【解決手段】結晶形態にあるプラバスタチンナトリウム塩の製造方法であって、以下の工程:a)低級脂肪族アルコール(例えばエタノール)中にプラバスタチンとナトリウム・カチオンを含有する溶液を用意し;b)上記溶液に酢酸エチルを添加し;c)上記溶液からプラバスタチンのナトリウム塩を結晶化させる、を含む方法。

Description
本発明は、式Iで示される化合物
約5,500種の原核および真核生物株を対象とするスクリーニングプログラムにより、コンパクチンに負けずにそれをヒドロキシル化することができる23種の微生物を選別した。これらの株のなかでは糸状カビが、公開特許で周知となっている株に比してコンパクチン耐性が高くプラバスタチンの生産により適すると判明した。分類学的調査により、この株はMortierella属に属する新たな代表種(Mortierella maculata n.sp.)であると判明した。選別したカビから新株を、一方では突然変異−選択法の適用により、他方では同株のヒドロキシラーゼ酵素の誘導により、単離したが、それは既公開株よりも高濃度でコンパクチン基質をプラバスタチンへとヒドロキシル化することができた。突然変異誘発物質には、物理的および化学的変異原(UV照射、メチルメタンスルホネート、N-メチル-N’-ニトロ-N-ニトロソグアニジン)を使用した。変異誘発処理の後、半数体細胞を作製するために胞子懸濁液をベノミル剤を混ぜた寒天プレート上にまき、次いでヒドロキシラーゼ酵素を誘導するために生育コロニーを100 μg/mlの8-デ-(2-メチル-ブチリル)-コンパクチンまたはコンパクチンを混ぜた寒天プレートに接種した。これらの方法を適用することにより、新株から、親株よりもずっと高度にコンパクチンをプラバスタチンへと変換することができる変異株を作製した。
でんぷん-カゼイン-麦芽エキス-寒天培地では、気中菌糸体がよく発達する(10 μmを越える厚さの被覆層をなして基底菌糸体を覆う)。それは初め、緻密な白色の菌糸網のように見えるが、やがてその中に数mm径の黄色がかった胞子形成点が現れる(新名称のmaculatusはこの状態を指し、「斑点のある」という意味である)。この黄色がかった色合いは気中網のもっと大きな連続した表面を占める場合もある。基底菌糸体の色はCzapek-、血液-Czapek-、チロシン-、でんぷん-カゼイン-、麦芽エキス-などの寒天培地上ではほとんどが無色であるかまたは薄黄色がかっている。基底菌糸体網の色は酵母エキス-グルコース-ペプトン培地上では薄赤みがかっている。拡散性および可溶性色素の産生は前掲培地上では見られないか、またはごくまれにわずかな黄色がった色合いが見られる。E-97株コロニーはその揮発油産生のために、また(Isabellina節の種を除く)他種Mortierellaと同様に、きわめて特徴的な強い芳香を発散させる場合がある。
本発明について以下の実施例との関連でさらに詳しく解説するが実施例を示すのは説明のためであって、本発明の範囲を制限するつもりはまったくない。
コンパクチンを6β位でヒドロキシル化することができるMortierella maculata nov.spec. E-97[NCAIM(P)F 001266]株の7〜10日齢の麦芽エキス−酵母エキス寒天斜面培養物から胞子懸濁液を0.9%塩化ナトリウム溶液5 mlで調整し、懸濁液を500 ml三角フラスコ中の滅菌100 ml接種培地PIに接種した。
培地PIの成分:
1000 ml水道水中
グルコース 50 g
大豆ミール 20 g
滅菌処理前に培地のpHを7.0に調整し、次いで121℃で25分間滅菌処理した。培養物をロータリーシェーカーにより3日間、28℃で振とうし(250 rpm、振幅2.5 cm)、次いで得られた培養物10 mlを500 ml三角フラスコ中の121℃、25分間滅菌処理した100-100 ml生物変換培地MU/4に移した。
培地MU/4の成分:
1000 ml水道水中
グルコース 40 g
大豆ミール 20 g
カゼイン−ペプトン 1 g
アスパラギン 2 g
リン酸二水素カリウム 0.5 g
滅菌処理前に培地のpHを7.0に調整し、次いで121℃で25分間滅菌処理した。
実験室規模の実効内容積5リットルの発酵槽にMU/S生物変換培地を準備し、培地成分は5リットルの容積に合わせて計算するが実際の充填は4.5リットルにとどめ、次いで121℃で45分間滅菌処理し、実施例1に従って調製した接種培養物500 mlをまいた。

実施例3
実施例1に述べた要領で、実験室規模の実効内容積5リットルの発酵槽に生物変換培地MU/4を準備した。実際の充填は4.5リットルにとどめたが、培地成分は5リットルで計算した。次いで121℃で45分間滅菌処理し、実施例1に従って調製した接種培養物500 mlを接種した。発酵は25℃、かく拌速度300 rpm、通気量50リットル毎時の条件で4日間行った。5 gのコンパクチン基質を培養物に供給した後、実施例2に準じて生物変換を行わせた。
10〜12日間培養したMortierella maculata nov.spec. E-97[NCAIM(P)F 001266]株の斜面培養物から胞子懸濁液を0.9%塩化ナトリウム溶液5 mlで調整し、懸濁液を3000 ml三角フラスコ中の滅菌500 ml接種培地VHIGに接種した。
培地VHIGの成分:
1000 ml水道水中
グルコース 30 g
食肉エキス 8 g
酵母エキス 1 g
Tween-80 0.5 g
(ポリオキシエチレン(20)ソルビタンモノオレアート)
滅菌処理前に培地のpHを7.0に調整し、次いで121℃で25分間滅菌処理した。培養物をロータリーシェーカーにより3日間振とうし(250 rpm、振幅2.5 cm)、次いで得られた培養物を、実効内容積5リットルの、生物変換培地PKを入れた実験室規模発酵槽に接種した。

10〜12日間培養したMortierella maculata nov.spec. E-97[NCAIM(P)F 001266]株の斜面培養物から胞子懸濁液を0.9%塩化ナトリウム溶液5 mlで調整し、懸濁液を実施例4で述べた要領で500 mlの接種培地に接種した。実効内容積5リットルの実験室規模発酵槽を使用して、生物変換培地PC/4を121℃で45分間滅菌処理してから、タネ培養物を接種した。
培地PC/4の成分:

麦芽エキス 5.0 %

滅菌処理前に培地のpHを7.0に調整した。接種後、培養と基質の供給を実施例2の要領で行い、次いでプラバスタチン濃度610 μg/mlのブロス5.1リットルを得た。
実施例4で述べた要領で、7〜10日間培養したMortierella maculata nov.spec. E-97[NCAIM(P)F 001266]株の斜面培養物から、3000 ml三角フラスコ中の滅菌500 ml接種培地MIへの接種を行い、ロータリーシェーカー上で3日間、28℃に保持した。
培地MIの成分:

Mortierella maculata n.sp. E-97[NCAIM(P)F 001266]株により、実施例2の要領で発酵、基質の供給および生物変換を行った。生物変換の結果として得られたプラバスタチンをブロスから次の要領で単離した。
自然の生息環境から単離された(コンパクチンを6β位でヒドロキシル化することができる)Mortierella maculata n.sp. E-97株のヒドロキシル化能を、以下に詳述する突然変異−選択および酵素誘導実験で発達させることにより、Mortierella maculata n.sp. E-97/15/13[NCAIM(P)F 001267]変異株を作り出した。
寒天培地MSの成分:
1000 ml蒸留水中
グルコース 4 g
麦芽エキス 10 g
酵母エキス 4 g
寒天 20 g
5 mlの0.9%塩化ナトリウム溶液により斜面培地から胞子を洗い流し、次いで胞子懸濁液を滅菌ペトリ皿に移した後、それに紫外線を1分間照射した。その後N’-メチル-N’-ニトロ-N-ニトロソグアニジンを胞子懸濁液に加え、最終濃度を2000 μg/mlとした。次いで懸濁液を100 ml三角フラスコに移し、28℃、150 rpmで20分間振とうした。その後、胞子を4000 rpm、10分間の遠心分離にかけて沈降させ、次いで滅菌処理した0.9%塩化ナトリウム溶液に懸濁させた。懸濁液を、10 μg/mlのベノミルと1%の脱線維素血液を混ぜた寒天プレートMU-VB上にまいた。
寒天培地MU-VBの成分
990 ml蒸留水中
グルコース 40 g
アスパラギン 2 g
ペプトン 2.5 g
リン酸二水素カリウム 0.5 g
寒天 20 g
この培地は滅菌処理後、10 mlウシ血液と10 mgベノミルを加えて仕上げた。
寒天培地PSの成分
1000 ml蒸留水中
グルコース 40 g
菌類ペプトン 10 g
寒天 15 g
滅菌処理に先立って培地のpHを5.6〜5.7に調整する。滅菌処理は121℃で20分間実施する。
Claims (4)
- 結晶形態におけるプラバスタチンのナトリウム塩。
- その融点が、170℃〜173℃の範囲内にある、請求項1に記載のプラバスタチンのナトリウム塩。
- 結晶形態にあるプラバスタチンのナトリウム塩の製造方法であって、以下の工程:
a) 低級脂肪族アルコール中にプラバスタチンとナトリウム・カチオンを含有する溶液を用意し;
b) 上記溶液に酢酸エチルを添加し;
c) 上記溶液からプラバスタチンのナトリウム塩を結晶化させる、
を含む方法。 - 前記低級脂肪族アルコールが、エタノールである、請求項3に記載の方法。
