Abstract
Se describe un método para producir un virus ADN citoplásmico eucariótico por clonación molecular directo de una molécula de ADN modificada que comprende un genoma de virus ADN citoplásmico modificado. El método inventivo comprende las etapas de (I) modificar bajo condiciones extracelulares una molécula de ADN que incluye un genoma de un primer virus ADN citoplásmico, para producir una molécula de ADn modificada que comprende el genoma del virus ADN citoplásmico; (II) introducir la molécula de ADN modificada dentro de una primer célula huésped la cual empaca la molécula de ADN modificada dentro del virión infeccioso: y (III) recuperar de la célula huésped, los viriones constituídos por el genoma viral modificado. La célula huésped es infectada con un virus de ayuda el cual es expresado para empacar el genoma viral modificado dentro del virión infeccioso. Se describen ejemplos de empacamiento de un genoma de poxvirus modificado por un poxvirus de ayuda del mismo o diferente género. También se discuten vectores de poxvirus novedosos para la clonación molecular directa de estructuras de lectura abiertas dentro de un sitio de anclaje para enzima de restricción que es único en el vector. En un modelo de vector de poxvirus, la estructura de lectura abierta es transcrita por un promotor localizado en un vector de ADN aguas arriba de un sitio de clonación múltiple que comprende algunos sitios de anclaje únicos.
