Description
やアルブミンの微小球(microsphere)の利用ではまず最初に液体懸濁液の形態で形成し、固体化させる過程で気体を封入する。しかし、気体を封入している固相の殻は、気体が生体内で満足できるだけ寿命をもつには脆弱あるいは透過性過ぎることが一般的に証明されている。更に、厚い殻(例えば、アルブミン、糖、あるいは他の粘性の物質)は小泡の圧縮率を減少させる。従って、それらが持続している短い時間における反射生成率を減少させる。注射時にガスあるいは泡を放出する固体粒子あるいは液体エマルジョンの小滴は、(Quay,PCT/US94/00422のように)血液が気体あるいは蒸気で過飽和になる危険が生じる。これは、予定された多数の小泡よりもむしろ、利用できる核となりうる部位がある場所で少数の塞栓形成性の大きな気泡の形成を導くことになろう。また、生体内でこのように形成された泡は「むき出し」であり、結果として前述したように補体活性化の問題を有するであろう。

次の噴霧乾燥のため、第一の界面活性剤として疎水性リン脂質と、それに加えて少なくとも1つ以上の親水性の界面活性剤とを含有する第一の水性溶液を調製する。好ましくは、疎水性リン脂質は、全部で約10個以上の炭素原子(例えばデカノイルリン脂質)を有する1つ以上のアシル鎖を有する。いくつかの実施態様では、リン脂質第一界面活性剤は約10あるいは14から約20あるいは24個の炭素原子のアシル鎖を有する。例えば、ジパルミトイルホスファチジルコリン(各々16個の炭素原子を含む2本のアシル鎖を含む)が使用できる。アシル鎖は水素化(hydrogenated)されていてもフルオロ化されていても良い。他のリン脂質の頭部グループもまた考えられる。たとえば、ホスファチジルセリン、ホスファチジルグリセロール、ホスファチジルエタノールアミンは、本発明に適した特
性を有している。このようなリン脂質の組合せもまた、卵あるいは大豆レシチンや肺界面活性物質のような天然物由来のリン脂質物質と同様に、「第一界面活性剤」を構成することができる。さらに、リン脂質第一界面活性剤は他の極めて水不溶性の界面活性剤、例えばスクロースジ、トリ、あるいはテトラエステルで補足しても良い。コレステロールも第一界面活性剤を補うことができ、コレステロール/リン脂質の重量比でおよそ0.01から0.5の範囲で供給された場合に安定性を増す効果があることが知られている。不飽和のアシル基も本発明の範囲内であるが、好ましくは、リン脂質のアシル基は飽和されているほうがよい。第一界面活性剤は、好ましくは溶液のおよそ0.005〜20%w/vで供給されるのがよく、0.02〜10%w/vが最も好ましい。

界面活性剤/膨張剤エマルジョンは、好ましくは、上記したタイプの親水性モノマー/ポリマーの溶液及び塩と合わされ、乾燥した中空でほぼ微小球状の構造を形成させるために噴霧乾燥させられる。市販されている噴霧乾燥器は当業者にはよく知られたものであり、具体的な前駆物質溶液に適したセッティングが、以下の例を参照して経験的な標準試験を通じて容易に決定されうる。例I〜VIIおよびIX〜XIIで使用される、「Niroポータブル噴霧乾燥器」は、水性界面活性剤溶液を直径2〜20ミクロンの範囲の小滴に粉砕するために圧縮した空気の高速度噴射を使用し、二つの流動性圧縮空気ノズルで界面活性剤含有溶液を霧に吹くことによって機能する。その後、それらの小滴は乾燥室の上部で熱い空気(典型的には200〜375℃)の流れの中に注入される。それから、界面活性剤溶液の小滴は、約100℃の沸点までほとんど即時に熱せられる。蒸気冷却が温度がより以上に上昇することを防ぐが、温度はリン脂質のガラス転移温度よりもまだ高く、Poloxamer188やサッカロースステアリン酸のような他の多くの界面活性剤の融点よりも高い。

比較的圧縮されない液体が加圧され、シール15を下部チャンバー30方向へ押すことになる。これによって水性溶液25が下部チャンバー30に放出され、粉末35を溶解し気体浸透剤を捕捉した安定化された微小泡45を形成することになる。過剰の気体浸透剤40は下部チャンバー30から上部チャンバー20へ放出される。この配置は、使用者にとって便利で、尚且つ、厚い(0.5〜1.25インチ)水性溶液の層によりチャンバー間のシールで覆うことによって、下部チャンバー中の少量の水不溶性の気体浸透剤を密封すると言う予期しない利点と、粉末チャンバーの圧力を約10%以上上昇させずに、水溶液を下部チャンバーに導入できるという利点をもたらす。従って、圧力ベントは必要ない。(これに対して、ベントなしに注射針と注射器を用いて単一チャンバーのバイアル中で再構築する通常の方法では、チャンバー内にかなりの圧力が発生し、微小泡をつぶしてしまう可能性がある。)

ouis, MO)で調製した。Szoka及びPapahadjopoulosの逆相蒸発法(Proc. Nat. Acad. Sci. 75 USA (1978), 4194)に従って、界面活性剤をジエチルエーテル/クロロホルムの1/1v/v溶液120mLに溶解した。脱イオン水40mLを加えた。混合物は3mmプローブソニケーター(50 W Vibra Cell, Sonics & Materials Inc., Danbury CT)を使用して、0〜4℃で10分間超音波処理し、エマルジョンを生成した。リポソーム分散液はロータリーエバポレーターで減圧下に溶媒を除去し、溶液を1.0μmポリカーボネートフィルターで、65℃で濾過することによって生成した。リポソーム溶液の分画(1mL)を、10mLの超音波バイアル中で、15%w/vマルトース(Sigma, St. Louis)(4mL)と混合し、−30℃で凍結させ凍結乾燥した(FTS Systems, Stone Ridge NY)。バイアルは窒素または13℃でパーフルオロヘキサンで飽和させた窒素で処理した。凍結乾燥した粉体は水(5mL)で再構成し下記の濃度とした。

Schneiderらの米国特許第5,380,519号の実施例4に記載の割合でリン脂質及びリン酸ジセチルを含有する処方を、下記のエマルジョンを噴霧乾燥することによって調製した。Schneiderの実施例のように、界面活性剤を薄層としなかった(リポソームに変換しなかった)。
し、溶液2は界面活性剤を含有した。溶液2は、高せん断ミキサーに添加し、氷浴中で冷却した。1,1,2−トリクロロトリフルオロエタン(フレオン133)の粗懸濁液を、溶液2(1L)中で調製した。この懸濁液を、マイクロフルイドダイザーMicrofluidizer(Microfluidics Corporation, Newton, MA;モデルM-110F)を使用して、10,000psi、5℃で5パスさせて、乳化した。得られたエマルジョンを溶液1に加え、噴霧乾燥する下記の処方を調製した。

噴霧乾燥粉体(800mg)を、20mLのWheaton RS-177FLWの二チャンバーのバイアル(図1)の下のチャンバーに量り入れた。バイアルはチャンバー間の封止材を挿入する前に、13℃でパーフルオロヘキサンで飽和させた窒素でフラッシュした。上のチャンバーは注射用滅菌水(10mL)で満たした。上のチャンバーのストッパーを挿入し、上のチャンバー内の全ての空気の泡を除去した。上のストッパーを押して、チャンバー間の封止材を下のチャンバー内に落とし、水を下のチャンバーに流し入れ、粉体を再構成した(図2)。光学顕微鏡で示されるように、無数の安定な微小泡が形成された。この方法によって、パッケージのこの形態が便利であること、及び水相を粉体に加えたときの圧力の上昇を除くためのベントを設ける必要が無くなることが示される。
噴霧乾燥粉体(100mg)を、5mL+5mLのHYPAK液体/乾燥デュアルチャンバーシリンジ(Becton Dickinson, Flanklin Lakes NJ)中に量り入れ、粉体チャンバー(針状末端)に振り入れた。次に、チャンバー間の封止材をバイパスチャネルの直ぐ上に配置した。次に、5μMのフィルターを含有する針をシリンジの上に取り付けた。次に、粉体を含有するチャンバーは、真空チャンバー内に器具を設置し、真空にし、13℃で気体浸透圧剤であるパーフルオロヘキサンで飽和させた窒素でチャンバーを再充填することによって、気体浸透圧剤で満たした。フィルター針によって、粉体含有チャンバー内の雰囲気を真空化し、再充填した。次に、封止針のカバーを針の上にかぶせた。次に、液体チャンバーを注射用水(4mL)で満たし、一時的なベント(全ての気泡を取り除くために、ガラスのシリンジとプランジャとの間に挿入した針金)を使用して、プランジャを固定した。
Claims (19)
- N2、CO2及びO2からなる群から選択される気体又はそれらの混合物を含む第一気体と、パーフルオロカーボンである第二気体とを含む気体混合物を含み、気体混合物は膜形成材料により囲まれている、液体媒体中の安定化気体封入微小泡製剤であって、第一気体及び第二気体は、1:100〜1000:1のモル比で存在し、第一気体は、37℃で少なくとも(760−x)mmHgの蒸気圧を有し、xは、第二気体の37℃での蒸気圧であり、第一気体及び第二気体のそれぞれの蒸気圧は37℃で75mmHgより大きく、第一気体及び第二気体は水蒸気ではなく、
膜形成材料が、10個以上の炭素原子を含む一つ以上のアシル鎖を有する第一リン脂質からなり且つ全界面活性剤の少なくとも5%w/wを構成する第一の界面活性剤と、脂肪酸の糖エステル、ポリオキシプロピレン−ポリオキシエチレンコポリマー、ポリオキシエチレン脂肪酸エステル、第二リン脂質及び非イオン性アルキルグルコシドからなる群から選ばれる少なくとも一つからなり且つ前記第一の界面活性剤より高い水溶性を有する第二の界面活性剤とを含む界面活性剤混合物である、前記製剤。 - パーフルオロカーボンが、パーフルオロメタン、パーフルオロエタン、パーフルオロプロパン、パーフルオロブタン、パーフルオロシクロブタン、パーフルオロペンタン、パーフルオロシクロペンタン、パーフルオロヘキサン、パーフルオロシクロヘキサン、パーフルオロヘプタン、パーフルオロシクロへプタン、パーフルオロオクタン及びパーフルオロシクロオクタンからなる群から選択されるか又はそれらの混合物である請求項1記載の安定化気体封入微小泡製剤。
- 膜形成材料が、さらにアルブミンを含む、請求項1又は2記載の安定化気体封入微小泡製剤。
- 第一気体及び第二気体が、1:10〜100:1のモル比で存在する請求項1〜3の何れか一項記載の安定化気体封入微小泡製剤。
- 第一リン脂質が、ホスファチジルコリン、ホスファチジルセリン、ホスファチジルグリセロール、及びホスファチジルエタノールアミンからなる群から選ばれる少なくとも一つである、請求項1〜4の何れか一項記載の安定化気体封入微小泡製剤。
- 第一リン脂質が、ジミリストイルホスファチジルコリンである請求項5記載の安定化気体封入微小泡製剤。
- 膜形成材料が、さらにコレステロールを含む、請求項1〜6の何れか一項記載の安定化気体封入微小泡製剤。
- 液体媒体が水系である請求項1〜7の何れか一項記載の安定化気体封入微小泡製剤。
- 液体媒体及び微小泡が容器中にある請求項1〜8の何れか一項記載の安定化気体封入微小泡製剤。
- 液体媒体がin vivoの血液である請求項1〜7の何れか一項記載の安定化気体封入微小泡製剤。
- 安定化気体封入微小泡製剤が液体媒体中に調製される時に微小泡が第一平均直径を有しており、微小泡が、第一気体が該媒体中へ失われるために前記液体媒体中で第一平均直径の75%よりも小さい第二平均直径に収縮し、該媒体に対する気体浸透圧の差のために第二平均直径又はその直径付近で少なくとも1分間安定化される請求項1〜10の何れか一項記載の安定化気体封入微小泡製剤。
- 第一平均直径が10ミクロンであり、第二平均直径が1〜6ミクロンである請求項11記載の安定化気体封入微小泡製剤。
- 微小泡が機械的振盪若しくは振動、又は、単純振盪、又は、温和な攪拌により形成された請求項1〜12の何れか一項記載の安定化気体封入微小泡製剤。
- パーフルオロカーボンが、パーフルオロプロパン、パーフルオロブタン、パーフルオロペンタン、パーフルオロヘキサン、パーフルオロヘプタン及びパーフルオロオクタンからなる群から選択されるか又はそれらの混合物である請求項1〜13の何れか一項記載の安定化気体封入微小泡製剤。
- 微小泡が浸透圧により安定化された請求項11〜14の何れか一項記載の安定化気体封入微小泡製剤。
- 微小泡が診断画像化に使用される請求項1〜15の何れか一項記載の安定化気体封入微小泡製剤。
- 微小泡が少なくとも20秒の半減期を有する請求項1〜16の何れか一項記載の安定化気体封入微小泡製剤。
- 第一気体が窒素であり、第二気体がパーフルオロヘキサンである請求項1〜17の何れか一項記載の安定化気体封入微小泡製剤。
- 第一リン脂質が微小泡上に単層形態で存在する請求項1〜18の何れか一項記載の安定化気体封入微小泡製剤。
