Claims (9)
- ワクチンの大規模生成のための方法であって、該方法は、
(a)無血清培地、無タンパク質培地、または化学的に規定された培地中で、MDCK懸濁培養物を増殖させる工程であって、ここで
(i)該増殖は、供給バッチ系で行われ;
(ii)培養容積は、新鮮な培地の添加により増加され;そして
(iii)100〜10000Lの生成容量が達成される、工程;
(b)該MDCK懸濁培養物をウイルスに感染させる工程;
(c)該ウイルスを、該MDCK懸濁培養物中で増殖させる工程;および
(d)該ウイルスまたは該ウイルスにより生成されるタンパク質を、該細胞培養物から単離する工程
を包含する方法。 - 前記ウイルスは、β−プロピオラクトンの使用により不活性化されることを特徴とする、請求項1に記載の方法。
- 前記細胞の前記増殖は、感染前に化学的に規定された培地中で、そして感染後に無タンパク質培地中で行われる、請求項1または2に記載の方法。
- 前記MDCK細胞株は、細胞株MDCK33016に由来する、請求項1〜3のいずれか一項に記載の方法。
- 前記ウイルスは、ssDNAウイルス、dsDNAウイルス、RNA(+)ウイルス、RNA(−)ウイルスまたはdsRNAウイルスであることを特徴とする、請求項1〜4のいずれか一項に記載の方法。
- 前記ウイルスは、アデノウイルス、オルトミクソウイルスもしくはパラミクソウイルス、レオウイルス、ピコルナウイルス、エンテロウイルス、フラビウイルス、アレナウイルス、ヘルペスウイルス、またはポックスウイルスから選択されることを特徴とする、請求項1〜5のいずれか一項に記載の方法。
- 前記ウイルスは、アデノウイルス、ポリオウイルス、A型肝炎ウイルス、日本脳炎ウイルス、ヨーロッパダニ媒介性脳炎ウイルス、および関連する東洋形態、デング熱ウイルス、黄熱病ウイルス、C型肝炎ウイルス、風疹ウイルス、流行性耳下腺炎ウイルス、麻疹ウイルス、RSウイルス、ワクシニアウイルス、インフルエンザウイルス、ロタウイルス、ラブドウイルス、肺炎ウイルス、レオウイルス、1型単純ヘルペスウイルスまたは2型単純ヘルペスウイルス2、サイトメガロウイルス、水痘帯状疱疹ウイルス、イヌアデノウイルス、エプスタイン−バーウイルス、ウシヘルペスウイルスまたはブタヘルペスウイルス、または仮性狂犬病ウイルスである、請求項6に記載の方法。
- 前記ワクチンは、アジュバント、補助剤、緩衝剤、希釈剤または薬物キャリアと混合される、請求項1に記載の方法。
- (d)前記ウイルスをCSクロマトグラフィーカラムおよび/またはスクロース勾配超遠心分離により精製する工程をさらに包含する、請求項1〜8のいずれか一項に記載の方法。
