Abstract
Description
(1)目的物質を得るまでに多種のカラムを用いなければならず、精製効率が悪い。
(2)分画成分中に目的物質が含まれているかどうかの確認試験を行う必要があることから、精製に多大な時間を要する。
(3)精製時のロスも大きいため多量のサンプルが必要となる。
(1)所望の目的物質が選択的に分離されない。すなわち、リガンドに捕捉される目的物質の他に、希望しない目的物質もカラムに吸着されてしまう。
(2)捕捉効率が低く、多量の液体試料が必要となる。
(1)所望の目的物質が選択的に分離されない。すなわち、リガンドに捕捉される目的物質の他に、希望しない目的物質もアフィニティー粒子に吸着されてしまう。
(2)比重が軽く、アフィニティー粒子を分離することが困難であった。
(3)担体の崩壊が起こり易く、耐久性が低い。
さらに、通常の粒子は塩により凝集する傾向があるため、例えば、血清から目的物を単離したい場合、血清中の各種の塩により凝集し精製効率が低下するが、本発明のアフィニティー粒子は塩の存在下でも凝集が少なく、効率的に目的物質を回収できる。
本発明において、アフィニティー粒子を構成する無機粒子は特に限定されない。無機粒子とは一般に平均粒径20nm〜500μm程度の無機の物体を意味する。具体的な粒子としては、例えば、タルク、カオリン、雲母、絹雲母(セリサイト)、白雲母、金雲母、合成雲母、紅雲母、黒雲母、パーミキュライト、炭酸マグネシウム、炭酸カルシウム、ケイ酸アルミニウム、ケイ酸バリウム、ケイ酸カルシウム、ケイ酸マグネシウム、ケイ酸ストロンチウム、タングステン酸金属塩、マグネシウム、シリカ、ゼオライト、硫酸バリウム、焼成硫酸カルシウム(焼セッコウ)、リン酸カルシウム、弗素アパタイト、ヒドロキシアパタイト、セラミックパウダー、金属石鹸(例えば、ミリスチン酸亜鉛、パルミチン酸カルシウム、ステアリン酸アルミニウム)、窒化ホウ素、酸化セリウム、金コロイド等の無機粒子が挙げられる。
例えば、シリカ、酸化チタン、亜鉛華、アルミナ、酸化鉄、タルク、マイカ、セリサイト、金コロイド等である。
リガンドが結合できる限り限定されない。例えば、共有結合形態では、アミド、エステル、ウレタン、エーテル、2級アミン、尿素結合、ジスルフィド結合などが好ましい。従って、リガンドがこれらの共有結合形態となり得る反応基が好ましく、アミノ基、水酸基、カルボキシル基、チオール基、アルデヒド基等が好ましい。また吸着形態は、アビジン−ビオチン、金属−キレート化合物などが好ましい。従って、リガンドがこれらの吸着形態となり得る吸着基が好ましく、アビジン、ビオチン、キレート化合物等が好ましい。
本発明において、リガンドとは、ある目的物質と特異的に結合する物質であり、各種抗体、抗原、酵素、基質、レセプター、ペプチド、DNA、RNA、アプタマー、プロテインA、プロテインG、アビジン、ビオチン、キレート化合物、各種金属イオンなどである。例えば、各種抗体はIgG、IgM、IgA、IgD、IgE、IgY、抗原は蛋白質、多糖類、酵素はグルタチオン−S−トランスフェラーゼ、基質はグルタチオン、レセプターはホルモンレセプター、サイトカインレセプター、キレート化合物はニトリロ三酢酸、各種金属イオンはNi2+、Co2+、Cu2+、Zn2+、Fe3+である。
式(1)で示されるホスホリルコリン基を無機粒子の表面に共有結合で有し、ある目的物質に特異的に親和性を持つリガンドと結合可能な反応基又は吸着基を無機粒子の表面に共有結合又は吸着で無機粒子の表面に直接的に存在していることが本発明の本質であるので、その製造方法は限定されず、いかなる方法により結合させてもよい。
但し、上述したように、ホスホリルコリン基及びリガンドが結合可能な反応基又は吸着基をあらかじめ有する重合体を用い、化学結合することなく粒子表面を単に被覆する態様は含まない。被覆された重合体が剥れてしまったり、被覆重合体による影響が生じたりするからである。
ステップ1:粒子に下記式(1)で示されるホスホリルコリン基とリガンドが結合可能な反応基又は吸着基を導入する。反応基又は吸着基は限定されないが、アミノ基や水酸基、カルボキシル基、アルデヒド基等である。
ステップ2:粒子に導入した反応基又は吸着基に対して、式(1)で示されるホスホリルコリン基とリガンドとを結合させる。なお、ホスホリルコリン基又はリガンドと反応基又は吸着基の間に存在する化学構造(スペーサー)は任意である。例えば、任意なスペーサーとして、メチレン鎖、オキシエチレン鎖などの他、アミノ基を1つまたは複数含むアルキレン鎖でも良い。
ステップ1:任意の粒子に、公知の方法若しくは今後開発される方法にてアミノ基を導入する。アミノ基は粒子表面に直接的に導入される。アミノ基は一級アミン若しくは二級アミンである。
ステップ2:アミノ基を有する粒子に対し、グリセロホスホリルコリンの酸化的解裂反応により得られたアルデヒド体又はハイドレート体を、還元的アミノ化反応によって、ホスホリルコリン基を粒子表面に直接的に付加させる。
すべてのアミノ基に対してホスホリルコリン基を結合させずに(反応量を調節する)、残存するアミノ基が、リガンドが結合可能な置換基となる。
または、アミノ基を有する粒子に対し、グリセロホスホリルコリンの酸化的解裂反応により得られたカルボキシル体を、アミド化反応によって、ホスホリルコリン基を粒子表面に直接的に付加させる。
すべてのアミノ基に対してホスホリルコリン基を結合させずに(反応量を調節する)、残存するアミノ基が、リガンドが結合可能な置換基となる。
粒子にアミノ基を導入する公知の方法(ステップ1)としては、下記が挙げられる。
1.プラズマ処理の表面反応によるアミノ基の導入
窒素ガス雰囲気下で低温プラズマにより粒子表面にアミノ基を導入する。具体的には粒子をプラズマ反応容器内に収容し、反応容器内を真空ポンプで真空にした後、窒素ガスを導入する。続いてグロー放電により、粒子表面にアミノ基を導入できる。プラズマ処理した無機材料を機械的に粒子化することも可能である。プラズマ処理に関する文献を下記に示す。

アミノ基を有するアルコキシシラン、クロロシラン、シラザンなどの表面改質剤を用いて、シラノール含有粒子等の無機粒子表面を処理する。
例えば、1級アミノ基を有する3−アミノプロピルトリメトキシシランにより、シリカ粒子を処理してアミノ基を導入する。具体的には、シリカを水−2−プロパノール混合液中に浸し、3−アミノプロピルトリメトキシシランを添加後、100℃に加熱し6時間反応させる。室温に冷却後、シリカをメタノールで洗浄し、乾燥してアミノ基がシリカ表面に直接導入された粒子が得られる。好ましく処理される粒子としては、シリカ以外に、ガラス、アルミナ、タルク、クレー、マイカ、アスベスト、酸化チタン、亜鉛華、酸化鉄等の粒子が挙げられる。
粒子表面をまず1.3.5.7−テトラメチルシクロテトラシロキサンにより処理し、表面に導入されたSi−H基と、アミノ基を有するモノマーを反応させてアミノ化された表面を得る。例えば、マイカと1.3.5.7−テトラメチルシクロテトラシロキサンをデシケーター中に入れ、アスピレーターで脱気する。80℃で16時間反応させた後、マイカを取り出し、120℃で乾燥させる。得られたマイカをエタノール中に分散し、アリルアミンを添加、続いて塩化白金酸のエタノール溶液を添加し、60℃で2時間攪拌する。反応終了後、濾過、エタノール洗浄、減圧乾燥してアミノ化マイカを得る。各種無機粒子(マイカ、タルク、カオリン、アルミナ、酸化チタン、酸化亜鉛、酸化鉄、各種無機顔料)が好ましく処理される。
本法に用いるモノマーは、アミン系モノマーを用いることが出来る。アミン系モノマーとは、アリルアミンに限られず、アミノ基及び重合可能なビニル、アクリル等の反応性部位を有していれば良い。アミノ基は、ブトキシカルボニル基、ベンジルオキシカルボニル基などにより保護されていても良い。
また、アミン系モノマーでなくても、エポキシ基のように、例えばジアミンとの反応により簡単にアミノ基を導入可能な官能基を有するモノマーでも良い。
次に、アミノ化された粒子表面にホスホリルコリン基を導入する方法(ステップ2)を以下に示す。
粒子をメタノール中に漬し、ホスファチジルグリセロアルデヒドを添加し、室温で6時間放置する。そして、シアノホウ素酸ナトリウムを0℃で添加、一晩加熱攪拌し、アミノ基にホスホリルコリン基を付加させる。粒子をメタノールで洗浄後、乾燥し、ホスホリルコリン基を表面に直接有する粒子が得られる。反応溶媒はメタノール以外にも水、エタノール、2−プロパノール等プロトン性溶媒であれば使用可能であるが、メタノールを用いた場合の導入率が高い傾向にある。
この方法は、ホスホリルコリン基の導入率が高く、また、様々な無機粒子の表面を修飾できるという大きな利点がある。
n=1〜12までの整数
具体的な方法としては、例えば、式(2)の化合物と塩化チオニルをN,N’−ジメチルホルムアミド中で反応させて酸塩化物とし、N,N’−ジメチルホルムアミド中でアミノ基を有する粒子と反応させ、アミド結合により式(1)で示されるホスホリルコリン基を導入できる。
式(2)の化合物は下記のスキームにより合成できる。
上記反応においては、すべてのアミノ基に対してホスホリルコリン基を結合させずに(反応量を調節する)、残存するアミノ基が、リガンドが結合可能な反応基又は吸着基となる。この粒子が請求項2記載のアフィニティー粒子であり、式(1)で示されるホスホリルコリン基とリガンドが結合可能な反応基又は吸着基とが無機粒子の表面に直接的に存在する粒子となる。そして、この残存するアミノ基にリガンドを結合させたものが、請求項3記載のアフィニティー粒子となり、式(1)で示されるホスホリルコリン基とリガンドとが無機粒子の表面に直接的に存在する粒子となる。
請求項2記載のアフィニティー粒子は、捕捉したい物質(目的物質)に応じて使用者が任意のリガンドを結合させることが可能となる製品態様である。請求項3記載のアフィニティー粒子はあらかじめリガンドを結合させた製品態様である。なお、請求項1記載のアフィニティー粒子は、少なくとも式(1)のホスホリルコリン基が粒子表面に存在しているアフィニティー粒子であり、リガンドやこれを結合できる反応基又は吸着基の有無に限らず、捕捉したい物質(目的物質)に応じて使用者が任意のリガンドを結合させることが出来る製品態様である。また、少なくとも式(1)のホスホリルコリン基が粒子表面に存在している限り、いかなる態様のアフィニティー粒子をも含むものであり、例えば請求項2及び請求項3の態様も含むものである。
なお、このアミノ基に任意の官能基を有する化合物を反応させて、その官能基をリガンドが結合可能な反応基又は吸着基としても良い。例えば、グルタルアルデヒド、アルキルジイミデート、アシルアジド類、イソシアネート類などが考えられる。
リガンドが蛋白質の場合、無機粒子上のアミノ基にグルタルアルデヒドの片方のアルデヒド基を反応させ、もう一方のアルデヒド基に蛋白質中のアミノ基を反応させ、蛋白質を結合させる。
無機粒子の多くは、その表面に水酸基が存在するので、上記のアミノ基のようなリガンドが結合可能な反応基又は吸着基を新たに導入することなく、粒子表面に存在する水酸基(OH)をそのまま利用して、ホスホリルコリン基及びリガンド又はリガンドが結合可能な反応基又は吸着基を導入する。本発明のアフィニティー粒子はこの方法により製造することが好ましい。
粒子表面の水酸基と、下記式(3)或いは(4)の化合物のSi−OMeから脱水によって化学結合を形成させる。この化学反応はほとんどの有機溶媒中で、加熱・還流を行うことで極めて容易に定量的に進行する。この脱水反応によって化学的、物理的に極めて安定なホスホリルコリン基を導入することが出来るので好ましい。なお、下記式(3)または(4)で示されるホスホリルコリン基含有化合物は新規化合物である。
式中、mは2〜6、nは1〜4である。OMeは、OEt、Clであってもよい。またSiと結合するOMeまたはOEtまたはClの内、2つまではメチル基、エチル基、プロピル基、イソプロピル基、ブチル基、イソブチル基でも良い。
下記式(5)に示したホスホリルコリン誘導体を蒸留水に溶解させる。下記式(5)のホスホリルコリン誘導体は公知の化合物であり市販品を入手できる。
また、(3)または(4)中のメトキシ基(OMe)がエトキシ基(OEt)の場合はメタノールをエタノールに変えて反応を行い、Clの場合はジメチルホルムアミドやジメチルスルホキシドに変更するだけでよい。
さらには、Siと結合するOMeまたはOEtまたはClの内、2つまたは1つがメチル基、エチル基、プロピル基、イソプロピル基、ブチル基、イソブチル基のいずれかで置換されている場合も上記の手法と全く同様に製造することができる。
下記式(5)に示したホスホリルコリン誘導体を蒸留水とアセトニトリルの混合液に溶解させる。下記式(5)のホスホリルコリン誘導体は公知の化合物であり市販品を入手できる。
式(5)の化合物の水溶液を氷水浴中で冷却し、過ヨウ素酸ナトリウム及び三塩化ルテニウムを添加し、3時間攪拌した。反応液を減圧濃縮、減圧乾燥し、メタノールにより下記式(7)に示すカルボキシル基を有するホスホリルコリン誘導体を抽出する。
次に、式(7)に塩化チオニルをN,N’−ジメチルホルムアミド中で添加して酸塩化物とし、次いで3−アミノプロピルトリメトキシシランを0.5当量及びトリエチルアミンを2当量添加する。この混合溶液を室温で所定時間撹拌し、式(4)のN,N’−ジメチルホルムアミド溶液を得る。
また、Siと結合するOMeまたはOEtまたはClの内、2つまたは1つがメチル基、エチル基、プロピル基、イソプロピル基、ブチル基、イソブチル基のいずれかで置換されている場合も上記の手法と全く同様に製造することができる。
上記反応においては、すべての水酸基に対してホスホリルコリン基を結合させずに(反応量を調節する)、残存する水酸基が、リガンドが結合可能な反応基又は吸着基となる。この粒子が請求項2記載のアフィニティー粒子であり、式(1)で示されるホスホリルコリン基とリガンドが結合可能な反応基又は吸着基とが無機粒子の表面に直接的に存在する粒子となる。そして、この残存する水酸基にリガンドを結合させたものが、請求項3記載のアフィニティー粒子となり、式(1)で示されるホスホリルコリン基とリガンドとが無機粒子の表面に直接的に存在する粒子となる。
請求項2記載のアフィニティー粒子は、捕捉したい物質(目的物質)に応じて使用者が任意のリガンドを結合させることが可能となる製品態様である。請求項3記載のアフィニティー粒子はあらかじめリガンドを結合させた製品態様である。なお、請求項1記載のアフィニティー粒子は、少なくとも式(1)のホスホリルコリン基が粒子表面に存在しているアフィニティー粒子であり、リガンドやリガンドが結合可能な反応基又は吸着基の有無に限らず、捕捉したい蛋白質(ある目的物質)に応じて使用者が任意のリガンドを結合させることが出来る製品態様である。また、少なくとも式(1)のホスホリルコリン基が粒子表面に存在している限り、いかなる態様のアフィニティー粒子をも含むものであり、例えば請求項2及び請求項3の態様も含むものである。
リガンドが蛋白質の場合、粒子上の水酸基を臭化シアンを用いて活性化する。ここに蛋白質中のアミノ基を反応させ、蛋白質を結合させる。
なお、この水酸基に、任意の官能基を有する化合物を反応させて、その官能基をリガンドが結合可能な反応基又は吸着基としても良い。
ステップ1:任意の粒子に、公知の方法若しくは今後開発される方法にてカルボキシル基を導入する。カルボキシル基は粒子表面に直接的に導入される。
ステップ2:カルボキシル基を有する粒子に対し、下記式(2)で示されるホスホリルコリン含有化合物を常法により反応させて、ホスホリルコリン基を酸アミド結合させ、残存するカルボキシル基をリガンドが結合可能な反応基又は吸着基としても良い。
すべてのカルボキシル基に対してホスホリルコリン基を結合させずに(反応量を調節する)、残存するカルボキシル基が、リガンドが結合可能な反応基又は吸着基となる。
粒子にカルボキシル基を導入する公知の方法(ステップ1)としては、下記が挙げられる。
1.表面改質剤によるカルボキシル基の導入
カルボキシル基を有するアルコキシシラン、クロロシラン、シラザンなどの表面改質剤を用いて、シラノール含有粒子等の無機粒子表面を処理する。
例えば、トリエトキシシリルプロピル無水コハク酸により、シリカ粒子を処理してカルボキシル基を導入する。具体的には、トリエトキシシリルプロピル無水コハク酸をN,N−ジメチルホルムアミドに溶解させ、蒸留水と4−ジメチルアミノピリジンを添加し、16時間室温で撹拌し、下記式(3)に示したカルボン酸を有するシランカップリング剤を得る。本反応は無水コハク酸の4−ジメチルアミノピリジンによる加水分解反応である。
カルボン酸を有するシランカップリング剤により、シリカ粒子を処理してカルボキシル基を導入する。具体的には、シリカを水−2−プロパノール混合液中に浸し、カルボン酸を有するシランカップリング剤を添加後、100℃に加熱し6時間反応させる。室温に冷却後、シリカをメタノールで洗浄し、乾燥してカルボキシル基がシリカ表面に直接導入された粒子が得られる。好ましく処理される粒子としては、シリカ以外に、ガラス、アルミナ、タルク、クレー、マイカ、アスベスト、酸化チタン、亜鉛華、酸化鉄等の粒子が挙げられる。
粒子表面をまず1.3.5.7−テトラメチルシクロテトラシロキサンにより処理し、表面に導入されたSi−H基と、カルボキシル基を有するモノマーを反応させてカルボキシル化された表面を得る。各種無機粒子(マイカ、タルク、カオリン、アルミナ、酸化チタン、酸化亜鉛、酸化鉄、各種無機顔料)が好ましく処理される。
本法に用いるモノマーは、カルボキシル系モノマーを用いることが出来る。カルボキシル系モノマーとは、カルボキシル基及び重合可能なビニル、アクリル等の反応性部位を有していれば良い。
次に、カルボキシル化された粒子表面にホスホリルコリン基を導入する方法(ステップ2)を以下に示す。
カルボキシル基が表面にある粒子をN−ヒドロキシスクシンイミド(NHS)、1−エチル−3−(3−ジメチルアミノプロピル)−カルボジイミドの溶液に浸すと、粒子の表面が活性エステル基で覆われる。ここに式(7)に示すアミノ基を有するホスホリルコリン誘導体溶液を入れ、ホスホリルコリン基を導入する。
上記反応においては、すべてのカルボキシル基に対してホスホリルコリン基を結合させずに(反応量を調節する)、残存するカルボキシル基が、リガンドが結合可能な反応基又は吸着基となる。この粒子が請求項2記載のアフィニティー粒子であり、式(1)で示されるホスホリルコリン基とリガンドが結合可能な反応基又は吸着基とが無機粒子の表面に直接的に存在する粒子となる。そして、このリガンドが結合可能な反応基又は吸着基にリガンドを結合させたものが、請求項3記載のアフィニティー粒子となり、式(1)で示されるホスホリルコリン基とリガンドとが無機粒子の表面に直接的に存在する粒子となる。
請求項2記載のアフィニティー粒子は、捕捉したい物質(目的物質)に応じて使用者が任意のリガンドを結合させることが可能となる製品態様である。請求項3記載のアフィニティー粒子はあらかじめリガンドを結合させた製品態様である。なお、請求項1記載のアフィニティー粒子は、少なくとも式(1)のホスホリルコリン基が粒子表面に存在しているアフィニティー粒子であり、リガンドやリガンドが結合可能な反応基又は吸着基の有無に限らず、捕捉したい物質(目的物質)に応じて使用者が任意のリガンドを結合させることが出来る製品態様である。また、少なくとも式(1)のホスホリルコリン基が粒子表面に存在している限り、いかなる態様のアフィニティー粒子をも含むものであり、例えば請求項2及び請求項3の態様も含むものである。
なお、このカルボキシル基に、任意の官能基を有する化合物を反応させて、その官能基をリガンドが結合可能な反応基又は吸着基としても良い。
リガンドが蛋白質の場合、無機粒子上のカルボキシル基にN−ヒドロキシスクシンイミド(NHS)、1−エチル−3−(3−ジメチルアミノプロピル)−カルボジイミドの溶液に浸し、粒子の表面を活性エステル化する。ここに蛋白質中のアミノ基を反応させ、蛋白質を結合させる。なお、この水酸基に、任意の官能基を有する化合物を反応させて、その官能基を蛋白質が結合可能な反応基又は吸着基としても良い。
上記で得られる本発明のアフィニティー粒子を用いて、本発明の目的物質のアフィニティー分離方法が行われる。
本発明の方法は、無機粒子を利用して、高精度な分離が簡便に行われるという点で画期的な目的物質の分離精製方法である。
本発明の方法は、下記の3つの工程を含むものである。なお、あらかじめリガンドが結合されているアフィニティー粒子の場合は(請求項2)、第1工程はすでに行われているので省略される。
1.式(1)で示されるホスホリルコリン基を無機粒子の表面に共有結合で有するアフィニティー粒子又は式(1)で示されるホスホリルコリン基を無機粒子の表面に共有結合で有し、ある目的物質に特異的に親和性を持つリガンドを無機粒子の表面に共有結合又は吸着で有することを特徴とするアフィニティー粒子に、任意のリガンドを化学結合させる第1工程。
例えば、式(1)で示されるホスホリルコリン基とリガンドが結合可能な反応基又は吸着基とを無機粒子の表面に、前者は共有結合で後者は共有結合又は吸着で有するアフィニティー粒子と任意のリガンドPBS溶液1mlを2mlエッペンチューブに入れて、30分間4℃で緩やかに振る。5000rpm、5分間遠心し、上清を捨てる。洗浄のため、PBS溶液1mlを加えて、緩やかに振り、5000rpm、5分間遠心して、上清を捨てる。この洗浄操作を3回繰り返す。
2.第1工程で製造したアフィニティー粒子を、任意のリガンドにより選択的に捕捉される目的物質を含む液体試料に分散させる第2工程。
例えば、第1工程で製造したアフィニティー粒子を、任意のリガンドにより選択的に捕捉される目的物質を含む液体試料に分散させ、30分間4℃で緩やかに振る。5000rpm、5分間遠心し、上清を捨てる。洗浄のため、PBS溶液1mlを加えて、緩やかに振り、5000rpm、5分間遠心して、上清を捨てる。この洗浄操作を3回繰り返す。
3.分離したアフィニティー粒子から捕捉した目的物質を回収する第3工程。
例えば、アフィニティー粒子から捕捉した目的物質を回収するため、溶出バッファー1mlを加え、30分間4℃で緩やかに振り、粒子から目的物質を溶出させ、上清を回収する。溶出バッファー1mlを加えて、緩やかに振り、5000rpm、5分間遠心して、上清を回収する。この操作を2回繰り返す。
図1は、本発明のアフィニティー粒子と従来のアフィニティー粒子による目的物質捕捉の選択性の違いを示す模式図である。
「ホスホリルコリン基を含有するアルデヒド化合物」
1−α−グリセロホスホリルコリン(6.29g)を蒸留水210mlに溶解し、氷水浴中で冷却した。過ヨウ素酸ナトリウム(10.23g)を添加し、5時間攪拌した。反応液を減圧濃縮、減圧乾燥し、メタノールにより目的物を抽出した。下記化合物(6)に構造を示す。
式(6)の化合物の重水中での1H NMRスペクトルを図2に示す。式(6)の化合物は水中において式(9)と平衡状態なので、実際のスペクトルは式(6)と式(9)の双方を反映したものとなる。
「ホスホリルコリン基を含有するカルボン酸化合物」
200mlフラスコ内に、グリセロホスホリルコリン5g、過ヨウ素酸ナトリウム17g、三塩化ルテニウム・n水和物81mg、および、イオン交換水70%、アセトニトリル30gを加える。室温にて2時間攪拌した後、ろ過し、濾液から溶媒を除去した。得られた固形物からメタノールにて目的化合物を抽出、続いてメタノールを除去することによって目的化合物(7)を得た。
式(7)の化合物の重水中での1H NMRスペクトルを図3に示す。
「式(10)の化合物」
合成例1の化合物5.0gを脱水したメタノール55mLに溶解させ、容器内を乾燥窒素で置換する。次に、化合物1のメタノール溶液に3−アミノプロピルトリメトキシシランを2.84g添加した。この混合溶液を、室温で1晩攪拌したのち、氷冷し、シアノヒドロホウ素化ナトリウム1.39gを添加し、室温に戻して5時間撹拌した。この間も反応容器には乾燥窒素を流し続けた。沈殿をろ過した後、目的物質である下記式(10)を含むメタノール溶液を得た。
「式(11)の化合物」
合成例4の化合物9.0gをN,N’−ジメチルホルムアミド300mlに分散させ、容器内を乾燥窒素で置換する。次に、塩化チオニル4.5gを添加し、15分間攪拌した後、3−アミノプロピルトリメトキシシランを2.84g、トリエチルアミンを9.5g添加した。この混合溶液を、室温で1晩攪拌し、沈殿をろ過した後、目的物質である下記式(11)の化合物を含むN,N’−ジメチルホルムアミド溶液を得た。
「ホスホリルコリン基を無機粒子の表面に共有結合で有するホスホリルコリン粒子(PC粒子(A))」
合成例3で製造した式(10)の化合物50μmolを含むメタノール溶液97.7μlをとり、メタノール47.5ml、蒸留水2.5mlを加え、さらに平均粒子径1.5μm、比表面積が6m2/gのシリカゲルを5g添加した。この粒子分散溶液を80℃で一晩還流させ、カップリングさせた。還流の後メタノールで遠心洗浄し、請求項1のPC粒子を得た(以後、PC粒子(A)と呼ぶ。)。以上の手順で、合成例3の表面改質剤でPC処理した粒子(A)のP定量測定を図4に示す。そこから求めたPC導入量は3.1μmol/g粒子になりPC基が粒子表面に導入されていることを確認した。
「ホスホリルコリン基を無機粒子の表面に共有結合で有するホスホリルコリン粒子(PC粒子(B))」
合成例4で製造した式(11)の化合物50μmolを含むジメチルホルムアミド溶液278μlをとり、ジメチルホルムアミド50mlを加え、さらに平均粒子径1.5μm、比表面積が6m2/gのシリカゲルを5g添加した。この粒子分散溶液を160℃で一晩還流させ、カップリングさせた。還流の後メタノールで遠心洗浄し、請求項1のPC粒子を得た(以後、PC粒子(B)と呼ぶ。)。以上の手順で、合成例4の表面改質剤で処理したPC粒子(B)のP定量測定を図4に示す。そこから求めたPC導入量は3.4μmol/g粒子になりPC基が粒子表面に導入されていることを確認した。
「ホスホリルコリン基を無機粒子の表面に共有結合で有するホスホリルコリン粒子(PC粒子(C))」
合成例4で製造した式(11)の化合物50μmolを含むジメチルホルムアミド溶液278μlをとり、ジメチルホルムアミド47.5ml、蒸留水2.5mlを加え、さらに平均粒子径1.5μm、比表面積が6m2/gのシリカゲルを5g添加した。この粒子分散溶液を160℃で一晩還流させ、カップリングさせた。還流の後メタノールで遠心洗浄し、請求項1のPC粒子を得た(以後、PC粒子(C)と呼ぶ。)。以上の手順で、合成例4の表面改質剤で処理したPC粒子(C)のP定量測定を図4に示す。そこから求めたPC導入量は7.3μmol/g粒子になりPC基が粒子表面に導入されていることを確認した。
参考例1で使用したホスホリルコリン基を導入していない未処理のシリカゲル粒子(未処理粒子と略す)と参考例1、2、3で製造したPC粒子(A)、(B)、(C)をそれぞれ25mgずつとり、蒸留水1ml加えて超音波処理を1分間行った。遠心で蒸留水を取り除き、アルブミン(100μg/ml)或いはリゾチーム(100μg/ml)を1ml加えて室温で1時間反応させ、PBSで遠心・精製(5000g)を5回行い、洗浄した。次にSDS(1%)を1ml加えて室温で1時間反応させ、遠心(5000g)して上清をMICRO BCA法で定量した。その結果を図8に示す。未処理粒子に比べホスホリルコリン基で処理されているPC粒子(A)はアルブミン、リゾチームともに吸着がかなり抑制されていることが分かった。PC粒子(B)、(C)では未処理粒子またはPC粒子(A)に比べ、アルブミン、リゾチームどちらともさらに吸着を抑制していた。
「ホスホリルコリン基とアミノ基を無機粒子の表面に共有結合で有するアフィニティー粒子(Af粒子(A))」
合成例3で製造した式(10)の化合物45μmolを含むメタノール溶液87.9μlと3−アミノプロピルトリメトキシシラン5μmolを含むメタノール溶液50μlをとり、メタノール47.5ml、蒸留水2.5mlを加え、さらに平均粒子径1.5μm、比表面積が6m2/gのシリカゲルを5g添加した。この粒子分散溶液を80℃で一晩還流させ、カップリングさせた。還流の後メタノールで遠心洗浄し、請求項1のアフィニティー粒子を得た(以後、Af粒子(A)と呼ぶ。)。以上の手順で、合成例3の表面改質剤で処理したAf粒子(A)のP定量測定を図4に示す。そこから求めたPC導入量は2.7μmol/g粒子になりPC基が粒子表面に導入されていることを確認した。
「ホスホリルコリン基とアミノ基を無機粒子の表面に共有結合で有するアフィニティー粒子(Af粒子(B))」
合成例4で製造した式(11)の化合物45μmolを含むジメチルホルムアミド溶液250μlと3−アミノプロピルトリメトキシシラン5μmolを含むジメチルホルムアミド溶液50μlをとり、さらに平均粒子径1.5μm、比表面積が6m2/gのシリカゲルを5g添加した。この粒子分散溶液を160℃で一晩還流させ、カップリングさせた。還流の後メタノールで遠心洗浄し、請求項1のアフィニティー粒子を得た(以後、Af粒子(B)と呼ぶ。)。以上の手順で、合成例4の表面改質剤で処理したAf粒子(B)のP定量測定を図4に示す。そこから求めたPC導入量は3.3μmol/g粒子になりPC基が粒子表面に導入されていることを確認した。
「ホスホリルコリン基とアミノ基を無機粒子の表面に共有結合で有するアフィニティー粒子(Af粒子(C))」
合成例4で製造した式(11)の化合物45μmolを含むジメチルホルムアミド溶液250μlと3−アミノプロピルトリメトキシシラン5μmolを含むジメチルホルムアミド溶液50μlをとり、ジメチルホルムアミド47.5ml、蒸留水2.5mlを加え、さらに平均粒子径1.5μm、比表面積が6m2/gのシリカゲルを5g添加した。この粒子分散溶液を160℃で一晩還流させ、カップリングさせた。還流の後メタノールで遠心洗浄し、請求項1のアフィニティー粒子を得た(以後、Af粒子(C)と呼ぶ。)。以上の手順で、合成例4の表面改質剤で処理したAf粒子(C)のP定量測定を図4に示す。そこから求めたPC導入量は6.3μmol/g粒子なりPC基が粒子表面に導入されていることを確認した。
次に、請求項6で示すアフィニティー分離法を示す。実施例1、2、3で作製したAf粒子(A)、(B)、(C)25mgに蒸留水1ml加えて、超音波処理を1分間行った。遠心で蒸留水を取り除き、グルタルアルデヒド溶液(8%)1mlとシッフ塩基の安定化のためシアノトリヒドロホウ酸ナトリウム10mgを加えて室温で5時間反応させ、MQ水で遠心・精製(5000g)5回で洗浄した。グルタルアルデヒドがリガンドが結合可能な反応基又は吸着基である請求項2のアフィニティー粒子を得た。次にウシアルブミン(1mg/ml)或いはヒトヘモグロビン(1mg/ml)1mlとトリヒドロホウ酸ナトリウム10mgを加えて室温で1日反応させ、PBSで遠心・精製(5000g)を4回行った。このウシアルブミン或いはヒトヘモグロビンがリガンドである。ここから、請求項7で示すアフィニティー分離法である。残っているグルタルアルデヒド基を不活性化するためエタノ−ルアミン塩酸塩(0.5M、pH7.1)1mlとトリヒドロホウ酸ナトリウム10mgを加えて室温で1時間反応させ、PBSで遠心・精製(5000g)を4回行い、請求項3のアフィニティー粒子を得た。次にHRP標識抗ウシアルブミン抗体(10μg/ml)或いはHRP標識抗ヒトヘモグロビン抗体(10μg/ml)を1ml加えて、室温で1時間反応させ、PBSで遠心・精製(5000g)を5回行った。さらにPBSを1ml加えて攪拌し、10μlを96穴ウェルプレートに移して基質TMBZを用いて発色試験を行い、450nmで測定を行った。その結果を図9、図10、図11に示す。Af粒子(A)はヒトヘモグロビン−HRP標識抗ヒトヘモグロビン抗体で選択性があった。またAf粒子(B)、Af粒子(C)ではウシアルブミン−HRP標識抗ウシアルブミン抗体、ヒトヘモグロビン−HRP標識抗ヒトヘモグロビン抗体どちらでも選択性があった。
ヒトヘモグロビンに対するヤギ抗血清を用いて選択性試験を行った。実施例1、3で作製したAf粒子(A)、(C)25mgに蒸留水1ml加えて、超音波処理を1分間行った。遠心で蒸留水を取り除き、グルタルアルデヒド溶液(8%)1mlとシッフ塩基の安定化のためシアノトリヒドロホウ酸ナトリウム10mgを加えて室温で5時間反応させ、MQ水で遠心・精製(5000g)5回で洗浄した。グルタルアルデヒドがリガンドが結合可能な反応基又は吸着基である請求項2のアフィニティー粒子を得た。次にヒトヘモグロビン(1mg/ml)1mlとトリヒドロホウ酸ナトリウム10mgを加えて室温で1日反応させ、PBSで遠心・精製(5000g)を4回行った。このヒトヘモグロビンがリガンドである。ここから、請求項7で示すアフィニティー分離法である。残っているグルタルアルデヒド基を不活性化するためエタノ−ルアミン塩酸塩(0.5M、pH7.1)1mlとトリヒドロホウ酸ナトリウム10mgを加えて室温で1時間反応させ、PBSで遠心・精製(5000g)を4回行い、請求項3のアフィニティー粒子を得た。次に100倍希釈したヤギ抗血清を1ml加えて、室温で1時間反応させた。次に遠心(5000g)して上清を得た(上清画分)。PBSで遠心・精製(5000g)を5回行った。次にGly−HClバッファー(0.2M、pH2.5)1mlを加えて室温で1時間反応させて抗ヒトヘモグロビン抗体を溶出し、遠心(5000g)して上清を得た(溶出画分)。この上清画分と溶出画分をSDS−PAGEで流して、銀染色を行った結果を図12に示す。Af粒子(A)、(C)どちらとも、溶出画分では抗体の重鎖のバンドが濃く見られその他にバンドは見られないことから、高選択的に抗体を捕捉している粒子であることがわかった。
抗ヒトヘモグロビンを混合したヤギ抗血清を用いて選択性試験を行った。実施例1、3で作製したAf粒子(A)、(C)25mgに蒸留水1ml加えて、超音波処理を1分間行った。遠心で蒸留水を取り除き、グルタルアルデヒド溶液(8%)1mlとシッフ塩基の安定化のためシアノトリヒドロホウ酸ナトリウム10mgを加えて室温で5時間反応させ、MQ水で遠心・精製(5000g)5回で洗浄した。グルタルアルデヒドがリガンドが結合可能な反応基又は吸着基である請求項2のアフィニティー粒子を得た。次にヒトヘモグロビン(1mg/ml)1mlとトリヒドロホウ酸ナトリウム10mgを加えて室温で1日反応させ、PBSで遠心・精製(5000g)を4回行った。このヒトヘモグロビンがリガンドである。ここから、請求項7で示すアフィニティー分離法である。残っているグルタルアルデヒド基を不活性化するためエタノ−ルアミン塩酸塩(0.5M、pH7.1)1mlとトリヒドロホウ酸ナトリウム10mgを加えて室温で1時間反応させ、PBSで遠心・精製(5000g)を4回行い、請求項3のアフィニティー粒子を得た。次に抗ヒトヘモグロビン50μgを混合した100倍希釈ヤギ血清を1ml加えて、室温で1時間反応させた。次に遠心(5000g)して上清を得た(上清画分)。PBSで遠心・精製(5000g)を5回行った。次にGly−HClバッファー(0.2M、pH2.5)1mlを加えて室温で1時間反応させて抗ヒトヘモグロビン抗体を溶出し、遠心(5000g)して上清を得た(溶出画分)。この上清画分と溶出画分をSDS−PAGEで流して、銀染色を行った結果を図13に示す。Af粒子(A)、(C)どちらとも、溶出画分では抗体の重鎖のバンドが濃く見られその他には薄いバンドしか見られないことから、高選択的に抗体を捕捉している粒子であることがわかった。またバンドの濃さから、抗体捕捉量は10〜20μg程度であった。また溶出画分の抗体活性をサンドイッチELISAで確認したところ、Af粒子(A)13.0μg、Af粒子(C)では10.1μg程度活性があった。
「アミノ基を無機粒子の表面に共有結合で有するアフィニティー粒子(アミノ粒子)」
3−アミノプロピルトリメトキシシラン50μmolを含むメタノール溶液500μlをとり、メタノール47.5ml、蒸留水2.5mlを加え、さらに平均粒子径1.5μm、比表面積が6m2/gのシリカゲルを5g添加した。この粒子分散溶液を80℃で一晩還流させ、カップリングさせた。還流の後メタノールで遠心洗浄し、アミノ粒子を得た。このアミノ粒子25mgに蒸留水1ml加えて、超音波処理を1分間行った。遠心で蒸留水を取り除き、グルタルアルデヒド溶液(8%)1mlとシッフ塩基の安定化のためシアノトリヒドロホウ酸ナトリウム10mgを加えて室温で5時間反応させ、MQ水で遠心・精製(5000g)5回で洗浄した。グルタルアルデヒドがリガンドが結合可能な反応基又は吸着基である。次にウシアルブミン(1mg/ml)或いはヒトヘモグロビン(1mg/ml)1mlとトリヒドロホウ酸ナトリウム10mgを加えて室温で1日反応させ、PBSで遠心・精製(5000g)を4回行った。このウシアルブミン或いはヒトヘモグロビンがリガンドである。残っているグルタルアルデヒド基を不活性化するためエタノ−ルアミン塩酸塩(0.5M、pH7.1)1mlとトリヒドロホウ酸ナトリウム10mgを加えて室温で1時間反応させ、PBSで遠心・精製(5000g)を4回行い、請求項3のアフィニティー粒子を得た。次にHRP標識抗ウシアルブミン抗体(10μg/ml)或いはHRP標識抗ヒトヘモグロビン抗体(10μg/ml)を1ml加えて、室温で1時間反応させ、PBSで遠心・精製(5000g)を5回行った。さらにPBSを1ml加えて攪拌し、10μlを96穴ウェルプレートに移して基質TMBZを用いて発色試験を行い、450nmで測定を行った。その結果を図14に示す。タンパクの非特異吸着が多く、選択性が低い。
Claims (7)
- 前記無機粒子が、シリカ、酸化チタン、亜鉛華、アルミナ、酸化鉄、タルク、マイカ、セリサイト、金コロイドからなる群から選ばれた一種又は二種以上の無機粒子であって、その平均粒子径が20nm〜500μm、比重が1.0g/cm2以上であることを特徴とする請求項1〜3記載のアフィニティー粒子。
- 前記リガンドが各種抗体、抗原、酵素、基質、レセプター、レクチン、ペプチド、DNA、RNA、アプタマー、プロテインA、プロテインG、アビジン、ビオチン、キレート化合物、各種金属イオンからなる群から選ばれた一種又は二種以上のリガンドであることを特徴とする請求項1〜4記載のアフィニティー粒子。
- (1)請求項1または2記載のアフィニティー粒子に任意のリガンドを結合させる第1工程、(2)第1工程で製造したアフィニティー粒子を、任意のリガンドにより選択的に捕捉される目的物質を含む溶液試料に分散させる第2工程、(3)アフィニティー粒子から捕捉した目的物質を回収する第3工程を含むことを特徴とする無機粒子による目的物質のアフィニティー分離方法。
- (1)請求項3記載のアフィニティー粒子を、任意のリガンドにより選択的に捕捉される目的物質を含む溶液試料に分散させる第1工程、(2)アフィニティー粒子から捕捉した目的物質を回収する第2工程を含むことを特徴とする無機粒子による目的物質のアフィニティー分離方法。
