インフルエンザA型ウイルスは、少なくとも10個のタンパク質をコードする8つの一本鎖マイナス鎖ウイルスRNA(vRNA)のゲノムを有する。インフルエンザウイルスライフサイクルは、宿主細胞の表面上におけるシアル酸含有受容体への赤血球凝集素(HA)の結合から開始した後、受容体依存性エンドサイトーシスが生じる。後期エンドソームにおける低pHによって、HAにおける立体配座シフトが引き起こされることにより、HA2サブユニット(いわゆる融合ペプチド)のN末端が曝露される。融合ペプチドは、ウイルス膜及びエンドソーム膜の融合を開始し、マトリックスタンパク質(M1)及びRNP複合体が細胞質に放出される。RNPは、vRNAをキャプシド形成する核タンパク質(NP)と、PAタンパク質、PB1タンパク質及びPB2タンパク質によって形成されるウイルスポリメラーゼ複合体とからなる。RNPは、転写及び複製が起きる核の中に移送される。RNAポリメラーゼ複合体は、3つの異なる反応、すなわち、5’キャップ及び3’ポリA構造を有するmRNAの合成、全長相補的RNA(cRNA)の合成、及び鋳型としてのcRNAを用いたゲノムvRNAの合成、を触媒する。次いで、新しく合成されたvRNAs、NP及びポリメラーゼタンパク質は、RNPに構築され、核から搬出され、子孫ウイルス粒子の出芽が生じる原形質膜に移送される。ノイラミニダーゼ(NA)タンパク質は、シアリルオリゴ糖からシアル酸を除去し、したがって、新しく構築されたビリオンを細胞表面から放出し、ウイルス粒子の自己凝集を防止することによって、感染における後期において重要な役割を果たす。ウイルス構築は、タンパク質-タンパク質相互作用及びタンパク質-vRNA相互作用を含むが、これらの相互作用の性質は、大部分が不明である。
インフルエンザウイルスを分離し且つ/又は増殖させるために、インフルエンザウイルスの効率的な複製を支える突然変異体細胞を含む任意の細胞、例えば、トリの細胞又はヒトなどの哺乳類の細胞、例えば、293T細胞若しくはPER.C6(登録商標)細胞、又は、イヌ、例えばMDCK、ウシ、ウマ、ネコ、ブタ、ヒツジ、齧歯動物、例えばミンク、例えばMvLu1細胞、若しくはハムスター、例えばCHO細胞、若しくは非ヒト霊長類、例えばVero細胞を用いることができる。再集合体ウイルスを調製するために、分離されたウイルスを用いることができる。一実施形態において、ワクチン作製のための宿主細胞は、哺乳類又はトリの連続細胞系又は連続細胞株である。最終産物の純度についての適切な試験を含むことができるように、用いられる前記細胞の完全な特性決定を行ってもよい。細胞の特性決定のために用いることができるデータは、(a)その由来、誘導及び継代歴に関する情報、(b)その成長及び形態学的特性に関する情報、(c)偶発因子の試験の結果、(d)細胞が他の細胞系の中で明確に認識されることを可能にする生化学的パターン、免疫学的パターン及び細胞遺伝学的パターンなどの目立った特徴、及び(e)腫瘍形成性についての試験の結果、を含む。一実施形態において、用いられる宿主細胞の継代レベル又は集団倍化は、できるだけ低い。
本開示のワクチンは、本開示の分離された組換えインフルエンザウイルス、及び場合により他の分離されたインフルエンザウイルスを含む1以上の他の分離されたウイルス、1以上の分離されたインフルエンザウイルス若しくは1以上の他の病原体の1以上の免疫原性タンパク質若しくは糖タンパク質、例えば、1以上の細菌、非インフルエンザウイルス、酵母若しくは真菌に由来する免疫原性タンパク質、又は本開示の分離されたインフルエンザウイルスの1以上の免疫原性タンパク質を含む1以上のウイルスタンパク質をコードする分離された核酸(例えば、DNAワクチン)を含む。一実施形態において、本開示のインフルエンザウイルスは、インフルエンザウイルス又は他の病原体のためのワクチンベクターであってもよい。
WT-Venus-PR8を産生するためにインフルエンザA型/Puerto Rico/8/34(PR8、H1N1)ウイルスのNSセグメントにVenus蛍光タンパク質についての遺伝子を挿入することによって、例えばインフルエンザウイルス誘導病理を理解するためにウイルス感染細胞の視覚化を可能にするレポータインフルエンザウイルスを調製した。挿入されたVenus遺伝子は、WT-Venus-PR8の連続的継代の間に欠失したが、WT-Venus-PR8は有意に弱毒化され、PB2-E712D突然変異は、Venus遺伝子を安定化させることが分かった。本明細書に開示されるように、Venus遺伝子欠失が起きる機序と、ポリメラーゼ突然変異がVenus遺伝子を安定化する方法とを検討した。大規模配列決定分析によって、PB2-E712Dが突然変異率の顕著な変化を引き起こさないことが明らかになったが、このことは、Venus遺伝子の安定性がポリメラーゼ忠実度による影響を受けないことを示唆している。定量的リアルタイムPCRを用いて、WT-Venus-PR8が、高レベルのインターフェロンβ(IFN-β)の発現を誘導することが分かった。IFN-β発現の誘導は、WT-Venus-PR8における修飾NSセグメントの転写/複製効率性の低下に起因すると思われた。対照的に、修飾NSセグメントの転写/複製効率性は、PB2-E712D突然変異によって増強された。WT-Venus-PR8におけるVenus遺伝子の喪失は、NSセグメントにおける内部欠失によって引き起こされると思われた。さらに、Venus安定化機序のさらなる理解のために、Venus遺伝子を安定化させるウイルスポリメラーゼ複合体における付加的なアミノ酸突然変異を同定した。これらのアミノ酸の一部がウイルスポリメラーゼの鋳型出口又は産物出口の近くに位置することが分かったが、このことは、これらのアミノ酸が、ポリメラーゼ複合体とRNA鋳型及び産物との間の結合親和性に影響を及ぼすことによってVenus遺伝子の安定性に寄与することを示唆している。

上記再集合体を調製するための複数のインフルエンザウイルスベクターを提供する。一実施形態において、vRNA産生のための前記ベクターにおける前記PB1 DNA、前記PB2 DNA、前記PA DNA、前記NP DNA、前記NS DNA及び前記M DNAは、配列番号1~6又は10~15によってコードされる対応するポリペプチドに対して少なくとも95%のアミノ酸配列同一性を有するポリペプチドをコードするものに対応する配列を有する。一実施形態において、PB2の540位における前記残基は、K、R若しくはHであり、PAにおける180位における前記残基は、R、K若しくはHであり、PAにおける200位における前記残基は、A、I、L、G若しくはVであり、PB1における149位における残基は、A、T、I、L若しくはGであり、684位における前記残基は、D若しくはNであり、又はPB1における685位における前記残基は、E若しくはまたQである。一実施形態において、前記ウイルスセグメントの内の少なくとも1つは、遺伝子産物をコードする異種の遺伝子配列を含む。一実施形態において、前記ベクターは、遺伝子産物をコードする異種遺伝子配列を含むウイルスセグメントを有するさらなるベクターを含む。
接種、例えば経鼻投与、非経口投与又は経口投与のために適切な本開示の医薬組成物は、1以上のインフルエンザウイルス分離株、例えば、1以上の弱毒化若しくは不活化インフルエンザウイルス、それらのサブユニット、それらの(1又は複数の)分離されたタンパク質、及び/又はそれらの1以上のタンパク質をコードする分離された核酸を含み、滅菌された水溶液又は非水溶液、懸濁剤及び乳剤を場合によりさらに含む。前記組成物は、本技術分野において知られている通りの補助剤又は賦形剤をさらに含むことができる。本開示の前記組成物は、概して個別の用量(単位用量)の形態で示される。
前記組成物(又はそれが誘発する抗血清)の投与は、「予防」又は「治療」目的のいずれかのためのものであってもよい。ワクチンである本開示の前記組成物は、予防的に提供される場合、病原体感染の病徴又は臨床徴候が明らかになる前に提供される。前記組成物の予防的投与は、いかなる後続の感染も予防するか又は弱める役割を果たす。本開示の前記遺伝子治療組成物は、予防的に提供される場合、疾患のいずれの病徴又は臨床徴候が明らかになる前に提供される。前記組成物の予防的投与は、前記疾患に関連する1以上の病徴又は臨床徴候を予防するか又は弱める役割を果たす。
本開示の組成物は、受動免疫又は能動免疫によって1以上の病原体、例えば1以上のインフルエンザウイルス株に対する耐性を付与してもよい。能動免疫において、弱毒化生ワクチン組成物は、予防的に宿主(例えば、哺乳動物)に投与され、投与に対する宿主の免疫応答によって、感染及び/又は疾患に対する保護が行われる。受動免疫のために、誘発された抗血清を回収して、少なくとも1つのインフルエンザウイルス株によって引き起こされる感染があることが疑われるレシピエントに投与することができる。本開示の遺伝子治療組成物は、能動免疫によって予防レベル又は治療レベルの所望の遺伝子産物を産生してもよい。
方法
色-インフルエンザの作製
eCFP、eGFP、Venus及びmCherryを含む異なる蛍光レポータ遺伝子と融合させたPR8のNSセグメントを、Manicassamyら(2010)に記載されているように重複融合PCRによって構築した。手短に言えば、アミノ酸リンカーGSGGをコードしている配列によって、終止コドンを有さないNS1遺伝子のオープンリーディングフレーム(ORF)を蛍光レポータ遺伝子のN末端と融合させた。蛍光レポータORFには、GSGリンカーをコードする配列、Manicassamyら(2010)におけるブタテシオウイルス-1に由来する57個のヌクレオチドを有する口蹄病ウイルスプロテアーゼ2A自己タンパク質分解部位、及び核外移行タンパク質(nuclear export protein)(NEP)のORFが続いた(図5)。加えて、サイレント突然変異をNS1遺伝子の内在性スプライス受容部位に導入してスプライシングを抑止した(Baslerら、2001)。続いて、Newmannら(1999)に記載されているように、構築されたNSセグメント(eCFP-NS、eGFP-NS、Venus-NS及びmCherry-NSと呼ばれる)をリバースジェネティクスのためのpPolIベクターにクローニングした。Venusレポータタンパク質をコードするプラスミドは、A.Miyawaki博士(Laboratory for Cell Function Dynamics、理化学研究所 脳科学総合研究センター、和光、日本)からの寄贈であった(Nagaiら、 2002)。Newmannら(1999)に記載されているようにリバースジェネティクスシステムを用いてWT-Venus-PR8を作製した。WT-Venus-PR8病原性レベル及びVenus発現レベルがマウスにおいてかなり弱くなったので、マウスにおいてWT-Venus-PR8の継代を連続的に行った。6回の継代の後、病原性が増加し、Venus発現が強くなった変異体(MA-Venus-PR8)が得られた。MDCK細胞において、MA-Venus-PR8のストックが生成された。典型的には動物における連続的継代によって遺伝子的変異体から構成されるウイルス集団が生じるので、リバースジェネティクスを用いることによってMA-Venus-PR8を再形成した。同様に、MA-Venus-PR8と同じ遺伝子的骨格を有するMA-eCFP-PR8、-eGFP-PR8及び-mCherry-PR8を生成した。
雌の6週齢のC57BL/6(’B6’)マウスを日本エスエルシー株式会社(静岡、日本)から購入した。セボフルラン麻酔下で50μLのPBS中において図のパネルに示された用量で色-インフルエンザウイルスをマウスに鼻腔内接種し、体重及び生存率を14日間モニタされた。図のパネルに示される時点で、ウイルス滴定、フローサイトメトリ分析及び組織学的実験のためにPBS接種マウス又は色-インフルエンザ感染マウスから肺を採取した。全ての動物実験は、東京大学動物実験委員会の規定に従って行われ、東京大学医科学研究所の動物実験委員会によって承認された。
4%パラホルムアルデヒド(PFA)リン緩衝溶液中に肺を固定した。固定された組織をOCT化合物(サクラファインテック、東京、日本)の中に包埋し、液体N2によって凍結し、-80℃で貯蔵した。クライオスタット6μm切片を、1%BSA(PBS-BSA)を含有するPBSで30分間処置して非特異的結合を阻止し、次いで、フィコエリトリン(PE)-Mac3(M3/84、BD Biosciences、サンノゼ、カリフォルニア州)でインキュベートした。MDCK細胞の細胞診断を検討するために、細胞を色-インフルエンザウイルスに感染させ、次いで、4%PFAリン緩衝溶液中に固定した。核をHoechst33342(Invitrogen、カールズバッド、カリフォルニア州)で染色した。NIS-Elementsソフトウェアによって制御される共焦点顕微鏡(Nikon A1、ニコン、東京、日本)を用いて、切片及び細胞を視覚化した。定量的多色撮像分析のために、InFormソフトウェア(PerkinElmer、ウォルサム、マサチューセッツ州)を有するNuance FX多スペクトル感応性イメージングシステムを有する倒立蛍光顕微鏡(Nikon Eclipse TS100)を用いることによってスライドを視覚化した。
マウスを安楽死させ、心臓内にPBSを潅流させて肺から血液細胞を除去した。4%PFAリン緩衝溶液による気管内潅流の後に肺を分離した。製造業者の使用説明書に従ってSCALEVIEW-A2溶液(オリンパス、東京、日本)で肺組織をきれいにした。デジタルカメラ(DFC365FX、Leica Microsystems)を備えた立体蛍光顕微鏡(M205FA、Leica Microsystems、ウェッツラー、ドイツ)を用いることによって画像を取得した。
合計で105PFUのMA-eGFP-PR8をB6マウスに鼻腔内接種した。肺マクロファージを標識するために、感染後Day3において、マウスに50μLのPE-CD11b(M1/70、BioLegend、サンディエゴ、カリフォルニア州)を静脈内注射した。抗体注射の30分後、マウスの肺を採取した。肺におけるeGFP陽性細胞及びPE陽性細胞のキネティクスを多光子顕微鏡(LSM 710 NLO、Carl Zeiss、オーバーコッヘン、ドイツ)で撮像した。分析の間、湿潤チャンバ(37℃、5%CO2)内で完全培地(10%ウシ胎仔血清を有するRPMI1640)中において肺を維持した。LSMソフトウェアZen2009(Carl Zeiss)でデータを処理した。HPAIウイルス感染肺組織の三次元撮像のために、B6マウスに105PFUのMA-Venus-HPAIウイルスを鼻腔内接種した。肺組織を、感染後Day2においてマウスから回収し、SCALEVIEW-A2溶液(オリンパス)で処置して、上記のように組織を透明にした。肺組織の三次元画像を多光子顕微鏡(Nikon A1R MP)から得た。
単細胞懸濁液を得るために、肺を、ナイロンフィルタ(BD Biosciences)によって組織を粉砕することによって、37℃で30分間、Collagenase D(Roche Diagnostics、マンハイム、ドイツ;終濃度:2μg/mL)及びDNase I(Worthington Biochemical、レークウッド、ニュージャージー州;終濃度:40U/mL)によって解離させた。RBC溶解緩衝剤(Sigma Aldrich、セントルイス、ミズーリ州)による処置によって赤血球(RBC)を溶解した。抗体の非特異的結合を阻止するために、精製抗マウスCD16/32(Fc Block、BD Biosciences、サンディエゴ、カリフォルニア州)によって細胞をインキュベートした。蛍光抗体の適切な組み合わせによって細胞を染色して各免疫細胞サブセットの集団を分析した。以下の抗体、すなわち、抗CD45(30-F11:eBioscience、サンディエゴ、カリフォルニア州)、抗CD11b(M1/70:BioLegend)、抗F4/80(BM8:eBioscience)及び抗CD11c(HL3:BD Biosciences)を用いた。死滅細胞の排除のために7-アミノアクチノマイシンD(Via-Probe、BD Biosciences)によって全ての試料をインキュベートした。標識細胞からのデータを、FACSAria II(BD Biosciences)において取得し、FlowJoソフトウェアバージョン9.3.1(Tree Star、サンカルロス、カリフォルニア州)によって分析した。Venus陽性マクロファージ及びVenus陰性マクロファージを肺から分離するために、染色された細胞を、FACSAria II(BD Biosciences)を用いて選別した。
選別されたマクロファージのトータルRNAを、TRIzol試薬(Life Technologies、カールズバッド、カリフォルニア州)を用いて抽出し、イソプロパノールによって沈殿させた。製造業者の使用説明書に従って、Arcturus Riboamp Plus RNA Amplification Kit(Life technologies)を用いてRNA増幅を行った。RNAを、Agilent Low Input Quick Amp Labelingキット(1色)(Agilent Technologies、サンタクララ、カリフォルニア州)を用いることによって標識し、SurePrint G3 Mouse GE 8X60Kマイクロアレイ(Agilent Technologies)にハイブリダイズした。アレイをSureScan High-Resolution Technology(G2565CA;Agilent Technologies)を有するDNA Microarray Scannerで走査し、Agilent Feature Extractionソフトウェアバージョン10.7.3.1(Agilent Technologies)を用いてデータを取得した。プローブアノテーションは、Agilent Technologies(AMADID 028005)によって提供された。プローブ強度を、それぞれ標準指数法及び分位点法を用いて、バックグラウンド補正し、正規化した。次いで、強度のlog2を、対象の群を比較した線形モデルに当てはめた34。全ての報告されたp値を、Benjamini-Hochberg法を用いて複数の仮説比較のために調整した。調整されたp<0.01を伴う対比の間に平均プローブ強度の少なくとも2倍の変化があった場合、転写物が差次的に発現したと考えた。階層的クラスタリングをRにおいて行った。次いで、得られた遺伝子クラスタをToppCluster(Kaimalら、2010)で分析して、各クラスタにおいて強化された遺伝子アノテーションを同定した。報告されたスコアは、Benjamini-Hochberg調整p値の-log10である。
MDCK細胞の全溶解物をSDS-ポリアクリルアミドゲル(Bio-Rad Laboratories、ハーキュリーズ、カリフォルニア州)によって電気泳動にかけ、PVDF膜(Miilipore、ビルリカ、マサチューセッツ州)にトランスファーした。膜をBlocking One(ナカライテスク、京都、日本)でブロックし、ウサギ抗GFPポリクローナル抗体(MBL、名古屋、日本)、マウス抗NS1抗体(188/5)、A/WSN/33(H1N1)(R309)に対するウサギ抗血清又はマウス抗アクチン抗体(A2228;Sigma-Aldrich)でインキュベートした後、HR共役抗マウス又は抗ウサギIgG抗体(GE Healthcare、ウォーキショー、ウィスコンシン州)でインキュベートした。PBS-Tweenで前記膜を洗浄した後、ECL Plus Western Blotting Dectection System(GE Healthcareを用いて特異的タンパク質を検出した。VersaDoc Imaging System(Bio-Rad)を用いることによって特異的タンパク質バンドを視覚化した。
NS1オープンリーディングフレームに融合されたレポータタンパク質を発現する蛍光インフルエンザウイルスを生成するために、Venusを選択した(GFPと比較して発色団形成を向上させ、輝度を増加させるF46Lを含む8つの突然変異を有するGFP変異体)(Wagaiら、2002)。レポータタンパク質を発現するインフルエンザウイルスについての弱毒化の以前の知見に基づいて予想されるように(Kittelら、2004;Shinhyaら、2004)、Venus(WT-Venus-PR8)を発現するA/Puerto Rico/8/34(PR8;H1N1)ウイルスのマウス病原性は、野生型PR8(WT-PR8)よりも実質的に低く、感染マウスの50%が死滅するために必要な用量(MLD50)は、WT-PR8についての102.5PFUと比較して、WT-Venus-PR8については104.5プラーク形成単位(PFU)超であった。C57BL/6(B6)マウスにおいて、WT-Venus-PR8の継代を連続的に行った。6回の連続的継代の後、変異体(MA-Venus-PR8と呼ばれる;赤血球凝集素タンパク質の380位におけるTからAへの突然変異とポリメラーゼサブユニットPB2の712位におけるEからDへの突然変異とを有する)は、WT-Venus-PR8と比較して、かなりより高い病原性(MLD50=103.5PFU)を有することが同定されたが、元のPR8ウイルスよりも依然として病原性が低かった(図1A)。マウス肺におけるMA-Venus-PR8の複製能を評価するために、104PFUのMA-Venus-PR8ウイルス又はPR8ウイルスをB6マウスに鼻腔内感染させた。試験された全ての時点において、肺ウイルス力価は、MA-Venus-PR8感染マウス及びPR8感染マウスについて同様であった(図1B)。Venus発現の安定性を試験するために、感染マウス由来の肺ホモジェネートを用いてプラークアッセイを行い、感染後(p.i.)におけるDay3、Day5及びDay7の各々において150のプラークの内の1つのみがVenus陰性であることが分かったが、このことは、この組換えウイルスにおけるVenus発現の高遺伝的安定性を証明している。対照的に、NS1-GFPウイルスの70%のみがレポータタンパク質を発現した(Manicassamyら、2010)。MA-Venus-PR8の堅牢な毒性及び遺伝的安定性は、このウイルスが、インビボにおいてインフルエンザウイルス感染細胞を視覚化するための非常に魅力的なレポータシステムを表すことを示す。
この研究において、細胞レベルでインフルエンザウイルス感染を研究するために色-インフルエンザウイルスを生成した。色-インフルエンザウイルスは、堅牢なウイルス複製、毒性、安定な蛍光タンパク質発現、及び同時に視覚化され得る一組の4つの異なる色を含む、既存のシステムを超えるいくつかの向上を組み合わせる。色-インフルエンザウイルスは、全てのインフルエンザウイルス株に適用可能である。これらの向上によって、感染細胞及びバイスタンダー細胞の全般的トランスクリプトミクス分析が可能になり、初めて、マウス肺におけるインフルエンザウイルス感染細胞のライブイメージングが可能になる。
実施例Iにおいて開示されたように、リバースジェネティクスを用いて、Venus(Nagaiら、2002)(eGFPの変異体)レポータ遺伝子を有するH5N1型ウイルス(野生型Venus-H5N1型ウイルスと呼ばれる、WT-Venus-H5N1ウイルスと略記される)を調製したが、このウイルスは、マウスにおいて中等度の毒性及び低いVenus発現を示した。マウスにおける6回の継代の後、インビボにおいて高レベルのVenusを安定して発現し、マウスにとって致死性であったマウス適応Venus-H5N1型ウイルス(MA-Venus-H5N1ウイルスと略記される)を取得し、感染マウスの50%を死滅させるために必要な用量(MLD50)は、3.2プラーク形成単位(PFU)であったが、その親WT-Venus-H5N1ウイルスについては103PFUであった。しかし、毒性及びVenus安定性のこの差異についての機序は不明だった。
細胞。10%ウシ胎仔血清を有するダルベッコ変法イーグル培地(DMEM)中においてヒト胎児腎臓293及び293T細胞を維持し、5%新生児ウシ血清が補充された最小必須培地(MEM)中においてマジンダービーイヌ腎臓(MDCK)細胞を維持した。5%CO2において37℃で全ての細胞をインキュベートした。
WT-Venus-H5N1ウイルス及びMA-Venus-H5N1ウイルスの間の比較。
実施例Iにおけるように、リバースジェネティクスを用いることによってA/Viet Nam/1203/2004(H5N1)(VN1203と略記される)のNSセグメントをVenus-PR8ウイルスのVenus融合NSセグメントと置換し、Venus蛍光レポータ遺伝子(WT-Venus-H5N1ウイルス)を発現したH5N1型ウイルスを取得した。マウスにおける病原性分析は、このウイルスが、103PFUのMLD50値を有する親VN1203と比較して(VN1203について0.7PFUと比較して)マウスにおける弱毒化された毒性を示したことを示した(図6A及び図6B並びにHattaら(2007))。その上、主に呼吸器官で複製されるWT-Venus-H5N1ウイルス(表3)、及びそのVenus発現は、MDCK細胞(図7)及びウイルス感染後のマウス(図8)の両方において非常に弱かった。
マウス適応後のVenus-H5N1ウイルスの毒性及びVenus発現の差異についての遺伝子的基準を検討するために、RG-MAウイルスと呼ばれたMA-Venus-H5N1ウイルスのためのリバースジェネティクスシステムを確立した。RG-MAウイルスは、その元のウイルス(MA-Venus-H5N1ウイルス)と同様の器官におけるウイルス力価、MLD50値(図6C、図6D及び図9)及びマウスにおけるVenus発現(図9)を示した。
これらのウイルスの複製能をMDCK細胞においてさらに検討し、MA-Venus-H5N1ウイルスが、RG-MAウイルスと同様の複製能を有し、WT-Venus-H5N1ウイルスよりも効率的に増殖し、MA-Venus-H5N1ウイルスの力価が、36hpi及び48hpiでWT-Venus-H5N1ウイルスの力価よりも有意に高かったことが分かった(図10)。次いで、2つのウイルスの複製に対するPB2ウイルスセグメント及びPAウイルスセグメントの寄与を検討した。感染後のいくつかの時点におけるWT-Venus-H5N1ウイルスの力価と比較して、WT+MA-PB2及びWT+MA-(PB2+PA)の有意により高い力価が認められたが、それでも、WT+MA-PAの複製効率は、WT-Venus-H5N1ウイルスの複製効率と同等だった(図10)。WT+MA-(PB2+PA)の力価は、36hpi及び48hpiでWT+MA-PB2の力価よりも高かったが、その差は統計学的に有意ではなかった(図10)。これらの結果は、MA-PB2遺伝子がMDCK細胞におけるVenus-H5N1ウイルスの複製を増強し、この増加はMA-PAの存在下でさらに増強され得るが、MA-PA単独はMDCK細胞におけるウイルス複製を改変しないことを示す。
ウイルスリボ核タンパク(RNP)複合体のポリメラーゼ活性は、ウイルスの複製及び毒性と相関してきた(Gabrielら、2005;Leungら、2010;Liら、2008;Salomenら、2006)。ルシフェラーゼ活性を測定することによって、WT-Venus-H5N1ウイルス又はMA-Venus-H5N1ウイルスに由来するPB1、PB2及びPAの8つのRNPの組み合わせの活性を決定した。マウス適合ウイルスのポリメラーゼ活性は、WT-Venus-H5N1ウイルスのポリメラーゼ活性よりも4倍近く低かった(図11)。任意のMA遺伝子の置換は、WT-Venus-H5N1ウイルスのポリメラーゼ複合体の活性を減少させたが、MA-PB2及びMA-PAの二重置換を含む複合体のポリメラーゼ活性は、WT-Venus-H5N1ウイルスのポリメラーゼ活性と比較して有意に減少し、MA-Venus-H5N1ウイルスのポリメラーゼ活性と同様であった。これらの結果は、RNP複合体のポリメラーゼ活性がマウス適応後にとりわけ減少したことを示すが、このことは、増強された複製及び毒性と一致していない。
インビトロにおけるWT-Venus-H5N1及びRG-MAウイルスにおけるVenus安定性を評価するために、MDCK細胞において2つのウイルスの継代を5回行った。これらの継代の間、RG-MAウイルスからではなく、WT-Venus-H5N1ウイルスからVenus陰性プラークを採取したが、これによって、Venus遺伝子がマウス適応後においてより安定していることが示唆された(表5)。このVenus安定性の分子決定因子を同定するために、MDCK細胞において各種再集合体の継代を5回行った。MA-PB1遺伝子、MA-NA遺伝子又はMA-M遺伝子を有する再集合体からVenus陰性プラークを取得したが、我々は、MA-PB2遺伝子、MA-PA遺伝子、MA-(PB2+PA)遺伝子又はMA-NS遺伝子を有するVenus-H5N1ウイルスの5回目の継代からいずれのVenus陰性プラークも得なかった(表5)。これらのデータは、MA-PB2遺伝子、MA-PA遺伝子及びMA-NS遺伝子がVenus安定性において役割を果たすことを示唆する。
前に、マウスに対してより致死性となり、且つマウス適応後により安定することになったVenusレポータ遺伝子を発現する視覚化可能なH5N1ウイルスを構築した(実施例I)。この研究において、このウイルス(MA-Venus-H5N1)の全ゲノムを配列決定し、WT-Venus-H5N1ウイルス配列とは異なった7つのアミノ酸を同定した。これらの2つのウイルスの間のマウスにおける毒性及びVenus発現の差異についての分子決定因子を探究するために、リバースジェネティクスを用いて両ウイルスの一連の再集合体を生成した。PB2(V25A)及びPA(R443K)の二重突然変異は、マウスにおけるWT-Venus-H5N1の病原性を著しく増強することが分かった。PB2のV25Aも、MDCK細胞及びマウスにおけるVenus発現及びウイルス複製を有意に増加させ、PAのそのR443Kは、これらの効果をさらに増強した。異なる再集合体の安定性をインビトロにおいて検討し、MA-PB2、MA-PA又はMA-NSを有する再集合体がより安定していることが分かった。これらの結果は、PB2タンパク質及びPAタンパク質が、哺乳動物宿主におけるVenus発現H5N1ウイルスの病原性及びVenus安定性において役割を果たすことを示唆する。
材料及び方法
細胞及びウイルス。5%の新生児ウシ血清(神経皮膚NCS)を含有する最少必須培地(MEM)中においてマジンダービーイヌ腎臓(MDCK)細胞を維持した。10%ウシ胎仔血清(FCS)が補充されたダルベッコ変法イーグル培地中においてヒト胎児腎臓293T(HEK293T)及びHEK293細胞を維持した。リバースジェネティクスを用いることによってA/Puerto Rico/8/34(H1N1;PR8)(Horimotoら、2007)及び各NS1-Venus PR8ウイルスを生成し、L-(トシルアミド-2-フェニル)エチルクロロメチルケトン(TPCK)処置トリプシン(0.8μg/mL)及び0.3%ウシ血清アルブミン(BSA)(Sigma Aldrich)を含有するMEM中において、48時間、37℃で、MDCK細胞において増殖させた。
マウス適応NS1-Venus PR8ウイルスの確立。リバースジェネティクスによってNS1-Venus PR8 WTウイルスを救出することに成功したが、このウイルスは、マウスにおいて無毒性であり(MLD50:>105PFU)、Venusの発現は、MDCK細胞及びこのウイルスを感染させたマウスの肺切片において非常に弱かった。NS1-Venus PR8 WTウイルスの毒性及びVenus発現を増加させるために、プラーク精製高Venus発現クローンによる鼻腔内感染によってマウスにおいて前記ウイルスの継代を連続的に行った(実施例I及び実施例II参照)。6回の連続的継代の後、ウイルスの毒性は増加したと思われ、ゆえに、突然変異を探索するために、このマウス適応NS1-Venus PR8 WTウイルスを配列決定した。
ウイルスNS1タンパク質を有する融合構築体から異種遺伝子産物を発現し得るベクターが上記に記載されている(図26)。特に、PB2-E712D突然変異は、異種遺伝子産物の発現を安定化した。試験ウイルス(WT-Venus-PR8)の継代を連続的に行って、安定化に寄与するポリメラーゼ複合体における他の突然変異を同定した(図27)。
ポリメラーゼサブユニットPB2の712位におけるEからDへの突然変異(PB2-E712D)は、挿入されたVenus遺伝子を安定化した(Fukuyamaら、2015;Katsuraら、2016)。また、PR8からNSセグメントに挿入されたVenus遺伝子を担持するH5N1ウイルス(Venus-H5N1)も調製した(Fukuyamaら、2015)。しかし、WT-Venus-PR8と同様に、WT-Venus-H5N1は中等度の毒性及び低Venus発現を示したが、我々は、マウスに対してより致死性になり、マウス適応後にVenusを安定して発現した変異体を取得した。ポリメラーゼサブユニットPB2の25位におけるVからAへの突然変異及びポリメラーゼサブユニットPAの443位におけるRからKへの突然変異は、Venus遺伝子の安定した維持に寄与した(Zhaoら、2015)。これらの結果は、ウイルスポリメラーゼの組成物が、挿入された外来遺伝子の安定化において役割を果たすことを示した。しかし、Venus遺伝子を欠失することができる機序と、ポリメラーゼ突然変異がVenus遺伝子を安定化する方法とは、不明なままであった。
細胞及びウイルス。5%CO2において37℃で5%新生児ウシ血清を有する最小必須培地(Gibco)中でマジンダービーイヌ腎臓(MDCK)細胞を培養した。10%のウシ胎仔血清が補充されたダルベッコ変法イーグル培地中においてヒト胎児腎臓293T(HEK293T)細胞を培養した。リバースジェネティクス(Neumannら、1999)を用いることによって、WT-Venus-PR8及びVenus蛍光タンパク質をコードするNSセグメントを有するVenus-PR8突然変異体(Fukuyamaら、2015)を生成し、37℃でMDCK細胞において増殖させた。
WT-Venus-PR8におけるVenus発現の喪失は、複製効率を回復させる。以前に記載されているように(Neumannら、1999)リバースジェネティクスを用いることによって、WT-Venus-PR8及びVenus-PR8-PB2-E712Dを調製した。図28Aに示されるように、Venus蛍光タンパク質の遺伝子をNSセグメントに挿入した(Fukuyamaら、2015)。最初に、Venus発現がWT-Venus-PR8においてどの程度迅速に喪失したのかについて、及びVenus欠失及びウイルス力価の間の関係について確認した。0.001の感染多重度(MOI)でMDCK細胞においてウイルスの継代を行い、Venus陽性プラークの割合を測定した(図28B)。WT-Venus-PR8においてVenusの発現は直ちに喪失したが、Venus-PR8-PB2-E712Dの全てのプラークは、4回の継代後にVenus発現を示したことが確認された。WT-Venus-PR8は、以前に記載されているように(Katsuraら、2016)、MDCK細胞におけるVenus-PR8-PB2-E712Dよりも低い力価を示したが、Venus陽性プラークの割合が減少するにつれてウイルス力価はウイルス継代の間に増加した(図28C)。この結果は、突然変異WT-Venus-PR8におけるVenus遺伝子の喪失がウイルスの複製効率を回復させたことを示唆する。
PB2-E712D突然変異は、継代の間、挿入されたVenus遺伝子を保持するために、ウイルスポリメラーゼ忠実度を増加させるということを仮定した。この仮説を試験するために、WT-PR8と、PB2の712位においてアスパラギン酸を有するので、このアミノ酸のみがWT-PR8と異なるPR8-PB2-E712Dとをリバースジェネティクスによって生成し、それらの突然変異率を比較した。ここで、突然変異率を測定することをより容易にするために、Venus遺伝子を含まなかったウイルスを用いた。忠実度が高い突然変異体ウイルスであることが報告されているPR8-PB1-V43I(Cheungら、2014;Naitoら、2015)及び忠実度が低い突然変異体ウイルスであることが報告されているPR8-PB1-T123A(Paulyら、2017)もリバースジェネティクスによって生成し、対照として用いた。突然変異率を推定するために、これらのウイルスの継代を、0.001のMOIで、MDCK細胞において行い、ゲノム全体の大規模配列決定を行い、継代を5回行ったウイルスについての配列決定データを、継代を行う前のウイルスの配列決定データと比較した。継代を行う前に存在しなかった継代を5回行ったウイルスにおけるヌクレオチド変化の数を計数した。5回の継代の間に導入された突然変異の数は、セグメントによって図29Aに示される。また、ヌクレオチド1つ当たりの突然変異の数も正規化のために算出し、8つの全セグメントについての平均値を比較した(図29B)。忠実度が高い対照であるPR8-PB1-V43Iは、WT-PR8よりも突然変異が少なく、忠実度が低い対照であるPR8-PB1-T123Aは、WT-PR8よりも突然変異が多いことが確認された。PR8-PB1-V43Iは、WT-PR8よりも突然変異が少なかったが、PR8-PB1-V43I及びWT-PR8の間の差は小さかった。以前の報告は、PB1-V43Iが突然変異率を改変しないことを示唆したので(Paulyら、2017)、突然変異率に対するPB1-V43Iの影響は、ウイルス株又は実験条件に依存した可能性があった。その上、WT-PR8及びPR8-PB2-E712Dの間に突然変異数の明瞭な差はなかった。PB2-E712D突然変異がウイルスポリメラーゼ忠実度に影響を及ぼすという可能性を除外することはできないものの、その効果は、Venus安定性のかなりの差を引き起こすために充分に大きいとは思われない。ゆえに、この結果は、Venus遺伝子の安定性がウイルスポリメラーゼの忠実度によって影響されないことを示唆する。
いくつかの報告は、それらのNSセグメントにおける外来遺伝子挿入を含む組換えウイルスが、MDCK細胞などのIFNコンピテント細胞におけるよりもインターフェロン(IFN)欠乏性Vero細胞において効率的に増殖する可能性があることを示唆する(Kittelら、2004;Ferkoら、2001;Kuznetsovaら、2014)。ゆえに、定量的リアルタイムPCRを用いることによって、ウイルス感染細胞におけるIFN-βの発現レベルを定量した。1のMOIでWT-Venus-PR8若しくはVenus-PR8-PB2-E712DをMDCK細胞に感染させたか、又は培地だけを模擬に感染させ、感染細胞におけるIFN-βの相対発現レベルを感染の9時間後に定量した。WT-Venus-PR8は、Venus-PR8-PB2-E712Dよりも高いIFN-β発現レベルを誘導した(図30A)。この結果は、WT-Venus-PR8がIFN-β発現を効率的に阻害しないことを示唆する。NS1が宿主におけるIFN発現及びIFN媒介抗ウイルス応答を抑制することにおいて主要な役割を果たすと仮定すると(Garcia-Sastreら、1998;Opitzら、2007)、インフルエンザウイルスストランド特異的リアルタイムPCRを用いることによって感染細胞におけるNS vRNAを定量した(Kawakamiら、2011;Kupkeら、2018)。WT-Venus-PR8感染細胞におけるNS vRNAの量は、Venus-PR8-PB2-E712D感染細胞におけるNS vRNAの量よりも90%低かったが(図30B)、それらのNP vRNA発現レベルにおける有意な差(dierence)はなかった(図30C)。WT-Venus-PR8感染細胞におけるNS vRNA/NP vRNA比は、Venus-PR8-PB2-E712D感染細胞におけるNS vRNA/NP vRNA比よりも80%低かった(図30D)。この結果は、NSセグメントの転写/複製がWT-Venus-PR8において特異的に損なわれることを示唆する。リバースジェネティクスによってウイルスを生成するために用いられるプラスミドの内の全ての配列を使用前に確認し、同じNS-VenusプラスミドからWT-Venus-PR8及びVenus-PR8-PB2-E712DのNSセグメントを誘導した。ゆえに、WT-Venus-PR8又はVenus-PR8-PB2-E712Dが、そのNSセグメントにおいて突然変異プロモータ配列を有する可能性は低い。したがって、NSセグメントの転写/複製効率性の差は、PB2-E712Dによって引き起こされた可能性がある。その上、ウエスタンブロット法によってNS1タンパク質の発現レベルが確認された(図30E)。NSセグメントの転写/複製効率性の低下のため、WT-Venus-PR8感染細胞におけるNS1タンパク質の発現レベルは、Venus-PR8-PB2-E712D感染細胞におけるNS1タンパク質の発現レベルよりも非常に低かった。対照的に、NPの発現レベルは、ほとんど同じであった。バンド強度に基づいて定量されたNS1/NP比は、WT-Venus-PR8感染細胞において有意に低下した(図30F)。ゆえに、NS1発現の低レベルがWT-Venus-PR8感染細胞におけるIFN-βの高発現をもたらすと思われる。さらに、IFN-βの高発現は、他の因子が含まれる可能性があるものの、WT-Venus-PR8の弱毒化を引き起こす。
どのようにしてVenus遺伝子の欠失が生じるのかについて探究するために、連続的継代後にVenus発現を喪失したWT-Venus-PR8におけるNSセグメントの配列を決定した。独立して継代を行った3つのWT-Venus-PR8ウイルスストックを用いてプラークアッセイを行い、大部分のプラークがVenus陰性であることが分かった。各ストックに由来する5超のプラークを配列決定し、各ウイルスストックにおいて1又は2つの欠失パターンがあることが分かった。NSセグメントにおいて大きな欠失が生じ、Venus配列の大部分が喪失した(図31A)。しかし、ヌクレオチド欠失の数、(1又は複数の)欠失の部位、又は欠失が生じた特異的配列などの欠失に関する特異的パターンは同定されなかった。前記大きな欠失は、不完全な干渉ウイルスRNA産生の知られた機序であるポリメラーゼ跳躍(Davisら、1980;Jenningsら、1983)又はウイルスゲノムの再配列によるRNAウイルス適応において役割を果たす遺伝子組換え(Xiaoら、2016;Simonら、2011;Mitanulら、2000;Khatchikianら、1989)によって引き起こされた内部欠失から生じたと仮定された。WT-Venus-PR8のNSセグメントの3’領域又は5’領域に同義突然変異を導入し(図31B)、次いで、これらの突然変異体ウイルスの各々を0.001又は5のMOIでMDCK細胞に感染させた後、それぞれ感染の2日後又は8時間後に上清を回収した。次いで、MDCK細胞と共に上清をインキュベートした。Venus陰性プラークを採取し、MDCK細胞において増幅し、次いで、我々は、Venus発現を喪失したウイルスにおけるNSセグメント配列を決定した。我々は、トランケートされたNSセグメントが一方の側だけにおいて同義突然変異を有したことを見出した(図31C)。両側において同義突然変異を有するか又は低MOI同時感染及び高MOI同時感染の両方における同義突然変異を有さないNSセグメントを有したウイルスは見出されなかった。この結果は、NSセグメントにおける大きな欠失が、各NSセグメントにおける内部欠失から生じたものであって、NSセグメント間の遺伝子組換えから生じたものではないことを示す。
Venusの欠失及び安定化の機序をさらに理解するために、Venus遺伝子を安定化するポリメラーゼ複合体において付加的突然変異を試みた。MDCK細胞に0.001のMOIでWT-Venus-PR8を感染させ、次いで、Venus陽性プラークを採取し、MDCK細胞において増幅させた。Venus陽性ウイルスの連続的継代及びプラーク精製の後、増強されたVenus蛍光を安定して発現した突然変異体を得た。配列分析によって、突然変異が各突然変異体のポリメラーゼ遺伝子PB2、PB1及びPAに導入されたことを示した(図32A)。PA-180及びPA-200は、PB2-712のようにポリメラーゼ複合体の表面上に位置するが、一方でPB2-540、PB1-149及びPB1-684は、前記複合体の内部に位置する。PA-180及びPA-200は、エンドヌクレアーゼドメイン内に位置し、PB1-149及びPB1-684は、RNA鋳型の出口の近くに位置し、PB2-540は、新しく合成されたRNA産物の出口の近くに位置するが(図32B)、PB2-712周辺の領域の機能は不明なままであった(Reichら、2014;Pflugら、2017;Gerlachら、2015)。これらの突然変異がVenus遺伝子の安定化に寄与するかどうかを決定するために、リバースジェネティクスを用いることによって突然変異の各々を含む突然変異体ウイルスを生成し、MDCK細胞における4回の継代の後でVenus保持比を測定した(図32C)。突然変異体ウイルスは、WT-Venus-PR8と比較して増強されたVenus安定性を示したが、このことは、これらのアミノ酸がVenus遺伝子の安定化において役割を果たすことを示す。PB2-540、PB1-149及びPB1-684が、鋳型及び産物が転写/複製反応の間に通るポリメラーゼ内部トンネルの近くに位置することを考慮すると(Reichら、2014;Pflugら、2017;Gerlachら、2015)、どのようにしてこれらのアミノ酸がVenus遺伝子の安定性に寄与するのかについて明らかにするためにさらなる分析が必要であるものの、これらのアミノ酸は、RNA鋳型、産物及びポリメラーゼ複合体の結合安定性に影響を及ぼす可能性がある。これらの突然変異がInfluenza Research Databaseにおいて以前に分離されたインフルエンザA型ウイルスにおいて見出されたかどうかを我々が検討した際(図32D)、我々は、これらのアミノ酸が極めてまれであることを見出したが、このことは、それらが進化的に有利ではないことを示唆する。
外来遺伝子を発現する組換えインフルエンザウイルスは、有用なツールであるが、しかし、ウイルスゲノムにおける長い挿入は、多くの場合不安定であり、組換えウイルスの弱毒化を引き起こす。我々は、インフルエンザウイルスポリメラーゼ複合体におけるアミノ酸が外来遺伝子挿入の安定化において重要な役割を果たすことを以前に見出したが、NSセグメントに挿入されるVenus遺伝子は、H1N1ウイルスにおけるPB2-E712D(Fukuyamaら、2015;Katsuraら、2016)及びH5N1ウイルスにおけるPB2-V25A及びPA-R443K(Zhaoら、2016)によって安定化された。しかし、これらのアミノ酸が前記安定化に寄与する機序は不明なままだった。本研究において、我々は、H1N1ウイルスのNSセグメントに挿入されたVenus遺伝子のPB2-E712D誘導安定化の機序を探究した。修飾セグメントの転写/複製効率性は、Venus-PR8-PB2-E712Dと比較してWT-Venus-PR8において有意に低下することが分かった。この知見は、PB2-E712D突然変異が、修飾RNAセグメントの転写/複製効率性の増強のために、挿入された外来遺伝子を安定化することを示唆する。対照的に、付加配列を含まないセグメントの転写/複製効率性は、PB2-E712D突然変異の有無にかかわらず変化しない。その上、ポリメラーゼ活性は、ミニレプリコンアッセイにおいて、PB2-E712D突然変異によって低下し、増強されない(Katsuraら、2016)。これらの結果は、PB2-E712Dによって引き起こされる転写/複製効率性の変化が修飾RNAセグメントに対して特異的であることを示す。外来遺伝子の挿入は、修飾セグメントの転写/複製を損なうと思われ、ポリメラーゼは、PB2-E712D突然変異の存在下でこの障害を克服する。

- PAウイルスセグメント、PB1ウイルスセグメント、PB2ウイルスセグメント、NPウイルスセグメント、NSウイルスセグメント、Mウイルスセグメント、NAウイルスセグメント及びHAウイルスセグメントを有する分離された組換えインフルエンザウイルスであって、前記ウイルスセグメントの内の少なくとも1つが、アスパラギンではない540位における残基を有するPB2をコードするPB2ウイルスセグメント、グルタミンではない180位における残基若しくはスレオニンではない200位における残基を有するPAをコードするPAウイルスセグメント、又はバリンではない149位における残基、グルタミン酸ではない684位における残基若しくはアスパラギン酸ではない685位における残基を有するPB1をコードするPB1ウイルスセグメント、又はそれらの任意の組み合わせであり、前記組換えインフルエンザウイルスが、アスパラギンであるPB2における540位における残基、グルタミンであるPAにおける180位における残基、スレオニンであるPAにおける200位における残基、バリンであるPB1における149位における残基、グルタミン酸であるPB1における684位における残基又はアスパラギン酸であるPB1における685位における残基を有する対応する組換えインフルエンザウイルスと比較して、安定性が増強されており、且つ/又は複製が増強されている、分離された組換えインフルエンザウイルス。
- PAウイルスセグメント、PB1ウイルスセグメント、PB2ウイルスセグメント、NPウイルスセグメント、NSウイルスセグメント、Mウイルスセグメント、NAウイルスセグメント及びHAウイルスセグメントを有する分離された組換えインフルエンザウイルスであって、前記ウイルスセグメントの内の少なくとも1つが、アスパラギンではない540位における残基又はグルタミン酸ではない712位における残基を有するPB2をコードするPB2ウイルスセグメントであり、前記他のウイルスセグメントの内の少なくとも1つが、グルタミンではない180位における残基若しくはスレオニンではない200位における残基を有するPAをコードするPAウイルスセグメント、又はバリンではない149位における残基、グルタミン酸ではない684位における残基若しくはアスパラギン酸ではない685位における残基を有するPB1をコードするPB1ウイルスセグメント、又はそれらの任意の組み合わせであり、前記組換えインフルエンザウイルスが、アスパラギンであるPB2における540位における残基、グルタミン酸である712位におけるPB2における残基、グルタミンであるPAにおける180位における残基、スレオニンであるPAにおける200位における残基、バリンであるPB1における149位における残基、グルタミン酸であるPB1における684位における残基又はアスパラギン酸であるPB1における685位における残基を有する対応する組換えインフルエンザウイルスと比較して、安定性が増強されており、且つ/又は複製が増強されている、分離された組換えインフルエンザウイルス。
- PAウイルスセグメント、PB1ウイルスセグメント、PB2ウイルスセグメント、NPウイルスセグメント、NSウイルスセグメント、Mウイルスセグメント、NAウイルスセグメント及びHAウイルスセグメントを有する分離された組換えインフルエンザウイルスであって、前記組換えウイルスが、アスパラギンではない540位における残基若しくはグルタミン酸ではない712位における残基を有するPB2をコードするPB2ウイルスセグメント、グルタミンではない180位における残基若しくはスレオニンではない200位における残基を有するPAをコードするPAウイルスセグメント、又はバリンではない149位における残基、グルタミン酸ではない684位における残基若しくはアスパラギン酸ではない685位における残基を有するPB1をコードするPB1ウイルスセグメント、又はそれらの任意の組み合わせを含む2つ以上のウイルスセグメントを有し、前記組換えインフルエンザウイルスが、アスパラギンであるPB2における540位における残基、グルタミン酸である712位におけるPB2における残基、グルタミンであるPAにおける180位における残基、スレオニンであるPAにおける200位における残基、バリンであるPB1における149位における残基、グルタミン酸であるPB1における684位における残基又はアスパラギン酸であるPB1における685位における残基を有する対応する組換えインフルエンザウイルスと比較して、安定性が増強されており、且つ/又は複製が増強されている、分離された組換えインフルエンザウイルス。
- PB2の540位における前記残基が、K、R、D、E、Q若しくはHであり、PB2の712位における前記残基が、D、N、Q、S、H、T、Y若しくはCであり、PAにおける180位における前記残基が、R、K、D、E、N若しくはHであり、PAにおける200位における前記残基が、A、I、L、C、S、M、F、P、G若しくはVであり、PB1における149位における前記残基が、A、T、I、L、C、S、M、F、P若しくはGであり、684位における前記残基が、D、Q、S、H、T、Y、C、K、R若しくはNであり、又はPB1における685位における前記残基が、E、N、R、H、K、S、T、Y、C若しくはQである、請求項1から3のいずれか一項に記載のウイルス。
- PB2の540位における前記残基が、K、R、H、D、S、H、T、Y若しくはCであり、PB2の712位における前記残基が、D、K、H、R、Q若しくは、Nであり、PAにおける180位における前記残基が、R、K、D、N、S、H、T、Y若しくはHであり、PAにおける200位における前記残基が、A、I、L、G、S、M若しくはVであり、PB1における149位における前記残基が、A、T、I、L、S、M若しくはGであり、684位における前記残基が、D、Q、H、L、R若しくはNであり、又はPB1における685位における前記残基が、E、N、R、H、K若しくはQである、請求項1から3のいずれか一項に記載のウイルス。
