Description
・Streptomyces lividans、例えば、グルコースイソメラーゼを発現し得る発現カセット、即ち、例えばActinoplanes missouriensis由来のグルコースイソメラーゼをコードする遺伝子を含有するS.lividans TK21;
・Penicillium chrysogenum、例えば、エクスパンダーゼ、および任意成分としてのヒドロキシラーゼおよび/またはアセチルトランスフェラーゼを発現し得る1個以上の発現カセット、即ち、例えばAcremonium chrysogenum由来のエクスパンダーゼ、ヒドロキシラーゼおよびアセチルトランスフェラーゼをコードする遺伝子を含有するP.chrysogenum CBS 455.95、または側鎖先駆体としてアジピン酸(EP 532341参照)または3−カルボキシメチルチオ−プロピオン酸(WO95/04148参照)を用いて、7−ADCAまたは7−ACA等のセファロスポリン化合物を産生し得るStreptomyces clavuligerus;
・Aspergillusniger、例えば、ヒトラクトフェリン(WO93/22348参照)またはウシキモシンを発現し得る発現カセットを含有するAspergillus nigerCBS 513.88;
・ウシキモシンまたはフォスフォィパーゼA2、インシュリンまたは組換えヒトアルブミンを発現し得る発現カセットを含有するKluyveromyces lactis。
・A.niger、例えば、フィターゼ(EP 420358参照)またはエンドキシラナーゼI(EP 463706参照)を重複発現し得る発現カセットを含有するA.niger CBS513.88。
Streptomyces lividansを用いてのグルコースイソメラーゼの工業的生産
グルコースイソメラーゼを産生するストレプトマイセス株の構築
実施例1で述べたようにして構築した産生株の業務用セルバンクを、チオストレプトン耐性コロニーをピッキングし、これを100mlの振盪フラスコ内のチオストレプトン(50mg/l)を含有する20mlのトリプトン ソイトーン ブロス(Tryptone Soytone Broth)中で30℃40〜48時間増殖させ、280rpmで振盪させることによって調製した。

Claims (11)
- 化学的に明確な構成成分からなる化学的に明確な培地である発酵培地において10m 3 以上の容量規模で繊維状微生物株の発酵を行うこと、および発酵培養液からアラキドン酸を含む脂質を回収することの各工程を含み、繊維状微生物株がモーティアレラ アルピナ(Mortierella alpina)である、アラキドン酸を含む脂質の生産方法。
- 化学的に明確な培地が本質的に少量の複合炭素および/または窒素源を含有し、本質的に少量の複合炭素および/または窒素源とは、化学的に明確な培地中に炭素および/または窒素(ケルダールN)総量が多くとも10%の量存在することを意味する、請求項1記載の方法。
- 複合炭素および/または窒素源が主発酵用の植込みの発酵過程において使用される、請求項2記載の方法。
- 主発酵の初期段階で複合炭素および/または窒素源が添加される、請求項2または3記載の方法。
- 化学的に明確な培地の化学的に明確な構成成分が、炭水化物、グルコース、ラクトース、フルクトース、スクロース、マルトデキストリン類、澱粉およびイヌリン;グリセリン;植物油;炭化水素類;アルコール類、メタノールおよびエタノール;有機酸、酢酸塩およびより高級なアルカン酸からなる群から選ばれる炭素源と、尿素;アンモニア;硝酸塩;アンモニウム塩、硫酸アンモニウム、リン酸アンモニウムおよび硝酸アンモニウム;並びにアミノ酸類、グルタミン酸塩およびリシンからなる群から選ばれる窒素源とを含む請求項1ないし4のいずれか1項記載の方法。
- 炭素源がグルコースであり、窒素源がアンモニアおよび/またはアンモニウム塩である請求項5記載の方法。
- 発酵がバッチ、繰返しのバッチ、給送バッチ、繰返しの給送バッチまたは連続発酵過程で生ずる請求項1ないし6のいずれか1項記載の方法。
- 発酵が給送バッチ過程で生ずる請求項7記載の方法。
- 炭素および/または窒素源を給送バッチ過程に給送する請求項8記載の方法。
- 炭素源がグルコースであり、窒素源がアンモニアおよび/またはアンモニウム塩である請求項9記載の方法。
- アラキドン酸を含む脂質がトリグリセリドである請求項1ないし10のいずれか1項記載の方法。
