Description
本出願は、2011年6月2日に出願された米国仮特許出願第61/492,626号に対する優先権を主張し、その全内容は、参照により本明細書に組み込まれる。

本発明は、有効な量のナノ粒子を含む新生物特異的免疫原性組成物を提供し、非細胞性材料を含む内核と、新生物細胞に由来する細胞膜を含む外面と、任意に合成膜も含む。ある実施形態において、細胞膜は良性新生物細胞、潜在的に悪性の新生物細胞、または癌細胞に由来する。ある実施形態において、細胞膜は、癌細胞株に由来する。他の実施形態において、細胞膜は、対象の癌細胞に由来する。本発明の新生物特異的免疫原性組成物は、ナノ粒子の外面中の細胞膜が、新生物細胞に対する免疫応答を誘発するために、その構造的完全性を実質的に保持する。本明細書で使用されるとき、構造的完全性は、細胞膜の一次、二次、三次、もしくは四次構造、またはその構成素を含む。

乳酸−グリコール酸共重合体(PLGA)粒子を、予形成されたRBC膜由来の小胞とともに押出することにより、発明者らは、100nmより小さいポリマー粒子を、脂質および対応する表面タンパク質の両方を含む二重層化RBC膜でコーティングする。このアプローチは、ポリマー核の適用性を保持しながら、長時間循環するために、ナノ粒子表面を赤血球外部で偽装することを目的とする。発明者らは、この生物模倣型ナノ粒子送達プラットフォームの物理的特徴、物理化学的特性、タンパク質含有量、薬物動態、および生分布を報告する。
赤血球細胞(RBC)ゴースト誘導。細胞内容を欠くRBCゴーストは、修正により以前に公開されたプロトコルに従って調製した(32)。最初に、ヘパリン溶液(Cole−Parmer、Vernon Hills,IL)の滴を含有するシリンジを使用する心穿刺を通じて、Charles River Laboratories(Wilmington,MA)から得られた雄ICRマウス(6〜8週間)から全血を採取した。次にその全血を2000rpmで5分間、4℃で遠心分離し、血清および白血球層を慎重に除去した。得られた被包されたRBCを、透析のための低張媒体処理に先立って、氷冷1X PBS中で洗浄した。洗浄したRBCを、0.25X PBS中に氷浴で20分間懸濁し、2000rpmで5分間遠心分離した。ヘモグロビンを除去する一方で、ピンク色の小球を収集した。位相差顕微鏡を使用して、得られたRBCゴーストを検証したところ、改変された細胞内容を持つ正常な細胞構造が明らかになった(図5)。

赤血球細胞膜(RBCm)により被覆されたポリマーナノ粒子(NP)は、長い循環寿命と、制御された薬剤保持および放出という複合利点を付与する。高度な送達アプリケーションのためのこの細胞模倣型NPプラットフォームの開発に向けて、本明細書において、発明者らは、その薬剤負荷、薬剤放出動態、および細胞に基づく有用性に関するより良い理解を得るために研究を行った。具体的に、RBCmで被覆されたNPからの薬剤放出を研究するために、発明者は、モデル抗癌剤であるドキソルビチン(DOX)をRBCmで被覆されたNPに負荷するための2つの方法:物理的カプセル化および化学共役を比較した。インビトロ有用性は、急性骨髄性白血病(AML)Kasumi−1細胞株を使用することにより調べた。
過去数十年にわたって、ナノメートルスケールの材料工学における進歩は、臨床適用における無数のナノ粒子(NP)ベースの薬剤送達系をもたらした[1,2]。これらのナノ医療に固有の利点、特に改変された薬物動態および生体分布プロファイルを通じた既存の治療薬剤に対するそれらの改善は、より長い期間の間、血流中を循環する能力による[3,4]。結果として、NPにマクロファージの取り込みおよび全身クリアランスを回避させることができる、天然および合成で作製される新規の材料の検索に相当の研究関心が集められている[5,6]。その中で、寸法、形状、変形、および表面特徴を含むNP物理化学的特性を修飾することにより、インビボの粒子滞留時間を延長させることを目的とする方法も広く探究されている[7,8]。
2.1.RBCゴーストの誘導
細胞内容を欠くRBCゴーストを、以前に公開されたプロトコルに従って調製した[9,10]。つまり、ヘパリン溶液(Cole−Parmer)の滴を含有するシリンジによる心穿刺を通じて雄ICRマウス(6〜8週間、Charles River Laboratories)から採取した全血を遠心分離し(800×gで5分間、4℃)、血清および白血球層を除去した。被包されたRBCを、氷冷1X PBS中で洗浄し、透析のために低張媒体処理した後、0.25X PBS中に氷浴で20分間懸濁した。懸濁液を800×gで5分間遠心分離することにより、ヘモグロビンを除去した。ピンク色の小球の形態のRBCゴーストを収集した。
収集したRBCゴーストを、蓋をしたガラス瓶中で5分間、FS30D音波洗浄器(Fisher Scientific)を使用して、周波数42kHzおよび電力100Wで音波処理した。後次に得られた小胞を、Avantiミニ押出機(Avanti Polar Lipids)を使用することにより、400nm、次に200nmのポリカーボネート多孔質膜を通じて連続して押出した。各押出後、RBC膜由来の小胞の寸法を、動的光散乱(DLS,Nano−ZS,モデルZEN3600)により監視した。
DOX−ポリ(ラクチド酸)(PLA)抱合体を、公開されたプロトコルに基づいて合成した[11,12]。つまり、L−ラクチド(Sigma−Aldrich,USA)の開環重合化を、アルゴンで満たされたグローブボックス中、室温でアルコキシ組み合わせ(BDI)ZnN(SiMe3)2により触媒した。(BDI)ZnN(SiMe3)2)(6.4mg、0.01mmol)およびDOX(Jinan Wedo Co.,Ltd.,Jinan,China)(5.4mg、0.01mmol)を、無水テトラヒドロフラン(THF、0.5mL)中で混合し、2mLの無水THF中に溶解したL−ラクチド(101mg、0.7mmol)を滴下添加した。1H NMR(Varian Mercury 400MHz分光計)により示されるように、L−ラクチドが完全に消費された後、粗生成物を冷たいジエチルエーテル中に沈殿させ、複数の溶解−沈殿サイクルにより精製した。1H NMRにより共役を確認し、抱合体は、ゲル透過クロマトグラフィ(GPC,Viscotek,USA)により決定されるように、約10,000g/molの分子量を有した。
最初に、DOX−PLA抱合体をアセトニトリルに溶解して1mg/mLの溶液を形成し、
1mLのかかる溶液を3mLの水に滴下添加した。次に混合液を外気中で2時間撹拌し、アセトニトリルを蒸発させた。得られたNP核の溶液を、Amicon Ultra−4遠心分離フィルタ(Millipore、10kDaカットオフ)により濾過した後、1mLの蒸留水中で再懸濁した。DOXを物理的にカプセル化するために、1mgの乳酸−グリコール酸共重合体(PLGA、0.67dL/g、カルボキシ末端、LACTEL Absorbable Polymers)を、最初に1mLのアセトニトリルに溶解し、続いて25μLのジメチルスルホキシド(DMSO)中に事前に溶解されたDOXを添加した。上記と同様の手順に従って、NP核を含有する懸濁液を生成した。
RBCm由来の小胞を前述のNP核と融合するために、1mgのNP核を含有する懸濁液を最初に、1mLの全血から調製されたRBCm由来の小胞と混合した。次に混合液を、Avantiミニ押出機を用いて、11回、100nmポリカーボネート多孔質膜を通じて押出した。1mgのNP核を完全にコーティングするために、血液の超過分を使用して、RBCゴーストの誘導および押出の間の膜喪失を補償した[9]。
重量比1:1のDOX−PLA抱合体およびPLA−PEG−COOH(10kDa,PDI=1.12)[13]を最初に、1mg/mLの濃度でアセトニトリル中に溶解し、上記と同一の手順に従って、NP懸濁液を生成した。DOXをPEG化NPに物理的にカプセル化するために、1mgの乳酸−グリコール酸共重合体(PLGA、0.67dL/g、カルボキシ末端、LACTEL Absorbable Polymers)を最初に、1mLのアセトニトリルに溶解し、続いて25μLのジメチルスルホキシド(DMSO)中に事前に溶解された100μgのDOXを添加した。上記と同一の手順を使用して、NP懸濁液を生成した。
DLSを使用して粒径を監視することにより、PBS中のNP安定性を評価した。具体的に、500μgのNPを1mLの1×PBS中に懸濁し、1週間の期間にわたって24時間毎に室温で3回寸法を測定した。測定の間に、試料をやさしく撹拌しながら37℃で培養した。560nmの波長での紫外線吸収度を監視することによりNP血清安定性を評価した。具体的に、最初にNPをPBS中で2mg/mLに濃縮し、続いて等容量の2×ウシ胎孔血清(FBS,Hyclone)を添加した。Infinite M200マルチプレートリーダーを37℃で、2時間の期間にわたって約1分毎に使用することにより、吸収度を測定した。
溶液中のDOX濃度を、480nmの励起波長を用いて、580nmでの蛍光強度を測定することにより決定した。NPのDOX負荷収率を決定するために、100μLのNP溶液をアセトニトリル中100μLの0.1M HClを用いて24時間培養した後、上記蛍光測定を行った。DOX放出プロファイルをプロットするために、200μLのNP溶液(1mg/mL)をSlide−A−Lyzer MINI透析微小管(Pierce,Rockford,IL、分子量カットオフ=3.5kDa)に入れ、次に2LのPBS(pH=7.4)に対して37℃で透析した。全体透析プロセスの間12時間毎に、PBS緩衝液を変えた。各規定の時点において、3つのミニ透析ユニットからのNP溶液を収集し、DOX濃度を測定した。
遊離DOXおよびDOX負荷したNPの細胞毒性を、MTTアッセイ(Promega Corporation,Madison,WI,USA)を使用して、急性骨髄性白血病(AML)患者の末梢血から確立したKasumi−1細胞株に対して評価した。最初に細胞を96ウェルプレートに播種し(約5×103/ウェル)、次に24時間培養した。遊離DOXおよびDOX負荷したNPを添加した後、細胞をさらに72時間培養した。次に製造者により提供されたプロトコルに従って、MTTアッセイを使用することにより細胞生存性を決定した。
3.1.RBCmで被覆されたNPの調製
RBCmで被覆されたNPの調製プロセスは、以前に公開されたプロトコルに基づき、図9に概略的に示される[9]。つまり、精製されたRBCは最初に、低張性環境において膜破裂を受けて、その細胞内容を除去した。次に、空のRBC(直径約2μm)を洗浄し、100nm多孔質膜を通じて押出して、RBC膜由来の小胞(直径約200nm)を形成した。一方PLAまたはPLGAから作製されたもの等のポリマー核(直径約70nm)は、溶媒置換法を使用することにより調製した。得られたポリマー核を、RBC膜由来の小胞と後次に混合し、混合物を100nmの孔を通じて物理的に押出して、2つの構成成分が機械力な力の下で融合し、RBCmで被覆されたNP(直径約90nm)を形成した。
この研究において、発明者らは、モデル薬剤としてのDOXをRBCm被覆されたNPに負荷するための2つの別箇の方法:物理的カプセル化および化学共役を調べた。物理的カプセル化は、最初にアセトニトリル中でDOXおよびポリマーを混合し、続いて水に沈殿させることにより達成する。この場合、薬剤負荷収率は、異なる製剤パラメータを通じて変動し得る。例えば、初期DOX:PLGA重量比を5%から20%に変動させるとき、負荷収率は0.9%から1.8%に増加した( 図10を参照)。
次に、発明者は、生理学的に関連する緩衝液中のDOX負荷したRBCmで被覆されたNPの安定性を研究した。不安定な粒子は凝集する傾向があり、それらの寸法が増加するため、PBS中のNP安定性は、異なる辞典でNP寸法を測定することにより監視する。この研究において(図11A)、物理的カプセル化および化学共役の両方を使用することによりDOX分子を負荷したNPは、1週間の期間にわたって著しく寸法が増大することなく、約90nmの類似の初期直径を有した。同様に、同一の期間にわたって、NPのPDIにおけるわずかな変化のみが観察され、PBS中のDOX負荷されたRBCm被覆されたNPの高い安定性を示す。粒子凝集の程度を反映する特徴的な波長である、560nmでの紫外線吸収度を監視することにより、血清中のNP安定性をさらに調べた[14,15]。物理的カプセル化または化学共役のいずれかによりDOX分子を負荷したRBCmで被覆されたNPは、2時間の期間にわたって、560nmでほぼ一定の吸収度を示し(図11B)、NPが100%ウシ胎孔血清(FBS)中で高度に安定することを示唆する。対照的に、RBCm被覆のないPLGAまたはPLA−DOX抱合体から作製された裸ポリマー核の吸収は、FBSへの添加時に即時に増加した。これらの結果は、RBCm被覆が、緩衝液および血清の両方においてNPを安定化させる際に重要な役割を果たすことを示した。実践的な観点から、緩衝液中のコーティングされていないポリマー粒子の高速凝集は、選択的な沈殿およびそれらの沈殿後のRBCmで被覆されたNPからのコーティングされていない粒子の除去を提供した。
安定したDOX負荷したRBCmで被覆されたNPの製剤に続いて、発明者らは、それらのDOX放出動態の調査に進んだ(図12)。発明者らは最初に、異なる薬剤負荷機構がRBCmで被覆されたNPからのDOX放出にどのように作用するかを調べた。結果は、DOX分子の20%が最初の2時間以内にRBCmで被覆されたNPから放出されたため、DOX分子がポリマーマトリックスに物理的にカプセル化されたときに、薬剤放出率が著しく速くなることを示した。対照的に、化学共役の製剤を調べたとき、最初の2時間以内に、DOX分子の5%のみが放出した。かかる差異は、DOX分子のポリマー骨格への共有結合が、ポリマーマトリックスの外に拡散して放出し得る前に、バルク浸食により最初に薬剤分子がポリマーから水和される必要があるという事実に起因した[11,12,16]。薬剤−ポリマー共有結合から得られるより持続性の放出プロファイルは、環境トリガに応答する化学リンカーが、高度の薬剤送達適用のためのRBCmで被覆されたNPを開発するときに、より良好に制御される薬剤放出を達成し得ることを示唆する[13,17]。
最後に、発明者らは、Kasumi−1細胞株に対するDOX負荷したRBCmで被覆されたNPの治療能力を調べた。血流中の白血球ブラストの未制御成長および蓄積を特徴とする疾病であるAMLを、血流中のRBCmで被覆したNPの長い循環寿命およびそれらの持続性薬剤放出プロファイルに起因して、疾患標的として選択した。AMLに対する現在の治療標準は、高用量のアントラサイクリンであり、心毒性に対する深刻な懸念を引き起こす[28]。持続性様式で治療化合物を放出する長期循環NPは、必須用量を低減し、治療有用性を改善する機会を提供する。DOXが物理的に負荷されるか、または共有結合されるかのいずれかであるRBCmで被覆されたNPは、72時間の培養期間にわたって遊離DOXと比較すると、より高い毒性を呈した(図13)。有用性のこの強化は、NPの細胞内取り込みに起因する可能性があり得、高い薬剤の積荷が細胞内領域に進入することを可能にする[29]。対照的に、遊離DOXは、細胞進入の受動的膜拡散に依存し、膜結合された薬剤流出ポンプの効率が低く、その影響を受けやすい[30〜32]。この研究は、延長した循環寿命、持続性薬剤放出、および改善された細胞内在化を伴うRBCmで被覆されたNPが、血液癌の治療に対するプラットフォームであることを示唆する。さらなる研究は、これらのNPのインビボの治療能力を調べることを保証する。
要約すると、本明細書において、発明者らは、薬剤をRBCmで被覆されたNP送達系に負荷するための2つの方法:物理的カプセル化および化学共役を調べた。放出研究は、化学共役法が、より持続性の高い薬剤放出プロファイルをもたらしたことを示唆した。発明者らは、RBCmで被覆したNPと比較して、同一のNP核を有するが、異なる表面コーティングを有するPEG化NPをさらに製剤した。これら2つの送達系の薬剤放出プロファイルを比較することにより、発明者らは、RBCm被覆が、カプセル化された薬剤分子の外側拡散を減速させる障壁を提供することを実証した。これらの結果は、NP核中の薬剤−ポリマー結合に対する化学修飾およびNP表面コーティング上の設計が、RBCmで被覆されたNPの薬剤放出に対してより優れた制御を得ることができることを提供する。AML Kasumi−1細胞株を使用することによる以下の有用性研究において、RBCmで被覆されたNPは、遊離DOXと比較して、より高い毒性を呈した。以前に観察された血流中の長い全身性循環寿命および本明細書で報告される持続性薬剤放出動態は、この生体模倣型薬剤送達系が、血液癌等の様々な疾患の治療に対して、実行可能な積荷の全身性送達を提供することを示す。これらのRBCmで被覆されたNPは、全身性ポリマーおよび天然の細胞膜の両方の利点を組み合わせる、頑強な薬剤送達系を提供する。
RBCの長い循環寿命とポリマー粒子からの制御された薬物保持および放出という利点を組み合わせるために、発明者らは、PLAまたはPLGAから作製された100nmより小さいポリマー核、および保存膜タンパク質を持つRBCmから作製された赤血球外部を含有する、100nmより小さい寸法のRBCmで被覆されたNPを製剤した。
発明者らは、RBCmで被覆されたNPに対するモデル薬剤としてDOXを負荷するための2つの別箇の方法を調べたところ、物理的カプセル化は、0.9%〜1.8%の範囲の負荷収率をもたらし、共有結合は、5%の近似負荷収率をもたらした。
NP寸法および紫外線吸収度を監視することにより、発明者らは、RBCmで被覆されたNPが、RBCm被覆のない裸ポリマー核と比較したときに、優れた安定性を有することがわかり、RBCm被覆が、生物学的溶液中でNPを安定化させる際に重要な役割を果たすことを示唆する。
放出研究は、薬物−ポリマー共有結合アプローチが、物理的カプセル化より持続性の高い放出プロファイルを有することを示し、環境的トリガに応答する化学リンカーが、高度な薬剤送達適用のためのRBCmで被覆されたNPを開発するときに、より良好に制御された薬剤放出を達成し得ることを実証する。
RBCmで被覆されたNPをPEG化NPと比較することにより、発明者らは、RBCmがDOX放出の拡散障壁として作用することを見出した。この観察は、Higuchi式を使用する定量分析と一致した。したがって、脂質膜コーティングを設計することを目的とする方法は、ある環境条件下で、RBCmで被覆されたNPからの応答性薬剤放出も可能にし得る。
DOX負荷したRBCmで被覆されたNPは、遊離DOXと比較して、AML Kasumi−1細胞に対する有用性を強化した。有用性のこの強化は、NPの細胞内取り込みに寄与する可能性があり得、細胞内領域に進入する薬剤の高い積荷を可能にする。

本実施例は、既存のアプローチを越えるいくつかの利点を有する、免疫治療系を提供する。
1) 癌細胞は、患者の切除された腫瘍または一般的な癌細胞株に由来する。
2) 膜材料は、断片化等の方法を使用して、細胞に由来する。例は以下のとおりである。
− 癌細胞を機械的に均質化して、膜を破裂させる。
− ホモジネートを遠心沈殿させて、細胞内容を小球化し、膜を有する上澄みを収集する。
3) ナノ沈殿等の方法を使用して、アジュバントを負荷したナノ粒子を調製する。例は以下のとおりである。
− ポリマー(例えば、PLGA)およびアジュバント(例えば、モノホスホリル脂質A)を有機相に溶解する。
− 有機相を水性相にナノ沈殿させて、所望の寸法のナノ粒子を形成する。
4) 最終免疫治療粒子は、以下のように作製される。
− 癌細胞から収集された膜材料を物理的に押出して、より小さい膜小胞を作製する。
− 予形成されたアジュバント負荷された核を、小胞とともに押出して、最終粒子を形成する(図15)。
治療薬投与:
1) ナノ粒子製剤を非か投与する。
2) あるいは治療薬を静脈内投与する。
作用機構(図16):
1) 患者に注射するとき、一次免疫応答が引き起される。
2) APCは炎症部位に移行し、粒子を取り込む。
3) 取り込み時に、粒子は分解され、免疫原性アジュバントを放出する。
4) 免疫原性アジュバントを検出するときに、APCは成熟する
5) ナノ粒子表面上に存在した抗原材料は、ここでAPCの外部上に提示される。
6) 成熟したAPC上の抗原は、CD8+T細胞に提示される。
7) 癌特異的抗原と相互作用するときに、CD8+T細胞は活性化し、細胞毒性T細胞になる。
8) 癌細胞抗原に対する細胞毒性T細胞は増殖し、腫瘍を攻撃する。

抗毒素治療は、細菌感染、有毒な傷害、および生物学的兵器を含む、多数の健康障害の根底にある病原性因子の集団を一層する可能性を提供する。しかし、抗毒素の開発において益々の努力が成されているにもかかわらず、安全かつ有効な治療オプションは依然として限定されている。本明細書において、発明者らは、生物模倣型ナノスポンジを構成し、黄色ブドウ球菌からのα毒素を吸収および中和するその能力を実証する。ポリマーナノ粒子に支持されるRBC膜二重層で構成される、これらのナノスポンジは、膜を損傷する毒素に容易に取り込まれ、それらの細胞標的から逸れてしまう。マウスモデルにおいて、ナノスポンジは、毒素の毒性を顕著に低減させる。この生物学的に触発されたナノ製剤は、抗毒素治療のためのナノ医療における進歩を提示する。
毒素ナノスポンジの調製
ナノスポンジ粒子は、以前に報告されているように合成した(2)。RBCを単離するために、6週齢の雄ICRマウス(Charles River Laboratories)から採取した全血を800×gで5分間遠心分離した。次にRBCを低張処理に供し、RBCゴーストを800×gで5分間遠心分離することにより収集した。得られたゴーストは、ミニ押出機(Avanti Polar Lipid)を使用して、400nmおよび100nmポリカーボネート多孔質膜を通じて連続的に押出した。PLGAポリマー核は、溶媒置換プロセスを使用して、0.67dL/gカルボキシ末端50:50ポリ(-DL−ラクチド−co−グリコリド)(LACTEL Absorbable Polymers)を用いて同時に調製した。PLGAをアセトニトリル中に1mg/mLで溶解した。1mgの粒子を形成するために、1mLのPLGA溶液を3mLの水に滴下添加した。得られた混合液を外気中で2時間攪拌し、10kDa分子量カットオフ Amicon Ultra−4遠心分離フィルター(Millipore)を使用して濃縮した。最終RBCナノスポンジを、100nmポリカーボネート膜を通じて新鮮な血液(1mg PLGA/1mL血液)から調製された小胞を持つPLGAナノ粒子を押し出すことにより合成した。窒素パージを使用して、必要に応じてナノ粒子を濃縮した。PLGAポリマーの重量を、ナノスポンジに引用されるすべての後次質量値に使用する。ナノスポンジの寸法は、Malvern ZEN3600を使用して、3回の反復動的光散乱から得られ、平均寸法85nmを示した。
100μgのRBCナノスポンジを、3μgの黄色ブドウ球菌α毒素(Sigma Aldrich)で15分間培養した。ナノ粒子溶液の滴を、4μg/mLのナノスポンジ濃度で、グロー放電された炭素コーティングされたグリッドの上に蒸着させた。蒸着から1分後、液滴を10滴の蒸留水で洗い流し、1%酢酸ウラニルで染色した。試料をFEI Sphera Microscopeを使用して200kVで撮像した。
PEG化ナノ粒子は、ナノ沈殿法に従って調製した。つまり、1mgのPEG−PLGAジブロックコポリマーを、1mLのアセトニトリルに溶解し、常に攪拌しながら3mLの水を含有するバイアルに添加した。次に有機溶媒をフード内で2時間蒸発させた。次にAmicon Ultra−4遠心分離フィルタ(Millipore)を使用して、分子量カットオフ10kDaでNP溶液を3回洗浄した。PEG化リポソームは、機械的押出から調製した。つまり、1mgのEgg PCおよび200μgのDSPE−PEG−カルボキシ(Avanti Polar Lipids)を1mLのクロロホルムに溶解した。次に有機溶媒を蒸発させて、乾燥脂質フィルムを形成した。脂質フィルムを1mLのPBSで再水和し、続いて1分間渦流させ、3分間FS30D音波器(Fisher Scientific)中で音波処理した。後次に、狭く分布するリポソームを形成するために、製剤を100nm孔寸法のポリカーボネート膜を通じて11回押出した。RBC膜小胞は、ナノスポンジ調製について記載される、RBC精製および膜押出プロトコルに従って調製した。ナノ製剤の寸法は、動的光散乱を使用し、3回測定を繰り返すことにより得られ、PEG化PLGAナノ粒子、PEG化リポソーム、およびRBC膜小胞についてそれぞれ90、105、および120nmの平均寸法を示した。
3μgのα毒素を、1mg/mLのRBCナノスポンジ、PEG化PLGAナノ粒子、PEG化リポソーム、およびRBC膜小胞を含有する200μLのPBS溶液で30分間培養した。負の対照をPBS中の9μgのα毒素で調製した。次に溶液を、1.8mLの5%精製されたマウスRBCでさらに30分間培養した。培養に続いて、各試料を、Beckman Coulter Microfuge(登録商標)22R遠心分離器内で10分間、14,000rpmで沈降させた。上澄み中のヘモグロビンの吸収度を、Tecan Infinite M200マルチプレートリーダーを使用して、540nmで分析し、RBC溶解の程度をアッセイした。実験は三回重複した行った。
9μgのα毒素を、1mg/mLのRBCナノスポンジ、PEG化PLGAナノ粒子、PEG化リポソーム、およびRBC膜小胞を含有する200μLのPBS溶液で30分間培養した。培養後、Sepharose(登録商標)CL−4Bを通じて試料を濾過し、非結合毒素を除去した。次に試料を凍結乾燥し、SDS試料緩衝液(Invitrogen)中で調製した。9μgの純粋なα毒素を、参照として濾過された試料とともに調製した。調製した試料を、Novex(登録商標)XCell SureLock Electrophoresis System(Invitrogen)を使用して、MOPS泳動緩衝液中の4〜12%ビス−トリス10−ウェルミニゲル上で分離した。試料を200Vで50分間泳動させ、得られたポリアクリルアミドゲルをSimplyBlue(Invitrogen)中で一晩染色して可視化した。毒素保持を定量するために、ImageJを使用し、0.3、1、3、および9μgの純粋なα毒素から調製された毒素標準を用いて、34kDaでの帯域強度を分析した。
PBS中の200μLの1mg/mLナノスポンジを、30、9、3.6、1.2、0.6、および0.3μgのα毒素で30分間培養した。対照群として、同一濃度のα毒素を含有する溶液もナノスポンジの不在下で調製した。次に試料および対照溶液を、PBS中の1.8mLの5%マウスRBCで30分間培養した。各試料は、14,000rpmで10分間沈降させた。上澄み中のヘモグロビンの吸収度を540nmで分析し、RBC溶解の程度をアッセイした。実験を三回重複して行った。
PBS溶液を、200、60、20、6、および2μgの様々な量の毒素ナノスポンジを含有するように調製した。各ナノスポンジ溶液を、9μgのα毒素と混合し、最終容量200μLに希釈した。30分間の培養に続いて、溶液をPBS中の1.8mLの5%マウスRBCに添加し、30分間培養した。次に溶液を14,000rpmで10分間沈降させた。上澄み中のヘモグロビンの吸収を540nmで分析して、RBC溶解の程度をアッセイした。実験を三回重複して行った。
細胞取り込み後のナノスポンジおよびRBC膜小胞の膜材料を調べるために、10μgのDMPE−ローダミン(Avanti Polar Lipids)を、膜小胞に機械的に押出する前に、1mLの全血に由来するRBCゴーストに添加した。得られる染料負荷されたRBC膜小胞を使用して、ナノスポンジを調製した。蛍光ナノスポンジおよび膜小胞を、1時間、ヒト臍帯静脈内皮細胞(HUVEC)(ATCC #CRL−1730)で、Hanks′BSS、L−グルタミン、HEPES、および100U/mLペニシリン、100μg/mLストレプトマイシン(Invitrogen)、および50μg/mL内皮細胞成長補助剤(Biomedical Technologies,Inc.)を補充した1.4g/L NaHCO3(Lonza)含む培地199中、300μg/mLの濃度で培養した。次に媒質を吸引し、細胞を新鮮な媒質中で1時間培養した。第2の培養期間に続いて、細胞をPBSで洗浄し、10%ホルマリン(Millipore)で定着させて、DAPI含有Vectashield(登録商標)(Invitrogen)とともに装填した。Applied Precision DeltaVisionデコンボリューション走査蛍光顕微鏡上で60X油浸対物レンズを使用して、細胞を撮像した。
RBCナノスポンジを、α毒素を用いて、PBS中0.67mg/mLのナノスポンジおよび12μg/mLα毒素の最終濃度で15分間培養した。次に容量150μLの混合物を、6週齢の雌nu/nu裸マウス(Charles River Laboratories)の側腹領域に皮下注射した。注射後3日目に、マウスを撮像した。皮膚および筋肉試料を5μmに切断し、H&Eを使用して組織像を染色した。
濃度20mg/mLのRBCナノスポンジおよび濃度60μg/mLのα毒素を、蒸留水中で事前に調製した。前接種研究の場合、6週齢の雄ICRマウスに尾静脈を通じて、80mg/kg(体重当たりの用量)のナノスポンジに続いて、2分後に75μg/kgのα毒素注射を静脈内注射した。後接種研究の場合、最初に6週齢の雄ICRマウスに75μg/kgのα毒素を注射し、2分後に80mg/kgのナノスポンジを注射した。対照には、75μg/kgのα毒素溶液のみを注射した。各群の試料サイズは9であった。
