Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung des neuen Camitinsäurechlorid-chlorides
(CH3)3N-CH2-CH-CH2-C0C1, ClW. I
OH DJL-Camitinchlorid ist ein vielseitiges Zwischenprodukt zur Herstellung von verschiedenen Camitinderivaten, z. B. Estern und Amiden, deren therapeutische Eigenschaften bekannt sind. (+)
Camitin, (CH3)3N-CH2-CH-CH2-COO^ enthält zusätzlich zu einer Carboxylgruppe eine Hydroxylgruppe,
OH welche, wie bekannt, empfindlich gegen saures Reaktionsmilieu ist
Es ist eine bekannte Tatsache, wie beispielsweise in Bull. Soc. Chim. Fr. (1960), 1196, beschrieben, daß ß-Hydroxysäuren und deren Ester in einem sauren Milieu leicht ein Wassermolekül freisetzen und so ungesättigte Verbindungen bilden. In Biochim. Biophys. Acta 122,98-106 (1967), und 152.559 (1968), ist beschrieben, daß die Camitindehydratisierung in saurem Milieu unter Erhitzen stattfindet. In J. Biol. Chem. 237/12, 3268 (1962) ist beschrieben, daß die Bildung von Crotonoylbetain als Nebenprodukt durch Erhitzen von Camitin in einem sauren Milieu erfolgt.
Da eine Carbonsäure unter sauren Bedingungen in ein Säurechlorid übergeführt wird, konnte nicht erwartet werden, daß die Carnitinchlorierung erfolgt, ohne vorher die Hydroxylgruppe zu schützen, um eine Zersetzung des Ausgangsmaterials und die Bildung unerwünschter Nebenprodukte zu verhindern.
Das vorher Gesagte ist in völliger Übereinstimmung mit dem Stand der Technik. Die Herstellung von Säurechloriden von hydroxysubstituierten Säuren, nachdem die Hydroxylgruppe vorher geschützt worden ist, ist in J. Org. Chem. 43/20, 3972 (1978) beschrieben. Die Chlorierung von ß-Hydroxybuttersäure (welche wie Camitin eine ß-Hydroxysäure ist) nach vorherigem Schutz der Hydroxylgruppe durch Acyliemng ist in J. Am. Chem. Soc. 25,4106 (1973) beschrieben. Überraschenderweise wurde nun gefunden, daß es möglich ist,D,L-Carnitin-hydrochlorid in das Säurechlorid von DJ.-Camitinhydrochlorid mit ausgezeichneten Ausbeuten ohne die Bildung von unannehmbaren Mengen von Nebenprodukten, vom industriellen Standpunkt aus betrachtet, (insbesondere Crotonoylbetain) übeizuführen. Dies erfolgt durch Chlorierung von D^-Camitin-hydrochlorid ohne vorherigen Schutz der Hydroxylgruppe (z. B. durch Überführen der Hydroxylgruppe in eine Acetylgruppe entsprechend dem Stand der Technik), vorausgesetzt, daß einige kritische Verfahrensbedingungen erfüllt werden.
Es wurde gefunden, daß das molare Verhältnis Camitin-hydrochlorid und Chlorierungsmittel, die Reaktionstemperatur und die Reaktionszeit kritische Parameter bei der Umsetzung von DJL-Camitin-hydrochlorid in das entsprechende Säurechlorid sind und daß die Werte der vorgenannten Parameter in gut definierte Bereiche fallen müssen.
Gemäß der Erfindung wird Camitin-hydrochlorid mit einem Chlorierungsmittel ausgewählt unter Thionylchlorid, Dichlormethylether und Oxalylchlorid bei Raumtemperatur während einer Reaktionszeit zwischen etwa 1,5 h und 12 h chloriert, wobei Camitin -hydrochlorid und Chlorierungsmittel in einem Molverhältnis zwischen 1:1 und 1:3 eingesetzt werden.
Phosphoipentachlorid könnte auch als Odorierungsmittel verwendet werden. Dieses Odorierungsmittel wird jedoch erfindungsgemäß nicht eingesetzt wegen der bemerkenswert höheren Reaktionszeiten (1-3 Tage) im Vergleich mit den vorher genannten.
Um hohe Ausbeuten bei der Umsetzung von Camitin-hydrochlorid in das Säurechlorid zu erhalten, ist es wesentlich, daß die vorgenannte Temperatur und die vorgenannte Reaktionszeiten genau erfüllt werden, da schon geringe Abänderungen die Bildung von Nebenprodukten mit sich bringen. Wenn z. B. Oxalylchlorid als Chlorierungsmittel verwendet wird, ergibt sich eine Reaktionszeit von 15 h die fast vollständige Zersetzung von Camitin in Crotonoylbetain.
Die folgenden, nicht begrenzenden Beispiele 1 und 2 illustrieren das erfindungsgemäße Verfahren.
Beispiel! (Chlorierungsmittel: Thionylchlorid) 2,25 cnr* (0,03 Mol) SOCL2 wurden zu 1,98 g (0,01 Mol) D,L-Camitinhydrochlorid gegeben. Nach 1 h war die Solubilisierung beendet und nach 1,5 h wurde weiter verarbeitet Der Überschuß an SOCI2 wurde abdestilliert und der Rückstand bestehend aus Camitinsäurechlorid-chlorid wurde mit wasserfreiem Ethylether gewaschen. Zum Zwecke der Identifikation wurde das Säurechlorid in den Methylester der Formel -2-

OH ungewandelt: die Reaktionsmischung wurde auf 0 °C gekühlt und 10 cm^ wasserfreier Methanol wurden tropfenweise zugesetzt; die entstandene Mischung wurde dann unter Vakuum bei 35 - 40 °C eingeengt, wobei ein gelatineartiges Rohprodukt erhalten wurde, welches getrocknet unter Vakuum erstarrt bleibt, jedoch sehr hygroskopisch istFür dieses Rohprodukt, den Methylester, wurden folgende Analysenwerte ermittelt;

Beispiel 2 (Chlorierungsmittel: 1,1 Dichlordimethylether) Das Verfahren des vorhergehenden Beispiels wurde wiederholt mit der Ausnahme, daß an Stelle von Thionylchlorid Dichlormethylether (1,78 cm^’ 0,02 Mol) verwendet wurde. Die Reaktionsmischung wurde über Nacht bei Raumtemperatur stehen gelassen. Der Überschuß an Dichlormethylether wurde abdestilliert und der Rückstand wurde mit wasserfreiem Ethylether gewaschen. Der Rückstand bestand aus Camitinsäurechlorid-chlorid. ch3 NMR CD3NC Delta 3,33 (9H, s, CH3~> CHo-^

Nach Reaktion des Säurechlorids mit D2O nahmen die Werte der chemischen Verschiebung die gleichen Werte ein wie jene des NMR-Spektrums von Camitin. Das rohe Säurechlorid wurde anschließend in das Camitinmethylester-chlorid umgewandelt, wie vorher beschrieben. Der Methylester von Camitin zeigte die chemischphysikalischen Kenndaten entsprechend dem vorher isolierten Produkt Die folgenden Vergleichsbeispiele A und B sollen illustrieren, daß die vorgenannten Verfahrensbedingungen absolut erfüllt werden müssen, um das Säurechlorid von Camitin zu erhalten. -3-
Claims (1)
- Nr. 390057 Verpleichsheispiel A (die richtige Reaktionstemperatur wird nicht eingehalten) Eine Mischung von Camitin und Thionylchloiid (Molverhältnis 1:1) wurde unter Rühren bei 50 °C gehalten: Proben der Reaktionsmischung wurden nach 0,5 h, 1 h und 1,5 h nach Beginn der Reaktion gezogen. Die Proben wurden mittels ILC (Chloroform 55/ CHgOH 35/ H2O 5/ NH4OH 5) nach Verdünnen mit Methanol, um das Säurechlorid, falls vorhanden, in den Methylester überzuführen, geprüft. Nach 0,5 h wurde das Verhandensein von Camitin und Camitinmethylester-chlorid (Rf 0,4 bzw. 0,8) festgestellt. Nach 1 h begann sich Crotonoylbetain, Camitin und Camitinmethylester-chlorid (Rf 0,2 bzw. 0,4 bzw. 0,8) zu bilden. Nach 1,5 h wurde das Vorhandensein von Crotonoylbetain unter anderen Zersetzungsprodukten, welche nicht identifiziert werden konnten, festgestellt Vergleichsbeispiel B (die richtige Reaktionszeit wurde nicht eingehalten) Thionylchlorid (2,3 cm^; 0,03 Mol) wurde zu Camitinhydrochlorid (1,98 g; 0,01 Mol) gegeben und die entstandene Reaktionsmischung wurde unter Rühren bei Raumtemperatur 24 h gehalten. Der Überschuß an Thionylchlorid wurde abdestilliert und der rohe Rückstand wurde mit wasserfreiem Ethylether gewaschen, wobei ein festes Produkt mit dem Fp. 217 - 218 °C erhalten wurde. Im Gegenteil erfolgte eine Dehydratisierung unter Bildung von Crotonoylbetain. ch3 ch3 n-ch2-ch=ch-cooW. ch3 PATENTANSPRUCH Verfahren zur Herstellung des neuen Camitinsäurechlorid-chlorides der Formel (+) (CH3)3N-CH2-CH-CH2-C0C1 ,1 I OH aw dadurch gekennzeichnet, daß Camitinhydrochlorid mit einem Chlorierungsmittel ausgewählt unter Thionylchlorid, Dichlormethylether und Oxalylchlorid bei Raumtemperatur während einer Reaktionszeit zwischen etwa 1,5 h und 12 h chloriert wird, wobei Camitinhydrochlorid und Chlorierungsmittel in einem Molverhältnis zwischen 1:1 und 1:3 eingesetzt werden. -4-
