PATENTANSPRUCH Verfahren zur Herstellung einer neuen Verbindung der Formel

R2 Wasserstoff, Alkyl mit 1-4 C-Atomen oder zusammen mit R3 und dem Kohlenstoffatom, an das sie gebunden sind, eine Carbonylgruppe bildet;

Chlorkohlenwasserstoffrest ist, m 0 oder 1, n und p je eine ganze Zahl von 0 bis 6 bedeuten, Y und Z je ein Sauerstoffatom oder zwei Wasserstoffatome bedeuten, mit der Massgabe, dass Y und Z nicht gleichzeitig je ein Sauerstoffatom s bedeuten können. Diese Verbindungen haben entweder den Prostaglandinen entsprechende biologische Eigenschaften oder sind Prostaglandin-Antagonisten.
Bolliger und Muchowski beschreiben in «Tetrahedron Letters» Band 1975, Seite 2931, die Herstellung von io 11 -Desoxy-8-azaprostaglandin Ex. In verschiedenen biologischen Versuchen ist das eine Epimere dieser Verbindung wirksamer als das andere.
Es wurde nun eine Klasse neuer Verbindungen aufgefunden, die wertvolle pharmakologische Aktivitäten besitzen. i5 So hemmen diese Verbindungen z. B. die Magensaftabsonderung, haben eine kardiovaskuläre Wirkung, d.h. eine antihypertensive Wirkung, eine die Blutplättchenaggregation hemmende Wirkung, eine Wirkung auf den Respirationstrakt, d.h. eine Bronchodilator-Wirkung, eine Antifertili-20 tätswirkung und eine Wirkung auf die glatte Muskulatur. Im allgemeinen kann man sagen, dass diese Verbindungen eine den natürlichen Prostaglandinen entsprechende pharmakologische Wirkung haben, dass diese Wirkungen jedoch selektiver sind.
25 Die vorliegende Erfindung betrifft nun die Herstellung von Ausgangsprodukten für die Herstellung solcher neuer zyklischer Amine und Amide, die wertvolle pharmakologische Eigenschaften aufweisen.
Gegenstand der Erfindung ist somit ein Verfahren zur 30 Herstellung neuer Verbindungen der allgemeinen Formel I
Die Erfindung betrifft ein Verfahren zur Herstellung neuer substituierter Aminosäuren oder Estern oder Amiden davon, die am Stickstoffatom und an einem a-Kohlenstoff-atom mit aliphatischen Resten substituiert sind.
Natürliche Prostaglandine und deren Analoga weisen eine grosse Anzahl von pharmakologischen Aktivitäten auf. In der DE-OS 2 323 193 sind Pyrazolidinderivate der Formel beschrieben

In Formel I bedeutet n oft eine ganze Zahl von 3 bis 8, R2 ein Wasserstoffatom oder die Methylgruppe, oder 60 R2 bildet mit R3 und dem Kohlenstoffatom, an das sie gebunden sind, eine Carbonylgruppe.
Für n in Formel I geeignet ist eine ganze Zahl von 1 bis 5, vorzugsweise bedeutet n jedoch 3,4 und 5, insbesondere 3. Vorzugsweise hat die a-Seitenkette der Verbindungen der 65 Formel I die Formel (CH2)nlR1, in der nl 6,7 oder 8, vorzugsweise 6, bedeutet.
Als Schutzgruppen für die Hydroxylgruppen geeignet sind leicht verseifbare Reste, wie Acylreste mit 1 bis 4 Koh-

lenstoffatomen, z. B. der Acetoxyrest, und leicht abspaltbare inerte Reste, wie der Benzylrest. Vorzugsweise sind Hydroxylgruppen der Verbindungen der Formel I jedoch nicht geschützt.
Für R! geeignet ist das Wasserstoffatom, die Hydroxy-methyl- oder Carbonsäuregruppe sowie deren Methyl-, Äthyl-, Propyl-, Butyl-, Pentyl-, Hexy 1-, Phenyl-, Benzyl-oder Toluylesterrest. Im allgemeinen bedeutet Rj jedoch das Wasserstoffatom, die Hydroxymethyl- oder Carbonsäuregruppe oder deren C^-Alkylester.
Für R2 in Formel I besonders geeignet ist das Wasserstoffatom, der Methyl- oder Äthylrest, wobei der Methyl-und Äthylrest bevorzugt sind. Im allgemeinen bedeutet R2 den Methylrest.
Für R3 bevorzugt ist das Wasserstoffatom oder die Hydroxylgruppe, insbesondere die Hydroxylgruppe.
Spezielle Beispiele für R4 sind Alkylreste mit 4 bis 9 Kohlenstoffatomen, die geradkettig oder mit ein oder zwei Methylgruppen am gleichen oder an verschiedenen Kohlenstoffatomen substituiert sein können, wie n-Butyl-, n-Pentyl-, n-Hexyl- oder n-Heptylreste, oder Reste der Formeln CH2R5, CH(CH3)R5 oder C(CH3)22R5, in denen R5 ein geradkettiger Alkylrest mit so vielen Kohlenstoffatomen ist, dass R4 4 bis 9 Kohlenstoffatome hat. Für R5 geeignet ist der n-Butyl- oder n-Pentylrest. Im allgemeinen sind für R4 n-Pentyl-, n-Hexy 1- und n-Heptylreste, insbesondere der n-Hexylrest bevorzugt.
Geeignete Reste C02R7 in Formel (I) sind die für R! genannten Estergruppen.
Von besonderem Interesse sind erfindungsgemäss hergestellte Verbindungen der Formel II
in der n1 eine ganze Zahl von 1 bis 5 bedeutet, R12 ein Wasserstoffatom oder ein Alkylrest mit 1 bis 4 Kohlenstoffatomen, R* 3 ein Wasserstoffatom oder eine gegebenenfalls geschützte Hydroxylgruppe und R\ ein Wasserstoffatom oder ein Alkylrest mit 4 bis 9 Kohlenstoffatomen ist und R7, Y und W wie in Formel I definiert sind.
In Formel II bedeutet vorzugsweise Y den Äthylenrest und n1 3 oder 5, insbesondere 3.
Für R12 in Formel II geeignet ist das Wasserstoffatom, der Methyl- oder Äthylrest. Bevorzugt ist der Methyl- oder Äthylrest, insbesondere der Methylrest.
Für Rx3 bevorzugt ist die Hydroxylgruppe.
Für R\ geeignet sind die oben beschriebenen geradkettigen oder verzweigten Alkylreste. Für Rx4 bevorzugt sind n-Pentyl-, n-Hexyl- und n-Heptylreste, insbesondere der n-He-xylrest.
Für W in Formel II bevorzugt ist das Wasserstoffatom oder ein Alkylrest mit 1 bis 4 Kohlenstoffatomen, wie der Methyl- oder Äthylrest.
Für R7 bevorzugt ist Ci-C4-Alkyl. Normalerweise sind R7 und W gleich und oft Ci-C4-Alkyl.
Unter den Verbindungen der Formel II sind Verbindungen der Formel III bevorzugt,

in der n11 6 bedeutet, Rn2 ein Wasserstoffatom, der Me-thyl- oder Äthylrest, R114. der n-Pentyl-, n-Hexyl- oder n-Heptylrest, R17 Alkyl mit 1 bis 4 C-Atomen und W1 ein Wasserstoffatom oder ein Alkylrest mit 1 bis 4 Kohlenstoffatomen ist.
In Formel III bedeutet R112 bevorzugt den Methyl- oder Äthylrest, insbesondere den Methylrest, und Rl x4 den n-He-xylrest. Für W1 bevorzugt ist der Methyl- oder Äthylrest, und für R17 wird C14-Alkyl bevorzugt.
Besonders geeignete Verbindungen der Formel I sind: Diäthyl-2[(N-3-hydroxy-3-methyl-n-octyl)]-aminoazelat, Diäthyl-2[(N-3-hydroxy-3-methyl-n-dodecyl)]-aminoazelat, Diäthyl-2[(-3-hydroxy-3-äthyl-n-nonyl)]-aminoazelat und Diäthyl-2[(N-3-hydroxy-3-methyl-n-nonyl)]-aminoazelat.
Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen der Formel I erfolgt durch Umsetzen eines Amins der Formel

mit einer Verbindung der Formel

in der Q ein durch eine elektronenreiche Gruppe leicht verdrängbarer Rest ist.
Für Q in Formel V geeignet ist das Jod-, Brom- oder Chloratom, die Methylsulfonyl- oder Toluolsulfonylgruppe.
Diese Verdrängungsreaktion kann unter herkömmlichen Bedingungen erfolgen, z.B. in einem alkoholischen Lösungsmittel in Gegenwart von Natriumcarbonat oder Pyridin.
Da die nach dem erfindungegemässen Verfahren hergestellten Verbindungen der Formel I asymmetrische Zentren haben, können sie in verschiedenen stereoisomeren Formen vorliegen, die voneinander durch herkömmliche Verfahren getrennt werden können. Die Erfindung betrifft daher auch die Herstellung dieser stereoisomeren Formen und deren Gemische.
Die Amine der Formel IV können beispielsweise nach folgenden Methoden A bis E erhalten werden.
Methode A (PhCH2)2NCH2CH2COR
N,N-Dibenzyl-2-aminoäthylmethylketon (R = CH3)
70,5 frisch destilliertes Methylvinylketon werden tropfenweise unter Rühren zu einer Lösung von 197 g Dibenzyl-amin in 50 ml trockenem Äthanol gegeben. Das Gemisch wird 30 min gerührt, dann wird das Lösungsmittel abgedampft. Der feste Rückstand wird mit einer geringen Menge Äthanol gewaschen. Man erhält 211,6 g (79%) der gewünschten Verbindung als schwach gelben Feststoff, Fp. 58 bis 59 °C.
N,N-Dibenzyl-2-aminoäthyläthylketon (R = C2H5)
wird entsprechend aus Äthylvinylketon und Dibenzylamin als schwach gelbes Öl erhalten.

3-Methyl-1 -(N,N-dibenzylamino)-nonan-3-ol (Rt = CH3; R2 = C6H13) 6,55 g Magnesium werden unter Stickstoffschutz mit 48,9 g Hexylbromid in 100 ml trockenem Tetrahydrofuran umgesetzt und mit einer Lösung von 50 g N,N-Dibenzyl-2-aminoäthyl-methylketon in 200 ml trockenem Tetrahydrofuran tropfenweise versetzt. Das Gemisch wird gerührt und über Nacht am Rückfluss erhitzt. Das Gemisch wird dann mit einer gesättigten Ammoniumchloridlösung versetzt und dreimal mit Äther extrahiert. Die organischen Phasen werden vereinigt, über Magnesiumsulfat getrocknet und eingedampft.
Die in Tabelle I zusammengestellten Produkte werden entsprechend hergestellt.

3-Benzyloxy-n-nonanitril (R = C6H13)
Eine Lösung von 6,5 g Natrium in 722 g Benzylalkohol wird bei Raumtemperatur tropfenweise unter Rühren mit 311g l-Cyano-non-2-en versetzt. Das Gemisch wird gerührt, 4 Stunden in einem Wasserbad erhitzt und dann bei Raumtemperatur über Nacht stehengelassen. Das Gemisch wird vorsichtig mit Eisessig neutralisiert, der überschüssige Benzylalkohol wird unter vermindertem Druck abgedampft. Der Rückstand wird in Äther aufgenommen und filtriert. Das Filtrat wird unter vermindertem Druck eingedampft. Das Produkt wird destilliert; man erhält 302 g (54%) des gewünschten Produkts als farbloses scharf riechendes Öl, Kp.0,6mm 166 bis 168 °C.
3-Benzyloxy-n-octanitril (R = C5Hu) wird entsprechend als farbloses Öl, Kp.0i25min 128 bis 130 °C erhalten.

l-Amino-3-methylnonan-3-ol (Rj = CH3; R2 = C6H13) Eine Aufschlämmung von 8 g lOprozentigem Palladium auf Kohle in Äthanol wird mit einer Lösung von 71g 3-Methyl-l-(N,N-dibenzyl-amino)-nonan-3-ol in 200 ml Äthanol versetzt. Das Gemisch wird bei 70 °C und einem Druck von 14 kg/cm2 3 Tage lang hydriert, dann durch Diatomeenerde filtriert und eingedampft. Das ölige Produkt wird fraktioniert destilliert. Man erhält 18,9 g (55%) des gewünschten Produkts als farblose Flüssigkeit, Kp.02mm 104 bis 106°C.
Die in Tabelle II zusammengefassten Produkte werden entsprechend hergestellt.

3-Benzyloxy-n-octylamin(R = C5Hh)
Eine gerührte Suspensionen 12,2 g Lithiumaluminiumhydrid in 450 ml trockenem Äther wird tropfenweise mit 30 74 g 3-Benzyloxy-n-octanitril versetzt. Das Gemisch wird 40 min am Rückfluss erhitzt, dann in einem Eisbad abgekühlt und tropfenweise mit Wasser versetzt, um das überschüssige Hydrid zu zerstören. Die Lösung wird filtriert, der feste Rückstand wird einige Male mit Äther gewaschen. Die ver-35 einigten Ätherlösungen werden über Magnesiumsulfat getrocknet und unter vermindertem Druck eingedampft. Das Produkt wird destilliert, man erhält 70,4 g (94%) eines farblosen Öls, Kp.04mm 129 °C.
3-Benzyloxy-n-nonylamin (R = C6H13) wird entspre-40 chend als farbloses Öl, Kp.01mm 134 bis 140 °C erhalten. Das folgende Beispiel erläutert das erfindungsgemässe Verfahren.

2-(N-3'-Benzyloxy-n-nonyl)-aminoazelainsäure-diäthylester (n = 6; Rj = C2H5; R2 = H; R3 = CH2Ph; R4 = C6H13) Eine am Rückfluss siedende Lösung von 80 g 60 3-Benzyloxy-n-nonyl-amin in 500 ml trockenem Äthanol, die 41 g wasserfreies Natriumcarbonat enthält, wird mit einer Lösung von 114 g 2-Bromazelainsäure-säurediäthylester in 200 ml trockenem Äthanol versetzt. Das Gemisch wird 12 Stunden am Rückfluss erhitzt und dann filtriert. Das Filtrat 65 wird unter vermindertem Druck eingedampft. Der Rückstand wird in 500 ml Äther aufgenommen, die ätherische Lösung wird mit gesättigter Natriumchloridlösung gewaschen, bis sie neutral ist, über Magnesiumsulfat getrocknet und un-
