可与治疗疾病的其它组合物和方法结合使用。例如,肿瘤通常可用与该抑制剂结合的手术、放射或化疗方法治疗,然后可接着给患者施用该抑制剂以延长微转移瘤的潜伏期并稳定和抑制任何残余初生肿瘤的生长。此外,该抑制剂、其片段、抑制剂特异性抗血清、抑制剂受体兴奋剂和拮抗剂、或其组合体,与药物上可接受的赋形剂,以及任选的缓释基质如可生物降解的聚合物组合而形成治疗组合物。
本文所用的缓释基质是一种由通常为聚合物的原料构成的基质,它们可通过酶促水解或酸解/碱解或通过溶解作用而降解。一旦插入体内,该基质就受酶和体液的作用。该缓释基质优选选自生物相容性原料如脂质体,聚丙交酯(聚乳酸),聚乙交酯(乙醇酸的聚合物),聚(丙交酯-乙交酯)(乳酸和乙醇酸的共聚物),聚酐,聚(原酸)酯,多肽,透明质酸,胶原,硫酸软骨素,羧酸,脂肪酸,磷脂,多糖,核酸,聚氨基酸,氨基酸如苯丙氨酸、酪氨酸、异亮氨酸,多核苷酸,聚乙烯基丙烯,聚乙烯基吡咯烷酮和聚硅氧烷。优选的可生物降解基质是下列基质之一:聚丙交酯、聚乙交酯、或聚(丙交酯一乙交酯)(乳酸和乙醇酸的共聚物)。
本发明的血管生成-调节治疗组合物可以是固体、液体或气溶胶,且可以任意已知的施用途径施用。固体治疗组合物的实例包括丸、乳膏和可植入的剂量单位。丸可经口施用,治疗性乳膏可以局部施用。可植入的剂量单位可以局部施用,例如在肿瘤部位施用,或者可植入它以便在全身释放治疗性血管生成-调节组合物,例如植入皮下。液体组合物的实例包括:适于皮下注射、静脉注射、动脉注射的调和物,以及适于局部和眼内施用的调和物。气溶胶调和物的实例包括对肺施用的吸入剂调和物。
本发明的抑制剂蛋白质还可被用于产生对该抑制剂及其受体具有特异性的抗体。这类抗体可以是多克隆抗体或单克隆抗体。这些特异性地结合该抑制剂或抑制剂受体的抗体可用于本领域普通技术人员熟知的诊断方法和试剂盒中以检验或定量分析体液或组织中该抑制剂含量或抑制剂受体的含量。这些试验结果可用于诊断或预测癌和其它血管生成介导的疾病的发生或复发。
该抑制剂还可用于开发诊断方法和试剂盒以检验和定量分析能结合该抑制剂的抗体。这些试剂盒应能检验循环的抑制剂特异性抗体。带有这类循环的抗抑制剂抗体的患者很可能长有多种肿瘤和癌,以及在治疗或缓解期之后癌很可能会复发。这些抗体的Fab片段可用作抗原以产生能用于中和抗抑制剂抗体的抗抑制剂特异性Fab-片段抗血清。这种方法会减少由抗抑制剂抗体对循环抑制剂的脱除,于是有效地提高循环抑制剂的含量。
本发明的另一方面是阻止过量内源性抑制剂的作用的方法。这可通过用对系统中不希望有的抑制剂呈特异性的抗体来被动地免疫人或动物而实现。这种治疗方法在治疗异常的排卵,行经和胎盘形成,以及血管发生方面会很重要。这就为检验抑制剂的除去对转移过程的效果提供了有用工具。抑制剂特异性抗体的Fab片段含有抑制剂的结合位点。应用本领域技术人员已知的技术从抑制剂特异性抗体分离该片段。抑制剂特异性抗血清的Fab片段被用作抗原以引起抗Fab片段血清的生产。灌注抗该抑制剂特异性的Fab片段的这种抗血清能防止该抑制剂与抑制剂抗体的结合。通过中和内源性抗抑制剂抗体通过阻止抑制剂与抗抑制剂的Fab片段结合从而达到治疗效果。这种治疗的纯效果是强化内源性循环抑制剂到达靶细胞的能力,于是减少转移瘤的扩散。
应明白,本发明旨在包括具有内皮细胞增殖抑制活性的抑制剂的任意衍生物。本发明包括整个抑制剂蛋白质、抑制剂蛋白质的衍生物和抑制剂蛋白质的生物活性片段。它们包括具有抑制活性的蛋白质,蛋白质带有氨基酸取代物或在氨基酸功能基上连接有糖或其它分子。本发明还包括编码该抑制剂和抑制剂受体的基因,以及被这些基因表达的蛋白质。
上述具有抑制活性的蛋白质和蛋白质片段可以分离的和大体上纯化的蛋白质和蛋白质片段的形式提供,它们处于应用本领域普通技术人员已知的调和方法调制的药物上可接受的调和物中。这些调和物可用标准途径施用。一般地,这类组合物可通过如下途径施用:局部的、经皮的、腹膜内的、颅内的、脑室内的、脑内的、阴道内的、宫内的、经口的、经直肠的或胃肠外的(例如静脉内的、脊柱内的、皮下的或肌内的)途径。此外,可将该抑制剂掺入可生物降解的聚合物使之缓释该化合物,这类聚合物被植入需要药物递送的地方附近,例如在肿瘤部位,或者植入能全身缓释该抑制剂的地方。还可应用渗透微型泵提供受控的递送高浓度的该抑制剂,即通过插管递送至感兴趣的部位例如直接递送入转移生长癌或递送入该肿瘤的血管。这类可生物降解的聚合物及其应用例如详细地描述于Brem等,J.Neurosurg.74:441-446(1991),该文献全文并入本文作参考。
本发明的抑制剂剂量将依赖于受治疗的疾病状态或情况以及其它临床因素如人或动物的体重和健康状况,还有该化合物的施用途径。至于治疗人或动物,可施用约0.5mg/kg至500mg/kg的该抑制剂。依赖于该抑制剂在特定动物或人中的半衰期,该抑制剂的施用频率可在每天数次至每周一次之间。应懂得,本发明的应用方面包括人和兽医应用。本发明方法期望单次施用和多次施用,可同时地或在持续的一段时间内。
该抑制剂调和物包括适合于下列施用途径的那些,即经口的,经直肠的,经眼的(包括玻璃体内的或眼房内的),经鼻的,局部的(包括颊的和舌下的),宫内的,阴道的或胃肠外的(包括皮下的、腹膜内的、肌内的、静脉内的、皮内的、颅内的、气管内的和硬膜外的)施用。该抑制剂调和物可便利地呈单位剂量形式并可通过惯常的药物技术制备。这类技术包括将活性成分与药物载体或赋形剂结合的步骤。大体上,可这样制备这类调和物,即将活性成分与液态载体或细碎的固态载体或与二者均匀地、紧密地结合,然后需要的话将该制品加工成形。
适合胃肠外施用的调和物包括:水性的和非水的无菌注射液,它可含有抗氧剂、缓冲剂、抑菌剂和能使该调和物与预期的受体血液等渗的溶质;以及水性的和非水的无菌悬浮液,它可包括悬浮剂和增稠剂。这类调和物可盛于单位剂量或多剂量容器中,例如密封的安瓿和小瓶,可在冷冻干燥的(冻干的)条件下贮存,就在应用前只需加入无菌液态载体,例如注射用水。现场注射的溶液或悬浮液可从前述种类的无菌粉末、颗粒和片配制。
优选的单位剂量调和物包括施用成分的日剂量或单位、日亚剂量或其适当的部分。应懂得,除了上面专门提到的成分之外,本发明的调和物还可包括本领域有关所述类型的调和物惯用的其它试剂。细胞毒性剂可任选掺入或以其它方式结合抑制剂蛋白质或其生物功能性肽片段,以给患者提供双重治疗。
本发明的血管生成抑制肽可在标准的微量化学设备中合成,并可用HPLC和质谱法检测纯度。肽合成法、HPLC纯化法和质谱法是本领域技术人员已知的。抑制剂肽和抑制剂受体肽还可在重组大肠杆菌(E.coli)或酵母表达系统中生产,再用柱色谱法纯化。
可合成该完整抑制剂分子的不同肽片段以供包括但不限于下列方面的数种应用:作为抗原供开发特异性抗血清,作为在抑制剂结合位点有活性的兴奋剂和拮抗剂,作为与细胞毒性剂连接或用于结合细胞毒性剂的肽供定向杀伤结合该抑制剂的细胞。包括这些肽的氨基酸序列基于它们在该分子外部区的位置来选择并易于结合抗血清。该抑制剂的氨基端和羧基端,以及该分子的中部区分别显示在待合成的片段之间。
利用蛋白质序列数据库如GenBank、Brookhaven Protein、SWISS-PROT和PIR将这些肽序列与已知序列比较以确定可能的序列同源性。该信息有助于消除表现为同其它分子高度的序列同源性的序列,于是增强开发该抑制剂的抗血清、兴奋剂和拮抗剂中高度特异性的潜能。
可应用标准方法将抑制剂和抑制剂衍生的肽与其它分子偶联。该抑制剂的氨基端和羧基端都含有酪氨酸残基和赖氨酸残基,都可用多种方法进行同位素标记和非同位素标记,例如应用惯常技术(酪氨酸残基-氯胺T,碘剂(iodogen),乳过氧化物酶;赖氨酸残基-Bolton-Hunter试剂)进行放射性标记。本领域技术人员熟知这些偶联技术。也可将酪氨酸或赖氨酸加入不具备这些残基的片段以促进对肽上的反应性氨基和羟基的标记。偶联技术的选择基于氨基酸上可获得的功能基,这些功能基包括但不限于氨基、巯基(sulfhydral)、羧基、酰胺、酚和咪唑。进行偶联的各种试剂包括戊二醛、重氮化的联苯胺、碳化二亚胺和对-苯醌等。
将抑制剂肽与同位素、酶、载体蛋白质、细胞毒性剂、荧光分子、化学发光剂、生物发光剂和其它化合物化学偶联供多种应用。偶联反应的效率可用适合于具体反应的不同方法测定。例如,应用氯胺T和高比活性的Na125I实现用125I对抑制剂肽的放射性标记。用焦亚硫酸钠终止该反应,将混合物在一次性使用的柱上脱盐。从柱上洗脱标记的肽并收集级分。从各级分采等分样再用γ计数仪测定放射性活度。按该方法将未反应的Na125I从标记的抑制剂肽分离。贮存具有最高比放射性活度的肽级分供后续应用例如分析它与抑制剂抗血清的结合能力。
肽缀合的另一个应用是生产多克隆抗血清。例如,应用戊二醛将含赖氨酸残基的抑制剂肽与已纯化的牛血清白蛋白连接。通过测定放射性标记肽的掺入而确定该反应的效率。通过透析分离未反应的戊二醛和肽。贮存该缀合物供后续应用。
可产生对于该抑制剂、抑制剂类似物、该抑制剂的肽片段和抑制剂受体呈特异性的抗血清。在肽合成和纯化之后,应用本领域技术人员已知的现有技术产生单克隆和多克隆抗血清。例如,可在兔、绵羊、山羊或其它动物内产生多克隆抗血清。将与载体分子如牛血清白蛋白缀合的抑制剂肽或抑制剂本身与佐剂混合物结合,乳化,皮下注入背部、颈部、胁腹和有时爪垫中的多个部位。在定期的间隔内如每2至4周加强注射。每次注射约7~10天后,通过静脉穿刺术获取血液样本,例如应用扩张术后的耳边沿静脉。在4℃下放置过夜使血液样本凝固,再在4℃和约2400Xg下离心约30分钟。取出血清,分成等分样,在4℃下贮存供立即使用或在-20~-90℃下贮存供后续的分析。
分析得自生产多克隆抗血清的所有血清样本或得自生产单克隆抗血清的中层样本(media samples)以测定抗体滴定。通过数种方法确定滴度,例如,应用斑点印迹法和密度分析法;还应用放射性标记的肽-抗体复合体沉淀法,其中运用蛋白质A、次级抗血清、冷乙醇或活性炭-葡聚糖,接着用γ计数仪测定活性。最高滴度的抗血清也用可商购的亲和柱纯化。在亲和柱中使抑制剂肽与凝胶偶联。抗血清样本流过该柱,而抗抑制剂抗体保持与该柱结合。然后洗脱这些抗体,收集并估测滴度和特异性。
测试最高滴度的抑制剂特异性抗血清以确定如下性能:a)抗原的最高特异性结合和最低非特异性结合的最佳抗血清稀释度,b)在标准置换曲线中结合增加量的抑制剂肽的能力,c)与相关的肽和相关种类的蛋白质潜在的交叉反应性,d)检测血浆、尿、组织的提取物中和细胞培养基中的抑制剂肽的能力。
测定抑制剂和抑制剂受体的试剂盒,也属于本发明的部分。进一步检验这种抗血清,即具有最高滴度和特异性并能检测血浆、尿、组织的提取物中和细胞培养基中的抑制剂肽,以证实易于应用试剂盒迅速、可靠、灵敏和特异性地测定和定位抑制剂。这些分析试剂盒包括但不限于下列技术:竞争性和非竞争性分析,放射免疫分析,生物发光和化学发光分析,荧光测定分析,夹心分析,免疫放射分析,斑点印迹法,包括ELISA的酶联分析、微量滴定板法,供快速监测尿或血液用的抗体涂覆的条或测杆,以及免疫细胞化学法。对于每种试剂盒,确定了其分析的范围、灵敏度、精确度、可靠性、特异性和重现性。确定了标准的置换曲线或活性曲线中20%、50%和80%的点上分析内和分析间变化。
常用于研究和临床中的分析试剂盒的一个实例是放射免疫分析(RIA)试剂盒。下文阐述了抑制剂RIA。在成功地放射性碘化并纯化抑制剂或抑制剂肽之后,在数个稀释度下将具有最高滴度的抗血清加入含有相对恒定量的放射性活度的试管,例如适当缓冲系统中的10,000cpm。其它试管含有缓冲剂或免疫前血清以测定非特异性结合。在4℃下保温24小时后,加入蛋白质A并旋转试管,在室温下保温90分钟,再在4℃和约2000-2500Xg下离心以沉淀同标记的抗原结合的抗体复合体。吸出上清液,用γ计数仪计数离心片中的放射性活度。扣除非特异性结合后,进一步鉴定结合约10%~40%标记肽的抗血清稀释度。
然后,通过将已知量的肽加入含放射性标记的肽和抗血清的试管而估测用于培养抗血清的抑制剂肽的稀释范围(约0.1pg~10ng)。再次保温例如24~48小时之后,加入蛋白质A,将试管离心,吸出上清液并计数离心片中的放射性活度。用未标记的抑制剂肽(标准物)置换放射性标记的抑制剂肽的结合而提供标准曲线。将数种浓度其它抑制剂肽片段、得自不同物种的抑制剂和同源性肽加入分析试管以鉴定该抑制剂抗血清的特异性。
制备各种组织的提取物,这些组织包括但不限于初生和次生肿瘤、Lewis肺癌、抑制剂生产细胞的培养物、胎盘、子宫、以及其它组织如脑、肝和肠。将组织提取物冻干或真空离心蒸发浓缩之后,加入分析缓冲剂,将不同的等分样置于RIA试管中。抑制剂生产细胞的提取物形成的置换曲线类似于标准曲线,但不产生抑制剂的组织的提取物不能从抑制剂置换放射性标记的抑制剂。另外,将得自患Lewis肺癌的动物尿、血浆和脑脊髓液的提取物以递增的量加入分析试管。类似的置换曲线显示该抑制剂分析法在测定组织和体液中的抑制剂方面的效用。
含抑制剂的组织提取物还通过反相HPLC分析等分样而被鉴定。收集洗脱级分,在真空离心蒸发浓缩器中干燥,在RIA缓冲剂中重建后再用该抑制剂RIA分析。抑制剂免疫反应性的最大量位于对应于抑制剂的洗脱位置的级分中。
该分析试剂盒提供说明书、抗血清、抑制剂或抑制剂肽,以及有可能放射性标记的抑制剂和/或用于沉淀结合的抑制剂-抑制剂抗体复合体试剂。该试剂盒适用于测定生物流体中以及患有和未患肿瘤的动物和人组织提取物中的抑制剂。
另一种试剂盒被用于定位组织和细胞中的抑制剂。这种抑制剂免疫组织化学试剂盒提供说明书,抑制剂抗血清,以及与荧光分子如异硫氰酸荧光素或与用于显示初级抗血清的某些其它试剂连接的可能的封阻血清和次级抗血清。本领域技术人员熟知免疫组织化学技术。这种抑制剂免疫组织化学试剂盒允许应用光学显微术和电子显微术来定位组织切片和培养细胞中的抑制剂。它可用于研究和临床应用。例如,对肿瘤进行活组织检查或者采集肿瘤并用切片机进行组织切片以检查抑制剂生产部位。这种信息适用于检测和治疗癌方面的诊断应用和可能的治疗应用。显示抑制剂生物合成位点的另一种方法包括放射性标记用于现场杂交的核酸以探测抑制剂信使RNA。同样,该抑制剂受体可用免疫组织化学技术定位、显示和定量分析。
具体实施方式
通过下述实施例而进一步阐释本发明,但不能认为这些实施例以任何方式对本发明的范围加以限制。反之,应清楚地懂得在阅读本说明书后,本领域技术人员能得出本发明的各种其它实施方案、改进形式、以及等效形式。
实施例1
用实验说明人纤溶酶原的Kringle 5的内皮细胞增殖抑制剂活性来源:用适当的酶消化人纤溶酶原而得Kringle 5片段。分离该肽片段后运用本领域技术人员熟知的蛋白质和肽纯化的标准方法纯化。分离后的Kringle 5的纯度如图2所示,该图示出在凝胶电泳上操作该肽制剂的结果。
分析:通过牛毛细内皮细胞中DNA合成([甲基-3H]胸苷掺合)的抑制而分析内皮细胞抑制活性。毛细内皮细胞用牛肾上腺制备后在明胶涂覆的48孔微量滴定板上生长。
将不同量已分离的Kringle 5加入培养细胞,再测定细胞数的变化。将细胞数的变化百分数作为加入培养物中Kringle 5的量的函数描点得图1的曲线图。
图3示出得自人纤溶酶原的Kringle 1、2、3、4和5区的相似80个氨基酸的序列之间的比较。
Claims (32)
1.体外抑制内皮细胞增殖的方法,它包括对内皮细胞施用有效量的具有纤溶酶原分子Kringle 5肽的氨基酸序列的蛋白质。
2.权利要求1的方法,其中该蛋白质包括约80个氨基酸。
3.权利要求1的方法,其中该蛋白质的分子量约为14kD。
4.权利要求1的方法,其中的纤溶酶原是人纤溶酶原。
