Abstract
본 발명은 휴지기의 공여체 세포 유래의 핵을 적당한 수용체 세포로 이식시키는 것을 포함하여, 동물의 배를 재구성하는 방법에 관한 것이다. 공여체 세포는 휴지기 세포로서, 즉 G1의 성장과 분열 사이클을 이탈하여 G0 상태에 정지시킨 것이다. 핵 이식은 세포 융합으로 수행할 수 있다. 그 다음 재구성된 배를 사용하여 1마리 이상의 동물을 만들 수 있다. 또한, 본 발명은 돌연변이 동물 및 유전자 이점이 많은 비돌연변이 동물의 제조 방법에도 유용하다.
Description
공여체 배 유래의 핵을 핵출된 난모세포나 하나의 세포 수정란으로 이식시켜 포유류의 배를 재구성하는 방법은 유전자적으로 동일한 개체를 생성시킨다. 이 방법은 연구용(즉, 생물학적 대조군)으로서, 그리고 상업용(즉, 유전자적으로 우수한 가축의 증식, 육제품의 균일성, 동물 관리)으로 유리하다. 그러나, 핵 공여체로서 초기 배 사용시의 한가지 문제점은 단일 배로부터 생성될 수 있는 자손의 수가 세포 수(사육 동물 종의 경우에는 32 내지 64 세포 단계의 배가 가장 널리 사용됨) 및 핵 이식 프로토콜의 효율성 면에서 제한을 받는다는 것이다.
핵 공여체로서 배의 사용과는 달리, 배양물중에서 유지될 수 있는 세포로부터 핵 이식에 의하여 정상 태아를 생산하는 능력은 동물 사육자들에게 얼마동안 추구되어 왔던 목적이다. 배양된 세포주로부터 클로닝된 태아를 생산하는 능력은 초기 배 사용시에 많은 이점을 제공할 것이다. 예컨대, 장기간동안 다수의 동일한 태아의 생산(배양 세포는 동결 보존할 수 있음) 및 유전자 변화시키고(또는) 배의 재구성전 필요한 유전자형(예, 성별)을 가진 세포 모집단을 선택할 수 있는 능력 등이다. 이러한 절차에 사용할 수 있는 하나의 세포 종류는 배의 간상부(ES) 세포이다. ES 세포는 마우스에서 분리되었었지만, 핵 이식에 사용된 이후 출산 만기까지 발육되었는지에 대해 보고된 바는 없다. 지금도, 돼지의 ES 유사 세포가 생체내 생성된 포배의 포배강으로 주입된 후 발육하였다는 보고서가 하나 있지만(Wheeler, Reprod. Fertil. Dev. 6 563-568(1994)), 다른 가축 종들에서 관찰한 키메라성에 대한 보고 및 임의의 소정의 세포주로부터 임의의 포유류 종내에 핵 이식 후 출산 만기까지의 발육 여부에 대한 보고는 전혀 없다.
ES 세포주를 사용할 수 있는 다른 여러 양태가 있는데, 이중의 하나는 핵 이식에 사용했을 때 발육을 촉진시킬 수 있는 다른 세포 모집단의 연구에 사용하는 것이다. 몇몇 보고서에서는 적당한 시험 세포로서 원시 배세포를 제공하였으나, 출산만기까지 발육하는지에 대해서는 보고된 바가 없었다. 초기 배로부터 세포주가 형성되었음이 보고되기도 하였으나, 여기에서 발육은 연장 배양시 양의 초기 계대 세포로부터 발생하는 것으로 보고되었고(Campbell et al., Therio 43 181(1995)), 통상적인 핵 이식 프로토콜을 사용한 경우에는 전혀 발육시키지 못했다(Campbell et al., J. Abstract Series (5) 31 (1995)).
핵 이식후 출산 만기까지 발육을 얻기 위하여, 이식된 핵의 발육 시계는 재조정되어야 한다. 이를 위하여 전사 유발이 억제되어야 하고, 그 후 발육 조절된 패턴으로 재개시되어야 한다. 종래에는 소, 양, 돼지, 토끼 및 마우스내의 각종 세포 종류를 이용하여 포배 단계로 발육시킬 수 있다는 것이 보고된 바 있다. 그러나, 이러한 모든 보고에서도 출산 만기까지의 발육에 대해서는 밝혀진 바가 전혀 없다. 9일된 양 배의 배 원반(ED)으로부터 형성된 초기 세포주(3 계대주까지)로부터 핵 이식 후 정상 새끼 양의 출생에 대하여 종래 보고된 바 있다(Campbell et al., Therio 43 181(1995)). 그러나, 그 이후의 배양물에서는 통상적인 핵 이식 프로토콜을 사용한 경우 출산 만기까지 어떤 발육도 일어나지 않았다(6 계대주 및 11계대주)(Campbell et al., J.Reprod.Fertil. Abstract Series(5) 31 (1995)). 이러한 결과는 다양한 의미로 해석할 수 있다; 첫째로 배양 초기 기간동안 얻어진 ED 유래의 모든 세포는 발육을 촉진할 수 있을 것으로 예상된다. 하지만, 배양된 세포주의 안정화동안 연장 배양시 이 세포들은 변화하며, 따라서 상기 문헌에 기재된 바와 같은 "유니버설 수용체"내로 핵 이식하기 위하여 핵 공여체로서 사용했을 때 발육을 조절할 수 없다. 두번째로, 초기 배양 기간동안 세포의 계대집단이 발육을 촉진시키는 성질을 보유하는 바, 이러한 초기 계대동안에 핵 이식 후 정상 태아의 생성을 설명할 수 있을 것으로 추정된다. 종래 한 연구 보고서에서는 핵 이식에 의해 재구성된 배의 발육에 있어서 공여체 핵과 수용체 세포질의 세포 주기 공조 역할에 대하여 강조하였다(Campbell et al., Biol. Reprod. 49 933-942(1993) 및 Biol. Reprod. 50 1385-1393(1994)).
공지된 핵 이식 프로토콜을 사용하는 핵 이식에 전능인 세포주의 분리에 의존적인 방법에 대안적인 가능한 2가지 전략은 다음과 같다:
(1) 기존의 핵 이식 절차를 변화시키는 방법; 또는
(2) 핵 이식 이전에 공여체 세포의 염색질을 변화시키는 방법.
전능 세포는 동물 전체의 발육을 유도할 수 있다(공여체 세포로부터 수용체 세포, 예컨대 핵출된 난모세포로의 핵 이식로 배를 제조한 경우에는, 공여체 세포의 핵이 전능성이다). 여기에는 배외 세포계, 즉 태반의 발육 유도도 포함된다. 이러한 의미로 말하면, 또한 수정된 수정란 및 일부 종에서는 각각의 난할구도 전능세포이다. 이와 대비적으로, 다능성 또는 다잠재성 세포(즉, 배 간상부 세포) 종류는 난할강으로 주입된 후 수태산물/태아내의 모든 조직을 형성할 수 있는 것으로 정의되었다.
전술한 핵 이식 방법 (1) 및 (2)에는 모두 이식된 핵의 유전자 발현을 재프로그램밍할 수 있는 방법이 필요로 된다. 즉, 이러한 방법에는 핵 공여체로서 분화 세포 또는 부분 분화 세포가 사용될 필요가 있으며, 따라서 그 세포 본래의 전능성이 유도될 것이다.
본 발명은 유전자 측면에서 선택되고(또는) 변형된 동물을 비롯한 동물의 생성방법에 관한 것이다.
본 발명은 휴지기 세포, 즉 세포 주기에 의하여 활동적으로 증식하지 않는 세포가 동물 배의 재구성시에 핵 공여체로서 유리하게 사용될 수 있다는 발견에 따른 것이다. 이 배들은 출산 만기까지 발육할 수 있었다. 배 재구성 후 관찰되고 효과적인 핵 이식에 필요한 공여체 핵의 변화는, 세포의 핵을 휴지기 상태로 만들어 핵 공여체로서 사용하기 이전에 세포의 핵내에 유도될 수 있는 것으로 관찰된다. 이러한 사실을 이용하여 본 발명을 완성하였다.
본 발명의 제1목적은 적합한 수용체 세포로 휴지기의 공여체 세포의 핵을 이식시키는 것을 포함하여, 동물의 배를 재구성하는 방법을 제공하는 것이다.
원리적으로, 본 발명은 모든 동물, 예컨대 가금류와 같은 조류, 양서류 및 어류종에 사용할 수 있다. 하지만, 실제로는 산업적 이용 가능성이 가장 클 것으로 예상되는 인간을 제외한 동물, 특히 포유 동물(인간 제외), 구체적으로 태반을 가진 포유 동물에 사용할 수 있을 것이다. 또한, 유제 동물, 특히 소, 양, 염소, 물소, 낙타 및 돼지와 같이 경제적면에서 중요한 유제 동물을 동물의 클로닝 수단 및 돌연변이 동물이나 유전자 변화된 동물의 생성 수단으로서 본 발명에 사용하는 것이 가장 유용할 것으로 생각된다. 또한, 유념해야 할 점은 본 발명이 경제적으로 중요한 다른 동물 종, 예를 들어 말, 라마 또는 설치류, 예를 들어 쥐 또는 생쥐, 또는 토끼에도 사용될 수 있을 것이라는 점이다.
본 발명은 돌연변이 동물 뿐만 아니라 비돌연변이 동물의 생성에 동등하게 적용할 수 있다. 돌연변이 동물은 유전자적으로 변화된 공여체 세포로부터 생성할 수 있다. 전체적인 과정은 돌연변이(즉, 유전적으로 변경된) 동물의 생성을 위한 통상적인 방법에 비해 하기하는 바와 같이 개략적으로 설명할 수 있는 다수의 이점을 보유한다:
(1) 더 적은 수의 수용체를 필요로 한다는 점;
(2) 클론성 공여체 세포를 이용하여 다수의 공통 유전자형 창시자(founder)를 생성할 수 있다는 점;
(3) 유전자 표적화에 의한 미세한 유전자 변화가 허용된다는 점;
(4) 본 발명에 의해 제조된 배로부터 생성된 모든 동물은 각 동물이 단일 핵으로부터 유도되기 때문에 배선(胚線)을 통해 관련된 유전자 변화를 전달하지만; 이에 반해, 포배 주입이나 다른 방법에 의한 변화된 간상 세포군의 주입후 전핵 주입 또는 키메라 현상(chimerism)에 의한 돌연변이 동물의 생성은 모든 세포가 상기 변화를 포함하지 않기 때문에 생성된 동물이 상기 배선을 통해 변화를 전달할 수 없는 일정 비율의 모자이크 동물을 생성한다는 점;
(5) 완전한 동물로 생성되기 이전에 유전자 변화[예를 들어, 통합(integration)] 부위에 대해 세포를 선택할 수 있다는 점.
동물과 관련하여 본 명세서에서 사용한 용어 "돌연변이의(transgenic)"는 많은 돌연변이 동물이 다른 종의 유전자(들)를 함유하지만, 동물의 배선 내에 다른 종의 유전자를 하나 이상 함유하는 동물만을 언급하는 것으로 한정하여 이해하여서는 안된다. 오히려, 상기 용어는 재조합 DNA 기법에 의한 기술적 개입의 대상인 배선을 가진 모든 동물을 광범위하게 언급하는 것이다. 예를 들어, 배선 내의 내인성 유전자가 결실, 중복, 활성화 또는 변화된 동물은 배선 내에 외인성 DNA 서열이 첨가된 동물 만큼 본 발명의 목적에 유용한 돌연변이 동물이다.
동물이 돌연변이 동물인 본 발명의 구체예에서, 공여체 핵은 유전자 변화되어 있다. 상기 공여체 핵은 하나 이상의 돌연변이 유전자(transgene)를 함유할 수 있으며, 핵 이식 및 배 재구성 이전에 유전자 변화를 일으킬 수 있다. 수정란의 웅성 또는 자성 전핵 내로의 주입과 유사 방법인 미량주입법은 유전자 변화의 한 방법으로 사용될 수 있지만, 본 발명은 이 방법에만 제한되지 않는다: 또한, 매스 형질전환 또는 형질감염 기법, 예를 들어 전기침투법, 비루스 형질감염 또는 리포펙션(lipofection)을 사용할 수도 있다.
상기한 본 발명의 방법에서, 핵은 휴지기의 공여체로부터 수용체 세포로 이식된다. 이 방법은 특정한 공여체 세포 종류에만 제한적으로 사용되지 않는다. 예컨대, 휴지기 상태로 유도될 수 있거나 생체내에서 휴지기 상태로 존재할 수 있는 배, 태아 및 성체의 체세포를 비롯하여 정상 핵형을 가진 모든 세포가 이 기법에 사용시 전능성을 나타낼 수 있을 것이다. 따라서, 본 발명에는 완전 분화 세포를 비롯하여 적어도 부분적으로 분화된 세포의 이용도 포함된다. 공여체 세포는 배양물일 수 있으나, 반드시 배양물 상태이지 않아도 된다. 이하에는, 배양된 소 원시 섬유아세포, 배 유래의 양 세포주(TNT4), 6년령의 성숙 양에서 얻은 양의 유방 상피 세포 유래의 세포주(OME), 태내 양 조직 유래의 섬유아세포의 세포주 및 9일령의 양 배에서 유래된 상피-유사 세포주를 예시하였다. TNT4 세포주를 포함하는 본발명에 유용한 배 유래의 세포주 군에 대해서는 본 출원과 동시 계류중인 PCT 특허 출원 번호 PCT/GB95/02095, 공개 번호 WO96/07732호에 자세히 기술되어 있다.
본 발명에 유용한 공여체 세포는 휴지기 세포, 즉 유사분열 세포 주기로 활동적으로 증식하지 않는 세포이다. 유사분열 세포 주기는 뚜렷한 4 단계, G1, S, G2 및 M을 갖고 있다. 개시기라고 하는 세포 주기의 초기 상태는 G1 단계에서 일어나고 독특한 기능을 갖고 있다. 다른 세포 주기를 선택하는 결정이나 유도는 개시기에서 이루어진다. 세포는 일단 개시기로 진행되면, 전(前)-DNA 합성 단계인 G1 단계의 후반 단계까지 진행된다. 제2 단계인 S 단계는 DNA 합성이 일어나는 시기이다. 이 시기 다음에는 DNA 합성과 유사분열 사이 기간인 G2 단계가 온다. 유사분열 자체는 M 단계에서 일어난다. 휴지기 세포(특정한 완전 분화 세포와 같이 천연적으로 휴지기인 세포 뿐만 아니라 휴지 상태가 유도된 세포 포함)는 일반적으로 상기 세포 주기의 4 단계중 어느 한 단계에 있는 것이 아닌 것으로 간주된다; 따라서, 이들이 세포 주기를 통해 정상적으로 진행하지 않는다는 것을 나타내기 위해 보통 G0 상태에 있는 것으로 표기한다. 휴지기의 G0 세포의 핵은 이배체 DNA 함량을 가진다.
배양된 세포는 화학 처리, 영양 결핍, 성장 억제 또는 유전자 발현의 조작을 비롯한 각종 방법에 의해 휴지 상태가 되도록 유도될 수 있다. 현재, 양 및 소의 세포주에서 휴지 상태를 유도하는 방법으로서 배양액내 혈청 농도를 감소시키는 방법이 성공적으로 이용되었다. 이러한 상태에서, 세포는 G1 단계동안 성장 주기를 이탈하여 전술한 바와 같이 소위 G0 단계에서 정지된다. 이 세포들은 재자극되어 성장 주기로 재진입될 때까지 수일동안(세포에 따라 더 장기간도 가능) 이 단계로 유지될 수 있다. G0 단계에서 정지된 휴지기 세포는 이배체이다. G0 단계는 세포가 분화할 수 있는 세포 주기의 한 시점이다. 휴지 상태에서는 많은 대사적 변화가 일어나는 것으로 보고되었고 그 예로는 다음을 포함한다: 일인산화된 히스톤, 섬모화된 중심소체, 모든 단백질 합성의 감소 또는 완전한 정지, 증가된 단백분해작용, 총 세포 RNA를 감소시키는 RNA 전사 감소 및 전환 증가, 폴리리보솜의 해리, 불활성 80S 리보솜의 축적 및 염색질 응축(문헌[Whitfield et al., Control of Animal Cell Proliferation, 1 331-365(1985)] 참조).
이러한 특징중 대부분은 핵출된 난모세포로 핵을 이식한 후 일어나야하는 것들이다. G0 상태가 세포 분화와 관련이 있다는 사실은 이것이 수용체 세포의 세포질에 의하여 재모델링 및/또는 재프로그래밍되는 것이 보다 용이한 핵/염색질 구조를 제공할 수 있음을 암시한다. 이러한 방식에서, 핵 공여체 세포는 휴지기 상태에 있음으로 인하여 공여체 핵의 염색질은 그 핵이 발육을 유도할 수 있도록 배의 재구성 또는 재구성전에 변화시킬 수 있을 것이다. 이러한 방법은 핵 공여체로서 공여체 세포를 사용하기 이전에 그 세포의 염색질을 변화시킨다는 점에서 종래 보고된 핵 이식 방법과는 다른 것이다.
공여체 세포 유래의 핵이 이식되는 수용체 세포는 난모세포 또는 다른 적합한 세포일 수 있다.
수용체 세포는 각종의 여러 발육 단계에 있는 세포로서 중기 I 내지 중기 II의 난모세포에서 부터 수정란에 이르는 세포 및 2-세포 배가 사용될 수 있다. 각 세포는 이점과 단점을 갖고 있다. 수정란을 사용하면 활성화가 충분한 반면, 난모세포에는 단성생식적 활성화가 필요로 된다(하기 참조). 일부 종에서는 난할-단계의 배 이용이 선호될 수 있는 다른 기작에 필요한 것으로서 단지 유전자 발현의 재프로그래밍만을 요구한다. 이 때, 전사는 마우스의 제2 세포 주기동안 개시되며, 포배 단계까지 2차원 전기영동으로 관찰되는 합성된 단백질 성질의 주요 변화는 전혀 없었다(Howlett & Bolton J. Embryol. Exp. Morphol. 87 175-206(1985)). 하지만, 대부분의 경우에는 수용체 세포는 난모세포일 것이다.
수용체는 핵출성인 것이 바람직하다. 핵 이식 과정에서 수용체 난모세포의 핵출은 필수적인 것으로 통념화되어 왔지만, 이러한 판단이 실험적으로 확인된 바는 없다. 유제 동물에 대해 기술된 최초의 방법은 세포를 1/2씩으로 나누는 단계를 포함하는데, 이들중 하나는 핵출성이기가 쉽다[참조: Willadsen Nature 320 (6) 63-65 (1986)]. 이 방법은 다른 미지의 1/2 세포가 여전히 중기 기구(metaphase apparatus)를 보유할 것이며, 세포질 부피의 감소로 새로운 배의 분화 패턴을 가속화시킬 수 있다는 단점이 있다[참조: Eviskov 등, Development 109 322-328 (1990)].
최근에는 최소량의 세포질과 함께 염색체를 분리하기 위한 시도에 다른 방법들이 이용되었다. 제 1 극체 및 주변 세포질의 흡출은 양 난모세포의 중기 II 기구를 67% 분리하는 것으로 확인되었다[참조: Smith & Wilmut Biol. Reprod. 40 1027-1035 (1989)]. 단지 DNA-특이성 플루오로크롬(획스트 33342)의 이용은 세포질 부피의 감소를 최소화하면서 핵출을 보장하는 한 방법이다[참조: Tsunoda 등, J. Reprod. Fertil. 82 173 (1988)]. 이 방법은 가축 종에서 통상 사용하고 있는 방법이다[참조: Prather & First J. Reprod. Fertil. Suppl. 41 125 (1990), Westhusin 등, Biol. Reprod. (Suppl.) 42 176 (1990)].
포유류에서 핵출을 위한 비침입성 기법에 대한 보고는 거의 없지만, 양서류에서는 자외선을 사용하는 조사가 일반적으로 사용되었다[참조: Gurdon Q. J. Microsc. Soc. 101 299-311 (1960)]. 포유류에서 상기 기법의 사용에 대한 상세한 보고는 없지만, DNA-특이성 플루오로크롬의 이용 중에도 생쥐 난모세포의 자외선에 대한 30초 이상 동안의 노출은 상기 세포의 발생 가능성을 감소시킨다는 점을 유념해야 한다[참조: Tsunoda 등, J. Reprod. Fertil. 82 173 (1988)].
공여체 세포 핵이 이식되는 수용체 숙주 세포는 핵출된 중기 II 난모세포, 핵출되고 불활성화된 난모세포 또는 핵출되고 예비활성화된 난모세포인 것이 바람직하다. 수용체가 적어도 핵출된 중기 II 난모세포인 경우에는, 활성화는 이식시에 일어날 수 있다. 또는, 수용체가 적어도 핵출되고 불활성화된 중기 II 난모세포인 경우에는 활성화는 차례로 일어날 수 있다. 전술한 바와 같이, 핵출화는 실제로 핵이나, 전핵, 또는 중기판(수용체 세포에 따라 결정)을 제거하여 물리적으로 수행하거나, 또는 예컨대 자외선 조사하거나 다른 핵출화 영향을 가하여 기능적으로 수행할 수 있다.
적합한 사이토플라스트(핵출된 난모세포) 수용체에는 다음과 같은 3가지가 있다:
1. 본 출원과 동일자 출원되고 동시계류중인 PCT 특허 출원 PCT/GB96/02098 (우선권 번호 GB9517779.6)에 기술된 "MAGIC 수용체"(중기 정지된 G1/G0 허용성 사이토플라스트).
2. "GOAT"(G0/G1 활성화 및 이식) - 활성화시의 MII(중기 II) 난모세포(Campbell et al., Biol. Reprod. 49 933-942(1993)).
3. "유니버설 수용체"(Campbell et al., Biol. Reprod. 649 933-942(1993), Biol. Reprod. 50 1385-1393(1994).
상기 3가지 수용체 모두는 재구성된 배에서 G0의 공여체 핵을 사용했을 때 정상 배수체를 생성할 것이다. 하지만, 최근 보고에 따르면 휴지기 세포 유래의 핵중 일부 핵은 핵 외피를 분리시키지 않고 S-단계 세포질에 도입시켰을 때 DNA 합성 단계로 진행될 수 없다고 주장하고 있다(Leno & Munshi, J. Cell Biol. 127(1) 5-14(1994)). 따라서, 일부 배는 "유니버설 수용체"를 이용하여 발육시킬 수 있지만, 방법 1 또는 2중 어느 한 방법에 따라서 고농도의 MPF(M-단계 촉진 인자 또는 성숙 촉진 인자)를 함유하는 MII 난모세포를 사이토플라스트 수용체로서 사용하므로써 발생 빈도수를 증가시킬 수 있을 것으로 추정하고 있다.
일단 적합한 공여체 및 수용체 세포를 선별하면 공여체 세포의 핵을 수용체 세포로 이식시켜야 한다. 보다 편리하게는 융합법으로 핵 이식을 실시할 수도 있다.
융합을 유도하는데 사용되어온 3가지 소정의 방법은 다음과 같다:
(1) 융합-촉진성 화합물, 예컨대 폴리에틸렌 글리콜에 세포를 노출시키는 방법;
(2) 센다이 비루스등과 같은 불활성화 비루스를 사용하는 방법; 및
(3) 전기 자극을 이용하는 방법.
폴리에틸렌 글리콜이나 다른 글리콜과 같은 융합-촉진 화합물에 세포를 노출시키는 방법은 체세포 융합의 경우에는 일반적인 방법이나, 배의 경우에는 널리 사용되지 않는 방법이다. 이것은 폴리에틸렌글리콜이 독성이므로 세포를 최소 시간동안 노출시켜야 하고 신속하게 화합물을 제거해야함에 따라 투명대의 제거도 수반할 수 있기 때문이다(Kanka et al., Mol. Reprod. Dev. 29 110-116(1991)). 마우스 배를 사용한 실험에서, 불활성화된 센다이 비루스는 난할 단계 배 유래의 세포 융합에 효과적인 수단을 제공하는데(Graham Wistar Inst. Symp. Monogr. 9 19(1969)), 활성화를 유도하지 않는다는 부가적인 실험 이점도 갖고 있다. 유제 동물에서는 일반적으로 단위생식 활성화를 유도하는데 사용된 것과 동일한 전기 충격에 의해 융합을 실시한다[참조: Willadsen Nature 320 (6) 63-65 (1986), Prather 등, Biol. Reprod. 37 859-866 (1987)]. 이들 종에서, 센다이 비루스는 대부분 융합을 유도하지만, 통상적으로 사용되던 용도에서 보다 신뢰도가 낮은 편이다[참조: Willadsen Nature 320 (6) 63-65 (1986)].
세포-세포 융합은 핵 이식을 수행하기 위한 바람직한 방법이지만, 사용할 수 있는 유일한 방법은 아니다. 기타 적합한 기법으로는 미량주입법을 들 수 있다[참조: Ritchie 및 Campbell, J. Reproduction and Fertility Abstract Series No. 15, p60].
핵 이식 전 또는 핵 이식 후(바람직함)(또는, 어떤 경우에는 적어도 핵 이식과 동시에), 일반적으로 적어도 세포가 난모세포인 경우에는 수용체 세포를 단성생식 활성화로 발생을 자극할 필요가 있다. 최근 실험에 따르면, 단위생식 활성화에 대한 필요 조건이 예상보다 훨씬 복잡한 것으로 밝혀졌다. 활성화는 전부-또는-무의 현상으로서, 전핵의 형성을 유도할 수 있는 다수의 처리들이 모두 전부 "활성화"를 유발하는 것으로 추정되었다. 그러나, 토끼의 난모세포를 전기 충격에 반복 노출시킨 시험에서는 적절한 일련의 충격의 선택과 Ca2+의 조절만이 이배체화된 난모세포를 임신 중기까지 발육 촉진시킬 수 있다는 것을 밝혀냈다(Ozil Development 109 117-127(1990)). 수정동안에는 세포내 칼슘 농도의 반복적이며 일시적인 증가가 나타났는데(Cutbertson & Cobbold Nature 316 541-542(1985)), 전기 충격이 칼슘 농도의 유사한 증가를 일으키는 것으로 생각된다. 칼슘 일시성의 패턴은 종마다 다르며, 예상컨대 전기 펄스의 최적 패턴도 유사한 방식으로 달라질 것이라는 것이 증명되었다. 토끼에 있어서 충격간의 간격은 약 4분이고(Ozil Development 109 117-127(1990)), 마우스에 있어서는 10 내지 20분인 한편(Cutbertson & Cobbold Nature 316 541-542(1985)), 소에 대하여 예비 관찰한 결과는 약 20 내지 30분이었다(Robl et al., in Symposium on Cloning Mammals by Nuclear Transplantation(Seidel ed.), 콜로라도 스테이트 유니버시티, 24-27(1992)). 대부분의 공지된 실험에서는 1회 전기 충격으로도 활성화가 유도되지만, 수차례의 충격에 노출시키면 재구성된 배의 발생 비율이 증가한다는 새로운 관찰 결과가 제시되었다(Collas & Robl Biol. Reprod. 43 877-884(1990)). 따라서, 충격의 수, 전기장 강도 및 충격의 지속 시간과 매질의 칼슘 농도를 최적으로 하기 위하여 각 경우마다 통상적인 방식으로 조정할 수 있다.
본 발명의 제 2 목적은 이미 기술한 방법으로 제조한 재구성된 동물 배를 제공하는 것이다.
본 발명의 제 3 목적은 동물을 제조하는 방법을 제공하는 것인데, 이 방법은
(a) 이미 기술한 바와 같이 동물 배를 재구성하는 단계; 및
(b) 동물을 상기 배로부터 출산 만기까지 발육시키는 단계; 및
(c) 필요에 따라, 이와 같이 형성된 동물을 사육하는 단계를 포함한다.
상기 단계 (a)는 이미 상술하였다.
이러한 목적의 본 발명의 방법에서 제 2 단계인 단계 (b)는 동물을 배로부터 출산 만기까지 발육시키는 단계이다. 이는 직접 또는 간접적으로 수행할 수 있다. 직접 발생에서, 상기 단계 (a)의 재구성된 배는 발생이 일어나는데 필요한 임의의 추가 조치없이도 단순히 발생된다. 그러나 간접 발생에서는, 상기 배는 완전한 발생이 일어나기 이전에 추가 조작될 수 있다. 예를 들어, 수율을 증가시키기 위해서상기 배는 난할하고, 상기 세포는 클론 단위로 증식시킬 수 있다.
대안으로 또는 부가적으로, 생육성 배 수율의 증가는 공여체의 클론 단위 증식에 의한 본 발명의 수단에 의해서 및/또는 그 사용이 일련의 (핵) 이식 과정으로 이루어지는 경우 획득할 수 있다. 현재 포배 형성의 획득률에 대한 제한점은 대부분의 배가 "재프로그램"되지 않는다는 사실에 기인할 수 있다(허용할 수 있는 수치가 획득됨에도 불구하고). 이러한 상황이 발생하는 경우, 그 후의 획득률은 다음과 같이 증가시킬 수 있다. 스스로 발생하는 각각의 배는, 예컨대 상실배 단계나 32 내지 64 세포 단계에서 핵 공여체로 사용할 수 있으며; 대안으로, 내부 세포괴 세포는 포배기에서 핵 공여체로 사용할 수 있다. 또한, 이러한 연속 이식으로 유래되는 배는 그 자체를 유력한 핵 공여체로서 사용하여 효율을 더욱 증가시킬 수 있다. 이들 배가 실제로 재프로그램된 유전자 발현을 보유하는 배를 반영하고, 이들 핵이 실제로 재프로그램된 경우(그러할 것으로 보임), 이후 각각의 발생하는 배는 상기 핵 이식 과정의 효율에 따라 증식할 수 있다. 획득될 것으로 생각되는 증가도는 세포 유형에 의존한다. 양에서, 16 세포기의 배로부터 미리 활성화된 "유니버설 수용체(universal recipient)" 난모세포로 하나의 할구를 이식하여 55% 포배기 배를 수득하는 것은 용이한 일이다. 따라서, 단세포에서 발생된 각각의 배가 16 세포기에서 8개의 세포를 산출할 수 있다는 가설은 설득력이 있다. 이러한 특징들은 단지 개략적인 지침에 불과하지만, 이후의 발생 단계에서 이점의 정도는 그 단계에서 상기 과정의 효율에 의해 결정된다는 것은 명백하다.
또한, 중요한 것은 핵 공여체 세포원으로 작용하는 새로운 세포주가 전술한 바에 따라 형성된 배 또는 이로부터 생성된 태아나 성체로부터 생산될 수 있다는 것이다.
어떤 경우에는, 재구성된 배를 출산 만기까지 발육시키는데 제한점이 있을 수 있다면 천연적으로 형성된 배와 핵 이식에 의해 재구성된 배에서 유래된 세포로부터 형성시킨 키메라성 동물을 발생시키는 것이 바람직할 수 있다. 이러한 키메라는 일정 비율의 천연 배 세포와, 포배 단계 이하의 임의의 단계에 있는 일정 비율의 재구성된 배 세포를 취한 뒤, 응집이나 주입에 의해 새로운 배를 형성시키므로써 만들 수 있다. 세포 비율은 50:50 이거나, 출산 만기까지 발육하는 배가 형성되기에 적합한 다른 비율을 사용할 수 있다. 이러한 경우에 정상 세포의 존재는 재구성된 배의 재구성 및 출산 만기까지의 발육과 출산이 성공적으로 이루어지도록 보조한다고 생각된다.
수율 개선의 방편에 대한 문제는 차치하고, 상기 재구성된 배는 생체 내에서 또는 시험관 내에서 포배로 배양할 수 있다.
핵 이식에 의해 유도된 배는 정상적인 배와는 다르며, 때로 배가 통상적으로 배양되는 배양 조건(적어도 생체내) 이외의 다른 배양 조건을 요구하거나 다른 배양 조건이 유리하다는 것을 경험적으로 알게 되었다. 그 이유는 아직 알려지지 않고 있다. 소 배의 일반적인 증식에서, 재구성된 배(동시에 이들중 다수)는 양의 수란관에서 5 내지 6일 동안 배양했었다[참조: Willadsen, In Mammalian Egg Transfer(Adams, E.E., ed.) 185 CRC Press, Boca Raton, Florida (1982)]. 그럼에도 불구하고, 배를 보호하기 위한 본 발명의 실시에서, 상기 배는 이식 이전에 한천과 같은 보호성 배지 내에 매립하고, 그후 일시적 수용체를 회수한 후 상기 한천으로부터 절개해낸다. 보호성 한천 또는 기타 배지의 기능은 2가지 인데, 그중 하나는 투명대를 함께 보유하므로써 배를 위한 구조 보조제로서 작용하는 것이고, 다른 하나는 수용체 동물의 면역계를 지닌 세포들에 대한 차단체로서 작용하는 것이다. 이러한 방법이 포배를 형성하는 배의 비율을 증가시킴에도 불구하고, 다수의 배가 상실될 수 있다는 단점이 있다.
시험관 내 조건이 사용되는 경우, 당업계에서 통상 사용되는 조건이 사용될 만 하다.
포배기에서, 출산 만기까지의 발생을 위한 적합성에 대해 배를 선별할 수 있다. 이것은 전형적으로 상기 배가 돌연변이이고, 적합한 통합제에 대한 선별 및 선택이 이루어져 온 경우 실시가능하다. 또한, 비돌연변이성 유전자 마커에 대한 선별도 이 단계에서 수행할 수 있다. 그러나, 본 발명의 방법은 초기 단계에서 공여체의 선별을 가능하게 하는데, 이는 일반적으로 바람직하다.
선별을 수행한 경우라면, 그 선별후, 포배기의 배는 출산 만기까지 발생할 수 있다. 이는 일반적으로 생체내에서 일어날 것이다. 포배까지의 발생을 시험관 내에서 수행한 경우에는, 이 단계에서 최종 수용체 동물 내로의 이식을 실시한다. 포배 발생이 생체 내에서 이루어진 경우에는, 원칙적으로 포배를 포배이전 숙주내에서 출산 만기까지 발생시킬 수 있음에도 불구하고, 실제로는 상기 포배를 통상 포배이전(일시적인) 수용체로부터 분리하고, 보호성 배지로부터 절제해 낸 후, 포배후(영구적인) 수용체로 이식될 것이다.
본원의 제 3 목적의 선택적인 단계 (c)에서, 동물은 선행 단계에 의해 제조된 동물로부터 사육된다. 이 방법에서, 동물은 목적하는 유전적 특성(들)을 보유하는 동물의 떼 또는 군집을 형성시키는데 사용할 수 있다.
유전자적으로 동일한 세포원으로부터 핵 이식에 의해 생성된 동물은 동일한 핵을 공유하나, 이들 동물이 상이한 난모세포로부터 유래되기 때문에 엄격한 의미로는 동일하지 않은 것이다. 이러한 상이한 기원의 중요성은 명백하지 않으나, 상업적인 특성에 영향을 미칠 수 있다. 아이오와 스테이트 유니버시티 브리딩 허드에서 실시한 젖소의 미토콘드리아 DNA의 최근 분석에 따르면, 이 DNA가 우유 및 증식능과 관련이 있는 것으로 확인되었다[참조: Freeman & Beitz, In Symposium on Cloning Mammals by Nuclear Transplantation(Seidel, G. E. Jr., ed.) 17-20, Colorado State University, Colorado (1992)]. 유사한 효과가 상기 젖소군 전체에 존재하는지의 여부는 확인해야 할 과제이며, 난모세포의 선택을 고려하는 것이 특정 상황에서 가능 또는 필요한지의 여부도 고려하여야만 한다. 소 사육 분야에서, 높은 유전자적 장점을 가진공여체로부터 다수의 배를 생성할 수 있는 능력은 유전자적 개선점을 전국적인 규모로 확산시킬 상당한 잠재적 가치를 보유할 수 있다. 적용의 범위는 배 각각의 비용 및 출산 만기까지 발생할 수 있는 이식된 배의 비율에 따라 달라질 것이다.
예시 및 개략용으로서, 돌연변이 및 비돌연변이 동물을 제조할 수 있는 전형적인 방법을 개략적으로 기술하면 다음과 같다:
(1) 핵형의 부과, 휴지기의 유도(G0 단계에 정지) 및/또는 발생 유도를 포함할 수 있는 적합한 공여체 세포를 선택 및 분리하는 단계;
(2) 유전자적으로 변화된 세포 모집단을 생산하기 위한 적합한 분자 생물학적 기법을 실시하는 단계, 이러한 기법으로는 유전자 삽입, 유전자 녹-아웃(knock-out), 유전자 녹-인(knock-in) 및 기타 다른 유전자 변화를 포함할 수 있다. 또한, 필요한 경우에는 적합한 마커를 갖거나 갖지 않은 적합한 작제물로 형질감염시켜 돌연변이화를 유도할 수 있다;
(3) 선택적으로, 필요한 유전자형/표현형(즉, 안정한 통합체)으로 변화된 세포 모집단이나 클론을 선별 및 선택하는 단계;
(4) 변화된 세포 모집단을 휴지기로 유도하는 단계;
(5) 핵 이식에 의해 배를 재구성하는 단계;
(6) 생체내 또는 시험관 내에서 포배까지 배양하는 단계;
(6a) 필요에 따라 안정한 통합체[(3)에서 수행된 경우에는 생략)] 또는 기타 목적하는 특성에 대해 선별 및 선택하는 단계;
(7) 필요에 따라 최종 수용체로 이식시키는 단계.
본원의 제 4 목적에 따라, 본 발명은 상기한 바와 같이 제조된 동물을 제공한다.
본 발명의 각 목적에 바람직한 특징은 각각 다른 목적에도 바람직하며, 필요에 따라 변화를 줄 수 있다.
본 발명은 예시하기 위하여 제공되는 하기 실시예를 참고로 하여 기술하지만, 본 발명은 이것에만 제한되지 않는다.
실시예 1: 공여체 세포의 휴지기 유도
배양된 세포주를 사용하여 휴지기를 유도하는 방법에는 예컨대, 접촉 억제 또는 혈청 결핍을 포함하여 다양한 방법이 제시되었다(참조: Whitfield et al., Control of Animal Cell Proliferation, 1 331-365(1985)). 휴지기 유도 방법은 중요한 사항은 아니지만, 세포를 성장 주기로부터 이탈시켜 이배체 DNA 함량을 가진 G0 상태에 정지시키고 생육성을 유지시키는 것이 중요한 단계이다. 실시예 3과 4에서는 혈청 결핍시킨 소의 원시 섬유아세포, 생체내에서 생성된 포배로 7일된 내부 세포괴로부터 안정화된 소의 세포주, 및 양 세포주(TNT4) 유래의 배를 사용하여 휴지기를 유도하고 세포를 세포 주기의 G0 단계에 정지시켰다. 혈청 결핍 방법은 실시예 5에 기술된 공여체 세포의 휴지기 유도 방법과 유사한 방법으로 실시하였다.
실시예 2: 난모세포의 분리 및 핵 이식
난모세포는 (i) 도살장에서 얻은 재료를 시험관내 성숙시키거나 경질적 소포 천공으로 얻거나; (ii) 생체내 성숙시킨 뒤 외과적으로 회수하거나; 또는 (iii) 다른 모든 적합한 방법으로 얻을 수 있다. 모든 생체내 성숙된 난모세포는 1.0% 태내 송아지 혈청(FCS)을 함유하는 칼슘 마그네슘 제거된 인산염 완충 식염수(PBS)로 수란관을 세척하므로써 수거할 수 있다. 시험관내 성숙된 난모세포는 수거한 뒤 1.0% FCS를 함유하는 칼슘 제거된 M2(Whittingham and Wales Aust.J.Biol.Sci. 22 1065-1068(1969))로 옮긴다. 난모세포는 소구 세포를 표피박락시킨 뒤 전술한 바와 같이 핵출시킨 것인데[(Campbell et al., Biol. Reprod. 49 933-942(1993) 및 Biol.Reprod. 50 1385-1393(1994)], 단 모든 절차에 칼슘이 제거된 매질을 사용하였다. 융합 절차는 전술한 방법[(Campbell et al., 1993, 1994 (상기 인용문헌)]의 변형 방법으로서, 하기 관련 부분에 기술된 바와 같으며, 대안적으로 공여체 세포를 핵출된 난모세포에 주입하여 핵을 도입시킬 수도 있다[(Ritchie & Campbell, J. Reprod. Fertil. Abstract Series (5) 60 (1995)]. 이러한 절차들의 시기는 종마다 다르며, 이하에는 생체내에서 성숙된 양의 난모세포와 시험관내에서 성숙된 소의 난모세포를 이용한 2가지 프로토콜을 개략한다.
실시예 3: 양에 대한 핵 이식법
3.1 공여체 암양의 과잉자극과 난모세포의 회수
스코틀랜드 블랙페이스 암양을 14일동안 프로제스타젠 스폰지로 동시처리하고(VeramixTM, Upjohn, UK), 연속 2일간 양의 소포-자극 호르몬(FSH)(OvagenTM, Immuno-chemical Products Ltd, New Zealand)을 3.0mg/일씩 일회 주입(총 6.0mg)하여 과잉배란하도록 유도하였다. 배란은 FSH를 2차 주입한 후 24시간이 지나서 8㎎의 고나도트로핀-방출 호르몬 유사물(GnRH ReceptalTM, Hoechst, UK)을 일회 투여량으로 투여하여 유도하였다.
GnRH를 주입한 후 24 내지 29 시간이 지나서 수란관으로부터 수정되지 않은 중기 II 난모세포를, 사용전까지 37℃에서 유지시킨 1.0% 태내 송아지 혈청(FCS)을 함유하는 둘베코 인산염 완충 식염수를 사용하여 세척해내어 회수하였다.
3.2 난모세포 조작
회수한 난모세포는 37℃에서 유지시키고, PBS 1.0% FCS로 세척한 뒤, 37℃하의 10% 태내 송아지 혈청(FCS)을 함유하는 칼슘이 제거된 M2 배지로 옮겼다. 염색체를 분리해내기 위하여(핵출화), 난모세포를 10% FCS, 7.5㎍/㎖의 사이토칼라신 B(시그마) 및 5.0㎍/㎖ 헥스트 33342(시그마)를 함유하는 칼슘 제거된 M2중에 넣고 37℃에서 20분동안 방치하였다. 그 다음 제1극체의 바로 아래로부터 소량의 세포질을 20μM의 유리 피펫을 사용하여 흡인 분리하였다. 이와 같이 흡인된 세포질을 자외선에 노출시켜 핵출화를 확인하고 중기판의 존재를 조사하였다.
3.3 배 재구성
10개 내지 20개의 난모세포 군을 핵출화하고, 5% CO2, 37℃에서 칼슘이 없는 M2 배지 20㎕ 적가물에 광유(SIGMA) 하에 첨가하였다. 배를 재구성하기 위하여 다음과 같은 3가지 프로토콜(a), (b) 및 (c) 각각을 사용하였다.
(a) "MAGIC"(중기 정지된 G1/G0 허용성 사이토플라스트)
핵출화 후 가능한 한 즉시 단세포를 유리 피펫을 사용하여 난모세포와 접촉시켜 투명대에 이전에 만들어진 구멍을 통해 상기 세포를 이식시켰다. 이러한 사이토플라스트/세포 결합체를 증류수중의 0.3M 만니톨 용액 200㎕를 함유하는 융합 챔버로 전달하고 이를 전극사이에 수작업으로 정렬시켰다. 5V의 AC 펄스는 3초 동안 가하고, 그 다음 1.25kV/㎝의 3DC 펄스를 80 μsec 동안 가하였다. 그 다음 결합체를 10% FCS를 함유하는 칼슘이 제거된 M2로 37℃에서 세정하고 오일이 중층된 동일 배지중에서 37℃, 5% CO2하에 항온배양하였다. 활성화시키기 30분전에 결합체를 5μM 노코다졸을 함유하는 10% FCS, 칼슘이 제거된 M2 배지로 옮겼다. 하기 기술되는 바와 같이 hCG 주입 후 32 내지 34 시간째 활성화를 유도하였다. 활성화 후, 재구성된 수정란은 10% FCS를 함유하는 배지 TC199(기브코)중에서 37℃, 5% CO2하에 추가 3시간동안 항온배양하였다. 그 다음 노코다졸을 함유하지 않는 동일 배지로 37℃에서 5분동안 3회 세정하고, 일시적 수용체 암양에 이식하기 전에 12 내지 15 시간동안 추가 배양하였다.
(b) "GOAT"(G0/G1 활성화 및 이식)
hCG 주입후 32 내지 34 시간째 단세포를 핵출된 난모세포와 접촉시켰다. 결합체를 증류수중에 0.3M 만니톨, 0.1mM MgSO4, 0.001mM CaCl2를 함유하는 용액 200㎕를 첨가한 융합 챔버(하기 참조)로 이식시켰다. 3V의 AC 펄스를 5초간 가한 다음 1.25kV/㎝의 3 DC 펄스를 80μsec간 가하여 융합 및 활성화를 유도하였다. 그 다음 결합체를 7.5㎍/㎖의 사이토칼라신 B를 함유하는 TC199 10% FCS로 세척하고, 이 배지중에서 37℃ 5% CO2하에 1 시간동안 항온배양하였다. 그 다음 결합체를 TC199 10% FCS로 세척하고 TC199 10% FCS중에서 37℃, 5% CO2하에 추가 12 내지 15 시간동안 배양하였다.
(c) "유니버설 수용체"
핵출된 난모세포를 hCG 주입한 지 32 내지 34 시간 후 활성화시키고(하기 참조) 그 다음 37℃ 5% CO2하에 TC199 10% FCS중에서 4 내지 6 시간동안 항온배양하였다. 그 다음 단세포를 난모세포와 접촉시키고 하기와 같이 융합을 유도하였다. 그 다음 결합체를 37℃, 5% CO2하에 TC199 10% FCS 7.5㎍ 사이토칼라신 B중에서 1 시간동안 항온배양하였다. 그 다음 결합체를 TC199 10% FCS로 세척한 뒤 37℃ 5% CO2하에 동일 배지중에서 추가 8 내지 11 시간동안 배양하였다.
3.4 융합 및 활성화
활성화시키기 위하여, 난모세포를 증류수중에 용해된 0.3M 만니톨, 0.1mM MgSO4, 0.001mM CaCl2용액 200㎕중에서 2개의 평행 전극 사이에 배치하였다(Willadsen, Nature 320 63-65(1986)). 1.25 kV/㎝의 1 DC 펄스를 80μs간 가하여 활성화를 유도하였다. 융합를 위해, 조작된 배를 유사한 방식으로 처리하였고, 그 외에도 핵출된 난모세포와 세포간의 접촉 표면을 전극사이에 평행하게 배열하였다. 3V의 AC 전류를 5초동안 가한 다음, 1.25kV/㎝의 3 DC 펄스를 80μs간 가하여 융합을 유도하였다.
3.5 배의 배양 및 분할(모든 군)
배양 기간이 경과한 후, 융합된 결합체를 PBS중의 1% 및 1.2% 아가(DIFCO)중에 이중 매립한 후, 동조화되지 않은 암양의 결찰된 수란관으로 이식시켰다. 결합체를 아가에 매립시켜 수용체 암양에 의한 배의 면역 거부반응을 방지 또는 감소시키고 결합체가 함께 보유되도록 보조하였다. 6일 후, 수용체 암양을 죽이고, PBS 10% FCS를 사용하여 수란관으로부터 배를 세정하여 회수하였다. 2개의 바늘을 사용하여 한천 조각으로부터 배를 절제해내고 발생을 현미경 조사하였다. 상실배/포배 단계로 발생한 모든 배를 가능한 한 즉시 동조화된 최종 수용체 암양의 자궁각으로 이식하였다.
또한, 배를 포배 단계로 발생시키는데에는 일시적 수용체 암양 대신에 시험관내 기법을 사용하는 것이 적합할 것이다.
실시예 4: 소에 대한 핵 이식법
4.1 시험관내 난모세포 성숙화
난소는 지방 도살장에서 입수하여 실험실로 운반해오는 동안 28 내지 32℃로 유지시켰다. 소구 난모세포 복합체(COC)는 피하 주사바늘(내경 1.2 ㎜)을 사용하여 직경이 3 내지 10 ㎜인 소포로부터 흡인하여 멸균 플라스틱 유니버설 용기에 주입하였다. 이 유니버설 용기를 가온실(35℃)에 방치하고, 10 내지 15분동안 소포 물질을 침전시킨 뒤 상청액의 3/4을 버렸다. 나머지 소포 물질을 10% 소 혈청이 보강된 해부용 배지(어얼스 염(기브코), 75.0 ㎎/ℓ의 카나마이신, 30.0 mM 헤페스를 함유한 TCM 199, pH 7.4, 몰삼투압 농도 280 mOsmol/H2O kg) 동등량으로 희석하여, 85 ㎜ 페트리 접시로 옮긴 뒤, 해부 현미경하에서 COC의 존재에 대해 조사하였다. 2 내지 3층 이상의 치밀한 소구 세포층을 가진 복합체를 선택하고 해부용 배지로 3회 세척한 뒤, 10% 소 혈청과 1 x 106과립막 세포/㎖가 보강된 성숙 배지(어얼스 염(기브코), 75 ㎎/ℓ의 카나마이신, 30.0 mM 헤페스, 7.69 mM NaHCO3를 함유한 TC 배지 199, pH 7.8, 몰삼투압 농도 280 mOsmol/H2O kg)로 옮긴 다음, 필요할 때까지 5% CO2를 함유하는 대기중에서 39℃하에 진동 테이블상에서 배양하였다.
4.2 난모세포 조작
성숙된 난모세포는 성숙 개시 18 시간 후 소구 세포를 박리하였다. 표피박락된 난모세포는 그 다음 10% 태내 송아지 혈청(FCS)을 함유하는 칼슘이 없는 M2 배지로 세정하고 이 배지중에서 37℃하에 유지시켰다. 염색체를 제거하기 위하여(핵출화), 난모세포를 10% FCS, 7.5㎍/㎖의 사이토칼라신 B(시그마) 및 5.0㎍/㎖ 헥스트 33342(시그마)를 함유하는 칼슘 제거된 M2중에 넣고 37℃에서 20분동안 방치하였다. 그 다음 제1극체의 바로 아래로부터 소량의 세포질을 20μM의 유리 피펫을 사용하여 흡인 분리하였다. 이와 같이 흡인된 세포질을 자외선에 노출시켜 핵출화를 확인하고 중기판의 존재를 조사하였다.
4.3 배 재구성
다음과 같은 3가지 재구성 방법 (a), (b) 및 (c) 각각에 핵출화된 난모세포를 사용하였다.
(a) "MAGIC"(중기 정지된 G1/G0 허용성 사이토플라스트)
핵출화 후 가능한 한 즉시 핵출화된 난모세포는 칼슘이 제거된 M2 10% FCS중에서 39℃하에 유지시키고, 단세포를 유리 피펫을 사용하여 난모세포와 접촉시켜 그 세포를 투명대에 이전에 만들어진 구멍을 통해 이식시켰다. 이러한 사이토플라스트/세포 결합체를 증류수중의 0.3M 만니톨 용액 200㎕를 함유하는 융합 챔버로 전달하고 이를 전극사이에 수작업으로 정렬시켰다. 3V의 AC 펄스는 5초 동안 가하고, 그 다음 1.25kV/㎝의 3DC 펄스를 80 μsec 동안 가하였다. 그 다음 결합체를 10% FCS를 함유하는 칼슘이 제거된 M2로 37℃에서 세정하고 오일을 중층시킨 동일 배지중에서 37℃, 5% CO2에서 항온배양하였다. 활성화시키기 30분전에 결합체를 5μM 노코다졸을 함유하는 10% FCS, 칼슘이 없는 M2 배지로 옮겼다. 하기 기술되는 바와 같이 활성화를 유도한 다음, 활성화 후 재구성된 수정란은 10% FCS를 함유하는 배지 TC199(기브코)중에서 37℃, 5% CO2하에 추가 3시간동안 항온처리하였다. 그 다음 노코다졸을 함유하지 않는 동일 배지로 37℃에서 5분동안 3회 세정하고, 일시적 수용체 암양에 이식하기 전에 12 내지 15 시간동안 추가 항온배양하였다(암양은 재구성된 배에 대한 일시적 수용체로서 보다 비용이 적게 드는 대안체이다).
(b) "GOAT"(G0/G1 활성화 및 이식)
핵출된 난모세포는 성숙 배지로 복귀시켰다. 성숙 개시 30 시간 또는 42 시간후 단세포를 핵출된 난모세포와 접촉시켰다. 결합체를 증류수중에 0.3M 만니톨, 0.1mM MgSO4, 0.001mM CaCl2를 함유하는 용액 200㎕를 첨가한 융합 챔버(하기 참조)로 옮겼다. 3V의 AC 펄스를 5초간 가한 다음 1.25kV/㎝의 3 DC 펄스를 80μsec간 가하여 융합 및 활성화를 유도하였다. 그 다음 결합체를 TC199 10% FCS로 세척하고, 이 배지중에서 37℃ 5% CO2하에 15 내지 20 시간동안 항온배양하거나(30hpm 군) 또는 4 내지 8 시간동안 항온배양하였다(42 hpm 군). [약어 "hpm"은 "후-성숙 시간"을 나타냄].
(c) "유니버설 수용체"
핵출된 난모세포는 성숙이 개시된지 30 시간 또는 42 시간 후 활성화시키고(하기 참조) 그 다음 37℃ 5% CO2하에 TC199 10% FCS중에서 8 내지 10 시간동안 항온배양하거나(30 hpm 군) 또는 4 내지 6 시간동안 항온배양하였다(42 hpm 군). 그 다음 단세포를 난모세포와 접촉시키고 하기와 같이 융합을 유도하였다. 그 다음 결합체를 37℃, 5% CO2하에 TC199 10% FCS중에서 12 내지 16 시간 동안 추가배양하거나(30 hpm 군) 또는 4 내지 6 시간동안 추가배양하였다(42 hpm 군).
4.4 융합 및 활성화

또한, 배를 포배 단계로 발생시키는데에는 일시적 수용체 암양 대신에 시험관내 기법을 사용하는 것이 적합할 것이다.
실시예 3의 결과(양 세포) 및 실시예 4의 결과(소 세포)
본 발명의 기법을 양 및 소의 배 재구성에 사용하였다. 현재 포배 단계의 배는 소에서 얻었으나; 이러한 배들이 최종 수용체로 이식된 적은 없었다. 양에서 7마리의 수용체 암양이 임신하여 5마리의 정상 새끼양을 출산하였다(이중 2마리는 출산 후 곧 죽었다). 이 실험들의 결과는 표 1 내지 3에 요약한다.
표 1은 휴지기의 TNT4 세포 모집단 및 3가지 상이한 사이토플라스트 수용체를 사용하여 재구성된 양의 배를 포배 단계까지 발생시킨 결과를 나타낸 것이다. 재구성된 배는 재구성한지 7일 후까지 일시적 수용체 암양의 결찰된 수란관에서 배양하였다. 그 결과는 회수된 배의 총 수에 대한 상실배/포배 단계의 배의 백분율로서 나타내었다.

실시예 5: OME, BLWF1 및 SEC1 세포를 사용한 양의 핵 이식 및 배 재구성
3가지 새로운 세포 종류, OME, BLWF1 및 SEC1을 사용하여 핵 이식을 수행하였다. OME(양의 유선 상피 세포) 세포는 문헌[Fin et al., Biochem. Soc. Trans. 24 369S(1996)]에 기술된 절차를 사용하여 6년령의 성숙한 핀-도르셋 암양의 유선에서 분리된 생검조직으로부터 형성시킨 상피 세포주이다. BLWF1(블랙 웰시 섬유아세포) 세포는 블랙 웰시 암양을 블랙 웰시 수양과 자연 교배후 형성된 26일령의 블랙 웰시 태아를 해부 및 배양하여 얻은 섬유아세포의 세포주이다. 원시 태내 섬유아세포의 분리 방법은 문헌[Robertson, E.J., Teratocarcinomas and embryonic stem cells: A practical approach, 71-112, IRL Press Oxford(1987)]에 기술된 바에 따랐다. SEC1(양의 배 세포)은 폴-도르셋(Pol-Dorset) 수양과 교배한 과잉배란성의 폴-도르셋(Pol-Dorset) 암양에서 얻은 9일령의 배에서 유래된 상피-유사 세포주이다. SEC1 세포는 동시계류중인 PCT/GB95/02095(공개번호 WO96/07732)에 기술된 바와 같은 TNT 세포와 다음과 같은 이유에서 상이한 것이다. 첫째, 두 세포주간에 세포 형태가 전혀 상이하고, 둘째, 세포주 분리에 사용된 방법이 다르다. SEC1 세포주는 단일 배에서 형성된 것인 반면, TNT 세포주는 세포 군으로부터 유래된 것이다.
모든 세포주는 핵형화하였고, 형식 염색체 수는 54(2n) 였다. 배를 재구성하기 위한 핵 공여체로서 사용하기 전에, 혈청 농도를 0.5%로 감소시킨 후 휴지기의 유도는 전술한 바와 같이 모니터하였다(Campbell et al., Nature 380 64-66(1996)). 재구성된 배의 제조는 상기 실시예에서 전술한 바와 같다.
표 4는 여러 세포 종류로부터 재구성된 핵 이식 배의 발생을 요약한 것이다. 이 표에는 재구성된 배의 수, 포배 단계로의 발생 및 3가지 세포 종류 각각에 대한 임신 빈도수를 나타내고 있다. 모든 세포주는 배 재구성에 사용하기 이전에 핵형화하였다. 이 세포주들의 형식 염색체 수는 54였다. 1개 내지 3개의 포배 단계 배를 각각 동조화된 최종 수용체 암양에게 이식하였다. 시험관내에서 배양한 재구성된 배는 10% 인체 혈청을 함유하는 SOFM(합성 수란관액) 10㎕(4가지 배) 적가액에 넣고, 5% O2, 5% CO2및 90% N2의 가습 대기중에서 39℃하에 배양하였다. 배양된 배는 2일마다 새 배지로 옮겼다. SOFM 배지는 문헌[Gardner et al., Biology of Reproduction 50 390-400(1994) 및 Thompson et al., Biology of Reproduction 53 1385-1391(1995)]에 따라 제조하였다.
표 5는 1996년 6월 24일에 임신상태를 유지하고 있는 수용체 암양, 배 재구성에 사용된 세포 종류 및 분만 예정일을 나타낸다. 임신은 동조화된 최종 수용체 암양에게 1 내지 3개의 상실배/포배 단계 배(재구성후 7일째)를 이식시켜 형성시켰다. 재구성된 수는 표 4에 구체적으로 나타내었다. 사용된 약어는 다음과 같다: PD = 폴-도르셋(Pol-Dorset), BW = 블랙 웰시(Black Welsh), FD = 핀-도르셋(Fin-Dorset), * = 포배 단계까지 시험관내에서 배양된 배.
