Werkwijze ter 'bereiding van carnitinechloride.
Be onderhavige uitvinding heeft betrekking op een werk-wijze ter bereiding van D,L-carnitinechloride,
(CH^^-GEg-CH-CHg-OOCl. B,L-carnitinechloride is een veelSijdig OH
5 tussenprodukt voor de bereiding van verschillende carnitinederi-vaten, bijvoorbeeld esters en amiden, waarvan therapeutische eigenschappen bekend zijn.

10 carboxylgroep een hydroxylgroep, die zoals bekend gevoelig is voor zure reactiemilieus.
Het is feitelijk bekend, bijvoorbeeld zoals beschreven in Bull. Soc. Chim. Pr. (i960), 1196, dat β-hydroxy-zuren en esters daarvan gemakkelijk een watermolecuul vrijmaken in een zure om-15 geving, derhalve onverzadigde verbindingen vormen. In. Biochim. Biophys. Aota 131, (1967) 98-106 en 1^2 (1968) 559, wordt be schreven dat dehydratering van carnitine onder verwarming in zure omgeving plaats heeft. In J. Biol. Chem. 257/12, (1962) 3268, wordt de vorming van orotonoylbetaine als bijprodukt beschreven 20 als plaats hebbend door verwarming van carnitine in een zure omgeving.
Aangezien een carbonzuur in zure omgeving wordt omgezet tot zuur-chloride, kan niet verwacht worden dat chlorering van carnitine plaats heeft zonder voorafgaande bescherming van de 25 hydroxylgroep met het oog op de vermijding van de afbraak van het uitgangsprodukt en de vorming van ongewenste bijprodukten.
Het voorgaande Wordt volledig bevestigd uit hetgeen kan worden afgeleid uit de stand der techniek. Be bereiding van zuur-chloriden van met hydroxy gesubstitueerde zuren nadat de hydroxyl-30 groep vooraf is beschermd, is beschreven in J· Org. Chem. 45/20, (1978) 3972. Meer in het bijzonder wordt de chlorering beschreven van β-hydroxyboterzuur (wat een β-hydroxyzuur is als carnitine) na voorafgaande bescherming van de hydroxylgroep met een acetyl-groep in J.Am. Chem. Soc. ££, (1975) 4106.
35 Verrassenderwijze werd nu gevonden, dat het mogelijk is 8202677 ’ - 2 - κ Ζ *'* B,L-earnitine in het zuurchloride van B,L-carnitine om te zetten* waarbij uitstekende opbrengsten verkregen worden zonder vorming van onaanvaardbare hoeveelheden bijprodukten vanuit induetrigel gezichtspunt (in het bijzonder crotonoylbetaine). Bit wordt uit-5 gevoerd door B,L-carnitine zonder voorafgaande bescherming van de hydroxylgroep (bijvoorbeeld door omzetting van de hydroxylgroep tot een acetylgroep zoals geleerd door de stand der techniek) te chloreren, op voorwaarde dat enkele kritische operationele omstan-digheden in acht worden genomen.
10 In feite werd gevonden, dat de molverhouding oarnitine/chlo- reringsmiddel, de reactietemperatuur en de reactietijd kritische parameters zijn, die de omzetting van B,1-carnitine tot het over-eenkomstige zuurchloride belnvloeden en dat de waarden van de voorgaande parameters binnen goed gedefinieerde trajeoten moeten 15 vallen.
Yolgens de uitvinding omvat de werkwijze voor de omzetting van carnitine tot het zuurchloride van carnitine de volgende trap-pen: chlorering van carnitine met een chloreringsmiddel (bij voorkeur 20 gekozen uit onder andere thionylchloride, dichloormethylether en oxalylchloride) bij kamertemperatuur gedurende reactietijden tussen ongeveer 1,5 en 12 iiren.
Poaforpentachloride kan ook als chloreringsmiddel gebruikt worden. Echter verdiend dit chloreringsmiddel niet de voorkeur 25 vanwege de merkbaar hogere reactietijden (1 tot 3 dagen) in ver-gelijking met het hiervoor aangegeven chloreringsmiddel·
Bij voorkeur ligt de molverhouding carnitine/chlorerings-middel tussen 1:1 en 1:3· Met het oog op het verkrijgen van grote opbrengsten bij de omzetting van carnitine tot het zuurchloride, 50 is het essentieel, dat de voorgaande temperatuur en reactietijden strikt in acht worden genomen aangezien zelfs ^ondergeschikte va-riaties de vorming van bijprodukten teweegbrengen. Wanneer bij-voorbeeld oxalylchloride als chloreringsmiddel wordt gebruikt, brengt een reactietijd van 15 uren de vrijwel volledige afbraak 55 van carnitine tot crotonoylbetaine teweeg.
Be volgende niet beperkende voorbeelden 1 en 2 lichten de werkwijze van de onderhavige uitvinding toe.
Yoorbeeld 1 (chloreringsmiddel: thionylchloride) 2,25 cm^ (0,03 mol) SOClg werden aan 1,98 gram (0,01 mol) 40 B,L-carnitinehydrochloride toegevoegd. Ma 1 uur was hun oplosbaar- 8202677 - 3 - making volledig en na 1,5 uren werd deze als voltooid beschouwd·
De overmaat SOClg werd afgedestilleerd en het residu, bestaande uit carnitinezuurchloride, werd met watervrije diethyl ether ge- wassen. Yoor identificatie doeleinden werd het zuurchloride omge- 5 zet tot de methylesters het reactiemengsel werd op 0°C gekoeld 5 en 10 cur watervrije methanol werden druppelsgewijze toegevoegd; daarna werd het verkregen mengsel onder een verminderde druk bij 35-40°C geconoentreerd, waarbij een gelatineus onzuiver produkt werd verkregen, dat onder vacutim gedroogd, vast hleek, echter zeer 10 hygroseopisch was.

Be methoden van het voorafgaande voorbeeld werden gevolgd, hehalve dat in plaats van thionylchloride 1,78 cm^ (0,02 mol) dichloormethylether werden gebruikt. Het reactiemengsel werd 1 nacht 50 bij kamertemperatuur bewaarcL. De overmaat dichloormethylether werd afgedestilleerd en het residu werd met watervrije diethylether gewassen. Het residu bleek uit carnitinezuurchloride te tiestaan.

Ha uitwisseling met DgO herkregen de chemische verschui-ving'en dezelfde waarden als die van het HMR spectrum van carnitine. Het onzuivere zuurchloride werd vervolgens tot de methylester omgezet, zoals hiervoor beschreven. De methylester van carnitine 5 vertoonde de chemisch-fysische eigenschappen, die overeenkomen met het hiervoor gelsoleerde produkt.
De volgende vodrbeelden A en B heogen toe te lichten dat aan de hiervoor vermelde operationele omstandigheden absoluut moet worden voldaan om het zuurchloride van carnitine te verkrijgen. 10 Yoorbeeld A (aan de geschikte reactietemperatuur wordt niet voldaan).
Een mengsel van carnitine en thionylchloride (molverhou-ding 1s1) werd bij 50°C geroerd. Monsters van het reaetiemengsel werden vanaf 0,5, 1 en 1,5 uur vanaf het begin van de reactie genomen. De monsters werden gecontroleerd met dunne laag 15 dhromatografie (chloroform 55/ CH^OH 35/ H2/ 5/ en HH^OH 5) na ' verdunning met methanol om eventueel zuurchloride tot methylester om te zetten.
Ha 0,5 uur werd een aanwezigheid van carnitine en de methylester van carnitine (R^ 0,4 nesp, 0,8) vastgesteld.
20 Ha 1 uur werden crotonoylbetaine, carnitine en earnitine- methylester (Rf resp. 0,2, 0,4 en 0,8) vastgesteld.
Ha 1,5 uur werd de aanwezigheid van crotonoylbetaine · tezamen met andere afbraakprodukten, die niet geidentificeerd . konden worden, vastgesteld.

2,3 cm^ (0,03 mol) thionylchloride werden aan 1,98 gram (0,01 mol) camitinehydrochloride toegevoegd en het verkregen mengsel werden 24 uren bij omgevingstemperatuur geroerd. De over-maat thionylchloride werd afgedestilleerd en het onzuivere residu 30 werd met watervrije diethylether gewassen, waarbij een vast produkt verkregen werd met een smpt. van 217-218°C.

Zoals DLC en het HMR spectrum laten zien wordt onder deze reactieomstandigheden het zuurchloride van carnitine niet gevormd. Daarentegen heeft een dehydratering plaats onder vorming van ^ crotonoylbetaine 8202677 , - 5 - CH* ΟηΓ^ t-CH2-CH=CH-G00H.

Claims (2)
1. Werkwijze ter bereiding van het zuurchloride van carnitine, met het kenmerk, dat men carnitine chloreert met een chloreringsmiddel gekozen uit thionylchloride, 5 dichloormethylether en oxalylchloride, bij omgevingstemperatuur gednrende reactietijden tussen ongeveer 1,5 nur en 12 uur.
2. Werkwijze volgens conclnsie 1, met het kenmerk, dat men een molverhouding carnitine.: chloreringsmiddel toepast, die ligtinssen 1:1 en 1:3· 10 8202677
