Description
(a)(i)発光部分を含んで成るプローブ;(ii)前記プローブに対して相補的であるかまたは部分的に相補的であるハイブリダイゼーション標的;および(iii)チアジン色素またはジアジン色素を含む可溶性発光修飾剤であって、前記発光部分をクエンチングできる前記可溶性発光修飾剤、を準備し;
(b)前記プローブと前記ハイブリダイゼーション標的とを、塩基対形成が起こり標的ハイブリダイゼーション複合体を形成できる条件下でアニーリングすること;
(c)前記可溶性発光修飾剤存在下で標的ハイブリダイゼーション複合体の温度を変化させ前記発光部分の放出を測定すること;
(d)前記発光部分の測定放出を前記標的ハイブリダイゼーション複合体の存在と温度関数として相関させ、それによって測定された放出に基づき標的ハイブリダイゼーション複合体のTmを決定すること、
を含んで成る方法を提供する。

(c)前記温度範囲にわたり試料からのシグナルを測定するための検出器;および
(d)前記検出器と作用可能に連結し、且つ前記温度範囲でシグナル測定値を受け取る相関モジュールであって、シグナル強度を前記プローブとハイブリダイゼーション標的を含むハイブリダイゼーション複合体の存在とチアジン色素またはジアジン色素との混合物中で温度関数として相関させ、それによって前記標的ハイブリダイゼーション複合体のTmを決定する相関モジュール、

本発明を詳細に説明する前に、本発明が特定のオリゴヌクレオチド、方法、組成物、キット、システム、コンピュータ、またはコンピュータ可読媒体に限定されず、もちろんそれらは変更可能であることを理解されたい。また、本明細書で使用した用語は、特定の態様のみを説明する目的のためであり、限定することを意図していない。さらに、特に断りがなければ、本明細書に記載の全ての技術的および科学的用語は、本発明が属する当該領域における当業者が通常理解する意味と同じ意味を有する。本発明の説明および請求において、下記の用語および文法上の変化も、下文に記載の定義に従い用いられることがある。
本発明は、標識核酸からの発光修飾のための簡易で優れた方法とそれに関連した面を述べる。例示すると、5’−ヌクレアーゼプローブ、分子ビーコン、SCORPION(登録商標)プライマー、蛍光共鳴エネルギー移動(FRET)プローブ等のような標識核酸は、当業者に周知の多くのなかでも遺伝子型分類、診断、法医学を含むさまざまな適用において核酸を検出するために頻繁に用いられる。これらのプローブの多くは、蛍光レセプター色素、および2個の部分が互いに適当に近接している時発光性部分からの検出可能な発光を低下させるクエンチャー部分のような発光性標識部分を含む。これらのクエンチャー部分は、検出可能な発光を低下させるが、この低下はしばしば不完全である。すなわち、これらの多重標識プローブは、頻繁に、関連の残留またはベースライン発光を有する。反応混合物中でプローブ量が増加するに伴い、多重適用における異なるプローブの複数セット使用によるのかまたは基本的に任意の適用においてもあるプローブの量が多いことによるのか、このベースライン発光はまた、増大する傾向がある。これらのようなベースライン発光は、典型的には、例えば、感度(すなわち、分析物質濃度のわずかの差異を識別するアッセイ能力)およびダイナミックレンジ(すなわち、定量的測定ができる最低濃度(定量限界すなわちLOQ)から校正曲線が直線性を逸脱する濃度(直線性限界すなわちLOL)まで広がるアッセイの有用範囲)を限定することによって、これらのプローブを用いるアッセイパフォーマンスにマイナスの影響を及ぼす。したがって、本明細書に記載のある発光修飾剤は、消滅させるというまでにはいかないが、これらのベースライン発光をいくつかの態様でさらに低下させるために用いられ、これらのタイプの標識プローブを伴うアッセイパフォーマンスを向上させる。
本発明の反応混合物は、標識核酸からの発光を修飾するのが望ましいさまざまな用途において使用できる。態様によっては、例えば、特に複数の標識プローブをプールした多重フォーマットにおいて本明細書に記載の反応混合物をホモジニアス増幅/検出アッセイ(例えば、リアルタイムPCR等)を実施する際に利用する。本明細書に記載の発光修飾剤のあるものは、温度を変えかつ多くのこれまでの公知の化合物と異なりこれらのアッセイタイプで典型的に使用される反応条件下で、標識プローブからのベースライン発光を低下させる。発光修飾剤および標識オリゴヌクレオチドのほかに、任意に本発明の反応混合物に含められる他の試薬もさらに詳細に下文で説明する。
本発明の反応混合物および他の面で使用する発光修飾剤には、さまざまなタイプの核酸増幅反応およびアッセイにおいて標識プローブからの発光を修飾するかまたは修飾するのに非常に適するようにしたさまざまな性質を含む。例えば、これらの発光修飾剤は、典型的には、一本鎖核酸(例えば、一本鎖プローブ)と二本鎖核酸(例えば、標的核酸とハイブリダイズした一本鎖プローブ)の両者に結合する。あらゆる特定の理論に限定されることなく、本明細書に記載の発光修飾剤は、さらに、核酸に通常結合し核酸に結合した標識物からの発光を長さ依存的に修飾する。すなわち、ある標識オリゴヌクレオチドからの発光を発光修飾剤が修飾する程度は、典型的には、そのオリゴヌクレオチドの長さに比例している。例えば、特定の発光修飾剤は、通常、オリゴヌクレオチドの標識断片からの発光を、インタクトなすなわち全長のオリゴヌクレオチド、例えば、5’−ヌクレアーゼ反応における開裂前のものからの発光よりも低い程度にまで修飾する。ある発光修飾剤は、また、典型的には、さまざまな異なる発光性部分からの発光を有効に修飾できる。すなわち、これらの発光修飾剤によって実行される前記修飾(すなわち、クエンチング)は、通常、スペクトル重複に依存せずすなわち普遍的であり、いかなる特定の作用理論にも限定されるわけでもないが、基底状態複合体形成を経由して出現する可能性がある。これは、例えば異なる蛍光団または他の標識部分で標識した複数のプローブを通常使用する多重アッセイにおいて、重要な性質である。
本発明の反応混合物は、発光修飾剤に加えて標識オリゴヌクレオチドを含む。さまざまな手法が、プローブ(例えば、5’−ヌクレアーゼプローブ、分子ビーコン、FRETプローブ等)及び/又はプライマーとして利用される。例示すると、DNASTAR社(Madison,WI,USA)から入手可能なDNAstarソフトウェアパッケージを配列アラインメントに使用できる。たとえば、個別ファイルとして標的核酸配列および非標的核酸配列をDNAstar EditSeqプログラムに、例えば、これらの配列における低配列類似性を有する領域を同定するプロセスの一部としてアップロードすることができる。さらに例示すると、配列ファイルペアをDNAstar MegAlign配列アラインメントプログラムで開いて、Clustal Wアラインメント方法を適用できる。Clustal Wアラインメントに用いたパラメータは、任意に、前記ソフトウェアのデフォルト設定である。MegAlignは、典型的には、2個の配列間の同一性パーセントの要約を示すことはない。これは、通常、手作業で計算される。前記アラインメントから、例えば互いに選択した同一性パーセント未満である領域を典型的に同定し、これらの領域におけるオリゴヌクレオチド配列を選択できる。多くの他の配列アラインメントアルゴリズムおよびソフトウェアパッケージもまた、任意に利用される。配列アラインメントアルゴリズムは、また、例えば、引用により組み入れられるNotredameら(2000)“T−coffee: a novel method for fast and accurate multiple sequence alignment”、J.Mol.Biol.302:205−217、Edgar(2004)“MUSCLE:a multiple sequence alignment method with reduced time and space complexity”、BMC Bioinformatics 5:113、Mount,Bioinformatics:Sequence and Genome Analysis,Cold Spring Harbor Laboratory Press(2001)、and Durbinら、Biological Sequence Analysis: Probabilistic Models of Proteins and Nucleic Acids、Cambridge University Press(1998)。

本発明の方法で用いた反応混合物またはその成分(例えば、プローブまたは発光修飾剤)は、任意にキットに梱包される。さらに、前記キットには、また、標的核酸ハイブリダイゼーション、増幅、および検出のために必要な緩衝液、酵素、DNAスタンダード、塩、金属イオン、プライマー、伸長可能ヌクレオチドまたはターミネーターヌクレオチド、グリセロール、ジメチルスルホキシド、ポリrA等のような適切に梱包された試薬および材料、ならびに、特定のアッセイを行うための説明書も含まれる。プローブおよび発光修飾剤のようなキット成分は、典型的には、1又は複数の容器で提供される。これらの態様の一部で、前記キットはさらに、例えばピロリン酸分解を最小にするために使用する少なくとも1つのピロホスファターゼ(例えば、熱安定性ピロホスファターゼ)、及び/又はキャリーオーバーコンタミネーションに対する保護が望ましい適用で使用するためのウラシルN−グリコシラーゼ(UNG)を含む。前記キット成分の2個以上は、同一容器に梱包することもできる。
本発明は、ハイブリダイゼーション複合体の融解温度(Tm)を決定するための方法を提供し、これらの方法では、本明細書で教示の可溶性発光修飾剤技術を用いる。Tm決定では、可溶性発光修飾剤(すなわち、可溶性クエンチャー)システムを用いて、二本鎖融解曲線またはアニーリング曲線をモニタリングする。
本発明は、例えば、Tm決定実施するための方法のような本発明の方法を促進するためのキットのような製造された製品を提供する。これらのキットは、本明細書に記載の方法を用いてTm決定を行うために必要な材料を提供する。これらのキットは、例えばウイルス遺伝子型決定を行うためにTm評価を使用できる臨床医のための用途を見出す。これらの方法を実行するための材料および試薬は、本方法実施を促進するキットで提供できる。
態様によっては、本発明は、Tm決定を行うための統合システムを提供する。前記システムは、特にTm誘導手段がアルゴリズム及び/又は電子的に保存された情報(例えば、採取蛍光データ、既定のTm相関等)を含む場合、採取データの解釈および分析のための機器および手段を含むことができる。統合システムの各部分は、機能的に内部で接続されており、場合によっては、物理的に接続されている。態様によっては、統合システムは自動化され、Tm分析開始後、試料または機器のオペレータによるいかなる操作も必要としていない。
さまざまな核酸技術が、核酸の増幅及び/又は検出において配列ミスマッチを起こしている。本発明は、この問題に対する解決策を提供し、本発明は、核酸二本鎖を安定化させる方法を提供する。これらの核酸二本鎖安定化方法は、単一のミスマッチ位置ならびに複数のミスマッチ位置を含む核酸二本鎖安定化に有効である。実際、本明細書に記載の方法は、さらに、マッチした核酸二本鎖を完璧に安定化させることさえできる。完璧にマッチした二本鎖のそれ以上の安定化は、二本鎖がそうでなければ解離した場合思われる条件下でインタクトな核酸二本鎖の保存を可能とするであろう。
蛍光クエンチングアッセイのための全般プロトコール
この章に含めた表は、特に断りがなければ下文に示した実施例で言及した分析において使用したそれぞれの反応成分、条件、および操作を記載している。通常、標識オリゴヌクレオチドの蛍光は、可溶性クエンチャー存在下および不在下で溶液中で測定した。ある分析では、例えば、5’末端においてFAM標識したプローブシリーズは、PCR反応緩衝液(下文で説明)およびさまざまな濃度の可溶性クエンチャーを含む溶液400μL中で測定した。それらの分析においてFAM標識の蛍光検出のため、励起光の波長は、485ナノメータとなるように選択し、蛍光は、波長520ナノメータで測定した。

下記の表、表XIV−XVIIは、本発明で使用した代表的なPCR熱サイクル反応を開示している。各表において、サイクリング反応は、両端に矢印をつけた線で示してあり、この反応は、示した総数のサイクルだけ繰り返した。温度範囲を示したところでは、その範囲内の単一の温度値を用いて異なる実験を行ったことを示している。反応完了時に、試料を4℃にいつまでも保持した。

一重標識プローブとニューメチレンブルーを用いたポリメラーゼ連鎖反応
図12は、HCV検出アッセイにおける一重標識ヌクレアーゼプローブとニューメチレンブルーを含むさまざまな5’−ヌクレアーゼ反応から得たデータを示す増幅プロット(縦軸は、絶対蛍光を示し、横軸は、サイクル数を示す)である。これらの反応混合物中で用いた一重標識ST650プローブ(配列番号5に対応)は、5’末端においてFAMで標識した。さらに、これらのトレースで示される反応混合物は、ポリrAを含んでおり、これらの反応で用いた変性温度(Tden)は、95℃であった。各反応混合物中にはHCV cDNAの20,000コピーが存在していた。これらの反応で用いたアニーリング温度は、58℃であった。これらの反応で用いたニューメチレンブルー濃度は、プロットに示したラベルで示されている。図13の増幅プロットは、このデータについての相対的蛍光を示している。

さまざまな濃度のメチレンブルーを用いたポリメラーゼ連鎖反応
図22(パネルAとB)は、標的HCV DNA,さまざまなプローブおよびさまざまな量のメチレンブルーによるPCR反応をポリアクリルアミドゲルで分析したものの写真である。ゲル中レーン上に示した数は、特定の反応混合物中に含まれたメチレンブルー濃度(μg/mL)を示している。0(−)で示したレーンは、ネガティブコントロールを流したものである。各反応混合物中は、ポリrAとHCV DNAの20,000コピーを含んでいた。さらに、これらの反応で用いたTdenは、95℃であった。示したように、用いたプローブは、ST650_5’−FAM&ST2325_5’−HEX、デュアルST650、およびデュアルST2535であり、それらは上文で説明してある。パネルAとBは、二重反応を示している。この分析は、PCR増幅が、発光修飾剤量が増え存在していても比較的影響を受けないことを示した。
アズールBを用いたポリメラーゼ連鎖反応
図25は、HCV検出アッセイにおいてアズールB不在下で実施した5’−ヌクレアーゼ反応から得たデータを示す増幅プロット(縦軸は、絶対蛍光を示し、横軸は、サイクル数を示す)である。これらのトレースで示した反応混合物は、アズールBを含まず、HCV由来の標的核酸の20,000コピーを含んでいた。これらの反応で用いた変性温度(Tden)は、95℃であった。付したトレースラベルに示したように、反応混合物中には、5’末端においてFAMで標識したプローブ(すなわち、5’−FAM)またはデュアル標識プローブ(すなわち、デュアル)を含み、ポリrAを含んでいるかまたは欠いていた。示したように、非クエンチ一重標識プローブからのわずかの蛍発光があった。これは、前記プローブ中のなんらかのG−クエンチングによるのかもしれない。対照的に、二重標識プローブは、良好なシグナルを産生した。さらに、可溶性クエンチャー存在下で、一重標識プローブはまた、良好なシグナルを産生した(例えば、下文に示した図26および27を参照のこと)。
メチレンブルーを用いた追加の増幅反応
図28と29は、HCV検出アッセイにおいて一重標識ヌクレアーゼプローブとメチレンブルーを含むDNAおよびRNA鋳型滴定から得たデータをそれぞれ示す増幅プロット(縦軸は、絶対蛍光を示し、横軸は、サイクル数を示す)である。これらのトレースで示した一重標識ST650プローブ(配列番号5に対応)は、5’末端においてFAMで標識した。これらのトレースで示した反応混合物はポリrAを含み、これらの反応で用いたTdenは、95℃であった。これらの反応混合物で用いたメチレンブルー濃度は、40μg/mLであった。プロットに付したラベルは、HCV由来の標的cDNAまたはRNAのコピー数を、各トレースについての他の反応条件とともに示した。これらの分析では、例えば、高感度で定量的なPCRおよびRT−PCR検出が、これらの方法を用いて達成できることを示している。
一重標識プローブとニューメチレンブルーを用いたポリメラーゼ連鎖反応
図32は、HCV検出アッセイにおける一重標識5’−ヌクレアーゼプローブとニューメチレンブルーを含むさまざまな5’−ヌクレアーゼ反応から得たデータを示す増幅プロット(縦軸は、絶対蛍光を示し、横軸は、サイクル数を示す)である。これらの反応混合物中で用いた一重標識ST650プローブ(配列番号5に対応)は、5’末端においてFAMで標識した。さらに、これらのトレースで示される反応混合物は、ポリrAを含んでおり、これらの反応で用いたTdenは、95℃であった。各反応混合物中にはHCV cDNAの20,000コピーが存在していた。これらの反応で用いたアニーリング温度は、40℃であった。これらの反応混合物中で用いたニューメチレンブルーチオニン濃度は、プロットに示したラベルで示されている。

この実施例および下記の他の実施例は、多重標識プローブの使用を含む5’−ヌクレアーゼアッセイにおける蛍光の修飾を示している。この実施例では、5’−ヌクレアーゼ反応においてメチレンブルーを使用して、5’−ヌクレアーゼプローブからのベースライン発光が修飾されることを示している。
多重標識プローブと1,9−ジメチルメチレンブルーを用いたポリメラーゼ連鎖反応
この実施例は、5’−ヌクレアーゼプローブからのベースライン発光を修飾するための5’−ヌクレアーゼ反応における1,9−ジメチルメチレンブルーの使用を示している。

この実施例は、5’−ヌクレアーゼプローブからのベースライン発光を修飾するための5’−ヌクレアーゼ反応におけるニューメチレンブルーの使用を示している。
HEX標識プローブを用いた定量化標準増幅と検出
この実施例では、HCV定量化標準(HCV−QS)増幅および検出アッセイにおけるHEX標識5’−ヌクレアーゼプローブからのベースライン発光を修飾するメチレンブルー使用を示している。HCV QS DNAは、HCVプライマー結合配列と独自のQS特異的プローブ結合領域を含んでいた。反応混合物は、HCV およびHCV QS DNAに特異的なプライマーペアを含んでおり、増幅DNAの検出は、増幅時に標的特異的QSプローブから放出される蛍光レポーター色素の発光強度を測定することによって、実施し、それにより、HCVとHCV QSをそれぞれ別々に識別できた。
HCV核酸検出におけるベースライン発光の修飾
この実施例では、HCV核酸検出を必要とするアッセイにおいて5’−ヌクレアーゼプローブからのベースライン発光を修飾するさまざまな発光修飾剤の使用を示している。
HCVモデルシステムにおける可溶性発光修飾剤と結合させた一重標識プローブを用いた融解曲線分析(Tm決定)
本実施例では、可溶性発光修飾剤と結合させた一重標識プローブを用いた融解曲線分析(すなわち、Tm決定)を述べている。HCVプローブとHCV合成鋳型を、実験システムで用いる。一重常識プローブを可溶性クエンチャーとともに用いることの有効性を明らかにする。
HCVモデルシステムを用いたチアジン存在下における核酸二本鎖安定化の実証
本実施例は、チアジン色素の二本鎖安定化性質を示している。HCVプローブとHCV合成鋳型を実験システムで用い、形成されたさまざまなハイブリダイゼーション複合体のTmを測定することによって、二本鎖安定化を実証する。
複数HCV鋳型標的を用いたチアジン色素存在下における核酸二本鎖安定化
チアジン色素の二本鎖安定化特徴を、さらに図60の棒グラフに示した。この棒グラフは、示したHCV遺伝子型に対応するヌクレオチド配列を有するさまざまな合成核酸で示唆したHCVプローブを用いてTm決定した要約を示している。この分析ではメチレンブルーの効果を調べ、融解曲線分析においてこれとは別にメチレンブルーゼロ、メチレンブルー10μg/mL、メチレンブルー20μg/mLを使用した。
チアジン色素存在下における単一ヌクレオチドミスマッチ安定化の実証
本実施例は、さらに、8種の異なるヌクレオチドミスマッチに対する安定化影響を実証することによって、チアジン色素の単一ヌクレオチドミスマッチ安定化性質を例示する。HCVプローブおよび合成鋳型を、実験システムで用い、単一ヌクレオチドミスマッチ安定化は、形成されたさまざまなハイブリダイゼーション複合体のTmを測定することによって、実証する。
HIVモデルシステムを用いたチアジン色素存在下における改良サブタイプ堅守の実証
本実施例では、ウイルス標的の増幅および検出におけるチアジン色素の二本鎖安定化性質の利点を示す。ミスマッチ二本鎖安定化のチアジン色素の能力により、多型サブタイプの検出感度が、通常、増幅及び/又は検出効率向上により大きく改善できることがわかる。先の実施例と対照的に、この実施例では、さまざまなTaqMan増幅反応のCT値を用いて、二本鎖安定性増強の利点を明らかにする。HIV増幅プライマー、HIV二重標識5’−ヌクレアーゼ定量化プローブおよびHIV合成鋳型を、図62に示したように実験モデルシステムで用いた。図62のプライマーおよびプローブ配列の下に、公知のHIV単離物由来の対応する相同ドメインを示し、可変位置も示した。
FAM−TCTGCAGCTBHQ2TCCTCATTGATGGTATCTTTTA−PO4(配列番号25)
を有する二重標識5’−ヌクレアーゼプローブGAG108FBHQ29Iを含んでおり、ここで、FAMは、蛍光標識であり、PO4は、末端リン酸であり、BHQ2は、ブラックホールクエンチャー(BHQ(商標))−2である。増幅反応はまた、サブクローニングし単離したHIV遺伝物質のインビトロ転写とオリゴ−dT−セファロースクロマトグラフィによって産生させた合成HIV RNA分子を含んでいた。特定のRNA転写物の100万コピーを各反応で使用した。PCR反応では、下記のサイクリングプログラムを用いた:
50℃/5分;59℃/30分;95℃/2分;95℃→58℃(2サイクル);91℃→58℃(60サイクル)。
HIVモデルシステムを用いたさまざまな濃度のチアジン色素存在下における用量依存性核酸二本鎖安定化の実証
本実施例では、色素をある濃度範囲で使用し、チアジン色素の二本鎖安定化性質を示している。この実施例では、実施例23に記載のものと同一のHIV実験モデルシステムと試薬を用いる。
SYBR(登録商標)グリーン アンプリコン検出を用いたチアジン色素の核酸二本鎖安定化の実証
本実施例では、チアジン色素の二本鎖安定化性質を示し、ここで、モデルシステムSYBR(登録商標)グリーンを用いてアンプリコン蓄積をモニタリングしている。HIV増幅産物のリアルタイム定量化のための増幅反応は、HIV増幅プライマーSK145BU(配列番号23)およびGAG152BU(配列番号24)を用いて行った。この実施例では、増幅反応は、HIV遺伝子型110−5合成RNA鋳型(106コピー)を標的にした。全反応物に、SYBR(登録商標)グリーンを添加し、二本鎖アンプリコン産物の蓄積をモニタリングした。5’−ヌクレアーゼ定量化プローブは用いなかった。

Claims (7)
- ハイブリダイゼーション複合体の融解温度(Tm)を決定する方法であって:
(a)(i)発光部分を含んで成るプローブ;
(ii)前記プローブに対して相補的であるかまたは部分的に相補的であるハイブリダイゼーション標的;および
(iii)チアジン色素またはジアジン色素を含む可溶性発光修飾剤であって、前記発光部分をクエンチングできる前記可溶性発光修飾剤、
を準備すること;
(b)前記プローブと前記ハイブリダイゼーション標的とを、塩基対形成が起こり標的ハイブリダイゼーション複合体を形成できる条件下でアニーリングすること;
(c)前記可溶性発光修飾剤存在下で標的ハイブリダイゼーション複合体の温度を変化させ前記発光部分の放出を測定すること;
(d)前記発光部分の測定された放出を前記標的ハイブリダイゼーション複合体の存在と温度関数として相関させ、それによって測定された放出に基づき標的ハイブリダイゼーション複合体のTmを決定すること、
を含んで成る方法。 - 前記ハイブリダイゼーション標的の準備が核酸標的に対応するアンプリコンの産生を含んで成る、請求項1に記載の方法。
- 前記アンプリコンの産生がポリメラーゼ連鎖反応による核酸の増幅を含んで成る、請求項2に記載の方法。
- 前記ポリメラーゼ連鎖反応が非対称ポリメラーゼ連鎖反応である、請求項3に記載の方法。
- 前記測定段階がある温度範囲での放出の測定を含んで成る、請求項1〜4のいずれか1項に記載の方法。
- ハイブリダイゼーション複合体の融解温度(Tm)を決定するキットであって、
(a)発光部分を含む少なくとも1つのプローブであって、ハイブリダイゼーション標的と相補的であるかまたは部分的に相補的であるプローブ;
(b)可溶性のチアジン色素またはジアジン色素を含む少なくとも1つの可溶性発光修飾剤であって、発光部分をクエンチングできる可溶性発光修飾剤;および
(c)(a)、(b)又は(a)及び(b)を含む1又は複数の容器、
を含むキット。 - ハイブリダイゼーション複合体の融解温度(Tm)を決定するシステムであって:
(a)(i)シグナルを放出する発光部分を含んで成る核酸プローブ;
(ii)前記核酸プローブと相補的であるかまたは部分的に相補的な標的核酸;並びに
(iii)可溶性のチアジン色素またはジアジン色素、
を含んで成る試料;
(b)(i)全プローブ分子が所定のハイブリダイゼーション条件下で前記ハイブリダイゼーション標的とアニーリングする温度;
(ii)標的ハイブリダイゼーション複合体の50%が前記ハイブリダイゼーション条件で解離する温度;および
(iii)どのプローブ分子も前記ハイブリダイゼーション標的と前記ハイブリダイゼーション条件下でアニーリングしておらず、且つ、どのハイブリダイゼーション複合体も前記ハイブリダイゼーション条件で存在していない温度、
を含む温度範囲にわたり試料の温度を制御するための熱制御デバイス;
(c)前記温度範囲にわたり前記試料由来のシグナルを測定するための検出器;および
(d)前記検出器と作用可能に連結し、且つ前記温度範囲でシグナル測定値を受け取る相関モジュールであって、シグナル強度を前記プローブとハイブリダイゼーション標的を含むハイブリダイゼーション複合体の存在とチアジン色素またはジアジン色素との混合物中で温度関数として相関させ、それによって前記標的ハイブリダイゼーション複合体のTmを決定する相関モジュール、
を含む、システム。
