Verfahren zur Herstellung von Carnitinchlorid
Die Erfindung betrifft ein Verfahren zur Herstellung von D/L-Carnitinchlorid
(CH3)3N-CH2-CH-CH2-COCl.

D,L-Carnitinchlorid ist ein wichtiges Zwischenprodukt
für die Herstellung von verschiedenen Carnitinderivaten, z.B. Estern und Amiden, die bekannte therapeutisehe
Eigenschaften aufweisen.

enthält ausser einer Carboxylgruppe eine Hydroxylgruppe, die bekanntlich gegenüber sauren Reaktionsbedingungen
empfindlich ist.
So ist es aus Bull. Soc. Chim. Fr. (1960), 1196, bekannt,
dass ß-Hydroxysäuren und deren Ester in einer
sauren Umgebung leicht ein Wassermolekül abgeben und ungesättigte Verbindungen bilden. In Biochem. Biophys.
Acta 1_3J7/ 98-106 (1967) und ^52, 559 (1968) wird beschrieben,
dass in saurer Umgebung unter Erwärmen eine Dehydratation des Carnitins stattfindet. In J. Biol.
Chem. 237/12, 3268 (1962) wird die Bildung von Crotonoylbetain
als Nebenprodukt beim Erhitzen von Carnitin in saurer Umgebung beschrieben.
Da eine Karbonsäure unter sauren Bedingungen in ein Säurechlorid überführt wird,war nicht zu erwarten,
dass die Chlorierung von Carnitin ablaufen würde, ohne dass zuvor die Hydroxygruppe geschützt wird, weil man
einen Abbau des Ausgangsmaterials und die Bildung von unerwünschten Nebenprodukten verhindern muss.

Dieses Vorurteil wird durch den Stand der Technik gestützt.
Die Herstellung von Säurechloriden von hydroxysubstituierten
Säuren, nachdem man die Hydroxylgruppe zuvor geschützt hat, wird in J. Org. Chem. 43/20, 3972
(1968) beschrieben. Weiterhin wird die Chlorierung von ß-Hydroxybuttersäure (die eine ß-Hydroxysäure ist, wie
Carnitin) nach vorhergehendem Schutz der Hydroxylgruppe mit einer Acetylgruppe in J. Am. Chem. Soc. 95/
4106 (1973) beschrieben.
überraschenderweise wurde nun gefunden, dass man D,L-Carnitin
in das Säurechlorid des D,L-Carnitins überführen kann, und zwar mit ausgezeichneten Ausbeuten
und ohne dass sich unerwünschte Mengen von Nebenprodukten bilden (insbesondere Crotonoyl-betain), wenn
man D,L-Carnitin ohne vorhergehenden Schutz der Hydroxylgruppe (d.h. ohne dass man die Hydroxylgruppe gemäss
dem Stand der Technik in eine Acetylgruppe überführt) chlorieren kann, vorausgesetzt, dass gewisse kritische
Betriebsbedingungen eingehalten werden.
Es wurde gefunden, dass sowohl das Molverhältnis von Carnitin/Chlorierungsmittel als auch die Reaktionstemperatur
und die Reaktionszeit kritische Parameter sind, die bei der Umwandlung von D,L-Carnitin in das entsprechende
Säurechlorid beachtet werden müssen, und dass die Zahlenwerte für die vorgenannten Parameter in
ganz bestimmt umrissene Bereiche fallen müssen.
Nach dem erfindungsgemässen Verfahren wird Carnitin
in das Säurechlorid des Carnitine durch folgende Stufen überführt:
Chlorieren von Carnitin mit einem Chlorierungsmittel (vorzugsweise ausgewählt aus Tionylchlorid, ..Dichlormethylether
und Oxalylchlorid) bei Raumtemperatur während einer Reaktionszeit zwischen etwa 1,5 und 12 Stunden.
Phosphorpentachlorid könnte auch als Chlorierungsmittel
verwendet werden. Dieses Chlorierungsmittel wird jedoch nicht bevorzugt wegen der wesentlich höheren
Reaktionszeiten (1 bis 3 Tage) im Vergleich zu dem zuvor genannten.
Vorzugsweise beträgt das Molverhältnis Carnitin/ Chlorierungsmittel 1:1 bis 1:3. Um eine hohe Umwandlung
des Carnitins und eine hohe Ausbeute an Säure-Chlorid zu erzielen, ist es wesentlich, dass die
vorerwähnte Temperatur und die vorerwähnten Reaktionszeiten ganz strikt eingehalten werden, weil schon geringere
Abweichungen die Bildung von Nebenprodukten begünstigen. Verwendet man beispielsweise Oxalylchlo-'
rid als Chlorierungsmittel, so ergibt sich bei einer
Reaktionszeit von 15 Stunden nahezu ein vollständiger
. Abbau von Carnitin zu Crotonoyl-betain.
Die nachfolgenden Beispiele 1 und 2 beschreiben die Erfindung.
Beispiel 1
(Chlorierungsmittel: Thionylchlorid)
(Chlorierungsmittel: Thionylchlorid)
2,25 cm3 (0,03 mol) SOCl2 wurden zu 1,98 g (0,01 mol)
DjL-Carnitinhydrochlorid gegeben. Nach 1 Stunde war
die Auflösung vollständig und nach 1,5 Stunden war die Umsetzung beendet. Der Überschuss an SOCl2 wurde abdestilliert
und der Rückstand, bestehend aus Carnitinsäurechlorid, wurde mit wasserfreiem Ethylether
gewaschen. Zur Identifizierung wurde das Säurechlorid
in der nachfolgenden Weise in den Methylester überführt: Das Reaktionsgemisch wurde auf 00C gekühlt und
dazu wurden 10 cm3 wasserfreies Methanol tropfenweise gegeben. Die Mischung wurde dann im Vakuum bei 35 bis
400C konzentriert/ unter Erhalt eines gelartigen Rohproduktes,
das nach dem Trocknen im Vakuum verfestigte/ aber sehr hygroskopisch war.

Das Verfahren des vorhergehenden Beispiels 1 wurde wiederholt, wobei jedoch anstelle von Thionylchlorid

Dichlormethylether verwendet wurde (1,78 cm3, 0,02 mol),
Das Reaktionsgemisch wurde über Nacht (jedoch höchstens 12 Stunden) stehen gelassen. Der Überschuss an
Dichlormethylether wurde abdestilliert und der Rückstand wurde mit wasserfreiem Ethylether gewaschen. Der
Rückstand bestand aus Carnitinsaurechlorid.

Nach einem Austausch mit D3O ergab die chemische Ver-Schiebung
die gleichen Werte wie für das NMR-Spektrum von Carnitin. Das rohe Säurechlorid wurde anschliessend
in den Methylester, wie zuvor in Beispiel 1 beschrieben, überführt. Der Methylester des Carnitins zeigte
die chemisch-physikalischen Eigenschaften wie das zuvor isolierte Produkt.
Die nachfolgenden Beispiele A und B zeigen, dass die vorstehenden Betriebsbedingungen genau eingehalten werden
müssen, um das Säurechlorid des Carnitins zu erhalten.

Eine Mischung aus Carnitin und Thionylchlorid (Molverhältnis 1:1) wurde unter Rühren bei 5O0C gehalten.
Proben des Reaktionsgemisches wurden 0,5 Stunden, 1 Stunde und 1,5 Stunden nach Beginn der Umsetzung gezogen.
Die Proben wurden durch TLC (Chloroform 55/ CH3OH 35/H2O 5/ NH4OH 5) nach Verdünnung mit Methanol
zur Umwandlung des Säurechlorids, falls vorhanden, in den Methylester untersucht.
Nach 0,5 Stunden wurde die Gegenwart von Carnitin und dem Methylester von Carnitin (Rf: 0,4 bzw. 0,8)
festgestellt.
Nach 1 Stunde begann sich Crotonoyl-betain, Carnitin
und Carnitinmethylester (Rf: 0,2; 0,4 bzw, 0,8) zu bilden.
Nach 1,5 Stunden wurde die Gegenwart von Crotonoylbetain, zusammen mit einigen nicht-identifizierten
Nebenprodukten, festgestellt.

Thionylchlorid (2,3 cm3, 0,03 mol) wurde zu Carnitinhydrochlorid
(1,98 g, 0,1 mol) gegeben und das Reaktionsgemisch wurde 2 Stunden bei Raumtemperatur während
24 Stunden gerührt. Das überschüssige Thionylchlorid
wurde abdestilliert und der rohe Rückstand wurde mit wasserfreiem Ethylether gewaschen, wobei man einen
Feststoff mit einem Schmelzpunkt von 217 bis 2180C

TLC und NMR-Spektrum zeigen, dass sich unter diesen Umsetzungsbedingungen kein Säurechlorid des Carnitins
bildet. Es findet vielmehr eine Dehydratation unter Bildung von Crotonoyl-betain

- 2. Verfahren gemäss Anspruch.1, dadurch gekennzeichnet , dass das Molverhältnis von Carnitin zu Chlorierungsmittel 1:1 bis 1:3 beträgt.
