본 발명은 특정 C-10 치환된 아르테미시닌 유도체의 기생충 감염에 의해 유발된 질병의 치료 및(또는) 예방에 있어서의 용도, 신규한 특정 C-10 치환된 아르테미시닌 유도체, 그의 제조 방법 및 이러한 C-10 치환된 유도체를 함유하는 제약 조성물에 관한 것이다.
말라리아는 오늘날 전 세계에서 가장 중요한 인간 기생충 질환이다. 전 세계에 걸쳐 대략 2억 7000만명이 말라리아에 감염되어 있으며, 매년 약 200만명이 사망한다. 감염된 적혈구의 표면에 다양한 항원을 발현시킴으로써 복잡한 생존 메카니즘을 만들어내는 기생충의 능력으로, 기생충은 이러한 항원에 대한 숙주의 면역 반응의 파괴 작용으로부터 벗어날 수 있게 된다. 또한, 말라리아 감염 비율의 증가는 플라스모디움 팔시파룸(Plasmodium falciparum)의 클로로퀸-내성 균주 및 다른 여러 약물에 내성을 나타내는 균주의 확산으로 인한 것이다.
동물 보건 분야에서, 기생충 질환, 특히 기능상 말라리아에 관련된 질환은 주요 문제거리이다. 예를 들어, 네오스포로시스(neosporosis)는 동물에서 네오스포라 종의 기생충, 특히 네오스포라 카니눔(Neospora caninum)에 의해 유발된 질환을 설명하기 위해 사용되는 용어이다. 네오스포라 감염은 개, 소, 양, 염소 및 말 에서 발생하는 것으로 알려져 있다.
네오스포라 카니눔을 비롯한 네오스포라 종에 대한 최종 숙주는 알려져 있지 않을뿐 아니라, 이 기생충에 대한 완전한 생육 싸이클도 밝혀지지 않고 있다. 그러나, 쉬조고니(schizogony)로 알려진 무성 생식 단계, 및 단세포인 타키조이트(tachyzoite)/브라디조이트(bradyzoite) 단계의 거동은 분명하게 밝혀져 있다. 타키조이트는 엔도디오제니라 불리는 세포내 생식 후에 형성된 약 (3 내지 7) ×(1 내지 5) mm 크기의 감염성 단세포 기생충 단계이다. 타키조이트를 통한 생식은 특히 근육 또는 신경 세포와 같은 세포 기관에서 일어난다. 감염 후에 나타나는 병리학적 징후는 주로 이러한 조직과 관계가 있다. 개에서 자연 감염 후 약 5 내지 6주 후의 상기 질환의 징후로는 신경 세포의 염증에 의해 유발된 과민증, 및 후각부(hind leg)의 과신전 경향의 증가가 있다. 조직병리학적 병변은 신경계, 특히 뇌 및 척수에서 분명하게 나타난다. 단핵 세포(마크로파지, 림프구 및 혈장 세포)의 광범위한 비-화농성 염증, 신경교 돌출 및 혈관주위 침윤이 우세하며, 또한 호산구 및 호중구에서도 일부 나타난다. 근육 조직에서는, 육안으로 관찰할 수 있는 괴사 및 퇴행성 변화가 나타난다. 증상이 다소 강하게 나타나는 위축증과는 달리, 길고 옅은 세로 줄무늬가 나타난다.
캘리포니아와 오스트레일리아에서, 네오스포라 카니눔 감염은 가축의 유산에 대한 주요 원인인 것으로 보인다. 소의 질환에 대한 징후는 개의 질환에 대한 징후와 유사하다. 운동실조증은 분명히 나타나고, 관절 반사는 약하며, 후각부 마비(일부는, 다리 4개 모두에서 일어남)를 관찰할 수 있다. 조직학적 형상은 개 의 조직학적 형상, 주로 비-화농성 수막염 및 척수염과 유사하다.
네오스포로시스에 대해 적합한 화합물의 생체내 활성에 대한 데이타가 부족한 것은 적합한 생체내 시험 시스템이 아직 개발되지 않았기 때문이다. 처치가 예방적이기만 하다면, 즉 감염 전에 처치를 시작하는 경우, (식수를 통해 투여되는) 술파디아진(sulfadiazine)은 실험상 감염된 마우스에게 효과적이다. 개의 경우에는, 술파디아진 및 클린다마이신(clindamycin)을 사용한 처치는, 처치를 일찍, 즉 신경의 염증으로 인한 임상 징후가 처음 나타났을때 시작하는 경우에만 성공적일 뿐이었다.
콕시디아증(coccidiosis), 즉 소장의 감염은 상대적으로 사람에서는 거의 진단되지 않는 것으로, 이소스포라 벨리(Isospora belli)에 의해 유발된다. 그러나, 사람은 적어도 2종의 낭포-형성 콕시디아목의 종(사르코시스티스 쉬호미니스(Sarcocystis suihominis) 및 에스. 보비호미니스(S. bovihominis))의 최종 숙주이기도 하다. 이러한 포낭을 함유하는, 날것이거나 또는 불충분하게 요리된 돼지고기 또는 소고기의 섭취는 심한 설사를 일으킬 수 있는데, 그 원인이 정확하게 진단될 가능성은 거의 없다. 밝혀진 사실상 모든 종류의 메타조아를 포함하는 콕시디아(coccidia)(아피콤플렉사(Apicomplexa) 문, 에이메리이나(Eimeriina) 아목)는 가장 성공적인 기생충 원충류 군의 하나이다. 인간에게 특히 중요한 것은 가축에 기생하는 60 내지 100 종이며, 어떤 경우에 이들은 특히 가금류, 및 양, 송아지, 새끼 돼지, 토끼 등의 동물(표 A 참조)에서도 매우 심한 손실을 일으킬 수 있다.

약물의 용도에 관한 한, 예방법은 가금류에서 주요한 연구 대상으로, 증가된 질병 상태 단계까지는 증상이 나타나지 않으며, 치료법이 포유류에서 주요 전략인 것이다[McDougald 1982]. 다른 약물 중에서 폴리에테르계 항생제 및 술폰아미드는 이러한 치료 및 예방용으로 현재 사용된다. 그러나, 에이메리아의 약물-내성 균주가 나타났으며, 약물 내성은 이제 심각한 문제가 되었다. 따라서, 새로운 약물이 시급히 필요하다. 병원체 및 숙주가 다양하다면, 항콕시디아증 물질을 확인 및 시험하기 위한 "이상적인 모델"은 없다. 예를 들어, 가금류에서 콕시디아증을 예방하는데 사용되는 많은 물질의 대부분은 그 효과가 불충분하거나 또는 포유류의 콕시디아증에 대해 효과가 전혀 없기도 하다[Haberkorn and Mundt; 1989; Haberkorn 1996]. 동물에서 항콕시디아증 효과, 면역화 등에 대해 활성 성분을 시험한 많은 연구물과 지침서들이 간행되었다. 특히 중요하고 포괄적인 한 예는 에커트(Eckert) 등의 문헌[the survey of current methods (1995a)]에 기재되어 있다.
킹하오수(qinghaosu)로 알려지기도 한 화합물인 아르테미시닌(1)은 아르테미시아 아누아(Artemisia annua)에서 생산되는 테트라시클릭 1,2,4-트리옥산이다. 아르테미시닌 및 그의 유도체인 디히드로아르테미시닌(2), 아르템에테르(3) 및 소듐 아르테수네이트(4)는 말라리아의 치료에 사용하여 왔다.

상기의 아르테미시닌 유도체는 성공적이기는 하지만, 안정성, 생체이용성 및 잠재적 신경독성에 관련된 문제가 있다. 또한, 아르테미시닌 유도체는 다양한 기생충에 대해 넓은 활성 스펙트럼을 나타내야 한다.
본 발명에 이르러, 특정 C-10 치환된 아르테미시닌 유도체는 기생충 감염에 의해 유발된 질환의 치료에 효과적인 것으로 밝혀졌다. 이 화합물들은 플라스모디움, 네오스포라 또는 에이메리아 속의 기생충, 특히 플라스모디움 팔시파룸(말라리아를 일으킴), 네오스포라 카니눔(네오스포로시스증을 일으킴) 및 에이메리아 테넬라(콕시디아증을 일으킴) 감염에 의해 유발된 질환의 치료에 특히 효과적이다. 따라서, 본 발명은 플라스모디움 속의 유기체 이외의 기생충 감염에 의해 유발된 질환의 치료 및(또는) 예방에 사용되는 하기 화학식 I의 화합물 또는 그의 염을 제공한다.
상기 식에서, Y는 할로겐 원자, 임의로 치환된 시클로알킬, 아릴, C-연결 헤 테로아릴 또는 헤테로시클릴알킬기, 또는 -NR1R2기이며, 여기서 R1은 수소 원자 또는 임의로 치환된 알킬, 알케닐 또는 알키닐기이고, R2는 임의로 치환된 알킬, 알케닐, 알키닐, 시클로알킬, 아릴 또는 아랄킬기이거나, 또는 R1 및 R2는 사이에 있는 질소 원자와 함께 임의로 치환된 헤테로시클릭기, 또는 임의로 치환된 아미노산 에스테르로부터 유도된 아미노기이다.
적합한 염으로는 산 부가 염을 들 수 있으며, 이는 적합한 화학식 I의 화합물과 유기산 또는 미네랄산과 같은 적합한 산의 반응에 의해 형성된다. 미네랄 산을 사용한 반응에 의해 형성된 산 부가 염, 특히 염산 또는 브롬화수소산을 사용한 반응에 의해 형성된 염이 특히 바람직하다. Y가 -NR1R2(R1 및 R2

헤테로시클릴알킬기는 헤테로시클릭기에 의해 치환된 임의 알킬기일 수 있다. 바람직하게는, 헤테로시클릭 잔기는 상기 정의된 바와 같이 3원 내지 18원, 특히 3원 내지 14원, 특히 5원 내지 10원 헤테로시클릭기이며, 알킬 잔기는 C1-6알 킬, 바람직하게는 C1-4알킬, 특히 메틸기이다.

예를 들어, 가금류, 특히 닭, 오리, 거위 및 칠면조에서 콕시디아증 및 이와 관련된 기생충에 대한 치료 및 예방을 위해서는, 상기 활성 화합물 0.1 내지 100 ppm, 바람직하게는 0.5 내지 100 ppm을 적당한 식품, 예를 들어 영양성 식품내에 혼합할 수 있다. 특히, 투여 대상이 본 발명의 활성 화합물을 잘 참아내는 경우, 원한다면 사용량을 증가시킬 수 있다. 따라서, 본 발명의 활성 화합물은 음료수와 함께 사용할 수 있다.

이러한 면으로 바람직한 화합물에는 화학식 I의 화합물이 포함되는데, 여기서 Y는 플루오르 원자이거나, 페닐, 디메톡시페닐 또는 트리메톡시페닐기이거나 또는 프로필아미노, 플루오로페닐아미노, 비페닐아미노, 벤질아미노, 페닐에틸아미 노, 페닐메톡시카르보닐메틸아미노 또는 디메틸아미노기이다.
또한, 본 발명은 플라스모디움 속의 유기체 이외의 기생충 감염에 의해 유발된 질환의 치료 방법을 제공하는데, 치료를 요하는 숙주에게 상기 맨처음에 정의된 화학식 I의 화합물을 치료 유효량 투여하는 것을 포함한다. 바람직하게는, 기생충은 네오스포라 속 또는 에이메리아 속의 유기체이다. 마찬가지로, 플라스모디움 속의 기생충 감염에 의해 유발된 질환의 치료 방법도 제공되는데, 이 방법은 치료를 요하는 숙주에게 상기 정의된 바와 같은 화학식 I의 신규 화합물을 치료 유효량 투여하는 것을 포함한다.

<실시예 1>: 10β-플루오로-10-데옥소-10-디히드로아르테미시닌(10β-플루오로-10-데옥소디히드로아르테미시닌)(화학식 I: Y는 F임)의 제조
디클로로메탄(24 ml) 중 디히드로아르테미시닌(1.136 g, 4 mmol)의 용액을 질소하에 0 ℃로 냉각하고, 디에틸아미노술푸르 트리플루오리드(DAST)(0.6 ml, 4.8 mmol)를 첨가하였다. 반응 혼합물을 실온으로 가온한 다음, 질소하에 24시간 동안 교반하였다. 이 황색 용액을 다시 0 ℃로 냉각하고, Na2CO3 용액(5 %, 20 ml)을 첨가하고 혼합물을 실온에서 2시간 동안 교반하였다. 그 다음, 2-상을 분리하여 유기층을 1M HCl, 5 % NaHCO3 및 물로 세척하고 MgSO4로 건조하였다. 용매를 증발시 킨 직후에, 잔류물을 플래쉬 컬럼 크로마토그래피(10 % 에틸 아세테이트/헥산)로 2회 정제한 다음, 헥산(289 mg, 50.5 %)으로부터 재결정화하였다.

(a) 10-(트리메틸실옥시)디히드로아르테미시닌(화학식 II: Q는 -Si(CH3)3임)의 제조
방법 1
0 ℃에서 질소하에 피리딘(20 ml) 중 디히드로아르테미시닌(1.51 g, 5.32 mmol)의 용액에 클로로트리메틸실란(5.20 ml, mmol)을 적가하였다. 혼합물을 실온에서 추가로 1시간 동안 교반하고 빙수 혼합물에 부었다. 용액을 디에틸 에테르(3×15 ml)로 추출하고, 건조하고(MgSO4), 감압(in vacuo) 농축하였다. 잔류물을 플래쉬 크로마토그래피(SiO2, 5 % 에틸 아세테이트/헥산)로 정제하여, 백색 고체인 10-(트리메틸실옥시)디히드로아르테미시닌(1.47 g, 78 %)을 수득하였다.
방법 2
10α-(트리메틸실옥시)디히드로아르테미시닌(화학식 II: Q는 -Si(CH3)3임)의 제조

0 ℃에서 디클로로메탄(5 ml) 중 상기 (a) 방법 2에 기재된 바와 같이 제조한 10α-(트리메틸실옥시)디히드로아르테미시닌(372 mg, 1.04 mmol)의 용액에 브로모트리메틸실란(140 ㎕, 1.06 mmol)을 적가하였다. 혼합물을 0 ℃에서 추가로 30분 동안 교반하여 반응중에 10-브로모아르테미시닌을 제조하였다.
(c) 10β-페닐-10-데옥소-10-디히드로아르테미시닌(10β-(페닐)디히드로아르테미시닌)(화학식 I: Y는 페닐임)의 제조
상기 (b)에서 제조한 용액을 감압 농축하였다. 잔류물을 디에틸 에테르(5 ml) 중에 용해시켰다. 0 ℃에서 질소하에 이 용액에 브롬화 페닐마그네슘(1.40 ml, 2.38 mmol, 1.7 M)을 첨가하였다. 그 후에, 혼합물을 0 ℃에서 교반한 다음, 밤동안 실온에 도달케 하였다. 그 후에, 용액을 염화 암모늄 포화 용액으로 켄칭하고, 건조하고(MgSO4), 감압 농축하였다. 잔류물을 플래쉬 크로마토그래피(SiO2, 8 % 에틸 아세테이트/헥산)로 정제하여 백색 고체인 10β-페닐-10-데옥소-10-디히드로아르테미시닌(10β-(페닐)디히드로아르테미시닌)(159 mg, 45 %)을 수득하였다. 에테르/헥산 혼합물로부터 재결정화하여 무색의 직사각형 결정을 수득하였다.
nOe-차이 실험: δ5.75에서 H-10의 이중선 신호의 방출은 δ2.75에서 H-9의 다중선 신호를 10 % 증대시켰다. 이 결과는 H-10 및 H-9의 입체화학이 서로 신(syn)임을 보여주었다.

(a) 9,10-안히드로-10-데옥소아르테미시닌(9,10-안히드로아르테미시닌)의 제조
0 ℃에서 질소하에 디클로로메탄(28 ml) 중 디히드로아르테미시닌(500 mg, 1.86 mol)의 용액에 4-(N,N-디메틸아미노)피리딘(37 mg) 및 트리플루오로아세트산 무수물(0.79 ml, 5.58 mmol)을 첨가하였다. 혼합물을 실온으로 가온하고 밤새 교반하였다. 그 다음, 용액을 감압 농축하였다. 잔류물을 플래쉬 크로마토그래피(SiO2, 에테르:헥산은 0.5:9.5 내지 1.5:8.5임)로 정제하여 백색 고체인 9,10-안히드로-10-데옥소아르테미시닌(9,10-안히드로아르테미시닌)(180 mg, 25 %)을 수득하였다.

실온에서 질소하에 디클로로메탄(10 ml) 중 상기 (a)에 기재된 바와 같이 제조한 9,10-안히드로-10-데옥소아르테미시닌(9,10-안히드로아르테미시닌)(191 mg, 0.71 mmol) 및 1,3-디메톡시벤젠(130 ㎕, 1.00 mmol)의 용액에 삼불화 붕소 디에틸 에테레이트(2 방울)을 가하였다. 용액을 추가로 1시간 동안 교반한 다음, 20 % 염산 용액(5 ml)으로 켄칭하였다. 혼합물을 디에틸 에테르(3×20 ml)로 추출하고, 에테르 추출물을 건조하고(MgSO4), 감압 농축하였다. 잔류물을 플래쉬 크로마토그래피(SiO2, 15 % 에틸 아세테이트/헥산)로 정제하여 백색 고체인 10-(2',4'-디메톡시페닐)-10-데옥소-10-디히드로아르테미시닌(10-(2',4'-디메톡시페닐)-디히드로아 르테미시닌)(89.5, 44 %)을 수득하였다.

(a) 10β-(2'나프톡시)-디히드로아르테미시닌의 제조
℃에서 질소하에 테트라히드로푸란(10 ml) 중 디히드로아르테미시닌(568 mg, 2.00 mmol) 및 2-나프톨(288 mg, 2.00 mmol)의 용액에 트리페닐포스핀(524 mg, 4.00 mmol) 및 디에틸 아조디카르복실레이트(330 ㎕, 2.00 mmol)를 첨가하였다. 혼합물을 실온에서 철야 교반하였다. 그 후에, 황색 용액을 감압 농축하고 잔류물을 플래쉬 크로마토그래피(SiO2, 5 % 에틸 아세테이트/헥산)로 정제하여 백색 고체인 10β-(2'-나프틸옥시)디히드로아르테미시닌(185 mg, 23 %)을 수득하였다.

상기 실시예 3(b)에 기재된 10α-(트리메틸실옥시)디히드로아르테미시닌(356 mg, 1.00 mmol)으로부터 제조한 브롬화물과 티오모르폴린(300 ㎕, 3.00 mmol)을 반응시켜 플래쉬 크로마토그래피(8 % 에틸 아세테이트/헥산) 후에 백색 고체인 10α- (티오모르폴리노)-디히드로아르테미시닌(243 mg, 66 %)을 제조하였다.
<실시예 7>: 10α-(4'-(S,S-디옥소티오모르폴린-1'-일)-10-데옥소-10-디히드로아르테미시닌(10α-(4'-모르폴리노술포닐)디히드로아르테미시닌)(화학식 I: Y는 4'-(S,S-디옥소티오모르폴린-1'-일)(4-모르폴리노술포닐))의 제조
실온에서 질소하에 디클로로메탄(10 ml) 중 상기 실시예 6에 기재된 바와 같이 제조한 10α-(4'-티오모르폴리노)-10-데옥소-10-디히드로아르테미시닌(10α-(티오모르폴리노)디히드로아르테미시닌)(388 mg, 1.05 mmol)의 용액에 NMO(369 mg, 3.15 mmol), 분말 분자체(525 mg, 4 Å) 및 TPAP(약 18.5 mg)을 첨가하였다. 혼합물을 실온에서 철야 교반한 다음, SiO2의 패드를 통해 여과하고, 잔류물을 에틸 아세테이트(3×15 ml)로 세척하였다. 여액을 감압 농축하였다. 그 후에, 잔류물을 플래쉬 크로마토그래피(SiO2, 35 % 에틸 아세테이트/헥산)로 정제하여 백색 고체인 10α-(4'-(S,S-디옥소티오모르폴린-1'-일)-10-데옥소-10-디히드로아르테미시닌(10 α-(4'-모르폴리노술포닐)-디히드로아르테미시닌)(421 mg, 100 %)을 수득하였다.

실시예 3(b)에 기재된 10β-(트리메틸실옥시)디히드로아르테미시닌(356 mg, 1.00 mmol)으로부터 제조한 브롬화물과 1-벤질피페라진(212.1 ㎕, 1.22 mmol)을 반응시켜 플래쉬 크로마토그래피(40 % 에틸 아세테이트/헥산) 후에 백색 고체인 10α-(4'-벤질피페라진-1'-일)-10-데옥소-10-디히드로아르테미시닌(144.3 mg, 40 %)을 제조하였다.
<실시예 9>: 10α-(2'-푸릴)-10-데옥소-10-디히드로아르테미시닌(화학식 I: Y는 2-푸릴임)의 제조
방법 1
20 ℃에서 디클로로메탄(10 ml) 중 디히드로아르테미시닌(284 mg, 1.0 mmol)의 용액에 트리클로로아세토니트릴(2.0 ml, 20.0 mmol) 및 1,8-디아자비시클로[5.4.0]운데칸 1 방울을 첨가하였다. 혼합물을 20 ℃에서 2시간 동안 교반한 다음 20 ℃에서 감압 농축하였다. 그 후에, 0 ℃에서 잔류물을 디클로로메탄(10 ml) 중에 용해시키고 -40 ℃로 냉각시켰다. 이 용액을 푸란(1.09 ml, 15.0 mmol)으로 처리한 다음, 삼불화 붕소 디에틸 에테레이트(123 ㎕, 1.0 mmol)로 처리하고, 생성된 혼합물을 -40 ℃에서 30분 더 교반하였다. 혼합물을 포화 NaHCO3 용액으로 켄칭하고, 디클로로메탄(2×10 ml)으로 추출하였다. 추출물을 건조하고(MgSO4), 감압 농축하였다. 잔류물을 플래쉬 크로마토그래피(SiO2, 15 % 에틸 아세테이트/헥산)로 정제하여 무색 오일인 표제 화합물(11.0 mg, 3.3 %)을 수득하였다. 분석 샘플은 헥산으로부터 재결정화에 의해 수득하였다.
방법 2
(a) 10β-벤조일옥시-10-디히드로아르테미시닌(10β-디히드로아르테미시닐 벤조에이트)의 제조
0 ℃에서 질소하에, 테트로히드로푸란 중 디히드로아르테미시닌(568 mg, 2.00 mmol) 및 벤조산(244 mg, 2.00 mmol)의 용액에 트리페닐포스핀(524 mg, 2.00 mmol) 및 디에틸 아조디카르복실레이트(ml)를 첨가하였다. 혼합물을 실온으로 가온하고 철야 교반하였다. 용액을 감압 농축하였다. 플래쉬 크로마토그래피(SiO2, 10 % 에틸 아세테이트/헥산)처리하여 백색 고체인 10β-디히드로아르테미시닐 벤조에이트(419 mg, 53 %)를 수득하였다.
(b) 10α-(2'-푸릴)-10-데옥소-10-디히드로아르테미시닌(화학식 I: Y는 2-푸릴임) 의 제조
-45 ℃에서 디클로로메탄(5 ml) 중 10β-벤조일옥시-10-디히드로아르테미시닌(193 mg, 0.50 mmol)의 용액을 푸란(542 ㎕, 7.5 mmol)으로 처리한 다음, 삼불화 붕소 디에틸 에테레이트(123 ㎕, 1.0 mmol)로 처리하였다. 생성된 혼합물을 -45 ℃에서 1시간 더 교반하였다. 혼합물을 포화 NaHCO3 용액으로 켄칭하고 디클로로메탄(3×10 ml)으로 추출하였다. 추출물을 건조하고(MgSO4), 감압 농축하였다. 잔류물을 플래쉬 크로마토그래피(SiO2, 15 % 에틸 아세테이트/헥산)로 정제하여 무색 오일인 표제 화합물(53.7 mg, 32 %)을 수득하였다.
<실시예 10>: 10α-(피롤-2'-일)-10-데옥소-10-디히드로아르테미시닌(화학식 I: Y는 2-피롤릴임)의 제조
-50 ℃에서, 디클로로메탄(30 ml) 중 실시예 9의 방법 2(a)에 기재된 바와 같이 제조한 10β-벤조일옥시-10-데옥소아르테미시닌(700.8 mg, 1.80 mmol)의 용액을 피롤(624 ㎕, 9.00 mmol)로 처리하고 삼불화 붕소 디에틸 에테레이트(332 ㎕, 2.70 mmol)로 처리한 다음, -50 ℃에서 1시간 동안 교반하였다. 혼합물을 포화 NaHCO3 용액으로 켄칭하고, 디클로로메탄(3×10 ml)으로 추출하였다. 추출물을 건조하고(MgSO4), 감압 농축하였다. 잔류물을 플래쉬 크로마토그래피(SiO2, 30 % 디에틸 에테르/헥산)로 정제하여 무색 오일인 표제 화합물(486.6 mg, 81 %)을 수득하였다.
<실시예 11>: 10α-(4'-벤질-4'-메틸피페라지늄-1'-일)-10-데옥소-10-디히드로아르테미시닌 요오드화물 염(화학식 I: Y는 4'-벤질-4'-메틸피페라지늄-1'-일임)의 제조
0 ℃에서 질소 분위기하에 디클로로메탄(1.8 ml) 및 디에틸 에테르(5.4 ml)의 혼합물 중에 상기 실시예 8에 기재된 바와 같이 제조한 10α-(4'-벤질피페라진-1'-일)-10-데옥소-10-디히드로아르테미시닌(272 mg, 0.62 mmol)의 용액에 요오도메탄(36.7 ㎕, 0.59 mmol)을 적가하였다. 혼합물을 교반하고 밤새 서서히 20 ℃로 가온하였다. 침전물을 수집하여 디에틸 에테르(2×5 ml)로 세척하고 고 감압하에 건조하였다. 메탄올/디에틸 에테르로부터의 재결정화 반응에 의해 추가로 정제하 여 직사각형-플레이트형 결정(87 mg, 24 %)을 생성하였다.

(a) 시험관내의 네오스포라 카니눔 세포 배양에 대한 화합물의 스크리닝
스크리닝을 96-웰 플레이트(팰콘(Falcon) 3872)에서 수행하였다. 단층 숙주 세포(VERO 또는 ED)를 세포 배양 플레이트에 놓았다. 감염되지 않은 단층 세포를 2개의 50 ml 조직 배양 용기(세포 배양 면적 50 cm3)에서 배양하였다. 37 ℃의 CO2-배양기에서 트립신-EDTA(5 ml. Gibco 45300-019)를 사용하여 세포층을 분리하였다. 10분 후에, 대부분의 세포가 분리되었다. 5 ml 피펫을 사용하여, 약 1 ml의 가온된 우 태아 혈청을 함유하는 50 ml 원심분리 튜브(그라이너(Greiner), B769331)로 세포를 옮겼다. 1500 rpm에서 5분 동안 원심분리한 다음(베리퓨즈(Varifuge) 3.0, Heraeus), 액체를 제거하고, 세포 펠릿을 RPMI 배지(100 ml, 95 % RPMI 1640, 2 % FCS, 1 % L-글루타민, 1 % 탄산수소나트륨, 1 % 페니실린/스트렙토마이신)에서 현탁시켰다. 세포 현탁액을 6개의 96-웰 플레이트에 웰 당 150 ㎕로 피펫팅하였다. 코팅된 세포 배양 플레이트를 37 ℃에서 5 % CO2
하에 24시간 동안 인큐베이션 장치에 넣었다. 그 다음, 세포를 네오스포라 카니눔 타키조이트로 웰당 48,000 마리 타키조이트의 농도로 감염시켰다. 그 후에, 37 ℃에서 5 % CO2하에 24시간 동안 인큐베이션하였다.
본 발명의 시험 화합물(0.5 내지 1.5 mg)을 계량하여 1.5 ml 에펜도르프(eppendorf) 용기에 넣고, 디메틸 술폭시드 1 ml 중에 용해시켜, 약 1×10-3 gㆍml-1의 농도가 되게 하였다. 추가로 희석하는데 사용된 배지는 87 % RPMI 1640, 10 % FCS, 1 % L-글루타민, 1 % 탄산 수소 나트륨, 1 % 페니실린/스트렙토마이신으로 이루어져 있다. 첫번째 스크리닝에서, 10-5, 10-6 및 10-7 gㆍml-1의 농도가 사용되었다. 그 다음, 네오스포라 카니눔에 감염시키고 24시간 후에, 희석된 제제를 웰당 150 ㎕의 용적에서 세포 배양 플레이트에 옮겼다. 첫번째 열의 경우, 처리하지 않은 배지를 사용하였다. 이 열은 감염된 세포 및 감염되지 않은 세포를 대조군으로 포함하였다. 세포 플레이트를 37 ℃에서 5 % CO2하에 5일 동안 인큐베이션하였다. 처리 후 4일 및 감염 후 5일 후에, 다음의 평가 계획에 따라 역상 현미경으로 25×10의 확대하여 평가하였다.

(b) 시험관내 에이메리아 테넬라 세포 배양에 대한 화합물의 스크리닝
19일령의 병아리의 신장으로부터의 세포를 96-웰 플레이트(팰콘 3872)에서 행크(Hank) 락트알부민 가수분해물, 5 % 우 태아 혈청, 1 % 글루타민 및 1 % 비필수 아미노산의 배지 중에 단층으로 배양하였다. 42 ℃에서 5 % CO2하에, 2일 후에, 배양물을 웰당 약 30.00의 에이메리아 테넬라의 잘린 스포로조이트로 감염시켰다. 시험 화합물을 DMSO 중에 용해시키고 배양 배지로 희석하여 최대 10 ㎍ㆍml-1의 최종 농도로 하였다. 희석 단계는 1:10이었다. 감염 후 5일째, 배양물을 100배 확대하여 현미경으로 평가하고, 숙주 세포의 상태 및 무손상 쉬존트의 양, 및 유리 메로조이트를 측정하였다. 효과의 등급은 하기와 같다.

(c) 플라스모디움 팔시파룸에 대한 시험관내 스크리닝
2가지 기생충 종, 즉 클로로퀸에 대해 내성인 W2 및 클로로퀸에 대해 감응성이나 메플로퀸에 대해 내성인 D6을 사용하였다. 하기 표 IV에서, 가장 좋은 본 발명의 화합물은 2가지 종 사이에서 교차 내성을 나타내지 않아야 한다. 이 분석은 기생충에 의한 방사성 표지된 히포크산틴의 흡수에 의존하는데, 흡수의 억제는 공 지된 또는 후보 항말라리아 약물의 활성때문이다. 각 분석에서, 클로로퀸, 메플로퀸, 퀴닌, 아르테미시닌 및 피리메타민과 같은 입증된 항말라리아제를 대조군으로 사용하였다. 인큐베이션 기간은 66시간이었고, 처음의 기생충혈증은 1 % 헤마토크리트로 0.2 %이었다. 배지는 RPMI-1640 배양 배지였는데, 엽산 또는 p-아미노벤조산을 함유하지 않았다. 알부맥스가 단백질 결합이 적고, 이 모델에서 화합물이 약간 높은 활성을 유도하기 때문에, 보통 열에 의해 실활화된 10 % 미만의 인간 혈장대신에 알부맥스(Albumax)를 사용했다. 활성에 대한 어떤 선행 지식 없는 화합물의 경우에는, 화합물을 디메틸 술폭시드(DMSO)에 직접 용해하여, 완전 배양 배지로 400배 희석하였다. 미지의 화합물을 최대 농도 50,000 ngㆍml-1에서 시작하여, 이어서 2배 희석을 11번 수행하여 1048배의 농도 범위를 제공하였다. 이 희석 과정은 96웰 마이크로타이터 플레이트에서 비오멕(Biomek) 1000 액체 핸들링 시스템에 의해 자동적으로 수행하였다. 그 후에, 희석된 약물을 시험 플레이트로 옮기고, 기생충에 감염된 적혈구 200 ㎕를 첨가하고, 37 ℃에서 5 % CO2, 5 % O2, 및 90 % N2의 조절된 환경에서 인큐베이션하였다. 42시간 후에, 3H-하이포크산틴 25 ㎕를 첨가하고 플레이트를 추가로 24시간 동안 인큐베이션하였다. 66시간 후에, 플레이트를 -70 ℃에서 동결하여 적색 세포를 용균시킨 다음, 해동하고, 96-웰 수획기에서 유리 섬유 필터 매트 상에 수획하였다. 그 후에, 섬광 계수기로 필터 매트를 카운팅하였다. 각 약물에 대해, 농도 반응 프로필을 결정하고, 비선형의 로그곡선 형 투여량 반응 분석 프로그램에 의해 50 %, 90 % 및 10 %의 억제 농도(IC50, IC90 및 IC10)를 결정하였다.
3-희석 분석을 이용하여 고 매질 또는 저농도에서 활성을 측정할 수 있는 예비 스크린 포맷을 사용할 수 있다. 농도는 50,000, 500 및 50 ngㆍml-1으로 선택된다. 이들은 플레이트 당 시험 화합물 14종 및 공지 (표준) 화합물 1종을 갖는 96-웰 포맷 플레이트에서 똑같이 수행하였다. 약물을 혼합 및 희석하고, 시험 플레이트에 약물 및 기생충을 첨가하기 위해, 바이오멕(Biomek) 희석장치를 사용하여 시스템을 자동화하였다. 예비 스크린 포맷에서, 분석 필드(AF)가 "<"를 갖는다면, 화합물은 "매우 활성적"이고, IC 값은 AF 다음에 나열되어 있는 최종 희석 값(ml 당 나노그램) 미만일 가능성이 가장 크다. 대부분의 경우, 이러한 화합물을 낮은 출발 농도에서 다시 수행하여 정확한 IC 값을 결정하였다. AF가 ">"를 갖는다면, IC 값은 예비 스크린 희석 값보다 크며, 따라서 "AF>250"은 IC 값이 250 ngㆍml-1보다 크며, 추가의 스크리닝은 수행되지 않음을 의미한다. 이러한 경우, IC 값에 대해 0.00 값을 기록하였다.
