Abstract
Description
本発明は、様々な癌型、特に卵巣癌において見いだされる腫瘍を標的とする抗体の使用に関する。抗体を腫瘍に向かわせるために、癌細胞表面で発現される腫瘍特異性抗原の同定を実施しなければならない。腫瘍特異性抗原を同定するために利用可能な技術がいくつかあり、卵巣腫瘍におけるKAAG1を同定するために使用された方法(サブトラクティブトランスクリプションに基づくmRNAの増幅(STAR)と呼ばれる革新的な発見の基盤)は、公開特許出願第PCT/CA2007/001134号に記載されている。
本出願者は、KAAG1がいくつかの腫瘍型において発現され、患者の血液及び腹水においても見いだされるという予想外の発見に至った。この抗原は従って、インビボで当該抗原を発現する腫瘍細胞を標的とするため、及びインビトロ又はインビボで腫瘍関連抗原を測定するための検出分析の開発に有用であり得る。血液中を循環するKAAG1抗原はシグナルペプチドがない。
抗体は、最初は興味のある抗原に対するそれらの特異性に関してFabライブラリから単離された。最大の特徴を示す抗体の軽鎖可変領域又は重鎖可変領域のアミノ酸配列の比較によって、CDR内及び可変領域内のコンセンサス配列を誘導することができた。CDRのコンセンサス配列は、配列番号74〜90に示される。

KAAG1に対するIgG1抗体はADCCを媒介することができる。
抗体依存性細胞傷害性(ADCC)は、細胞媒介性免疫機構であり、これによって免疫系のエフェクター細胞、典型的にはナチュラルキラー(NK)細胞は、特異的抗体により結合された標的細胞を積極的に溶解する。NK細胞と抗体との間の相互作用は、抗体の定常Fcドメイン及びNK細胞の表面上の高親和性Fcγ受容体によって起こる。IgG1は、Fc受容体に関して最高の親和性を有し、一方、IgG2mAbは非常に低い親和性を示す。この理由のために、KAAG1を標的とするキメラ抗体はIgG1として設計した。このように媒介されるこの種類のエフェクター機能によって、多くの場合、自身の細胞表面上で高レベルの抗原を発現する癌細胞を選択的に殺すことができるようになる。

KAAG1は皮膚腫瘍及び腎臓細胞癌で発現され、これらの適応症における治療上の標的である。
実施したmRNAプロファイリング研究から、KAAG1抗原をコード化する転写物は、黒色腫試料及び腎臓癌由来の細胞系で高度に発現されたことがわかった。これらの結果は、Sooknananら(2007年)で開示された。これらの癌型におけるKAAG1遺伝子の転写調節を確認するために、免疫組織化学法を、扁平上皮細胞癌及び黒色腫から単離されたいくつかの切片を含むヒト皮膚腫瘍組織マイクロアレイ(Pantomics Inc.、カリフォルニア州リッチモンド)で抗KAAG1抗体を用いて実施した。このアレイの分析は、扁平上皮細胞癌及び黒色腫から単離された生検において非常に強い染色を示した(図14、上のパネル)。これらの型はどちらも皮膚癌の最も一般的な形態に含まれ、また興味深いことに、扁平上皮細胞癌は最も転移性が強く、この事実もKAAG1の発現と侵襲的表現型とを結びつける。すでに観察されたように、アレイ上に含まれる3つの正常な皮膚試料上のKAAG1の存在は非常に微弱であるか又は存在しなかった。同様に、KAAG1は、腎臓癌のアレイに含まれる試料の多くで検出された。陽性試料のほとんどは、主に乳頭細胞癌型のものであり、若干の明細胞癌がKAAG1タンパク質を発現した。乳頭癌は腎臓癌症例の約20%に相当する。
KAAG1は卵巣癌細胞の表面上で発現される。KAAG1をコード化するcDNAの一次構造のバイオインフォーマティック分析、生化学研究、及び上皮細胞中のタンパク質の免疫組織化学的検出から得られた総合結果は、KAAG1抗原が細胞表面上に位置することを示唆した。しかし、KAAG1が実際に膜結合タンパク質であることを証明するためにはさらに直接的な証拠が必要であった。一方法において、KAAG1を発現することが知られている卵巣癌細胞系をマイクロタイタープレート中に播種し、細胞を透過しない条件下で固定し、抗KAAG1キメラ抗体の濃度を増加させつつインキュベートした。細胞をよく洗浄した後、修飾された細胞ベースのELISAにおいて二次抗体としてHRP結合抗ヒトIgGを用いて結合抗体を検出した(図16Aを参照)。これらの実験から得ることができる最初の観察結果は、抗体を細胞によって特異的に捕獲することができることであり、このことは、KAAG1が細胞表面で存在することを示唆した。第2に、結合の量は、SKOV−3細胞上で最強であり、TOV−21G細胞は最弱の結合を示した。これは、KAAG1 mRNAがこれらの細胞系において同様の割合で発現されることを証明したRT−PCRデータ(不図示)と完全に一致した。さらに、3D3抗体は最強のシグナルを生じ、このことは、この抗体によって標的とされるエピトープがこの分析では最もアクセス可能であったことを意味する。3G10は、最高レベルのAB−0447を発現した細胞系においてKAAG1を検出できるだけであった(SKOV−3細胞、図16Aの右パネルを参照)。使用した第2の方法はフローサイトメトリーであった。この場合、マウス3D3抗KAAG1抗体をSKOV−3卵巣癌細胞とともに飽和条件でインキュベートし、続いてよく洗浄し、結合した3D3抗KAAG1抗体を、フローサイトメーターでFITCと結合した抗マウスIgGを用いて検出した。図16Bで示すように、SKOV−3細胞の表面でのシグナルは、バックグラウンドの読みと同じ蛍光レベルである、非哺乳動物非関連性タンパク質について特異的な負の対照である抗KLH(キーホールリンペットヘモシアニン)抗体で標識された同じ細胞と比較してはるかに高かった。まとめると、これらの結果は、KAAG1が細胞表面上に位置することを示す。
ヒト化抗KAAG1抗体の使用法。
インビトロ及び予備インビボ結果の両方に基づいて、2つのマウス抗KAAG1抗体候補(3D3及び3C4と表示)を、当業者に周知の方法を使用するコンピュータ内でのモデリングを用いたヒト化について選択した。手短に言うと、ネズミ抗体の可変領域を、高い配列相同性と類似したCDRループ長さを呈するマウス、ヒト化、及び完全ヒト可変領域の利用可能な結晶構造に基づいて3Dでモデル化した。CDRは、抗原結合に寄与するアミノ酸配列であり、抗体鎖上に3つのCDRが存在する。さらに、CDR間に介在するアミノ酸配列であるフレームワーク領域を、マウスとヒト抗体配列との標準的相同性比較によって修飾し、その結果、「最良適合」ヒト配列を得た。これらの修飾は、潜在的なN及びO結合グリコシル化部位の保存はもちろんのこと、ヒト化構造における最大抗原結合を保証するために、これらの修飾はCDRの適切なポジショニングが確実に維持されるようにした。ヒト化(h)3D3及び3G4における重鎖及び軽鎖可変領域の両方の配列の結果として、それぞれ96%及び94%がヒト化された。各抗体の3D3及び3C4モデルの構造をそれぞれ図17A及び17Bに示す。これらの構造に示されるように、3D3は、3つの異常なアミノ酸(図17A、重鎖上のMet93及びGly94、並びに軽鎖上のSer57)がCDRモデルに近いので、これらのアミノ酸の維持を必要とした。モデリングは、これらのマウスアミノ酸のヒト等価物での置換が抗体のKAAG1抗原との結合を相殺し得ることを予想した。3C4の場合、6個のアミノ酸が異常と見なされた(図17B、重鎖上のGlu1、Gln72及びSer98、並びに軽鎖上のThr46、Phe49及びSer87)。両図において、軽鎖CDRは、CDR1、CDR2、及びCDR3についてそれぞれL1、L2、及びL3により示され、一方、重鎖CDRは、CDR1、CDR2、及びCDR3についてそれぞれH1、H2、及びH3で示される。

- 前記腫瘍細胞が、卵巣癌細胞、皮膚癌細胞、腎臓癌細胞、結腸直腸癌細胞、肉腫細胞、白血病細胞、脳腫瘍細胞、甲状腺腫瘍細胞、乳癌細胞、前立腺癌細胞、食道腫瘍細胞、膀胱腫瘍細胞、肺腫瘍細胞及び頭頸部腫瘍細胞からなる群から選択される、請求項164で定義される使用。
- 前記癌細胞が悪性であることを特徴とする、請求項166で定義される使用。
- 前記癌細胞がアンカレッジ依存性成長を特徴とする、請求項166で定義される使用。
- KAAG1発現細胞を含む癌を治療する方法であって、必要とする哺乳動物に請求項1〜28のいずれか1項に記載の抗体を投与することを含む、方法。
- 前記癌が、卵巣癌、皮膚癌、腎臓癌、結腸直腸癌、肉腫、白血病、脳腫瘍、甲状腺腫瘍、乳癌、前立腺癌、食道腫瘍、膀胱腫瘍、肺腫瘍及び頭頸部腫瘍からなる群から選択される、請求項169記載の方法。
- KAAG1発現細胞を含む腫瘍を検出する方法であって、必要とする哺乳動物に請求項1〜128のいずれか1項に記載の抗体を投与することを含む、方法。
- KAAG1が前記KAAG1発現細胞の表面で発現される、請求項171記載の方法。
- 前記癌が、卵巣癌、皮膚癌、腎臓癌、結腸直腸癌、肉腫、白血病、脳腫瘍、甲状腺腫瘍、乳癌、前立腺癌、食道腫瘍、膀胱腫瘍、肺腫瘍及び頭頸部腫瘍からなる群から選択される、請求項172記載の方法。
- 配列番号2と少なくとも80%の配列同一性を有するKAAG1(配列番号2)又はKAAG1変異体を検出する方法であって、KAAG1若しくはKAAG1変異体を発現する細胞、或いはKAAG1若しくはKAAG1変異体を含むか又は含むことが疑われる試料を請求項1〜128のいずれか1項に記載の抗体と接触させ、結合を測定することを含む、方法。
- 前記試料が哺乳動物由来である、請求項174記載の方法。
- 前記哺乳動物が癌を有するか又は癌を有することが疑われる、請求項175記載の方法。
- 前記試料が、前記哺乳動物から得られる血清試料、血漿試料又は血液試料である、請求項174〜176のいずれか1項に記載の方法。
- 前記試料が前記哺乳動物から得られる組織試料である、請求項174〜176のいずれか1項に記載の方法。
- 前記試料が細胞培養物又は上清である、請求項174記載の方法。
- KAAG1又はKAAG1変異体に結合した抗体の量を定量化することを含む、請求項174〜179のいずれか1項に記載の方法。
- 請求項1〜129のいずか1項に記載の抗体を含むキット。
- 抗体の産生におけるKAAG1の分泌された形態又はKAAG1の細胞外領域の使用。
- KAAG1はシグナルペプチドが欠失している、請求項182で定義される使用。
- KAAG1はアミノ酸1〜30が欠失している、請求項182で定義される使用。
- 配列番号2NO.:NO.:NO.:の少なくとも10個のアミノ酸を含むエピトープの、癌を診断または治療するための抗体を生成させるための使用。
- 前記エピトープが、KAAG1の細胞外領域由来である、請求項185で定義される使用。
- KAAG1の分泌された形態又はKAAG1の細胞外領域に対する抗体を生成させるための組成物であって、配列番号2のアミノ酸30〜84内に含まれるKAAG1のエピトープ及び担体を含む、組成物。
- KAAG1の分泌された形態又はKAAG1の細胞外領域に対する抗体を生成させるための医薬組成物であって、配列番号2のアミノ酸30〜84内に含まれるKAAG1のエピトープ及び薬剤的に許容される担体を含む、医薬組成物。
- KAAG1の分泌された形態又はKAAG1の細胞外領域に対する抗体を生成させるための方法であって、配列番号2のアミノ酸30〜84内に含まれるKAAG1のエピトープを含むポリペプチドを投与することを含み、前記エピトープはKAAG1シグナルペプチドが欠失している、方法。
- 配列番号2のアミノ酸30〜84内に含まれるKAAG1のエピトープの、KAAG1の分泌された形態又はKAAG1の細胞外領域に対する抗体を生成するための使用。
- 腫瘍の拡大、腫瘍浸潤若しくは腫瘍形成を減少させるため又は腫瘍溶解を誘発するための請求項1〜128のいずれか1項に記載の単離された抗体の使用であって、前記腫瘍がKAAG1又はKAAG1変異体を発現する、使用。
- 前記腫瘍が悪性腫瘍である、請求項191記載の使用。
- 前記悪性腫瘍が転移する能力を有する、請求項192記載の使用。
- 前記腫瘍がアンカレッジ依存性成長を特徴とする腫瘍細胞を含む、請求項191記載の使用。
- 腫瘍の拡大、腫瘍浸潤、腫瘍形成を軽減するため又は腫瘍溶解を誘発するための方法であって、請求項1〜128のいずれか1項に記載の単離された抗体を必要とする哺乳動物に投与することを含む方法であって、前記腫瘍がKAAG1又はKAAG1変異体を発現する、方法。
- 必要とする前記哺乳動物が悪性腫瘍を有する、請求項195記載の方法。
- 前記悪性腫瘍が転移する能力を有する、請求項196記載の方法。
- KAAG1を発現する腫瘍細胞の移動又は生存を損なう、配列番号1、配列番号1の10〜884ヌクレオチドのフラグメント及び補体からなる群から選択される核酸の使用。
- KAAG1を発現する前記腫瘍細胞が、アンカレッジ依存性成長を特徴とする、請求項198で定義される使用。
