N-ヒドロキシメチル-2-クロロアセトアミド(以下、NMCA)2.3gをニトロベンゼン31gと98重量%硫酸31gの混合溶液に添加、NMCAが溶解するまで10℃で攪拌して、NMCA溶液とした。次に、ニトロベンゼン2.0g、98重量%硫酸2.0gにパラホルムアルデヒド(以下、PFA)0.2gを添加し、PFAが溶解するまで20℃で攪拌し、PFA溶液とした。PFA溶液4.2gを5℃に冷却し、NMCA溶液64.3gに混合、5分間攪拌し、編地A1gを添加して2時間含浸した。含浸後の編地Aを0℃のニトロベンゼン200mL中に浸して反応を停止させた後、当該編地に付着しているニトロベンゼンをメタノールで洗浄した。
まず、有機物吸着用担体1を凍結包埋し、ミクロトームにより有機物吸着用担体1に含まれる海島型中実複合繊維の繊維軸方向に垂直な断面を作製した。得られた観察面に導電処理を行い、観察試料とした。次に日立ハイテクノロジーズ製 電界放射型走査電子顕微鏡S-5500を用いて観察試料の断面をランダムに観察し、断面画像を100点撮影した。得られた断面画像中の繊維断面に対して最小内包円を作成し、最小内包円の直径を算出した。この操作を断面画像100点全てに行い、得られた直径の平均値を繊維径とした。結果を表1に示す。
水に含浸された有機物吸着用担体1を約6mg、DSC測定直前に取り出し、過剰な表面付着水を除去してから、アルミニウム製密封型試料容器に封入した。次に、TA Instruments社製DSC Q100を用い、湿潤状態のまま-55℃に急冷し、5℃まで0.3℃/分で昇温させて示差走査熱量を測定し、ピークトップ温度を融点として、DSC曲線を得た。なお、温度、熱量校正は純水を用いた。得られたDSC曲線から、Ishikiriyamaらの方法(JOURNAL OF COLLOID AND INTERFACE SCIENCE、1995年、171巻、p.92-102やp.103-111)に従い、有機物吸着用担体1に含まれる海島型中実複合繊維の細孔体積を算出した。結果を表1に示す。
有機物吸着用担体1に含まれる海島型中実複合繊維の塩基性官能基の含量は、当該海島型中実複合繊維中の塩基性官能基を、酸塩基逆滴定することより決定した。なお、有機物吸着用担体1には補強材が含まれないことから、有機物吸着用担体1の重量を海島型中実複合繊維の重量とみなして測定を実施した。200mLナスフラスコに有機物吸着用担体1を1.5g、乾燥機にて80℃で48時間静置することで乾燥処理をした有機物吸着用担体1を得た。次に、ポリプロピレン製容器に対し、上記の有機物吸着用担体1を1.0g、6M水酸化ナトリウム水溶液50mLを添加して30分攪拌し、濾紙を用いて有機物吸着用担体1をろ別した。次にイオン交換水50mLにろ別した有機物吸着用担体1を添加して30分間攪拌し、濾紙を用いてろ別した。有機物吸着用担体1を添加したイオン交換水のpHが7になるまでイオン交換水に添加、ろ別を繰り返すことで脱塩後の有機物吸着用担体1を得た。脱塩後の有機物吸着用担体1を80℃常圧条件で48時間静置した後、ポリプロピレン製容器に有機物吸着用担体1を1.0gと0.1M塩酸を30mL添加し、10分間攪拌した。攪拌後、溶液のみを5mL抜き取って、ポリプロピレン製容器に移した。次に、得られた溶液に対して、0.1Mの水酸化ナトリウム水溶液を0.1mL滴下した。滴下後10分間攪拌し、溶液のpHを測定した。滴下後10分間の攪拌、pHの測定を同様に100回繰り返した。溶液のpHが8.5を越えた際の水酸化ナトリウム水溶液滴下量を1g当たりの滴定量とした。1g当たりの滴定量と以下の式2を用いて、有機物吸着用担体1に含まれる海島型中実複合繊維1g当たりの塩基性官能基の含量を算出した。結果を表1に示す。

有機物吸着用担体4のアシッドオレンジ7吸着率測定:
粉末のアシッドオレンジ7を0.0875g、酢酸ナトリウム三水和物0.41g、酢酸0.96mL、イオン交換水24mLを混合し、さらにその混合物をイオン交換水で100倍に希釈することで、アシッドオレンジ7濃度が1.0×10-4Mのアシッドオレンジ7緩衝溶液を調製した。次に、酢酸1.14mLとイオン交換水200mLの混合物170mLと、酢酸ナトリウム三水和物1.36gと水100mLの混合物30.6mLをさらに混合し、酢酸緩衝溶液を調製した。上記アシッドオレンジ7緩衝溶液をイオン交換水で2倍、4倍、8倍、16倍に希釈した標準溶液を調製し、紫外可視分光光度計を用いて、測定波長450nm、参照波長600nmとして標準溶液の吸光度を測定した。得られた吸光度と標準溶液中のアシッドオレンジ7濃度から、検量線を作成した。次に、直径6mmの円板状に切り抜いた有機物吸着用担体4を一枚、1.5mLの酢酸緩衝溶液に含浸し、20分間転倒混和した。混和後、有機物吸着用担体4を濾紙でろ別し、付着している酢酸緩衝溶液を150rpmで15分間遠心分離することで除去した。遠心分離後の有機物吸着用担体4を、1.0mLアシッドオレンジ7緩衝溶液を添加したポリプロピレン製容器に添加し、2時間転倒混和した。混和後、ピンセットで有機物吸着用担体4のみを取り出した。有機物吸着用担体4を添加、転倒混和する前後のアシッドオレンジ7緩衝溶液の吸光度を同様に測定し、検量線を用いてアシッドオレンジ7濃度を算出した。有機物吸着用担体4の一枚当たりのアシッドオレンジ7吸着率を、以下の式3を用いて算出した。結果を表2に示す。

有機物吸着用担体4のIL-8吸着性能を確認するため、IL-8を含む液体に有機物吸着用担体4を所定時間含浸後に取り出し、含浸前後の液体中のIL-8量の差分からIL-8吸着率を測定した。以下に測定方法を示す。

有機物吸着用担体4のIL-6吸着性能を確認するため、IL-6を含む液体に有機物吸着用担体4を所定時間含浸後に取り出し、含浸前後の液体中のIL-6量の差分からIL-6吸着率を測定した。以下に測定方法を示す。

有機物吸着用担体4のHMGB-1吸着性能を確認するため、HMGB-1を含む液体に有機物吸着用担体4を所定時間含浸後に取り出し、含浸前後の液体中のHMGB-1量の差分からHMGB-1吸着率を測定した。以下に測定方法を示す。

健常ウサギを用いて、有機物吸着用担体4のカラム通液可能時間を測定した。まず、NZW系雄ウサギ(体重:3~3.5kg)をペントバルビタールナトリウム(25mg/mL、ナカライテスク株式会社)を30mg/kg静脈内投与により導入麻酔した後、頸部及び腹部を毛刈りした。リドカイン(キシロカイン注射液「0.5%」、アストラゼネカ株式会社)を皮下投与したのち、頸部より気管を露出する。気管用カニューレ(16Fr、テルモ株式会社)を挿管、固定した。換気は、人工呼吸器(EVITA300、ドレーゲル・メディカルジャパン株式会社)により実施した。換気条件は、呼気終末陽圧型人工呼吸(PositiveEnd-expiratory pressure、PEEP)をかけた状態で頸動脈から採取した血液をi―STAT(カートリッジCG4+、アボットジャパン株式会社)により血液ガスパラメータを測定し、測定値(体温補正値)がpCO2:35~45mmHgの範囲内になるように換気回数を変更することで調節した。吸気酸素濃度は100%とし、換気条件を設定した後に被験機器の評価を開始し、評価中に換気条件変更は行わなかった。輸液は、0.06mg/kg/hrでベクロニウム加生理食塩液(ベクロニウム静注用4mg:富士製薬工業株式会社、生理食塩液:大塚製薬工場株式会社)を2mL/kg/hrで持続注入した。また、三方活栓を介して、インフュージョンポンプ(55-1111、HARVARD社)に接続し維持麻酔経路とした。維持麻酔は、ペントバルビタール(12.5mg/mL、ナカライテスク株式会社)を2~8mg/kg/hrで持続注入(状態により増減させる)した。ここで、有機物吸着用担体4を充填体積11cm3(充填高さ:4.7cm、充填直径1.9cm)の円筒状のミニカラムに0.35g/cm3の密度で充填し、ウサギ体外循環用カラムを作製した。本カラムは生理食塩液で洗浄し、ヘパリンをプライミングした後に、5mL/分の流速で健常ウサギに施行した。圧上昇の評価は、ウサギ体外循環用カラムの入口側にかかる圧力から出口側にかかる圧力の差分(以下、差圧)を指標に、循環を行ってから差圧が100mmHgを越えるまでの時間(最大60分間)をカラム通液可能時間として測定した。結果を表2に示す。
有機物吸着用担体4を直径26mmの円形に切り出し、孔サイズ0.3μmのHEPAフィルターを通過させたイオン交換水(フィルター水)50mLとともに清浄な容器に入れて10回転倒混和してから液を排出し、洗浄した。この洗浄操作をさらにもう1回繰り返した。洗浄した被験編地を攪拌型ウルトラホルダーUHP-25K(ADVANTEC社製)付属のベースプレートに載せてO-リングを重ねたのち直径18mmの円筒状容器(セル)の間に挟みこみ、ベース取付金具により固定した。ベースプレートの液出口をシリコーンチューブで塞ぎ、編地を底面側にして10mLのフィルター水を加え、水漏れがないことを確認した。ここにUHP-25K付属の攪拌セットを取りつけ、マグネティックスターラーRCN-7(東京理化器械社製)上で、攪拌セットが編地に接触しない状態で回転数600rpmにて5分間攪拌を行った。この液を採取し、光遮蔽型自動微粒子測定装置KL-04(リオン社製)で3mL測定し、1mL当たりの10μm以上の微粒子量を測定し、微粒子発生量(単位:個/mL)とした。結果を表2に示す。
有機物吸着用担体5を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、IL-6吸着率、HMGB-1吸着率、カラム通液可能時間、微粒子発生量を測定した。結果を表2に示す。
有機物吸着用担体7を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、IL-6吸着率、HMGB-1吸着率、カラム通液可能時間、微粒子発生量を測定した。結果を表2に示す。
有機物吸着用担体8を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、IL-6吸着率、HMGB-1吸着率、カラム通液可能時間、微粒子発生量を測定した。結果を表2に示す。
有機物吸着用担体9を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、IL-6吸着率、HMGB-1吸着率、カラム通液可能時間、微粒子発生量を測定した。結果を表2に示す。
有機物吸着用担体11を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、IL-6吸着率、HMGB-1吸着率、カラム通液可能時間、微粒子発生量を測定した。結果を表2に示す。
有機物吸着用担体13を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、微粒子発生量を測定した。結果を表3に示す。
有機物吸着用担体14を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、微粒子発生量を測定した。結果を表3に示す。
有機物吸着用担体15を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、微粒子発生量を測定した。結果を表3に示す。
有機物吸着用担体16を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、微粒子発生量を測定した。結果を表3に示す。
有機物吸着用担体17を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、微粒子発生量を測定した。結果を表3に示す。
有機物吸着用担体18を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、微粒子発生量を測定した。結果を表3に示す。
有機物吸着用担体19を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、微粒子発生量を測定した。結果を表3に示す。
吸着カラム1の圧力損失測定試験:
吸着カラム1を図3のように接続し、37℃(外温)でFBS溶液を保温した。次に2000pg/mLになるようにIL-8が添加されたFBS溶液を2500mL恒温水層中にプールした。続けて流量100mL/minで吸着カラム1に上記のFBS溶液を2時間通液し、通液中の入口圧測定装置9及び出口圧測定装置10によりそれぞれの圧力を測定し、入口圧測定装置で測定した圧力から出口圧測定装置で測定した圧力を差し引いた値を圧力損失として測定した。さらに、吸着カラム1を2時間通液後のFBS溶液中のIL-8濃度をELISAにより測定し、2000pg/mLからの減少率をIL-8循環吸着率として得た。圧力損失とIL-8循環吸着率の結果を表4に示す。
吸着カラム2を用いて、実施例14と同様の測定を行うことで、圧力損失とIL-8循環吸着率を得た。結果を表4に示す。
吸着カラム3を用いて、実施例14と同様の測定を行うことで、圧力損失とIL-8循環吸着率を得た。結果を表4に示す。

有機物吸着用担体4、5、7~9、11、13~19の真円度の測定:
有機物吸着用吸着担体4に含まれる海島型中実複合繊維の、繊維軸方向に対して垂直に切断した試料の断面を撮影した画像を、SEMで島成分が明瞭に観察できる倍率として画像を10ヶ所無作為に選定し、撮影した。得られた繊維断面画像に対して最小内包円(図1の2)を作成し、半径を0.1μm単位で算出した。さらに、最小内包円と同心であり、繊維内部に内包できる最大の円(以下、最大内接円,図1の3)を繊維断面内に作成し、半径を0.1μm単位で算出した。得られた最小内包円の半径から最大内接円の半径を差し引いた値を真円度とした。さらに、有機物吸着用担体4と同様の方法で有機物吸着用担体5、7~9、11、13~19についてもそれぞれ真円度を測定した。その結果、有機物吸着用担体4、5、7~9、11、13~19の真円度はいずれも0~15μmの範囲に入った。
有機物吸着用担体1を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、IL-6吸着率、HMGB-1吸着率、カラム通液可能時間、微粒子発生量を測定した。結果を表2に示す。
有機物吸着用担体2を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、IL-6吸着率、HMGB-1吸着率、カラム通液可能時間、微粒子発生量を測定した。結果を表2に示す。
有機物吸着用担体3を用いて、実施例1と同様の測定を行うことでアシッドオレンジ7吸着率、IL-8吸着率、IL-6吸着率、HMGB-1吸着率、カラム通液可能時間、微粒子発生量を測定した。結果を表2に示す。

有機物吸着用担体4同様の方法で有機物吸着用担体1~3、6、10、12の真円度を測定したところ、有機物吸着用担体12の真円度は18μmであり、有機物吸着用担体1~3、6、10の真円度はいずれも0~15μmの範囲に入った。

Claims (7)
- 細孔体積が0.05~0.5cm3/g、かつ、繊維径が25~60μmである海島型中実複合繊維を含む、有機物吸着用担体。
- 前記海島型中実複合繊維の表面に酸性官能基又は塩基性官能基を有するリガンドを含み、
前記酸性官能基又は前記塩基性官能基の含量は前記海島型中実複合繊維の乾燥重量1g当たり0.5~5.0mmolである、請求項1記載の有機物吸着用担体。 - 前記海島型中実複合繊維の海成分は、単一の熱可塑性樹脂からなり、
前記海島型中実複合繊維の島成分は、ポリオレフィンからなる、請求項1又は2記載の有機物吸着用担体。 - 前記海島型中実複合繊維の繊維軸方向に対し垂直な断面における、該海島型中実複合繊維の表面から最外島成分までの距離が1μm以上30μm未満であり、かつ、該海島型中実複合繊維の島成分の最大島径が0.1~2μmである、請求項1~3のいずれか一項記載の有機物吸着用担体。
- 血液成分の吸着除去用である、請求項1~4のいずれか一項記載の有機物吸着用担体。
- 請求項1~5のいずれか一項記載の有機物吸着用担体を備える、吸着カラム。
- 請求項1~5のいずれか一項記載の有機物吸着用担体を備え、該有機物吸着用担体の充填密度が0.15~0.40g/cm3である、吸着カラム。
