Description
療用途のための、生体に投与するのに適切な水性液体中
の空気もしくは気体封入微小球の安定な分散体又は溶液
の製造のための方法に関する。
物及び以下の方法を実施することにより得られる所望の
安定微小球溶液又は懸濁物に関する。
イクロボディー)又は微小球体(マイクロカプセル)、
例えば微小気泡(マイクロバフル)又は微小中空球(マ
イクロバルーン)はエコグラフィーのために特に有効な
超音波反響体であることがよく知られている。本明細書
において「微小気泡」なる語は、分割状態において空気
又は気体を液体に導入することにより一般的に得られ
る、液体中において懸濁状態における空気又は気体の球
体を特に表わし、該液体は好ましくはこの気泡の表面特
性及び安定性をコントロールするために界面活性剤(サ
ーファクタント又はテンサイド)も含む。「微小カプセ
ル」又は「微小中空球」なる語は好ましくは物質的境界
もしくは囲い、即ち、ポリマー膜壁を有する空気又は気
体を意味する。微小気泡及び微小中空球の両方共超音波
造影剤として有用である。例えば搬送液中の微小気泡又
は微小中空球の懸濁物(0.5〜10μmに範囲する)の生
体の血流への導入は超音波エコグラフィーイメージング
を強く助成するであろう。従ってこのことは内臓の識別
化に役立つ。血管及び内臓のイメージングは医療診断、
例えば心臓血管及びその他の疾患の検査に大いに役立つ
ことができる。
の微小気泡の懸濁物の状態の形成は、加圧のもとでこの
液体中に溶解している気体の放出により、あるいは気体
性の生成物を発生せしめる化学反応により、あるいは空
気もしくは気体を中に取り込んでいる又は吸着して含む
液体に可溶性又は不溶性な固形物の混合により提供され
うる。
無菌注射可能液体キャリア中の気体微小気泡の懸濁物の
調製に関する一連の異なる技術であって、(a)搬送液
(水性)中の界面活性剤の溶液と(b)安定剤としての
粘度上昇剤の溶液を利用する技術が開示されている。気
泡を発生させるため、これに開示されている技術には、
小さな開口部に(a),(b)及び空気の混合物を高速
にて通すこと、又は(a)を(b)の中に生理学的に受
け入れられている気体と一緒に利用する直前に、注入せ
しめること;あるいは(a)に酸を加えそして(b)に
炭酸塩を加え、両方の配合物を利用直前に一緒に混合せ
しめることにより、酸が炭酸塩と反応してCO2気泡を発
生せしめること;あるいは(a)と(b)の混合物の保
存品の中に過剰加圧気体を加えることにより、この混合
物を注射のために利用する時にこの気体が微小気泡とな
って放出されることが含まれている。
であって、安定注射可能搬送液、例えば塩の水溶液又は
マルトース、デキトロース、ラクトースもしくはガラク
トースのような糖類の水溶液を、封入空気を含む同じ糖
類の固形微粒子(0.1〜1μmの範囲における)と混合
せしめる方法を開示している。この液体キャリア中の気
泡の懸濁物を作るため、この液体及び固形成分の両者を
数秒間無菌状態のもとで一緒に攪拌する。これが終了し
たなら、この懸濁物を直ちに使用する、即ち、これはエ
コグラフィー検査のために5〜10分以内に注射しなけれ
ばならない。実際にこれらは消失し易いため、この時間
を過ぎるとこの気泡濃度は実用的には低すぎるようにな
る。従って、注射のための微小気泡溶液の利用に関する
1つの重要な問題は、時間に伴う安定性の欠如である。
本発明の目的はこの欠点を解消することにある。
問題はサイズである。一般に認められている通り、細い
血管中にわたる移送を可能にする微小気泡の有用なサイ
ズは約0.5〜10μmに範囲し、大きい気泡は凝結及びそ
の結果としての塞栓症の危険性を伴う。例えば、US−A
−4,446,442号(Schering)に開示されている気泡懸濁
物であって、界面活性剤例えばレシチン、脂肪酸のエス
テル及びエーテル、並びにポリオキシエチレン及びポリ
オキシエチル化ポリオール例えばソルビトール、グリコ
ールとグリセロール、コレステロール又はポリオキシ−
エチレン−ポリオキシプロピレンポリマーを有する脂肪
アルコールの水溶液を、粘度上昇化合物並びに安定化化
合物、例えば単−及び多糖類(グルコース、ラクトー
ス、スクロース、デキストラン、ソルビール)、ポリオ
ール例えばグリセロール、ポリグリコール;並びにポリ
ペプチド例えばタンパク質、ゼラチン、オキシポリゼラ
チン及び血漿タンパク質の溶液と強く攪拌せしめたもの
において、40〜50μmより小さい微小中空球は約50%の
みであり、従ってこのような懸濁物はあらゆるエコグラ
フィー用途に適していない。本発明はまた、この気泡の
サイズを機械的に小さくすることによってこの問題を解
決することを可能にする。
試みにおいて、微小中空球、即ち、物質的な壁を有する
微小球が開発された。前述した通り、微小気泡は無形又
は消失性の囲いのみを有すが(即ち、それらは液状の壁
であってその表面張力は界面活性剤の存在により改質さ
れているもののみに囲まれている)、微小中空球又は微
小カプセルは一定の機械的強度を有す物質的な材料、例
えばポリマー膜より作られた有形な囲いを有する。言い
換えるならば、それらは空気又は気体が多かれ少なかれ
強く封入されている物質の微小球である。
イメージを増強せしめるための気体を含む表面膜微小カ
プセルの利用を開示し、この膜は複数の無毒及び非抗原
性有機分子を含む。開示されている態様において、これ
らの微小気泡は凝集を防ぐゼラチン膜を有し、そしてそ
れらの好ましいサイズは5〜10μmである。これらの微
小気泡の膜はエコグラフィー検査を行なうのに十分に安
定であると言われている。しかしながら、一定の時間が
経過したら、その中に封入されている気体は血流中にお
いて溶解し、そしてこの気体は徐々に消失するであろう
ということも言われており、これはおそらくゼラチンの
ゆっくりとした溶解に起因するであろう。従ってこの技
術によってこの不安定性の問題は完全には解決されてお
らず、従って利用する直前迄この微小カプセルはそれら
が保存安定性であるゼラチン溶液の中に保存される。し
かしこの懸濁物を注射物の作製のために利用する際、こ
のゼラチンは液状となるように加熱及び溶融される必要
があり、これは不便である。
−A−4,718,433号(Feinstein)に開示されている。こ
れらの微小球はタンパク質溶液例えば5%の血液アルブ
ミンの高周波処理(ソニケーション)(5〜30KHz)に
より作られ、そして2〜20μm、主として2〜4μmの
範囲における粒径を有する。この微小球は高周波処理後
の膜形成タンパク質の変性、例えば加熱又は化学的手
段、例えばホルムアルデヒドもしくはグルタルアルデヒ
ドとの反応を用いることにより安定化されている。この
方法により得られる安定微小球の濃度は2〜4μmの範
囲において約8×106/mlであり、4〜5μmの範囲にお
いて約106/mlであり、そして5〜6μmの範囲において
5×105以下であると言われている。これらの微小球の
安定な時間は48時間又はそれより長いものと言われてお
り、そしてそれらは静脈内注射後に左心臓イメージング
を好適に可能とした。例えば、末梢血管の中に注入せし
めた際のこの高周波処理アルブミン微小気泡は経肺動脈
通過(transpulmonary passage)が可能である。このこ
とは左心室及び心筋組織のエコーグラフ不透明化をもた
らす。
れた微小中空球がEP−A−324,938号(Widder)におい
て報告されている。この明細書において、数ヶ月又はそ
れより長い半減期を有する、10μm以下の高濃度(108
以上)空気封入タンパク質被覆化微小球が開示されてい
る。これらの微小中空球の水性懸濁物は熱変性しうるタ
ンパク質、例えばヒト血清アルブミンの溶液の超音波キ
ャビテーションにより作られ、この操作は膜形成タンパ
ク質のある程度の発泡化及び加熱によるその後の硬化を
もたらす。その他のタンパク質、例えばヘモグロビン及
びコラーゲンもこの文献の方法において好都合であると
言われているが、我々の操作においてそのようではなか
った。
い保存安定性は、即ち液体キャリア相と一緒にそれらが
市販されることを可能とし、このことは使用前の調製が
もはや必要としなくなったために強い商業価値を有す
る。しかしながら、この膜物質の極端な強さ及び安定性
はいくつかの欠点を有する。例えばその硬さの理由によ
り、この膜はこの微小球のさらされうる突然の圧力変化
に耐えることができない。この圧力変化は例えば血流を
移動する際、心臓の脈動に起因しうる。従って実際上の
超音波検査のもとでは、一部の微小球は破裂することが
あり、これはイメージングの再現性を悪くするであろ
う。更に、柔軟な壁を有する微小球は、硬質の壁を有す
る対応の微小球よりも反響性であることも知られてい
る。
る材料の過剰な安定性は検査される生体によるその生物
分解を遅らせ、従って代謝の問題をもたらしうる。従っ
て、柔かく且つ弾性な膜により覆われている圧力耐久性
微小中空球であって種々の圧力のもとで一時的に変形で
き、且つ高い反響性の付与されているものを開発するこ
とがより好ましい。
物の調製の方法は2段階のヒト血清アルブミン(HSA)
の高周波処理を包含する。第1段階において、細いチュ
ーブ中における数mlの該タンパク質の希釈溶液を、その
中にソニケーター(高周波器)ヘッドを浸すことによっ
て高周波処理にかける;第2段階において、このソニケ
ーターヘッドをこの溶液のレベルより高く持ち上げて高
周波処理をかけ、これにより発泡が起こりうる。次にこ
の高周波処理溶液をデカント処理のために一夜放置し、
そしてこの上層に集まっている微小中空球を集める(フ
ロート分離)。
量のインクレメントによるタンパク質溶液の処理を更に
採用している(EP−A−324,938号の頁4、行9)。従
ってこれは工業生産に必要とされる大量の微小中空球を
得るには明らかに不都合である。
気泡の剪断の採用による機械的な縮小が連続的又はほぼ
同時に行なわれる本発明の方法並びに組成物は以上の欠
点を解決するであろう(請求の範囲を参照のこと)。以
下に定義する条件のもとで、水性懸濁物の気泡のサイズ
は有意なる気泡の崩壊及び気泡計測値の損失を伴うこと
なく所望の値の範囲に迄小さくされうることは実際に非
常に驚くべきことであり且つ予想し得ないことである。
(filmogenic)タンパク質の水溶液を任意的に発泡剤の
存在下において泡へと変換せしめること、即ち、該溶液
に空気又は気体の気泡の分散体を付与し、そしてその後
又は同時に、摩擦又は振動によって該気泡のサイズ(通
常はまず100μm又はそれより大きい範囲である)を所
望の値、通常は0.5〜10μmの範囲、好ましくは4〜10
μmの範囲迄機械的に小さくすることより成る。これは
この泡を強い剪断にかけること、例えば機械的もしくは
気体圧力のもとでこれを小さな開口部又は通路に通すこ
とにより成し遂げられる。該通路は任意的にこの剪断効
果を高めるためのバッフル(baffle)手段を含む。これ
はホモジナイザー−乳化装置によるか、又はバッフル化
(遮断)されているもしくはねじれている通路のネット
ワークの施されている無機又は有機媒体に通すことによ
りもたらすことができ、例えばこの材料は非常に粗い多
孔性材料でありうる。多孔性無機媒体は種々のサイズの
無機粒子の層又はセラミックフィルターのような濾過助
剤を含む。有機媒体は当業者によく知られているフィル
タースクリーンを含む。バッフル化通路を通すことによ
る気泡のサイズ縮小の際、剪断効果は徐々に適用するべ
きである。即ち、この処理は連続的に少量づつの泡にの
み適用すべきである。同時に大量の泡に剪断を適用する
こと、即ち、大量の部分に高すぎる圧力をかけること
は、単に圧縮による一時的な気泡サイズの縮小をもたら
し、従って最終的にこの圧力の解放に基づいてもとの気
泡のサイズに復帰しうる。発泡及び気泡サイズ縮小は、
1工程において発泡及びサイズ減縮の両方を連続的に行
なうことができる適切なホモジナイザー−乳化機を用い
る場合に同時に行うことができることが明らかである。
我々は以降にこの装置の好ましい機械的条件について示
す。音波又は超音波振動も気泡サイズ縮小に有効であり
うる。
好ましくは0.1〜5重量%、最も好ましくは0.2〜4.9重
量%の膜形成タンパク質、即ち、溶液の中に存在してい
る場合に泡の気泡の廻りに膜を形成せしめるのに役立つ
ことができるタンパク質を含んで成り、このような膜は
気泡のサイズ縮小の際にわたるホモジナイザーの機械的
作用のもとで壊れることのないほど十分に安定であるべ
きである。上記の条件を満足せしめるタンパク質は数多
くあり、そしてヒト血清アルブミン(HSA)、牛及び羊
血清アルブミン(BSA及びOSA)、卵アルブミン、ヘモグ
ロビン、フィブリノーゲン、グロブリン、コラーゲン、
ゼラチン、発泡性タンパク質例えば加水分解野菜タンパ
ク質、グルテンの加水分解画分(ハイフォーマ:Hyfoam
a)、加水分解カゼイン等が含まれる。タンパク質が本
質的に発泡特性を有さない又はわずかにしか有さない場
合、発泡剤例えば生理学的に受け入れられる界面活性剤
を任意的に加えることができる。広範囲にわたる常用の
界面活性剤が存在し、全てではないがその一部として
「ツイーン」(Tween:商標)、「スパン」(Span:商
標)が挙げられる。
オール粘度上昇剤も含み、この量は100〜600の範囲のCP
をこれに授けるのに十分である。この粘度上昇剤は好ま
しくは一分子当り少なくとも4個のヒドロキシ基を有
し、且つ前記の濃度の値を満足せしめる水溶解度を有す
るポリオールを含む。低濃度で得られる泡は一般に薄す
ぎてしまい、しかしながら高濃度では粘度は有効な均一
化及び気泡サイズ縮小のために高すぎてしまう。好まし
い粘度上昇剤はデキストラン、ポリデキストロース、グ
ルコース、フルクトース、マルトース、コーンシロッ
プ、糖分シロップ(モノマー及びオリゴマー糖の混合
物)、部分加水分解炭水化物(例えばデンプン)、グル
コース及びフルクトースのような糖の合成ポリマー;ソ
ルヒドール、マンニトール等のような還元糖、少なくと
も4個のグリセロール単位を有するポリグリセロール、
並びにその他の水に十分に可溶性の類似のポリオールを
含む。スクロースはその不十分な粘度の理由により単独
では利用できないことが分る。しかしながらこれはその
他のより可溶性な糖、例えばグルコース又はフルクトー
スと組合せて利用できる。発泡すべき溶液の粘性パラメ
ーターが単独でその気泡に保存安定性を授けるには不十
分の場合、有効な粘度及び増強剤の濃度の好ましい組合
せは低Mw(〜180)と平均的な分子量(〜300から2000)
の炭水化物の混合物を利用することにより得られること
が分る。全ての範囲のデンプン加水分解物もこの範疇に
入る。多糖類の中で本発明において好ましく、且つ市販
されているものとして、米国又は英国におけるPfizerの
ポリデキストロースN;Amylum(ベルギー国)からミロー
ス(Mylose:商標)CN、ミロースHM−43−38−250又はRo
quette,Lestrem(フランス国)からフロリス(Flolys:
商標)B−6080Sの名のもとで販売されているデンプン
加水分解;AmylumからのSOS−LUもしくはSIR−O−SWIT
(商標)の名のもとで販売されている高フルクトースコ
ーンシロップが挙げられる。
泡のサイズを約0.5〜10μmの所望のサイズに迄小さく
するのに十分な剪断力を提供する任意の市販されている
ヘッドを有するものでありうる。発泡と気泡サイズ縮小
を同時に行なう、即ち1工程において行なう場合、好ま
しい乳化ヘッドは高速切断、空気又は気体の吸引、及び
気泡の最終サイズをコントロールすることを可能とする
サイズの開口部を通過する液体循環を同時に提供するも
のである。食品工業において有用な多数の乳化機(例え
ばPOLYTRON)が適切である。軸流カッター(axial cut
ter)及び約1.5mmの幅の外周上の縦(peripheral longi
tudinal)スリットを有す管状乳化ヘッドが優れた結果
を提供する。
ストロース(PFIZER)を含み、20℃で約400cPの粘度を
有し、約3osmolの浸透圧を有する0.2〜1%のHSA溶液
を、POLYTRON20TS乳化ヘッドにより、10〜15000rpmにて
0.2〜5分間にわたり発泡且つホモジナイズせしめる。
これにより、週数間にわたり完全に安定(有意なる測定
変化が認められない)である、1ml当り約108〜109個の
微小気泡(0.5〜5μm)の気泡又は泡の分散体が得ら
れる。この分散体はエコグラフィー検査のために直接注
射せしめるには粘性が高すぎ、従ってある時間(例えば
10〜20時間)にわたり室温で任意的に放置せしめた後、
これを0.01〜0.1%のグルタルアルデヒド溶液によって
1:10〜1:15に希釈せしめる。希釈後、この濃度は未だ10
7〜108個の範囲であるが、その粘度は数cPのみとなり、
従ってこの溶液は注射及びエコグラフィー検査のために
適する。この希釈溶液も少なくとも数週間は安定であっ
た。
ド(又はその他の任意の薬学的に受け入れられている架
橋剤)を利用することもできる。その他の架橋剤には、
システイン、アセチルシステイン、メルカプトエタノー
ル、エタンスルフィドのような硫化物;カルボジイミ
ド、ポリイソシアネート;多酸;及び無水マレイン酸の
ような無水物;並びに低級脂肪酸とのスクロースもしく
はグルコースのエステルのような炭水化物ポリエステル
又は脂肪酸が含まれる。安定化は、希釈のために1〜10
%のBSA又はHSAの溶液を用いることによっても成し遂げ
られうる。通常、所望するならば、安定化は約60〜120
゜、好ましくは約65〜80゜の温度範囲において数秒から
数分間にわたり注意深く加熱せしめることによっても行
なわれることができ、その正確な温度は所望するタンパ
ク質の硬化の粘度に依存する。
バーの外周スリットに発泡すべき溶液(又は縮小すべき
気泡)を押し通し、そして同時に溶液の中心に渦を形成
せしめることによってその周辺から空気(又は気体)を
取り込む。これによって強力な液/空気混合作用及び泡
の気泡のサイズを小さくせしめる強い剪断効果が提供さ
れる。通常空気は任意の生理学的に受け入れられている
気体、例えばN2、Ar及びその他の貴ガス、N2O、炭化水
素蒸気、CH4等に代替されうる。
記載の通りに小さくする場合、この発泡工程は任意の常
用のブレンダー又は泡立て装置により行なわれる。例え
ばある装置態様は、底部排出部を有すパイレックス反応
槽であって、温度コントロールのための冷却マントル、
発泡する前及びしている際中にて組成物を攪拌するため
のブレード攪拌器、並びに剪断せしめ且つ発泡、ホモジ
ネーション及び気泡サイズの縮小を行なうためのカッタ
ークラッシャーヘッドの装備しているものを含んで成
る。配合物の無菌化はこの反応槽にて直接行うことがで
きる。通常、上記の反応槽による操作は、以下の段階を
好適に含む。

とが好ましく、もし温度が変性の起こるレベル迄高まる
と、収率は非常に悪くなりうる。包括されるホモジネー
ション時間は数分間、即ち5〜10分間であることがで
き、これは利用する材料の量に依存する。剪断エネルギ
ーに基づく高すぎる温度は発泡又は気泡サイズ縮小の際
に有害でありうるにもかかわらず、所望するならば微小
気泡の完了懸濁物を有害な作用を伴わずに50℃より高く
加熱せしめることがあることにも注目すべきである。実
際、希釈する前後の完了気泡懸濁物の加熱は気泡壁を成
す膜形成タンパク質の発展的硬化をもたらし、このこと
は気泡特性を多少変化せしめ、従って保存期間のもとで
の安定性に役立つ。事実、例えば60〜120℃で数秒間か
ら数分間加熱することは、変性によるタンパク質の有用
な特性範囲への硬化をもたらすであろう。
技術によって乾燥したもろい固形物へと乾燥されること
ができる。この固形物は変化を伴うことなく乾燥条件の
もとで無限の期間保存されることができ、そしてもとの
注射可能な懸濁物は単にこれに対応する量の水又は注射

ることによって安全に再生することができる(気泡の損
失を伴わず)。注射可能な微小球懸濁物を提供するため
に利用する水に関し、泡の安定性の欠如のため、完全に
脱気せしめたタイプのもの(例えば蒸留水)は避けるべ
きであることが理解される。泡の希釈を及ぼす好ましい
種類の水は無菌であり且つ空気にさらされているもので
あるべきである;通常、水道水又は天然のミネラル飲料
水がより適切である。少量の溶解無機物質は(家庭ライ
ンから入手できる水の中に通常存在)、保存のもとでの
この懸濁物における気泡の安定に役立つようである。
施例において、粘度の値はHAAKE回転式粘度計、ヘッドM
E−30(回転速度に従って1〜1000cP)により25℃にて
測定し;気泡濃度は格子付顕微鏡スライドを含んでなる
ヘマトサイトメーターにより測定した(測定面積は1×
1mm、厚さ100μm(0.1μl))。計測すべき溶液はこ
の計測面積中に約50〜100個の気泡となるよう予め適当
に希釈した。他方、常用のカルターカウンター(Coulte
r Counter)も用いた。

塩水による希釈(1:13)の実施は約7.7×104個の気泡/
立方mmを有する容易に注射できる液体を提供する(粘
度、約5〜10cP)。

の溶液窒素の雰囲気下でPOLYTRON乳化ヘッドにより3分
間発泡且つ乳化せしめ、その温度は硬化、及び変性によ
るタンパク質の水への不溶性化を防ぐために40℃以下に
保った。
アルデヒドを含む水により約1/10に希釈し、そして気泡
濃度について計測した(約5×107個/ml、の希釈懸濁物
であった)。この気泡サイズ分布は、0.5〜3μmの範
囲において80%であった(粘度は数cPであった)。この
濃い及び希釈気泡懸濁物の両者は、有意な気泡計測値変
化を伴わずに少なくとも4週間安定であった。
水性HSA及び粘性上昇成分として42重量%のデキストロ
ースを用いて繰り返した(粘度は25℃にて518cP)。発
泡及びホモジネーションは35℃以下の温度にて1分間行
った。この微小気泡懸濁物(約109/ml)は少なくとも15
日間安定であった。5%のHSA溶液による希釈の後(1/1
0)、その粘度は約15〜20cP迄落ち、そしてこれは注射
及びエコグラフィー検査に適していた。時間に伴う安定
性は70〜75℃で2分間加熱することにより更に高まっ
た。
代用することで繰り返した。その他のパラメーターは実
施例1と同様とした。以下の表に、未希釈懸濁物の微小
気泡濃度の結果を報告する。食塩水(0.9%のNacl水溶
液)又はその他の薬学的に受け入れられる水性希釈剤の
いづれか、及び安定剤による希釈の後、反響検査に適す
る微小気泡の注射可能な懸濁物が得られた。
%水性ヒト血清アルブミン(HSA)36gを順に加えた。攪
拌機を作動させ(25rpm)そして温度を20℃にコントロ
ールした。次にポリトロン(商標)ヘッド(スリット3m
m)を11,000rpmにて作動させ、その間ゆっくりとした攪
拌をし続け、そしてこの反応を25分間行い、その間この
温度は40℃を超えないようにコントロールした。次に50
%の水性グルタルアルデヒド1.29mlを加え、そしてホモ
ジネーションを数分間続け、ここでこの温度は45℃以下
に保った。このカッターヘッドを停止させ、そしてこの
混合物を室温で約10分間冷却した。この気泡懸濁物をこ
の反応槽において約12時間放置し、その後これを底から
回収せしめ(反応槽上部の泡は除去した)、そしてガラ
ス容器に保存した。この気泡濃度は108〜109/mlの範囲
にあり、そして90%以上の気泡は1〜10μmのサイズの
範囲にあった。
希釈し、そして冷い囲いの中で−20〜−40℃で瞬間凍結
させた。次にこれを室温にゆっくり戻しながら減圧した
(10-3〜10-4mm)(真空フラスコとポンプの間に位置す
るコンデンサーは約−40℃に保った)。完全な蒸発の後
に薄く且つもろい固形の塊が残り、これは有意なる変化
を伴わずに乾燥条件(吸湿性)のもとで無期限的に保存
できる。長期間保存後(数週間)、この固形物に12mlの
水を加え、これはもとの気泡計測値(約107/ml)を伴う
気泡懸濁物を再生せしめた。
主たる方法に従って行った。但し、いくつかの操作パラ
メーターは発展的に改良した。操作条件及び結果を添付
した表にまとめた。この表において、頭にある一連の縦
の列は以下のパラメーターについて記した。即ち、サン
プル番号;粘度上昇のための糖溶液の種類;溶解糖の重
量%(乾燥体として計算);処理にかける混合物のpH;
その中に用いているアルブミンの重量%;架橋剤の重量
%を示す。次の4つの縦列については更なるコメントは
必要ないであろう。それに続く残りの縦列(11列目よ
り)は以下のパラメーターに関する:ポリトロンカッタ
ーヘッドにおけるスロットの幅;回収の前にこの調製物
が放置される温度(及び任意的に時間);ヘマトサイト
メーターにより確認される、濃い懸濁物中の気泡の濃
度;希釈懸濁物のサンプルの吸光度(希釈は水における
固形分10%に調製);微小気泡の「容量」平均サイズ;
前記の気泡の「数」平均サイズ(両者ともマスターサイ
ザーにより測定);そして最後に長所の数値化である
「分散性」パラメーターであって、数値が小さいほど明
らかにサイズ分布が狭いパラメーターを示す。
Claims (20)
- 【請求項1】少なくとも1種類の膜形成タンパク質の膜
により覆われている生体における超音波適用のために適
切な空気もしくは気体封入微小球又は微小中空球の安定
なる水性懸濁物を調製するための方法であって、該タン
パク質の粘性水溶液を空気又は薬学的に受け入れられる
気体と攪拌せしめることにより空気又は気体が封入され
ている気泡の泡へと変換せしめ、そして同時に又はその
後、該泡における該気泡のサイズを注射に適切な0.5〜1
0μmの範囲の値へと小さくするために該発泡溶液を十
分な時間をかけて、バッフル手段を任意的に含む狭い開
口部又は通路に押し通すことによって強い剪断にかける
ことを特徴とする方法。 - 【請求項2】外周上に平行に軸方向に並んでいるスリッ
ト及び中央部に軸回転するカッターを有する管状ヘッド
を含んで成るホモジナイザー及び乳化装置において、前
記の泡を強い剪断にかける前記方法であって、この作用
は、周辺の空気がこの液体と混合されること及びこの空
気−液体混合物が該スリットに軸方向に押し通されるこ
とを引き起こし、これによって該泡は強い剪断作用にか
けられるものである、請求項1に記載の方法。 - 【請求項3】前記ホモジナイザー及び乳化装置により発
生する熱によるタンパク質変性を回避するため、前記の
発泡及び気泡サイズ縮小の際の温度を50℃以下に保ち続
ける、請求項2に記載の方法。 - 【請求項4】得られる前記懸濁物が108〜109個/mlの範
囲における微小気泡の濃度を有す、請求項1に記載の方
法。 - 【請求項5】前記得られる懸濁を、生体の血流に直接注
射することを可能とするようなわずかなcPへとその粘度
を下げるために水又は生理学的に受け入れられる液体に
より更に希釈せしめる、請求項1に記載の方法。 - 【請求項6】前記希釈溶液を、乾燥状態において無期限
にわたり変化することがなく且つ対応する量の水又は注
射のために適切な生理学的に受け入れられている液体と
混合せしめることにより微小中空球の懸濁物を再生せし
める固形物へと凍結乾燥せしめる、請求項5に記載の方
法。 - 【請求項7】前記希釈液が前記の希釈懸濁物の保存安定
性を高める物質を含んで成る請求項5に記載の方法。 - 【請求項8】前記希釈液が有機アルデヒドである、請求
項7記載の方法。 - 【請求項9】変性による膜タンパク質の硬化を及ぼすた
めに前記希釈溶液を数秒から数分間60〜120℃の温度に
加熱することを含んで成る請求項5に記載の方法。 - 【請求項10】前記泡に摩擦作用を提供する媒体、即
ち、多数のバッフル化通路を有する無機又は有機材料に
該泡を押し通すことにより、該泡を強い剪断にかける請
求項1に記載の方法。 - 【請求項11】タンパク質で覆われている前記の気泡の
特性を漸進的な変性により改善せしめるため、得られる
前記懸濁物を数秒間から数分間60〜120℃で更に加熱す
る請求項4に記載の方法。 - 【請求項12】請求項1に記載の方法により超音波エコ
グラフィーに適切な微小気泡の懸濁物へと変換される超
音波エコグラフィーのための水性組成物であって、溶液
中に膜形成タンパク質0.1〜5重量%及び周囲温度粘度
を100〜600cPに迄高める水溶性ポリオール粘度上昇剤40
〜80重量%、並びに任意的に発泡増強剤としての界面活
性剤及び該泡の安定性を高めるための架橋剤を含んで成
ることを特徴とする組成物。 - 【請求項13】前記のポリオール粘度上昇剤が1分子当
りに少なくとも4個のOH基を有する、請求項12に記載の
組成物。 - 【請求項14】前記の粘度上昇剤がポリアルコール、モ
ノー、ポリー及びオリゴサッカライド並びにその混合物
から選ばれる、請求項13に記載の組成物。 - 【請求項15】前記の粘度上昇剤が、グルコース、フル
クトース、マンノース、デキストロース、デキストラ
ン、ポリデキストロース、糖分シロップ、コーンシロッ
プ、少なくとも4個のグリセロール単位を有するポリグ
リセロール及びその混合物より選ばれる、請求項14に記
載の組成物。 - 【請求項16】安定剤が加えられている、請求項12に記
載の組成物。 - 【請求項17】前記安定剤がホルムアルデヒド又はグル
タルアルデヒドである、請求項16記載の組成物。 - 【請求項18】少なくとも4週間にわたり安定であるこ
とを特徴とする、請求項1に記載の方法を実施すること
により得られる超音波エコグラフィーのための微小気泡
の懸濁物。 - 【請求項19】その安定性が膜形成タンパク質の硬化を
及ぼすために数秒から数分間60〜120℃に更に加熱する
ことによって高められている請求項18に記載の懸濁物。 - 【請求項20】ヒトを除く生体のエコグラフィー検査の
ための請求項18に記載の懸濁物の利用方法であって、該
懸濁物を生理学的に受け入れられる液体により希釈せし
めてその粘度を2〜30cPに迄下げ、そしてこの状態にお
いてこれを検査すべき該生体の血流に注射せしめ、これ
により該生体において反響的応答が発生できるようにな
る方法。
