Abstract

Un método para obtener una eritropoyetina mono-PEGilada en forma sustancialmente homogénea, que comprende los pasos siguientes: a) PEGilar eritropoyetina empleando un reactivo PEGilante activado de un peso molecular entre 20 kDa y 40 kDa para producir eritropoyetina PEGilada, b) purificar la eritropoyetina PEGilada obtenida en el paso a) mediante dos pasos consecutivos de cromatografía de intercambio catiónico, en donde dichos pasos primero y segundo de la cromatografía de intercambio catiónico emplean el mismo tipo de material de intercambio catiónico, c) recuperar la eritropoyetina mono-PEGilada de la segunda columna de cromatografía de intercambio catiónico en una forma sustancialmente homogénea, d) regenerar la columna de cromatografía de intercambio catiónico mediante los pasos siguientes: i) sacar los polipéptidos residuales fijados de la columna de intercambio catiónico con una solución acuosa tamponada que contiene cloruro sódico, ii) lavar la columna con agua purificada, iii) aplicar a la columna una solución de hidróxido sódico, iv) lavar la columna con agua purificada, v) aplicar a la columna una solución que contiene dihidrogenofosfato sódico y ácido fosfórico, vi) lavar la columna con agua purificada, vii) aplicar a la columna una solución 0,5M de hidróxido sódico durante por lo menos 4 horas, y viii) regenerar la columna de cromatografía de intercambio catiónico lavando la columna con agua purificada.
