Abstract
Description
炭水化物は、アノマー炭素から最も遠いキラル炭素の配置によって決定されるD−およびL−鏡像異性配置を有し得る。炭水化物の直線状形態がフィッシャー投影として描かれる場合、キラル炭素に結合したヒドロキシル基は、炭素の左側または右側のいずれかに配置され得る。ヒドロキシルがキラル炭素の右側にあれば、炭水化物はD−炭水化物として注釈が付けられ、ヒドロキシル基がキラル炭素の左側にあれば、炭水化物はL−炭水化物として注釈が付けられる。以下の例は、グルコースのD−およびL−鏡像異性体の構成を示す。
ポリカプロラクトン(Mw:45,000)、L−ラムノース、およびNaOHを、Sigma−Aldrich(USA)から購入した。キトサン(CS)を、Mian Scientific Company、Lahore、Pakistanから購入して、本発明者らのラボにおいてさらに精製した(脱アセチル化度(DD)84%;Mw:87047.26g/mol)(Farooq et al., Materials Science and Engineering: C. 2015)。コラーゲンを、Mian ScientificのサプライヤーLahoreから購入した。ポリ(ビニルアルコール)(PVA)(Mw:72,000、加水分解度98%)、塩酸(HCl)、および硫酸(H2SO4)を、Merck(Germany)から購入した。オルトギ酸トリエチル(98%)を、Alfa Aesar(Germany)から購入した。氷酢酸(CH3COOH)を、AnalaR BDH Laboratory Supplies(UK)から購入した。エタノール(99.8%)を、Sigma−Aldrich(Germany)から購入した。2−デオキシ−D−リボース、D−フコース、およびL−フコースを、それぞれSigma−Aldrich China、UK、およびSlovakiaから購入した。2−デオキシ−L−リボースおよびD−ラムノースは、Carbosynth(UK)の製品であった。Brain Heart Infusionブロスおよびトリプトンソイブロス(1%w/v)を、Oxoid Ltdから購入した。フェノールレッドを、Sigma−Aldrich UKから購入した。
PCL(10wt%)を5ml DMF/DCM(4:1、10%w/v)中に溶解させて、2−デオキシ−D−リボース(1.8wt%)をDMF(0.5mL)中に溶解させた。双方の溶液を組み合わせて、ロッキングステーション上に一晩放置して、室温にて均一な溶液が生じた。翌日、この溶液を、4×5mLシリンジを用いて電界紡糸した。これを、プログラム可能なシリンジポンプ(Kent Scientific、USA)上に水平に配置して、40μL/minの速度で放出した。シリンジには、高電圧電源(Genvolt、UK)を用いて、17kVを供給した。繊維状足場を、針先から17cmの距離にて、直径16cm×6cmの回転ドラム上に収集した。このドラムを300rpmにて回転させて、18×16cmのマットを生じさせた。足場を、約20℃の室温にて製造した。
CS(2.5%w/v)を酢酸(1%)溶液中に溶解させて、12時間磁気撹拌した。別個のフラスコ内で、PVA(10%w/v)を、磁気撹拌下で80℃にて蒸留水中に溶解させた。次に、2つの溶液をそれぞれ、CSおよびPVAの80:20の比率で混合して、さらに24時間撹拌した。次に、溶液を別々のペトリ皿に流し込んで、皿を−80℃にて24時間冷凍した。サンプルを、凍結乾燥機内で−40℃にて24時間凍結乾燥させた。次に、凍結乾燥ヒドロゲルを再水和させて、硫酸(17%w/v)を有する様々な濃度のTEOF(すなわち、0、4、8、および16%w/v)中に24時間浸した。次に、ヒドロゲルをペトリ皿から取り出して、NaOH(12%w/v)で1時間処理してから、蒸留水で3回洗浄して、24時間凍結乾燥させた。
CSおよびコラーゲン(それぞれ0.5g)を、酢酸(0.5M、20mL)中に溶解させて、完全に溶解して透明な溶液が得られるまで、室温にて撹拌した。これに、オルトギ酸トリエチル(4%v/v、0.8mL)を架橋剤として加えて、溶液をさらに一晩撹拌して、材料の均一性および架橋性をより良好にした。次に、混合液をペトリ皿に注いで、−20℃にて一晩凍結した。次に、凍結した膜を、予め冷却した水酸化ナトリウムのエタノール溶液(3M)中に浸して、再び−20℃にて24時間置いた。その後、凍結した膜を、50%(v/v)エタノール溶液中で洗浄してから、蒸留水で洗浄して水酸化ナトリウムを除去した。膜を最終的に無水エタノール中で15分間、2回洗浄して、室温にて乾燥させて多孔質足場を得た。
赤外線(FT−IR)スペクトルを、Thermo−Nicolet 6700 P FTIR Spectrometer(USA)で8cm−1の解像度にて、4000〜400cm−1の周波数範囲の光音響モードで、256回の連続スキャンにより記録した。
複合マトリックスの表面形態を、SEM、モデルJEOL JSM 6480下で調査した。サンプルを、金でスパッタコーティングしてから、広範な倍率にて試験した。
糖担持のために、ヒドロゲルを、37℃の2−デオキシ−D−リボース(1mg/ml)の水溶液中に、全ての液体がヒドロゲルによって吸収されるまで浸した。−20℃にて一晩保存した後に、ヒドロゲルを凍結乾燥機(Christ、Alpha 1−2 LD plus、Germany)内で−40℃にて乾燥させてから、さらに特性評価した。
ニワトリ(Gallus domesticus)受精卵を、Big Bird(Raiwind road、Lahore、Pakistan)から購入して、加湿孵卵器(HHD、435、China)内で受精2日目から8日目まで37℃にてインキュベートした。8日目に、正方形の窓(1cm2)を殻中にカットして取り外し、2cm2片の電界紡糸足場またはヒドロゲル(16%PCL電界紡糸足場/CS/PVAヒドロゲル/CS/コラーゲンヒドロゲル)をCAM上に配置した。各卵に、1つの足場またはヒドロゲルのみを移植した。
in vivo実験のために、ヒドロゲルの直径20mmおよび厚さ1.2mmの円を準備した。このサンプルを、エタノール(70%)溶液を用いて滅菌して、風乾させて、PBS中で短時間平衡化させてから、創傷上に配置した。動物試験用に、体重140〜170gの雄Sprague−Dawley(SD)ラットを研究に用いた。動物は、食物および水を自由に摂取できるように、Molecular Biology (CEMB)、Lahore内のCentre of Excellenceの動物ハウス施設内で飼育した。全ての動物を、CEMB、Lahore、PakistanのInstitutional Animal Ethics Committeeによって承認された手順に従って処置した。動物を、ケタミン(100mg/体重kg)およびキシラジン(10mg/体重kg)で麻酔してから、背側の毛をヘアトリマー(Dingling professional hair clipper、RF−608、China)で除去した。粘着テープ固定と包帯用に、ラットの背中を完全に剃毛した。剃毛後、背中をエタノールできれいにし、その後、清潔な滅菌テーブルに移して、手術用ハサミ(Noorani Surgical Medical Supplies、Pakistan)で、マークした3つの領域の皮膚を除去することによって、3つの円形の創傷を生じさせた。切除プロセスの直後に、これらの創傷の一方を、滅菌した2−デオキシ−D−リボース担持(70%エタノールを使用)膜円で覆い、他方を同じ材料膜で覆ったが、2−デオキシ−D−リボースは全く含まなかった。移植した膜を双方とも、滅菌編み絹糸(Mersilk、直径220ミクロン)を用いて所定の場所に縫合した。足場を、絆創膏(Mepore、Sweden)で覆って、さらに綿ガーゼで覆って、最後に、白色の手術用粘着テープ(日東電工株式会社、日本)で固定して、自己グルーミングによる破損を防いだ。これらの創傷の1つを開いたままにして、包帯(Mepore、Sweden)のみで覆って陰性対照として機能させた。
パラフィン包埋サンプルからミクロトーム(Leica TP 1020 Automatic Tissue Processor)で厚さ6μmの切片をカットして、Superfrost(登録商標)plusスライド(Menzel−Glaser、Denmark)上に配置した。従来のヘマトキシリンおよびエオシン(H&E)染色およびGouldnerのトリクローム染色を実行してから(Yar et al., International Journal of Polymeric Materials and Polymeric Biomaterials. 2016)、DPXマウンティングメディア(Fisher Scientific)中にカバースリップと共にマウントした。
この調査において研究したLおよびDデオキシ糖を代謝する細菌の能力を評価するために、糖を発酵して酸を生成する能力を判定した。
5つの細菌株を本研究に採用した;黄色ブドウ球菌(Staphylococcus aureus)の4株、および緑膿菌(Pseudomonas aeruginosa)の1株。黄色ブドウ球菌(S.aureus)株はS235であった−以前このラボから報告した細菌感染皮膚モデルの開発に用いた最近の臨床分離株である(Shepherd J et al., Tissue Engineering Part C: Methods. 2009)。NCTC 6571(Oxford)−抗生物質感受性試験用の参照株として一般的に用いられる参照株である。ニューマン株−臨床株由来であるが、表面フィブロネクチン結合タンパク質に欠陥がある。L−9879−別の臨床分離株であるが、親水性である。
細菌を、Brain Heart Infusionブロス内で37℃にて16時間増殖させた。フェノールレッド(0.02%w/v;Sigma−Aldrich UK)および適切な糖(1%最終濃度)を補充したトリプトンソイブロス(1%w/v)のアリコート(100ml)を、滅菌96ウェルプレートに加えた。次に、各細菌株の一晩ブロス培養液5lをウェルに加えて、37℃にて16時間インキュベートした。各糖を発酵する各株の能力を、酸の生成およびフェノールレッドpH指示薬の変化によって判断した。
免疫染色についての群間差異を、ノンパラメトリッククラスカル・ワリス検定で検定して、個々の群間の多重比較を、ダン検定を用いて検定した。
電界紡糸PLC膜中へのD−デオキシ糖の組込み
2−デオキシ−D−リボースを電界紡糸ナノファイバ中にカプセル化して、DCM中DMFを電界紡糸用の溶媒として用いた。電界紡糸により、糖を担持した柔らかい繊維状材料のシートを製造した(図1)。糖を含まないPCL繊維の平均直径は、224nm±82nmであったが、2−デオキシ−D−リボースを担持した繊維の平均直径は、323nm±25nmに有意に増大した(40本のランダムな繊維の平均直径)(p≦0.05)。しかしながら、両タイプの繊維は、相互関係のある孔と共にランダムに配向されており、その表面は滑らかであり、いかなるビーズもなかった。糖担持後、膜の色は明るい青色となった。
オルトギ酸トリエチル(TEOF)を用いてCSとPVAを架橋する方法は、2015年に本発明者らのグループが以前に報告した(Yar et al., Materials Science and Engineering: C. 2015; Shahzadi et al., Journal of biomaterials applications. 2016)。本研究では、本発明者らは、2−デオキシ−D−リボースを、物理的吸着によって、16%TEOFで架橋したCS/PVAヒドロゲル上に担持させた。本研究では、2−デオキシ−D−リボースを担持した非架橋ヒドロゲルを対照として用いた。SEM顕微鏡写真は、架橋と糖担持の双方が、このヒドロゲルの孔径と形態に影響を与えるようであることを示した(図2)。架橋前に、ヒドロゲルは、相互連結した孔ではなく層状構造を有した。平均孔径は、103μm±52μmであった。架橋後、孔径は有意に縮小した(p≦0.05)。架橋の不在化で糖を加えると、層状構造が、歪んだ孔に変化した。架橋と糖の双方の導入により、平均孔径は68μm±31μmに有意に縮小した(p≦0.05)。これらの相互連結した新しい孔は、より均一な形態を有しており、孔は、ヒドロゲルマトリックスの全体に十分に分布していた。
キトサンおよびコラーゲンを、オルトギ酸トリエチルを用いて架橋させ、その後、糖水溶液(1mg/ml)中に浸して2−デオキシ−D−リボースを担持させた。生理食塩水中に沈める前のヒドロゲルの平均厚さは、1.11mmであり、浸水24時間後は1.37mmに増大した。値は、3つの独立した実験の平均である。
FTIR
FTIR分光法を用いて、糖とPCLのマイクロ繊維との相互作用を調べた。図5Aは、紡糸前に糖をポリマーに加えた場合に、約3400cm−1にて見られるDリボースの主要なピークがPCL繊維において検出されたことを示す。PCLのカルボニルエステルに対応する1720cm−1にて主要な吸収バンドが観察された。D糖とPCLの双方におけるアルキル基のピークは、2700〜2900cm−1に吸収ピークを示した。D−糖担持PCL繊維のスペクトルはまた、糖が電界紡糸混合物中に均一に分布したことも示している。
D−デオキシリボースを担持した電界紡糸繊維およびヒドロゲル
CAMアッセイを用いて、合成された材料の血管新生の潜在性を調査した。2−デオキシ−D−リボース担持繊維の場合、材料の下に広範な新しい血管の成長が観察された(図6A(b)を参照)。これにより、2−デオキシ−D−リボース担持材料の化学誘引および血管新生の潜在性が確認される。
一般的に、血管の急速な成長および浸潤の一助となる生体材料は、血管新生の遅延および炎症の継続を示す生体材料よりも優れた生体適合性を示すと考えられている(Wolf et al., Biomaterials. 2014)。その結果、これは、組織の生存を確実にするために組織工学的材料にとって絶対に不可欠な特性である。
ラットにおける創傷治癒を、2−デオキシ−D−リボース−担持ヒドロゲルを当てることによって促進した。20mmの領域をマークしてから、外科用ハサミを用いて麻酔下で切除を実行することによって、全層創傷を生じさせた。それぞれの材料を創傷に当てて、適所で縫合して、最初にMepore(Sweden)絆創膏で、そして最後に綿包帯で覆った。全てのラットに、創傷の3日後に除去したのと同じ包帯を着けた。特定の時点にて各創傷をデジタル写真撮影して、Image Jソフトウェアを用いてさらに定量化した。創傷の代表的な巨視的写真を図8に示す。創傷後3、9、11、14、および17日目の対照ヒドロゲル(架橋CS/コラーゲン足場)および2−デオキシ−D−リボース−担持ヒドロゲルを示す。
全ての群由来のサンプルを17日目に収穫して、パラホルムアルデヒド中で一晩固定した。脱水後、サンプルをパラフィンで包埋してブロックを作製し、これを6μmにて切片化した。次に、スライドを、H&E、Gouldnerのトリクローム、およびDABペルオキシダーゼキット(3つの抗原(前駆内皮細胞についてCD34、M1マクロファージについてCD80、そしてM2マクロファージについてCD163)の検出を調べる)で染色した。最後に、スライドをDPX封入剤およびガラス製カバースリップで覆ってから、光学顕微鏡で撮像した。偽(いかなる処置もしない創傷)群、対照(キトサンおよびコラーゲン足場で処置した創傷)群、およびD−デキソリボース(2−デオキシ−D−リボースを担持した足場で処置した創傷)群を、非処置ラット由来の正常な皮膚と比較した。
調査中の6つの糖を、細菌代謝用の基質としての作用に関して、グルコースと比較した。黄色ブドウ球菌(S.aureus)の4株および緑膿菌(Pseudomonas aeruginosa)の1株を、創傷感染における一般的な病原体の代表として調査した。黄色ブドウ球菌(S. aureus)の全4株の結果は、全てが発酵用の基質としてグルコースを用いることができるという点で同じであったが、これらの株はいずれも、調査中の3つのデオキシ糖のLまたはD異性体を用いることができなかった。しかしながら、緑膿菌(P.aeruginosa)は、デオキシフコースおよびデオキシラムノースのL異性体を除く全ての糖を発酵させることができた。
本研究は、電界紡糸PCL繊維またはキトサンベースのヒドロゲルのいずれかで送達された場合の、2−デオキシ−D−リボースの血管新生促進能力を実証した。これは、試験した2−デオキシ糖の3つ全てのL異性体が血管新生性であったものの、デオキシフコースまたはデオキシラムノースのD異性体によって共有されなかった。2−デオキシ−D−リボースは、緑膿菌(Pseudomonas aeruginosa)によって代謝され得たものの、黄色ブドウ球菌(Staphylococcus aureus)の4株によって代謝され得なかった。キトサン/コラーゲンヒドロゲルへのD−デオキシ−リボースの添加は、ラット内の全層創傷における創傷治癒および毛包の回復を促進した。
Ex−ovo CAMアッセイ
卵のインキュベーション
全てのCAM実験を、英国内務省のガイドラインに従って実行した。ex−ovo CAMアッセイの概略図を図13に示す。ニワトリ(Gallus domesticus)受精卵を、Henry Stewart & Co.(MedEggs、Norwich、UK)から購入した。ハンドペーパータオルを用いて、20%の工業用メチル化スピリット(IMS)溶液で卵を注意深く拭いて、殻から汚れおよび羽毛を取り除いた。次に、卵を、加湿孵卵器(RCOM King SURO、P&T Poultry、Powys、Wales)内に水平に配置して、胚発生日(EDD)3日目まで37.5℃にてインキュベートした。
EDD3に、卵の上面をフェルトペンでマークした。卵を水平に(マークした表面を上にして)保持して、1000mlのガラスビーカーの端で割って、ペトリ皿の底面近くに維持した。次に、胚を滅菌ペトリ皿中に静かに移して、38℃の加湿インキュベータ(Binder、Tuttlingen、Germany)内に保管した。
プラスチックリング(直径約6.5 mm)を、物質用のリザーバ、および移植領域用のマーカーとして用いて、CAM上に配置して、20μl容量の物質をCAM上に1日2回付与して、EDD7からEDD11の間に40μlの総容量を与えた。EDD11にデジタル顕微鏡を用いてリングの画像を得、その後、20%レンズマメアグルチニン(lens culinaris agglutinin)(LCA)(Vector Laboratories、Peterborough、UK)を、解剖顕微鏡(Wild Heerbrugg、Heerbrugg、Switzerland)下で30G針を用いて循環系中に注射して、血管内内皮細胞を標識した。その後、CAMを取り出して、3.7%ホルムアルデヒド溶液中で固定した。胚を、EDD11の終わりに犠牲にした。次に、固定CAMサンプルを共焦点顕微鏡(Zeiss LSM 510 Meta、Jena、Germany)下で撮像して、CAMの微小血管構造に及ぼす物質の影響を調査した。
識別可能な全ての血管を含むように画像を分割してから[C. Roma−Rodrigues, A. Heuer−Jungemann, A.R. Fernandes, A.G. Kanaras, P. V. Cheresh, Use of the 10−Day−Old Chick Embryo Model for Studying Angiogenesis, Integrin Protoc. 129 (1999) 257−269. doi:10.1385/1−59259−249−X:257]、血管を定量化した。最初に、リングの内部領域を切り取って、Adobe Photoshop CS6(ADOBE Systems Inc.、San Jose、California、USA)を用いて、赤、緑、および青色のチャンネルを分割した。次に、緑色のチャンネルを、ImageJ(Wayne Rasband、米国国立衛生研究所)にインポートして、アンシャープマスクフィルタリング、ローカルコントラストの強調、ノイズ除去、およびセグメンテーションを含む更なる分析を行った。最後に、定量化ソフトウェア(AngioTool、米国国立癌研究所)を用いて、分岐点の数を定量化して(図14A)、バイナリ画像ヒストグラムを用いて、ImageJ(Wayne Rasband、米国国立衛生研究所)内の既知のピクセル/mm比により平均血管長を算出した。
特に記載がない限り、全ての化学物質をSigma Aldrichから購入した。組換えVEGF165ストック溶液を、2ng/μl(VEGF−80ng)の濃度に希釈した。E2をメタノール中に溶解させてから、ワーキングソルーションをリン酸緩衝生理食塩水(PBS)で調製して、(a)100ng/日(E2−100ng)、(b)200ng/日(E2−200ng)、および(b)600ng/日(E2−600ng)の濃度にした。PBS中に溶解させることによって、最終濃度が(a)20μg/日(2dDR−20)、(b)200μg/日(2dDR−200)、および(c)1000μg/日(2dDR−1000)になるように、2dDR溶液を調製した。リンゴ酸スニチニブ(スニチニブ)をDMSO中に溶解させて、PBSで希釈して、50ng/μlの最終濃度にした。全ての物質のワーキングソルーションを、EDD7の直前に調製した。様々な物質濃度の血管新生効率を、物質をCAMに直接付与(2用量/日/胚)することによって評価した。分岐点の数を定量化し、かつ平均血管長を算出することによって、血管新生活性を判定した。
E2および2dDRを担持したPHBV足場の電界紡糸
溶液の調製
溶液(約10ml)を、内径0.6mmのシリンジチップを取り付けた10mlシリンジ中にロードした。次に、シリンジを、シリンジポンプ(GenieTMPlus、KentScientific、Connecticut、USA)内に入れた。アルミニウム箔をコレクタとして用いて、針先から17cm離して配置した。ポンプを40μl/分にセットして、コレクタおよびチップの双方に17kVの電圧を印加した。電界紡糸を、全てのポリマー溶液が使い切られるまで、室温にて行った。
E2放出足場および2dDR放出足場の表面形態を、SEM(Philips/FEI XL−20 SEM;Cambridge、UK)下で観察した。サンプルを、金スパッタ(EdwardsスパッタコーターS150B、Crawley、England)を用いて金でコーティングしてから撮像した。ImageJを用いて繊維の直径および孔径を測定した。
足場を、6ウェルプレート中にフィットするように断片に切り分けて、重量を量って、4 mlのPBS中に沈めた。各群(25mg E2、50mg E2、250mg 2dDR、500mg 2dDR)から放出されたE2および2dDRの濃度を、UV−VIS分光光度計(Thermo Fischer Evolution 220、Massachusetts、USA)を用いて、2dDRについて238nmにて、そしてE2について220nmにて蛍光定量的に測定した。E2および2dDRの知られている濃度の標準曲線を用いて、吸光度値を濃度に変換した。
足場を、レーザー切断機(Epilog Laser Cutter、Clevedon、UK)を用いて直径5.5mmの円にカットして、1時間UV光下で滅菌してから移植した。2つの円形の足場を、EDD8にCAM上に配置した。足場の画像を、EDD12およびEDD14に、デジタル顕微鏡を用いて得た。循環系中へのローダミン標識レクチンのマイクロインジェクションを実行して、CAMを取り出して、3.7%ホルムアルデヒド溶液中で固定した。次に、胚を、EDD14の終わりに犠牲にした。移植片に向かって収束した全ての血管をカウントすることによって、血管新生を定量化した。
ヘマトキシリンおよびエオシン(H&E)染色を、標準的なプロトコル[47]を修正することによって、細胞含浸足場に実行した。簡単に言えば、固定サンプルを、最適な切断温度のOCT内に包埋して、液体窒素中で3分間凍結した。クリオスタット(Leica Biosystems Nussloch、Germany)を−20℃にて用いて、切片を厚さ8〜10μmにカットした。次に、切片を、ヘマトキシリンで90秒間、そしてエオシンで5分間染色した。最後に、H&E画像を、光学顕微鏡(Motic DM−B1、Xiamen、China)下で得た。足場に隣接する血管の総数を、H&E切片内の血管をカウントすることによって定量化した[A. Minajeva, M. Kase, M. Saretok, A. Adamson−Raieste, S. Kase, K. Niinepuu, M Vardja, T. Asser, J. Jaal, Impact of Blood Vessel Quantity and Vascular Expression of CD133 and ICAM−1 on Survival of Glioblastoma Patients, Neurosci J. 2017 (2017) 8 pages]。簡単に言えば、足場に隣接する全ての識別可能な血管を、2人の独立した調査者が、2つの独立した顕微鏡を10倍の倍率で用いて、各群について合計6つの異なるスライド、および各スライド由来の6つの異なる注目領域からカウントした。
足場から20mm×10mmの断片をカットすることによって、生体力学的試験サンプルを調製した。装置のクランプを互いに10mm離して配置して、各足場の幅および厚さを測定した。試験サンプルを、張力計(BOSE Electroforce Test Instruments、Minnesota、USA)内の2つのグリップでクランプ留めした。各サンプルに、サンプルが破損するまで、0.1mm/秒の速度にて引張試験を実行した(n=4)。これらの試験の生データを用いて、応力−歪みグラフおよび荷重−変位グラフを描いた。極限引張強度(UTS)を、各サンプルの応力(σ)および応力(ε)曲線から算出した一方、剛性を、荷重(F)および変位(ΔL)曲線から算出した。
統計分析を、対応のないスチューデントt検定を用いて実行した。P値<0.05を統計的に有意とみなし、有意性の程度を星の数で示した(****P≦0.0001、***P≦0.001、**P≦0.01、*P≦0.05、nsP≧0.05)。
CAMに対するE2および2dDRの血管新生活性の評価
CAMのマクロ画像の定量化により、E2−100ng、E2−200ng、およびE2−600ngの群について、分岐点の数がそれぞれ1.3倍、1.5倍、1.4倍増大し、そして全ての濃度について、平均血管長が、4日にわたって対照と比較して、1.2倍増大することが示された。同様に、2dDR−20μgおよび2dDR−200μgについて、分岐点の数がそれぞれ1.3倍および1.4倍増大した。双方の濃度について、対照足場と比較して、平均血管長が1.2倍増大した一方、2dDR−1000μg群について、有意差はなかった。分岐点および平均血管長の定量化を、それぞれ図15Aおよび図15Bに示す。E2および2dDRの最も効果的な濃度でのCAMのマクロ画像を、図16に示す。
E2および2dDR放出PHBV足場のSEM画像を、図17に見ることができる。図17の右下隅のグラフに示すように、繊維の直径は、PHBV対照群(0.66±0.16μm)と比較した場合、全ての群内に物質を加えることによって(25mg E2(0.83±0.17μm)、50mg E2(0.98±0.35μm)、250mg 2dDR(0.89±0.19μm)、500mg 2dDR(1.22±0.28μm)をPHBV足場に加えた)、有意に増大した。
図18に示すように、足場からのE2および2dDRの放出速度を、30日にわたって評価した。7日目までに、足場からの2dDR放出は、250mgおよび500mgの2dDR足場について、ポリマー溶液中に存在する2dDRのそれぞれ81.3%および86.5%であった(図18A)。対照的に、7日以内の足場からの総E2の放出は、25mgおよび50mgのE2足場用のポリマー溶液中に存在する初期E2のそれぞれ1.3%および1.6%を表した(図18B)。
図19Aからわかるように、全ての物質を加えると、プレーンなPHBV足場と比較した場合、PHBV足場のUTSが有意に増大した。UTSの最も有意な増大は、2dDR 250mg群について観察された。同様に、2dDRおよびE2を担持した足場の剛性は、担持していないPHBV足場と比較して、有意に高かった。
CAM上のE2放出足場および2dDR放出足場の評価により、図20においてわかるように、プレーンなPHBV足場と比較して、全ての群について、足場に向かって成長する識別可能な血管の数が少なくとも2倍になることが示された。25mg E2担持足場および50mg E2担持足場についての平均血管数は、対照群(平均血管数:23.1(±1.24))と比較した場合、それぞれ49.5(±0.92)(****P≦0.0001)および37.9(±1.05)(****P≦0.0001)であった一方、250mgおよび500mgの2dDR担持足場について、それぞれ48.6(±1.02)(****P≦0.0001)および37.1(±1.37)(****P≦0.0001)であった。担持された物質はいずれも、胚生存率に影響せず、各群について75%を超えた。
足場に隣接する血管の平均数は、対照群およびCAMのみの群と比較した場合、E2放出足場および2dDR放出足場の双方の全ての濃度に応じて、有意に増大した(図21および図22参照)。
データから、2dDRおよびE2の直接的付与、ならびにPHBV繊維からのこれらの因子の漸次放出の双方が、ex−ovo CAMアッセイにおいて血管新生を刺激したと結論付けることができる。これらの2つの小さな安定因子は、例えば、組織工学的構築体に用いられ、かつ血管新生をin vivoで促進するTE足場の官能化に用いられる高い潜在性を有するような電界紡糸繊維中に容易に組み込まれた。

- 創傷治癒を促進するのに使用されるD−デオキシリボース糖であって、前記糖が、担体内に用意され、前記担体が、生体適合性マトリックス材料またはヒドロゲルである、D−デオキシリボース糖。
- 前記D−デオキシリボースが、2−デオキシリボースである、請求項1に記載の使用用のD−デオキシリボース糖。
- 前記担体が生分解性担体である、請求項1または2に記載の使用用のD−デオキシリボース糖。
- 前記マトリックス材料が、電界紡糸された足場である、請求項1〜3のいずれか一項に記載の使用用のD−デオキシリボース糖。
- 前記電界紡糸された足場が、ポリ乳酸(PLA)、ポリグリコリド(PGA)、ポリ(乳酸−co−グリコール酸)(PLGA)、またはポリ(3−ヒドロキシブチラート−co−3−ヒドロキシバレラート)PHBVの少なくとも1つを含む、請求項4に記載の使用用のD−デオキシリボース糖。
- 前記ヒドロゲルが、架橋ヒドロゲルである、請求項1または2に記載の使用用のD−デオキシリボース糖。
- 前記ヒドロゲルが、キトサン、ゼラチン、アルギナート、アガロース、メチルセルロース、ヒアルロナン、またはそれらのあらゆる組合せの少なくとも1つを含む、請求項1または6に記載の使用用のD−デオキシリボース糖。
- 前記ヒドロゲルが、キトサンおよびコラーゲンを含む、請求項7に記載の使用用のD−デオキシリボース糖。
- 前記ヒドロゲルが、ポリビニルアルコール、ポリアクリル酸ナトリウム、アクリラートポリマー、またはそれらのあらゆる組合せを含む、請求項7または8に記載の使用用のD−デオキシリボース糖。
- 前記ヒドロゲルが、キトサンおよびポリビニルアルコールを含む、請求項6〜9のいずれか一項に記載の使用用のD−デオキシリボース糖。
- 前記担体が、抗菌剤をさらに含む、請求項1〜10のいずれか一項に記載の使用用のD−デオキシリボース糖。
- 前記創傷が慢性創傷である、請求項1〜11のいずれか一項に記載の使用用のD−デオキシリボース糖。
- 前記創傷が全層創傷である、請求項1〜12のいずれか一項に記載の使用用のD−デオキシリボース糖。
- 前記創傷が火傷である、請求項1〜13のいずれか一項に記載の使用用のD−デオキシリボース糖。
- D−デオキシリボース糖を含む生体適合性マトリックス材料。
- 前記マトリックス材料が、電界紡糸された足場である、請求項15に記載の生体適合性マトリックス材料。
- 前記電界紡糸された足場が、ポリ乳酸(PLA)、ポリグリコリド(PGA)、ポリ(乳酸−co−グリコール酸)(PLGA)、またはポリ(3−ヒドロキシブチラート−co−3−ヒドロキシバレラート)PHBVの少なくとも1つを含む、請求項17に記載の生体適合性マトリックス材料。
- D−デオキシリボース糖を含むヒドロゲル。
- 前記ヒドロゲルが、キトサン、ゼラチン、アルギナート、アガロース、メチルセルロース、ヒアルロナン、またはそれらのあらゆる組合せの少なくとも1つを含む、請求項18に記載のヒドロゲル。

- 創傷治癒の促進に使用され、または脱毛症の処置に使用される、請求項15〜17のいずれか一項に記載の生体適合性材料。
- 創傷治癒の促進に使用され、または脱毛症の処置に使用される、請求項18〜22のいずれか一項に記載のヒドロゲル。
- 創傷床において血管化を増大させ、または誘導する方法に使用される、請求項15〜17のいずれか一項に記載の生体適合性材料。
- 創傷床において血管新生を増大させ、または誘導する方法に使用される、請求項18〜22のいずれか一項に記載のヒドロゲル。
- 脱毛症の処置に使用されるD−デオキシリボース糖であって、前記糖が、担体内に用意され、前記担体が、生体適合性マトリックス材料またはヒドロゲルである、D−デオキシリボース糖。
- 前記D−デオキシリボースが、2−デオキシリボースである、請求項27に記載の使用用のD−デオキシリボース糖。
- 前記担体が生分解性担体である、請求項27または28に記載の使用用のD−デオキシリボース糖。
- 前記マトリックス材料が、電界紡糸された足場である、請求項27〜29のいずれか一項に記載の使用用のD−デオキシリボース糖。
- 前記電界紡糸された足場が、ポリ乳酸(PLA)、ポリグリコリド(PGA)、ポリ(乳酸−co−グリコール酸)(PLGA)、またはポリ(3−ヒドロキシブチラート−co−3−ヒドロキシバレラート)PHBVの少なくとも1つを含む、請求項30に記載の使用用のD−デオキシリボース糖。
- 前記ヒドロゲルが架橋ヒドロゲルである、請求項27または28に記載の使用用のD−デオキシリボース糖。
- 前記ヒドロゲルが、キトサン、ゼラチン、アルギナート、アガロース、メチルセルロース、ヒアルロナン、またはそれらのあらゆる組合せの少なくとも1つを含む、請求項27または32に記載の使用用のD−デオキシリボース糖。
- 前記ヒドロゲルが、キトサンおよびコラーゲンを含む、請求項33に記載の使用用のD−デオキシリボース糖。

- 毛の再生を促進する非治療的方法であって、前記方法が、D−デオキシリボース糖の投与を含み、前記糖が、担体内に用意され、前記担体が、生体適合性マトリックス材料またはヒドロゲルである、方法。
- 前記D−デオキシリボースが、2−デオキシリボースである、請求項38に記載の毛の再生を促進する非治療的方法。
- 前記担体が生分解性担体である、請求項38または39に記載の毛の再生を促進する非治療的方法。
- 前記マトリックス材料が、電界紡糸された足場である、請求項38〜40のいずれか一項に記載の毛の再生を促進する非治療的方法。
- 前記電界紡糸された足場が、ポリ乳酸(PLA)、ポリグリコリド(PGA)、ポリ(乳酸−co−グリコール酸)(PLGA)、またはポリ(3−ヒドロキシブチラート−co−3−ヒドロキシバレラート)PHBVの少なくとも1つを含む、請求項41に記載の毛の再生を促進する非治療的方法。
