Abstract
Description
出物、特に成分ヘデラコサイドCおよびα−ヘデリン含量が貯蔵期間中、安定である乾燥抽出物を得ることができる方法を提供することにある。
a)一定量のツタの葉を粉砕し、
b)粉砕したツタの葉を熱水蒸気で蒸熱し、
c)抽出剤を加えて抽出し、そして
d)適宜、抽出物を乾燥する、
を含む方法により達成される。
図1aは、抽出物の成分スペクトルに及ぼす薬剤蒸熱処理の影響のグラフを示し、図1bは、ヘデラサポニン(Hedera saponin)の抽出収率に対する薬剤の蒸熱処理効果のグラフを示す。
ヘデラコサイドCのα−ヘデリンに対する割合を調節する根拠は、抽出工程の間に、ヘデラコサイドCのC28位の糖が選択的に脱離することを防止することにある。薬剤バッチを選択する際に、α−ヘデリン低含量の葉のバッチが確実に使用されるように注意を払う必要がある。乾燥薬剤中のα−ヘデリン含量が、0.5%未満であるように指定することが薦められる。
ヘデラコサイドC: 6.37%
α−ヘデリン: 0.85%
この薬剤を出発原料として、夫々の場合、三種類の抽出が以下の抽出プロトコルに従って2回実施された。
予め乾燥し、3×3mmに粉砕した3gの薬剤を熱水蒸気(約100℃)で数秒間、蒸熱処理した。このようにして処理した薬剤を18gの抽出剤(30%エタノール(m/m))で約6時間、予備膨潤させた。得られるミセルから水を除き、残存する薬剤をさらに18gの抽出剤で浸出した。このミセルを真空乾燥オーブン中で乾燥した。
この抽出プロトコルは、蒸熱処理の工程を除き、抽出プロトコル1に相当する。
Claims (19)
- 以下の工程:
a)一定量のツタの葉を粉砕し、
b)粉砕したツタの葉を熱水蒸気で蒸熱し、
c)抽出剤を加えて抽出し、そして
d)適宜、抽出物を乾燥する、
を含むことを特徴とし、抽出剤を使用する、貯蔵安定性を有し少なくともその成分であるヘデラコサイドCおよびα−ヘデリンを含むツタの葉抽出物の製造方法。 - ツタの葉が乾燥薬剤として使用されることを特徴とする、請求項1に記載の製造方法。
- 工程a)において、ツタの葉が、5×5mm以下に粉砕されることを特徴とする、請求項1また2に記載の製造方法。
- 蒸熱が1〜30秒間、行なわれることを特徴とする、請求項1〜3のいずれかに記載の製造方法。
- 蒸熱が1〜10秒間、特に5秒間、行なわれることを特徴とする、請求項4に記載の製造方法。
- 水蒸気による蒸熱が、約90〜140℃で行なわれることを特徴とする、請求項1〜5のいずれかに記載の製造方法。
- 水蒸気による蒸熱が、約100〜約120℃、特に110℃で行なわれることを特徴とする、請求項6に記載の製造方法。
- 使用される抽出剤が、アルコール/水の混合物であることを特徴とする、請求項1〜7のいずれかに記載の製造方法。
- 使用される抽出剤が、30%エタノールであることを特徴とする、請求項8に記載の製造方法。
- 抽出が予備膨潤および浸出により、行なわれることを特徴とする、請求項1〜9のいずれかに記載の製造方法。
- 予備膨潤が約1時間〜約30時間、行なわれることを特徴とする、請求項10に記載の製造方法。
- 予備膨潤が約6時間、行なわれることを特徴とする、請求項11に記載の製造方法。
- 乾燥が薄膜蒸発およびその後の噴霧乾燥により行なわれることを特徴とする、請求項1〜12のいずれかに記載の製造方法。
- 高含量のヘデラコサイドCを含むツタの葉抽出物を製造する、請求項1〜13のいずれかに記載の製造方法。
- 工程b)の前に、少なくともヘデラコサイドCの一部が選択的にα−ヘデリンに変換されることを特徴とする、請求項1〜13のいずれかに記載の製造方法。
- 請求項1〜15のいずれかに記載の製造方法を用いて製造されることを特徴とする、ツタの葉抽出物。
- 医薬を製造するための請求項16に記載の抽出物の使用。
- 医薬が気道疾患の治療のために使用されることを特徴とする、請求項16に記載の使用。
- 請求項16に記載の抽出物を含む医薬。
