Claims (13)
- 蛍光色素を用いて生体関連分子を定量する分析装置であって、
支持基体と、この支持基体上に生体関連分子を捕捉した磁気微粒子を接着するために設けられた接着層と、蛍光色素を励起する励起手段と、蛍光を検出する検出手段と、前記支持基体に対して磁力のオン/オフ、あるいは磁力の強弱の切り替えが可能な磁場発生手段と、を備え、
前記接着層の材料として、熱や光に応答して疎水性と親水性を可逆的に変化させる物質を用いた分析装置。 - 請求項1において、
支持基体を移動させる可動ステージを設けた分析装置。 - 請求項1において、
支持基体上に流路を備え、該流路は送液ポンプを接続した取水口と排水口を備える分析装置。 - 請求項1において、
前記蛍光色素として量子ドットを用いる分析装置。 - 請求項1において、
前記接着層の材料として、アルキル鎖状分子、ビオチン分子のうちいずれかから選ばれる物質を用いた分析装置。 - 請求項1において、
前記磁場発生装置として、電磁石、可動式の永久磁石、可動式の磁場遮蔽物が組み合わさった電磁石、および可動式の磁場遮蔽物が組み合わさった永久磁石のうちいずれかを用いる分析装置。 - 請求項1において、
前記検出手段として、落射型光学顕微鏡を用いた分析装置。 - 請求項1において、
2種類以上の蛍光色素を同時に識別する落射型光学顕微鏡を用いた分析装置。 - 請求項1において、
前記生体関連分子が抗原分子であり、前記磁気微粒子による捕捉が抗原抗体反応によって実施される分析装置。 - 請求項1において、
前記生体関連分子が核酸分子であり、前記磁気微粒子による捕捉がハイブリダイゼーションによって実施される分析装置。 - 請求項1において、
前記蛍光色素が、解析対象の生体分子の種類ごとに、配合割合が異なった複数種類の蛍光体を含むことを特徴とする分析装置。 - 請求項1において、
特定の生体関連分子以外の生体関連分子に対しては同一の蛍光色素を用い、前記生体関連分子ごとに蛍光輝点数を計数した上で、総輝点数に対する特定の生体関連分子ごとの輝点数の比を算出することで、前記特定の生体関連分子ごとの存在量を評価することを特徴とする分析装置。 - 蛍光色素を用いて生体関連分子を定量する分析方法であって、
生体関連分子を捕捉した磁気微粒子を接着するための接着層が設けられた支持基体上に磁気微粒子を配し、磁力により、磁気微粒子を支持基体に衝突させ、磁力をオフ又は弱くし、蛍光色素を励起して、蛍光を検出する分析方法であって、
前記接着層の材料として、熱や光に応答して疎水性と親水性を可逆的に変化する物質を用いることを特徴とする、分析方法。
