Abstract
엔도테린 길항제를 제외한 생리 활성 물질의 수용성 펩티드 유형의 수불용성 또는 약간의 수용성 다가금속 염 및 생분해성 폴리머를 함유하고 있는 서방성 제제. 본 발명은 아주 효과적으로 엔도테린 길항제를 제외한 수용성 펩티드 유형의 생리활성물질이 첨가되어 있으며, 엔도테린 길항제를 제외한 수용성 펩티드 유형의 초기 생리활성물질의 급격한 방출을 억제하는 서방성 제제에 관한 것이다. 본 발명의 서방성 제제는 생체내에서 투여된 후 그의 생활성을 유지하면서 엔도테린 길항제를 제외한 수용성 펩티드 유형의 생리활성물질을 방출할 수 있다. 더욱이, 서방성 제제에서 엔도테린 길항제를 제외한 수용성 펩티드 유형의 생리활성물질은 생활성을 거의 잃지않으면서 장기간 동안 안정하게 유지될 수 있다.
Description
생리활성물질, 특히 펩티드 또는 그의 유도체는 생체내에서 다양한 약리 효과를 나타낸다는 것이 공지되어 있다. 일부는 화학 합성에 의해, 또는 유전 공학과 세포 공학의 발전에 의해 대장균, 효모, 동물 세포 및 햄스터와 같은 유기체를 사용하여, 대량으로 제조되어 의약 분야에 사용되고 있다. 그러나, 이들 펩티드는 일반적으로 짧은 생물학적 반감기를 가지고 있기 때문에 자주 투여해야하고, 그 결과 환자는 물리적으로 상당한 부담을 안게된다. 이러한 문제점을 해결하기위한 서방성 제제를 개발하기위해 다양한 시도가 이루어져왔다.
수용성 생리활성물질, 특히 수용성 펩티드(이후 "펩티드"라함)의 서방성 제제를 개발하는 데 해결해야할 첫 번째 과제는 펩티드 용해도 조절 즉, 펩티드 방출 속도를 조절하는 것이다.
국제 특허 출원 번호 제 500286/1991 호의 일어 번역 공보에는 불용성 아연-프로타민-α-인터페론 복합체가 개시되어 있다.
일본 특허 공개공보 제 2930/1988 호에는 고분자 폴리펩티드가 분산되어 있는 폴리락티드를 함유하고 있는 계가 개시되어 있다.
일본 특허 공개공보 제 221855/1993 호와 제 172208/1994 호에는 수용성 펩티드를 수불용성 펩티드 염으로 전환시킨 다음, 생분해성 폴리머를 함유하고 있는 유기 매질에 현탁시켜 효율적으로 미세 과립에 수용성 펩티드를 첨가하는 기술이 개시되어 있다. 이들 특허 공보에 사용된 수용성 펩티드는 수용성 펩티드 분자의 주요 부분에 형성된 유기산 염이며, 그 예로는 파모에이트, 탄닌산, 스테아린산 또는 팔미틴산염을 들 수 있다.
상기에서 서술한 바와 같이 수용성 생리활성물질의 서방성 제제를 제조하기위해 다양한 시도가 이루어졌지만, 만족할만한 서방성 제제가 수득되지 않았다. 따라서, 초기 수용성 생리활성물질의 급격한 방출을 억제하고, 일정한 수용성 생리활성물질의 방출 속도를 제공하며, 수용성 생리활성물질의 생활성을 유지할 수 있는, 수용성 생리활성물질을 고효율로 함유하고 있는 서방성 제제의 개발이 요구되어 왔다.
본 발명은 엔도테린(endothelin) 길항제를 제외한 수용성 펩티드 유형의 생리활성물질의 수불용성 또는 약간의 수용성 다가 금속 염 및 생분해성 폴리머를 함유하고 있는 서방성 제제에 관한 것이다.
상기 문제를 해결하기위한 광범위한 조사를 통해, 본 발명자들은 산성기를 갖는 엔도테린 길항제를 제외한 수용성 펩티드 유형의 생리활성물질 또는 그의 수용성 염(이하 "생리활성물질"이라함)과 수용성 다가금속염의 결합으로부터 형성된, 엔도테린 길항제를 제외한 수용성 펩티드 유형의 생리활성물질의 수불용성 또는 약간의 수용성 다가 금속염(이하 "복합체"라함)을 제조하고, 이를 생분해성 폴리머에 분산시키거나 용해시킴으로써 생분해성 폴리머에 함유된 엔도테린 길항제를 제외한 수용성 펩티드 유형의 생리활성물질의 효율을 극적으로 증가시키고, 생체에 투여된 직후에 약물이 급격하게 방출되지 않는 서방성 제제가 수득될 수 있다는 것을 알아내었다. 이러한 발견을 바탕으로 더욱 연구를 거듭하여, 본 발명자들은 본 발명을 완성하기에 이르렀다.
따라서, 본 발명은 하기에 관한 것이다.

부수적으로, 본 발명에서 사용된 아미노산, 펩티드 등의 약어는 IUPAC-IUB 위원회의 생화학 명명법에 따르거나 관련분야에서 종래부터 사용되어온 약어를 기초로 한 것이다. 아미노산의 가능한 광학 이성질체는 다른 특별한 언급이 없다면 L-이성질체이다.
수불용 또는 약간의 수용성 다가 금속염중의 생리활성물질은 산성기를 가지고 있는 생리활성물질이다. 여기에서 산성 기로는 카르복실기와 술포기를 들 수 있다. 생리활성물질은 바람직하게는 펩티드 결합 또는 아미노산 및 산성기를 가지고 있는 생리활성물질이다.
산성기는 아미노산으로부터 유도될 수 있다. 더욱 바람직하게는, 생리활성물질은 산성기를 갖고 있는 수용성 펩티드 또는 그의 유도체이다. 생리활성물질의 수용해도는 25℃에서 1%(w/w) 이상이다.
생리활성물질은 바람직하게는 두 개 이상의 카르복실기를 가지고 있다.
생리활성물질의 분자량은 약 200 내지 약 200,000, 바람직하게는 약 200 내지 약 50,000, 더욱 바람직하게는 약 500 내지 약 40,000 이다.
생리활성물질의 대표적인 활성은 호르몬 작용이다. 생리활성물질은 천연, 합성, 반 합성 또는 유전공학제품, 또는 그의 유도체가 될 수 있다. 작용의 메카니즘에 있어서, 이들 생리활성물질은 작용성 또는 길항성일 수 있다.
생리활성물질, 특히 본 발명의 수용성 펩티드 또는 그의 유도체는 호르몬, 사이토카인, 조혈 인자, 성장 인자, 효소, 가용성 또는 가용화된 수용체, 항체 또는 그의 단편, 항원 함유 펩티드, 혈액 응고 인자, 접착 분자, 생리활성물질의 수용체에 결합할 수 있는 작용물질 또는 길항 물질 등을 들 수 있다.
호르몬의 예로는 인슐린, 성장 호르몬, 나트륨 배설증가 펩티드, 가스트린, 프로랙틴, 부신피질자극성 호르몬(ACTH), 갑상선 자극 호르몬(TSH), 황체형성호르몬(LH), 림프소절 자극 호르몬(FSH), 인간 융모막 고나트로핀(HCG), 모틸린, 칼리크레인 등을 들 수 있다. 호르몬은 바람직하게는 인슐린과 성장 호르몬이다.
사이토카인의 예로는 림포카인, 모노카인 등을 들 수 있다. 림포카인의 예로는 인터페론(알파, 베타, 감마), 인터류킨(IL-2 에서 IL-12) 등을 들 수 있다. 모노카인의 예로는 인터류킨 1(IL-1), 종양 괴사 인자 등을 들 수 있다. 사이토카인은 바람직하게는 림포카인이며, 더욱 바람직하게는 인터페론(알파, 베타, 감마)이다.
조혈 인자의 예로는 에리트로포이에틴, 과립세포 콜로니 자극 인자(G-CSF), 대식세포 콜로니 자극 인자(M-CSF), 트롬보포이에틴, 혈소판 성장 자극 인자, 거핵세포 상승 인자 등을 들 수 있다.
성장 인자의 예로는 염기성 또는 산성 섬유아세포 인자(FGF), 그러한 류의 구성원(예, FGF-9), 신경 세포 성장 인자(NGF) 또는 그러한 류의 구성원(예, IGF-1, IGF-2), 뼈 형태발생 단백질(BMP) 또는 그러한 류의 구성원 등을 들 수 있다.
효소의 예로는 슈퍼옥사이드 디스뮤테이스(SOD), 조직 플라스미노젠 활성화제(TPA) 등을 들 수 있다.
가용성 수용체의 예로는 가용성 IL-6 수용체, 유사 인슐린 성장 인자 결합 단백질(IGFBP), 가용성 TNF 수용체, 가용성 EGF 수용체, 가용성 IL-1 수용체 등을 들 수 있다.
가용화된 수용체의 예로는, IL-1 수용체, IL-6 수용체, TNF 수용체 또는 유전공학기법에 의해 가용화된 파스(Fas) 리간드 등과 같은 공지된 수용체를 들 수있다.
항체의 예로는 인간 단클론 항체, 쥐로부터 유도된 항체의 변종 부위가 사람으로부터 유도된 항체의 일정한 부위에 결합되어 있는 인간-쥐 키메라성 단클론 항체, 또는 그의 단편 등을 들 수 있다. 항체 유형의 예로는, IgM, IgG, IgE 등을 들수 있다. 상기에 기술된 항체에 의해 확인된 항원의 예로는 혈소판, 바이러스 등을 들 수 있다.
혈액 응고 인자의 예로는 인자 VIII 등을 들수 있다.
접착 분자의 예로는 피브로넥틴, ICAM-1 등을 들 수 있다.
더욱이, 생리활성물질의 예로는 엔도테린, Arg-Gly-Asp-Ser(RGDS), 하수체 아데닐산 사이클라제 활성화 폴리펩티드(PACAP) 등을 들수 있다.
상기 생리활성물질은 수용성 다가 금속과 접촉되어 수불용성 또는 약간의 수용성인 그의 다가 금속염으로 전환된다.
수용성 다가 금속염의 다가 금속은 알칼리 토금속(예컨대, 칼슘, 마그네슘), 전이금속(예컨대, 철(II,III), 구리(II), 아연(II) 등), IIIb족 금속(예컨대, 알루미늄(II,III)등), IVb족 금속(예컨대, 주석(II,IV) 등) 등과 같은 이가, 삼가 또는 사가 금속 등을 들 수있다. 다가 금속은 바람직하게는 알칼리 토금속 또는 전이금속이며, 더욱 바람직하게는 칼슘 또는 아연이고, 아주 바람직하게는 아연이다.
수용성 다가 금속염은 다가 금속과 산의 염, 예컨대, 다가 금속과 무기산의 염, 그리고 다가 금속과 유기산의 염을 들 수 있다.
다가 금속과 산의 염은 바람직하게는 실온(20℃)에서 수용해도가 약 20 mg/ml 이상, 더욱 바람직하게는 약 100 mg/ml 이상, 아주 바람직하게는 약 200 mg/ml 이상인 염이다.
다가 금속과 염을 형성하는 무기산은 염산, 황산, 질산, 티오시안산 등을 들 수 있다.
다가 금속과 염을 형성하는 유기산은 지방족 카르복실산과 방향족 산을 들 수 있다. 지방족 카르복실산은 바람직하게는 탄소수 2 내지 9의 지방족 카르복실산이다. 지방족 카르복실산은 지방족 모노카르복실산, 지방족 디카르복실산, 지방족 트리카르복실산 등을 들 수 있다. 이들 지방족 카르복실산은 포화되거나 포화되지 않은 것이다.
지방족 모노카르복실산의 예로는 탄소수 2 내지 9의 포화 지방족 모노카르복실산(예컨대, 아세트산, 프로피온산, 부탄산, 발레르산, 카프론산, 엔안틴산, 카프릴산, 페라르곤산, 카프린산 등) 및 탄소수 2 내지 9의 불포화 모노카르복실산(예컨대, 아크릴산, 프로피오릴산, 메타크릴산, 크로톤산, 이소크로톤산 등)을 들 수 있다.
지방족 디카르복실산의 예로는 탄소수 2 내지 9의 포화 지방족 디카르복시산(예컨대, 말론산, 숙신산, 글루타르산, 아디프산, 피멜산 등) 그리고 탄소수 2 내지 9의 불포화 지방족 디카르복실산(예컨대, 말레인산, 푸마르산, 시트라콘산, 메사콘산 등)을 들수 있다.
지방족 트리카르복실산의 예로는 탄소수 2 내지 9의 포화 지방족 트리카르복실산(예컨대, 트리카르발릴린산, 1,2,3-부탄트리카르복실산 등)들 수 있다.
상기 언급된 지방족 카르복실산은 1 또는 2개의 히드록실기를 가질 수도 있다. 그러한 지방족 카르복실산로는 글리콜산, 락트산, 글리세린산, 타르트론산, 말레산, 타르타르산, 시트르산 등을 들 수 있다.
지방족 카르복실산은 바람직하게는 지방족 모노카르복실산, 더욱 바람직하게는 탄소수 2 내지 9의 지방족 모노카르복실산, 아주 바람직하게는 탄소수 2 또는 3의 포화 지방족 모노카르복실산이다. 특히 바람직한 지방족 카르복실산의 예로는 아세트산 등을 들 수 있다.
방향족산의 예로는 벤조산, 살리실산 등을 들 수 있으며, 벤조산이 바람직하다. 다가 금속과 무기 산의 염, 즉, 무기산 다가 금속염의 예로는, 할로겐화물(예컨대, 염화아연, 염화칼슘), 황산염, 질산염, 티오시안산염 등을 들 수 있다.
다가 금속과 지방족 카르복실산의 염, 즉 지방족 카르복실산 다가 금속 염의 예로는, 칼슘 아세테이트, 아연 아세테이트, 칼슘 프로피오네이트, 아연 글리콜레이트, 칼슘 락테이트, 아연 락테이트, 아연 타르트레이트 등을 들 수 있다. 바람직한 지방족 카르복실산 다가 금속염은 칼슘 아세테이트와 아연 아세테이트이다. 보다 바람직한 것은 아연 아세테이트 등이다.
다가 금속과 지방족 방향족산의 염, 즉 방향족 다가금속 염의 예로는, 벤조에이트, 살리실산염 등을 들 수 있다. 보다 바람직한 것은 아연 벤조에이트이다.
생리활성물질의 수불용성 또는 약간의 수용성 다가 금속염은 용매에 수용성생리활성물질과 수용성 다가 금속염을 혼합함으로써 제조된다. 혼합 과정은 바람직하게는 물에서 수행된다.
물에서 생리활성물질과 수용성 다가 금속염의 혼합비(몰비)는, 예컨대 1:1 내지 1:1000, 바람직하게는 1:1 내지 1:100, 더욱 바람직하게는 1:1 내지 1:50, 아주 바람직하게는 1:1 내지 1:10이다. 물에서 양 성분의 농도는 수득한 복합체의 용해도를 초과한다면, 각각의 용해도 범위에서 조정가능하다.
상기 혼합으로부터 수득된 수용액의 pH는 생리활성물질의 생활성이 영향을 받지 않으며, 생리활성물질과 수용성 다가 금속염의 용해도가 과도하게 낮아지지 않는 그러한 것이어야한다. 혼합 과정은 통상적으로 증류수에서 수행되지만, 필요에 따라 약산성, 중성, 또는 약알칼리 pH로 조정된 물에서 수행될 수 도 있다.
여기에 언급된 "수불용성 또는 약간의 수용성이란" 비가역적이 아닌 가역적인 것으로, 수용해도가 아주 낮다는 것을 의미한다. 수용해도는, 실온(20℃)에서 약 0 내지 약 0.1%(w/w), 바람직하게는 약 0 내지 약 0.01%(w/w)이다.
이렇게 수득된 수용성 생리활성물질의 수불용성 또는 약간의 수용성 다가 금속염은 필요에 따라 진공 건조되거나 동결 건조된 후 사용된다. 본 발명의 서방성 제제에서, 생리활성물질의 수불용성 또는 약간의 수용성 다가 금속염의 함량은 통상적으로 약 0.1 내지 약 50%(w/w), 바람직하게는 약 1 내지 약 30%(w/w)이다.
생분해성 폴리머는 지방족 폴리에스테르(예컨대, 글리콜산, 락트산, 히드록시부탄산 등과 같은 하나 이상의 α-히드록시카르복실산, 말산 등과 같은 히드록시디카르복실산, 시트르산 등과 같은 히드록시트리카르복실산 등으로부터 합성된 단일 중합체, 공중합체 또는 그의 혼합물), 그리고 폴리-α-시아노아크릴산 에스테르, 폴리-γ-벤질-L-글루타민산 등과 같은 폴리아미노산의, 약간의 수용성이거나 수불용성인 고분자량의 폴리머를 들 수 있다. 이들은 적합한 비율로 혼합되어 사용될 수도 있다. 중합의 유형은 랜덤, 블록, 또는 그라프트일 수 있다.
생분해성 폴리머는 바람직하게는 지방족 폴리에스테르(예컨대, 글리콜산, 락트산, 히드록시부탄산 등과 같은 하나 이상의 α-히드록시카르복실산, 말산 등과 같은 히드록시디카르복실산, 시트르산 등과 같은 히드록시트리카르복실산 및 기타로 부터 합성된 단일 중합체, 공중합체 또는 그의 혼합물)이다.
상기 언급된 지방족 폴리에스테르 중에서, 신뢰할 수 있는 생분해성과 생체적합성의 측면에서 하나 이상의 α-히드록시카르복실산(예컨대, 글리콜산, 락트산, 히드록시부탄산 등)으로부터 합성된 단일 중합체 또는 공중합체가 바람직하다. 더욱 바람직하게는, 지방족 폴리에스테르는, 하나 이상의 α-히드록시카르복실산(예컨대, 글리콜산, 락트산, 히드록시부탄산 등)으로부터 합성된 공중합체이다. 또한 이들 공중합체는 혼합되어 사용될 수도 있다.
본 발명의 생분해성 폴리머는 일반적으로 알려진 방법에 의해 제조된다.
상기 기술된 α-히드록시카르복실산은 D-, L- 또는 D,L-배열을 가질 수 있지만, D-/L-배열(몰%)가 약 75/25 내지 약 25/75의 범위내에 있는 것이 바람직하다. D-/L-배열(몰%)의 비는 약 60/40 내지 약 30/70이 더욱 바람직하다.
상기 기술된 α-히드록시카르복실산의 공중합체의 예로는 글리콜산과 다른 α-히드록시산, 더욱 바람직하게는 락트산 또는 2-히드록시부탄산의 공중합체를 들수 있다.
α-히드록시카르복실산 공중합체는 바람직하게는 락트산-글리콜산 공중합체 또는 2-히드록시부탄산-글리콜산 공중합체이다.
더욱 바람직하게는, α-히드록시카르복실산 공중합체는 락트산-글리콜산 공중합체이다.
락트산-글리콜산 공중합체에 있어서, 함량비(락트산/글리콜산)(몰%)는 약 100/0 내지 약 40/60이다. 함량비는 더욱 바람직하게는 약 90/10 내지 약 45/55, 아주 바람직하게는 약 80/20 내지 약 45/55 이다. 락트산-글리콜산 공중합체의 중량평균 분자량은 약 3,000 내지 약 20,000, 바람직하게는 약 3,000 내지 약 14,000, 더욱 바람직하게는 약 3,000 내지 약 12,000 이다.

상기 기재된 2-히드록시부탄산-글리콜산 공중합체는 폴리락트산과 혼합되어 사용될 수도 있다. 폴리락트산은 D- 또는 L-배열이거나 혹은 그의 혼합물일 수 수 있지만, D-/L-배열(몰%)가 약 75/25 내지 약 20/80의 범위내에 있는 것이 바람직하다. D-/L-배열(몰%)의 비는 더욱 바람직하게는 약 60/40 내지 약 25/75이며, 아주 바람직하게는 약 55/45 내지 약 25/75이다. 폴리락트산의 중량평균 분자량은 바람직하게는 약 1,500 내지 약 20,000이며, 더욱 바람직하게는 약 1,500 내지 10,000이다. 또한 폴리락트산의 분산도는 바람직하게는 약 1.2 내지 약 4.0, 더욱 바람직하게는 약 1.5 내지 약 3.5 이다.
폴리락트산의 제조를 위해, 락티드 즉 락트산의 이량체의 개환 중합체, 그리고 락트산의 탈수 중축합 반응의 두 가지 방법이 알려져 있다. 본 발명을 위해 상대적으로 낮은 분자량의 폴리락트산을 수득하기위해서, 락트산의 직접 탈수 중축합이 바람직하다. 이 방법은, 예컨대 일본 특허공개공보 제 28521/1986 호에 기재되어 있다. 2-히드록시부탄산-글리콜산 공중합체와 폴리락트산이 혼합되어 사용될 경우, 이들의 혼합비는 약 10/90 내지 약 90/10(중량%)이다. 혼합비는 바람직하게는 약 20/80 내지 80/20이며, 더욱 바람직하게는 약 30/70 내지 70/30 이다.
본 발명의 명세서에서, 중량평균 분자량은 중량평균 분자량이 각각 120,000, 52,000, 22,000, 9,200, 5,050, 2,950, 1,050, 580 및 162인 9개의 폴리스티렌을 기준 물질로 한 겔투과 크로마토그래피(GPC)에 의해 수득한 분자량으로 정의된다. 또한 GPC 측정을 기초로 수평균 분자량이 계산되었다. 분산도는 중량평균 분자량과 수평균 분자량으로부터 계산된다. 상기 측정은 이동상으로 클로로포름을 사용하고 GPC컬럼 KF804L × 2(쇼와 덴코제)와 RI 검출기 L-3300(히다찌사제)를 사용하여 수행하였다.
촉매 유리 탈수 중축합 반응에 의해 합성된 상기 기재된 공중합체는 일반적으로 말단 카르복실기를 가지고 있다.
본 발명에서, 생분해성 폴리머는 바람직하게는 말단 카르복실기를 가지고 있다. 말단 카르복실기를 가지고 있는 생분해성 폴리머는 GPC측정에 의한 수평균 분자량과 말단기 측정에 의한 수평균 분자량이 거의 같은 폴리머이다.
말단기 정량에 의해, 수평균 분자량이 하기와 같이 계산될 수 있다:
약 1 내지 3 g의 생분해성 폴리머를 아세톤(25 ml)와 메탄올(5 ml)의 혼합 용매에 용해시키고, 말단 카르복실기 함량을 측정하기위해 지시약으로서 페놀프탈레인을 사용하여 실온에서 교반하는 동안 상기 용액을 재빨리 수산화칼륨의 0.05 N 알콜계 용액으로 적정한다. 말단기 정량을 기초로 한 수평균 분자량은 하기 식을 사용하여 계산되었다:
말단기 정량에 기초한 수평균 분자량 = 20,000 A/B
A: 생분해성 폴리머의 중량 (g)
B: 적정 끝점이 도달될 때까지 첨가된 수산화칼륨의 0.05 N 알콜계 용액의 양(ml).
예컨대, 말단 카르복실기를 가지고 있으며, 촉매 유리 탈수 중축합 반응에 의해 하나 이상의 α-히드록시 산으로부터 합성된 폴리머의 경우, GPC 측정에 기초로 한 수평균 분자량과 말단기 정량을 기초로 한 수평균 분자량은 거의 일치하였다. 한편, 필수적으로 어떠한 말단 카르복실기를 가지고 있지 않으며, 촉매를 사용한 개환 중합에 의해 환형 이량체로부터 합성된 폴리머의 경우, 말단기 정량을 기초로한 수평균 분자량이 GPC 측정을 기초로 한 수평균 분자량보다도 상당히 높았다. 이러한 차이점은 말단 카르복실기를 가지고 있는 폴리머와 말단 카르복실기를 가지고 있지 않은 폴리머의 차이를 명확하게 드러나게 한다.
말단기 정량에 기초한 수평균 분자량은 절대값이지만, GPC 측정에 기초한 수평균 분자량은 각종 분석 조건(예컨대, 이동상의 종류, 컬럼의 종류, 기준 물질,선택된 슬라이스 넓이, 선택된 기준선 등)에 따라 변화하는 상대치이다. 따라서, 후자를 절대수치로 나타내는 것은 어렵다. 그러나, GPC에 기초한 수평균분자량과 말단기 정량에 기초한 수평균 분자량이 거의 일치하다는 사실은 말단기 정량에 기초한 수평균 분자량이 GPC 측정에 기초한 수평균 분자량의 약 0.5 내지 약 2배, 바람직하게는 약 0.8 내지 약 1.5배라는 것을 의미한다. 또한, 말단기 정량에 기초한 수평균 분자량이 GPC 측정에 기초한 수평균 분자량 보다 상당히 높다는 사실은 말단기 정량에 기초한 수평균 분자량이 GPC 측정에 기초한 수평균 분자량의 약 2배 이상이라는 것을 의미한다.
본 발명의 서방성 제제는 생분해성 폴리머에 생리활성물질과 수용성 다가 금속염을 혼합하여 수득한 생리활성물질의 수불용성 또는 약간의 수용성 다가 금속염을 분산시킴으로써 제조된다. 서방성 제제의 제조 방법은 수중 건조법, 상분리법, 분사 건조법, 그의 개량법이 있다.
서방성 제제, 예컨대 마이크로캡슐의 제조 방법은 하기에 기재되어 있다.
(i) 수중 건조법(o/w 방법)
이 방법에서, 유기 용매중의 생분해성 폴리머의 용액을 먼저 제조한다. 본 발명의 서방성 제제를 제조하기위해 사용되는 유기 용매는 바람직하게는 120℃℃이하의 비점을 가지고 있다. 그러한 유기 용매로는 할로겐화 탄화수소(예컨대, 디클로로메탄, 클로로포름, 사염화탄소 등), 알콜류(예컨대, 에탄올, 메탄올), 아세토니트릴 등을 들 수 있다. 이들은 적절한 비율로 혼합되어 사용될수도 있다. 유기 용매는 바람직하게는 디클로로메탄과 아세토니트릴이며, 아주 바람직한 것은 디클로로메탄이다. 유기 용매 용액에서 생분해성 폴리머의 농도는 생분해성 폴리머의 분자량, 유기 용매의 종류 등에 따라 통상적으로 약 0.01 내지 약 80%(w/w)이며, 바람직하게는 약 0.1 내지 약 70%(w/w)이며, 더욱 바람직하게는 약 1 내지 약 60%(w/w)이다.
이렇게 수득된 생분해성 폴리머의 유기용매 용액에 필요에 따라 동결건조되거나 진공건조된 생리활성물질의 수불용성 또는 약간의 수용성 다가 금속염을 첨가하거나 용해시킨다. 이러한 조작에서, 첨가된 복합체의 양은 복합체:생분해성 폴리머 중량비가 약 1:2, 바람직하게는 약 1:3 이하가 되도록 조정된다.
이렇게 제조한 유기용매 용액을 수상에 첨가하고 터빈 유형 기계 교반기 등을 사용하여 o/w 에멀젼을 형성한 다음, 오일상에 용매를 증발시켜 마이크로캡슐을 얻는다. 수상의 부피는 통상적으로 오일상 부피의 약 1 내지 약 10,000 배, 바람직하게는 약 2 내지 약 5,000 배, 더욱 바람직하게는 약 5 내지 약 2,000 배이다.
유화제가 외부 수상에 첨가될수도 있다. 유화제는 안정한 o/w 에멀젼을 형성할 수 있는 한 어떠한 것이라도 사용할수 있다. 그러한 유화제의 예로는, 음이온성 계면활성제, 비이온성 계면활성제, 폴리옥시에틸렌 캐스터 오일 유도체, 폴리비닐피롤리돈, 폴리비닐알콜, 카르복시메틸셀룰로스, 레시틴, 겔라틴, 히알우론산 등을 들 수 있다. 이들은 적절하게 조합되어 사용될 수도 있다. 외부 수상에서 유화제의 농도는 바람직하게는 약 0.001 내지 약 20%(w/w), 더욱 바람직하게는 약 0.01 - 약 10%(w/w), 아주 바람직하게는 약 0.05 - 약 5%(w/w)이다.
상기 기술된 o/w 방법에서, 마이크로캡슐은 복합체가 생분해성 폴리머의 유기 용매 용액에 분산되는 방법, 즉 s/o/w 방법에 의해 제조될 수 있다.
(ii) 수중건조법(w/o/w 방법)
이 방법에서, 유기 용매중의 생분해성 폴리머의 용액을 먼저 제조한다. 유기 용매 용액에서 생분해성 폴리머의 농도는 생분해성 폴리머의 분자량, 유기 용매의 종류 등에 따라 통상적으로 약 0.01 내지 약 80%(w/w)이며, 바람직하게는 약 0.1 내지 약 70%(w/w)이고, 더욱 바람직하게는 약 1 내지 약 60%(w/w)이다. 복합체의 수상 분산액이 내부 수상으로 사용된다. 수상 분산액에서 복합체의 농도는 예컨대 약 10 내지 약 90%(w/v)이다. 상기 기술된 복합체의 수상 분산액이 생분해성 폴리머의 유기용매 용액에 유화되고 분산되고 터빈 유형 기계 교반기, 호모나이저 등을 사용한 공지의 분산 방법에 의해 w/o 에멀젼을 형성한다.
이러한 조작은 내부 수상과 생분해성 폴리머의 중량비를 약 1:2, 바람직하게는 약 1:3 이하로 하는 방식으로 수행된다. 내부 수상과 생분해성 폴리머의 유기용매 용액의 비율은 1:1,000 내지 1:1(v/v), 바람직하게는 1:100 내지 1:5(v/v), 더욱 바람직하게는 1:50 내지 1:5(v/v)이다.
이렇게 제조된 w/o 에멀젼은 이후에 다른 수상에 첨가되어 w/o/w 에멀젼을 형성한 다음, 오일상에서 용매를 증발시켜 마이크로캡슐을 수득한다. 이러한 조작은 상기 (i)에 따라 수행된다.
본 발명의 서방성 제제는 바람직하게는 미세한 입자의 형태로 사용된다. 이것은 서방성 제제가 통상적인 피하 또는 근내 주입을 위한 주사 바늘을 통해 투여될 때 환자에게 과중한 고통을 일으키지 않는다. 서방성 제제의 평균 입자직경은 예컨대, 약 0.1 내지 약 300 ㎛, 바람직하게는 약 1 내지 약 150 ㎛, 더욱 바람직하게는 약 2 내지 약 100 ㎛이다.
본 발명의 명세서에서, 미세 입자 형태의 서방성 제제는 또한 마이크로캡슐로 부른다.
여기에서 사용된 "마이크로캡슐"이란 용어는 "미세구"로서 부를 수도 있다.
본 발명의 서방성 세제는, 예컨대 그와 같은 마이크로캡슐로서, 또는 비경구 제제(예컨대, 근내, 피하 또는 내장 주사 또는 내재 제제, 코, 직장 또는 자궁 투과점막 제제 등) 또는 경구 제제(예컨대, 경질 캡슐, 연질 캡슐 등과 같은 캡슐, 과립 및 분말 등의 고상 제제, 현탁액 등의 액상 제제)의 각종 투여 형태로 투여될 수 있다.
본 발명에서, 서방성 제제는 바람직하게는 주사용으로 사용된다. 예컨대 서방성 제제가 마이크로캡슐일 경우, 실제 사용할 수 있는 주사용 서방성 제제를 수득하기위해 분산제(예컨대, Tween 80과 HCO-60과 같은 계면활성제, 카르복시메틸 셀룰로스, 나트륨 알기네이트 및 나트륨 히알우론산염 등과 같은 다당류), 보전제(예컨대, 메틸 파라벤, 프로필 파라벤 등), 등장제(예컨대, 염화나트륨, 만니톨, 소르비톨, 글루코스 등)과 함께 물에 마이크로캡슐을 현탁시킴으로써 수성 현탁액을 제조할 수 있다. 다른 방법으로는, 실제 사용할 수 있는 주사용 서방성 제제를 수득하기위해 본 발명의 서방성 제제를 레시틴과 같은 포스포리피드가 첨가되거나 첨가되지 않은 참기름 또는 옥수수 기름과 같은 식물성 오일, 또는 중급 지방산 트리글리세리드(예컨대, MIGLYOL 812)와 함께 마이크로캡슐을 분산시켜 오일성 현탁액을 제조한다.
예컨대 서방성 제제가 마이크로캡슐일 경우, 그의 평균 입자 크기는 주사용 현탁액으로 사용될 때 분산도와 바늘 투과에 대한 조건이 충족되는 한 약 0.1 내지 약 300 ㎛에서 선택된다. 바람직하게는 입자 크기는 약 1 내지 약 150 ㎛, 더욱 바람직하게는 약 2 내지 약 100 ㎛이다.
상기 기술된 마이크로캡슐은 전체 생산 공정이 살균되어 있는 방법, 감마선이 살균제로 사용되는 방법, 그리고 방부제가 첨가되는 방법에 제한받지 않고 살균 제제로 제조될 수 있다.
낮은 독성을 가진 본 발명의 서방성 제제는 안전하게 포유류(사람, 소, 돼지, 개, 고양이, 생쥐, 쥐, 토끼 등)에 사용될 수 있다.
본 발명의 서방성 제제의 적용은 사용되는 생리활성물질에 따라 다양하다. 예컨대, 본 발명의 서방성 제제는 생리활성물질이 인슐린일 경우 진성 당뇨병 등을, 생리활성물질이 인터페론 알파일 경우 직장암, C형 간염, 생리활성물질이 에리트로포이에틴일 경우 빈혈 등, 생리활성물질이 성장 호르몬일 경우 성장 장애, 생리활성물질이 과립세포 콜로니 자극 인자일 경우 항암 치료요법후 호중구 감소증 등을 치료 또는 예방하는 데 효과적이다. 생리활성물질이 에리트로포이에틴일 경우, 본 발명의 생리활성물질은 또한 자가혈액수혈을 위한 조혈을 증진하는 데 효과적이다.
생리활성물질의 종류와 함량, 생리활성물질 방출의 지속, 대상 질병, 대상 동물 및 다른 인자에 따라, 서방성 제제의 투여량은 생리활성물질이 그의 작용을나타내는 그러한 수준으로 조정될 수도 있다. 생리활성물질의 투여량은 제제가 1주일 제제일 경우 각 성인에 대해 약 0.0001 내지 약 10 mg/kg 체중의 범위에 걸쳐 적절하게 선택할 수있다. 더욱 바람직하게는 투여량은 약 0.0005 내지 약 1 mg/kg 체중의 범위에 걸쳐 적절하게 선택할 수 있다.
서방성 제제의 투여량은 바람직하게는 각 성인에 대해 약 0.0005 내지 약 50 mg/kg 체중의 범위에 걸쳐 적절하게 선택할 수 있다. 더욱 바람직하게는, 투여량은 약 0.0025 내지 약 10 mg/kg 체중의 범위에 걸쳐 적절하게 선택할 수 있다. 투여 빈도는 생리활성물질의 유형, 함량 및 투여 형태, 대상 질병, 대상 동물 및 다른 인자에 따라 예컨대 일주에 한 번, 매 2주에 한 번 또는 매 4주에 한 번으로 적절하게 선택할 수 있다.
본 발명의 제제는 상온 또는 냉장된 곳에 저장할 수 있지만, 냉장된 곳에 보관하는 것이 바람직하다. 여기에서 언급할 수 있는 상온과 냉장된 곳은 일본 약전에 정의된 것과 같이 구체적으로 상온에 대해서는 15 내지 25℃, 냉장된 곳에 대해서는 15℃ 이하를 말한다.
본 발명은 이하에 하기 실시예에 의해 더욱 상세히 기술된다. 하지만 제한하려는 의도로 주어진 것은 아니다.
참고예 1
100 mM 수산화나트륨 수용액 22 ml중의 돼지 인슐린(27.3 U/mg, DIOSYNTH, 네덜란드) 0.5 g의 용액과 증류수 10 ml중의 아연 아세테이트(이수화물) 1 g의 용액을 함께 혼합하여 실온에서 1시간 유지한다. 약 3,000 rpm(05PR-22, 히다찌사)에서 원심분리한 후, 상청액을 버린다. 잔류물을 다시 증류수에 분산시키고 원심분리시킨다. 상청액을 버린후, 소량의 증류수를 잔류물에 첨가한 다음, 동결건조시켜 건조 분말로서 돼지 인슐린의 조 아연 염 약 1 g을 수득한다.
이렇게 수득한 분말내 인슐린 함량을 측정하기위해, 상기 분말을 약 3시간 동안 흔든 30% 아세토니트릴을 함유하고 50 mM EDTA로 추출한 다음, 고속 액체 크로마토그래피(HPLC)에 의해 정량 분석한다. 건조 분말 100 mg당 돼지 인슐린이 47.6 mg이 함유되어있었다.
참고예 2
수산화칼륨의 40% 수용액 168 ml와 에틸 에테르 1,000 ml의 혼합물에 니트로소에틸우레아 104 g을 조금씩 첨가하면서, 이 혼합물을 빙냉 조건하에서 교반한다. 수득한 황색 에테르 층을 분리하고 과립상 수산화칼륨을 첨가하여 건조시킨다. 이후 수산화칼륨을 제거하여 약 900 ml의 디아조에탄 용액을 수득한다.
락트산-글리콜산 공중합체(중량평균 분자량이 약 5,800인 락트산/글리콜산=50/50 (몰%)) 130 g을 염화메틸렌 1,900 ml에 용해시키고, 교반하고 냉각시킨다. 빙냉조건하에서, 상기 디아조메탄 용액을 적가한 다음, 실온에서 2 시간동안 교반시킨다. 혼합물을 하룻밤 방치한 후, 이 용매를 감압하 증류제거하고, 잔류물을 실온에서 감압건조시켜 락트산-글리콜산 공중합체의 에틸 에스테르 131 g을 수득한다.
참고예 3
증류수 0.9 ml에 인간 성장 호르몬(바이오테크놀러지 제네랄, USA) 1 mg의 용액과 증류수 0.1 ml중의 아연 아세테이트(이수화물) 9.98, 29.43, 49.88, 69.84, 79.81 또는 99.77 μg의 용액을 함께 혼합한다. 아연 원자 대 성장 호르몬의 몰비는 1, 3, 5, 7, 8 및 10 이다. 몰비가 5인 경우, 약 60%의 인간 성장 호르몬이 침전되었다. 몰비가 7 이상인 경우, 거의 100%의 인간 성장 호르몬이 침전되었다.
참고예 4
류프롤리드 아세테이트(TAP-144) 1 g과 겔라틴 157.5 mg을 70 내지 80℃에서 1 ml의 증류수에 용해시킨다. 수용액의 겔화 온도보다 약간 높은 온도로 데운 수용액에, 디클로로메탄 13.15 g에 락트산-글리콜산 공중합체 7.85 g[락트산/글리콜산:75/25(몰%), 점도:0.142 내지 0.169 cP]를 용해시킴으로써 제조한 락트산-글리콜산 공중합체 21 g의 용액을 첨가한다. 이 혼합물을 수분 이상 동안 소형 호모나이저로 유화시켜 W/O 에멀젼을 제조한다. 수득된 W/O 에멀젼을 10 내지 20℃로 냉각한다. 에멀젼을 10 내지 20℃로 조정한 0.1%(w/v) 수성 폴리비닐알콜 용액 5000 ml에 붓고, 이 혼합물을 터빈 호모나이저를 사용하여 유화시켜 W/O/W 에멀젼을 제조한다. 이 W/O/W 에멀젼을 실온(15 내지 30℃)에서 교반하고, 디클로로메탄을 증발시켜 내부 W/O 에멀젼을 고화시키고, 이후에 마이크로캡슐을 원심분리하여 수집한다. 이 마이크로캡슐을 증류수에 분산시키고, 더욱 원심분리시켜 과량의 약물과 폴리비닐알콜을 세정제거한다. 회수된 마이크로캡슐을 소량의 증류수에 현탁시킨다. 이 현탁액에 D-만니톨 1.5 g을 첨가하고 용해시킨다. 수득된 현탁액을 감압하에 동결 건조시켜 분말상 마이크로캡슐을 수득한다.
동결건조후, 수득한 분말상 마이크로캡슐을 감압하에 50℃에서, 바꿔말해 매트릭스 성분 락트산-글리콜산 공중합체의 Tmg보다 3℃ 높은 온도에서, 24, 48, 96 또는 120 시간동안 추가로 건조하여 분말상 서방성 마이크로캡슐을 제조한다.
실시예 1
인터페론 알파(40 조 IU 함유)의 수용액 200 ml에, 아연 아세테이트(이수화물)(200 mg/ml)의 수용액 1 ml와 1N 수산화나트륨 1 ml를 첨가하고, 혼합한 후 이 혼합물을 4℃에서 하룻밤 유지시킨다. 3,000 rpm에서 원심분리한 후, 불용성 복합체를 회수하고 동결건조하여 인터페론 알파의 조 아연염 약 200 mg을 수득한다.
디클로로메탄 4 ml중의 락트산-글리콜산 공중합체(락트산/글리콜산=50/50, 분자량 5,800, 와꼬 순수화학회사제) 1.5 g과 참고예 2에서 수득한 락트산-글리콜산 공중합체의 에틸 에스테르 1.5 g의 용액에, 상기에서 기술한 인터페론 알파의 조 아연염 200 mg을 첨가한 다음, 호모나이저(Polytron)를 사용하여 약 30 초간 교반하여 s/o 에멀젼을 수득한다. 이 에멀젼을 18℃로 미리 조정한 폴리비닐알콜(EG-40, 니뽄 합성 화학회사제)의 0.1%(w/w) 수용액 700 ml에 부은 다음, 6,000 rpm에서 터빈 호모나이저로 교반하여 s/o/w 에멀젼을 수득한다. 이 에멀젼을 실온에서 3시간 교반하여 디클로로메탄을 휘발시키고 오일상을 고화시킨다. 이후, 약 2,000 rpm(05PR-22, 히다찌사)에서 원심분리시킨 후, 상청액을버린다. 수득한 잔류물을 다시 증류수에 분산시키고 원심분리시킨다. 수집한 마이크로캡슐을 D-만니톨 50 mg의 존재하에서 소량의 증류수에 다시 분산시킨 후 분산액을 동결건조시켜 분말 마이크로캡슐을 수득한다.
실시예 2
락트산-글리콜산 공중합체(락트산/글리콜산=75/25(몰%), 중량평균 분자량 13,585, 수평균 분자량 4,413, 와꼬 순수화학회사제) 3.6 g을 디클로로메탄 6.6 g(5 ml)에 용해시킨다. 참고예 1에서 수득한 돼지 인슐린의 조 아연염(200 mg의 돼지 인슐린 함유) 420 mg을 디클로로메탄 6.6 g(5 ml)에 분산시킨다. 이 두 성분을 혼합하고 호모나이저(Polytron)에서 약 10 초간 교반시켜 s/o 에멀젼을 수득한다. 이 에멀젼을 18℃로 미리 조정한 폴리비닐알콜(EG-40, 니뽄 합성 화학회사제)의 0.1%(w/w) 수용액 800 ml에 부은 다음, 6,000 rpm에서 터빈 호모나이저로 교반하여 s/o/w 에멀젼을 수득한다. 이 에멀젼을 실온에서 3시간 교반하여 디클로로메탄을 휘발시키고 오일상을 고화시킨다. 이후, 약 2,000 rpm(05PR-22, 히다찌사)에서 원심분리시킨 후, 상청액을 버린다. 수득한 잔류물을 다시 증류수에 분산시키고 원심분리시킨다. 수집한 마이크로캡슐을 D-만니톨 50 mg의 존재하에서 소량의 증류수에 다시 분산시킨 후 분산액을 동결건조시켜 분말 마이크로캡슐(약 3 g 회수)을 수득한다.
이렇게 수득한 마이크로캡슐의 인슐린 함량을 측정하기위해, 상기 분말을 3 시간동안 30% 아세토니트릴을 함유한 50 mM EDTA 용액과 흔들어 추출한 다음, 고속 액체 크로마토그래피(HPLC)로 정량분석한다. 마이크로캡슐 100 mg당 6.2 mg의인슐린이 함유되어 있었다.
실시예 3
에리트로포이에틴 주사 용액(EspoTM주사 3000, 상꾜 제)(12,000 IU 함유) 8 ml에 염화아연 1 g을 조금씩 첨가한 후, 이 혼합물을 실온에서 1시간 유지시킨다. 이 혼합물을 3,000 rpm에서 원심분리시킨 후, 침전물을 다시 증류수에 분산시키고 원심분리시켜 침전물을 수득한다. 이 침전물에 소량의 증류수를 첨가한 후, 동결건조시켜 에리트로포이에틴의 조 아연염과 알부민의 조 아연염의 혼합물 60 mg을 분말로서 수득한다.
디클로로메탄 1.5 ml중의 락트산-글리콜산 공중합체(락트산/글리콜산=50/50, 분자량 14,000, 와꼬 순수화학회사제) 0.5 g의 용액에, 상기에 기재된 에리트로포이에틴의 조 아연염과 알부민의 조 아연염의 혼합물 60 mg을 첨가한 다음, 호모나이저(Polytron)를 사용하여 약 30 초동안 교반시켜 s/o 에멀젼을 수득한다. 이 에멀젼을 실시예 1과 동일한 방법으로 처리하여 분말 마이크로캡슐 152 mg을 수득한다.
실시예 4
인간 성장 호르몬(GenotropinTM161U, 스미또모제약 제조)를 증류수 1 ml에 용해시킨다. 이 용액에, 염화아연 수용액(10 mg/ml)의 100 μl을 첨가한 후, 이 혼합물을 실온에서 1시간 유지시킨다. 이 혼합물을 원심분리시킨 후, 침전물을 다시 증류수에 분산시키고 원심분리시켜 침전물을 수득한다. 이 침전물에소량의 증류수를 첨가한 후, 동결건조시켜 인간 성장 호르몬의 조 아연염 5.6 mg을 분말로서 수득한다.
디클로로메탄 1.5 ml중의 락트산-글리콜산 공중합체(락트산/글리콜산=75/25, 분자량 9,800, 와꼬 순수화학회사제) 0.5 g의 용액에, 상기에 기재된 인간 성장 호르몬의 조 아연염 5.6 mg을 첨가한 다음, 호모나이저(Polytron)를 사용하여 약 30 초동안 교반시켜 s/o 에멀젼을 수득한다. 이 에멀젼을 실시예 1과 동일한 방법으로 처리하여 분말 마이크로캡슐 121 mg을 수득한다.
실시예 5
과립구 콜로니 자극 인자(G-CSF) 주사용액(Filgrastin Neupogen 상표명, Amgen, USA) 10 ml(3×108IU 함유)를 수산화나트륨의 희석 수용액으로 중화시킨 후, 염화아연 수용액(10 mg/ml)의 1 ml를 첨가하고, 이 혼합물을 실온에서 1시간 유지시킨다. 이 혼합물을 원심분리시킨 후, 침전물을 다시 증류수에 분산시키고 원심분리시켜 침전물을 수득한다. 이 침전물에 소량의 증류수를 첨가한 후, 동결건조시켜 과립구 콜로니 자극 인자의 조 아연염 4 mg을 분말로서 수득한다.
디클로로메탄 1.5 ml중의 락트산-글리콜산 공중합체(락트산/글리콜산=50/50, 분자량 8,000, 와꼬 순수화학회사제) 0.5 g의 용액에, 상기에 기재된 과립구 콜로니 자극 인자의 조 아연염 4 mg을 첨가한 다음, 호모나이저(Polytron)를 사용하여 약 30 초동안 교반시켜 s/o 에멀젼을 수득한다. 이 에멀젼을 실시예 1과 동일한 방법으로 처리하여 분말 마이크로캡슐 110 mg을 수득한다.
실시예 6
사람 인슐린(사람 재조합 인슐린, 와꼬 순수 화학회사로부터 구입) 5.21 mg(26 U/mg)을 57 mM 염산 수용액 0.63 ml에 용해시킨 후, 수산화나트륨 0.05 N 수용액 0.35 ml를 첨가하여 거의 중성 pH의 사람 인슐린 용액을 수득한다. 이 사람 인슐린 용액에, 아연 아세테이트의 수용액(20 mg/ml) 0.2 ml를 첨가하고, 이 혼합물을 4℃에서 하릇밤 유지시킨다. 이 혼합물을 약 3,000 rpm에서 원심분리시킨 후, 침전물을 다시 증류수에 분산시키고 원심분리시켜 침전물을 수득한다. 이 침전물에 소량의 증류수를 첨가한 후, 동결건조시켜 사람 인슐린의 조 아연염 11 mg을 분말로서 수득한다.

비교예
디클로로메탄 1.5 ml중의 락트산-글리콜산 공중합체(락트산/글리콜산=50/50(몰%), 중량평균 분자량 6,000, 와꼬 순수화학회사제) 0.9 g의 용액에, 실질적으로 아연이 없는 사람 인슐린[아연 함량이 0.0001% (w/w)이하] 100 mg을 첨가한 다음, 호모나이저(Polytron)를 사용하여 약 10 초동안 교반시켜 s/o 에멀젼을 수득한다. 이 에멀젼을 실시예 1과 동일한 방법으로 처리하여 분말 마이크로캡슐 (470 mg)을 수득한다.
이렇게 수득한 마이크로캡슐의 인슐린 함량을 측정하기위해, 상기 분말을 3 시간동안 아세토니트릴을 함유한 50 mM EDTA 용액과 흔들어 추출한 다음, 고속 액체 크로마토그래피(HPLC)로 정량분석한다. 마이크로캡슐 100 mg당 8.7 mg의 인슐린이 함유되어 있었다.
실험예 1
실시예 2에서 수득한 분말 마이크로캡슐 323 mg을 주사용 분산제 (증류수 1 ml에 용해된 5 mg의 카르복실메틸 셀룰로스, 폴리소르베이트 80 1 mg 및 만니톨 50 mg) 1 ml에 분산시킨다. 수득한 분산액을 6주된 수컷 SD 쥐의 등에 피하 투여한다(쥐당 약 20 mg의 인슐린 투여). 투여후, 혈액을 일정한 간격으로 꼬리를 통해 수집하여, 효소 면역측정(EIA) 키트(상꾜 제약제조)를 사용하여 혈청 돼지 인슐린 농도를 측정하였다. 투여후 1 주일 이상동안 혈청에서 활성 돼지 인슐린이 검출되었다.
실험예 2
실시예 4에서 수득한 분말 마이크로캡슐 70 mg을 주사용 분산제 (증류수 1리터에 용해된 5 g의 카르복실메틸 셀룰로스, 폴리소르베이트 80 2 g 및 만니톨 50 g) 0.5 ml에 분산시킨다. 수득한 분산액을 6주된 수컷 SD 쥐의 등에 피하 투여한다(쥐당 약 3 mg의 성장 호르몬 투여). 투여후, 혈액을 일정한 간격으로 꼬리를 통해 수집하고, 방사성 면역측정법에 의해 혈청 성장 호르몬 농도를 측정하였다. 투여후 1 주일 이상동안 혈청에서 활성 성장 호르몬이 검출되었다.
비교 실험예
비교예에서 수득한 분말 마이크로캡슐 154.7 mg을 주사용 분산제 (증류수 1 ml에 용해된 5 mg의 카르복실메틸 셀룰로스, 폴리소르베이트 80 1 mg 및 만니톨 50 mg) 1.75 ml에 분산시킨다. 수득한 분산액을 6주된 수컷 SD 쥐의 등에 피하 투여한다(쥐당 약 44 mg의 인슐린 투여). 투여후, 혈액을 일정한 간격으로 꼬리를 통해 수집하여, 효소 면역측정(EIA)에 의해 사용하여 혈청 인슐린 농도를 측정하였다. 투여후 단지 1일만에 혈청에서 활성 인슐린이 검출되었다.
본 발명에 따라, 아주 효과적으로 생리활성물질이 첨가되어 있으며, 초기 생리활성물질의 급격한 방출을 억제하는 서방성 제제를 제공할 수 있다. 본 발명의 서방성 제제는 생체내에서 투여후 그의 생활성을 유지하면서 생리활성물질을 방출할 수 있다. 더욱이, 서방성 제제에서 생리활성물질은 생활성을 거의 잃지않으면서 장기간 동안 안정하게 유지될 수 있다.
Claims (28)
- (a) 산성기를 갖는, 엔도테린 길항제를 제거한 수용성 펩티드유형의 생리활성물질 또는 그 수용성 염과 수용성 다가금속염의 결합으로 형성되는, 20℃에서의 수중의 용해도가 0 내지 0.1%(w/w)인 상기 수용성 펩티드 유형의 생리활성물질의 수불용성 또는 수난용성 다가금속염, 및(b) 생분해성 폴리머를 함유하고 있는 서방성 제제.
- 제 1 항에 있어서, 생리활성물질이 수용성 펩티드 또는 그의 유도체인 것을 특징으로 하는 제제.
- 제 2 항에 있어서, 펩티드가 호르몬, 사이토카인, 조혈 인자, 성장 인자, 효소, 가용성 또는 가용화된 수용체, 항체, 항원 함유 펩티드, 혈액 응고 인자 또는 접착 분자인 것을 특징으로 하는 제제.
- 제 2 항에 있어서, 펩티드가 호르몬인 것을 특징으로 하는 제제.
- 제 4 항에 있어서, 호르몬이 성장 호르몬인 것을 특징으로 하는 제제.
- 제 4 항에 있어서, 호르몬이 인슐린인 것을 특징으로 하는 제제.
- 제 2 항에 있어서, 펩티드가 사이토카인인 것을 특징으로 하는 제제.
- 제 7 항에 있어서, 사이토카인이 인터페론인 것을 특징으로 하는 제제.
- 제 2 항에 있어서, 펩티드가 성장 인자인 것을 특징으로 하는 제제.
- 제 1 항에 있어서, 다가 금속염이 전이 금속염인 것을 특징으로 하는 제제.
- 제 1 항에 있어서, 다가 금속염이 아연 염인 것을 특징으로 하는 제제.
- 제 1 항에 있어서, 다가 금속염의 수용해도가 20℃에서 약 0 내지 약 0.1%(w/w)인 것을 특징으로 하는 제제.
- 제 1 항에 있어서, 다가 금속염의 수용해도가 약 0 내지 약 0.01%(w/w)인 것을 특징으로 하는 제제.
- 제 1 항에 있어서, 약 0.1 내지 약 50%(w/w)의 다가 금속염을 함유하고 있는 것을 특징으로 하는 제제.
- 제 1 항에 있어서, 약 1 내지 약 30%(w/w)의 다가 금속염을 함유하고 있는 것을 특징으로 하는 제제.
- 제 1 항에 있어서, 생분해성 폴리머가 지방족 폴리에스테르인 것을 특징으로 하는 제제.
- 제 16 항에 있어서, 지방족 폴리에스테르가 락트산과 글리콜산의 중합체인 것을 특징으로 하는 제제.
- 제 17 항에 있어서, 락트산과 글리콜산의 조성비가 100/0 내지 약 40/60(몰%)인 것을 특징으로 하는 제제.
- 제 18 항에 있어서, 조성비가 약 90/10 내지 약 45/55(몰%)인 것을 특징으로 하는 제제.
- 제 17 항에 있어서, 중합체의 중량 평균 분자량이 약 3,000 내지 약 20,000인 것을 특징으로 하는 제제.
- 제 17 항에 있어서, 중합체의 중량 평균 분자량이 약 3,000 내지 약 14,000인 것을 특징으로 하는 제제.
- 제 16 항에 있어서, 지방족 폴리에스테르가 락트산의 단일 중합체인 것을 특징으로 하는 제제.

- 산성기를 갖는, 엔드세린 길항제를 제거한 수용성 펩티드유형의 생리활성물질 또는 그 수용성 염과 수용성 다가금속염의 결합으로 형성되는, 20℃에서의 수중의 용해도가 0 내지 0.1%(w/w)인 상기 수용성 펩티드유형의 생리활성물질의 수불용성 또는 수난용성 다가금속염을 생분해성 폴리머를 함유하고 있는 유상(油相)으로 분산시켜 오일에 분산된 고체 에멀젼(s/o형 에멀젼: solid-in-oil emulsion)을 제조하고, 상기 s/o형 에멀젼을 수상(水相)으로 첨가하여 수중유에 분산된 고체 에멀젼 (s/o/w형 에멀젼: solid-in-oil-in water emulsion)을 제조한 후, 상기 s/o/w형 에멀젼을 수중 건조시키는 것을 포함하는 서방성 제제의 제조방법.
