制限酵素処理、アガロースゲル電気泳動、ゲル抽出キット(Gel Extraction Kit, QIAGEN)、プラスミドDNA精製、ポリメラーゼ連鎖反応(PCR)、DNA断片の結合、大腸菌の形質転換などの分子生物学で一般に用いる方法は、非特許文献6に紹介された方法に最小限の変更を加えて行った。
<2−1>pGL3−BAベクターの作製
CHO細胞から得た全ゲノムDNAを鋳型とし、配列番号1及び2のプライマーを用いてPCRを行うことにより、β−アクチンプロモーター遺伝子を増幅し、その増幅した産物をNheI及びHindIIIで切断した。こうして得られたβ−アクチンプロモーターDNA断片(3.0kb)を、同じ制限酵素で処理されたベクターpGL3−Basic(Promega)に挿入してpGL3−BAベクターを作製した(図3)。ここで、PCRは、94℃で5分間変性し、その後94℃で1分間の変性、55℃で1分間のアニーリング、及び72℃で3.5分間の重合の過程を25回繰り返し、最終的に72℃で7分間伸長した。

pcDNA3.1ベクター(インビトロジェン)を鋳型とし、配列番号4及び5のプライマーを用いてPCRを行うことにより、CMVプロモーターのTATAボックス領域(130bp)を増幅し(図4)、その増幅した産物をSalI及びHindIIIで切断した。こうして得られたTATAボックス領域DNA断片を、pGL3−BAベクターをXhoI及びHindIIIで切断した部位に挿入してpGL3−B/CTAベクターを作製した(図5)。ここで、作製されたベクターpGL3−B/CTAは、SalI及びXhoI制限酵素部位がない。ここで、前記PCRは、最初に94℃で5分間変性し、その後94℃で1分間の変性、55℃で1分間のアニーリング、及び72℃で2.5分間の重合の過程を25回繰り返し、最終的に72℃で7分間伸長した。

実施例2−1で作製されたpGL3−BAベクターを鋳型とし、配列番号6及び2のプライマーを用いてPCRを行うことにより、β−アクチンイントロン部分を増幅し、その増幅した産物をSacI及びHindIIIで切断してβ−アクチンイントロンDNA断片(1kb)を得た。また、実施例2−2で作製されたpGL3−B/CTAベクターをEcoRV及びSacIで切断してTATAボックス領域DNA断片(370bp)を得た。pGL3−B/CTAベクターを制限酵素EcoRV及びHindIIIで処理し、次いでβ−アクチンイントロンDNA断片(1kb)とTATAボックス領域DNA断片(370bp)を前記ベクターに挿入し、pGL3−B/CTA/Binベクターを作製した(図6)。ここで、作製されたpGL3−B/CTA/BinベクターにはSacI制限酵素が2つ存在する。前記PCRは、94℃で5分間変性し、その後94℃で1分間の変性、55℃で1分間のアニーリング、及び72℃で3.5分間の重合の過程を25回繰り返し、最終的に72℃で7分間伸長した。

Claims (18)
- CMV(Cytomegalovirus)エンハンサー、β−アクチンプロモーター、CMVプロモーターのTATAボックス、及び配列番号12の塩基配列からなるβ−アクチンイントロンを含むハイブリッドプロモーターであって、前記CMVエンハンサー、前記β−アクチンプロモーター、前記CMVプロモーター及び前記β−アクチンイントロンは、5‘から3’方向にこの順序で作動可能に連結されたハイブリッドプロモーター。
- 前記β−アクチンプロモーターが、
i)配列番号9で表されるヌクレオチド配列を有するDNA断片、
ii)配列番号10で表されるヌクレオチド配列を有するDNA断片、及び
iii)DNA断片i)及びii)のヌクレオチド配列において少なくとも1つのヌクレオチドの欠失、置換又は挿入を有するDNA断片であり、プロモーター活性、及びプロモーターの下流に作動可能に連結された標的遺伝子の発現調節活性を有するDNA断片とからなる群から選択される少なくとも1つのDNA断片からなるものである、請求項1に記載のハイブリッドプロモーター。 - 前記β−アクチンプロモーターがCHO(Chinese Hamster Ovary)細胞のβ−アクチンプロモーターである、請求項1に記載のハイブリッドプロモーター。
