Abstract
La presente invención se relaciona a un método para sintetizar una molécula de ARN de una secuencia dada, que comprende el paso de determinación de la fracción (1) para cada uno de los cuatro nucleótidos G, A, C y U en dicha molécula de ARN, y el paso de síntesis de dicha molécula de ARN mediante transcripción in vitro en una mezcla de reacción optimizada por secuencia, donde dicha mezcla de reacción optimizada por secuencia comprende los cuatro ribonucleótidos GTP, ATP, CTP y UTP, donde la fracción (2) de cada uno de los cuatro ribonucleótidos en la mezcla de reacción optimizada por secuencia corresponde a la fracción (1) del nucleótido respectivo en dicha molécula de ARN, un amortiguador, un plantilla de ADN y una ARN polimerasa. Además, la presente invención se relaciona a un biorreactor (1) para la síntesis de moléculas de ARN de una secuencia dada, siendo que el biorreactor (1) posee un módulo de reacción (2) para llevar a cabo las reacciones de transcripción in vitro en una mezcla de reacción optimizada por secuencia, un módulo de captura (3) para capturar temporalmente las moléculas de ARN transcritas, y un módulo de control (4) para controlar la alimentación de la mezcla de reacción optimizada por secuencia en el módulo de reacción (2), donde el módulo de reacción (2) comprende una membrana de filtración (21) para separar los nucleótidos de la mezcla de reacción optimizada por secuencia, y el control de la alimentación de componentes de la mezcla de reacción optimizada por secuencia mediante el módulo de control (4) se basa en una concentración medida de nucleótidos separados.
