Description
本発明は、体細胞で突然変異した少なくとも1つのアミノ酸配列を生殖系列アミノ酸配列に変換する修飾ヒトモノクローナル抗体またはその抗原結合部分を提供する。好ましくは置き換えられる残基はその抗体の可変領域中に含有され、またより好ましくは置き換えられる残基は可変領域のフレームワーク領域中に含有される。
一実施形態ではこの抗体の軽鎖の可変領域の配列は、生殖系列A30遺伝子がコードするアミノ酸配列に戻った3つのフレームワーク突然変異を含む。好ましい実施形態ではこの軽鎖の可変領域は、SEQ ID NO: 5のアミノ酸配列のアミノ酸23番から130番を含む。さらに一層好ましい実施形態ではこのヒト抗体の軽鎖は、SEQ ID NO: 5のアミノ酸23番から236番を含む。

本発明をより良く理解することができるように下記の実施例を示す。これらの実施例は単に例示の目的に過ぎず、いかなるやり方でも本発明の範囲を限定するものと解釈されるべきではない。
本発明の抗体を次のように調製、選択、検定した。すなわち、
8から10週の老いたXENOMICETMを、腹膜内に免疫するか、あるいはヒトIGF−IRの細胞外ドメイン(10 μg/用量/マウス)、または3T3−IGF−IR もしくは300.19−IGF−IR細胞(これらはその形質膜上にヒトIGF−IRを発現させる2種類の移入細胞株である)(10×106細胞/用量/マウス)で後部フットパッド中に免疫した。この用量を3から8週間にわたって5から7回繰り返した。融合の4日前にこれらマウスは、PBSに溶かしたヒトIGF−IRの細胞外ドメインの最後の注射を受けた。免疫したマウスから採った脾臓およびリンパ節リンパ球を非分泌性骨髄腫P3−X63−Ag8.653細胞株と融合させ、以前に述べたHAT選択にかけた(GalfreおよびMilsteinの論文Methods Enzymol. 73: 3〜46, 1981)。IGF−IR に特効のあるモノクローナル抗体を産生するハイブリドーマ2.12.1を更なる検討用に選択し、2000年12月12日に下記の寄託番号でAmerican Type Culture Collection (ATCC), 10801 University Boulevard, Manassas, VA 20110−2209に寄託した。
2.12.1 PTA−2792
細胞を用いた検定でIGF−IがIGF−IRと結合するのを阻害する本発明の抗体の能力を定量するためにELISA実験を行った。IGF−IRを移入したNIH−3T3細胞(5×104 /mL)を、96ウェルU底プレートを用いてLグルタミン(0.29 mg/mL)と、熱で不活性化した10%FBSと、ジェネティシン、ペニシリン、およびストレプトマイシンのそれぞれ500 μgとを補足したDMEM高グルコース培地100 μL中で平板培養した。細胞を付着させるためにこのプレートを37℃、5%CO2中で夜通しインキュベートした。この培地をプレートからデカントし、1ウェル当たり100 μLの新鮮な培地で置き換えた。試験する場合、この抗体を検定培地(Lグルタミンと、熱で不活性化した10%FBSと、200 μg/mLのBSAと、ジェネティシン、ペニシリン、およびストレプトマイシンのそれぞれ500 μg/mLとを補足したDMEM高グルコース培地)中で所望の最終濃度まで希釈した。すべての試料は3組で行った。これらのプレートを37℃で10分間インキュベートした。[125I]−IGF−Iを検定培地中で1 μCi/mLの濃度まで希釈し、プレートの1ウェル当たり50 μL を加えた。バックグラウンド放射能用の対照として低温IGF−I を100 ng/mLの最終濃度まで加えた。これらプレートを37℃で10分間インキュベートし、この培地をペーパタオル上に静かに吸い取り、検定培地で2度洗浄することによってデカントした。50 μLの0.1N NaOH、0.1%SDSを加えることによって細胞を溶解し、そのプレートを室温で5分間振とうした。試料をシンチレーションプレートに移し、OptiPhase Supermix 150 μLを加え、Wallace Micro-Beta計数管を用いてシグナルを読み取った。
表1および図5は、本発明の抗体を用いて行ったこの実験の結果を示す。この実験は本発明の抗体が、IGF−IRを過剰発現させる細胞と[125I]−IGF−I の結合を特異的に阻害することを実証した。
本発明の抗体がIGF−I介在性のIGF−IR活性化を遮断できるかどうかを判定するためにELISA実験を行った。IGF−I介在性のIGF−IRの活性化は、受容体が関連するチロシンのリン酸化の低下により検出した。
ELISAプレートの調製:
遮断緩衝液(トリス緩衝塩類溶液[TBS]に溶かした3%ウシ血清アルブミン[BSA])を、ReactiBind Protein Gを塗布した96ウェルのプレートの各ウェルに加えることによってELISA捕捉プレートを調製した(Pierce)。これらプレートを室温で30分間振とうすることによってインキュベートした。ウサギの汎特異的SC−713抗IGF−IR抗体(Santa Cruz)を遮断緩衝液中で5 μg/mLの濃度まで希釈した。希釈した抗体100 μLを各ウェルに加えた。これらプレートを室温で60〜90分間振とうしながらインキュベートした。これらプレートを洗浄緩衝液(TBS+0.1%トゥイーン20)で5回洗浄し、残りの緩衝液をペーパータオル上に吸い取った。これらのプレートは、ライゼートの添加までは乾燥しないようにした。
IGF−IRを移入したNIH−3T3細胞(5×104 /mL)を、96ウェルU底プレートを用いて成長培地(Lグルタミン(0.29 mg/mL)と、熱で不活性化した10%FBSと、ジェネティシン、ペニシリン、およびストレプトマイシンのそれぞれ500 μg/mLとを補足したDMEM高グルコース培地)100 μL中に置いた。細胞を付着させるためにこのプレートを37℃、5%CO2中で夜通しインキュベートした。この培地をプレートからデカントし、1ウェル当たり100 μLの新鮮な培地で置き換えた。試験する場合、これらの潜在的抗IGF−IR抗体を成長培地中で所望の最終濃度の5倍まで希釈し、それを1ウェル当たり25 μL加えた。すべての試料は3組で行った。これらプレートを37℃で1時間インキュベートした。この細胞を600 ng/mLのIGF−1(成長培地中で調製した)の25 μL/ウェルで刺激し、室温で10分間インキュベートした。プレートを逆さにし、ペーパタオル上に静かに吸い取ることによってこの培地をデカントした。溶解緩衝液(50 mM HEPES(pH 7.4)、10 mM EDTA、150 mM NaCl、1.5 mM MgCl2、1.6 mM NaVO4、1%トリトンX−100、1%グリセロール、および使用直前に50 mL当たり1個のEDTAを含まないプロテアーゼ阻害薬の錠剤[Roche Molecular Sciences]を補足した)50 μLを加えることによって付着細胞を溶解した。この細胞を室温で5分間振とうした。各ウェルに希釈緩衝液(50 mM HEPES(pH 7.4)、1.6 mM NaVO4)200 μLを加え、ピペットで移すことによって混合した。ライゼート100 μLを各ウェルから、上記で調製したELISA捕捉プレートの各ウェルへ移し、室温で2時間静かに振とうしながらインキュベートした。
プレートを逆さにし、そのプレートを洗浄培養液で5回洗浄し、ペーパタオル上に過剰の液体を吸い取ることによって細胞ライゼートを取り出した。1ウェル当たり100 μLのpTYR特異抗体(HRP−PY54)を加え、遮断緩衝液中で0.2 μg/mLの濃度まで希釈した。この細胞を、プレートを振とうすることによって室温で 30分間インキュベートした。次いでこのプレートを洗浄緩衝液で5回洗浄し、ペーパタオル上に吸い取った。
HRP−PY54抗体の結合は、1ウェル当たり100 μLのTMBペルオキシダーゼ基質溶液(Kirkegaard & Perry)を加え、発色としての振とうを行いながらインキュベートする(約2〜10分)ことによって検出した。発色反応を1ウェル当たり100 μLのTMB停止液(Kirkegaard & Perry)を加えることによって停止した。プレートをこの溶液と混ぜるために室温で10分間振とうし、OD450nmを測定することにより定量した。
本発明の抗体の種交差反応を求めるためにイムノプレシピテーション、IGF−Iが誘発する受容体リン酸化の抗体介在性遮断、およびFACS分析を含めた幾つかの実験を行った。
イムノプレシピテーション実験を行うために細胞を、Lグルタミン(0.29 mg/mL)と、熱で不活性化した10%FBSと、ジェネティシン、ペニシリン、およびストレプトマイシンのそれぞれ500 μg/mLとを補足したDMEM高グルコース培地中でT25フラスコを用いて50%集密するまで平板培養した。本発明の抗体100 μLをハンクス緩衝塩類溶液(HBSS、Gibco BRL)中に1 μg/mL の濃度で加えた。このプレートをインキュベーター中で37℃において30分間インキュベートし、次いで細胞を100 ng/mLのIGF−I により室温で10分間刺激した。この細胞をRIPA緩衝液(前掲のHarlowおよびLaneの著書)中で溶解し、このIGF−IRを汎特異的SC−713抗IGF−IR抗体(Santa Cruz)2 μgプラスプロテインAアガロースビーズで4℃において1時間、免疫沈降させた。このビーズをペレット化し、PBS/T(PBS+0.1%トゥイーン20)で3回洗浄し、次いで5%βMEを含有するLaemmli緩衝液40 μL中で煮沸した。
FACS分析を行って他の動物、具体的には上記旧世界のサルから採ったIGF−IRに対する本発明の抗体の親和性を求めた。ヒトおよびサル(カニクイザル)細胞の分割量(5×105)を氷上で1時間インキュベートして本発明のビオチニル化した抗IGF−IR抗体の濃度を増した。この試料をスプレプトアビジン複合RPE(フィコエリトリン)と一緒に氷上で30分間インキュベートした。結合は、フローサイトメトリーにより測定し、CellQuestソフトウェアを用いて蛍光強度(FI2−H)対細胞数(計数)の柱状グラフにより解析した。平均蛍光強度対抗体濃度のグラフから結合(Kd)を各抗体について計算した。大部分の実験において結合は、培養ヒトMCF−7細胞およびカニクイザル組織培養細胞中で測定した。抗体の消耗は、或る範囲の細胞濃度にわたって結合を測定することにより制御した。
上述のFACS分析を行って本発明の抗体がヒトおよびカニクイザル細胞と結合する能力を試験した。試験されたすべての細胞株に対して0.1 μg/mLの最大の2分の1の結合(Kd)が観察された。
本発明の抗体が細胞上でIGF−1Rのダウンレギュレーションを引き起こすことができるかどうかを調べるためにMCF7細胞を、T75フラスコ中でLグルタミン(0.29 mg/mL)、熱で不活性化した10%FBS、ペニシリン、およびスプレプトマイシンを補足したDMEM/F12培地中で50%集密するまで平板培養した。その細胞に本発明の抗体を1 μg/mLの最終濃度で加えた。これらのプレートをインキュベーター中で37℃において指定された時間インキュベートし、次いで50 mM HEPES(pH 7.4)、10 mM EDTA、150 mM NaCl、1.5 mM MgCl2、1.6 mM NaVO4、1%トリトンX−100、1%グリセロール中で溶解した。汎特異的SC−713抗IGF−IR抗体(Santa Cruz)を用いたウェスタンブロット分析により細胞抽出物内の総全IGF−IRのレベルを求めた。図6Bを参照されたい。本発明の抗体によるMCF7細胞の処理は、1〜2時間でIGF−1Rの60〜70%のダウンレギュレーションを引き起こした。
さきの実施例中で述べたIGF−IRに及ぼす本発明の抗体の効果がin vivoで起こるかどうかを判定するために実験を行った。公表されている方法(V. A. Pollack等の論文「CP−358,774によるヒト癌腫中の表皮成長因子受容体が関連するチロシンリン酸化の阻害: 無胸腺マウスにおけるin situでの受容体阻害の動態および抗腫瘍効果(Inhibition of epidermal growth factor receptor-associated tyrosine phosphorylation in human carcinomas with CP-358,774: Dynamics of receptor inhibition in situ and antitumor effects in athymic mice)」J. Pharmacol. Exp. Ther. 291: 739-748 (1999))に従って無胸腺マウス中に腫瘍を誘発させた。手短に言えばIGF−IR移入NIH−3T3細胞(5×106)を3〜4週の老いた無胸腺(nu/nu)マウスにMatrigel製剤0.2 mLを用いて皮下注射した。次いで確立(すなわち約400 mm3)腫瘍が生じた後、このマウスに本発明の抗体を腹膜内注射した。
本発明の抗体が腫瘍成長を阻害するように作用するかどうかを判定するために抗体を試験した。腫瘍は前述(実施例VI)と同様に誘発され、それが確立されると触感できる腫瘍が生じた(すなわち6〜9日以内に250 mm3)。このマウスを腹膜内注射による抗体0.20 mLの単一回投与量で処理した。腫瘍の大きさをVernierカリパスにより3日ごとに2ヶ所の直径で測定し、体積をGeran等の論文「動物の腫瘍および他の生体器官系に対して化学物質および天然産物をスクリーニングする実験計画(Protocols for screening chemical agents and natural products against animal tumors and other biological systems)」Cancer Chemother. Rep. 3: 1-104によって確立された方法を使用して式、(長さ×〔幅〕2)/2を用いて計算した。
本発明の抗体による分析の後で、この抗体のみによる単一処理が、IGF−IR移入NIH−3T3細胞の誘発する腫瘍の成長を阻害することが観察された(図8A)。さらに静脈内に与えた単一回投与量のアドリアマイシン7.5 mg/kgとの併用の検討で、単一回投与量の抗体の投与が、周知の腫瘍成長の阻害薬であるアドリアマイシンの効力を高めることが観察された。本発明の抗体とアドリアマイシンの併用は、抗体またはアドリアマイシンのみによる処理に対して7日の成長の遅延を示した(図8B)。
実施例VIで述べたと同様にヌードマウス中で腫瘍を誘発させた。次いでこのマウスを実施例VIで述べたと同様にこの抗体125 μgで腹膜内注射により処理した。腫瘍を摘出し、ELISAによりIGF−IR レベルを測定した。図9は、或る期間にわたっての血清抗体レベルおよびIGF−IR 受容体レベルを示す。この実験は、IGF−IRがこの抗体によってダウンレギュレートされること、およびIGF−IR 阻害の程度が用量による抗体の血清濃度に比例することを実証する。
Colo 205細胞(ATCC CCL 222)を使用したことを除いて実施例VIで述べたと同様にヌードマウス中で腫瘍を誘発させた。確立した約250 mm3の皮下腫瘍を有するマウスを、様々な量の抗体(i.p.)または100 mg/kgの5−フルオロデオキシウリジン(5−FU、i.v.)により、単一薬品としてまたは併用のいずれかで実施例VIIで述べたと同様に処理した。図10Aおよび図10Bは、様々な処理について或る期間にわたっての腫瘍の大きさを示す。この実験は、単一薬品として与えられた場合に、その一度与えられた抗IGF−IR抗体による処理がヒトの直腸結腸癌細胞の成長を阻害し、また周知の腫瘍阻害薬である5−FUの有効性を高めることを実証する。
抗IGF−IR抗体の薬物動態を評価するためにカニクイザルに酢酸緩衝液に溶かした抗体5 mg/kgを静脈内注射した。サルから様々な時点で血清を回収し、10週までの期間のあいだサル中の抗IGF−IR抗体濃度を求めた。機能血清抗体レベルを定量するためにヒトIGF−IRの細胞外ドメイン(IGF−I−sR、R&D Systems、カタログ番号391GR)を96ウェルプレートと結合させた。各試料が基準曲線の線形範囲内に入るようサルの血清(1 : 100から1: 15,000の間で希釈した)を検定プレートに加え、どの抗IGF−IR抗体もIGF−I−sRと結合する条件下でインキュベートした。プレートを洗浄した後、標識した抗ヒトIgG抗体をこのプレートに加え、その抗ヒトIgG抗体が抗IGF−IR抗体と結合する条件下でインキュベートした。次いでプレートを洗浄、発色させ、対照基準曲線および線形回帰の当てはめを用いて抗IGF−IR抗体の量を定量化した。図11は、或る時間にわたっての血清中の抗体の濃度を示す。この実験は、この抗IGF−IR抗体の半減時間(t1/2)が6.1日であり、また0.054L/kgの体積配分(Vdss)を有することを実証する。
Claims (10)
- ヒトインスリン様成長因子I受容体(IGF−IR)に特異的に結合するヒトモノクローナル抗体又はその抗原結合部分であって、前記抗体の可変領域のフレームワーク領域中に、対応する生殖系アミノ酸残基により置換されている体細胞突然変異アミノ酸残基を含み、軽鎖の前記可変領域が、アミノ酸配列番号5のアミノ酸23番〜130番を含み、かつ、重鎖の可変領域が、配列番号3のアミノ酸20番〜144番を含む、前記ヒト抗体又は前記抗原結合部分。
- 前記軽鎖が配列番号5のアミノ酸23番〜236番を含む、請求項1に記載のヒト抗体。
- 前記重鎖が配列番号3のアミノ酸20番〜470番を含む、請求項1又は2に記載のヒト抗体。
- 前記抗体の前記重鎖が配列番号3のアミノ酸20番〜469番を含み、かつ、前記軽鎖が配列番号5のアミノ酸23番〜236番を含む、請求項1又は2に記載のヒト抗体。
- 請求項1〜4のいずれか一項に記載の抗体又はその抗原結合部分と、薬学的に許容される担体を含む癌治療用医薬組成物。
- 抗新生物薬、化学療法薬又は抗腫瘍薬をさらに含む、請求項5に記載の医薬組成物。
- 請求項1〜4のいずれか一項に記載の抗体の重鎖又はその抗原結合部分あるいは軽鎖又はその抗原結合部分をコードする核酸配列を含む単離ポリヌクレオチド。
- 請求項7に記載の単離核酸分子を含むベクター。
- 請求項8に記載のベクターを含む宿主細胞。
- 請求項9に記載の宿主細胞を培養するステップと、前記抗体を回収するステップとを含むモノクローナル抗体又はその抗原結合部分の製造方法。
