Abstract
【解決手段】蛋白質1グラム当たり15,000μg未満の免疫学的に活性なエンドウ豆抗原を有するエンドウ豆蛋白加水分解物を含む、栄養製品。該エンドウ豆蛋白加水分解物が、蛋白質1グラム当たり10,000μg未満の免疫学的に活性なエンドウ豆抗原を有することが好ましい栄養製品。
【効果】前記栄養製品は牛乳蛋白質と大豆蛋白質に対して不耐症及び/又はアレルギーの個体、並びに/或いは吸収不良、消化不良若しくは他の胃腸障害をもつ個体の栄養源として使用することができる。

Description
エンドウ豆蛋白加水分解物を含有する本発明の栄養組成物は任意の公知の経口製品形態又は適切な経口製品形態で製剤化及び投与することができる。同様に本願に記載する必須成分を個体に安全且つ有効に経口送達できるものであれば、任意の固形、液体又は粉末形態とその組合せ又は変形が本願で使用するのに適している。
液体飲料としては、濃縮液とそのまま摂取可能な液剤が挙げられる。本願で使用するのに適した液体製品形態の非限定的な例としては、乳児用液体調合乳(そのまま摂取可能な製剤と濃縮製剤の両方を含む)、幼児用液体調合乳(そのまま摂取可能な製剤と濃縮製剤の両方を含む)、液体母乳強化剤(そのまま摂取可能な製剤と濃縮製剤の両方を含む)、スナック及び代用食製品、温飲料又は清涼飲料、炭酸飲料又は非炭酸飲料、ジュース又は他の酸性飲料、シェーク、コーヒー、紅茶、経腸栄養組成物等が挙げられる。これらの液体組成物は最も典型的には懸濁液又は乳剤として製剤化されるが、透明溶液、液体ジェル等の任意の他の適切な形態で製剤化することもできる。
栄養乳剤はエンドウ豆蛋白加水分解物を主要又は単独蛋白質源として含有しており、更に脂質と糖質を含有する水性乳剤である。これらの乳剤は約1℃〜約25℃で流動性又は飲用可能であり、典型的には水中油型、油中水型又は複合型水性乳剤の形態であるが、このような乳剤は最も典型的には連続水相と不連続油相をもつ水中油型乳剤の形態である。
本発明の実質的に透明な栄養液剤は下記のように少なくとも蛋白質と糖質を含有する低粘性液剤であり、前記蛋白質の少なくとも一部はエンドウ豆蛋白加水分解物である。前記実質的に透明な栄養液剤は実質的に無脂肪であり、即ち前記液剤は原料に内在する脂質又は液剤の製造を助長するために低濃度で添加された脂質以外に脂質を添加されていない。この文脈で、「無脂肪」なる用語は前記液剤が典型的には栄養液剤重量当たり1.0%未満、より典型的には0.5%未満、より典型的には0.1%未満しか脂質を含有せず、0%でもよいことを意味する。これらの実質的に透明な栄養液剤は約1℃〜約25℃で流動性又は飲用可能である。
前記栄養粉末はエンドウ豆蛋白加水分解物を主要又は単独蛋白質源として含有しており、典型的には流動性もしくは実質的に流動性の粒状組成物、又は少なくとも粒状組成物の形態である。特に適切な製品形態としては、噴霧乾燥、凝集又はドライブレンド粉末組成物が挙げられる。前記組成物はスプーン又は他の同等器具で容易にすくって測定することができ、前記組成物は対象使用者が適切な水性液体、典型的には水で容易に再構成し、即時経口又は経腸用栄養製剤を形成することができる。この文脈で「即時」用とは典型的に再構成後約48時間以内、最も典型的には約24時間以内、好ましくは直後を意味する。
前記栄養製品は固形栄養バー及び食品の形態とすることもできる。例えば、前記栄養製品はバー、スティック又はクッキー、パン、ケーキもしくは他の焼き菓子製品として製剤化することができる。
前記栄養製品が乳児用調合乳であるとき、前記栄養製品は上記のような液体又は固形栄養製品とすることができる。本発明の液体又は固形乳児用調合乳製品は対象乳児に適した任意のカロリー密度としてもよいし、粉末態様の再構成後又は濃縮液態様の希釈後にこのような密度を提供するようにしてもよい。例えば、本発明の乳児用調合乳態様に最も一般的なカロリー密度は一般に少なくとも約19kcal/fl oz(660kcal/リットル)、より典型的には約20kcal/fl oz(675〜680kcal/リットル)〜約25kcal/fl oz(820kcal/リットル)、更に典型的には約20kcal/fl oz(675〜680kcal/リットル)〜約24kcal/fl oz(800〜810kcal/リットル)である。一般に、早産児又は出生時低体重児では22〜24kcal/fl oz製剤がより一般に使用され、満期産児では20〜21kcal/fl oz(675〜680から700kcal/リットル)製剤を使用することが多い。
前記栄養製品はエンドウ豆蛋白加水分解物を主要又は単独蛋白質源として含有する。低アレルギー性組成物が所望される栄養製品態様(例えば低アレルギー性乳児用調合乳)において、栄養組成物は蛋白質成分重量当たり100%のエンドウ豆蛋白加水分解物を含有する。本発明の製品で使用されるエンドウ豆蛋白加水分解物は大豆蛋白質等の他の植物性蛋白加水分解物や牛乳蛋白質に比較して免疫反応性が低く、風味と官能特性が改善されていることが判明した。例えば、図1に示すように、本発明の栄養製品で使用されるエンドウ豆蛋白加水分解物はフィトエストロゲン(即ちイソフラボン類であるゲニステインとダイゼイン)、アルミニウム、マンガン及びフィチン酸塩の濃度が低いため、最終栄養製品の栄養素プロファイルが改善される。更に、本願に記載するように、前記エンドウ豆蛋白加水分解物は抗原含量が低く、及び/又は免疫反応性が低い。従って、前記エンドウ豆蛋白加水分解物は牛乳蛋白質及び/又は大豆蛋白質に対して不耐症及び/又はアレルギーの個体を対象とすると共に、吸収不良、消化不良及び他の胃腸障害をもつ個体の栄養の必要を満たすように意図された栄養製品の良好な蛋白質源を提供する。これらの栄養製品は更に風味と乳剤安定性が改善されている。
本発明の栄養製品は場合によりエンドウ豆蛋白加水分解物以外に更に他の1種以上の蛋白質源を含有することができる。経口栄養製品用として適切であると共にこのような製品の必須成分及び特徴に適合可能な任意の他の蛋白質源がエンドウ豆蛋白加水分解物と併用するのに適切である。
前記栄養製品は更に、経口栄養製品用として適切であると共にこのような製品の必須成分及び特徴に適合可能な任意糖質を含有することができる。栄養液体飲料中の糖質濃度は例えば栄養液体飲料重量当たり約5%〜約40%、例えば約7%〜約30%、例えば約10%〜約25%の範囲とすることができる。1特定態様において、糖質は栄養乳剤中に栄養乳剤の約10.2重量%の量で存在する。
前記栄養製品は更に、最も典型的には乳化脂質としての脂質を含有することができる。脂質は栄養液体飲料中に栄養乳剤重量当たり0%〜約30%、例えば1%〜約30%、例えば約1%〜約20%、例えば約1%〜約15%、例えば約1.5%〜約5%の量で存在することができる。1特定態様において、栄養乳剤は栄養乳剤の約1.6重量%の量の脂質を含有する。
本発明のエンドウ豆蛋白加水分解物を含有する乳児用調合乳を含む栄養製品は更に、製品の物理的、化学的、審美的又は加工特性を改変することや、対象集団で使用時に医薬成分又は付加栄養成分として機能することが可能な他の非必須成分を含有することができる。多数のこのような非必須成分が公知であり、あるいは公知ではないが、医療食品もしくは他の栄養製品又は医薬製剤で使用するのに適しており、経口投与に安全であると共に選択された製品形態の必須成分及び他の成分と適合可能である限り、このような非必須成分も本願に記載する組成物で使用することができる。
本発明の栄養製品は選択された固形又は液体製品形態を製造するための製造技術として公知であるか又は公知でないとしても有効である任意技術により製造することができる。栄養液剤又は粉末又は栄養バー等の任意の所定製品形態用として多数のこのような技術が公知であり、当業者は本願に記載の栄養組成物に容易に適用することができる。
エンドウ豆蛋白加水分解物を単独蛋白質源として含有する前記栄養製品は牛乳蛋白質、大豆蛋白質に対して不耐症及び/又はアレルギーであるか、及び/又は他の食物全般アレルギーをもつ早産児、乳児、幼児、小児、成人及び高齢者用の低アレルギー性栄養源及び/又は消化態栄養源として有用である。更に、前記栄養製品は吸収不良、消化不良及び他の胃腸障害をもつ個体による使用に特に適切であると思われる。
本実施例では、本願に記載の方法に従って製造した濾過(F)及び非濾過(N)エンドウ豆蛋白加水分解物のサンプル10個を無傷のエンドウ豆蛋白質分離物(ISO)サンプルと各種物理的特性について比較評価する。濾過(F)はサンプルを上記第2の濾過工程に供することを示す。全サンプルは上記第1の濾過工程に供する。対照サンプル(無傷)は加水分解工程に供しない。具体的には、標準分析技術を使用して加水分解度、分子量分布、イソフラボン含量及びフィチン酸塩含量についてサンプルを評価する。評価の結果を下表に示す。
本実施例では、ウサギの過剰免疫を使用し、ウサギに以下のように35日間複数回免疫し、実施例1で分析したエンドウ豆蛋白加水分解物の低アレルギー性を実施例1の無傷のエンドウ豆蛋白質分離物(ISO)サンプルと比較評価する。次にエンドウ豆蛋白加水分解物の免疫原性をELISA(酵素免疫定量法)により試験し、無傷のエンドウ豆蛋白質/エンドウ豆蛋白加水分解物免疫原に対するウサギの抗原特異的IgG応答を測定する。
本実施例では、阻害ELISA法により測定した抗原含量を使用し、実施例1で分析したエンドウ豆蛋白加水分解物の低アレルギー性を実施例1の無傷のエンドウ豆蛋白質分離物(ISO)サンプルと比較評価する。このアッセイでは、無傷のエンドウ豆蛋白質をELISAプレートにコーティングする。別に、サンプル又は標準濃度のエンドウ豆蛋白質溶液+抗エンドウ豆蛋白質抗体を入れたインキュベーションチューブを準備する。インキュベーション時間後に、コーティングしたプレートにサンプル(又は標準)+抗体溶液のアリコートを加える。インキュベーションと洗浄後に抗ウサギIgG酵素共役抗体を加える。プレートをインキュベーションし、洗浄し、酵素基質を加える。サンプル中のエンドウ豆抗原の濃度と反比例する発色反応が生じ、標準曲線に基づいて定量する。サンプル10個のこの試験の結果を下表に示す。
本実施例では、2期間のベンチスクリーニング期間で感覚評価を行い、水中とAlimentum(登録商標)マトリックス(Abbott Laboratories、Columbus、Ohio)中の両者でレトルト殺菌した実施例1のエンドウ豆蛋白加水分解物サンプルの味を判定する。Alimentum(登録商標)マトリックスは低アレルギー性調合乳の生化学をシミュレーションするように糖質、脂質、ビタミン及びミネラルを含有する。これを使用し、レトルト殺菌処理後に前記製品ナトリックス内で加水分解物及び最終調合物に及ぼす相互官能的効果を評価する。サンプルをAlimentum(登録商標)で使用されている現在の加水分解物(カゼイン加水分解物)及び実施例1の無傷のエンドウ豆蛋白質分離物(ISO)と比較する。特に、加水分解蛋白質は一般に苦味、酸味、塩味及び/又は豆の味がすることが知られている。従って、製品の味を著しく損なわずにこの機能成分を栄養製剤に配合することは困難である。
実施例5〜34は少なくとも1種の蛋白質源、場合によっては単独蛋白質源としてエンドウ豆蛋白加水分解物を含有しており、下表に示す原料からなる本発明の各種乳児用調合乳栄養粉末を例証する。前記エンドウ豆蛋白加水分解物は約7.0%から19.0%未満までの加水分解度(調整後)である。これらの製品は別々のバッチで各種方法により製造し、使用前に所望の目標原料濃度まで水で再構成する。エンドウ豆蛋白加水分解物を単独蛋白質源として含有する下記態様は低アレルギー性態様である。
実施例35〜54は少なくとも1種の蛋白質源、場合によっては単独蛋白質源としてエンドウ豆蛋白加水分解物を含有しており、下表に示す原料からなる本発明の栄養乳剤態様を例証する。前記栄養乳剤を製造するためには、少なくとも3種類の別個のスラリー(例えばCHO−MINスラリー、PIWスラリー、PIFスラリー)を調製後、相互にブレンドし、熱処理し、ホモジナイズし、標準化する。得られた組成物をプラスチックボトルに無菌充填包装又はレトルト殺菌する。エンドウ豆蛋白加水分解物を単独蛋白質源として含有する下記態様は低アレルギー性態様である。
実施例55〜64は下表に示す原料からなる本発明の栄養乳剤を例証する。前記栄養乳剤を製造するためには、少なくとも3種類の別個のスラリー(例えばCHO−MINスラリー、PIWスラリー、PIFスラリー)を調製後、相互にブレンドし、熱処理し、ホモジナイズし、標準化する。得られた組成物をプラスチックボトルに無菌充填包装又はレトルト殺菌する。エンドウ豆蛋白加水分解物を単独蛋白質源として含有する下記態様は低アレルギー性態様である。
実施例65〜74は下表に示す原料からなる本発明の実質的に透明な栄養液剤を例証する。前記栄養液剤を製造するためには、少なくとも2種類の別個のスラリー(例えばCHO−MINスラリーとPIWスラリー)を調製後、相互にブレンドし、熱処理し、ホモジナイズし、標準化する。得られた組成物をプラスチックボトルに無菌充填包装又はレトルト殺菌する。エンドウ豆蛋白加水分解物を単独蛋白質源として含有する下記態様は低アレルギー性態様である。
実施例75〜84は下表に示す原料からなる本発明の濃縮液母乳強化剤を例証する。前記濃縮液母乳強化剤を製造するためには、少なくとも3種類の別個のスラリー(例えばCHO−MINスラリーとPIWスラリー)を調製後、相互にブレンドする。エンドウ豆蛋白加水分解物を単独蛋白質源として含有する下記態様は低アレルギー性態様である。
以下の実施例は本発明の方法に従って母乳に添加して乳児に投与することができる母乳強化剤を例証する。粉末母乳強化剤8,172kgを製造するための原料を下表に示す。
これらの実施例では、本発明に従ってゲル化母乳強化剤を製造する。前記ゲル化母乳強化剤の原料を下表に示す。
実施例105〜114は下表に示す原料からなる本発明の栄養粉末を例証する。これらの製品は別々のバッチで噴霧乾燥法により製造し、使用前に所望の目標原料濃度まで水で再構成する。特に指定しない限り、全原料量は製品バッチ1000kg当たりのkgとして示す。

- 蛋白質1グラム当たり15,000μg未満の免疫学的に活性なエンドウ豆抗原を有するエンドウ豆蛋白加水分解物を含む、栄養製品。
- エンドウ豆蛋白加水分解物が、蛋白質1グラム当たり10,000μg未満の免疫学的に活性なエンドウ豆抗原を有する、請求項1に記載の栄養製品。
- エンドウ豆蛋白加水分解物が、約7.0%から19.0%未満の調整後加水分解度を更に有する、請求項1に記載の栄養製品。
- 早産児用調合乳、母乳強化剤及び乳児用調合乳から構成される群から選択される低アレルギー性製品である、請求項1に記載の栄養製品。
