Abstract
【解決手段】クロマトグラフィー操作からの溶離液試料中の完全なウイルス粒子の数を測定する方法であって、
a)選択された吸光度においてウイルス粒子の数の標準曲線を作成し;
b)ウイルス粒子を含む試料についてアニオン交換樹脂にてクロマトグラフィーを行い;c)上記の選択された吸光度における段階(b)のクロマトグラフィーをモニターし;
d)段階(c)で得られた吸光度値を段階(a)の標準曲線と比較することによって溶離液のウイルス粒子の総数を測定する

Description
遺伝子療法による疾患の治療は、理論上の事から実際的な領域に移行した。最初の人間の遺伝子療法の試みは、1990年9月に開始され、そして、アデノシン・デアミナーゼ(ADA)遺伝子を、この酵素が欠損する患者のリンパ球に転移させることが含まれた。ADA活性の不足は、免疫欠損を生じさせる。治療遺伝子を罹患した細胞に届けるための幾つかの方法において、ウイルスベクターは、特に有望であった。腫瘍サプレッサー遺伝子は、癌細胞を治療するために研究されている。そのような腫瘍サプレッサー遺伝子を含むウイルスベクターは、潜在的な治療物としてガン療法の分野において評価されている。最近、アデノウイルス・ベクターは、そのような腫瘍サプレッサー遺伝子と他の生体応答調節剤の産生のために有利なベクターとして、特に注意を払われた。ガン遺伝子療法の研究が臨床試験に移るにつれて、ますます多量の精製されたウイルスベクターが必要とされる。そのような試験のために適当な量のベクターを産生する際の一つの問題は、ウイルス粒子が産生される細胞溶菌液からの粒子の精製である。

遺伝子療法の成功した実例における一つの障害は、治療遺伝子を摘出するための精製されたウイルスベクターの入手可能性である。本発明は、治療遺伝子を含むウイルスベクターを含む細胞溶菌液を酵素処理し;第一の樹脂上で、酵素処理された細胞溶菌液をクロマトグラフィーに付し;そして、第一のカラムからの溶出液を、第二の樹脂上でクロマトグラフィーに付すこと:からなる三段階法を使用することにより、治療遺伝子を含むウイルスベクターを、治療の及び/又は予防の用途のために充分に精製することができるという、予想外で驚くべき発見により、前記の問題を解決する。治療遺伝子を有する、結果として得られる精製されたウイルスベクターは、クロマトグラフィー的処理の間及び後に、その感染性を保持し、そして、遺伝子転移を行なうことができる。本発明の精製方法を使用することにより初めて、治療遺伝子を含むクロマトグラフ的に精製されたウイルスベクターは、三日超遠心分離法(three day ultracentrifugation)を使用して精製されたウイル
スベクターと同程度に純粋で且つ活性であることが判った。本発明の精製方法は、精製さ
れたウイルスベクターの品質、一貫性、減少された工程時間、及び多量の試料を加工する能力を包含する、既存の方法を越える幾つかの利点を提供する。更に、この新しい精製スキームは、ビリオンの表面特性に基づくので、この新しい精製スキームは、異なる治療遺伝子で異なる内部DNA構造物を含むビリオンにだけでなく、本発明の教示を使用する他のビリオンに広く適用できる。この破過時間精製方法(break-through purification method)は、遺伝子療法のバルクな商業化において重要な観点である。

d)段階(c)で得られた吸光度値を段階(a)の標準曲線と比較することによって溶離液のウイルス粒子の総数を測定する
ことからなる方法である。
本発明の好ましい態様は、クロマトグラフィー操作がポリスチレン/ジビニルベンゼンコポリマー支持体に共有結合で結合したQAE基をもつアニオン交換樹脂において行われる前記方法である。
したがって、本発明は、遺伝子療法の際に使用するための治療遺伝子を含むウイルスベクターの精製方法に関連する。一つの態様において、本発明は、治療遺伝子を含む組換え体ウイルスベクターの、細胞溶菌液からの精製方法であって、a)非被包性(unencapsulated)のDNAとRNAの両方を選択的に分解する酵素的試薬を用いて前記溶菌液を処理し;b)第一の樹脂上で、工程a)からの処理された溶菌液をクロマトグラフィーに付し;次いでc)第二の樹脂上で、工程b)からの溶出液をクロマトグラフィーに付し;この場合、一方の樹脂はアニオン交換樹脂であり、そして、他方の樹脂は、固定化金属イオンクロマトグラフィー(IMAC;immobilizedmetal ion chromatography)樹脂である、方
法に関連する。
別の関連する態様において、疎水的相互作用クロマトグラフィー(hydrophobic interaction chromatography)樹脂で固定化金属イオン・クロマトグラフィー樹脂を代用してよく、及び/又はカチオン交換樹脂でアニオン交換樹脂を代用してよい。

そのようなDNA構築体は選択された正に作用する成長調節遺伝子、例えば(1)c−myc,c−fos,c−jun,c−myb,c−ras,KcおよびJEからなる群から選択されるが、それに限定されない腫瘍遺伝子または原(プロト)腫瘍遺伝子;(2)表皮成長因子、血小板誘導成長因子、トランスフェリンおよびインシュリンからなる群から選択されるが、それに限定されない成長因子レセプター遺伝子のRNAに結合し、そして破壊するRNA酵素をコードする。

工程1 ACN53標準物質(CsCl−ACN53)
ACN53と命名されている標準組換えアデノウイルス5型はヒトサイトメガウイルスプロモータにより駆動される全長1.4−kbのp53cDNAを持つF1をコードしてい
る配列を持つ5型アデノウィルスから誘導されたベクターである(Wills et al.Human Gene Therapy 5:1079−1088(1994))。ウイ
ルスは、(Laver et al.,Virology 45:598−614(197
1))に記述されている3段階遠心分離法を下記のように変形したものである。感染した細胞を、凍結−解凍の3周期により溶菌しそしてSorvall RC−5B遠心分離機
中、15,000rpmで4℃で10分間にわたり遠心分離した。ペレットを廃棄し、上澄
み液をBenzonaseTMを使用して133U/mL、室温で30分間にわたり処理した。次いで、処理した物質を、10mMTris pH8.1中の1.25g/mLと1.40g/mLのCsCl非連続段階勾配上に層状に置き、Sorvall TST 41−14ロータ中、30,000rpm、10℃で75分間にわたり遠心分離した。各々の管からのウイル
スバンドを集め、1.35g/mLCsCl(10mMTris pH8.1)と混合し、BeckmanVTi50ロータ中、45,000rpm,10℃で遠心分離した。各々の管から
のウイルスバンドを集め、前と同様にして45,000rpmで更に4時間にわたり遠心分
離した。この段階からの最後のウイルスプールを、スクロース2%と2mMMgCl2を
添加したリン酸塩緩衝食塩水(PBS)に対して広範囲に4℃で透析した。精製したウイルスプールをヒト胎芽腎臓293細胞(ATCC(American Type Culture Collection))から購入)に、下記の工程2に記述の通り感染するの
に使用した。
ATCC293(ATCC CRL 1573)細胞をCO2インキュベータ中、1.5Lの下記の組成を持つ培養液中、Cell FactoryTM(Nunc,Ruskilde
,デンマーク)中で培養した〔定義した10%Hycloneウシ血清を含むDME高グルコース培養液、2mMグルタミン(Irvine Scientific,Santa Ana,Califonia),1mMピルビン酸ナトリウム(Irvine Scie
ntific)で補填した。抗生物質は培養液に添加してない。〕。
Cell FactoryTM接種、二日ないし二日半後に、細胞単層の約50−60%が
合流した時に、細胞に新鮮培養液500mLに細胞当り5ないし10感染単位を複数回感染(MOI)で感染した。ウイルスを培養液に添加し、十分に攪拌し、ユニット中の細胞に導入した。
調製(Preparation)1からの細胞単層がCell FactoryTMの表面
からの剥離の徴候を示した時に(普通は感染後3ないし4日)、ゆっくりと出して細胞を採取し、Beckman TJ−6中、1500rpmで5分間にわたり遠心分離した。それらを血清を含まない培養液で一度洗浄し、再度ペレット化し、超遠心分離−誘導標準ウイルスの調製に使用するための25mLの50mMTris緩衝液pH8.0/150mMN
aCl,2mMMgCl2,2%スクロースに再懸濁した。実施例1ないし3を通して使
用する予定の試料は、25mLの50mMHEPES緩衝液pH7.5/150mMNaCl,2mMMgCl2,2%スクロースに再懸濁した。細胞をこの時点で凍結−解凍の3
周期により溶菌した。3回目の周期に続いて、細胞破片を、Beckman TJ−6中
、1500rpm、4℃での5分間にわたる遠心分離により除いた。
SDC−PAGEゲルを実施例3、工程Fに記載したのと同様に、銀染色したゲルに大体同じ量を添加して、実施した。次いでバンドを100%メタノールで予湿潤しそしてTris緩衝した生理食塩水(TBS)中平衡させたPVDF膜に移した。そのゲルもTBS中で平衡させた。タンパク質をBio−Rad半乾燥移動装置を25Vで30分間使用してその膜に移した。次いでその膜を1%カゼイン/0.01%窒化ナトリウムで一夜、4℃又は室温で1時間ブロックし、TBSで3回洗浄した。その膜を一番目の抗体(Cytimmune rabbit IgGa-アデノウイルス5型,Lee Biotechnology Research:SanDiego,CA)5μg/mL(TBS中)と共に
1時間にわたり室温で保温した。一番目の保温に続いて、膜をTBSで3回洗浄し2番目の抗体(Amersham Life Sciences,Arlington Heig
hts,IL)
Horserradishperoxidase conjugated anti−rabbit Ig)1μLストック抗体/1mLTBSに希釈した)と共に室温で1時間保
温した。最後の3回の洗浄をTBSで実施しそして膜をAmersham ECL検出試
薬と共に1分間保温し、暗所でいろいろの時間(いろいろの対照を選択をするために数秒ないし数分)、ハイパーフィルム(Hyperfilm)−ECL(Amersham)に露光し、そしてX線フィルム現像液中で現像した。
このウエスタンブロット分析を使用して、いろいろの精製状態にあるACN53のバンドパターンの間の相違を図7で見ることができる。
この測定のために、濃厚ウイルスをリン酸塩緩衝食塩水(PBS)中、0.1%SDS中
の10中の1に希釈する。試料を1分間渦攪拌し、Ependorfmicrofuge中で14,000rpmで遠心分離し沈澱物を除いた。キュベットの合致する対を、PBS
緩衝液中0.1%SDSを使用して、島津UV160U分光光度計(Shimadzu Scientific Instrument,Columbia,MD.)上にベースライ
ン走査線を記載することによりブランクを決める。次いでSDS処理したウイルス試料を試料キュベット中に置きそして220nmから340nmまで走査した。310ないし320の間に吸光がある場合は、試料は濃過ぎるので更に希釈しそして再測定した。A250
/A280比もこの走査から決め、そしてそれは粒子数を測定するために十分に純粋である
ことを確実にするためには1.2と1.3の間になければならない。もしこの条件が満たされるならば260nmだけにおける吸光度はヴィリオン/mL数を計算するのに使用される。260nmにおける吸光単位当り1.1×1012粒子の変換係数(Maizel et al
.,Virology 36:115−125(1968))
は、粒子数を計算するのに約10%の誤差で使用されていた。アニオン交換クロマトグラフィーだけにかけた試料(即ち、DEAE樹脂を使用する実施例1と2)の典型的な値は1.14ないし1.19の間であった。そのような試料を実施例3に記載したIZACに処すると、その比は1.22ないし1.25の範囲にあった。
感染した細胞の溶菌液は初めは宿主細胞とウイルスの両方の汚染物からなる。
これらの汚染物のあるものは、クロマトグラフィーに先立って除かれる。特に宿主細胞、未カプセル化又は不完全ACN53核酸はBenzonaseTMのようなヌクレアーゼを使用する工程のこの段階で酵素的に分解された。これは、二つの理由により工程の早期に実施されてよい。初期のBenzonaseTM処理はアニオン交換樹脂における収率を良くする。BenzonaseTMを次の工程段階で除いた。その工程の間のBenzonaseの存在は市販のELISAキット(American Internationa
l Chemical,Natick,MD)により測定し得る。
処理した溶菌液の清澄化は、遠心分離よりはむしろろ過により実施された。遅い回転を細胞破片を除くのに使用した(図1)。次に1番目のカラム上に載せるための生成物を調製するためにろ過を使用した。使用したろ過材の型(即ち、組成と孔径)とその効果的な生成物回収は実施例3、工程Aに記載されている定量アニオン交換測定法により評定された。
遠心分離の次は、Gelman SciencesのAcrodiscTM0.8/0.2μm
の2段円筒ろ過器を通過するものであった。ACN53の回収は、ろ過に使用された孔径と膜の型に依存した。ポリスルホン膜、PVDF膜と酢酸セルロース膜を基材とする膜のようなろ過材が使用された。ポリスルホンとPVDFが好ましかった。
この段階での上澄み液を、MgCl22mM、スクロース2%(wt/vol)及びβ
−シクロデキストリン2.5%(wt/vol)にした。BenzonaseTM(Amer
ican International Chemical,Inc.)は最終濃度100単位/mLになるまで添加しそして1時間室温で保温した。処理した物質を、Beckman TJ−6中、3000rpmでの10分間にわたる遠心分離とGelman Scie
ncesのAcrodiscTM0.8/0.2μmのろ過器を通過するろ過により清澄化した。得られた上澄み液を実施例2用に採取した。
全体を通して、DEAEクロマトグラフィーの特性は一貫していることが見出され、そして高濃度溶菌液(3×1012ACN53粒子/mL)による負荷試験は注入された体積と回収されたACN53ピーク領域の間で一次応答(linear response)
を示した。一次塩勾配(linear salt gradient)の導入によるDEAEカラムの溶離は3個の主要なピークを示した。これらピークの一番目は約0.5のA260/A280比を持つタンパク質ピークであった。次はACN53ピーク(A260/A280=1.23
)であり、それに続いて、勾配(グラジエント)の末端にDNA(A260/A280=2)ピークがあった。
同じピークが、HEPES、Tris緩衝液又はリン酸塩緩衝系のいずれでも得られた。pHは変更できるが、HEPESpH7.5/NaCl/スクロース/MgCl2を使
用してpH7.5で操作するとより汚染の少ない物質がカラムに吸着される。DEAEクロ
マトグラフィーは高程度の初期精製をもたらす。不動化金属イオン精製クロマトグラフィー段階はこれらの汚染物を除く。
カラム樹脂をその分離性能について、6.6×50mm(1.7mL)の硼珪酸OmnifitTMカラム(Omnifit Ltd.,ケンブリッジ、イングランド)中で試験した。
カラムはPerSeptiv Biosystems BiocadTM(ケンブリッジ、MA)クロマトグラフィーワークステーションで取り付けた。クロマトグラフィーはオンラインで、pH、電導度と280nmと260nmの二つの波長における光学密度について追跡、調査された。
アニオン交換樹脂は、1mL/分において、50mMHEPES,pH7.5,300m
MNaCl,2mMMgCl2,2%スクロースで平衡化された。50mMTris緩衝
液pH8.0(300mMNaCl,2mMMgCl2,2%スクロースを含む)も使用された。細胞溶菌液を充填した後、吸光度により追跡してベースラインまで洗浄し、溶離は20カラム容積の300−600mM一次NaCl勾配で実施し、0.5mLのフラクション
を採取した。将来の使用のために、次にカラムを2カラム容量の0.5MNaOH,1.5MNaCl洗浄液で清浄にし次に再平衡化した。
適当な対照保持時間を得るために、50mMTrispH8中で平衡化したDEAEカラム上に、2mL/分(350cm/hr)で、CsCl−ACN53を、注入しそして10分間(11.7カラム容積の)0−1.5M一次NaCl勾配で溶離した。1.23のA260/A280が検出された。このフラクション中に存在するタンパク質バンドはスロット−ブロッ
ト(slot−blot)分析上のAd5ポリクロナール抗体と反応した。
幾つかのピークを、感染細胞溶菌液試料をDEAEカラム(図2)に適用した時に分割した。ピークの組成はA260/A280吸光比から推測した。例えば1番目のピークは、0.5
のA260/A280比を持つので主にタンパク質である。三番目のピークは2のA260/A280比を持つので、この物質は核酸であることを暗示している。ACN53ウイルスピークは1.23の比を持って二番目に溶離した。これらのピークの同定は、実験を一部変更しそして各々のピークをSDSゲルに通すことにより確認した。上述の条件を使用する典型的な試験では、1.23±0.8のA260/A280比がウイルスピークの特性であることが見出された。
DEAEクロマトグラフィーの精製能を評価するために、感染しなかったATCC−293細胞溶菌液とCsCl−ACN53の両方をカラムに適用した(図3)。宿主細胞物質の殆どは負荷の間にカラムを通過するか又はACN53の保持時間より早い時間に溶離したというものの、小さいピークがACN53と同じ保持時間に溶離した。これらのデータから、ACN53ピークのウイルス性でない汚染物は、宿主細胞物質からであると期待される。汚染ピーク、即ち図3に示した細胞溶菌液試料中の最後に溶離するピークの検査は、約2のA260/A280nm比であることを明らかにした。これは、そのピークは高い核酸含量を持ちそしてヌクレアーゼ処理により減少又は排除することが可能であることを示している。BenzonaseTM前処理をした又はしないDEAE操作は、その酵素がACNフラクションプール中の宿主細胞からの汚染物の量を減少することを示した。
二価カチオンとして亜鉛原子を使用する(IZAC)固定化金属親和性クロマトグラフィー(IMAC)は、高い生成物の回収率を与え、またロードする前のDEAD画分プールのあらゆる試料操作を必要としなかった。この方法によって除去される不純物は流出分ピーク中に溶出し、また生成物から良く溶解し、シンンプラープール基準に導く。IZACは再生可能で、またDEAEを伴って使用されたとき、SDS−PAGEゲルおよびウエスタンブロッド、A260/A280比および感染しない粒子に対する感染する粒子の比によって特定される、純粋なACN53を得ることが可能である2カラム精製プロトコルを与える。要約された全体のプロトコルは以下のようである。

固定化亜鉛親和性クロマトグラフィー(IZAC)系を、それぞれの段階後の水でのフラッシングを伴う100mM EDTA1体積部および0.2M NaOH1体積部で連続してカラムを洗浄することにより、金属チャージすることに備えた。マトリクスを続いて亜鉛原子で、0.5μL/mL氷酢酸で酸性化されたH2O中の100mM ZnCl2の

ACN53の金属親和性カラムとの相互作用は金属特有の性質(亜鉛原子が好まれる)を示し、チャージされていないカラム(亜鉛原子で前ロードされていないカラム)へのCsCl−ACN53の注入は生成物ピークの流出分へのシフトの結果となった。実施例2画分プールからのIZACで精製されたACN53−DEAEを図5に示す。IZAC画分プールの分析は49ないし65%の収率および1.22ないし1.25のA260/A280比を生じた。CsCl−ACN53、DEAE精製およびDEAE/IZAC精製された物質のゲルおよびウエスタンブロット比較を図6および7に見ることができる。CsCl−ACN53およびDEAE/IZAC物質はとても類似しており、またDEAEのみで精製された物質はこれらの基準よりも純粋でない。
異なるIZACバッファおよび溶出系の効果を、DEAE−ACN53プールを半分に分けそして両方の半分をIZACでHEPES pH7.5およびTrispH8バッフ
ァ系中で精製することによって評価した。IZACはまた分離に影響することのないスクロースおよびMgCl2の存在中でも行った。金属イオンとしての銅および溶出試薬とし
てのイミダゾールの使用もまた試験した(金属親和性クロマトグラフィーの一般的な批評について、Belew et.al.1987、Kato et.al.1986を見よ。)。様々な系、亜鉛/グリ
シン、亜鉛/イミダゾール、銅/グリシンおよび銅/イミダゾールの全てが金属親和性精製カラムで作用した。ACN53をpH7からpH9への勾配液を使用して溶出することができる。
1mL Resource Q(Pharmacia,Piscataway,NJ)アニオ
ン交換カラムを、様々な試料中のウイルス粒子の数を定量するために使用した。カラムを300mM NaCl、50mM HEPES、pH7.5中で1mL/分の流速で、717プラスオートサンプラーおよび991発光ダイオード配列検出器(PDA)を備えたWaters625クロマトグラフィー系で平衡化した。クロマトグラフィーは210から
300nmへ走査するPDA検出器(Milford,MA)で監視した。標準線を、選択された
吸光度、0.1%SDS中のA260で全粒子について評価された、CsCl精製されたA
CN53ビリオンを注入することによって作成した。
アッセイは注入された試料の体積と無関係であった。試料をロードした後、カラムを平衡化バッファの2カラム体積、続いて50mM HEPES、pH7.
5中の300から600mMへのNaCl線形勾配液10カラム体積以上で洗浄した。勾配液の次に50mM HEPES、pH7.5中の600mM NaClでの2カラム体積の洗浄を行った。
それぞれのクロマトグラフィー実行の後に、カラムを50mM HEPES、pH7.
5中の1.5M NaClの2.6カラム体積で清浄し、そしてその後次の注入のために
再平衡化した。カラムは粗試料の注入後は0.5N NaOHの0.25ないし1カラム
体積の注入、続く1.5M 塩での洗浄によって、より強く清浄した。NaOHの注入お
よびその後の勾配液の流出は清浄を達成するための便利な方法であった。アデノウイルス粒子の存在の全数を測定することにおける前記アッセイの使用を以下の表1に説明する。表1:全ACN53粒子に基づいた収率および精製データ

バッファ条件の変化を7.0ないし8.0のpH範囲における本研究のカラムクロマトグラフィーによるACN53の精製に使用した(例えば、Seth,Virology 68:1204-1206
(1994))。様々なpH限界での崩壊についての調査を、崩壊についてTCID50(後述の方法C)および分析的アニオン交換分析(上述の方法A)によってアッセイすることによって行う。ACN53の生存率についてのバッファ塩濃度の効果は、イオン強度の急激な変化はウイルスの損失を導くことになることを示した。塩濃度は注意深く制御および監視されるべきである。
これらの実施例において教示される精製法の前、間および後の感染するアデノウイルスType5粒子の定量を、終点タイターアッセイ(組織培養感染率−50%、略号TCID50)によって行う(Philipson,Virology15: 263-268 (1961)を見よ。)。試薬、物質
リストおよび説明はChemicon International,Inc(cat.#3130:"Adenovirus Direct Immunofluorescence Assayi",Temecula,CA)から用いることができる。
ATCC293細胞を96穴プレートに、完全MEM(10%子牛血清、1%グルタミン)培養液中にそれぞれの穴について100μLの5×105細胞/mLをプレートする
。個々のプレートにおいて、1:106に希釈されたウイルス試料の250μLのアリコ
ートを第一のカラムに添加し、そしてプレートにわたり連続して2倍に希釈する。一つの列をポジティブ制御(CsCl精製されたACN53)のために、一つの列をネガティブ制御のために保存する。それぞれの穴の100μLのアリコートをATCC−293種付けプレートにおける同一の位置へ移動し、37℃で加湿インキュベーターにおいて2日間培養した。培養液をその後反転によってデカントし、そして細胞を50%アセトン/50%メタノールの添加で固定した。PBSでの洗浄後、固定された細胞を45分間、キットの説明に従って製造されたFITCラベルされたアンチ−ad5抗体(Chemicon International #5016)と共に培養する。PBSでの洗浄後、プレートを蛍光顕微鏡(490nm励起、520nm放射)下で検査し、そしてラベルの存在について得点をつけた。タイターはTiterprint Analysisプログラム(Lynn,Bio Techniques,12:880-881(1994))を使
用して定量した。本方法に従って行われたアッセイからの結果を以下の表2に説明し、該表は上述の表1からの値を使用して計算された比を包含する。

ウイルス製造物の活性をまたSaos−2細胞、p53−ネガティブ骨肉腫細胞ラインにおけるp53遺伝子生成物の発現についてアッセイすることによって試験した。Saos−2細胞を6穴組織培養プレート中に5×105細胞/穴の濃度で培養液、2mM L−グルタミンおよび10%牛胎児血清を補充されたKaighn's栄養混合物F12:D
ME High Glucose(1:1混合物)3mL中に種付けした。細胞を加湿空気
において、7±1%CO2、37℃で16ないし24時間培養した。使い終った培養液を
除去し、そして1mLの新鮮な培養液で置き換え、そして細胞を精製されたウイルスの20、40、または60MOIで感染させた。1時間の培養後、さらなる2mLの培養液を添加しそして8時間培養させた。細胞をその後一度Dulbecco's−PBSで洗浄
し、そして50mM tris/0.5%Noridet P−40/250mMNaCl/5mM EDTA/5mM NaF/5μg/mL Leupeptinおよび5μg/
mL Aprotinin/2mM PMSFの250μLの添加により溶解した。プレートを氷上で5分間培養し、その後溶解液を個々の1.7mLマイクロ遠心分離チューブに移した。それらを45秒間14000rpmでミクロヒュージ(microfuge)において回
転降下し、そして上清をp53タンパク質の存在についてウエスタンブロット分析によってプライマリーアンチ−p53モノクローナル抗体1801(Vector Laboratories,Burlingame,California)および羊アンチ−マウスIgG−HRP並びにストレプトアビジ
ン(streptavidin)−HRP(Amersham)の1:1混合物でアッセイした。p53タンパク質バンドをAmersham's ECL検出キットを使用して、製造者説明に従って検出した。前記アッセイの使用の結果を図8に示す。p53の発現は、DEAEクロマトグラフィーによって並びにDEAEおよびIZACクロマトグラフィーの両方によってのみ精製されたACN53において見ることができる。(レーン5および6における低いレベルの発現は、これらの試料がDEAE試料よりも低いMOIで行われた事実のためである。)
宿主細胞汚染物をウエスタンブロッド分析によってATCC−293細胞成分に対して発生分化されたポリクローナル抗体を使用してアッセイした。ポリクローナル抗体を商業的に得、そして様々な293種の細胞抗原(HTI,Ramona,CA)に対して育成した。結果
はCsClまたはDEAE−IZAC精製の最終精製物は、検出可能な宿主細胞汚染物を含まないことを示した。半精製DEAE−ACN53の場合、精製プールにおいて見られる少量の免疫性汚染物、3本の主なバンドおよびいくつかの小さいものが存在した。宿主細胞汚染物の大部分はDEAE段階の流出分中に回収される。ACN53でDEAE段階において補助精製された汚染物を亜鉛親和性クロマトクラフィーによって除去し、そしてグリシン勾配液の挿入前にIZAC流出画分中に回収した。
クーマシ−青着色のために、100ないし200μLのアデノウイルス含有試料(約1×1011粒子/mL)を収集し、トリクロロ酢酸沈殿により、または透析および続くスピード−バック(speed-vac)における濃縮により脱塩する。試料をその後SDS−PAG
E還元バッファ(125mM Tris−HCL、pH6.8、20%グリセロール、4
%(w/v)SDS、0.005%ブロモフェノール青、0.5%β−メルカプトエタノール)中で約30μLに再懸濁し、5分間煮沸しそして1mm×10穴Novex 8な
いし16%勾配Tris−Glycine minigelにロードした。試料を1.5
時間140Vで電気泳動した。ゲルをその後40%メタノール/10%酢酸中で30分間固定し、そしてその後クーマシーをPro−Blue着色系(Integrated Separation Systems,Natick MA)で販売者の方法に従って着色した。
銀着色されるゲルに5ないし15μLの試料をロードした。試料を当体積の還元バッファと共に煮沸し、そしてクーマシー検出について記載したように電気泳動した。固定はゲルを10%トリクロロ酢酸中で1時間処理し、続いて18megohmに精製された水中で3回洗浄することにより行った。ゲルはDaiichi銀着色キットで与えられた説明(Integrated Separation Systems)に従って着色された。
ビリオンがTCDID50によって測定されたようにクロマト処理の間にそれらの感染性を
保ち(表1、2)、そしてSaos−2細胞におけるP53タンパク質発現によってアッセイされたように遺伝子移動を生じることができるこを示している。このDEAE−IZAC−ACN53のCsCl−ACN53に対するSDS−PAGEにおける比較は、CsCl−ACN53において存在するより低分子量のタンパク質バンドが存在することを示している。異なるCsCl−ACN53ロットの互いに横に並べた比較は、クロマトグラフィー的に生成された物質がとても良く合致するのに対して、いくらかのバッチ毎の変化性を示している。感染する粒子に対する全体の比および両方の種類の物質の吸収スペクトルは直接比較できる。クロマトグラフィー的に生成されたACN53の純度および活性についての評価は、1日の2カラム方法は3日の超遠心分離プロトコルに置き換わることができることを示している。
ACN53について上述で示されたような好ましい精製計画は、クロマトグラフィー段階の前に感染された細胞溶解液をBenzonaseTMで処理するものである。清浄がその後0.8μmおよび続く0.2μm膜を通るろ過段階によって行われる。もし必要ならば、大孔(例えば、5μm)前ろ過段階を、さらなる粘性懸濁のために追加することができる。pH7.5/300mMNaClへの調整がその後DEAEカラムへのロードのための準備において行われる。A260/A280nm比によって、または特性発光ダイオード配列スペクトルによって検出されるような精製物ピークはプールされ、そして亜鉛−チャージされ等浸透圧に平衡化された金属親和性カラムに直接に注入される。バッファのイオン強度はその後、グリシン勾配液での生成物の溶出の前に、ほぼホスフェートバッファされた食塩水(およそ150mM NaCl)に徐々に低下される。この物質がその後最終配
合物へと透析される。
Claims (2)
- クロマトグラフィー操作からの溶離液試料中の完全なウイルス粒子の数を測定する方法であって、
a)選択された吸光度においてウイルス粒子の数の標準曲線を作成し;
b)ウイルス粒子を含む試料についてアニオン交換樹脂にてクロマトグラフィーを行い;c)上記の選択された吸光度における段階(b)のクロマトグラフィーをモニターし;
d)段階(c)で得られた吸光度値を段階(a)の標準曲線と比較することによって溶離液のウイルス粒子の総数を測定する
ことからなる方法。 - クロマトグラフィー操作がポリスチレン/ジビニルベンゼンコポリマー支持体に共有結合で結合したQAE基をもつアニオン交換樹脂において行われる請求項1に記載の方法。
