Abstract
En la presente se proporcionan métodos para la fabricación de células T; en ciertas modalidades se proporcionan métodos para fabricar células T que expresan un receptor de superficie celular que reconoce una porción antigénica específica en la superficie de una célula objetivo; dichos métodos pueden incluir, pero no se limitan a, los pasos de (1) enriquecer una población de linfocitos obtenidos de un sujeto donador; (2) estimular la población de linfocitos con uno o más agentes estimuladores de la célula T para producir una población de células T activadas, en donde la estimulación se realiza en un sistema cerrado utilizando un medio de cultivo libre de suero; (3) transducir la población de células T activadas con un vector viral que comprende una molécula de ácido nucleico que codifica para el receptor de superficie celular, usando una transducción de ciclo sencillo para producir una población de células T transducidas, en donde la transducción se realiza en un sistema cerrado utilizando un medio de cultivo libre de suero; y (4) expandir la población de células T transducidas durante un tiempo predeterminado para producir una población de células T diseñadas, en donde la expansión se realiza en un sistema cerrado utilizando un medio de cultivo libre de suero; también se proporcionan en la presente poblaciones de células T diseñadas producidas por los métodos que describen en la presente y sus composiciones farmacéuticas.
