Description
本発明は、感染性PIV 中に、定義された予定の構造及び表現型変化を導入するための新規手段及び方法を提供する。本発明の1つの態様においては、作用可能に連結された転写プロモーター、PIV ゲノム又はアンチゲノムをコードするポリヌクレオチド配列、及び転写ターミネーターを含んで成る単離されたポリヌクレオチド分子が提供される。
図1は、HPIV3ゲノムRNA の完全なコピーをコードするプラスミドp218 (131) の構成を示す。p218 (A A'CC'D E)のダイアグラム(上部左)は、HPIVゲノムのヌクレオチド1に隣接するδリボザイム及びT7ターミネーターの位置を示す。p(E FF'GHIJKL)の上部右のダイアグラムは、7個の配列決定マーカー(星印)(下記図2を参照のこと)、及びHPIV3のヌクレオチド15462 に隣接するT7プロモーターの位置を示す。十分な長さのゲノムcDNAを構成するために使用される15のオーバーラップするクローンが、2種のプラスミドの外部上に文字により示される。ユニーク制限部位 NgoMI及び XhoIが、底部に示されるHPIV3の完全なネガティブ−センスのゲノムRNA をコードするp218 (131) をクローン化するために使用された。JSと組換えJSとを区別するp218 (131) における個々の配列決定マーカーの位置は星印により示される。
1)7903での非コードnt置換は、JS wt から HgaI部位を除く。7913及び7915でのJS cDNA におけるヌクレオチド置換は、 ScaI部位を創造し、そしてアミノ酸370 をプロリンからトレオニンに変え、mAb170/7及び423 /6により認識される抗体エピトープを除去する。また、HN遺伝子は、これまで認識されたことがないnt7593での追加の点突然変異を含むことが見出された。これは、HNタンパク質におけるアミノ酸位置263 でのトレオニンのイソロイシンへの変化をもたらす。
2)プラスミド構成又はアセンブリーの間に生じた、JS cDNA のL遺伝子における3種の偶発的な非コード突然変異。
プラスミド:T7プロモーターは、転写の方向を示す黒の矢印として示される。位置は、δリボザイム、T7転写ターミネーター、挿入されたワクシニアウィルス転写ターミネーター(T7CT及びT5AT)、制限酵素部位、及びコードされたミニゲノムの種々のドメインのためのコード配列に関して示される。
図11は、ネガティブ−センスの組換え PIV3の配列確認を示す。7903,7913及び7915での突然変異を包含する1379bpのフラグメント(ヌクレオチド7334−8713)を、感染された細胞からのRNA のRT−PCR により生成し、そしてサイクル−配列決定により分析した。JSから組換えPIV を分化する突然変異は、矢印により示される。nt7897〜7922の完全な配列が、太字で示される3種のヌクレオチド差異と共に、個々のゲルの次の端に示される。
図14は、 PIV3 HN 及びF ORFが PIV1のそれらにより置換されているキメラ PIV3- PIV1アンチゲノムをコードするcDNAの構成を示す。最初に(底部左から開始して)、 PIV3 F及びHN遺伝子を、十分な長さの PIV3 cDNA クローンp3/7(131)2G(パネルA)からサブクローン化し(BspEII−XhoI及び EcoRI−SpeIフラグメントとして)、それぞれpLit. 例I
陰性センスPIV ゲノムRNA をコードするプラスミドp218 (131) の構成
p218 (131)(図1:配列番号1)(American Type Culture Collection (ATCC), 12301 Parklawn Drive, Rockville, Maryland 20852, USA にブタベスト条約に基づいて寄託され、そして寄託番号第 97991号が得られた)と命名された十分なcDNAクローンを、HPIV3 JS 株の完全な15461nt のゲノム配列をコードするよう構成した。
HPIV3のための転写及びRNA 複製系
この例は、ヒトパラインフルエンザウィルスタイプ3(HPIV3)のための再構成された転写及びRNA 複製系を生成するための組成物及び方法を記載する。この典型的なシステムを、ワクシニアウィルス組換え体により供給されるT7 RNAポリメラーゼにより駆動される、トランスフェクトされたプラスミドから細胞内発現される成分を用いて開発した。この系は、ウィルス遺伝子が欠失され、そして細菌クロラムフェニコールアセチルトランスフェラーゼ(CAT)をコードするポリヌクレオチドにより置換されているHPIV3ゲノムRNAのネガティブ−センス類似体に基づかれている。N,P及びLタンパク質を、効果的な転写及びRNA 複製を方向づけるために、同時トランスフェクトされたプラスミドから発現せしめる。この例に従っての転写は、たとえばoligo (dT)クロマトグラフィーにより容易に単離され得る、ポリアデニル化されたサブゲノムmRNAを生成する。この例に従ってのRNA 複製は、ミクロコッカスヌクレアーゼによる消化に対する耐性に基づいてカプシド封入されることが示されている、ミニ−アンチゲノム及び子孫のミニゲノムを生成する。
バクテリオファージT7 RNAポリメラーゼを発現するワクシニアウィルス組換え体、すなわち vTF7-3を、Fuerstなど. (Proc. Natl. Acad. Sci. USA. 83 : 8122-8126, 1986 ; これは引用により本明細書に組込まれる)により記載のようにして調製した。 HEp−2単層培養物を、2%ウシ胎児血清(FBS)、50μg/mlのゲンタマイシンスルフェート及び2mMのグルタミンにより補充されたOptiMEM 1 (Life Technologies, Gaithersburg, MD)により、5%CO2 の下で37℃で維持した。
HPIV3のJS株(GenBank #Z11575 ; Stokesなど., 1992)のN,P及びL遺伝子のORF に対応するcDNAは、T7 RNAポリメラーゼ、及び外来性ORF の前の内部リボソーム侵入部位による翻訳により仲介される(Elroy-Steinなど., Proc. Natl. Acad. Sci. USA, 86 : 6126-6130 (1989); 引用により本明細書に組込まれる)。個々の遺伝子をまず、ポリメラーゼ鎖反応(PCR)により修飾し、下流の末端上の翻訳開始部位及びSa'lI部位で NcoI又は NcoI−適合性部位を配置した。
HEp−2細胞を、6ウェルプレートにおいて、90%集密性に増殖した。6ウェルプレートの個々のウェル(1.5×106 個の細胞)を、0.4μgのpTM(P), 0.4μgのpTM(N) , 0.05μgのpTM(L)、及び 0.4μgのミニゲノムプラスミドによりトランスフェクトした。前記プラスミドを、 0.1mlのOpti MEM(Life Technologies)に添加し、そして12μlのLipofect ACE (Life Technologies)を含むOpti MEM 0.1mlと共に混合した。室温で約15分間のインキュベーションの後、2%ウシ血清及び 1.5×107pfuの vTF7−3を含むOpti MEM1(0.8 ml)を、個々のウェルに添加した。プレートを37℃で12時間インキュベートし、その後、培地を、2%ウシ胎児血清を含む新鮮なOpti MEMにより変換した。次に、細胞を37℃で合計48時間インキュベートし、そしてRNA 分析及びCAT アッセイのために収穫した。個々のミニゲノムを、三重反復して表わし(3個のウェル)、これを培地に削り取り、そしてプールした。
上記収穫された細胞の個々のプールされたサンプルの3.33%(1.5×105 個の細胞)を表わすアリコートを、CAT アッセイのために取り出した。そのアリコートを1,000rpmで5分間、遠心分離し、そして上清液を捨てた。細胞懸濁液を、40mMのトリス(pH7.5)、1mMのEDTA、150mM のNaClの溶液1mlにより洗浄し、そして0.25Mのトリス(pH7.5)溶液50mlに再懸濁した。溶解物を、3回の凍結及び融解により調製し、そして8,000rpmでの5分間の遠心分離により透明にした。溶解物1μlを、従来のアッセイを用いて、D−トレオー〔ジクロロアセチル1−14C〕クロラムフェニコール(Amersham)をアセチル化する能力についてアッセイした(Gormonなど., Mol. Cell. Biol. 2 : 1044-1051 (1982); これは引用により本明細書に組込まれる。アセチル化を、薄層クロマトグラフィーにより可視化し、そしてホスホイメージャー分析(Molecular Dynamics, Sunnyvale, CA)により定量化した。
個々のプールされたサンプルの残る細胞収穫物を、カプシド化されたRNA 、合計RNA 及びmRNAの単離のために3つの等しい部分に分けた。それらの3つのアリコートを、1,000rpmで5分間、遠心分離し、そして上清液を捨てた。細胞懸濁液の2つのアリコートを、1%Triton X-100, 0.5%DOC を含むRSB(10mMのNaCl, 10mMのトリス、pH7.5, 1.5mMのMgCl2)50μlに再懸濁した。次に10mMのトリス(pH7.5), 1mMのCaCl2 及び20μg(1mg/mlのストック)のミクロコッカスヌクレアーゼの溶液50μlを1つのアリコートに添加し、そして他はミクロコッカスヌクレアーゼを有さない同じ混合物を受けた(Baker & Moyer, J. Virol. 62 : 834-838 (1988) ; 311pにより本明細書に組込まれる)。ミクロコッカスヌクレアーゼの目的は、カプシド化されなかったRNA を破壊することであり、そして使用される条件は、予備実験において最適化された。
修飾された PIV3ミニゲノムの構成及び発現
この例においては、cDNAのパネルが、単一のヌクレオチドインクレメントにより長さが異なる PIV3ミニゲノムをコードするよう構成された。この再構成されたシステムにおける転写及びRNA 複製は、その長さがちょうど6の倍数であるミニゲノムに関して最とも効果的であった。これに関して、パラミキソウィルス及びモルビリビラス(Morbillivirus)属のメンバーは、典型的には、“6の規則”により守られ、すなわちゲノム(又はミニゲノム)は、それらのヌクレオチドの長さが6の倍数である場合のみ、効果的に複製する(NPタンパク質のカプシド化に対してのヌクレオチド残基の正確な間隔開けのための必要条件であると思われる)。しかしながら、この発見は、長さが6のちょうど倍数よりも1つのヌクレオチド大きいか又は少さいミニゲノムが驚くべきことには、特にRNA 複製においてのみ活性的であることを示す。
PIV ミニゲノムによるポジティブ−センスRNA の合成
PIV ミニゲノムの転写及び複製を、両方法のRNA 生成物の検出により確かめた。前記例に記載されるように、細胞懸濁液の3つの等しいアリコートを、RNA 分析のために採取した。1つのアリコートを、上記のように、oligo (dT)分析のために使用した。他の2つのアリコートを、界面活性剤により溶解し、そしてミクロコッカスヌクレアーゼと共にインキュベートするか又は擬似−処理した。次に、RNA を、単離し、ホルムアルデヒドアガロースゲル上での電気泳動により分離し、ニトロセルロースに移し、そしてネガティブ−センスCAT リボプローブによるハイブリダイゼーションにより分析した。ミクロコッカス−処理された及び擬似−処理された溶解物からのRNA が、それぞれ、図5Aの上部及び下部パネルに示される。
PIV ミニゲノムによるネガティブセンスのRNA の合成
前記例に記載される種々の PIV3−CAT ミニゲノムを、mRNA及びポジティブ−センスのカプシド化されたミニ−アンチゲノムの合成に向け、後者は、RNA 複製における第1段階を表わす。RNA 複製における第2段階は、ミニ−アンチゲノム生成物からのカプシド化された子孫ミニゲノムの合成を包含する。この後者の工程を評価するために、前記例に記載される擬似(mock)−処理された及びヌクレアーゼ−処理された溶解物を、ポジティブ−センスCAT リボプローブによるノザンブロットハイブリダイゼーションにより分析し(図7A)、そしてホスホルイメージャーにより定量化した(図7B)。
cDNAからの感染性組換えRNA の構成
次の例は、4種のプラスミド担持のcDNAの細胞内同時発現による感染性組換えPIV (rPIV)の生成を記載する。それらのcDNAは、完全なHPIV3ゲノム及びHPIV3ヌクレオカプシドタンパク質N、リンタンパク質P、及びポリメラーゼタンパク質Lを別々にコードする。
修飾されたワクシニア株Ankara (MVA)、すなわちバクテリオファージT7 RNAポリメラーゼを発現するワクシニアウィルス組換え体を、Wyatt など., Virol. 210 : 202-205, 1995(引用により本明細書に組込まれる)に従って調製した。 HEp−2単層培養物を、2%FBS,50μg/mlのゲンタマイシンスルフェート及び2mMのグルタミンにより補充されたOpti MEM1 (Life Technologies)により、5%CO2 下で37℃で維持した。HPIV3のJS wt 株及びその弱毒化されたts誘導体、すなわちJS cp45 を、Hallなど., Virus Res. 22 : 173-184, (1992)(引用により本明細書に組込まれる)により記載のようにして、 LLC−MK2細胞において増殖した。
HPIV3の完全なゲノム配列をコードする、p218 (131)(図1、配列番号1)と称する十分なcDNAクローンを、上記のようにして構成した。p3/7 (131)(配列番号14)(ATCCにブタペスト条約の下で寄託され、そして寄託番号第 97990号を付与されている)と称する第2のcDNAクローンを、HPIV3の完全なアンチゲノム配列をコードするよう構成した。p3/7 (131)は、p218 (131)と、HPIV3 cDNA 挿入体に対してのT7プロモーター及びリボザイム/T7ターミネーターの位置が相互変更されていることにおいて異なる。従って、p3/7 (131)において合成される第1のヌクレオチドは、アンチゲノムの5'末端、すなわちゲノム位置1のポジティブ−センス相補体であり、そしてリボザイム分解により定義される3’アンチゲノム末端はゲノム位置15462 の相補体である。
HEp−2細胞を、6ウェルプレートにおいて90%集密性 (confluence) まで増殖せしめた。6−ウェルプレートの個々のウェル(1.5×106 個の細胞)を、3種の前記支持プラスミド、すなわち 0.4μgのpTM(P), 0.4μgのpTM(N), 0.05μgのpTM(L)、及び5μgの十分な長さのゲノム又はアンチゲノムHPIV3 cDNA によりトランスフェクトした。プラスミドを、12μlのLipofect ACE (Life Technologies)を含むOpti MEM1 (Life Technology)0.2mlに添加した。室温で約15分間のインキュベーションの後、2%ウシ胎児血清及び 1.5×107pfuの MVA−T7を含む 0.8mlのOpti MEMを個々のウェルに添加した。その培養物を32℃で12時間インキュベートし、その後、培地を、2%ウシ胎児血清を含む新鮮なOpti MEM1 により交換した。
組換えPIV のHN及びL遺伝子におけるヌクレオチド配列マーカーの存在を、回収されたビリオンから単離されたRNA のRT−PCR により決定した。1 mlのpPIV(1×105pfu/ml、継代レベル2)を、氷上での1時間のインキュベーションにより、25%ポリエチレングリコール 200μlにより沈殿せしめ、そして12,000gで15分間、遠心分離した。RNA を、TRIzol試薬(Life Technologies) により、製造業者の推薦方法に従って精製した。RT−PCR を、Superscript キット(Life Technologies) により、製造業者の推薦方法に従って実施した。対照反応は同一であったが、但しPCR 生成物がウィルスRNAからのみ由来し、そして可能性ある汚染性cDNAプラスミドからではないことを確認するために逆転写酵素を反応から削除した。4種のプライマー対を用いて、nt7334-8715, 9364-10854, 10939-15392及び13623-15392 からPCR 生成物を生成した。次に、それらの得られるPCR 生成物を、サイクルジデオキシヌクレオチド配列分析(NewEngland Biolabs, Beverly, MA)を用いて配列決定した。
ネガティブ−センスのゲノムRNA をコードするcDNAからの組換えウィルスの回収
プラスミドp218 (131) 、並びに3種の支持プラスミドpTM(N),pTM(P)及びpTM(L)を、T7 RNAポリメラーゼを発現するMVAと共に、 HEp−2細胞中にトランスフェクトした。pTM(N), pTM(P),pTM(L)及びMVA から成る対照グループを、 HEp−2細胞中に、p218 (131) を伴わないで同時トランスフェクトした。4日目、トランスフェクタントを収穫し、新鮮な HEp−2細胞単層上に5日間、継代し、そして再び LLC−MK2培養物において5日間、継代した(継代2)。
ポジティブ−センスのアンチゲノムRNA をコードするcDNAからの組換えウィルスの回収
上記でより詳細に記載されたように、p3/7 (131)及びp3/7 (131)2Gを、組換えPIV を生ぜしめるポジティブ−センスのアンチゲノムをコードするよう構成した。プラスミドp3/7 (131)2Gは、p3/7 (131)と同一であるが、但しT7プロモーターとアンチゲノムの第1ヌクレオチドとの間の2つのG残基が付加されている。T7プロモーターと第1HPIV3ヌクレオチドp3/7 (131)2Gとの間への2つのG残基の付加は、2つのG残基の存在(0,1又は3個の付加される残基に反して)はミニレプリコン複製の実質的に高められたレベルを生成することを示す前記例に基づかれている。
許容できる温度及び制限的温度でのrPIVのプラーク形成の効率
p218 (131) 及びp3/7 (131)2Gの両者に由来するrPIV及び対照ウィルスのインビトロ複製の温度感受性レベルを、次の変性を伴って、Hallなど., Virus Res. 22 : 173-184 (1992)(引用により本明細書中に組込まれる)により前に記載されたようにして LLC−MK2単層培養物において、32℃、37℃、39℃及び40℃で決定した。ウィルスを、室温で1時間、吸着せしめられた連続した10倍希釈溶液を含む24−ウェルプレート上の LLC−MK2単層上に接種した。
ハムスターにおけるrPIVの複製
36匹の生後16週のgolden Syrian ハムスターを、4つのグループ(それぞれ9匹のハムスター)に分け、そして105.5pfuの、ネガティブ−センスcDNAから回収されたrPIV、ポジティブ−センスcDNAから回収されたrPIV, JS cp45 、又はJS wt ウィルスを含む 0.1ml溶液を鼻腔内接種した。4日目、ハムスターを殺し、そして肺及び鼻甲介を収穫した。肺を、 2.5μg/mlのアンホテリシンB(Guality Biologicals, Gaithersburg, MD)、 200μg/mlのピペリシリン(Lederle Laboratories, Pearl River, NY)及び50μg/mlのゲンタマイシン(Quality Biologicals)を含む20%(w/v)L−15懸濁液において均質化した。鼻甲介を同様に、10%(w/v)L−15懸濁液において均質化した。均質化の後、サンプルをアリコートし、そしてドライアイス−エタノール槽において急速に凍結せしめた。サンプルに存在するウィルスを、後日、32℃で LLC−MK2細胞の96ウェルプレートにおいて滴定し、接種の後、5及び7日目でCPEを計測した。平均 log10TCID50/gを、9匹のハムスターの個々のグループについて計算した。
弱毒化された表現型を特定するHPIV3中のアミノ酸置換の同定、 及び感染性の弱毒化されたPIV クローン中への弱毒化突然変異の組込み
cDNAから感染性PIV を生成する能力は、弱毒化された生パラインフルエンザウィルスワクチンの開発を促進する。より詳しくは、本明細書に開示される方法及び手段を用いることによって、PIV 候補ワクチンの弱毒化の遺伝的基礎が容易に決定され、そしてcDNAから生成される感染性PIV ワクチンが、精細に調節されたレベルの弱毒化及び免疫原性を達成するよう企画され得る。
PIV3 JS wt及びcp45ウィルスを、前記のようにして、 LLC−MK2細胞において増殖した(Hallなど, Virus Res. 22 : 173-184 (1992) ;引用により本明細書中に組込まれる)。 vTF7−3組換えワクシニアウィルスは、Fuerstなど, Virology 225 : 419-422 (1996) に記載されており、そしてT7ポリメラーゼを発現する修飾されたワクシニアウィルスAnkara (MVA)は、Wyatt など, Virology 210:202-205 (1995)に記載される(個々の引例は、引用により本明細書中に組込まれる)。 HEp−2(ATCC CCL23)及び LLC−MK2(ATCC CCL7.1)細胞は、2%FBS 、及びゲンタマイシンスルフェート(50μg/ml)により補充されたOpti MEM (Life Technologies)において維持された。
部位特定突然変異誘発により点突然変異をL遺伝子中に導入するために、 pUC19を JS wt PIV3 L遺伝子のフラグメントを受容するよう修飾した。第1に、ユニーク NheI制限部位を、pUC19(N)を創造するために pUC19のMindIII 部位中に、 NheI制限部位を含む1対の相補的オリゴヌクレオチド(5' GATCGATGCTAGCCC 3'(配列番号23)及び5' GATCGGGCTAGCATC 3'(配列番号24))を連結することによって、 pUC19中に導入した。
3種の支持プラスミドpTM(N),pTM(P)及びpTM(L)と共に、1又は複数のcp45 L遺伝子突然変異を担持する個々の十分な長さのアンチゲノムcDNAを、上記のようにして、Lipofect ACE (Life Technologies)及び MVA−T7を用いて、6−ウェルプレート(Costar, Cambridgo, MA)上の HEp−2細胞中にトランスフェクトした。32℃で4日間のインキュベーションの後、トランスフェクション収穫物を、6−ウェルプレート上の HEp−2細胞上に継代し、これを32℃で4日間インキュベートした。個々の継代1の上清液を収穫し、そして LLC−MK2細胞のT−25フラスコ上に通し、これを32℃で5〜6日間インキュベートした。
対照及び組換えウィルスのインビトロプラーク形成の感温性のレベルを、 LLC−MK2単層培養物において、32℃、37℃、38℃、39℃、40℃及び41℃で決定し、そしてプラークを、メチルセルロース被膜の除去に続いて、テンジクネズミの赤血球細胞による血球吸着により数えた。他方では、その単層に存在するウィルスプラークを、1:500 に希釈された、2種の PIV3−特異的抗−HNネズミmAb101/1及び 454/11の混合物によるイムノペルオキシダーゼ染色により同定した(Murphyなど, 前記 (1990))。
6匹のグループにおける生後4〜16週間のgolden Syrian ハムスターに、105.5pfuのrPIV3 JS wt,PIV3 cp35 ウィルス、又は1もしくは複数のcp45 Lタンパク質置換を含むrPIV3を含むOpti MEM1(動物当たり1ml)を、鼻腔内接種した。感染の4日後、ハムスターを殺し、肺及び鼻甲介骨を収穫し、そしてウィルスを上記のようにして定量化した。平均 log10TCID50/gを、6匹のハムスターの個々のグループについて計算した。
wt JS rPIV3中への PIV3 cp45 Lタンパク質アミノ酸置換の導入
上記のようにして、cp45のLタンパク質に存在する3種のアミノ酸置換(表5)を、そのwt親、すなわち PIV3 JS 株をコードするアンチゲノムcDNA中に個々に又は選択された組合せで導入した。個々の導入された突然変異を、回収されたrPIV3における突然変異のモニターを促進するために、近位の天然に存在する制限酵素部位を削除したサイレント突然変異により識別されるよう構築した(表5、図12)。アミノ酸1558でのコード変化が、インビトロ、又はインビボ複製の間、この部位での復帰の機会を低めるために、cp45における1つのnt置換と比較して、r1558における2つのヌクレオチド変化を含むよう企画された。
cp45 Lタンパク質アミノ酸置換の種々の組合せを担持する7種のrPIV3を、32℃、37℃、38℃、39℃、40℃及び41℃で、 LLC−MK2単層上でのプラークを形成するそれらの能力についてアッセイした。表6に示されるように、cp45 aa 置換を担持する個々のrPIV3はtsであり、そしてJS wt rPIV3親は試験されたいづれの温度でも、プラーク形成において制限されなかった。
6匹のGolden Syrrianハムスターのグループを、JS wt rPIV3、生物学的に誘導されたcp45、又は1もしくは複数のcp45 Lタンパク質アミノ酸置換を含むrPIVにより鼻腔内接種し、そして肺及び鼻甲介骨におけるウィルス複製を決定した。この実験〔表7〕においては、単一のアミノ酸置換を担持するrPIV3の各々を、上部及び下部呼吸器官における複製において制限した〔表7〕。しかしながら、r942 、すなわち最小のtsウィルスが、第2の実験において上部及び下部呼吸器官における複製においてわずかに抑制されたに過ぎなかった。それらのデータは、3種のアミノ酸置換のうち2種の置換が、単一の損傷された組換えウィルスとして存在する場合、att表現型に寄与することを示す。
感温性、低温適合化及び弱毒化表現型を特定する、生物学的に誘導された、生存−弱毒化されたHPIVタイプ3ウィルス(cp45)における突然変異の直接的な同定、及び組換えワクチンウィルス中への再構成
上記例は、cp45のLポリメラーゼタンパク質における3種のアミノ酸置換の各々が感温性(ts)及び弱毒化(att) 表現型を付与するが、しかし低温−適合性(ca)表現型を提供しないことを示す(また、Skiadopoulosなど, J. Virol. 72 (3) : 1762-8, 1998 を参照のこと)。cp45は、他のタンパク質(N,C,M,F及びHN)又は可能性あるシス−作用性配列(N遺伝子のリーダー領域及び転写遺伝子開始{GS}シグナル)において12の追加の突然変異を含み、そしてそれらの表現型に対するそれらの寄与は前記のように下線が引かれている。
この例に記載されるrPIV3,PIV3 JS wt及びcp45ウィルスを、上記のようにして、サル LLC−MK2細胞(ATCC CCL7.1) において増殖した(また、Durbinなど, Virology 235 : 323-332, 1997a; Hallなど, Virus Res. 22 (3) : 173-184, 1992; Skiadopoulos など., J. Virol. 72 (3) : 1762-8, 1998を参照のこと)。修飾されたワクシニアウィルスAnkaraを上記のようにして調製した。HEp−2(ATCC CCL 23)及び LLC−MK2細胞を、2%FBS 及びゲンタマイシンスルフェート(50μg/ml)により補充されたOpti MEM1 (Life Technologies, Gaithersburg, MD)、又は10%FBS 、ゲンタマイシンスルフェート(50μg/ml)及び2mMのグルタミンにより補充されたEMEMにおいて維持した。L−132 細胞 (ATCC CCL 5) を、10%FBS,2mMのグルタミン、20mMのHepes,1mMの非必須アミノ酸、及び 100単位のストレプトマイシン−ネオマイシン/mlにより補充されたEarl's MEM (Life Technologies)において増殖せしめた。
3/7(131)2Gp、すなわち感染性ウィルスを回収するために上記で使用された PIV3 JS wtのアン3/7 (131)2GチゲノムcDNAクローンのサブゲノムフラグメント(また、Durbinなど., Virology 235 : 323-332, 1997a; Skiadopoulosなど, J. Virol. 72 (3) : 1762-8, 1988を参照のこと)を、標準的分子クローニング技法を用いて、それらのフラグメントを受容するよう修飾されたpUC19 ベクター中にクローン化した。cp45に同定される突然変異に対応する点突然変異、及びサイレント制限酵素認識配列を創造し又は削除する突然変異(表8)を、前記のようにして、Transformer 突然変異誘発キット(Clontech, Palo Alto, CA)を用いて導入した。

対照及び組換えウィルスのインビトロででのプラーク形成の感温性のレベルを、上記のようにして、 LLC−MK2単層培養物において、32℃、35℃、36℃、37℃、38℃、39℃、40℃及び41℃で決定した(また、Hallなど, Virus Res. 22 (3) : 173-184, 1992 を参照のこと;これは、引用により本明細書中に組込まれる) 。プラークを、メチルセルロースオーバレイの除去に続いて、テンジクネズミの赤血球細胞による血球吸着により数え、又は他方では、単層に存在するウィルスプラークを、1:250 に希釈された、2種の PIV3−特異的抗−HNネズミmAb101/1及び 454/11の混合物によるイムノペルオキシダーゼ染色により同定した(Murphなど., Vaccine 8 (5) : 497-502, 1990 ; van Wyke Coelinghなど., Virology 143 (2): 569-582, 1985 を参照のこと)。
変異体及び wt rPIV3ウィルスの増殖を、24−ウェル組織培養プレートにおいて調製された集密的L−132 細胞単層上で、32℃及び20℃で決定した。2つのプレートの個々のウェルを、 0.2mlの個々の変異体又は wt rPIV3ウィルスにより、0.01の感染の多重度で二重反復して接種した。室温での1時間の吸着の後、接種物を吸引し、そして単層を、ウェル当たり1mlのPBS により洗浄した。
5つのグループにおける生後5週目のGolden Syrian ハムスターを、106.0pfuのJS wt rPIV3,PIV3 cp45 ウィルス、又は変異体rPIV3の1つを含むOpti MEM 1 0.1ml/動物により鼻腔内接種した。感染の4日後、ハムスターを殺し、肺及び鼻甲介を収穫し、そしてウィルスを上記のようにして定量化した。32℃での平均 log10TCID50/gを、個々のグループのハムスターについて計算した。
JS wt rPIV3中への PIV3 cp45 突然変異の導入
cp45の3’リーダー、N GS シグナル、並びにN,C,M,F,HN及びLタンパク質における15の突然変異(表8)を、所望の突然変異を担持するcp45 cDNA のセグメントの部位特定突然変異誘発により又は直接的なPCR 増幅により、完全な PIV3アンチゲノムcDNA中に導入した。次のアンチゲノムcDNAを製造した(図13を参照のこと):(i)リーダー領域の4つの点突然変異、N GS シグナルにおける点突然変異、及びNタンパク質における2つのアミノ酸変化を含むrcp45 3'N ; (ii)Cタンパク質における単一のアミノ酸変化を含むrcp45 C;(iii )Mにおける単一のアミノ酸変化を含むrcp45 M;(iv)Fにおける2つのアミノ酸変化を有するrcp45 F;(v)HNにおける単一のアミノ酸変化を含むrcp45 HN ;(vi)上記のように、Lにおける3個のアミノ酸変化を含むrcp45 L;(vii )上記(i)及び(vi)からの突然変異を含むrcp45 3'XIL ; (viii)Lにおける3個の突然変異を除くすべての突然変異を含むrcp45 3'NCMFHN; 及び(ix)表8に列挙されるすべての15個のcp45突然変異を含むrcp45 。
いくつかの組換えウィルスは、 LLC−MK2細胞に対して試験される場合、特徴的なプラーク形態、すなわち平均サイズ1mmのJS wtrPIV3プラークを示し、そして生物学的に誘導された JS wt PIV3からはサイズ的に区別でなかった。cp45及びrcp45 ウィルスのプラークは、wtよりも大きく、wtよりも直径にして、平均2〜3倍大きく、そしてお互いとは区別できなかった。これは、この表現型に関しての生物学的に誘導された及び組換え体のcp45ウィルスの適合性を示した。
rPIV3を、32℃〜41℃の範囲の許容できる温度及び制限的温度でプラークを形成するそれらの能力についてアッセイした(表9)。
cp45の個々の成分を担持するウィルスのts表現型の分析は、rcp453'N 及びrcp45 Mウィルスが40℃の遮断温度を有し、そしてrcp45C変異体が41℃の遮断温度を有したことを示した。rcp45 F及びrcp45 HN変異体の遮断温度は、41℃以上であった。rcp45 Lウィルスが39℃の遮断温度を有したことは、上記結果と一致した。40℃での複製のその低下(すなわち32℃での力価−40℃での力価)が40℃でのwtウィルスの力価の約 100倍以上である場合、ウィルスはts表現型を有すると思われる。この定義の適用により、この結果は、cp45の少なくとも2つの領域(3’N、及びL)がts表現型に寄与することを示した。
L突然変異がリーダー及び/又はN突然変異と相互作用するかどうかを調べるために、ウィルスrcp45 3'NLを構成した。なぜならば、それらのすべての要素は、RNA 合成の間、相互作用すると思われるからである。このウィルスは、それぞれ、rcp45 3'N 及びrcp45Lに関して40℃及び39℃であるの比較して、36℃の遮断温度を有した。それらの結果は、感温性の増大をもたらす、3’NとL突然変異との間に相互作用が存在することを示唆する。それらの結果はまた、cp45突然変異のサブセットが全組の突然変異を含むrcp45 に関して観察される感温性よりも高レベルの感温性を特定するもう1つの例を提供する。
cp45の遺伝子要素がca表現型を特定したことを測定するために、rPIV3を分析した(表10)。rcp45 Lはts及びatt であるが、しかしcaではないことが上記に示されている。これは、ca表現型の大きな部分を特定する遺伝子要素がLの外部に位置することを示し、そしてこれは、本発明の研究において確かめられた。3’リーダー及びN突然変異を有するrPIVの各々は、中間表現型を示したrcp45 3'NCMFHNを除いて、caであった。これは、ca表現型が3’N領域内にほとんど特定されることを示す。
5匹のGolden Syrian ハムスターのグループを、106TCID50の組換え体又は生物学的に誘導されたウィルスにより鼻腔内接種し、そして肺及び鼻甲介骨におけるウィルス複製のレベルを、4日後に決定した(表11)。接種の14日目、wt及びcp45ウィルスの両者に関して、ハムスターにおいてウィルス複製のピークが存在することがこれまでに示されている(Crookshanks and Belshe, J. Med. Virol.13 (3) : 243-9, 1984 を参照のこと;引用により本明細書に組込まれる)。rcp45 ウィルスは、鼻甲介における複製が約40倍、及び肺における複製が1000倍低められ、そして従って、生物学的に誘導されたcp45ウィルスほどに弱毒化された。それらの結果は、cp45の弱毒化表現型が、その組換え体バージョンにおいて都合良く再生されたことを示す。cp45の個々の表現型は十分にrcp45 において再生されたので、この例に包含されないcp45における追加の5つの突然変異はたぶん、cp45の性質にほとんど寄与しなかった。
赤血球凝集素及び融合糖タンパク質が PIV1からの対応する糖タンパク質により置換されている組換え、キメラ PIV3の回収
この例においては、異なったrPIVバックグラウンド中に、1つのPIV からの1もしくは複数の異種遺伝子又は大きなポリヌクレオチド配列を組込んでいるキメラrPIVウィルスが生成され、そして選択される。本発明のこの観点においては、1つのPIV の個々の遺伝子又は遺伝子セグメントが、異種PIV 又は他の源からのカウンターパート配列により置換される。この例に記載される1つの態様においては、弱毒化された生ワクチンを開発するために適切なキメラ組換え体を生成するために、組換えHPIPV 中にHPIV1又はHPIV2のHN及び/又はF保護抗原を置換するための手段及び方法が提供される。
この研究に使用される PIV1株、 PIV1/Washington/20993 /1964(PIV1/Wash64)が、1964年、Washington DC において単離され、そして成人ヒトボランティアにおける臨床研究においてビルレント野生型ウィルスであることが確認された(Murphyなど, Infect. Immun. 12 : 62-8 (1975);引用により本明細書に組込まれる)。それを、50μg/mlのゲンタマイシンサルフェート、2mMのグルタミン及び0.75μg/mlのトリプシンを含むOpti-MEM 1 (Life Technologies)中の LLC−MK2細胞(ATCC CCL7.1) において増殖せしめた(カタログNo. 3941, Warthington Biochemical Corp., Freehold, NJ)。
PIV3 HN 及びF ORFがそれらの PIV1カウンターパートにより置換されている十分な長さの PIV3アンチゲノムRNA をコードするcDNAを、図14に示されるようにして構成した。 PIV1/Wash64のHN及びF遺伝子のcDNAクローンを、 PIV1/Wash64野生型ウィルスにより感染された LLC−MK2細胞から抽出されたRNA から調製した。cDNAを、ランダムヘキサマープライマー(Life Technologies) を用いて、SuperScript Preamplification System により生成した。 PIV1 F及びHN cDNA を、GenBank に存在するコンセンサス配列に基づいて、遺伝子特異的プライマー対を用いて、Vent DNAポリメラーゼ(New England Biolabs, Beverly, MA)により増幅した。
HEp−2細胞単層を、6−ウェルプレート(Costar Corp. Cambridge, MA)において集密性まで増殖し、そしてトランスフェクションを上記のようにして実施した。細胞単層を、50μg/mlのゲンタマイシン及び2mMのグルタミンを含む無血清Opti-MEM 1(0.8ml)における MVA−T7により、その感染の多重度で感染せしめ、そして次に、3種の支持プラスミド、すなわち 0.4μgのpTM(N), 0.4μgのpTM(P)及び0.05μgのpTM(L)、並びに PIV3−1アンチゲノムcDNA5μgによりトランスフェクトした。プラスミドを、12μlのLipofect ACE (Life Technologies)を含むOpti-MEM 1(0.2ml)において混合し、そして個々のウェルに添加した。
キメラcDNA構造体pSE. PIV3−1.hcを、Circumuent Sequencing Kit (New England Biolabs)により配列決定し、その後、最終的に、十分な長さのクローンpFLC.2G+.hc中にアセンブルした。RNA の分析のために、適切なPIV を、 LLC−MK2細胞のT75フラスコにおいて増幅した。ウィルスを感染の5日目に収穫し、そしてポリエチレングリコール沈殿法(Mbiguinoなど., J. Virol. Methods 31 : 161-70 (1991))により濃縮した。ウィルスRNA を、ウィルスペレットから抽出し、そしてSuperscript Preamplification System (Life Technologies) による逆転写(RT)に使用した。RT−PCRを、Advantage cDNA合成キット(Clontech)、及び PIV1又は PIV3特異的プライマー対を用いて行なった。RT−PCR 生成物を、電気泳動、及びNA45 DEAE ニトロセルロース膜(Schleicher & Schnuell, Keene, NH)のストリップからの溶出により精製し、そして配列決定した。
LLC−MK2単層上のプラークの計数を、前記のようにして行 なったが、但し、トリプシンが、 PIV1及びrPIV3−1ウィルスの場合に添加された(Hallなど. Virus Res. 22 : 173-84 (1992))。手短に言及すれば、連続的に希釈されたウィルスを、6−ウェルプレートにおける LLC−MK2単層上に接種し;ウィルスを32℃で1時間吸着せしめ;培養物を、 0.8%のアガロースを含むL−15培地(Life Technologies)(添加されたトリプシンを有する又は有さない)によりオーバーレイし、そして32℃で6日間インキュベートし;そしてプラークを、アガロースオーバーレイの除去に続いて、テンジクネズミ赤血球細胞による血球吸着(HAD)により同定した。
12匹のグループにおけるGolden Syrian ハムスターにそれぞれ、105pfuのrPIV3/JS, rPIV3−1又は PIV1/Wash64を含むL−15培地 0.1mlを鼻腔内接種した。接種の4及び5日後、個々のグループからの6匹のハムスターを殺し、そしてそれらの肺及び鼻甲介を収穫し、そして均質化し、そして LLC−MK2細胞単層からのサンプルに存在するウィルスを32℃でタイトレーションした。力価は、6匹のハムスターの個々のグループに関して、平均 log10TCID50/gとして表わされた。
十分な長さのキメラ PIV3−1アンチゲノムRNA をコードするcDNAクローンの構成がキメラウィルスrPIV3−1の回復をもたらした
上記に示されるようにして、JS野生型 PIV3 HN 及びF糖タンパク質遺伝子のORF が PIV1/Wash64コード配列のORF により置換されている(図14)、 PIV3及び PIV1キメラcDNAの最終構造体は、6の規則に従う、15,516ntの PIV3−1キメラアンチゲノムRNA をコードする(Durbinなど., Virology 234 : 74-78 (1997))。キメラ PIV3−1アンチゲノムをコードするpFLC.2G+.hc cDNA を、N,P及びL支持プラスミドと一緒に、 HEp−2細胞上にトランスフェクトした。 JS wt PIV3アンチゲノムをコードするp3/7 (131)2G cDNA を、rPIV3対照親ウィルスを生成するために、同時にトランスフェクトした。ウィルスを、 LLC−MK2細胞上での第2の増幅に続いて、個々のトランスフェクションから回収し、そして個々の組換えウィルスがcDNAに由来することを確かめるために、研究を開始した。
Sendaiウィルスのようであるが、しかし PIV3とは異なる PIV1は、組織培養細胞の連続した系上で多サイクル複製を受けるために、そのF糖タンパク質の切断のためにトリプシンを必要とする(Frankなど, J. Clin. Microbiol. 10 : 32-6 (1979)) 。さらに、 FIV1は非細胞障害性ウィルスであり、そして FIV3は広範なCPE を容易に生成する(Collinsなど, Fields Virology, 3rd ed., 1 : 1205-43 (1996)) 。rPIV3−1,rPIV3及び PIV1/Wash64を、それらの性質に基づいて比較した。 PIV1/Wash64のようにrPIV3−1は、テンジクネズミよりもむしろニワトリRBC を用いて高いHA力価を有し、その PIV1/Wash64親のように、rPIV3−1は流体オーバーレイを伴っての培養において効果的な複製のために及び効果的なプラーク形成のためにトリプシンを必要とした。
rPIV3,rPIV3−1及び PIV1 Wash /64の多サイクル複製を、0.01のMOI での LLC−MK2組織培養細胞の接種に続いて評価した(図18)。3種のウィルスの複製の速度動態及び程度は非常に類似することが見出され得る。これは、 PIV1のHN及びF遺伝子の、 PIV3のそれらによる置換が、インビトロでの複製のためにウィルスを弱毒化しなかったことを示す。rPIV3−1がハムスターの上部及び下部呼吸器官における複製のために特にインビボで弱毒化されるかどうかが次に決定された(表14)。rPIV3−1の複製の観察されるレベルは、ハムスターの上部及び下部呼吸器官において、いづれかの親に類似し、そうでなければ、親よりもわずか高かった。
PIV タイプ3の内部タンパク質及びPIV タイプ1の表面糖タンパク質をコードする生存−弱毒化されたキメラ組換えPIV の回収
この例においては、rPIV3−1. cp45Lと称する、生存−弱毒化cp45 PIV3候補体ワクチンウィルスのL遺伝子に見出される変化をコードする3種の感温性及び弱毒化アミノ酸を担持するrPIV3−1の誘導体は、同じ3種の突然変異を有する組換えrPIV3 cp45 Lの表現型に類似する、38℃の遮断温度を有する感温性表現型を表わすことが示されている。rPIV3−1.cp45Lは、rPIV3 cp45 Lと同じ程度に、ハムスターの呼吸器官において弱毒化される。rPIV3−1.cp45Lによるハムスターの感染は、wt PIV1に対して中位のレベルの血球凝集−阻害抗体を生成し、そして野生型 PIV1による攻撃に対して完全な耐性を誘発する。
この研究に使用されるwt PIV1株は、 PIV1/Washington/20993 /1964(PIV1/Wash64)(たとえば、Murphyなど, Infect. Immun. 62 : 1975を参照のこと;引用により本明細書に組込まれる)、及び PIV3 F及びHN ORFが、上記のようにして及びTao など, J. Virol. 72 : 2955, 1998(引用により本明細書に組込まれる)に記載のようにして、 FIV1/Wash64のORF により置換されているキメラ PIV3 cDNA から回収されたキメラrPIV3−1である。それらのウィルスは、50μg/mlのゲンタマイシンスルフェート、及び0.75μg/mlのトリプシン(カタログNO. 3741, Washington Biochemical Corp., Freehold, NJ) により補充されたOpti-MEM 1 (Life Technologies, Gaithersburg, MD)における LLC−MK2細胞において増殖された。
上記(また、Skiadopoulosなど, J. Virol. 72 : 1762, 1998 も参照のこと)、pTM(L)942 /992 /1558プラスミドに存在するcp45の3種のL突然変異が、 PIV1 F及びHN ORF、及び3種のcp45 L遺伝子突然変異を担持する十分な長さのpFLC. 2G+.hc.cp45Lを生成するために、 2.8kbの SphI− NheIフラグメント(PIV3アンチゲノムcDNAにおいてnt11313 〜14092)として、キメラcDNA pFLC.2G+.hc(上記に記載される;また、Tao など, J. 6−ウェルプレートにおける HEp−2細胞単層を、集密的に増殖し、そしてトランスフェクションを、上記のようにして(また、Tao など, J. Virol. 72 : 2955, 1988 も参照のこと)実施した。4日目、収穫の前、トリプシンを、トランスフェクションの3日後、添加し、0.75μg/mlの最終濃度にした。細胞培養物上清液を澄まし、そして新鮮な LLC−MK2細胞単層上に継代した(継代1として言及される)。一晩の吸着の後、培地を、0.75μg/mlのトリプシンを有する新鮮なOpti-MEM 1により交換した。継代1培養物を32℃で4日間インキュベートし、そして上清液に存在するウィルスを収穫し、そして再び、同じ条件下で継代した(継代2として言及される)。継代2収穫物に存在するウィルスを、上記のようにして血球凝集−阻害(HAI)アッセイにより PIV1 HN タンパク質の存在について試験した(また、Tao など, J. Virol. 72 : 2955, 1998 も参照のこと)。
LLC−MK2単層上のプラークの数え上を、 PIV1,rPIV3−1及びrPIV3−1.cp45Lの場合、アガロースオーバーレイに添加される0.75μg/mlのトリプシンを伴って、上記のようにして実施した(また、Tao など, J. Virol. 72 : 2955, 1998 を参照のこと)。種の温度での6日間のインキュベーションの後、アガロースオーバーレイを除去し、そしてプラークを、テンジクネズミ赤血球(RBC)による血球吸着(HAD)により同定した。
5匹のハムスターのグループを、106 プラーク形成単位(PFU)のrPIV3/JS, rPIV3 cp45 L,cp45, PIV1/Wash64, rPIV3−1又はrPIV3−1.cp45Lを含むL15培地 0.1mlにより鼻腔内接種した。ハムスターを、感染の4日後、殺し、そしてそれらの肺及び鼻甲介を収穫し、そして均質化した。組織サンプルに存在するウィルスを、上記のようにして、及びTao など, J. Virol. 72 : 2955, 1998 に記載のようにして、32℃で LLC−MK2細胞単層上で滴定した。力価は、個々のグループに関して、逆数平均 log10TCID50/g組織として表わされる。
ハムスターにおける免疫化及び攻撃の研究
10匹のハムスターのグループを、上記のようにして、動物当たり106 のPFU により鼻腔内免疫化した。血清を、HAI アッセイのために、感染の前、及び33日目に採取した。10匹のハムスターの個々のグループの血清に存在するHAI 抗体のレベルを、抗原として PIV1/Wash64及び PIV3/JSを用いて決定し、そして決定されたHAI 力価を、平均log2として表わす(また、Tao など, J. Virol. 72 : 2955, 1998 を参照のこと)。
組換えキメラウィルスrPIV3−1.cp45Lの回収及び特徴化
上記に示されるように、cDNAクローンpFLC.2G+.hc、すなわちF及びHN糖タンパク質をコードするORF が PIV1のORF により置換されている PIV3の十分な長さのアンチゲノムcDNAを、cp45のL遺伝子に同定され、そして独立したts及び弱毒化突然変異であることが示されている3種のアミノ酸コード変化(Lタンパク質におけるアミノ酸位置に従って、942, 992及び1558と称する)の導入により修飾した(図19;また、Skiadopoulosなど, J. Virol. 72 : 1762, 1998 も参照のこと)。
cp45の3種のL遺伝子突然変異は、wt PIV3中に導入される場合、ts表現型を付与することが上記に示された(また、Skiadopoulosなど, J. Virol. 72 : 1762, 1998)。キメラウィルスにおけるそれらの存在が同じ効果を有するかどうかを評価するために、rPIV3−1.cp45Lのプラーク形成効率を種々の温度で決定した。表15に示されるように、3種のL突然変異は実際、キメラウィルスに表現型を付与した。組換えウィルスrPIV3 cp45 L及びrPIV3−1.cp45Lにおけるcp45 L突然変異により特定される感温性のレベルは等しく(表15)、このことは、突然変異の効果が、 PIV3又は PIV1 HN 及びF糖タンパク質と無関係であることを示す。rPIV3 cp45L及びrPIV3−1.cp45Lの感温性のレベルは、後者のウィルスがLの外部の突然変異を有する事実にかかわりなく、生物学的に誘導されたcp45ウィルスのレベルに匹敵した(Stokesなど, Virus Res. 30 : 43, 1993を参照のこと;引用により本明細書に組込まれる)。
cp45の3種のL遺伝子突然変異は、wt PIV3中に導入される場合、ハムスターの上部及び下部呼吸器官においてウィルス複製の弱毒化を付与することが上記で示された(また、Skiadopoulosなど, J. Virol. 72 : 1762, 1998 を参照のこと)。キメラウィルスに対するそれらの効果を、表16に示されるように、ハムスターの鼻腔内感染により評価した。それらの発見は、rPIV3−1.cp45Lが実際、両部位で弱毒化され、そしてさらに、弱毒化のそのレベルがrPIV3 cp45 Lのレベルに匹敵したことを示す。従って、感温性のように、弱毒化を付与するcp45 L突然変異の能力は、表面糖タンパク質の抗原特異性に無関係である。
キメラrPIV3−1ウィルス及びその弱毒化されたrPIV3−1.cp45L誘導体を、ハムスターにおいて免疫原性及び保護効率について評価した。表17に示されるように、いづれかのウィルスによる感染は、 PIV3に対してではなく、 PIV1に対してHAI 抗体を誘発し、このことは、それらのキメラウィルスが PIV1 HN 糖タンパク質を有し、そして高い免疫原性であることを確認する。rPIV3−1.cp45Lにより誘発されるHAI 抗体のレベルはrPIV3−1によるレベルよりも2倍近く、このことは、その弱毒化が免疫原性において中位の低下をもたらしたことを示す。同様に、rPIV3及びrPIV3 cp45Lは PIV1に対してではなく、 PIV3に対してHAI 抗体を誘発し、そしてその弱毒化されたウィルスにより誘発されるレベルは約2倍、低かった。cp45 L突然変異を担持する組換えウィルスのハムスターにおける制限された複製にもかかわらず、rPIV3 cp45 L又はrPIV3−1.cp45Lのいづれかによる感染は、相同糖タンパク質を担持する攻撃ウィルスの複製に対して完全な耐性を誘発した。
耐性におけるPIV 中の非HN又はF糖タンパク質(すなわち、内部タンパク質)の役割に対する情報は制限されている。本明細書におけるrPIV3−1及びrPIV3−1.cp45Lの開示及び使用は、免疫化及び攻撃ウィルスにより共有される遺伝子のみが内部タンパク質遺伝子であるので、内部タンパク質が PIV3による攻撃に対する耐性において演じる役割を試験する機会を提供する。 PIV3攻撃ウィルスの複製は、rPIV3−1又はrPIV3−1.cp45Lによるハムスターの先の感染により上部及び下部呼吸器官において有意に制限される(表17)。従って、それらのデータは、HN及びFタンパク質のように、 PIV3の内部タンパク質が攻撃 PIV3の複製に対して部分的な耐性を誘発できることを示す。
Claims (15)
- PIV ゲノム又はアンチゲノムをコードするポリヌクレオチド配列を含んで成るポリヌクレオチド分子であって、前記PIV ゲノム又はアンチゲノムをコードする前記ポリヌクレオチド配列が、JS cp45のTyr942、Leu992及び/又はThr1558に対応するアミノ酸をコードする位置においてヌクレオチド置換により、アミノ酸置換が生ずるように修飾されることを特徴とするポリヌクレオチド分子。
- 前記PIV ゲノム又はアンチゲノムをコードする前記ポリヌクレオチド配列が、Nタンパク質におけるJS cp45のアミノ酸Val96又はSer389に対応する位置の少なくとも1つのアミノ酸置換をコードする請求項1に記載のポリヌクレオチド分子。
- 前記PIV ゲノム又はアンチゲノムをコードする前記ポリヌクレオチド配列が、Cタンパク質におけるJS bcp45のアミノ酸Ile96に対応する位置のアミノ酸置換をコードする請求項1に記載のポリヌクレオチド分子。
- 前記PIV ゲノム又はアンチゲノムをコードする前記ポリヌクレオチド配列が、Fタンパク質におけるJS cp45のアミノ酸Ile420又はAla450に対応する位置の少なくとも1つのアミノ酸置換をコードする請求項1に記載のポリヌクレオチド分子。
- 前記PIV ゲノム又はアンチゲノムをコードする前記ポリヌクレオチド配列が、HNタンパク質におけるJS cp45のアミノ酸Val384に対応する位置のアミノ酸置換をコードする請求項1に記載のポリヌクレオチド分子。
- 前記PIV ゲノム又はアンチゲノムをコードする前記ポリヌクレオチド配列が3’リーダー配列におけるJS cp45のヌクレオチド23、24、28又は45に対応する位置の少なくとも1つの突然変異を組込む請求項1に記載のポリヌクレオチド分子。
- 前記PIV ゲノム又はアンチゲノムをコードする前記ポリヌクレオチド配列が、N遺伝子開始配列におけるJScp45のヌクレオチド62に対応する位置の変異を組込む請求項1に記載のポリヌクレオチド分子。
- 前記PIV ゲノム又はアンチゲノムをコードする前記ポリヌクレオチド配列が、rcp45又はrcp45 3'NLに存在する変異を含む、請求項1に記載のポリヌクレオチド分子。
- 請求項1〜8のいずれか1項に記載のPIV ゲノム又はアンチゲノムをコードするポリヌクレオチド分子を含んで成る発現ベクター、及びPIV のN,P及びLタンパク質をコードする1又は複数のポリヌクレオチド分子を含んで成る少なくとも1の発現ベクターを包含し、それにより、前記PIVゲノム又はアンチゲノム、及びN,P、及びLタンパク質の発現が感染性PIV 粒子を生成する細胞又は細胞−フリー組成物。
- 前記PIV ゲノム又はアンチゲノムをコードするポリヌクレオチド、及びPIV のN,P及びLタンパク質をコードする1又は複数のポリヌクレオチドが、少なくとも2の発現ベクター内に組込まれる請求項9に記載の細胞又は細胞−フリー組成物。
- 請求項1〜8のいずれか1項に記載のポリヌクレオチド配列を含んでなる組換えPIV ゲノム又はアンチゲノム、Nタンパク質、Pタンパク質、及びLタンパク質を含んで成る感染性PIV 粒子。
- 前記組換えPIV ゲノム又はアンチゲノムが、
i)p218(131)(配列番号1);
ii)p3/7(131)(配列番号14);又は
iii )p3/7(131)2G(配列番号15)、
から選択される請求項11に記載の感染性PIV 粒子。 - 前記組換えPIV ゲノム又はアンチゲノムがRSV 又は麻疹ウィルスからの異種配列を組込む請求項11に記載の感染性PIV 粒子。
- 医薬的に許容できるキャリヤーに、請求項11〜13のいずれか1項に記載の感染性PIV 粒子の免疫原的有効量を含んで成る免疫原組成物。
- 請求項1〜8のいずれか1項に記載のポリヌクレオチド、並びに作用可能に連結されたプロモーター及び転写ターミネーターを含んでなる発現ベクター。
