Abstract
본 발명은 휴지기의 공여체 세포 유래의 핵을 적당한 수용체 세포로 이식시키는 것을 포함하여, 동물의 배를 재구성하는 방법에 관한 것이다. 공여체 세포는 휴지기 세포로서, 즉, G1에서의 성장과 분열 사이클을 이탈하여, G0기에 정지된 것이다. 핵 이식은 세포 융합으로 수행될 수 있다. 그 다음, 재구성된 배를 사용하여 1마리 이상의 동물을 제조할 수 있다. 또한, 본 발명은 돌연변이 동물 및 유전자 이점이 많은 비돌연변이 동물의 제조 방법에도 유용하다.
Description
본 발명은 유전학적으로 선택되거나 또는 변형된 동물을 비롯한 동물의 생산에 관한 것이다.
공여체(供與體)의 배(胚)의 핵을 핵출된 난모 세포나 하나의 세포 수정란에 이식시켜 포유류의 배를 재구성하는 방법은 유전학적으로 동일한 개체의 생성을 가능하게 해준다. 이 방법은 연구용(즉, 생물학적 대조군)으로서, 그리고 산업용(즉, 유전학적으로 우수한 가축의 증식, 육제품의 균일성, 동물 관리)으로 유리하다. 그러나, 핵 공여체로서 초기배(初期胚)의 사용에 따른 한 가지 문제점은 단일배로부터 생산될 수 있는 자손의 수가 세포 수(사육 동물종의 경우에는 32 내지 64 세포 단계의 배(胚)가 가장 널리 사용됨) 및 핵이식 프로토콜의 효율성의 양자에 의하여 제한을 받는다는 것이다.
핵 공여체로서의 배의 사용과는 달리, 배양액 중에서 유지될 수 있는 세포로부터의 핵이식에 의한 살아있는 자손의 생산능은 동물 사육자들이 그 동안 추구하여 왔던 대상이다. 배양된 세포주로부터 클로닝된 자손을 작제하는 능력은 초기배의 사용에 비하여 다수의 이점을 제공하게 된다. 이들에는, 장기간에 걸친 다수의 동일한 자손의 작제(배양된 세포는 동결하여 보존할 수 있음) 및 배의 재구성 전에 요구되는 유전자형(예컨대, 성별)의 세포 모집단을 유전적으로 변형 및/또는 선별하는 능력이 포함된다. 이들 과정에서 사용하기 위한 하나의 가능한 세포형은 배의 간상부(ES) 세포이다. ES 세포는 마우스에서 분리되어 왔지만, 핵이식에 이들을 사용한 후에 출산 만기까지의 발생에 관한 보고는 없다. 현재로서는, 돼지의 ES 유사 세포를 생체내 생성된 포배의 포배강으로 주입한 다음, 발생까지 도달하였다는 유일한 보고가 있으나[Wheeler, Reprod . Fertil . Dev. 6 563-568(1994)], 다른 가축종들에서 관찰한 키메라성에 대한 보고 및 임의의 소정의 세포주로부터 임의의 포유류종 내에 핵이식 후 출산 만기까지의 발생 여부에 대한 보고는 전혀 없다.
ES 세포주의 사용에 관한 다른 양태가 몇 가지 있는데, 이 중의 하나는 핵이식용으로 사용시 발생을 촉진시킬 수 있는 다른 세포 모집단의 연구에 관한 것이다. 몇몇 보고서에서는 적당한 시험 세포로서 원시 배세포(Primordial Germ Cell)가 제공되었으나, 출산 만기까지 발생하는지에 대해서는 아직 보고된 바가 없었다. 초기배로부터 형성된 세포주가 제안된 바 있고, 연장 배양시 양(羊)에 있어서 초기 계대 세포로부터의 발생이 보고되어 있으나[Campbell et al., Therio 43 181(1995)], 통상적인 핵이식 프로토콜을 사용하는 발생을 얻지 못하였다[Campbell et al., J. Abstract Series (5) 31 (1995)].
핵이식 후 출산 만기까지의 발생을 달성하려면, 이식된 핵의 발생 시계(developmental clock)는 재조정되어야 한다. 이를 위하여는 전사가 멈춰진 후 발생 조절된 패턴으로 재개시되어야 한다. 종전의 보고들에 의하면, 포배기 단계까지의 발생은 소, 양, 돼지, 토끼 및 마우스 내의 광범위한 세포 타입으로부터 얻을 수 있다는 것을 나타낸 바 있다. 그러나, 이들 모든 보고에 있어서, 출산 만기까지의 발생에 대해서는 보고된 바가 없다. 9일령(令)의 양의 배의 배원반(ED)으로부터 형성된 초기 세포주(3 계대주까지 포함)로부터의 핵이식 후 정상적인 새끼 양의 출생이 이미 보고된 바 있다[Campbell et al., Therio 43 181(1995)]. 그러나, 후속 배양시 통상적인 핵이식 프로토콜을 사용하여 출산 만기까지의 발생은 달성되지 않았다(6 계대주 및 11 계대주)[Campbell et al., J. Reprod . Fertil . Abstract Series (5) 31 (1995)]. 이들 결과는 다양한 방식으로 해석될 수 있다. 첫째로, 배양의 초기 기간 동안에 얻은 ED 유래의 모든 세포는 발생을 촉진할 수 있을 것으로 예상할 수 있다. 그러나, 배양된 세포주의 안정화 동안의 연장 배양시, 이들 세포는 변화하며, 따라서 상기 문헌들에서 언급된 "유니버설 수용체" 내로 핵이식하기 위한 핵공여체로서 사용시 발생을 조절할 수 없다. 둘째로, 초기 배양 기간 동안 세포의 계대 집단이 발생 촉진능을 보유하고, 이것은 이들 초기 계대 동안에 핵이식 후 정상 태아의 발생을 설명하게 되리라고 예상된다. 종래의 연구들은 핵이식에 의해 재구성된 배의 발생에 있어서 공여체 핵과 수용체 세포질의 세포 주기 공조의 역할을 강조하여 왔다[Campbell et al., Biol . Reprod. 49 933-942(1993) 및 Biol. Reprod. 50 1385-1393(1994)].
공지된 핵이식 프로토콜을 사용하여 핵이식에 전능성(全能性)인 세포주의 분리에 의존하는 방법에 가능한 2 가지 전략적 대안은 다음과 같다.
(1) 기존의 핵이식 절차를 변화시키는 방법; 또는
(2) 핵이식 이전에 공여체 세포의 염색질을 변화시키는 방법.
전능성 세포는 동물 전체의 발생을 유도할 수 있다(공여체 세포로부터 수용체 세포, 예컨대 핵출된 난모 세포내에 핵이식에 의하여 배를 구성하는 경우, 공여체 세포의 핵이 전능성이다). 여기에는 배외 세포계, 즉 태반의 발생 유도도 포함된다. 이러한 정의에 있어서, 수정된 수정란과, 일부 종에 있어서, 각각의 난할구도 역시 전능성 세포이다. 이에 반하여, 다능성 또는 다잠재성 세포(즉, 배 간상부 세포)의 형은 난할강 내에 주입된 후 수태 산물/자손 내의 모든 조직을 형성할 수 있는 것으로 정의되어 왔다.
전술한 핵이식 방법 (1) 및 (2)의 양자에 있어서, 이식된 핵의 유전자 발현을 재프로그래밍할 수 있는 방법이 요구된다. 즉, 그러한 방법은 핵 공여체로서 분화 세포 또는 부분 분화 세포의 사용을 가능하게 하고, 따라서 이들의 고유한 전능성이 "발휘"되게 할 것이다.
본 발명은 휴지기 세포, 즉 세포 주기에 의하여 활동적으로 증식하지 않는 세포가 동물의 배를 재구성할 때 핵공여체로서 유리하게 사용될 수 있다는 발견에 따른 것이다. 이들 동물배는 출산 만기까지 발생하는 것이 가능하게 될 것이다. 배의 재구성 후에 관찰되고 효율적인 핵 이식에 필요한 공여체 핵의 변화는 이들 세포의 핵을 휴지기 상태로 되게 함으로써, 핵 공여체로서 사용하기 전에 상기 세포의 핵 내에서 유발시킬 수 있는 것으로 보인다. 이러한 사실을 본 발명에서 이용하기에 이르렀다.
본 발명의 제1 목적은 적합한 수용체 세포로 휴지기의 공여체 세포의 핵을 이식시키는 것을 포함하여, 동물의 배를 재구성하는 방법을 제공하는 것이다.
원칙적으로, 본 발명은 모든 동물, 예컨대 가금류와 같은 조류종, 양서류종 및 어류종에 적용할 수 있다. 그러나, 실제로는 산업적 이용 가능성이 현재로서는 가장 클 것으로 예상되는 것은 인간을 제외한 동물, 특히 포유 동물(인간 제외), 구체적으로 태반을 가진 포유 동물일 것이다. 또한, 유제(有蹄) 동물, 예컨대 소, 양, 염소, 물소, 낙타 및 돼지와 같이 특히 경제적으로 중요한 유제 동물을 동물의 클로닝 수단 및 돌연 변이 동물이나 유전자 변화된(genetically modified) 동물의 생성 수단으로서 본 발명을 적용하는 것이 가장 유용할 것으로 생각된다. 또한, 유념해야 할 점은 본 발명을 경제적으로 중요한 다른 동물종, 예를 들어 말, 라마 또는 설치류, 예를 들어 랫트 또는 마우스, 또는 토끼에도 사용할 수 있을 것이라는 점이다.
본 발명은 돌연 변이 동물 뿐만 아니라 비돌연 변이 동물의 작제에 동등하게 적용할 수 있다. 돌연 변이 동물은 유전자 변화된 공여체 세포로부터 작제될 수 있다. 전체적인 과정은 돌연 변이(즉, 유전자 변화된) 동물의 작제를 위한 통상적인 방법에 비해 하기와 같이 개략적으로 설명할 수 있는 다수의 이점이 있다.
(1) 더 적은 수의 수용체를 요한다는 점.
(2) 클론성 공여체 세포를 이용하여 다수의 동조(同調) 유전자형(syngeneic) 파운더(founder)를 생성시킬 수도 있다는 점.
(3) 유전자 표적화에 의한 미세한 유전자 변화가 허용된다는 점.
(4) 본 발명에 의해 얻은 배로부터 작제된 모든 동물은 각 동물이 단일핵으로부터 유도되기 때문에 배선(胚線)을 통해 관련된 유전자 변화를 전달하지만, 반대로 포배 주입이나 다른 과정에 의해 변화된 간상 세포군의 주입 후 키메라 현상(chimerism) 또는 전핵 주입에 의한 돌연 변이 동물의 작제는 모든 세포가 상기 변화를 포함하지 않고 그 결과 작제된 동물은 상기 배선을 통해 변화를 전달할 수 없는 일정 비율의 모자이크 동물을 발생시킨다는 점.
(5) 완전한 동물의 작제 전에 유전자의 변화[예를 들어, 통합(integration)] 부위에 대해 세포를 선택할 수 있다는 점.
동물과 관련하여 본 명세서에서 사용한 "돌연 변이(transgenic)"라는 용어는 많은 돌연 변이 동물이 다른 종의 유전자(들)를 함유하게 되지만, 동물의 배선 내에 그러한 유전자를 1개 이상 함유하는 동물만을 지칭하는 것으로 한정하여 이해하여서는 아니 된다. 오히려, 상기 용어는 배선이 재조합 DNA 기법에 의한 기술적 개입의 대상이 되어 왔던 모든 동물을 광범위하게 지칭하는 것이다. 예를 들어, 배선 내의 내인성 유전자가 결실, 중복, 활성화 또는 변화된 동물은 배선 내에 외인성 DNA 서열이 첨가된 동물 만큼 본 발명의 목적에 유용한 돌연 변이 동물이다.
동물이 돌연 변이인 본 발명의 구체예에 있어서, 공여체 핵은 유전자 변화되어 있다. 상기 공여체 핵은 1개 이상의 돌연 변이 유전자(transgene)를 함유할 수 있으며, 핵이식 및 배의 재구성 전에 유전자 변화를 일으킬 수 있다. 수정란의 웅성 또는 자성 전핵(前核) 내로의 주입과 유사 미세주입법이 유전자 변화의 한 가지 방법으로 사용될 수 있지만, 본 발명은 이 방법에만 제한되지 않는다. 즉, 대량 형질 전환법 또는 형질 감염법, 예를 들어 전기 침투법, 바이러스 형질 감염법 또는 리포펙션법(lipofection)을 사용할 수도 있다.
전술한 본 발명의 방법에 있어서, 핵은 휴지기의 공여체로부터 수용체 세포에 이식된다. 이 방법의 사용은 특정한 공여체 세포 타입에만 한정되는 것이 아니다. 이 기법을 이용하여, 예컨대 휴지기 상태로 도입되거나 생체내에서 휴지기 상태로 존재하도록 유도할 수 있는 배, 태아 및 성체의 체세포를 비롯한 모든 정상 핵형의 세포가 전능성이 있다는 것을 탐지할 수 있다. 따라서, 본 발명에는 완전 분화 세포를 비롯한 적어도 부분적으로 분화된 세포의 이용도 포함된다. 공여체 세포는 배양중에 있을 수 있으나, 반드시 그러하지 않아도 좋다. 이하에는, 배양된 소의 배양된 원시 섬유 아세포, 배 유래의 양의 세포주(TNT4), 6년령의 성숙한 양에서 얻은 양의 유방 상피 세포 유래의 세포주(OME), 태내 양 조직 유래의 섬유 아세포의 세포주(BLWF1) 및 9일령의 양의 배에서 유래된 상피 유사 세포주(SEC1)가 예시되어 있다. TNT4 세포주를 포함하는 본 발명에 유용한 배 유래의 세포주들도 역시 본 출원과 동시 계류 중인 PCT 특허 출원 번호 PCT/GB95/02095, 공개 번호 WO96/07732호에 자세히 기술되어 있다.
본 발명에 있어서 유용하기 위하여, 공여체 세포는 말하자면 휴지기 세포, 즉 유사 분열 세포 주기에 의하여 활동적으로 증식하지 않는 세포이다. 유사 분열 세포 주기는 뚜렷한 4 단계, G1, S, G2 및 M이 있다. 개시기라고 하는 세포 주기의 초기 상태는 G1 단계에서 일어나고 독특한 기능을 갖고 있다. 다른 세포 주기를 진행하는 결정이나 이행은 개시기에서 이루어진다. 세포는 일단 개시기로 진행되면, DNA 합성 전 단계인 G1 단계의 후반 단계까지 진행된다. 제2 단계인 S 단계는 DNA 합성이 일어나는 시기이다. 이 시기 다음에는 DNA 합성과 유사 분열 사이의 시기인 G2 단계가 온다. 유사 분열 자체는 M 단계에서 일어난다. 휴지기 세포(특정한 완전 분화 세포와 같이 천연적으로 휴지기인 세포 뿐만 아니라 휴지 상태가 유도된 세포를 포함)는 일반적으로 상기 세포 주기의 4 단계 중 어느 한 단계에 있는 것이 아닌 것으로 간주된다. 따라서, 이들이 세포 주기를 통해 정상적으로 진행하지 않는다는 것을 나타내기 위해 보통 G0 상태에 있는 것으로 표기한다. 휴지기의 G0 세포의 핵은 이배체 DNA 함량을 가진다.
배양된 세포는 화학 처리, 영양 결핍, 성장 억제 또는 유전자 발현의 조작을 비롯한 각종 방법에 의해 휴지 상태로 도입되도록 유도될 수 있다. 현재로서는, 양 및 소의 세포주에서 휴지 상태를 유도하는 방법으로서 배양배지 내의 혈청 농도를 감소시키는 방법이 성공적으로 이용되어 왔다. 이러한 상태에서, 세포는 G1 단계 중에 성장 주기를 이탈하여 전술한 바와 같이 소위 G0 단계에서 정지된다. 이 세포들은 성장 주기로 재도입시 재자극될 때까지 이 상태에서 수일간(세포에 따라 더 장기간도 가능) 유지될 수 있다. G0 단계에서 정지된 휴지기 세포는 이배체이다. G0 단계는 세포가 분화할 수 있는 세포 주기의 한 시점이다. 휴지 상태에서는 다수의 대사성 변화가 일어나는 것으로 보고되어 왔는데, 그 예로는 일인산화 히스톤, 섬모화 중심 소체, 모든 단백질 합성의 감소 또는 완전한 정지, 증가된 단백 분해 작용, 총세포 RNA를 감소시키는 RNA 전사 감소 및 회전율(turnover) 증가, 폴리리보솜의 해리, 불활성 80S 리보솜의 축적 및 염색질 응축이 있다(문헌[Whitfield et al., Control of Animal Cell Proliferation, 1 331-365(1985)] 참조).
이러한 특징 중 다수는 핵출된 난모 세포로의 핵이식 후에 일어나야 하는 것들이다. G0 상태가 세포 분화와 관련이 있다는 사실은 이것이 수용체 세포의 세포질에 의하여 재모델링 및/또는 재프로그래밍되는 것이 보다 용이한 핵/염색질 구조를 제공할 수 있다는 것을 암시한다. 이러한 방식에 있어서, 핵 공여체 세포가 휴지기 상태에 있는 덕분에 공여체 핵의 염색질을, 그 핵이 발생을 유도할 수 있도록 배의 재구성 전에 변화시킬 수 있다. 이 방법은 핵 공여체로서 상기 공여체 세포를 사용하기 전에 그 세포의 염색질이 변화된다는 점에서 종래에 보고된 모든 핵 이식 방법과는 다른 것이다.
공여체 세포 유래의 핵이 이식되는 수용체 세포는 난모 세포 또는 다른 적절한 세포일 수 있다.
수용체 세포는 각종의 여러 발생 단계에 있는 세포로서 중기 I 내지 중기 II의 난모 세포에서부터 수정란에 이르는 세포 및 2-세포 배가 사용될 수 있다. 각 세포에는 장단점이 있다. 수정란을 사용하면 효율적인 활성화가 보장되지만, 난모 세포를 사용하면 단성 생식 활성화를 요한다(하기 참조). 일부 종에서는 난할 단계의 배를 사용하는 것이 유리할 수 있는 다른 기작은 유전자 발현의 재프로그래밍을 요구하는 정도이다. 전사는 마우스에 있어서 제2 세포 주기 동안에 개시되며, 포배 단계까지 2차원 전기 영동법에 의하여 합성된 단백질 속성의 주요 변화는 나타나지 않는다[Howlett & Bolton J. Embryol . Exp . Morphol . 87 175-206(1985)]. 그러나, 대부분의 경우, 수용체 세포는 난모 세포일 것이다.
수용체는 핵출성인 것이 바람직하다. 핵이식 과정에서 수용체 난모 세포의 핵출은 필수적인 것으로 통념화되어 왔지만, 이러한 판단이 실험적으로 확인된 바는 없다. 유제 동물에 대해 설명된 최초의 방법은 세포를 1/2씩으로 분할하는 단계를 포함하는데, 이들 중 하나가 핵출될 가능성이 높았다[참조: Willadsen Nature 320 (6) 63-65 (1986)]. 이 방법은 다른 미지의 1/2 세포가 여전히 중기 기구(metaphase apparatus)를 가지며, 세포질 부피의 감소로 새로운 배의 분화 패턴을 가속화시킬 수 있다는 단점이 있다[참조: Eviskov 등, Development 109 322-328 (1990)].
최근에 이르러, 최소의 세포질과 함께 염색체를 제거하기 위한 시도에 상이한 방법들이 이용되어 왔다. 제1 극체 및 주변 세포질의 흡출은 양의 난모 세포의 67%에 있어서 중기 II 기구를 제거하는 것으로 확인되었다[참조: Smith & Wilmut Biol. Reprod . 40 1027-1035 (1989)]. 단지 DNA 특이성 플루오로크롬(Hoechst 33342)을 사용하는 방법만이 제공되었는데, 이에 의하면 세포질 부피의 감소를 최소화하면서 핵출이 보장되게 된다[참조: Tsunoda 등, J. Reprod . Fertil. 82 173 (1988)]. 이 방법은 가축종에서 현재 통상 사용되고 있는 방법이다[참조: Prather & First J. Reprod . Fertil . Suppl. 41 125 (1990), Westhusin 등, Biol . Reprod . (Suppl.) 42 176 (1990)].
포유류에 있어서 핵출을 위한 비침투성 기법에 대한 보고는 거의 없지만, 양서류에 있어서는 자외선을 사용하는 조사법이 일반적으로 사용되고 있다[참조: Gurdon Q. J. Microsc . Soc. 101 299-311 (1960)]. 포유류에 있어서의 상기 기법의 사용에 대한 상세한 보고는 없지만, DNA 특이성 플루오로크롬의 이용 동안 마우스 난모 세포의 자외선에 대한 30초 이상 동안의 노출은 상기 세포의 발생 가능성을 감소시킨다는 점이 주목되었다[참조: Tsunoda 등, J. Reprod . Fertil. 82 173 (1988)]. 공여체 세포핵이 이식되는 수용체 숙주 세포는 핵출된 중기 II 난모 세포, 핵출된 불활성화 난모 세포 또는 핵출된 예비 활성화 난모 세포인 것이 바람직하다. 수용체가 적어도 핵출된 중기 II 난모 세포인 경우에는, 활성화는 이식시에 일어날 수 있다. 또는, 수용체가 적어도 핵출된 불활성화 중기 II 난모 세포인 경우에는 활성화는 뒤따라 일어날 수 있다. 전술한 바와 같이, 핵출은 실제로 핵이나, 전핵, 또는 중기판(수용체 세포에 따라 결정)을 제거하여 물리적으로 수행하거나, 또는 예컨대 자외선을 조사하거나 다른 핵출화 영향을 가하여 기능적으로 수행할 수 있다.
적합한 사이토플라스트(핵출된 난모 세포) 수용체에는 다음과 같은 3 가지가 있다.
1. 본 출원과 동일자 출원되고 동시 계류 중인 PCT 특허 출원 PCT/GB96/02098 (우선권 번호 GB9517779.6)에 기술된 "MAGIC 수용체"(중기 정지된 G1/G0 허용성 사이토플라스트).
2. "GOAT"(G0/G1 활성화 및 이식) - 활성화시의 MII(중기 II) 난모 세포[Campbell et al., Biol . Reprod. 49 933-942(1993)].
3. "유니버설 수용체"[Campbell et al., Biol. Reprod. 649 933-942(1993), Biol. Reprod. 50 1385-1393(1994)].
상기 3 가지 수용체는 모두 재구성된 배에서 G0의 공여체 핵을 사용했을 때 정상 배수체를 생성할 것이다. 그러나, 최근의 보고들은 휴지기 세포 유래의 핵 중에서 일부의 핵은 핵의 외피를 분리시키지 않고 S 단계 세포질에 도입시 DNA 합성 단계로 진행될 수 없다고 주장하여 왔다[Leno & Munshi, J. Cell Biol. 127(1) 5-14(1994)]. 따라서, 일부의 배는 "유니버설 수용체(universal recipient)"를 이용하여 발생시킬 수 있지만, 방법 1 또는 2 중의 어느 한 가지 방법에 따라서 고농도의 MPF (M 단계 촉진 인자 또는 성숙 촉진 인자)를 함유하는 MII 난모 세포를 사이토플라스트 수용체로서 사용함으로써 발생 빈도수를 증가시킬 수 있을 것으로 예상된다.
일단 적합한 공여체 및 수용체 세포가 확인되면, 공여체 세포의 핵을 수용체 세포로 이식시켜야 한다. 융합법으로 핵이식을 실시하는 것이 가장 편리하다.
융합을 일으키는 데 사용되어온 3 가지 확립된 방법은 다음과 같다.
(1) 융합 촉진 화합물, 예컨대 폴리에틸렌 글리콜에 세포를 노출시키는 방법.
(2) 센다이 바이러스(Sendai virus) 등과 같은 불활성화 바이러스를 사용하는 방법.
(3) 전기 자극을 이용하는 방법.
폴리에틸렌 글리콜이나 기타 글리콜류와 같은 융합 촉진 화합물에 세포를 노출시키는 방법은 체세포 융합의 경우에는 일반적인 방법이나, 배의 경우에는 널리 사용되지 않는 방법이다. 이것은 폴리에틸렌글리콜이 독성이므로 세포를 최소 시간 동안 노출시켜야 할 필요성이 있고, 또 신속하게 화합물을 제거해야할 필요성이 투명대의 제거를 불가피하게 할 수도 있다[Kanka et al., Mol . Reprod. Dev. 29 110-116(1991)]. 마우스 배를 사용한 실험에 있어서, 불활성화 센다이 바이러스는 난할 단계의 배로부터 얻은 세포의 융합에 효율적인 수단을 제공하는데[Graham Wistar Inst. Symp . Monogr. 9 19(1969)], 활성화가 유도되지 않는다는 부가적인 실험상의 이점도 있다. 유제 동물에 있어서는, 일반적으로 단위 생식 활성화를 유도하는 데 사용된 것과 동일한 전기 충격에 의해 융합을 실시한다[참조: Willadsen Nature 320 (6) 63-65 (1986), Prather 등, Biol . Reprod. 37 859-866 (1987)]. 이들 종에 있어서, 센다이 바이러스는 대부분 융합을 유도하지만, 통상적으로 적용할 만큼의 충분한 신뢰성은 없다[참조: Willadsen Nature 320 (6) 63-65 (1986)].
세포-세포 융합은 핵이식의 양호한 방법이지만, 사용할 수 있는 유일한 방법은 아니다. 기타 적합한 기법으로는 미세주입법(microinjection)을 들 수 있다[참조: Ritchie 및 Campbell, J. Reproduction and Fertility Abstract Series No. 15, p60].
핵이식 전 또는 (바람직하게는) 핵 이식 후(또는, 어떤 경우에는 적어도 핵이식과 동시에), 일반적으로 적어도 세포가 난모 세포인 경우에는, 수용체 세포를 단성생식 활성화에 의하여 발생을 자극할 필요가 있다. 최근의 실험에 따르면, 단위 생식 활성화에 대한 필요 조건은 예상하였던 것보다 훨씬 복잡한 것으로 밝혀졌다. 활성화는 과단성 있는(all-or-none) 현상이며, 전핵의 형성을 유도할 수 있는 다수의 처리들이 모두 "활성화"를 유발하는 것으로 추정되어 왔다. 그러나, 토끼의 난모 세포를 전기 펄스에 반복 노출시키면 일련의 적절한 펄스(pulse)의 선택과 Ca2+의 조절만이 이배체화 난모 세포의 발생을 임신 중기까지 촉진시킬 수 있다는 것이 밝혀지게 되었다[Ozil Development 109 117-127(1990)]. 수정 동안에는 세포내 칼슘 농도의 반복적이며 과도적인 증가가 나타나고[Cutbertson & Cobbold Nature 316 541-542(1985)], 전기 펄스는 칼슘 농도의 유사한 증가를 일으키는 것으로 생각된다. 칼슘의 과도성의 패턴은 종마다 다르며, 전기 펄스의 최적 패턴도 유사한 방식으로 달라질 것이라는 증거가 있다. 토끼에 있어서 펄스간의 간격은 약 4분이고[Ozil Development 109 117-127(1990)], 마우스에 있어서는 10 내지 20분인 반면에[Cutbertson & Cobbold Nature 316 541-542(1985)], 소에 대하여 예비 관찰한 결과는 약 20 내지 30분이었다[Robl et al., in Symposium on Cloning Mammals by Nuclear Transplantation (Seidel ed.), Colorado State University, 24-27(1992)]. 대부분의 발표된 실험에 있어서, 단일 전기 펄스에 의하여 활성화가 유도되었지만, 수 차례의 펄스에 노출시키면 재구성된 배의 발생 비율이 증가한다는 새로운 관찰 결과가 제시되었다[Collas & Robl Biol . Reprod. 43 877-884(1990)]. 따라서, 펄스의 수효, 전기장 강도 및 펄스의 지속 시간과 매질의 칼슘 농도를 최적으로 하기 위하여 각 경우마다 통상적인 조정을 행할 수 있다.
본 발명의 제2 목적은 앞에서 설명한 방법으로 생산한 재구성된 동물의 배를 제공하는 것이다.
본 발명의 제3 목적은 동물을 작제하는 방법을 제공하는 것인데, 이 방법은
(a) 전술한 바와 같이 동물의 배를 재구성하는 단계
(b) 동물을 상기 배로부터 출산 만기까지 발생시키는 단계, 및
(c) 필요에 따라, 이와 같이 형성된 동물로부터 사육하는 단계
를 포함한다.
상기 단계 (a)는 앞에서 상세히 설명하였다.
이러한 목적의 본 발명의 방법에 있어서 제2 단계인 단계 (b)는 동물을 배로부터 출산 만기까지 발생시키는 단계이다. 이는 직접 또는 간접적으로 수행할 수 있다. 직접 발생시, 상기 단계 (a)로부터의 재구성된 배는 발생이 일어나는 데 필요한 것 이상의 추가 조치없이도 간단히 발생된다. 그러나, 간접 발생시, 상기 배는 완전한 발생이 일어나기 이전에 추가 조작될 수 있다. 예를 들어, 수율을 증가시키기 위해서 상기 배를 난할하고, 상기 세포를 클론 단위로 증식시킬 수 있다.
별법으로 또는 추가적으로, 공여체의 클론 단위 증식에 의한 본 발명의 수단에 의하여 및/또는 그 사용이 일련의 (핵)이식 과정으로 이루어지는 경우, 생존 가능한 배의 수율 증가가 달성될 수 있다. 현재로서 포배 형성의 성공률에 있어서의 제약은 대부분의 배가 "재프로그램"되지 않는다는 사실에 기인할 수 있다(허용할 수 있는 수치가 재프로그램되기는 하지만). 경우가 그러하다면, 이 때 그 성공률은 다음과 같이 증가시킬 수 있다. 그 자체가 발생하는 각각의 배는, 예컨대 상실배(桑實胚) 단계나 32 내지 64 세포 단계에서 핵 공여체로 사용될 수 있으며, 별법으로서는, 내부 세포괴 세포를 포배기에서 핵 공여체로 사용할 수 있다. 이들 후속 이식으로 유래되는 배는 그 자체를 유력한 핵 공여체로서 사용함으로써 역시 효율을 더욱 증가시킬 수 있게 된다. 이들 배가 사실상 유전자 발현을 재프로그램한 배를 반영하고, 이들 핵이 실제로 재프로그램되는 경우(가능할 것으로 보임), 발생되는 각각의 배는 이 방식으로 상기 핵 이식 과정의 효율에 따라 증식될 수 있다. 달성될 가능성이 있는 증가도는 세포 타입에 의존한다. 양에 있어서, 16 세포기의 배로부터 미리 활성화된 "유니버설 수용체" 난모 세포에 하나의 할구를 이식하여 55% 포배기 배를 얻는 것은 쉽게 가능한 일이다. 따라서, 단일 세포에서 발생된 각각의 배가 16 세포기에서 8개의 세포로 발생할 수 있다는 가설은 타당성이 있다. 이러한 수치는 단지 개략적인 지침에 불과하지만, 후의 발생 단계에서의 유리한 점의 정도는 그 단계에서의 상기 과정의 효율에 의해 결정된다는 것은 명백하다.
또한, 핵 공여체 세포원으로 작용하기 위한 새로운 세포주가 전술한 바에 따라 형성된 배 또는 이로부터 생성된 자손이나 성체로부터 작제될 수 있다는 것도 역시 기대된다.
어떤 경우에는, 재구성된 배를 출산 만기까지 발생시키는 데 제약이 있게 되면, 천연적으로 형성된 배와 핵이식에 의해 재구성된 배로부터 유래된 세포로부터 형성된 키메라 동물을 생성시키는 것이 좋을 수도 있다. 이러한 키메라는 일정 비율의 천연 배 세포와, 포배 단계 이하의 임의의 단계에 있는 일정 비율의 재구성된 배세포를 취하고, 응집이나 주입에 의해 새로운 배를 형성함으로써 형성시킬 수 있다. 세포 비율은 50:50 또는 출산 만기까지 발생하는 배가 형성되기에 적합한 다른 비율을 사용할 수 있다. 이러한 경우에 정상 세포의 존재는 재구성된 배를 구하고 출산 만기까지의 발생과 출산이 성공적으로 이루어지도록 보조한다고 생각된다.
수율 개선의 수단에 대한 문제는 차치하고, 상기 재구성된 배는 생체 내에서 또는 시험관 내에서 포배로 배양시킬 수 있다.
핵 이식에 의해 유도된 배는 정상적인 배와는 상이하며, 때로는 배가 통상적으로 배양되는 배양 조건(적어도 생체내) 이외의 생체내 배양 조건으로부터 이득을 얻거나 또는 그러한 배양 조건을 요하기까지도 한다는 사실을 경험적으로 알게 되었다. 이에 대한 이유는 아직 알려져 있지 않다. 소의 배를 통상적으로 증식함에 있어서, 재구성된 배(일시에 이들의 다수)는 양의 수란관에서 5 내지 6일 동안 배양된 바 있다[참조: Willadsen, In Mammalian Egg Transfer(Adams, E.E., ed.) 185 CRC Press, Boca Raton, Florida (1982)]. 그러나, 본 발명을 실시함에 있어서, 배를 보호하기 위하여, 임시 수용체로부터 회수한 후 상기 배를 이식 전에 한천과 같은 보호 배지 내에 매립하고, 이어서 상기 한천으로부터 절개해내는 것이 좋다. 보호 한천 또는 기타 배지의 기능은 2 가지이다. 즉, 첫째로 투명대를 지탱함으로써 배를 위한 구조 보조제로서 작용하는 것이고, 둘째로 수용체 동물의 면역계 세포들에 대한 차단체로서 작용하는 것이다. 이러한 접근 방식은 포배를 형성하는 배의 비율을 증가시키지만, 다수의 배가 상실될 수 있다는 단점이 있다.
시험관내 조건이 사용되는 경우, 당업계에서 통상 사용되는 조건들이 다소 사용될 수 있다.
포배기 단계에서는, 출산 만기까지의 발생에 적합한지에 대하여 배를 선별할 수 있다. 통상, 이것은 상기 배가 돌연 변이이고, 구성 요소에 대한 선별 및 선발이 수행되어 있는 경우에 행하게 된다. 또한, 비돌연 변이성 유전자 마커에 대한 선별도 역시 이 단계에서 수행할 수 있다. 그러나, 본 발명의 방법에 의하면 초기 단계에서 공여체의 선별이 가능하게 되는데, 이는 일반적으로 바람직하다.
선별이 수행된 경우, 그 선별 후, 포배기 단계의 배는 출산 만기까지 발생할 수 있다. 이는 일반적으로 생체내에서 일어나게 된다. 포배까지의 발생이 시험관 내에서 수행된 경우에는, 이 단계에서 최종 수용체 동물에 대한 이식이 실시된다. 포배 발생이 생체내에서 이루어진 경우에는, 원칙적으로 포배를 포배 전 숙주내에서 출산 만기까지 발생시킬 수 있지만, 실제로는 상기 포배를 통상 포배 전 (임시적) 수용체로부터 분리하고, 보호 배지로부터 절제해 낸 후, 포배 후(영구적인) 수용체에 이식하게 된다.
본 발명의 선택적인 단계 (c)에서, 동물은 선행 단계에 의해 작제된 동물로부터 사육된다. 이 방식으로, 하나의 동물을 사용하여 목적하는 유전적 특성(들)을 보유하는 동물군 또는 동물집단을 형성시킬 수 있다.
유전학적으로 동일한 세포원으로부터 핵이식에 의해 작제된 동물은 동일한 핵을 공유하지만, 이들 동물은 상이한 난모 세포로부터 유래되기 때문에 엄격한 의미로는 동일하지 않은 것이다. 이러한 상이한 기원의 중요성은 명백하지 않으나, 상업적인 특성에 영향을 미칠 수 있다. 아이오와 주립 대학 사육장(Iowa State University Breeding Herd)에서 행한 젖소의 미토콘드리아 DNA에 대한 최근의 분석에 따르면, 이 DNA가 우유 및 증식능과 관련이 있는 것으로 확인되었다[참조: Freeman & Beitz, In Symposium on Cloning Mammals by Nuclear Transplantation(Seidel, G. E. Jr., ed.) 17-20, Colorado State University, Colorado (1992)]. 유사한 효과가 상기 젖소군 전체에 걸쳐 존재하는 지가 확인되어야 하고, 또 난모 세포의 선발을 고려하는 것이 특정 상황에서 가능하거나 또는 필요한 지의 여부도 고려하여야 하는 숙제이다. 소의 사육 분야에 있어서, 많은 유전학적 장점이 있는 공여체로부터 다수의 배를 생산하기 위한 능력은 유전학적 개량을 전국적으로 확산시킴에 있어서 상당한 잠재적 가치가 있을 수 있다. 적용 규모는 각 배의 비용 및 이식된 배가 출산 만기까지 발생할 수 있는 비율에 따라 달라질 것이다.
예시 및 요약에 의하여, 돌연 변이 및 비돌연 변이 동물을 생산할 수 있는 전형적인 방법을 이하에 기재한다. 이 방법은 7 개의 단계를 포함하는 것으로 간주된다.
(1) 핵형의 평가, 휴지기의 유도(G0 단계에서 정지) 및/또는 발생 유도를 포함할 수 있는 적당한 공여체 세포를 선발 및 분리하는 단계.
(2) 유전자 변화된 세포 모집단을 생산하기 위한 적당한 분자 생물학적 기법의 적용 단계. 이러한 기법으로는 유전자 삽입, 유전자 녹-아웃(knock-out), 유전자 녹-인(knock-in) 및 기타 다른 유전자 변화를 포함할 수 있다. 또한, 필요한 경우에는 적당한 마커가 있거나 없이 적당한 구성물을 사용하는 형질감염에 의한 돌연 변이법을 채용하여도 좋다.
(3) 선택적으로, 필요한 유전자형/표현형(즉, 안정한 구성요소)에 대하여 변화시킨 세포 모집단이나 클론을 선별 및 선발하는 단계.
(4) 변화된 세포 모집단을 휴지기에 유도하는 단계.
(5) 핵이식에 의해 배를 재구성하는 단계.
(6) 생체내 또는 시험관내에서 포배까지 배양하는 단계.
(6a) 필요에 따라, 안정한 구성 요소[(3)에서 수행된 경우에는 생략)] 또는 기타 목적하는 특성에 대해 선별 및 선발하는 단계.
(7) 필요에 따라, 최종 수용체에 이식하는 단계.
본 발명의 제4 목적에 따르면, 본 발명은 상기한 바와 같이 작제된 동물을 제공한다.
본 발명의 각 목적에 대하여 양호한 특징은 각각 다른 목적에도 마찬가지이고,그 반대도 또한 같다.
예시의 목적으로 제공되고 본 발명을 한정하지 아니하는 하기 실시예를 참고로 하여 본 발명을 설명하겠다.
실시예
1: 공여체 세포의 휴지기 유도
배양된 세포주에 있어서 휴지기를 유도하는 방법에는, 접촉 억제 또는 혈청 결핍을 비롯한 다양한 방법이 제시되어 왔다[참조: Whitfield et al., Control of Animal Cell Proliferation, 1 331-365(1985)]. 휴지기 유도 방법은 중요한 것으로 간주되지는 않지만, 중요한 단계는 세포를 성장 주기로부터 이탈시켜 이배체 DNA 함량을 가진 G0 상태에 정지시키고 생육력을 유지시키는 것이다. 실시예 3과 4에서는, 혈청 결핍시킨 소의 원시 섬유 아세포, 생체내에서 생성된 7일된 포배의 내부 세포괴로부터 안정화된 소의 세포주 및 배에서 유래된 양의 세포주(TNT4)를 사용하여 휴지기를 유도하고 이들 세포를 세포 주기의 G0 단계에서 정지시켰다. 혈청 결핍법을 실시예 5에 기재된 공여체 세포의 휴지기 유도 방법에 유사하게 사용하였다.

난모
세포의 분리 및
핵이식
난모 세포는 (i) 도살장에서 얻은 재료를 시험관내 성숙시키거나 경질성 소포 천공으로부터 얻거나; (ii) 생체내 성숙시킨 다음 외과적으로 회수하거나, 또는 (iii) 기타의 적절한 방법으로 얻을 수 있다. 모든 생체내 성숙된 난모 세포는 1.0% 태내 송아지 혈청(FCS)을 함유하는 무칼슘 및 무마그네슘 인산염 완충 식염수(PBS) 내에서 수란관을 세척함으로써 수거할 수 있다. 시험관내 성숙된 난모 세포는 수거하여 1.0% FCS를 함유하는 무칼슘 M2[Whittingham and Wales Aust . J. Biol. Sci. 22 1065-1068(1969)]에 옮긴다. 난모 세포는 소구 세포를 표피 박리시킨 다음 모든 절차에 무칼슘 매질을 사용하는 것을 제외하고는 앞서 설명한 바와 같이 하여 핵출시킨 것이다[(Campbell et al., Biol . Reprod. 49 933-942(1993) 및 Biol. Reprod. 50 1385-1393(1994)]. 융합 절차는 전술한 방법[(Campbell et al., 1993, 1994 (상기 인용 문헌 중)]의 변형법으로서 하기 관련 부분에 기술된 바와 같으며, 별법으로서는 공여체 세포를 핵출된 난모 세포에 주입하여 핵을 도입시킬 수도 있다[(Ritchie & Campbell, J. Reprod . Fertil . Abstract Series (5) 60 (1995)]. 이러한 절차들의 시기는 종마다 다르며, 이하에는 생체내에서 성숙시킨 양의 난모 세포와 시험관내에서 성숙시킨 소의 난모 세포를 이용한 2 가지 프로토콜을 요약한다.

3: 양에 대한
핵이식법
3.1 공여체
암양의
과잉 자극과
난모
세포의 회수
스코틀랜드 블랙페이스 종(Scottish Blackface) 암양들을 14일간 프로제스타젠 스폰지로 동조화하고(VeramixTM, Upjohn, UK), 연속 2일간 양의 소포 자극 호르몬(FSH)(OvagenTM, Immuno-chemical Products Ltd, New Zealand)을 3.0 mg/일씩 단일 주입(총 6.0 mg)하여 과잉 배란하도록 유도하였다. 배란은 FSH를 2차 주입한 후 24 시간 만에 8 ㎎의 고나도트로핀 방출 호르몬 유사물(GnRH ReceptalTM, Hoechst, UK)을 단일 투여량으로 투여하여 유도하였다.
수정되지 않은 중기 II 난모 세포는 사용 전까지 37℃에서 유지시킨 1.0% 태내 송아지 혈청(FCS)을 함유하는 둘베코 인산염 완충 식염수를 사용하여 세척함으로써 상기 GnRH의 주입 후 24 내지 29 시간만에 수란관으로부터 회수하였다.

난모
세포 조작
회수된 난모 세포는 37℃에서 유지시키고, PBS 1.0% FCS로 세척하여, 37℃에서 10% 태내 송아지 혈청(FCS)을 함유하는 칼슘이 제거된 M2 배지로 옮겼다. 염색체를 분리해내기 위하여(핵출), 난모 세포를 10% FCS, 7.5 ㎍/㎖의 사이토칼라신 B(Sigma) 및 5.0 ㎍/㎖ 획스트(Hoechst) 33342(Sigma)를 함유하는 무칼슘 M2 중에 넣고 37℃에서 20분 동안 방치하였다. 그 다음, 제1 극체의 바로 아래로부터 소량의 세포질을 20 μM의 유리 피펫을 사용하여 흡인·분리하였다. 이와 같이 흡인된 세포질을 자외선에 노출시켜 핵출화를 확인하고 중기판의 존재를 조사하였다.
3.3 배의 재구성
10개 내지 20개의 난모 세포군을 핵출하고, 광유(SIGMA) 존재하에 5% CO2, 37℃에서 무칼슘 M2 배지 20 ㎕ 적가물에 첨가하였다. 배를 재구성하기 위하여 다음과 같은 3 가지 프로토콜(a), (b) 및 (c)를 각각 사용하였다.

3.5 배의 배양 및 평가 (모든 군)
상기 배양 기간 후에, 융합된 결합체를 PBS 중의 1% 및 1.2% 한천(DIFCO) 중에 이중 매립하고, 비동조화 암양의 결찰된 수란관에 이식시켰다. 상기 결합체를 한천에 매립시켜 수용체인 암양에 의한 배의 면역 거부 반응을 방지 또는 감소시키고, 그 결합체가 함께 보유되도록 보조하였다. 6일 후, 수용체인 암양을 죽이고, PBS 10% FCS를 사용하여 수란관으로부터 배를 세척하여 회수하였다. 2개의 바늘을 사용하여 한천 조각으로부터 배를 절제해내고 발생을 현미경으로 평가하였다. 상실배/포배 단계까지 발생된 모든 배를 가능한 한 즉시 동조화한 최종 수용체인 암양의 자궁각에 이식시켰다.
또한, 배를 포배 단계까지 발생시키는 데에는 임시 수용체인 암양 대신에 시험관내 기법을 사용하는 것이 적합할 수도 있다.

성숙화
난소는 지방 도살장에서 입수하여 실험실로 운반해오는 동안 28 내지 32℃로 유지시켰다. 소구 난모 세포 복합체(COC)는 피하 주사 바늘(내경 1.2 ㎜)을 사용하여 직경이 3 내지 10 ㎜인 소포로부터 흡인하여 멸균 플라스틱 유니버설 용기에 주입하였다. 이 유니버설 용기를 가온실(35℃)에 방치하고, 10 내지 15분 동안 소포 물질을 침전시킨 다음 상징액의 3/4을 버렸다. 나머지 소포 물질을 10% 소 혈청이 보강된 해부용 배지[어얼스 염(Gibco), 75.0 ㎎/ℓ의 카나마이신, 30.0 mM 헤페스를 함유한 TCM 199, pH 7.4, 몰삼투압 농도 280 mOsmol/H2O kg] 등부피로 희석하여, 85 ㎜ 페트리 접시에 옮긴 다음, 해부 현미경하에서 COC의 존재를 조사하였다. 적어도 2 내지 3층의 치밀한 소구 세포층을 가진 복합체를 선발하고, 해부용 배지로 3회 세척한 다음, 10% 소 혈청과 1 x 106 과립막 세포/㎖가 보강된 성숙 배지[어얼스 염(Gibco), 75 ㎎/ℓ의 카나마이신, 30.0 mM 헤페스, 7.69 mM NaHCO3를 함유한 TC 배지 199, pH 7.8, 몰삼투압 농도 280 mOsmol/H2O kg]로 옮기고, 필요할 때까지 공기 중의 5% CO2 분위기에서 39℃하의 진동 테이블상에서 배양시켰다.

세포 조작
성숙된 난모 세포는 성숙 개시 18 시간 후 소구 세포를 박리시켰다. 이어서, 표피 박리시킨 난모 세포는 10% 태내 송아지 혈청(FCS)을 함유하는 무칼슘 M2 배지로 세척하고, 이 배지 중에서 37℃하에 유지시켰다. 염색체를 제거하기 위하여(핵출화), 난모 세포를 10% FCS, 7.5㎍/㎖의 사이토칼라신 B(시그마) 및 5.0㎍/㎖ 헥스트 33342(시그마)를 함유하는 무칼슘 M2 중에 넣고 37℃에서 20분 동안 방치하였다. 이어서, 제1 극체의 바로 아래로부터 소량의 세포질을 20μM의 유리 피펫을 사용하여 흡인 분리하였다. 이와 같이 하여 흡인시킨 세포질을 자외선에 노출시켜 핵출을 확인하고 중기판의 존재를 조사하였다.

본 발명의 기법을 양 및 소의 배를 재구성하는 데 적용시켜 보았다. 현재로서는, 포배 단계의 배는 소에서 얻어 왔으나, 이들 배가 최종 수용체에 이식된 적은 없었다. 양에 있어서 7마리의 수용체 암양을 임신시킨 결과 5마리의 정상 새끼양이 탄생되었다(이 중 2마리는 출산 후 곧 죽었다). 이 실험들의 결과를 표 1 내지 3에 요약한다.
표 1은 휴지기의 TNT4 세포 모집단 및 3 가지 상이한 사이토플라스트 수용체를 사용하여 재구성된 양의 배를 포배 단계까지 발생시킨 결과를 나타낸 것이다. 재구성된 배는 재구성 7일 후까지 임시 수용체인 암양의 결찰된 수란관에서 배양시켰다. 그 결과는 회수된 배의 총수에 대한 상실배/포배 단계의 배의 백분율로서 나타내었다.

및 배의 재구성
3 가지 새로운 세포형, OME, BLWF1 및 SEC1을 사용하여 핵이식을 수행하였다. OME(양의 유선 상피 세포) 세포는 문헌[Fin et al., Biochem . Soc . Trans. 24 369S(1996)]에 기술된 절차를 사용하여 6년령의 성숙한 핀-도르셋 종(Fin-Dorset) 암양의 유선에서 분리된 생검 조직으로부터 형성시킨 상피 세포주이다. BLWF1(블랙 웰시 섬유 아세포) 세포는 블랙 웰시 종(Black Welsh) 암양을 블랙 웰시 종 숫양과 자연 교배 후 형성시킨 26일령의 블랙 웰시 태아를 해부 및 배양하여 얻은 섬유 아세포의 세포주이다. 원시 태내 섬유 아세포의 분리 방법은 문헌[Robertson, E.J., Teratocarcinomas and embryonic stem cells: A practical approach, 71-112, IRL Press Oxford(1987)]에 기재된 것에 따랐다. SEC1(양의 배세포)은 폴-도르셋 종(Pol-Dorset) 숫양과 교배한 과잉 배란성의 폴-도르셋종 암양에서 얻은 9일령의 배에서 유래된 상피 유사 세포주이다. SEC1 세포는 동시 계류 중인 PCT/GB95/02095(공개 번호 WO96/07732)에 기재된 바와 같은 TNT 세포와는 다음과 같은 이유에서 상이한 것이다. 첫째로, 두 세포주의 세포 형태학이 전혀 상이하고, 둘째로, 세포주 분리에 사용된 방법이 다르다. SEC1 세포주는 단일배에서 형성된 것인 반면, TNT 세포주는 세포군으로부터 유래된 것이다.
모든 세포주는 핵형화하였고, 형식 염색체 수는 54(2n)이었다. 배를 재구성하기 위한 핵 공여체로서 사용하기 전에, 혈청 농도를 0.5%로 감소시킨 후 휴지기의 유도는 전술한 바와 같이 하여 모니터하였다[Campbell et al., Nature 380 64-66(1996)]. 재구성된 배의 제조는 앞서의 실시예들에서 이미 설명한 바와 같다.
표 4는 여러 세포형으로부터 재구성된 핵이식 배의 발생을 요약한 것이다. 이 표에는 재구성된 배의 수, 포배 단계까지의 발생 및 3 가지 세포형의 각각에 대한 임신 빈도수를 나타내고 있다. 모든 세포주는 배의 재구성에 사용하기 전에 핵형화시켰다. 이 세포주들의 형식 염색체 수는 54이었다. 1개 내지 3개의 포배 단계의 배를 각각 동조화한 최종 수용체인 암양에게 이식하였다. 시험관내에서 배양한 재구성된 배는 10% 인체 혈청을 함유하는 SOFM(합성 수란관액) 10㎕(4가지 배)의 적가액에 넣고, 5% O2, 5% CO2 및 90% N2의 가습 대기 중에서 39℃하에 배양시켰다. 배양된 배는 2일마다 새 배지에 옮겼다. SOFM 배지는 문헌[Gardner et al., Biology of Reproduction 50 390-400(1994) 및 Thompson et al., Biology of Reproduction 53 1385-1391(1995)]에 따라 제조하였다.
표 5는 1996년 6월 24일에 임신 상태를 유지하고 있는 수용체인 암양, 배의 재구성에 사용된 세포형 및 분만 예정일을 나타낸다. 임신은 동조화한 최종 수용체인 암양에게 1 내지 3개의 상실배/포배 단계의 배(재구성후 7일째)를 이식시켜 행하였다. 재구성된 수는 표 4에 구체적으로 나타내었다. 사용된 약어는 다음과 같다. PD = 폴-도르셋종(Pol-Dorset), BW = 블랙 웰시종(Black Welsh), FD = 핀-도르셋종(Fin-Dorset), * = 포배 단계까지 시험관내에서 배양시킨 배.

Claims (7)
- 인간이외의 성체 포유동물의 세포 배양물과 배 클론의 조합물로서,(1) 상기 세포 배양물은 인간이외의 성체 포유동물 유래의 체세포를 포함하며,(2) 상기 배 클론은 핵이식에 의해 배양물내 공여체 세포로부터 생성되고, 상기 배는 출산만기까지 발생될 수 있는 것인 조합물.
- 인간이외의 성체 포유동물의 세포 배양물과 살아있는 자손 클론의 조합물로서,(1) 상기 세포 배양물은 인간이외의 성체 포유동물 유래의 체세포를 포함하며,(2) 상기 살아있는 자손 클론은 핵이식에 의해 배양물내 공여체 세포로부터 생성되는 것인 조합물.
- 제1항 또는 제2항에 있어서, 상기 체세포가 소, 양, 돼지, 염소, 말, 마우스 또는 랫트 유래인 것인 조합물.
