Abstract
Description
1)ビフィドバクテリウムブレーベ
ビフィドバクテリウムブレーベは、本発明の本質的な要素である。この細菌は、人工栄養保育された乳児の場合に限られた量で存在することが、本出願人の検出方法によって見出された。したがって、この細菌を炭水化物混合物と共に投与することにより、母乳で保育された乳児の胃腸管に存在する場合と同等のレベルまで、ビフィドバクテリウム種集団を正常化することが可能になる。
難消化性炭水化物プレバイオティックスの混合物も、本発明の本質的な要素である。「難消化性」とは、炭水化物が、胃腸管内消化されないままであり、吸収されずに大腸に到達することを意味する。
(i)炭水化物の単糖単位の(平均)数であり、成分Aは、その平均鎖長が、成分Bの鎖長よりも少なくとも5単糖単位だけ少ない;これは、A及びBの炭水化物が同じ構造単位を有する場合、即ち鎖長のみ異なる同族体の混合物を形成する場合、この同族体の分布は2つの最大値を持たねばならず、1つの最大値は7よりも下であり、且つ1つは7よりも上であり、これら2つの最大値は、少なくとも5単位離れていることを意味し;6単位までの炭水化物(六糖)は成分Aの一部であり、7単位から先の炭水化物(七糖)は成分Bの一部である。
(ii)炭水化物の単糖単位の構造であり、成分Aは、成分Bとは異なる構造単位から構成され;A及び/又はBが、異なる単糖単位を繰り返した組合せから構成される場合、例えばガラクトマンナン及びアラビノガラクタンの場合、2成分の単糖単位の少なくとも50%は、異なるべきである(上記の例では、どちらか又は両方が、50%未満の無水ガラクトースを有するべきである)。
(iii)両方、即ち成分A及びBは、その(平均)鎖長と構造に差があり、この実施形態が好ましい。
炭水化物成分A及びBの他に、その他の炭水化物も同様に存在してよい。その中には、1)上述のように消化性のある消化性炭水化物、及び2)吸収性/消化性があり、又は吸収性/消化性も無い不溶性炭水化物がある。乳児栄養サプリメントに使用される、典型的な不溶性の難消化性炭水化物は、大豆多糖であり、抵抗性デンプン、セルロース、及びヘミセルロースであり、より好ましくは、この炭水化物は大豆多糖及び抵抗性デンプンから選択される。
本発明による製剤は、人工栄養保育され又は一部母乳で保育される乳児、特に未熟児、成熟児として生まれた赤ちゃん、並びに固形食物への適応期間にある乳児の胃腸管において、「標準」と見なされる母乳で保育される乳児での種の分布に従って、ビフィズス菌集団を正常化するのに特に有用であることがわかった。本発明の製剤は、母乳での保育から哺乳瓶での保育に変わる乳児にも適している。
本明細書では、種特異的オリゴヌクレオチドプライム及びプローブを使用して、ビフィドバクテリウム種、特にヒトに見られるもの、即ちビフィドバクテリウムカテニュレイタム及びB.シュードカテニュレイタム、B.アドレセンティス、B.ブレーベ、B.ロンガム、B.ビフィダム、B.アンギュレイタム(B.angulatum)、B.インファンティス、及びB.デンティウムを定量する方法も提供する。

ビフィズス菌用に開発されたプローブ及びプライマーの確認
種々のビフィドバクテリウム種の相対量に関するアッセイを、確認するのに使用される細菌株を、表2に列挙する。
b再現性は、個体培養物(100%)10個について試験を行い、得られた結果に基づいてCV(%)を計算することによって決定する。
c反復性は、個体培養物(100%)4個についてそれぞれ3回試験を行い、得られた結果に基づいてCV(%)を計算することによって決定する。
臨床試験
この研究は、2つの介入群に関する二重盲検プラセボ対照多施設治験であった。生後28日から90日の、完全に調合乳で保育した乳児を、ドイツの4カ所の病院から採用した。乳児を、その出生時体重が2600gから4500gの間にある場合には研究に組み入れ、少なくとも4週間完全に調合乳で保育し、その後、介入期間を開始した。先天性異常のある乳児、或いは牛乳アレルギーが確認され又は疑われる乳児、多子出産によって生まれた乳児、この研究の開始前に抗生物質を2週間未満与えた乳児、及びこの研究の開始前に任意のプロバイオティック又はプレバイオティック調合乳で1カ月未満保育された乳児は、この研究から除外した。登録後、乳児を無作為に、2つの治療群の一方に、即ち0.8g/100mlのガラクト−オリゴ糖及びフルクト−多糖が補われた乳児用調合乳を与えた群(GFSF群)と、標準的な乳児用調合乳を与えた群(SF群)との一方に割り当てた。この調合乳の多量養素組成を、表5に示す。
アレルギーに関する動物実験
特定の病原体を持たないオスBALB/cマウスを、Charles River(Maastricht、オランダ)から得た。食品及び水を随意に与え、そのマウスが6〜9週齢になったときに使用した。全ての実験は、ユトレヒト大学(オランダ)の動物倫理委員会により承認済みであった。

Claims (25)
- ビフィドバクテリウムブレーベ(Bifidobacterium breve)と、少なくとも2種の難消化性可溶性炭水化物成分A及びBの混合物とを含み、前記炭水化物成分Aは、炭水化物成分A及びBの合計の5〜95重量%の量で存在し、前記難消化性可溶性炭水化物全体の少なくとも50%は、二糖からエイコサ糖までの中から選択され、成分A及びBは、
(i)炭水化物の単糖単位の(平均)数が異なっており、成分Aの平均鎖長が成分Bの平均鎖長よりも少なくとも5単糖単位少ないものであり、又は
(ii)炭水化物の単糖単位の構造が異なっており、又は
(iii)上記(i)と(ii)の両方である製剤。 - 前記炭水化物成分Aが、前記炭水化物成分Bとは異なる構造を有する、請求項1に記載の製剤。
- 前記炭水化物成分A及びBの、単糖単位の(平均)数が異なっており、成分Aは、同じ炭水化物構造の不消化性単糖から六糖までの中から選択され、成分Bは、同じ炭水化物構造の不消化性七糖及びそれ以上の多糖から選択される、請求項1又は2に記載の製剤。
