Definitions
- the present invention relates to a CHO cell-derived protein secretion factor, a nucleic acid sequence encoding the protein secretion factor; and a gene encoding a target protein; an expression cassette operably linked to, an expression vector comprising the expression cassette, a transformant cell into which the expression vector is introduced, and a method for producing a target protein using the transformant cell is about
- Recombinant proteins can be mass-produced using microorganisms or animal cell systems through genetic recombination technology to produce useful protein components that are difficult to obtain in vivo.
- Recombinant proteins can be regulated to be expressed intracellularly or to be secreted extracellularly.
- intracellular expression often accumulates proteins as insoluble masses, and there are disadvantages in that productivity is lowered due to difficulties in separation and purification.
- extracellular secretion can readily yield soluble proteins with correct protein folding. Therefore, for more suitable extracellular secretion in the production yield and quality control of the protein, an optimized recombinant protein expression system is important.
- components such as host cells, target genes, expression vectors, selective markers, promoters, and signal peptide sequences are essential.
- the quality and productivity of the recombinant protein vary according to the selection of the components.
- the signal peptide In the case of the signal peptide, it is located at the N-terminal site of the recombinant protein to be produced, is involved in the expression level of the recombinant protein, and is a component that allows it to be secreted out of the cell. A difference in expression level is seen depending on which signal peptide is used, and the signal peptide is mis-cleaved and some signal peptide sequences remain at the N-terminus of the protein, which may affect the quality of the recombinant protein.
- a signal peptide which is a polypeptide consisting of a 17 to 31 amino acid sequence derived from a novel CHO cell.
- the present invention was completed by confirming that the signal peptide was 100% cleaved at the significantly increased expression level and the cleavage site to prevent erroneous cleavage.
- One object of the present invention is to provide a protein secretion factor consisting of the amino acid sequence of SEQ ID NO: 1, SEQ ID NO: 2 or SEQ ID NO: 3.
- Another object of the present invention is a nucleic acid sequence encoding a protein secretion factor consisting of the amino acid sequence of SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4 or SEQ ID NO: 5; and a gene encoding a target protein; to provide an expression cassette operably linked to.
