Description
I.組織再生用基材
本発明の組織再生用基材は、コラーゲン及びゼラチンを含む生体吸収性の多孔質基材に細胞成長因子を含有してなる。本発明の組織再生用基材とは、広く再生医療に用いられる基材を意味し、例えば、皮膚、骨、軟骨、心筋、脂肪などの再生医療に用いられる基材である。特に、皮膚(真皮)の組織再生用基材、つまり人工真皮基材として好適に用いられる。
式中、Wsは、組織再生用基材を25℃においてリン酸緩衝生理食塩水中に1時間浸漬したときの重量(湿潤重量)を表し、Wdは、癒着防止材を真空乾燥機を用いて完全に乾燥したときの重量(乾燥重量)を表す。
II.組織再生用基材の製法
本発明の組織再生用基材は、コラーゲン及びゼラチンを含む水性混合物を凍結乾燥する工程、得られる乾燥体を架橋処理する工程、及び得られる架橋体に細胞成長因子を含有させる工程を含む製造方法により製造される。
[実施例1]
(1)コラーゲンとゼラチンの混合スポンジの作製
豚腱由来タイプIコラーゲンと豚皮膚由来ゼラチンとを混合して水溶液とした。水溶液を型枠に入れて−40℃で1時間凍結し、その後凍結乾燥することによりスポンジを作製した。
(2)in vitro徐放試験
実験に使用したスポンジは、ゼラチン含量が0、10、30、50重量%のものを用いた。
ゼラチン含量0、10、30、50、100重量%のスポンジを、モルモット背部全層皮膚欠損層に埋植したときの組織切片像を図2〜6に示す。ゼラチン100重量%のスポンジでは、周囲組織の細胞の侵入が少なく、またスポンジの下に細胞が潜り込むダウングロースの像が観察された。一方で、コラーゲン含量が高くなるにつれて、細胞の侵入が多く、組織の再生も良好であったが、コラーゲン100重量%の場合、細胞成長因子の徐放性能が低下するため、周囲組織の侵入を妨げることなく、かつ細胞成長因子の徐放機能を発揮させるためには、ゼラチンを最適な混合割合で混合させることが必要となる。
Claims (5)
- コラーゲン及びゼラチンを含む生体吸収性多孔質基材に塩基性繊維芽細胞成長因子(bFGF)を含有してなる組織再生用基材であって、該生体吸収性多孔質基材のゼラチンの含有量が10重量%であり、該生体吸収性多孔質基材がコラーゲン及びゼラチンを含む水性混合物を凍結乾燥して得られたものであり、該生体吸収性多孔質基材が化学架橋されており、該生体吸収性多孔質基材が平均孔径10〜500μmの連続孔を有する組織再生用基材。
- 37℃のリン酸緩衝食塩水(PBS)に3日間浸漬させた後の該PBS中への細胞成長因子の放出量が組織再生用基材中に含侵させた初期の細胞成長因子の総量に対し7重量%以下である請求項1に記載の組織再生用基材。
- 37℃のリン酸緩衝食塩水(PBS)に7日間浸漬させた後の該PBS中への細胞成長因子の放出量が組織再生用基材中に含侵させた初期の細胞成長因子の総量に対し15重量%以下である請求項1又は2に記載の組織再生用基材。
- 皮膚の組織再生用である請求項1〜3のいずれかに記載の組織再生用基材。
- 組織再生用基材の製造方法であって、(1)コラーゲン及びゼラチンを含む水性混合物を凍結乾燥して、ゼラチンの含有量が10重量%であり、平均孔径10〜500μmの連続孔を有する生体吸収性多孔質基材を製造する工程、(2)得られる生体吸収性多孔質基材を化学架橋処理する工程、及び(3)得られる架橋体に塩基性繊維芽細胞成長因子(bFGF)を含有させる工程を含む製造方法。
