Description
본 발명은 신규의 디스폰산 유도체, 이들의 제조방법 및 이들을 함유하는 제약 조성물에 관한 것이다.
DE-OS 18 13 659호에는 파제트병의 치료를 위한 제제로서 중요한 1-히드록시에탄-1,1-디포스폰산과 같은 디포스폰산 유도체가 기술되어 있다. DE 896,453호, EP-A-175,315호, DE-A-24 05 254호, DE-OS 25 34 391호 및 EP 96,931호에는 증가된 골흡수의 치료를 위해 사용할 수 있는 우수한 칼슘착물 형성제로서, 아미노알칸-1,1-디포스폰산을 기술되어 있다. 그러나, 치료적으로 효과적인 투여량으로 사용하는 경우에, 이러한 물질들은 자주 부작용을 나타낸다.
본 발명의 목적은 가장 낮은 가능한 투여량으로 치료작용을 나타내는 아미노알칸-디포스폰산염을 발견하는 데에 있다.
본 발명자들은 질소원자가 완전히 알킬화되고, 그로 인해 알킬기중 1개가 C4이상을 함유하는 상기 화합물의 유사체들이 이러한 목적을 달성하고, 칼슘대사 장애의 폭넓은 치료를 위한 우수한 칼슘착물 형성제로서 적합함을 발견하였다. 특히, 이러한 화합물은 골격형성 및 분해가 장애되는 경우에 유용하게 사용할 수 있으며, 예를 들면 골다공증, 파제트병, 베크테레프병등과 같은 골격계통의 질병의 치료를 위해 사용할 수 있다.
그러나 이러한 성질들에 기초하여, 상기 화합물은 골전이, 요석중의 치료 및 이소골화의 예방을 위해 사용할 수도 있다. 칼슘대사의 이러한 영향때문에 상기 화합물은 또한 류머티스성 관절염, 골관절염 및 변형과절증의 치료를 위한 주약을 형성한다.
즉, 본 발명의 주제는 하기 일반식(Ⅰ)의 디포스폰산염 및 약리적으로 허용될 수 있는 이들의 염이다:
상기 식에서, R1은 메틸, n-프로필, 이소프로필, 3-메틸부틸, 펜틸, 노닐 또는 시클로헥실메틸 라디칼이고; R2는 부틸, 이소부틸, 3-메틸부틸, 펜틸, 헵틸, 노닐, 운데실, 도데실, 테트라데실, 헥사데실, 옥타데실, 시클로헥실, 시클로헥실메틸 또는 벤질기이고; R3는 수소원자 또는 메틸, 에틸, 이소프로필 또는 이소부틸이고; X는 메틸렌, 에틸렌 또는 프로필렌기이고; Y는 수소원자, 이미노 또는 히드록실이다.
바람직하게는, 치환기 R1은 메틸, n-프로필, 이소프로필, 3-메틸부틸, 펜틸 또는 노닐기이다.
R2는 바람직하게는 부틸, 이소부틸, 3-메틸부틸, 펜틸, 헵틸, 노닐, 운데실, 도데실, 테트라데실, 헥사데실, 옥타데실, 시클로헥실, 시클로헥실메틸 또는 벤질기이다.
치환기 R3는 바람직하게는 수소원자, 메틸기, 에틸기 또는 이소부틸기이다.
R1, R2및 X에서 존재하는 비대칭탄소 원자들은 R-, S- 또는 R,S-배열을 가질 수 있다.
본 발명의 바람직한 화합물로는 R1이 메틸기이고, R2가 C3-C5기인 화합물, 특히 1-히드록시-3-(N-메틸-N-펜틸아미노)-프로판-1,1-디포스폰산 및 1-히드록시-3-(N-이소부틸-N-메틸아미노)-프로판-1,1-디포스폰산이다.
일반식(Ⅰ)의 화합물들은 다음과 같은 공지된 방법에 의해 제조된다.
Ⅰ. 일반식(Ⅰ)에서 Y가 수소원자를 나타내는 경우에, 화합물들을 바람직하게 다음과 같이 제조된다:
a) 다음 일반식(Ⅱ)의 화합물을 다음 일반식(Ⅲ)의 화합물과 반응시켜 다음 일반식(Ⅳ)의 디포스포네이트를 얻고, 얻어진 테트라에스테르를 임의로 비누화시켜 일반식(Ⅰ)의 디에스테르 또는 산을 얻거나; 또는 다음 일반식(Ⅴ)의 화합물은 모노-또는 디알킬화시키고, 얻어진 테트라에스테르를 비누화시켜서 일반식(Ⅰ)의 디에스테르 또는 산을 얻고, 이렇게 해서 제조된 화합물은 약리학적으로 허용될 수 있는 염으로 전환시킨다.
상기 식에서, R1, R2, R3및 X는 상기 정의한 바와 같고, B는 반응성 잔기, 예를 들어 할로겐 또는 술포네이트이고, R'는 C1-C4-알킬기, 바람직하게는 메틸, 에틸 또는 이소부틸이고, R4는 수소 또는 R2의 기이다.
Ⅱ. 일반식(Ⅰ)에서 Y가 알킬기에 의해 치환될 수 있는 아미노기인 경우에, 다음 일반식(Ⅵ)의 카르복실산 유도체를 다음 일반식(Ⅶ)의 인 화합물과 반응시키고, 계속해서 비누화시켜 일반식(Ⅰ)의 화합물을 얻고, 이렇게 해서 제조된 화합물을 약리학적으로 허용될 수 있는 염으로 전환시킨다.
상기 식에서, R1, R2및 X는 상기에 정의한 바와 같고, A는 저급알킬기에 의해 질소 원자위에 치환된 나트릴기, 아미노에테르기 또는 카르복사미드 기이고, T는 할로겐원자, OH 또는 OR1기이고, 여기서 R1은 상기에 정의한 바와 같다.
Ⅲ. 일반식(Ⅰ)의 Y가 OH인 경우에,
(a) 다음 일반식(Ⅷ)의 카르복실산을 아미산 또는 인산과 할로겐화 인의 혼합물과 반응시키고, 계속해서 비누화시켜 일반식(Ⅰ)의 유리 디포스폰산을 얻거나; 또는,
(b) 다음 일반식(Ⅸ)의 카르복실산 염화물을 다음 일반식(Ⅹ)의 아인산 트리알킬과 반응시켜 다음 일반식(XI)의 포스폰산 아실을 얻고, 계속해서 다음 일반식(XII)의 아인산 디알킬과 반응시켜 다음 일반식(XIII)의 디스포스폰산염을 얻고, 얻어진 테트라에스테르를 비누화시켜서 일반식(Ⅰ)의 디에스테르 또는 산을 얻거나; 또는
(c) 다음 일반식(XIV)의 화합물을 모노-또는 디알킬시키고 얻어진 테트라에스테르를 비누화시켜서 일반식(Ⅰ)의 디에스테르 또는 산을 얻고;
이렇게해서 제조된 화합물을 약리학적으로 허용될 수 있는 염으로 전환시킨다:

방법 Ⅰ a)의 경우에, 일반식(Ⅲ)의 메틸렌-디포스폰산 에스테르는 이것의 나트륨 또는 칼륨염의 형태로 사용된다. 이를 위해 상기 에스테르는 0 내지 40℃, 바람직하게는 25℃의 온도에서 벤젠, 톨루엔 또는 디메틸포름아미드와 같은 비활성용매중에서 나트륨, 칼륨 또는 적당한 수소화물과 반응한다. 단리없는 알칼리금속염은 적당한 할로겐화물 또는 술폰산염과 반응하고, 사용된 온도는 20 내지 110℃이다.
환원성 알킬화(방법 Ⅰ(b))의 경우에, 일반식(Ⅴ)의 1차 또는 2차 아민 및 카르보닐 화합물 또는 이것의 아세탈의 혼합물은 예를 들면 팔라듐-목탄 또는 니켈과 같은 수소화촉매의 존재하에, 대기압 또는 증가된 압력하에 수소로 처리되거나, 또는 환원제로서 포름산이 첨가된다. 계속하여, 일반식(Ⅴ)의 2차아민의 알킬화는 황산디알킬의 사용으로 상전이 공정에 따라 특히 유리하게 수행될 수 있다.
공정 Ⅱ의 경우에, 일반식(Ⅵ)의 니트릴은 110°내지 180°의 온도에서 아인산과 반응한다. 반응은 디글리콜 디메틸에테르 또는 디글리콜 디에틸에테르와 같은 비양자성 용매의 존재하에 또는 용매없이 수행될 수 있다. 그러나, 니트릴은 20 내지 80℃의 온도에서 임의로 물의 첨가와 더불어 디옥산 또는 테트라히드로푸란과 같은 비활성 용매중에서 삼브롬화인 또는 삼염화인과 같은 삼할로겐화인과 반응할 수 있다. 일반식(Ⅵ)의 아미노에테르는 바람직하게는 디에틸에테르, 디옥사 또는 벤젠과 같은 비활성 용매중에서 동몰량의 나트륨의 존재하에 아인산디알킬과 반응하며, 이 반응은 일반적으로 상응하는 용매의 환류온도에서 수행된다. 일반식(Ⅵ)의 산아미드는 할로겐화된 탄화수소 또는 디에틸에테르와같은 에테르와 같은 비활성 용매중에서, 오염화인/아인산 또는 염화옥살릴/아인산트리알킬의 혼합물과 반응할 수 있다.
방법 Ⅲ a)에 사용된 일반식(Ⅷ)의 카르복실산은 80 내지 130℃, 바람직하게는 100 내지 110℃의 온도에서 1 내지 2몰, 바람직하게는 1.5몰의 아인산 또는 인산 및 1 내지 2몰, 바람직하게는 1.5몰의 삼할로겐화인과 반응한다. 또한 반응은 할로겐화된 탄화수소, 특히 클로로벤젠 또는 테트라클로로에탄 또는 디옥산과 같은 희석제의 존재하에 수용될 수 있다. 물에 의해 그러나 유리하게는 반-농축 염산 또는 브롬화 수소산에 의해 후속 가수분해가 수행된다.
방법 Ⅲ b)의 경우에, 일반식(Ⅸ)의 산 염화물은 0 내지 60℃, 바람직하게는 20 내지 40℃의 온도에서 일반식(Ⅹ)의 아인산 트리알킬과 반응한다. 반응은 용매없이 또한 디에틸에테르, 테트라히드로푸란, 디옥산, 또는 염화메틸렌과 같은 할로겐화된 탄화수소의 존재하에 수용될 수 있다. 중간 생성물로서 생성된 일반식(XI)의 포스폰산아실은 단리되거나 또는 직접 반응할 수 있다. 0 내지 60℃, 바람직하게는 10 내지 30℃의 온도에서, 약염기, 바람직하게는 디부틸아민과 같은 2차아민의 존재하에 후속 반응이 수행된다.
상기 언급한 방법에서 삼할로겐화의 예로는, 삼염화인 또는 삼브롬화인이 있다.
방법 Ⅲ c)의 경우에, 방법 Ⅰ b)에 제시된 방법이 유사하게 적용된다.
방법 Ⅰ 및 Ⅲ에서 얻을 수 있는 테트라알킬 에스테르는 해당하는 디에스테르 또는 유리 테트라산으로 비누화될 수 있다. 통상적으로, 디에틸테르로의 비누화는 주변온도에서 아세톤과 같은 적당한 용매중에서 테트라알킬 에스테르를 알킬리금속 할로겐화물, 바람직하게는 요오드화나트륨으로 처리함으로써 수행된다.
바람직하게는, 산성이온교환기에 의해 디에스테르/2산으로 전환될 수 있는 대칭적인 디에스테르/2나트륨염이 얻어진다. 통상적으로 유리 디포스폰산으로의 비누화반응은 염산 또는 브롬화수소산과 함께 가열함으로써 수행된다. 그러나, 트리메틸실릴 할로겐화물, 바람직하게는 브롬화물 또는 요오드화물에 의한 분해가 수행될 수 있다. 한편, 유리 디포스폰산은 오르토포름산 알킬 에스테르와 함께 가열되어 테트라아알킬 에스테르로 다시 전환될 수 있다. 일반식(Ⅰ)의 유리 디포스폰산은 유리산으로서 또는 이것의 모노-또는 디알칼리금속염의 형태로 단리될 수 있다. 통상적으로 알킬리 금속염은 물/메탄올 또는 물/아세톤으로부터 재침전에 의해 정제될 수 있다.
약리학적으로 허용될 수 있는 염으로서는, 특히 탄산수소나트륨 또는 칼륨과 같은 무기 또는 유기염기, 수산화나트륨 수용액 또는 수산화 칼륨 수용액, 또는 트리메틸-또는 트리메틸-또는 트리에틸아민과 같은 수성아민 또는 암모니아로 화합물을 적정하여 통상적으로 제조되는 알칼리금속 또는 암모늄 염들이 사용된다.
본 발명에 따른 일반식(Ⅰ)의 신규화합물 및 그의 염들은 액체 또는 고체형태로 경구 또는 비경구적으로 투여될 수 있다. 이러한 목적을 위해, 정제, 캡슐, 당의정, 시럽, 용액, 현탁액등과 같은 모든 통상적인 투여형태가 사용될 수 있다. 주사용액의 경우에 주사매질로서, 예를 들면 안정제, 용해제 및 완충제와 같은 통상적인 첨가제를 함유하는 물을 사용하는 것이 바람직하다. 이러한 종류의 첨가제로는 예를 들면 타르타르산염 및 시트르산염 완충제, 에탄올, 착물형성제(에틸렌디아민-테트라아세트산 및 이것의 비독성염) 및 점도조절용 고분자량 중합체(액체 폴리산화 에틸렌)을 포함한다. 주 사용액용 액체담체 물질은 무균상태이어야 하고, 바람직하게는 앰푸울속에 담겨진다. 고체담체 물질의 예로는, 전분, 락토오스, 만니톨, 메틸 셀룰로오스, 탈크, 고분산규산, 고분자량 지방산(예:스테아르산), 젤라틴, 한천, 인산칼슘, 스테아르산 마그네슘, 동·식물 지방, 고체 고분자량 중합체(예:폴리에틸렌 글리콜)등이 포함되며, 경구투여용으로 적당한 조성물은, 바람직하게는 향미제 및 감미제를 함유할 수 있다.
투여량은 투여방식, 성별, 나이 및/또는 개별적인 조건과 같은 여러가지 인자들에 의존하여 변할 수 있다. 하루 투여량은 인체의 경우에 약 1 내지 1000mg, 바람직하게는 10 내지 200mg이며, 하루에 한번 또는 여러번에 나누어서 투여될 수 있다.
하기의 특정 실시예에 언급한 화합물 및 특허청구의 범위에 제시된 모든 의미의 조합에 의해 유도할 수 있는 화합물과는 별도로, 본 발명에 따른 바람직한 화합물들은 다음과 같은 디포스폰산염 및 그의 메틸 및 에틸에스테르이다:

다음 실시예들은 본 발명에 따른 화합물의 합성을 위해 사용할 수 있는 여러가지 방법중의 일부를 설명한다. 그러나, 실시예는 본 발명의 주제의 제한을 나타내지는 않는다. 이들 화합물의 구조는 H- 및 P-NMR 분광법으로 확인하였고, 순도는 P-NMR, 얇은 막 전기영동(셀룰로오스, pH 4.0의 옥살산염 완충제) 및 C, H, N, P 및 Na 원소분석에 의해 확인하였다. 각각의 화합물들의 특성에 대해, 피로인산염(Mrel=1.0)과 관련된 Mrel값(상대이동성)을 제시하였다.
[실시예 1]
1-히드록시-3-(N,N-디펜틸아미노)-프로판-1,1-디포스폰산
67ml의 클로로벤젠 중에서 7.1g의 아인산 및 14.8ml이 삼염화인과 함께 13.3g의 3-N,N-디펜틸아미노프로피온산을 100℃에서 20시간 유지시킨다. 그 다음 용매를 경사분리시키고, 잔류물을 180ml의 6N-염산과 함께 8시간동안 환류하에 교반시킨다. 불용성 물질을 여과제거시키고, 여과액을 농축시킨 후 앰버라이트 컬럼 IR 120(H+형태)에 가한다. 물로 용리시키고 전기영동을 모니터한다. 원하는 분획을 합치고, 증발시키고, 아세톤과 교반시키고, 얻어진 결정을 단리시킨다.

출발물질은 다음과 같은 얻는다:
톨루엔중에서 디펜틸아민과 아크릴산메틸 에스테르를 1:3의 몰비로 반응시킨다. 이론치의 28%의수득률로 유상의 디펜틸아미노프로피온산 에스테르를 얻고 1N 수산화나트륨 수용액으로 비누화시켜 이론치의 56%의 수득률로 원하는 산을 얻는다:융점 47-49℃

실시예 1에 제시한 것과 유사한 방법으로, 3-N-메틸-N-노닐아미노프로피온산으로부터 이론치의 10%의 수득률로 해당하는 디포스폰산염을 얻는다:융점 159℃, 소결, 178-184℃(Mrel=0.22)
출발물질은 다음과 같이 얻는다:
노닐아민을 벤조알데히드와 반응시켜 유상의 쉬프염기를 이론치의 96%의 수득률로 얻는다. 이를 팔라듐-목탄촉매로 수소화시켜 이론의 94%의 수득률로 유상으로서 N-벤질-N-노닐아민을 얻는다. 이로부터, 포름알데히드 및 포름산과 반응시켜 98%로 유상의 N-벤질-N-메틸-N-노닐아민을 얻는다. 팔라듐-목탄 촉매에 의한 벤질라이칼의 가수소분열로, 실시예 1에 기술된 바와 같이, 아크릴산메틸과 반응하고, 비누화되는 2차아민을 오일로서 정량적으로 얻는다. 유상에스테르의 수득률은 이론치의 81%이고,페이스트 상태의 산의 수득률은 이론치의 95%이다.
[실시예 3]
3-(N-시클로헥실-N-메틸아미노)-1-히드록시-1프로판-1,1-디포스폰산
톨루엔중에서 N-시클로헥실-N-메틸아민(시판용 제제) 및 아크릴산메틸로부터 제조된 15g의 3-N-시클로헥실-N-메틸아미노프로피온산(에스테르의 수득률 이론치의 76%, 융점 131-134℃, 산의 수득률 이론치의 92%, 융점 101-105℃)을 13.3g의 아인산과 함께 80℃로 가열한다. 용융물을 14.1ml의 삼염화인과 혼합하고, 같은 온도에서 16시간동안 유지시킨다. 그 다음 여기에 물 240ml을 첨가하고, 1일동안 100℃에서 교반시킨다. 그 다음 여과시키고, 진공으로 농축시키고, 얻어진 오일을 1ℓ의 아세톤에 따르고, 결정화를 개시시킨다. 얻어진 결정을 물에 용해시키고, 실시예 1에 제시한 방법으로 이온교환 크로마토그래피로 정제시킨다. 1수화물로서 수득량 4.5g(이론치의 16.9%):융점 142℃, 소결, 182℃(Mrel=0.3)

1g의 3-N-시클로헥실아미노프로판-1-히드록시-1,1-디포스폰산을 30ml의 염화메틸렌에 현탁시키고, 여기에 2.5ml의 진한 수산화나트륨 수용액을 가하고, 냉각하면서 1g의 황산수소 테트라부틸암모늄 및 0.3ml의 황산디메틸과 혼합시킨다. 혼합물을 주변온도에서 여러시간동안 교반시킨다. 통상 방법으로 처리한 후, 실시예 3에 따라 제조된 화합물과 함께 얻어진 생성물의 확인을 실릴화후의 질량 분광법으로 나타낸다.
사용된 디포스폰산은 다음과 같이 얻었다:피리딘중에서 시클로헥실아민을 아크릴산과 반응시킨다. 이론치의 70% 수득률로 3-N-시클로헥실아미노프로피온산(융점 170-171℃)을 얻는다. 아인산 및 삼염화인과 반응시켜서, 이론치의 31% 수득률로 디포스폰산(융점 164℃, 분해)을 얻는다.

3-(N-시클로헥실메틸-N-메틸아미노)-프로판-1-히드록시-1,1-디포스폰산
(3-N-시클로헥실메틸)-N-메틸아미노)-프로피온산(N-벤질-N-메틸아민으로부터, 팔라듐촉매에 의해 수소화(수득률 이론치의 70%:융점 60℃/160mmHg)시키고, 톨루엔중에서 아크릴산메틸과 반응시켜 이론치의 37%의 수득률로 메틸 3-(N-시클로헥실메틸-N-메틸아미노)-프로피온네이트을 얻고:수산화나트륨으로 비누화시켜 이론치의 63%의 수득률로 산(융점 98-102℃)을 얻음으로써 제조됨)을 실시예 3과 유사하게 아인산/3염화인과 반응시켜 이론치의 34%의 수득률로 디포스폰산을 얻는다:융점 180-194℃(분해):Mrel=0.31

4g의 4-아미노-1-히드록시-1,1-디포스폰산을 64ml의 1N 수산화나트륨 수용액중에 용해시키고, 3.3ml의 이소발레로알데히드와 혼합시키고, 2.5g의 팔라듐-목탄(10%)의 첨가후에, 5bar에서 수소화시킨다. 출발물질의 사라질때까지, 반응경로를 모니터한다.
앰버라이트 R 120(H+형태)으로 산성화시킨 후에 여과시키고, 결정화가 시작될때까지 증발시키고, 1.3g의 결정(20%)을 단리시킨다. m.p. 225-227℃, 분쇄(Mrel=0.39). 모액중에 잔류한 1-히드록시-4-(N-3-메틸부틸아미노)-부탄-1,1-디포스폰산이 중간 생성물로서 생성되며, 환전성 알킬화를 위해 다시 사용될 수 있다. 모액중에 남아있는 1-히드록시-4-(N-3-메틸부틸아미노)-부틸-1,1-디스폰산은 환원 알킬화를 위해 다시 사용할 수 있다.

유상 중간 생성물은 증류없이 직접 더 반응한다. 구조는 스펙트럼에 의해 확인하였다. 이온교환 크로마토그래피에 의해 최종생성물을 정제하였다.
단일 복용 독성 연구
급성 독성 연구를 마우스 및 쥐를 대상으로 수행하였다. 모든 연구에서 단일 복용 후에, 관찰 기간은 14일이었다.
경구 및 정맥내 연구로, 비포스폰산염의 부류에 대해 저수준의 경구생체 이용성이 확인되었다. 개략적인 LD50값을 하기의 표에 기재하였다.

마우스에서 급성 정맥내 독성을 8.7 내지 70mg/kg(체중)으로부터 시험하였다. 가장 높은 비-치사 용량인 17.4mg/kg까지, 동물은 임상적 증상을 나타내지 않았다. 더 높은 용량에서, 8 내지 14일 만에 치사가 일어났으며, 모든 동물은 거친 피부, 진정, 하수증, 및 70mg/kg에서 비틀림거림을 나타내고, 느린 운동 및 중단된 자세와 함께 뒷다리 마비를 나타내었다. 죽은 마우스는 폐수종, 기관 웅혈 및 장내 물 함유물을 갖는다.
쥐를 대상으로 급성 정맥내 독성을 8.7 내지 87mg/kg으로부터 시험하였다. 8.7 내지 17.4mg/kg에서는 임상적증상이 없었다. 더 높은 용량에서 동물은 거친 피부 및 진정을 나타내었으며, 55mg/kg에서는 간헐적 경련성 발작을 나타내었으며, 5분내에 사망하였다. 다른 모든 경우에, 사망기간은 2일 내지 7일로 나타났다. 사망한 동물에게서, 흉수증, 폐수종, 간 응혈, 및 흑색 또는 적색장내 물 함유물에 의한 장 팽창이 관찰되었다.
단일 경구 투여를 마우스를 대상으로 87 내지 1391mg/kg에서, 그리고 쥐를 대상으로 87 내지 870mg/kg에서 시험하였다. 마우스 696mg/kg이하에서 독성을 나타내지 않았으며, 쥐는 348mg/kg 이하에서 독성을 나타내지 않았다.
마우스 및 쥐에서 각각 1044 및 557mg/kg에서 사망이 시작되었으며, 투여 후 2 내지 7일 만에 사망이 일어난다. 임상적 표시는 주로 거친 피부 및 현저한 진정 및 정맥내 연구에서의 검시 결과와 유사한 검시결과이다.
