본 명세서는 성분 함량이 변화된 녹차 추출물을 포함하는 순환기 질환 개선용 조성물에 관한 것으로서, 본 명세서를 이용하여 부작용 없이 순환기 질환을 효과적으로 예방, 개선 및 치료할 수 있으므로, 순환기 질환 환자들에게 큰 도움을 줄 수 있고, 관련 산업 발전에도 기여할 수 있을 것이다.
Description
본 명세서는 성분 함량이 변화된 녹차 추출물을 포함하는 순환기 질환 개선용 조성물에 관한 것이다.
생체 내에서 혈액은 복잡하고 정교한 조절 시스템에 의해 항상 평형을 이루고 있어 정상적인 상태에서는 출혈이나 혈전 등에 의해 흐름이 방해 받지 않는다. 그러나, 여러 가지 요인으로 이러한 평형상태가 깨지면 혈관의 흐름이 원활하지 못하여 순환기 질환이 발생된다. 순환기 질환은 미국, 유럽뿐만 아니라 우리나라에서도 주요 사망 원인으로 대두되고 있으며, 특히 동맥경화성 질환인 허혈성 심질환(협심증, 심근경색증)과 뇌혈관 질환에 의한 사망률이 큰 폭으로 증가되고 있다.
순환기 질환 중 가장 대표적인 경우가 동맥경화증인데, 동맥경화증은 심장이나 뇌 등 중요장기에 허혈성 상태를 초래하여 심근경색이나 뇌경색을 일으키는 매우 위험한 질환이다. 질병관리본부에서 2006년 발표한 한국인의 사망원인에 대한 자료에 의하면, 뇌출혈, 동맥경화증, 심근경색 등과 같은 순환기 장애로 인해 사망하는 환자의 수가 암 이상으로 높은 것으로 나타나 있다. 이러한 순환기 장애 질환의 높은 발병률의 원인은 식생활의 서구화와 스트레스 등 여러 요인에 의해 동맥경화증, 비만, 당뇨병 등의 만성 성인병 유병률의 증가에 있을 것으로 생각된다.
또한, 고지혈증은 우리 몸의 혈중 콜레스테롤과 중성지방(triglyceride) 농도가 정상 이상으로 증가되어 있는 상태를 나타내는 것으로서, 고혈압 및 흡연과 더불어 관상동맥질환의 3대 위험 인자로 알려져 있다. 고지혈증은 혈액검사에서 총 콜레스테롤이 240 ㎎/㎗ 이상이거나, 중성지방이 200 ㎎/㎗ 이상일시 진단되는 질환으로서, 근래에 돌연사가 젊은 층에 빈발하면서 더욱 큰 관심이 모아지고 있다.
특히, 고혈압, 고지혈증과 같은 순환기 질환이 돌연사의 주된 원인으로 지목 되고 있으며, 순환기 환자는 지속적으로 증가하고 있는 실정이다. 또한, 나이가 많을수록 환자가 더 많은 것으로 나타나, 수명 연장 및 고령화 시대 진입에 따른 대비 및 관리의 중요성이 더욱 높아진 실정이다.
심혈관계 질환의 치료에 대해서는 그간 많은 연구가 있어 왔으나, 부작용이 적으면서도 효과적인 천연물 유래의 물질에 대한 연구는 만족할만한 성과를 내지 못하고 있는 실정이다.
본 명세서는 부작용이 극히 적으면서도 효과가 뛰어난 천연물 유래의 순환기 질환 개선용 조성물을 제공하는 것을 과제로 한다.
상기 과제를 해결하기 위하여, 본 명세서는 차 추출물을 포함하는 순환기 질환 예방 또는 개선용 조성물을 제공한다.
특히, 본 명세서는 조성물 총 중량을 기준으로, 8 내지 25 중량%의 (-)-갈로카테킨 갈레이트((-)-gallocatechin gallate, GCG), 1 내지 8 중량%의 (-)-카테킨 갈레이트((-)-catechin gallate, CG), 및 7 내지 15 중량%의 (-)-에피갈로카테킨 갈레이트((-)-epigallocatechin gallate, EGCG)를 포함하는 녹차 추출물을 유효성분으로 포함하는 순환기 질환 예방 또는 개선용 조성물을 제공한다.
본 명세서를 이용하여, 혈관에 영향을 미치는 다양한 효과들을 통해 부작용 없이 순환기 질환을 효과적으로 예방, 개선 및 치료할 수 있으므로, 순환기 질환 환자들에게 큰 도움을 줄 수 있고, 관련 산업 발전에도 기여할 수 있을 것이다.

본 명세서에서 “녹차 추출물”은 추출방법, 추출용매, 추출된 성분 또는 추출물의 형태를 불문하고, 차과에 속하는 상록수인 차(카멜리아 시넨시스; Camellia sinensis)로부터 추출한 것을 포함하고, 상기 추출한 것을 특정 용매로 분획한 분획물을 포함한다. 상기 차는 차나무 잎, 꽃, 줄기, 열매, 뿌리, 줄기, 및 뿌리의 심재로 구성된 군에서 선택되는 하나 이상이 포함되며, 바람직하게는 잎일 수 있다. 또한, 상기 추출물의 형태는 바람직하게는 분말형태일 수 있다. 상기 추출 또는 분획은 물, 유기 용매, 또는 이들의 혼합용매를 사용하여 할 수 있다. 유기용매는 알코올, 아세톤, 헥산, 에틸아세테이트, 이산화탄소, 또는 이들 중 둘 이상의 혼합용매를 사용할 수 있으나 이에 한정되지는 않으며, 녹차의 유효 성분이 파괴되지 않거나 최소화된 조건에서 실온 또는 가온하여 추출 또는 분획할 수 있다. 상기 알코올은 C1~C5의 저급 알코올일 수 있다. 추출 또는 분획의 횟수나 방법은 특별히 제한되지 않으며, 예를 들어 냉침 추출, 초음파 추출, 환류 냉각 추출, 열수 추출 등의 방법을 사용할 수 있고, 바람직하게는 냉침 또는 가온하여 유효성분을 추출 또는 분획하고 여과한 다음, 그 여과액을 감압농축하여 본 발명의 녹차 추출물을 얻을 수 있다.
본 명세서에서 “총 에피카테킨”이란, (-)-에피갈로카테킨(EGC), (-)에피카테킨(EC), (-)-에피갈로카테킨 갈레이트(EGCG), 및 (-)-에피카테킨 3-O-갈레이트(ECG)을 포함하는 것이다.
본 명세서에서 “순환기 질환”은 순환기, 즉 심장, 동맥계 및 정맥계의 이상에 따른 질환을 말하며, 심부전, 협심증, 심근경색 등의 심장병, 동맥경화, 고지혈증, 고콜레스테롤혈증, 고혈압, 뇌졸중 및 혈전증을 포함한다.
본 명세서에서 “산화 스트레스”란, 유해산소의 부작용을 일컫는다. 우리 몸은 체내의 활성산소 양을 자체적으로 조절하는데, 여러 가지 상황으로 인해 유해산소의 생성이 급격히 증가하거나, 이들을 제거하는 기능이 저하될 경우 유해산소에 의한 각종 질병이 유발된다. 이에 따른 유해산소의 부작용을 산화 스트레스라 한다. 활성산소가 과잉 생성되어 산화스트레스가 체내에 지속적으로 축적되면 세포의 유전자에 영향을 미치거나 손상이 발생한다. 예를 들어, H2O2에 의해 과잉생성된 활성산소종은 자유 라디컬(free radical)을 가져 안정되지 못한 상태가 되며, 그로 인해 강한 활성을 가진다. 그리하여 세포내의 단백질, 지질 등을 산화시킴으로써 세포의 항상성을 파괴하고, 세포를 사멸시키는 것이다.
본 명세서에서 "약학적으로 허용 가능"이란 통상의 의약적 복용량(medicinal dosage)으로 이용할 때 상당한 독성 효과를 피함으로써, 동물, 더 구체적으로는 인간에게 사용할 수 있다는 정부 또는 이에 준하는 규제 기관의 승인을 받을 수 있거나 승인 받거나, 또는 약전에 열거되거나 기타 일반적인 약전에 기재된 것으로 인지되는 것을 의미한다.
본 명세서에서 "약학적으로 허용 가능한 염"은 약학적으로 허용 가능하고 모 화합물(parent compound)의 바람직한 약리 활성을 갖는 본 발명의 일측면에 따른 염을 의미한다.
혈액 속에 콜레스테롤 같은 지질이 증가하면 혈관 벽에 조금씩 쌓여서 혈관 벽이 두꺼워지고 혈관 내부가 좁아지며, 지질이 쌓인 곳에 혈전이 생기게 된다. 동맥경화는 이와 같이 혈관벽이 두꺼워지고 혈관 내강이 좁아지는 심혈관 질환으로서, 돌연사의 주요 원인 중 하나이다. 동맥경화는 다양한 산화적 스트레스와 염증인자들에 혈관내피세포가 자극받으면서 시작되는데 이런 자극에 의해 혈관벽이 느슨해 지고 혈중에 있는 과도한 지질이 쌓이면서 지방선조 (fatty streak)을 생성한다. 또한 혈중에 생성된 지질과산화물은 대식세포에 포식되어 거품세포를 생성하게 되는데 이런 거품세포는 동맥경화 발달에 중요한 요인으로 작용한다. 그러므로 산화적 스트레스를 막고 지질 생성을 억제하는 것이 동맥경화 예방에 중요하기 때문에 본 발명은 일 측면에서, 기존 열수 녹차 추출물에 비해 효능을 높이는 조성물을 찾고자 하였다.

일 측면에서, 상기 추출물은 물, C1 내지 C6의 알코올 및 이들의 혼합물 중 어느 하나 이상에 의해 1회 이상 추출한 것 일 수 있다. 일 구현예로서, 상기 알코올은 에탄올일 수 있다.

순환기 질환 중에서도 동맥경화증은 지질대사의 이상과 혈관의 염증, 동맥혈전의 형성으로 대변되는 질환으로서, 특히 동맥혈전의 형성은 동맥경화증의 급·만성 합병증으로 나타나고, 고혈압, 심근경색, 뇌졸중을 일으키는 주요 위험인자로 알려져 있다. 혈소판은 이러한 동맥혈전의 주요 구성 요소이며, 또한 동맥경화 플라크(plaque)의 발생과 진행에도 관여한다고 알려져 있다.
혈중에 콜레스테롤 및 지질이 증가하면 동맥경화증이나 고지혈증이 유발되기 쉬우며, 동맥경화증, 고지혈증 및 고콜레스테롤 혈증에 의해 혈류가 방해되면 심장마비, 뇌졸중 등이 유발될 수 있다. 현대인의 식생활 환경의 향상에 따른 과다한 영양 섭취로 인해 과다 섭취된 콜레스테롤은 신체 내의 세포 요구를 충족시키고 남게 된다. 이러한 콜레스테롤은 저밀도 지단백질(low-density lipoprotein, LDL)을 통해 떠돌다가 변형된 형태로 바뀌어 혈관 내막에 흡착되고, 포말세포(foam cell), 지방반(fatty streak), 아테롬(atherom)으로 전환되기도 하며, 이들은 혈관의 수축, 팽창 능력을 떨어뜨려 동맥경화, 관상 동맥 질환, 뇌졸중, 말초혈관 협착, 고혈압 등을 일으키는 원인이 되고 있다. 포말 세포들은 염증 반응과 세포성장을 반복하면서 혈관벽을 파괴하거나 비후하여, 혈액중의 혈소판이 점착됨으로 혈전을 형성하게 한다. 이러한 진행과정은 결국, 동맥경화에 의한 혈관의 협착과 혈전에 의한 혈관의 폐쇄로 이어진다.
본 발명의 일 측면에 따른 추출물 및 조성물은 자유라디칼 생성 억제, HMG-CoA 리덕타아제(HMG-CoA reductase)의 양 또는 활성 감소, 안지오텐신 전환 효소의 양 또는 활성 감소, 및 혈관내피세포에서의 산화질소 생산 감소 등의 효과를 발휘하므로, 이를 통해 지질과산화물 생성 억제, 콜레스테롤 생성 억제, 지방선조 생성 억제, 혈전 생성 억제, 혈관강 직경 또는 체적의 증가, 혈압 감소, 혈액순환 개선, 및 혈관 이완 등을 이룰 수 있고, 이를 통해 동맥경화, 고지혈증, 고콜레스테롤혈증, 고혈압, 뇌졸중 및 혈전증과 같은 순환기 질환을 효과적으로 예방, 치료 및 개선할 수 있는 것이다.
상기 조성물은 식품 또는 약학 조성물일 수 있다.
상기 식품 조성물의 제형은 특별히 한정되지 않으나, 예를 들어, 정제, 과립제, 환제, 분말제, 드링크제와 같은 액제, 캐러멜, 겔, 바, 티백 등으로 제형화될 수 있다. 각 제형의 식품 조성물은 유효 성분 이외에 해당 분야에서 통상적으로 사용되는 성분들을 제형 또는 사용 목적에 따라 당업계의 통상의 기술자가 어려움 없이 적의 선정하여 배합할 수 있으며, 다른 원료와 동시에 적용할 경우 상승 효과가 일어날 수 있다. 또한 상기 식품은 건강기능식품일 수도 있다.
상기 조성물은 단순 섭취, 음용, 주사 투여, 스프레이 투여 또는 스퀴즈 투여 등 다양한 방법으로 투여될 수 있다.
본 발명의 일측면에 따른 식품 조성물에 있어서, 상기 유효 성분의 투여량 결정은 당업계의 통상의 기술자의 수준 내에 있으며, 투여하고자 하는 대상의 연령, 건강 상태, 합병증 등 다양한 요인에 따라 달라질 수 있다.
본 발명의 일측면에 따른 식품 조성물은, 예를 들어, 츄잉껌, 캐러멜 제품, 캔디류, 빙과류, 과자류 등의 각종 식품류, 청량 음료, 미네랄 워터, 알코올 음료 등의 음료 제품, 비타민이나 미네랄 등을 포함한 건강기능식품 제품일 수 있다.
상기 외에, 본 발명의 일 측면에 따른 식품 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 증진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 포함할 수 있다. 그 밖에 본 발명의 일측면에 따른 식품 조성물들은 천연 과일 쥬스 및 과일 쥬스 음료 및 야채 음료의 제조를 위한 과육을 포함할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 그렇게 중요하진 않으나 본 발명의 일측면에 따른 조성물 100 중량부 당 0 내지 약 60 중량부의 범위에서 포함되는 것이 일반적이다.
본 발명의 일측면에 따른 상기 약학 조성물은 경구, 비경구, 직장, 국소, 경피, 정맥 내, 근육 내, 복강 내, 피하 등으로 투여될 수 있다. 경구 투여를 위한 제형은 정제(錠劑), 환제(丸劑), 연질 및 경질 캅셀제, 과립제(顆粒劑), 산제, 세립제, 액제, 유탁제(乳濁濟) 또는 펠렛제일 수 있으나, 이에 제한되는 것은 아니다. 비경구 투여를 위한 제형은 용액제, 현탁제, 유액제, 겔, 주사제, 점적제, 좌제(坐劑), 패취 또는 분무제일 수 있으나, 이에 제한되는 것은 아니다. 상기 제형은 당해 분야의 통상적인 방법에 따라 용이하게 제조될 수 있으며, 계면 활성제, 부형제, 수화제, 유화 촉진제, 현탁제, 삼투압 조절을 위한 염 또는 완충제, 착색제, 향신료, 안정화제, 방부제, 보존제 또는 기타 상용하는 보조제를 추가로 포함할 수 있다.
본 발명의 일 측면에 따른 상기 조성물은 약학적으로 허용가능한 염을 포함할 수도 있는데, 상기 염은 (1) 염산, 브롬화수소산, 황산, 질산, 인산 등과 같은 무기산으로 형성되거나; 또는 아세트산, 프로파이온산, 헥사노산, 시클로펜테인프로피온산, 글라이콜산, 피루브산, 락트산, 말론산, 숙신산, 말산, 말레산, 푸마르산, 타르타르산, 시트르산, 벤조산, 3-(4-히드록시벤조일) 벤조산, 신남산, 만델산, 메테인설폰산, 에테인설폰산, 1,2-에테인-디설폰산, 2-히드록시에테인설폰산, 벤젠설폰산, 4-클로로벤젠설폰산, 2-나프탈렌설폰산, 4-톨루엔설폰산, 캄퍼설폰산, 4-메틸바이시클로 [2,2,2]-oct-2-엔-1-카르복실산, 글루코헵톤산, 3-페닐프로파이온산, 트리메틸아세트산, tert-부틸아세트산, 라우릴 황산, 글루콘산, 글루탐산, 히드록시나프토산, 살리실산, 스테아르산, 뮤콘산과 같은 유기산으로 형성되는 산 부가염(acid addition salt); 또는 (2) 모 화합물에 존재하는 산성 프로톤이 치환될 때 형성되는 염을 포함할 수 있다.
본 발명의 일측면에 따른 상기 약학 조성물의 적용량 또는 투여량은 투여 받을 대상의 연령, 성별, 체중, 병리 상태 및 그 심각도, 투여 경로 또는 처방자의 판단에 따라 달라질 것이다. 이러한 인자에 기초한 유효성분 투여량 결정은 당업계 통상의 기술자의 수준 내에 있다.
이하, 실시예, 비교예, 실험예, 및 제형예를 들어 본 명세서의 구성 및 효과를 보다 구체적으로 설명한다. 그러나 이들 예는 본 명세서에 대한 이해를 돕기 위해 예시의 목적으로만 제공된 것일 뿐 본 명세서의 범주 및 범위가 하기 예에 의해 제한되는 것은 아니다.
[실시예 1] 성분 함량이 변화된 녹차 추출물의 제조
100g의 녹차(Camellia sinensis, 제주 오설록 농장)에 50% 에탄올 1000ml 를 가하여 60도씨에서 1시간 환류 교반하였다. 상기 시료의 온도를 실온으로 낮추고 여과하여 얻은 용액을 감압증류(에탄올 제거)하여 진한 갈색분말의 추출물 23g을 수득하였다.(수율 23%) 이후, 상기 시료 1 10g을 물 90ml에 녹인 후 80도씨에서 30분 ~ 8시간 교반시켰다. 이후 온도를 실온으로 낮추고 불용성물질을 여과하고 감압농축하여 열 숙성 녹차 추출물 10g을 수득하였다. 이 때, 상기 교반 시간 별로 얻은 상기 열숙성 녹차 추출물에 대해 하기 표 1과 같은 장치를 이용하여 시간대별로 GCG 등의 함량 변화를 측정하면서, GCG가 가장 많은 양 존재하는 시간대를 확인하였고, 그 시간대에서 교반을 멈추고 열 숙성 녹차 추출물(GT-LE-10GCG, 실시예 1, 시료 2) 10g을 수득하였다.
수득된 상기 추출물의 조성을 분석한 조건은 하기 표 1과 같고, 교반 시간별 성분 분석 결과는 표 2와 같다. 상기 추출물의 경우 기존의 녹차 추출물과는 조성이 다르다는 것을 확인하였으며, 구체적으로, 8 내지 25 중량%의 (-)-갈로카테킨 갈레이트((-)-gallocatechin gallate, GCG), 1 내지 8 중량%의 (-)-카테킨 갈레이트((-)-catechin gallate, CG), 및 7 내지 15 중량%의 (-)-에피갈로카테킨 갈레이트((-)-epigallocatechin gallate, EGCG)가 포함되는 것을 알 수 있었고, GCG가 10 내지 13 중량%인 시간대에서 EGCG는 8 내지 10 중량%, CG는 1 내지 3 중량%인 것을 확인하였다.

[비교예 1] 일반녹차추출물(식품의약품안전처 고시형 녹차추출물)의 제조
1g의 녹차(Camellia sinensis, 제주오설록농장)에 물 1000ml 를 가하여 80도씨에서 1시간 환류교반하였다. 이후 온도를 실온으로 낮추고 여과하여 얻은 용액을 감압하여 진한갈색분말의 고시형 녹차추출물(비교예 1) 0.29g을 수득하였다. (수율 29%)
상기 실시예 1과 같은 방법으로 성분 분석을 실시하였다. 실시예 1에 따라 추출물을 제조하되 다른 로트(LOT)의 녹차를 이용하여 시간에 따라 성분 함량을 추적(tracking)하며 4 내지 5시간 교반한 때의 추출물(표 3의 '실시예 1')과 비교예 1의 일반 녹차 추출물(표 3의 '비교예 1')을 대비한 결과 표 3과 같았다.

[실험예 1] 항산화능 효능 비교
제조 방법에 따른 추출물의 항산화능 활성 차이를 비교하였다.
DPPH(2,2-diphenyl-1-picrylhydrazyl)는 그 자체가 매우 안정한 자유라디칼(Free radical)로서 특징적인 광흡수를 나타내는 보라색 화합물이다. 상기 실시예 1 및 비교예 1에 DPPH 100μM의 에탄올 용액을 첨가하여 37℃에서 30분간 반응시킨 후 라디칼이 제거되는 효능을 515nm에서 흡광도로 평가하였다.
그 결과는 하기 표 4와 같으며, 비교예 1의 추출물에 비해 실시예 1의 추출물이 항산화 효과가 훨씬 뛰어남을 확인하였다.

[실험예 2] HMG-CoA reductase (3-hydroxy-3-methyl-glutaryl-CoA reductase 또는 HMGR)억제 효능 평가
콜레스테롤을 합성하는 주요 효소인 HMGR에 대한 억제효능을 평가하였다. 이 효소의 억제 효능은 HMG-CoA를 메발론산염(mevalonate)과 코엔자임 A(coenzyme A)로 변환시킬 때 필요한 NADPH의 감소로 인한 흡광도 변화로 측정하였다. 실시예 1 및 비교예 1의 추출물(각각 30 μg/ml)에 HMGR을 섞고 HMG-CoA와 NADPH를 넣은 완충액을 가한 후 즉시 2분마다 흡광도를 340nm에서 측정하였다. 필요한 시료와 장비는 통상의 기술자가 시중에서 용이하게 입수 가능한 것으로 사용하였다.
그 결과는 표 5와 같으며, 비교예 1의 추출물과 달리 실시예 1의 추출물은 콜레스테롤 합성 효소를 현저하게 억제함을 알 수 있었다.

[실험예 3] 고콜레스테롤 식이에 따른 지질과산화물 생성 억제
실시예 1의 추출물에 대해 고콜레스테롤 식이에 의해 유도되는 지질과산화물 생성 억제효능을 평가하였다.
구체적으로, 약 550g정도 몸무게를 가진 10주령 수컷 기니피그에 대해 정상 사료를 급이한 군, 고콜레스테롤 사료(사료 총중량 대비 1 중량% 콜레스테롤과 10 중량% 코코넛 오일이 첨가된 것)를 급이한 군, 상기 고콜레스테롤 사료에 사료 총 중량 대비 0.15 중량%의 실시예 1 추출물을 첨가하여 급이한 군으로 구분하여 8주 뒤 마지막 날 절식 후 채혈하였다.
혈장에 있는 지질과산화물을 도데실황산나트륨(sodium dodecyl sulfate) 완충액으로 분해하여 생성되는 말론디알데하이드(malondialdehyde, MDA)에 티오바르비툴산(thiobarbituric acid)를 가하여 95℃에서 1시간 동안 반응시킨 후 생성된 TBARS(Thiobarbituric acid reactive substances)를 여기파장 540nm, 방출파장 590nm에서 형광을 측정하여 정량하였다. 이때 사용되는 실험시료와 장비는 통상의 기술자가 지질과산화물 정량을 위해 시중에서 용이하게 입수할 수 있는 것들을 사용하였다.
그 결과는 도 1과 같으며, 고콜레스테롤의 사료를 급이한 군에 비해 고콜레스테롤 사료에 실시예 1의 추출물을 첨가하여 급이한 군에서 혈중 지질과산화물의 양이 현저하게 감소하였음을 알 수 있었다.
[실험예 4] 안지오텐신 전환 효소(Angiotensin-converting enzyme, ACE) 억제 효능 평가
안지오텐신 전환 효소(Angiotensin-converting enzyme, ACE)는 안지오텐신 I(Angiotensin I)을 혈관을 수축시키는 활성형 안지오텐신 II(angiotensin II)로 전환시키는 효소로 혈압강하제의 주요 타겟 효소이다. 이 효소의 억제 효능은 실시예 1 및 비교예 1의 추출물과 ACE, 기질인 3-hydroxybutylyl-Gly-Gly-Gly를 반응시켜 생성된 3-hyroxybutylic acid (3HB)양을, 반응에 필요한 NADH의 감소로 인한 발색시스템의 450nm 흡광도 변화를 측정하여 평가하였다. 실험에 필요한 시료와 장비는 시중에서 입수할 수 있는 것들을 이용하였다.
그 결과, 하기 표 6과 같이 실시예 1의 ACE 억제 효과가 비교예 1보다 현저함을 확인하였다.

[실험예 5] 혈관이완인자인 산화질소 (nitric oxide, NO) 생성 증가 평가
혈관내피세포에서 혈관내피산화질소합성효소(endothelial nitric oxide synthase, eNOS)에 의해 생성되는 산화질소(nitric oxide)는 정상적인 혈관기능에서 혈관을 이완시켜 혈행을 촉진하고 혈압을 낮추는 중요한 생리활성 인자이다. 사람 혈관내피세포인 HUVEC 세포에 실험예 1 및 비교예 1의 추출물을 처리하고 3시간, 6시간 후 증가하는 nitric oxide의 양을 0.2% naphthylethylenediamine (Sigma, USA) 수용액과 2% sulfanilamide (Sigma, USA) 의 5% 인산 용액을 측정 직전 동량부피를 혼합하여 제조한 Griess reagent를 이용하여 550nm에서 흡광도를 측정해서 평가하였다.
그 결과, 도 2와 같이, 비교예(비교예 1) 실시예(실시예 1) 모두 동일 용매만 처리한 대조군(vehicle)에 비해 생성량이 증가하였고, 비교예 보다 실시예를 처리하였을 때 2배 정도의 더 높은 생성을 나타내었다.
[제형예 1] 연질 캡슐
상기 실시예 1에 따른 추출물을 150mg으로 준비하고, 락토오스 440mg, 옥수수 전분 430mg 및 스테아린산 마그네슘 2mg을 혼합하여 연질 캡슐 충진액을 제조하였다. 그리고, 상기와 별도로 젤라틴 66 중량부, 글리세린 24 중량부 및 솔비톨액 10 중량부의 비율로 연질 캡슐 시트를 제조하고 상기 충진액을 충진시켜 연질 캡슐을 제조하였다.
[제형예 2] 정제
상기 실시예 1에 따른 추출물을 150mg으로 준비하고, 비타민 E 15mg, 비타민 C 15mg, 갈락토올리고당 250㎎, 유당 60㎎ 및 맥아당 140㎎을 혼합한 뒤 유동층 건조기를 이용하여 과립화한 후 당 에스테르(sugar ester) 8㎎을 첨가하였다. 이 조성물을 통상의 방법으로 타정하여 정제를 제조하였다.
[제형예 3] 드링크제
상기 실시예 1에 따른 추출물을 80mg 준비하고, 비타민 E 9mg, 비타민 C 9mg, 포도당 10g, 구연산 0.6g, 및 액상 올리고당 25g을 혼합한 후 정제수 400㎖를 가하여 충진하였다. 병에 충진한 후 30℃에서 4~5 초간 살균하여 드링크제를 제조하였다.
[제형예 4] 과립제
상기 실시예 1에 따른 추출물을 150mg 준비하고, 비타민 E 9mg, 비타민 C 9mg, 무수결정 포도당 250㎎ 및 전분 550㎎을 혼합한 후, 유동층 과립기를 사용하여 과립으로 성형한 뒤 포에 충진하여 과립제를 제조하였다.
[제형예 5] 건강 식품
상기 실시예 1에 따른 추출물을 150 mg 준비하고, 비타민 혼합물(비타민 A 아세테이트 70 ㎍, 비타민 E 1.0 ㎎, 비타민 B1 0.13 ㎎, 비타민 B2 0.15 ㎎, 비타민 B6 0.5 ㎎, 비타민 B12 0.2 ㎍, 비타민 C 10 ㎎, 비오틴 10 ㎍, 니코틴산아미드 1.7 ㎎, 엽산 50 ㎍)과 무기질 혼합물(황산제1철 1.75 ㎎, 산화아연 0.82 ㎎, 탄산마그네슘 25.3 ㎎, 제1인산칼륨 15 ㎎, 제2인산칼슘 55 ㎎, 구연산칼륨 90 ㎎, 탄산칼슘 100 ㎎, 염화마그네슘 24.8 ㎎)을 조합하여 건강식품을 제조하였다.
[제형예 6] 건강 음료
상기 실시예 1에 따른 추출물을 50 mg 준비하고, 구연산 1000 ㎎, 올리고당 100 g, 매실농축액 2 g, 타우린 1 g, 정제수 잔량을 첨가하여 900 mL의 건강음료를 제조하였다.
이상으로 본 명세서의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현예일 뿐이며, 이에 본 명세서의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 명세서의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.

- 조성물 총 중량을 기준으로, 8 내지 25 중량%의 (-)-갈로카테킨 갈레이트((-)-gallocatechin gallate, GCG), 1 내지 8 중량%의 (-)-카테킨 갈레이트((-)-catechin gallate, CG), 및 7 내지 15 중량%의 (-)-에피갈로카테킨 갈레이트((-)-epigallocatechin gallate, EGCG)를 포함하는 녹차 추출물을 유효성분으로 포함하는 순환기 질환 예방 또는 개선용 조성물.
- 제1항에 있어서, 상기 추출물 내의 총 에피 카테킨 함량은 상기 조성물 총 중량을 기준으로 20 중량% 이하인, 조성물.
- 제1항에 있어서, 상기 추출물 내의 GCG 및 CG의 합계량은 상기 조성물 총 중량을 기준으로 11 내지 25 중량%인, 조성물.
- 제1항에 있어서, 상기 추출물은 물, C1 내지 C6의 알코올 및 이들의 혼합물로 이루어진 군에서 선택된 하나 이상에 의해 1회 이상 추출한 추출물인, 조성물.

- 제1항 내지 제11항 중 어느 한 항에 있어서, 상기 조성물은 식품 또는 약학 조성물인, 조성물.
