Description
本発明は、表面上または溶液中に存在する核酸を不活化する際に使用するための剤を含むか、またはこの剤を使用する処方物、方法、およびキットに関する。
試験サンプルにおいて特定の生物もしくはウイルスの存在を定性的または定量的に決定するための手順は、慣例的に、核酸ベースのプローブ試験に依存している。これらの手順の感度を上昇させるために、試験サンプル中に存在する潜在的な核酸標的配列のコピー数を増加させるための増幅工程が、しばしば包含される。増幅の間、標的配列および/またはその相補体を含むポリヌクレオチド鎖は、ポリメラーゼとして公知のヌクレオチジルトランスフェラーゼを使用して、リボヌクレオシド三リン酸またはデオキシヌクレオシド三リン酸から鋳型依存様式で合成される。今日、一般的に使用される多くの増幅手順が存在する。これらの手順としては、ポリメラーゼ連鎖反応(PCR)、Q−βレプリカーゼ、自立的配列複製(3SR)、転写媒介性増幅(TMA)、核酸配列ベースの増幅(NASBA)、リガーゼ連鎖反応(LCR)、鎖置換増幅(SDA)およびループ媒介性等温増幅(LAMP)が挙げられ、これらの各々は、当該分野で周知である。例えば、以下を参照のこと:Mullis、「Process for Amplifying Nucleic Acid Sequences」(特許文献1);Erlichら、「Kits for Amplifying and Detecting Nucleic Acid Sequences」(特許文献2);非特許文献1;非特許文献2;Kacianら、「Nucleic Acid Sequence Amplification Methods」(特許文献3);Daveyら、「Nucleic Acid Amplification Process」(特許文献4);Birkenmeyerら、「Amplification of Target Nucleic Acids Using Gap Filling Ligase Chain Reaction」(特許文献5);Marshallら、「Amplificationof RNA Sequences Using the Ligase Chain Reaction」(特許文献6);Walker、「Strand Displacement Amplification」(特許文献7);Notomiら、「Process for Synthesizing Nucleic Acid」(特許文献8);Dattaguptaら、「Isothermal Strand Displacement Amplification」(特許文献9);および非特許文献3。
本発明は、溶液中または固体表面上で処方物と接触する核酸を不活化するのに十分な量の核酸不活剤(「不活剤」)を含むか、またはこの不活剤と合わせられ得る処方物を提供することによって、この目的を満たす。「不活化する」によって、核酸が、その核酸が不活化される前に機能したようにはもはや機能し得ないように変更されることを意味する。例えば、その核酸は、もはや、増幅反応において鋳型として機能することも、または別の方法で(例えば、プライマー−ダイマー形成を通して)干渉することも、別の核酸もしくはタンパク質へ結合することも、あるいは酵素の基質として機能することもできない可能性がある。用語「不活化する」は、不活剤の処方物が核酸を変更する任意の特定の機構を意味しない。上記処方物の成分としては、腐食阻害剤、湿潤剤、可溶化剤、および必要に応じて不活剤が挙げられる。処方物が4種全ての成分からなる場合、腐食阻害剤は、不活剤の腐食特性を低減させるために十分な量で存在し、湿潤剤は、不活剤の分散特性を改善するため、そして/あるいは不活剤および/または固体表面上もしくは溶液中に存在し得る他の物質の溶解度を増加させるために十分な量で存在し、そして可溶化剤は、不活剤もしくは腐食阻害剤もしくは湿潤剤、または種々のそれらの組み合わせの溶解度を増加させるために十分な量で存在し、そして不活剤は、その処方物に接触された核酸を実質的に不活化するのに十分な量で存在する。上記処方物が不活剤を含まない場合、腐食阻害剤、湿潤剤および可溶化剤の量は、その不活剤と、核酸を不活化し得る最終作業溶液を作製するために使用され得る任意の希釈剤(例えば、水)とが合わされたときに、それらの濃度の減少を計算して濃縮される。
本発明は、一部には、核酸を不活化するために有用な処方物、方法およびキットに関する。これらの処方物、方法およびキットは、上記に記載されており、そして以下の実施例および特許請求の範囲にも記載されている。さらに、実施例は、加工表面上、実験機器上および/もしくは溶液中の核酸を不活化するために有用であるか、または例えば消毒剤として使用され得る本発明の処方物を選択するためのスクリーニング方法を記載する。このような処方物はまた、タンパク質および脂質のような生物学的分子を不活化するためにも有用であり得る。実施例では、増幅前の適用および増幅後の適用の両方における多くの例示的な処方物の効果をさらに考察する。
(ゲル電気泳動によるDNAの可視化に対する種々の濃度の漂白剤の効果)
71マーのDNAオリゴヌクレオチドを、6.15%(w/v)の次亜塩素酸ナトリウムの濃度において、種々の濃度のUltra Chlorox(登録商標)Bleach(The Chlorox Company;Oakland,CA)と反応させて実験を実施し、そしてポリアクリルアミドゲル電気(PAGE)を使用してその反応生成物を分析した。173μg/mLの濃度の2μLのDNAオリゴヌクレオチドをサンプルバイアル中の希釈水と混合することによって10個のサンプルを調製し、その後種々の濃度の漂白剤を添加して各サンプルの総容量を20μLとした。このサンプルを約10秒間ボルテックスによって混合し、次いで180mM Tris塩基、180mM ホウ酸、4mM エチレンジアミン四酢酸(EDTA)(pH8.0)(Invitrogen Corporation,Carlsbad,CA;カタログ番号LC 6876)を含む2×TBE尿素サンプル20μLを提供して、各サンプルの総容量を40μLとした。再度、このサンプルを約10秒間ボルテックスして混合した。サンプル中の漂白剤の最終濃度は、以下の表1に記載されるように0%〜50%の漂白剤の範囲であった。各サンプルの10μLアリコートを、10%ポリアクリルアミドTBE尿素ゲルの10レーンのうちの1つにロードし、そのゲルを、40分間180Vで泳動した。泳動を完了させて、ゲルをそのキャストからはずし、蒸留水で10,000分の1に希釈したSYBR(登録商標)Green I核酸ゲル染色(Molecular Probes,Eugene OR;カタログ番号S7563)100mLを接触させ、そして30分間10rpmで混合した。染色の後、ゲルをChemiImagerTM System 4400(Alpha Innotech Corporation,San Leandro,CA)を使用して撮影した。ゲル上で染色した分離した生成物は、通常、バンドといわれる。結果の電気泳動図のコピーを図1に提示する。
(ゲル電気泳動によるDNA/RNAキメラの可視化に対する種々の濃度の漂白剤の効果)
実施例1の実験のDNAオリゴヌクレオチドを200μg/mlの濃度の60マーDNA/RNAキメラオリゴヌクレオチドで代用して、実施例1の実験を繰り返した。このキメラのRNAは、2’−O−メチルリボヌクレオチドからなった。結果の電気泳動図のコピーを図2に示す。これは、オリゴヌクレオチドバンドの大部分は0.1%の漂白剤で消え、次亜塩素酸ナトリウム対ヌクレオシドの1:1のモル比について再び示す。この実験のゲルの種々のレーンにおいて使用した漂白剤の濃度は、実施例1の実験で記載したものと同じである。
(ゲル電気泳動によるDNAの可視化に対する種々の濃度の漂白剤の効果)
ジクロロイソシアヌル酸ナトリウム塩(DCC)(Sigma−Aldrich,Milwaukee,WI;Prod.番号21,892−8)およびUltra Chlorox(登録商標)Bleach(6.15%(w/v)次亜塩素酸ナトリウム)を、この実験において、核酸と反応するその比較能力について、種々の利用可能な塩素濃度で試験した。試験した塩素濃度を以下の表2に記載する。71マーDNAオリゴヌクレオチドの使用を包含する全ての他の局面において、この実験は、実施例1に詳述した実験と同一であった。
(ゲル電気泳動によるDNA可視化に対する種々の濃度の過酸化水素および過酸化水素+硫酸銅の効果)
この実験において、53μg/mLの濃度で存在する71マーDNAオリゴヌクレオチドを、種々の濃度の30%(w/v)過酸化水素(Fisher Scientific,Tustin,CA;カタログ番号BP2633−500)および30%(w/v)過酸化水素+硫酸銅(Sigma−Aldrich,Milwaukee,WI;Prod.番号45,165−7)と反応させ、その反応生成物をポリアクリルアミドゲル電気泳動(PAGE)を使用して分析した。DNAと水とを合わせた後に30%(w/v)過酸化水素(8.8M)および/または1mM硫酸銅を添加して、10個のサンプルを以下の表3に示す様式で調製した。この実験の残りの手順の詳細は、実施例1に記載したものと同じである。以下の表4に各レーン中の過酸化物の濃度を記載する。
(ゲル電気泳動によるDNAの可視化に対する、硫酸銅の存在下での種々の濃度の過酸化水素の効果)
実施例4の実験を、ゲルの全てのレーンでより低い濃度の過酸化水素成分および100μM硫酸銅を使用して繰り返した。ゲルの各レーンの過酸化物の最終濃度を以下の表5に記載する。
(漂白剤のDNAとの反応に対するNALCの効果)
漂白剤は、種々の有機物質と反応することが公知である。従って、これらの物質は、漂白剤と反応し、その漂白剤を消費することによって、核酸の不活化に干渉し得る。従って、これらの有機物質の存在は、DNAと有機物質との両方と反応するのに十分な漂白剤の存在によって補償されなければならない「有機負荷」を構成する。この実験において、有機負荷化合物(すなわち、漂白剤を消費することが予想され得る化合物)であるN−アセチル−L−システイン(NALC)の効果の除去を、種々の濃度のUltra Chlorox(登録商標)Bleachの存在下で試験した。NALCは、増幅反応での使用が意図されるいくつかの酵素試薬において見出される還元剤である。この実験において、10個のサンプルの2つのセットを調製し、各サンプルは、173μg/mLの濃度で2μLの71マーDNAオリゴヌクレオチドを含んだ。第1のセットのサンプルは、NALCを含まなかったが、第2のセットのサンプルの各サンプルは、11.4μg/mlの濃度で16μLのNALCを含んだ。このサンプルを、まずDNAおよびNALC(存在する場合)をサンプルバイアルに提供し、約10秒間のボルテックスによりNALCを含有するサンプルを混合することによって調製した。次いで、種々の濃度で両方のセットのサンプルに漂白剤を添加し、蒸留水を加えて各サンプルの総容量を20μLにした。このサンプルを約10秒間ボルテックスすることによって混合し、その後20μLの2×TBE尿素サンプル緩衝液(Invitrogen Corporation;カタログ番号LC 6876)を添加して各サンプルの総容量を40μLにした。このサンプルを約10秒間ボルテックスすることによって再度混合した。このサンプル中の漂白剤の最終濃度は、以下の表6に記載するように、0%〜50%の漂白剤の範囲であった。10%ポリアクリルアミドTBE尿素ゲル(2セットのサンプルの各々について提供する分離ゲル)の10レーンのうちの1つに、各サンプルの10μLアリコートをロードし、そしてそのゲルを180Vで40分間泳動した。泳動を完了し、ゲルをそのキャストからはずし、蒸留水で10,000分の1に希釈したSYBR(登録商標)Green I核酸ゲル染色(Molecular Probes;カタログ番号S7563)100mLと接触させ、そして30分間10rpmで混合した。染色の後、ゲルをChemiImagerTM System 4400を使用して撮影した。結果の電気泳動図のコピーを図6に提示する。
(過酸化水素および硫酸銅の混合物とDNAとの反応に対するNALCおよびヒト血清の効果)
この実験において、種々の濃度の過酸化水素および硫酸銅とDNAとの反応に対する、NALCおよびヒト血清の効果を試験した。11個のサンプルのセットを調製し、各サンプルは、53μg/mLの濃度で2μLの71マーDNAオリゴヌクレオチドを含んだ。このサンプルの他の成分は、100μM硫酸銅、30%(w/v)過酸化水素、および11.4mg/mLの濃度のNALCを含んだ。サンプルバイアル中の各成分の量を以下の表7に記載する。サンプルバイアル中で過酸化水素を除いた全てのサンプル成分を合わせ、約10秒間のボルテックスにより混合することによって、サンプルを調製した。混合の後、過酸化水素を種々の濃度でサンプルに添加して、サンプル1の総容量を20μLとし、そしてサンプル2〜11を22μLとして、以下の表8に示す最終濃度とした。このサンプルを約10秒間ボルテックスすることによって再度混合し、その後20μLの2×TBE尿素サンプル緩衝液(Invitrogen Corporation;カタログ番号LC 6876)を添加して、サンプル1の総容量を40μLとし、そしてサンプル2〜11のサンプルを42μLとした。この手順の残りおよび試薬の供給源は、上記の実施例6に記載したものと同一である。結果の電気泳動図のコピーを図7に提示する。
(純粋系における核酸の漂白剤との反応)
この実験を実施して、純粋系において種々の漂白剤濃度がNeisseria gonorrhoeae(「標的」)に由来する精製リボソームRNAを不活化する能力を評価した。4μLの標的含有水および表9に示される濃度の4μLの漂白剤を含むように、8個のサンプルチューブを最初にセットした。サンプルチューブ6および8については、漂白剤の代わりに4μLの水を使用した。この実験において使用した漂白剤は、Ultra Cholorox(登録商標)漂白剤(6.15%(w/v)次亜塩素酸ナトリウム)であった。セットした後、サンプルチューブの内容物を室温で5分間インキュベートした。
(純粋漂白剤系における核酸不活化のさらなる特徴付け)
多重アッセイを使用して、核酸を不活化する際の有効性について、数個の処方物を試験した。
純粋系においてNeisseria gonorrhoaea(Ngo)リボソームRNA(rRNA)を、0〜20%の市販の漂白剤と反応させ(最も低い漂白剤濃度は、0.2%であった)、反応生成物をリアルタイムTMAアッセイによって分析した(実施例8を参照のこと)。最も低い漂白剤濃度においてさえ、リアルタイムアッセイの感度の制限内でrRNAを不活化した(図9A)。
リボソームRNAを溶液中の漂白剤と反応させ、生成物をキャピラリー電気泳動によって分析した。Agilent 2100 Bioanalyzerを利用して、不活化溶液に曝した核酸を特徴付けた。10μLの総反応物に、以下のものを(順番に)添加した:(a)Milli−Q H2Oまたは緩衝液、(b)指示された量の試薬(例えば、漂白剤またはH2O2由来の−OCl)、および(c)0nM(ブランク)または150nM(718μM=470ng/μL nt)Mycobacterium tuberculosis(Mtb)rRNAまたは15nM(71.8μM=47.0ng/μL nt)Mtb rRNA。この反応物を10分間室温(約23℃)でインキュベートし、次いで90μLの1mM アスコルビン酸ナトリウム(最終900μM)を添加した。LabChip(登録商標)プロトコル(Agilent Technologies,Inc.;Palo Alto,CA)においてと同様に、RNAラダーを70℃で2分間変性し、次いで各反応物の1μLを、5μLのサンプル緩衝液を含むRNA 6000 Nano LabChip(登録商標)またはPico LabChip(登録商標)(Agilent Technologies,Inc.;Palo Alto,CA)のウェルにロードした。この成分を混合し、アッセイの原核生物の総RNAをBio Sizingプログラム(Agilent Technologies,Inc.;Palo Alto,CA)で泳動した。
純粋系における漂白剤(次亜塩素酸塩)と核酸との反応は速く、本質的に1:1の比の次亜塩素酸塩対ヌクレオチドで完了する。これらのデータは、漂白剤を使用して実験室中の核酸の汚染除去が全く観察されないことは、次亜塩素酸塩と核酸とのゆっくりとした反応に起因するのでも、漂白剤が核酸に対して非常にモル過剰であることについての必要性に起因するのでもないことを示唆した。
(有機負荷の存在下における漂白剤による核酸不活化のさらなる特徴付け)
漂白剤とオリゴヌクレオチドとの間の反応に対する、有機負荷物質であるN−アセチル−L−システイン(NALC)の効果を、実施例6のPAGEによって特徴付けた。この後に提示するのは、PAGEおよび他の特徴付け方法を使用した、オリゴヌクレオチドと漂白剤との間の反応に対するNALCおよび他の有機負荷物質の効果の特徴である。
異なる量の種々の有機負荷物質の存在下において、リボソームRNAを漂白剤と反応させた。次いで、このRNAが増幅する能力をリアルタイムTMAを使用して試験した。有機負荷物質は、以下を含んだ:APTIMA COMBO 2(登録商標)アッセイキット(カタログ番号1032;Gen−Probe Incorporated;San Diego,CA)からの増幅試薬、ハイブリダイゼーション試薬、酵素試薬、および選択試薬、ならびにこれらの混合物、尿運搬(urine transport)培地(UTM;カタログ番号1040、男性および女性の尿標本のためのAPTIMA COMBO 2(登録商標)アッセイ尿標本収集キット;Gen−Probe Incorporated)、スワブ運搬(swab transport)培地(STM)、KOVA−TrolTM(Hycor Biomedical Inc.;Garden Grove,CA)、ウシ血清アルブミン(BSA)、ラウリル硫酸リチウム(LLS)およびヒト血漿。これらの化合物のうち、UTMおよび酵素試薬は、漂白剤とRNAとの反応を干渉するのに最も有効であった。1つの実験において、20%の市販の漂白剤が、UTMの効果を克服するのに必要とされ、これは、有機負荷物質の非存在下におけるrRNAと0.016%の漂白剤との非常に迅速で完全な反応と対照的である(図10A〜図10F)。
実施例1に開示した手順と類似の手順を使用して、PAGEを実施した。手短には、既知量の71マーオリゴヌクレオチドを、既知濃度の候補試薬を含む処方物とインキュベートした。2×TBE尿素ローディング緩衝液(180mM Tris,180mM ホウ酸,4mM EDTA,pH8.0)の1×容量を、混合物溶液に添加して10秒間ボルテックスした。10% ポリアクリルアミドTBE尿素ゲルの各レーンに、10μLのサンプルをロードした。このゲルを、オリゴヌクレオチドの長さに依存して、35〜40分間180Vで1×TBEランニング緩衝液中で泳動した。次いで、このゲルをキャストからはずし、1/10,000のSYBr Green I色素溶液中で20分間染色した。染色したゲルをChemiImagerTM 4400を使用して画像化した。
逆相(RP)HPLCを利用して、標準的な手順を使用して不活化溶液に曝された核酸を特徴付けた。HPLC機器についての規格および利用した方法論は、以下の通りであった。4.6mmの内径および15cmの長さを有するZORBAX(登録商標)Eclipse XDB C−8逆相カラム(Agilent Technologies,Inc.;Palo Alto,CA)を利用した。トリアセチルアンモニウムアセテート(TEAA)/アセトニトリルを移動相として利用した。ここで緩衝液Aは、0.1M TEAAを含み、緩衝液Bは、100%アセトニトリルを含んだ。15分の時間間隔で0.5ml/分の流速において、5%〜100%の緩衝液Bの勾配を利用した。2.0 OD(オリゴヌクレオチド26マー=10μM)の光学密度を有する50μLのオリゴヌクレオチドサンプルをカラムに注入し、カラム排出量を254nmの波長で検出した。
漂白剤と核酸との反応に効果的に干渉する物質は、尿運搬培地(UTM)および酵素希釈緩衝液(EDB)中のNALCであった。漂白剤と核酸との反応に適度に干渉する物質は、スワブ運搬培地(STM)、ハイブリダイゼーション試薬、増幅試薬およびヒト血清であった。漂白剤と核酸との反応に弱く干渉する(または全く干渉しない)物質は、選択試薬、APTIMA COMBO 2(登録商標)アッセイ標的捕捉試薬、ラウリル硫酸リチウムおよびKOVA−TrolTMであった。このアッセイから、有機負荷物質(特に、一次アミンおよびスルフヒドリル基を含む物質)が、漂白剤と反応して漂白剤を消費し、その結果、核酸を不活化するために全く利用できなくなることが決定された。より低い濃度における漂白剤の汚染除去力の喪失は、反応速度が遅いことに起因してもいないし、ヌクレオチドに対して過剰の次亜塩素酸塩が必要とされることに起因してもいないが、それよりは他の化合物による漂白剤の消費に起因する。

漂白剤に対する代替的な処方物(例えば、ジクロロイソシアヌル酸(DCC)または過酸化水素および銅イオンを含む溶液)を、PAGEによって実施例3および実施例4において特徴付けた。この後に記載されるように、PAGEおよび他のアッセイによって、これらの代替的処方物およびさらなる代替的処方物を特徴付けた。
71マーオリゴヌクレオチドを種々の候補化合物と反応させ、その生成物をPAGEを使用して分析した。他の処方物のなかで、DCCまたは過酸化水素と硫酸銅とを含む溶液を試験した。実施例3に示すように、漂白剤よりも腐食性が低いDCCは、より有効とはいかないまでも、オリゴヌクレオチドを不活化するための漂白剤と同程度に有効であった。スカベンジャー(酵素希釈緩衝液(EDB)および血清を含む)のDCCに対する効果をまた、試験し、そして漂白剤に対する効果と比較した。同様の効果を図12に示すように観察した(結果は、EDBについてのもので、血清についての結果は示さず)。実施例4、5、および7に示すように、過酸化水素および硫酸銅を含む溶液(これは、においがなく、かつ腐食性もない)は、(1)オリゴヌクレオチドの移動またはオリゴヌクレオチドのバンドの保持の変化、および(2)有機負荷の効果の克服において合理的に有効であった。
RP−HPLC保持シフトアッセイ(前述)を使用して、有機負荷物質(NALC)の存在下または非存在下において、いくつかの漂白剤の代替的候補をスクリーニングした。以下は、10%漂白剤と比較した場合の試験した代替的処方物の有効性の要約である。ここで、「=」は、おおよそ等価なものであり、「<」は、効果が低いものであり、そして「>」は、効果が高いものである。
リボソームRNAを溶液中で種々の候補処方物と反応させ、その生成物をキャピラリー電気泳動アッセイを使用して分析した。このアッセイにおいて、別々に試験した1mM ジクロロイソシアヌレート(DCC)および17.5mM ペルオキシモノサルフェート(Virkon S(登録商標);DuPont Animal Health Solutions,United Kingdom)は、0.72mM rRNAオリゴヌクレオチドに対応するピークを実質的に消去した。インサイチュで産生されたCl2(10mM ペルオキシモノサルフェート+20mM KCl)は、部分的に72μM rRNAオリゴヌクレオチドを消失した。別々に試験すると、(a)インサイチュで産生されたBr2(10mM ペルオキシモノサルフェート+20mM KBr)、(b)10μMと100μMとの間のジクロロヒダントインまたはジブロモヒダントイン、(c)10μMと100μMとの間の次亜臭素酸塩、および(d)10mM ペルオキシモノサルフェート+金属イオン(1mM Cu2+、1mM Fe2+または10mM Fe2+)は、実質的に72μM rRNAオリゴヌクレオチドを消失した。
リボソームRNAを溶液中で種々の化合物と反応させ、次いで、RNAが増幅する能力を実施例8に記載したリアルタイムTMAアッセイを使用して試験した。特定の処方物の有効性を以下に記載する。

以下は、特徴付け手順のために使用したいくつかの分析プロセスの記載である。
油状試薬(200μl)を80個の反応チューブ(12×75mm)に添加した。Chlamydia trachomatis(CT)およびNeisseria gonorrhoeae(GC)のrRNAの4.2×1010コピーを、3.15mlの再構成した増幅試薬に加えた。この加えた増幅試薬の75μl(1×109コピー(約2.5ng)を反応チューブのうちの40個に添加した(ポジティブサンプル)。標的を含まない増幅試薬(ネガティブサンプル)75μlを残りの40個のチューブに添加した。全ての80個のサンプルを10分間62℃でインキュベートし、次いで5分間42℃でインキュベートした。25μlの再構成した酵素試薬を各チューブに添加し、そのラックを水浴から取り出し、ラックを振盪してチューブの内容物を混合し、次いでラックをすばやく水浴に戻した。反応チューブの内容物を60分間42℃でインキュベート(増幅)し、次いで10分間80℃でインキュベートした(酵素の不活性化)。ポジティブサンプルのうちの38個とネガティブサンプルのうちの38個とをプールし、各プールから油を除いた。2個の残ったポジティブサンプルおよびネガティブサンプルを、標準的なAPTIMA COMBO 2(登録商標)マニュアルアッセイプロトコル(上記に記載される)に従ってアッセイした。
標準的な手順によって調製したCTおよびGCのrRNAの5×108コピーを、100μlのUTM:KOVA−TrolTM(1:1比)に添加した((以下の表に示した)いくつかの場合において、サンプルを100μlのSTMに添加した)。この混合物の所望の数の複製物を、表面上にスポットする前のプールとして調製し得る。
表面。汚染除去アッセイを、ChemSurf実験室ベンチの2×4ft切片上(「表面」)で行った。種々の実験の前、その間、およびその後に、その表面を50%の漂白剤溶液(水で1:1希釈した家庭用液体漂白剤(例えば、Chlorox(登録商標)Ultra Bleach))で洗浄し、その後水でリンスした。ペーパータオルまたは大きなKimwipeで拭った。

2)ネガティブコントロールのために、直径約1.5インチの円形領域からサンプルを収集し、表面をランダムに選択して、その後任意のポジティブサンプルを表面に適用した。
3)UTM:KOVA−TrolTM(1:1)(またはSTM)サンプル中のおよそ8個の複製物のCT rRNAおよびGC rRNA(各100μl)をスポットし、表面に均一に空間を空けて配置した。
4)以下のように、スポット1を上記の汚染除去条件「a」(10%漂白剤、1回適用)で処理した:サンプルを含む領域(中心で約7×7インチ平方のサンプル)を、およそ2mlの試薬で湿らせ((下の表に示すような)いくつかの場合では、およそ3mlを使用した)、次いで乾燥するまでペーパータオルまたは大きなKimwipeで即座に拭った(完全に乾燥させるために必要な場合、プロセスの間そのタオルを時々ひっくり返した)。タオルおよび拭いを行った手にはめたグローブを注意深く廃棄した(汚染をする可能性がある場合、もう一方のグローブも廃棄した)。適用したもとのスポットからのサンプルを、上記に記載した頸管内スワブを使用して収集した。
5)上記「4」で記載したものと同じ一般的方法を使用して、スポット2を条件「b」で処理したが、汚染除去試薬の第2の適用によっても処理した。
6)次いで、表面上の全てのサンプルを処理するまで、上記に列挙した汚染除去条件でサンプルスポットを処理した。
7)上記に記載のように表面を洗浄し、そして十分な数のサンプル複製物を適用して汚染除去条件+1つのさらなるスポット(ポジティブコントロールとして使用した)の試験を行った。
8)次いで、汚染除去条件の試験を行った。
9)最後に残ったサンプルスポット(ポジティブコントロール)に対して、汚染除去試薬をいずれも適用することなくそのスポットを直接拭った。
10)ネガティブ増幅サンプルおよびポジティブ増幅サンプルに対して、工程1〜9を行った。
1.ドッキングカラー(docking collar)を使用して試薬を再構成する。試薬再構成溶液を含む増幅試薬、酵素再構成試薬を含む酵素試薬、およびプローブ再構成試薬を含むプローブ試薬を再構成する。
2.標的捕捉試薬(TCR)成分Bを標的捕捉試薬に1:100希釈に希釈し、手で十分に混合する。
3.100μLのTCR:成分Bの混合物をTen−Tube Unit(TTU,カタログ番号TU002;Gen−Probe Incorporated)の各反応チューブに分配する。
4.キャップに穴を開け、以下の順番で400μLのコントロールを適切なチューブにピペットで入れる:チューブ1(CTポジティブコントロール)、次いでチューブ2(GCポジティブコントロール)。
5.各サンプルの400μLをTTUの適切なチューブに移す。
6.全てのサンプルを適切なラック(カタログ番号4579;Gen−Probe Incorporated)にロードし、TTU上にシーリングカードを配置し、そしてサンプルを手で穏やかに振盪して混合する。ラックをボルテックスしてはならない。
7.62℃の水浴で30分間インキュベートする。
8.ラックをベンチに置き、30分間インキュベートする。
9.Ten−Tipカセット(カタログ番号4578;Gen−Probe Incorporated)を含むTarget Capture System(TCS、カタログ番号5210、Gen−Probe Incorporated)をロードする。洗浄ボトルが、ポンプに接続されているのを確実にする。
10.洗浄試薬を2回流して、ポンプラインを満たす。
11.TCS磁気ベース上にラックを配置し、シーリングカードを除いて新しいカードで覆う(下に置いてはならない)。5分間インキュベートする。
12.アスピレーターのバキュームのスイッチを入れる。真空計は、そのシステムを正しくセットして9インチHgと11インチHgの間でなければならない。吸引マニフォールドを低くすることによって、全ての液体をチューブの底からゆっくりと吸引する。チューブの底をチップで軽く叩く。チューブから全ての気泡を除くまで吸引する。
13.洗浄ボトルを一度ポンピングすることによって、1.0mlの洗浄試薬を各チューブに添加する。
14.チューブをシーリングカードで覆い、マルチボルテックス(multi−vortexer)でボルテックスする。
15.TCS磁気ベース上に5分間ラックを置く。
16.全ての液体を吸引する。
17.75μLの再構成した増幅試薬を添加する。
18.200μLの油状試薬を添加する。
19.チューブをシーリングカードで覆い、マルチボルテックスでボルテックスする。
20.62℃の水浴で10分間ラックをインキュベートする。
21.42℃の循環水浴にラックを移し、5分間インキュベートする。
22.水浴中のラックに関して、シーリングカードを除き、全ての反応物に25μLの酵素試薬を添加する。
23.直ちにシーリングカードで覆い、しばらくの間水浴から出して、手で穏やかに振盪して反応物を混合する。
24.42℃で60分間ラックをインキュベートする。
25.ラックを水浴から除き、HPA領域に移す。100μLの再構成したプローブ試薬を添加する。
26.マルチボステックスでボルテックスする。
27.62℃の循環水浴中で20分間、ラックをインキュベートする。
28.水浴からラックを出し、ベンチの上部で室温で5分間インキューベートする。
29.250μLの選択試薬を添加する。
30.チューブをシーリングカードで覆い、マルチボルテックスでボルテックスする。
31.62℃の循環水浴中で10分間ラックをインキュベートする。
32.ベンチの上部で室温で15分間ラックをインキュベートする。
33.LEADER(登録商標)HC+Luminometer(カタログ番号4747;Gen−Probe Incorporated)Comboソフトウェアにおいて反応物を明らかにする。
以下の表は、上記に記載のプロトコルを使用して収集した結果を示す。「NA供給源」とは、核酸供給源であり、「#App」とは試薬適用の回数であり、「kRLU」とは、1000で割った相対発光単位(relative light unit)であり、そして「pip」とは、ピペラジンである。予想したCtrおよびNgoの結果は、Ngo/Ctr rRNAについてネガティブであり(Neg)、Pos増幅についてNegであり、そしてNeg増幅についてポジティブ(Pos)であった。この試験からのCtrおよびNgoの結果の大部分は、有効であり、無効の結果は、表にふくまない。
(核酸不活化処方物の特徴付け)
核酸不活化処方物において、腐食インヒビター、界面活性剤および芳香剤を含むことの効果をアッセイした。漂白剤、および漂白剤ほどではないにせよ依然として顕著な程度のDCCは、金属および他の物質を腐食させる。種々の候補の抗腐食性化合物(リン酸(PB)、ホウ酸、炭酸水素およびドデシル硫酸(SDS)のナトリウム塩が挙げられる)を、リアルタイムTMAおよびPAGEを使用した分析の前に試験した(例えば、実施例8および実施例1)。候補の腐食インヒビターと一緒に混合した場合の漂白剤およびDCCの活性を試験するための研究を行った。pH6.4およびpH7.5のリン酸は漂白剤を不安定化したが(活性の喪失が時間とともに増加した)、pH9.1およびpH9.5のリン酸は漂白剤を不安定化しなかった。DCCについては、逆が真であり、高いpH(9.1および9.5)のリン酸は不活化される一方、低いpH(6.4および7.5)のリン酸は不安定化されなかった。漂白剤は、pH7.6またはpH9.1のホウ酸中で安定であり、そしてpH9.3の炭酸水素中で安定であった。SDSは、漂白剤の活性に対していずれの明らかな効果も有さなかった。
腐食インヒビター/洗剤/芳香剤(6.7×濃度):600mM炭酸水素塩(pH9.3)、0.1% SDS、0.05% 2141−BG
完成した汚染除去試薬:0.6%次亜塩素酸塩、90mM炭酸水素塩(pH9.3)、0.015% SDS、0.0075% 2141−BG。
(核酸不活化処方物の安定性)
選択した試薬を種々の条件下で保存した。選択した時点で、溶液アッセイを使用して、標的核酸を不活化する能力について、その処方物をアッセイした。この溶液アッセイにおいて、rRNAを、溶液中で試薬とインキュベートし、希釈し、そいてリアルタイムTMA(実施例8)を使用してアリコートをアッセイした。インキュベーション条件は、光から保護せずに、室温だった。結果を以下に提供する。
(A.600mM 炭酸水素ナトリウム(pH9.3)/0.1% SDS/0.05% 2141−BG(6.7×溶液))
(実験機器の核酸の不活化)
実験機器のいくつかの部品(真空トラップ系、吸引マニフォールド、ラックおよびデッキが挙げられる)の核酸を不活化するための処方物および手順を、有効性について評価した。
吸引マニフォールド、2つのトラップ、インラインフィルター、および直列でチュービングによって接続した真空ポンプを備える真空系を、増幅アッセイを行うために利用し、その後、複数の標的捕捉を行う(Ctr rRNAおよびNgo rRNAの両方)。第1のトラップに漂白剤を添加せずに、汚染を評価した。この後、スワブサンプルを真空系の種々の位置から採取し、実施例8に提示したリアルタイムTMAアッセイを使用してCtr rRNAおよびNgo rRNAの存在についてアッセイした。Ngo rRNAによる検出可能な汚染がないことを、第1のトラップの外面で同定した。Ctr rRNAによる汚染を、第1のトラップと第2のトラップとの間のチュービング、第2のトラップ、および第2のトラップとインラインフィルターとの間のチュービングで同定し、そしてインラインフィルターの後ろで汚染は検出しなかった。これらの結果は、周囲から漏れた検出可能なNgo rRNAまたはCtr rRNAはなく、従って使用する間に、第1のトラップ中に漂白剤を含まないようである。
TMAアッセイ(APTIMA COMBO 2(登録商標)アッセイ、カタログ番号1032、Gen−Probe Incorporated、San Diego、California)に利用した標的捕捉吸引マニフォールドを汚染除去するための1つのプロトコルは、マニフォールドを50%漂白剤に10分間浸漬し、次いで水で完全に洗浄する工程を包含した。この手順により、マニフォールドは腐食し、比較的頻繁にそのマニフォールドを置き換える必要性が生じた。
主要な漂白剤の代替候補を、DTSTM TECAN GENESIS System(カタログ番号5216またはカタログ番号5203;Gen−Probe Incorporated;San Diego,CA)のデッキの汚染除去について試験した。以下の結果を観察した。
(2つの実験部位における核酸不活化の処方物および方法の有効性)
臨床実験の設定において2種の汚染除去の試薬および方法の有効性を、2つの部位において特徴付けた。試薬1(3% H2O2、2mM 硫酸銅)および試薬2(0.6%次亜塩素酸塩、90mM炭酸水素塩、0.015% SDS、0.0075% 2141−BG)を、各部位に提供された所定のプロトコル(以下を参照のこと)に従って使用した。これらは、APTIMA COMBO 2(登録商標)アッセイキット(Gen−Probe Incorporated カタログ番号1032)についてのパッケージ挿入物およびhttpアドレスwww.gen−probe.com/pdfs/pi/IN0037−04RevA.pdfにおいて記載された50% 漂白剤を使用するプロトコルと等価であり、臨床実験設定における核酸アッセイ手順に対して、有効な核酸の不活化および汚染除去のコントロールをもたらした。
以下は、各部位において利用した材料のリストである。

各部位において、以下の手順を利用した。APTIMA COMBO 2(登録商標)アッセイを行った各ラックのサンプル(10個までのTen−Tube Units(TTUs;カタログ番号TU0022;Gen−Probe Incorporated)について、通常のツーラン(two−run)コントロール(ポジティブコントロール、CTおよびポジティブコントロール、GC)、二重ポジティブコントロール(材料を参照のこと)、16個のネガティブコントロール(材料を参照のこと)、および80個までの患者標本を含んだ。このアッセイを、標準的なプロトコル(パッケージ挿入物)に従って行った。
各実験室に、サンプルのラックをセットアップするために以下の手順を利用するように指示した。
2)400μLのポジティブコントロール、CTを反応チューブ1に添加する
3)400μLのポジティブコントロール、GCを反応チューブ2に添加する
4)400μLの二重ポジティブコントロールを反応チューブ3〜4に添加する
5)400μLのネガティブコントロールを反応チューブ5〜20に添加する
6)400μLの患者標本を反応チューブ21〜100までに添加する。
各作業スペースを汚染除去しながら実験室における汚染の蔓延に対して注意するために、各実験室に、良好な物理的封じ込め技術を適用するように指示した。洗浄のために使用した手にはめた手袋は汚染されること、およびこの手で清潔な物体を触れることは避けられなければならないことを、各実験実に警告した。一方の手は洗浄のためのみに残しておかれるべきであること、そして他方の手(清潔)は試薬の適用のためのみに残しておかれるべきであることを推奨した。使用したタオルおよび手袋は、十分に封じ込められた容器に廃棄されて、汚染除去を受ける領域とその容器との間に漏れが生じないことを確実にするべきであることもまた推奨した。
各実験室に、以下に概要を述べる手順を使用して以下の試薬を調製するように指示した。
i)30mlのMilli−Q水(または等価な品質の水)を、試薬1B(乾燥試薬)の1バイアルに添加する
ii)きつくキャップし、30回反転させる。1時間静置する。さらに30回反転させる。全ての乾燥試薬が溶解するのを確実にする
iii)使用の間(以下の節2bを参照のこと)、試薬1B(液体)を2〜8℃の暗所で保存する(乾燥試薬は、室温で保存することができる)
iv)2週間保存した後、試薬1B(液体)を廃棄し、新鮮な溶液を調製する
b)試薬1を調製する(毎日)
i)150mlの試薬1Aを通気式の蓋を備える噴出ボトル(添付)に添加する
ii)1.5mlの試薬1Bを噴出ボトルに添加する
iii)上部を置き換え、10〜15秒間内容物を旋回させることによって完全に混合する
iv)以下に記載のように内容物を使用する。使用の間に噴出ボトルから試薬1が少しでも漏れた場合、上部をゆるめ、次いで使用を再開する前に、再度直ちに締める
v)最後に使用した日または12時間後に、どれを最優先にするとしても、噴出ボトルに残った試薬1を全て処分する。必要な場合、上記に記載のように新鮮な試薬を調製する
c)試薬2を調製する(2週間ごと)。
ii)容器を閉めて15〜20秒間内容物を旋回させることによって完全に混合する
iii)必要な場合、内容物を使用する
iv)2週間の最後に、全ての未使用の試薬2を廃棄し、上記に記載のように新鮮は溶液を調製する
(4.前アッセイ手順)
各実験室に、以下の前アッセイ手順を行うように指示した。
b)以下の通りに(列挙した順番で)増幅前領域における全ての表面を洗浄する:
Tecan。噴出ボトルを使用して、ペーパータオルを試薬1で、タオルが飽和するが浸漬しない程度まで湿らせる。湿らせたタオルでTecanデッキを完全に湿らせて洗浄し(現在の標準的プロトコルにあるような1分のインキュベーション時間を含まない)、全ての表面が乾くまで拭い続ける。一旦表面を洗浄して乾燥させ、試薬1の第2の適用によりこの手順を繰り返す。水でリンスしてはいけない。
各実験室に、以下の標本調製後手順を行うように指示した。
b)Tecan、浸漬するべき品目(以下を参照のこと)、標本処理領域で使用したベンチ表面、および使用した全てのピペットを、以下のように洗浄する:
i)Tecan.上記に記載のように試薬1で洗浄し、注意深く両方の手袋を廃棄する。
各実験室に、以下の標的捕捉後手順を使用するように指示した。
b)TCS、ベンチ表面およびピペット。上記に記載のように、試薬1で洗浄する。両方の手袋を注意深く廃棄する
c)真空トラップ廃液。必要な場合(以下を参照のこと)、廃棄物ボトル中の液体を汚染除去する。廃棄物ボトルを空のTCSユニットに取り付ける(すなわち、漂白剤を添加しない)。廃棄物ボトルが25%満たされるまでか(すなわち、利用可能な容量は、25%を超えない)、または1週間(どちらか早いほう)、廃棄物ボトルを使用する。廃棄物ボトルを系からはずし、400mlの希釈していない漂白剤を注意深く添加する(所望の場合、この手順は、実験室中に蒸気が放出されるのを避けるために換気フード中で行われ得る)。廃棄物ボトルに蓋をして、完全に混合するまで内容物を穏やかに旋回する。5分間インキュベートし、次いで廃棄物をシンクに注ぐ。空の廃棄物ボトルをTCSユニットに再度接続する。液体廃棄物および固体廃棄物を取り扱う場合および処分する場合の一般的な注意を使用する。地域の規則、州の規則、および連邦政府の規則に従って、液体廃棄物および固体廃棄物を処分する。廃棄物ボトルの内容物は、アッセイを汚染する可能性のある供給源として処理されるべきである。作業表面の汚染を避けるために注意されたい。両方の手袋を注意深く廃棄する。
各実験室に、各増幅反応(これは、増幅前領域において行われる最後の工程である)を開始した後に以下の手順を行うように指示した。反応を開始した後、各実験室に、上記に記載の手順に従って試薬1を使用して、水浴の周囲のベンチトップ、水浴の蓋のハンドル、およびピペットを洗浄するように指示した。各実験室に、これらの手順を行った後に両方の手袋を注意深く廃棄するように指示した。
各実験室に、増幅後領域手順に関する以下の指示を提供した。増幅後領域における最後の洗浄が完了し、新しい手袋を身につけた(adorn)後、各実験室に、増幅前領域に入った後直ちに、62℃の水浴にスイッチを入れるように指示した。上記に記載の特異的な手順に従って試薬2を使用して、増幅後領域(実験室ベンチ、ピペット、ハンドル、およびその他のもの)の全ての表面を前洗浄し、次いで両方の手袋を注意深く廃棄することもまた指示した。

各実験室に、検出後手順に関する以下の指示を提供した。指示は、以下のためのものであった:(a)照度計からTTUを除き、製品挿入物中の現在の手順を使用して反応物を不活化する;(b)上記に記載の特異的な手順に従って試薬2を使用して、全ての表面(ベンチ表面、ピペット、水浴の蓋のハンドル、LEADER(登録商標)HC+照度計の外面、および他のもの)を洗浄する;(c)2週間ごとか、または必要な場合、現在操作者のマニュアルに記載されているようにDI水でHC+の内面を洗浄し、HC+カセットを試薬2に30〜60分間浸漬する;および(d)両方の手袋を注意深く廃棄する。
各実験室に、以下の合格基準を使用するように指示した。
所定のプロトコルに従って使用した試薬1および試薬2は、APTIMA COMBO 2(登録商標)アッセイキットで提供される50% 漂白剤を使用したプロトコルと等価であり、臨床的設定においてAPTIMA COMBO 2(登録商標)アッセイについての有効な汚染除去コントロールをもたらした。
Claims (48)
- 核酸不活剤と合わせたときに、核酸が増幅反応における鋳型として作用することを防ぐための均質な溶液であって、該溶液は、以下:
i)ホスフェート、ホウ酸塩、または炭酸水素ナトリウムを含む、腐食阻害剤、
ii)洗剤または界面活性剤を含む、湿潤剤、および
iii)有機溶媒または乳化剤を含む、可溶化剤
を含有し、該腐食阻害剤が10mM〜750mMの濃度で該溶液中に存在し、該湿潤剤が0.005%〜1%(w/v)の濃度で該溶液中に存在し、そして該可溶化剤が0.001%〜20%(v/v)の濃度で該溶液中に存在し、これらの剤の各々は、22℃で実質的に溶液のままであり、
該溶液は、核酸不活剤を含まない、溶液。 - 前記溶液に対して核酸を実質的に不活化するのに十分な量の前記不活剤をさらに含む、請求項1に記載の溶液。
- 前記不活剤が、塩素または臭素を含む、請求項2に記載の溶液。
- 前記不活剤が、次亜塩素酸ナトリウムを含む、請求項3に記載の溶液。
- 前記溶液中の次亜塩素酸ナトリウムの濃度が、0.06%(w/v)〜3%(w/v)である、請求項4に記載の溶液。
- 前記溶液中の次亜塩素酸ナトリウムの濃度が、0.18%(w/v)〜1.8%(w/v)である、請求項4に記載の溶液。
- 前記溶液中の次亜塩素酸ナトリウムの濃度が、0.6%(w/v)〜1.5%(w/v)である、請求項4に記載の溶液。
- 前記溶液中の次亜塩素酸ナトリウムの濃度が、0.6%(w/v)〜1.2%(w/v)である、請求項4に記載の溶液。
- 前記不活剤が、ジクロロイソシアヌレートである、請求項3に記載の溶液。
- 前記溶液中のジクロロシアヌレートの濃度が、5mM〜400mMである、請求項9に記載の溶液。
- 前記溶液中のジクロロシアヌレートの濃度が、10mM〜200mMである、請求項9に記載の溶液。
- 前記溶液中のジクロロシアヌレートの濃度が、20mM〜100mMである、請求項9に記載の溶液。
- 前記溶液中のジクロロシアヌレートの濃度が、40mM〜80mMである、請求項9に記載の溶液。
- 前記腐食阻害剤が炭酸水素ナトリウムを含み、前記湿潤剤が洗剤を含み、そして前記可溶化剤が有機溶媒を含む、請求項1に記載の溶液。
- 前記防腐阻害剤が、炭酸水素ナトリウムである、請求項1に記載の溶液。
- 前記溶液が、前記不活剤を含み、該溶液中の前記腐食阻害剤の濃度が、10mM〜750mMである、請求項15に記載の溶液。
- 前記腐食阻害剤が炭酸水素ナトリウムを含み、前記湿潤剤が洗剤を含み、そして前記可溶化剤が乳化剤を含む、請求項1に記載の溶液。
- 前記腐食阻害剤および前記湿潤剤が、同じものである、請求項1〜13のいずれか1項に記載の溶液。
- 前記湿潤剤が、ドデシル硫酸ナトリウム、ラウリル硫酸リチウム、サポニン、CTAB、Alconox、およびポリオキシエチレン洗剤からなる群より選択される、請求項1または18に記載の溶液。
- 前記湿潤剤が、ドデシル硫酸ナトリウムまたはラウリル硫酸リチウムである、請求項19に記載の溶液。
- 前記溶液が、前記不活剤を含み、該溶液中の前記湿潤剤の濃度が、0.005%(w/v)〜1%(w/v)である、請求項20に記載の溶液。
- 前記腐食阻害剤が10mM〜750mMの濃度で前記溶液中に存在し、前記湿潤剤が0.005%〜1%(w/v)の濃度で該溶液中に存在し、そして前記可溶化剤が0.001%〜20%(v/v)の濃度で該溶液中に存在する、請求項2に記載の溶液。
- 前記可溶化剤が、酢酸ベンジル、PS20およびイソプロパノールからなる群より選択される有機溶媒である、請求項1〜22のいずれか1項に記載の溶液。
- 前記可溶化剤が、ポリオキシエチレンソルビタンモノパルミテート、レシチン、およびジステアリン酸エチレングリコールからなる群より選択される乳化剤である、請求項1〜22のいずれか1項に記載の溶液。
- 前記可溶化剤が、芳香剤である、請求項1〜24のいずれか1項に記載の溶液。
- 前記溶液が、前記不活剤を含み、該溶液中の前記可溶化剤の濃度が、0.001%(v/v)〜20%(v/v)である、請求項25に記載の溶液。
- 核酸不活剤と合わされたときに、核酸が増幅反応における鋳型として作用することを防ぐための溶液であって、該溶液は、以下:
i)ホスフェート、ホウ酸塩、または炭酸水素ナトリウムを含む、腐食阻害剤、

を含有し、該腐食阻害剤が10mM〜750mMの濃度で該溶液中に存在し、該湿潤剤が0.005%〜1%(w/v)の濃度で該溶液中に存在し、そして該可溶化剤が0.001%〜20%(v/v)の濃度で該溶液中に存在し、これらの剤の各々は、22℃で実質的に溶液のままであり、

- 前記腐食阻害剤および前記湿潤剤のうちの少なくとも1つが、乾燥形態で提供される、請求項29に記載のキット。
- 前記乾燥形態が、錠剤である、請求項30に記載のキット。
- 前記キットが、前記不活剤を備える、請求項29〜31のいずれか1項に記載のキット。
- 前記不活剤、前記腐食阻害剤、および前記湿潤剤のうちの少なくとも1つが、乾燥形態で提供される、請求項32に記載のキット。
- 前記乾燥形態が、錠剤である、請求項33に記載のキット。
- 核酸増幅反応を行う際に使用するための1種以上の増幅試薬をさらに備える、請求項29〜34のいずれか1項に記載のキット。
- 請求項1〜28にのいずれか1項に記載の溶液を作製する方法であって、該方法は、以下の順序の工程:
a)前記腐食阻害剤および前記湿潤剤の固体形態を別々に溶解する工程;
b)該腐食阻害剤および該湿潤剤の溶解形態を一緒に合わせて混合物を形成する工程;ならびに
c)可溶化剤と該混合物とを一緒に合わせて、該溶液を形成する工程
を包含し、該腐食阻害剤が10mM〜750mMの濃度で該溶液中に存在し、該湿潤剤が0.005%〜1%(w/v)の濃度で該溶液中に存在し、そして該可溶化剤が0.001%〜20%(v/v)の濃度で該溶液中に存在し、該剤の各々が、22℃で実質的に溶液のままである、方法。 - 工程c)の前に、前記可溶化剤の固体形態を溶解する工程をさらに包含する、請求項36に記載の方法。
- 前記不活剤と前記溶液とを一緒に合わせる工程をさらに包含する請求項36または37に記載の方法であって、ここで該不活剤は、該溶液に対して核酸を実質的に不活化するのに十分な量で存在し、そして該剤の各々は、22℃で実質的に溶液のままである、方法。
- 前記不活剤と前記溶液とを合わせる工程の前に、該不活剤の固体形態を溶解する工程をさらに包含する、請求項38に記載の方法。

- 前記表面に前記第1の溶液を適用する工程の前に、
