Description
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung neuer ML-236B-Carbonsäurederivate, welche als antihyperlipämische Mittel verwendet werden können.
Die erfindungsgemäss erhältlichen Verbindungen entsprechen der Formel:

In der japanischen Offenlegungsschrift Nr. 155 690/1975 (die am 16. d.h. «ML-236B selbst, durch Züchtung von Pénicillium ci-trinum SANK 18 767 hergestellt werden kann und eine pharmakologische Aktivität als hypocholesterämisches und hypolipämisches Arzneimittel hat.
55 Weitere Untersuchungen haben ergeben, dass die ML-236B-Carbonsäurederivate der Formel I leicht von ML-236B abgeleitet werden können und eine unerwartet höhere biologische Aktivität gegen Hyperlipämie zeigen als ML-236B.
6o Gegenstand der Erfindung sind also Verfahren zur Herstellung der neuen ML-236B-Carbonsäurederivate der Formel I, die eine wertvolle Aktivität als antihyperlipämische Mittel haben.
Die Metallsalze der ML-236B-Carbonsäure sind neue
65 Substanzen und werden erfindungsgemäss durch Verseifen von ML-236B mit einem schwachen Alkali und gegebenenfalls Ersatz des so eingeführten Metalles durch ein anderes Metall hergestellt.

Beispiele von ML-236B-Carbonsäuremetallsalzen sind Alkalimetallsalze, z.B. das Natrium- oder Kaliumsalz, Erdalkalimetallsalze, z. B. das Calcium- oder Magnesiumsalz, des Aluminiumsalz, das Eisensalz, das Zinksalz, das Kupfersalz, das Nickelsalz, das Kobaltsalz u.dgl. Unter anderem werden die Alkalimetallsalze, die Erdalkalimetallsalze und das Aluminiumsalz bevorzugt, und das Natriumsalz,das Calciumsalz und das Aluminiumsalz werden besonders bevorzugt.
Die ML-236B-Carbonsäurederivate der Formel I vermögen die Biosynthese von Cholesterin in der Leber in ähnlicher Weise zu hemmen wie ML-236B selbst. Jedoch haben die genannten Derivate eine viel stärkere Aktivität als ML-236B. Eine so starke Hemmwirkung der ML-236B-Carbon-säurederivate auf die Cholesterinbiosynthese konnte auf Grund der Wirkung von ML-236B selbst nicht erwartet werden. Demgemäss sind die ML-236B-Carbonsäurederivate sehr wirksame Antihyperlipämika.
Im folgenden werden die Hemmwirkung auf die Cholesterinbiosynthese, die senkende Wirkung auf den Serum-cholesteringehalt, die wirksame Dosis und die Toxizität der ML-236B-Carbonsäurederivate der Formel I erläutert.
1. Hemmwirkung auf die Cholesterinbiosynthese
Es wurde gefunden, dass die ML-236B-Carbonsäure-derivate 3-Hydroxy-3methylglutaryl-CoA-reductase, die bekanntlich ein geschwindigkeitsbestimmendes Enzym bei der Biosynthese von Cholesterin ist, spezifisch hemmen. Die Hemmwirkung auf die Cholesterinbiosynthese [bestimmt nach dem Verfahren, das in J. Biol. Chem. 234,2835 (1959) offenbart ist] und die Hemmwirkung auf 3-Hydroxy-3-methylglutaryl-CoA-reductase [bestimmt nach dem Verfahren, das in Anal. Biochem. 31, 383 (1969) offenbart ist] dieser Verbindungen sind in Tabelle I zusammengefasst.

Es ist ersichtlich, dass die ML-236B-Carbonsäure-derivate der Formel I eine mehrere Male stärkere, senkende 25 Wirkung auf den Blutcholesterinspiegel haben als ML-236B.
Die senkende Wirkung auf den Blutcholesterinspiegel bei Ratten wurde ebenfalls unersucht. Der Text bestand darin, dass eine in Wasser suspendierte Testprobe oral verabreicht wurde, nach 18 Stunden eine Blutprobe entnommen wurde 30 und der Cholesterinspiegel im Serum nach einem herkömmlichen Verfahren bestimmt wurde. Die Ergenisse sind in Tabelle III zusammengefasst.

2. Senkende Wirkung auf den Serumcholesteringehalt Die senkende Wirkung von ML-236B-Carbonsäure-derivaten der Formel I auf den Blutcholesteringehalt bei kleinen Spürhunden (Körpergewicht ca. 10 kg) wurde bestimmt. Der Test bestand darin, dass eine eingekapselte Testprobe zweimal täglich, nämlich am Morgen (9 Uhr) und am Nachmittag (16 Uhr) an 5 aufeinanderfolgenden Tagen verabreicht wurde, nach 5 Tagen am Morgen eine Blutprobe entnommen wurde und der gesamte Cholesterinspiegel in dem Serum nach einem herkömmlichen Verfahren bestimmt wurde. Die Ergebnisse sind in Tabelle II wiedergegeben.
Es ist ersichtlich, dass die ML-236B-Carbonsäure-60 derivate der Formel I eine sehr befriedigende senkende Wirkung auf den Blutcholesterinspiegel haben, wenn man sie mit ML-236B vergleicht.
3. Akute Toxizität 65 Die akute Toxizität von ML-236B-Carbonsäure-derivaten der Formel I wurde bei oraler und intraperitonealer Verabreichung (beide Male als wässrige Suspension) bestimmt. Die Ergebnisse sind in Tabelle IV wiedergegeben.

Wie aus den oben wiedergegebenen Versuchen ersichtlich ist, können die ML-236B-Carbonsäurederivate oral, durch intravenöse Injektion oder in ähnlicher Weise verabreicht werden, und ihre Wirkungen sind deutlich besser als diejenigen von ML-236B. Die Dosis für die Behandlung von Erwachsenen hängt von der Art und der Häufigkeit der Verabreichung ab, liegt aber gewöhnlich im Bereich von 100 bis 3000 mg pro Tag, vorzugsweise 1500 mg pro Tag.
Die ML-236B-Carbonsäurederivate der Formel I können analog wie andere bekannte antihyperlipämische Mittel, wie Clofibrate, Sinfibrate u.dgl., zu verschiedenen Präparaten formuliert werden, die nach beliebigen herkömmlichen Methoden verabreicht werden können.
Die Präparate werden zweckmässig in der Form hergestellt, die für die Resorption im Magen-Darm-Trakt bevorzugt wird. Tabletten und Kapseln für die orale Verabreichung liegen in Form einer Dosierungseinheit vor und können herkömmliche Träger, wie Bindemittel, z. B. Sirup, Gummiarabikum, Gelatine, Sorbit, Tragantgummi oder Polyvinylpyrrolidon, Ecipientien, wie Lactose, Saccharose, Maisstärke, Calciumphosphat, Sorbit oder Glycin, Gleitmittel, wie Magnesiumstearat, Talkum, Polyäthylenglycol oder Siliciumdioxyd, Sprengmittel, wie Kartoffelstärke, oder Netzmittel, wie Natriumlaurylsulfat, enthalten. Tabletten können nach beliebigen, dem Fachmann bekannten Verfahren beschichtet werden. Flüssige Präparate für die orale Verabreichung können wässrige oder ölige Suspensionen, Lösungen, Sirupe, Elixiere od.dgl. sein, oder sie können in beliebiger getrockneter Form vorliegen, die bei der Anwendung wieder in Wasser oder anderen geeigneten Trägern aufgelöst werden kann. Derartige flüssige Präparate können herkömmliche Additive, wie Suspendiermittel, wie Sorbitsirup, Methylcellulose, Glucose/Zuckersirup, Gelatine, Hy-droxyäthylcellulose, Carboxymethylcellulose, Aluminium-stearatgel oder ein hydriertes essbares Fett, Emulgatoren, wie Lecithin, Sorbitanmonooleat oder Gummiarabikum, nicht wässrige Träger, wie Mandelöl, fraktioniertes Kokos-nussöl, ölige Ester, Propylenglycol oder Äthanol, oder Konservierungsmittel, wie p-Hydroxybenzoesäuremethylester, p-Hydroxybenzoesäurepropylester oder Sorbinsäure, enthalten.
Injizierbare Präparate werden vorzugsweise in einer Ampullen-Dosierungseinheit oder in einem Gefäss für mehrere Dosierungen mit zugesetzten Konservierungsmitteln zur Verfügung gestellt. Die Präparate können in Form von Suspensionen, Lösungen oder Emulsionen in öligen oder wäss-rigen Trägern vorliegen und auch Formulierungsmittel, wie Suspendiermittel, Stabilisatoren und/oder Dispergiermittel, enthalten. Die Wirkstoffe können auch in Form eines Pulvers vorliegen, das bei der Anwendung wieder in einem geeigneten Träger, z. B. pyrogenfreiem, steriliertem Wasser, aufgelöst wird.
Diese Präparate können je nach der Art der Verabreichung nicht weniger als 0,1%, vorzugsweise 10 bis 60%, des s Wirkstoffes enthalten. Eine Dosierungseinheit des Präparates enthält vorzugsweise 50 bis 500 mg Wirkstoff.
Wenn die Aminosäure- oder Metallsalze der ML-236B-Carbonsäure in wässriger Lösung vorliegen, werden sie im sauren pH-Bereich zu ML-236B lactonisiert. Daher werden io wässrige Lösungen der Salze vorzugsweise im neutralen bis schwach alkalischen pH-Bereich gehalten.
Die folgenden Beispiele erläutern die Erfindung.
Beispiel 1
15 Natrium-ML-236B-carboxylat
Eine Suspension von 39 g ML-236B in 500 ml einer 0,2-normalen wässrigen Natriumhydroxydlösung wurde 2 Stunden lang bei 50 °C gerührt, wobei sich eine praktisch klare wässrige Lösung bildete. Eine geringe Menge restlicher un-20 löslicher Materialien wurde abfiltriert und das Filtrat gefriergetrocknet, wobei 40,2 g des Natriumsalzes von ML-236B gebildet wurden.

Calcium-ML-236B-carboxylat Eine Lösung von 4,7 g Calciumhydroxyd in 1 Liter Wasser wurde mit 50 g ML-236B versetzt und das Gemisch 2 Stunden lang bei 50 °C gerührt. Die ganze Lösung, die einen 35 in situ gebildeten Niederschlag enthielt, wurde gefriergetrocknet. Die getrocknete Probe (ca. 55 g) wurde in 500 ml Äthylacetat suspendiert. Die Suspension wurde 1 Stunde lang bei Raumtemperatur gerührt und der Niederschlag durch Filtration isoliert. Der Niederschlag wurde ferner mit 40 1 Liter Äthylacetat gewaschen und im Vakuum getrocknet, wobei 49,5 g des Calciumsalzes von ML-236B erhalten wurden.

Aluminium-ML-236B-carboxylat Eine Lösung von 10 g des Natriumsalzes von ML-236B, das in Beispiel 21 hergestellt wurde, in 100 ml Wasser wurde mit Salzsäure auf pH = 2 eingestellt und mit 100 ml Benzol 55 versetzt. Nach dem Schütteln wurde die Benzolschicht isoliert und zur Trockene eingeengt. 8 g des Rückstandes und 1,3 g Aluminiumisopropylat wurden in 200 ml wasserfreiem Benzol gelöst, worauf die resultierende Lösung 2,5 Stunden lang unter Rückfluss gekocht wurde. Dann wurde das Lö-60 sungsmittel abdestilliert, wobei 9,0 g des Aluminiumsalzes von ML-236B zurückblieben.

- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Verbindung der Formel:mit einem schwachen Alkali verseift und das so eingeführte Metall durch ein anderes Metall ersetzt.
