Abstract

본 발명은 인간 면역결핍 바이러스 타입 1(HIV-1)의 항원 단백질 p24를 코딩하는 유전자를 포함하는 발현백터에 의해 형질전화된 대장균을 배양하여 상기 유전자를 발현시키고, 상기 p24를 함유하는 상기 대장균 배양물의 추출물을 이온교환 크로마토그래피 및 젤 여과 크로마토그래피하는 단계를 포함하는 HIV-1의 항원 단백질 p24를 제조하는 방법에 관한 것으로, HIV 감염 진단시 강도와 정확도가 높은 항원 단백질을 제공한다.

Claims (4)

  1. 인간 면역결핍 바이러스 타입 1(HIV-1)의 항원 단백질 p24를 코딩하는 유전자를 포함하는 발현백터에 의해 형질전화된 대장균을 배양하여 상기 유전자를 발현시키고, 상기 p24를 함유하는 상기 대장균 배양물의 추출물을 이온교환 크로마토그래피 및 젤 여과 크로마토그래피하는 단계를 포함하는, HIV-1의 항원 단백질 p24를 제조하는 방법.
  2. 제1항에 있어서, 상기 대장균이 플라스미드 pYCM-p24 또는 플라스미드 pYKM-p24에 의해 형질전환된 대장균임을 특징으로 하는 방법.
  3. 제1항 또는 제2항에 있어서, 상기 대장균의 배양 후 단계에서 히드록실아민으로 절단하는 단계를 추가로 포함하는 방법.
  4. 제1항에 있어서, 상기 이온 교환 크로마토그래피에 DEAE 세파로스 컬럼을 사용하고, 상기 젤 여과 크로마토그래피에 세파크릴 S-200 HR 칼럼을 사용하는 것을 특징으로 하는 방법.
