- 【特許請求の範囲】 1.遺伝子治療のためにレンチウイルスを基にした複製能欠損ベクター粒子を作 製するためのレトロウイルスベクター作製システムであって、 前記ベクター粒子は哺乳動物の非分裂標的細胞に感染及び形質導入することが でき、 前記システムは前記ベクターの成分をコードする一組の核酸配列を含んでなり 、 HIV-l補助遺伝子vpr,vif,tatおよびnefから、または前記ベクター粒子の基 になるレンチウイルスに通常存在する他のレンチウイルスの類似補助遺伝子から 選択された1以上の機能性遺伝子が前記システムに存在しない、前記レトロウイ ルスベクター作製システム。 2.機能性vprおよびvpu遺伝子が両方とも存在しない場合には、vif,tatおよび nefから選択された1以上の補助遺伝子も存在しないという条件のもとで機能性 補助遺伝子vpuも存在しない、HIV-1を基にしたベクター粒子を作製するための、 請求項1に記載のレトロウイルスベクター作製システム。 3.機能性vprおよびtat遺伝子、またはベクター粒子の基になったレンチウイル スに通常存在する類似遺伝子が両方ともシステムに存在しない、請求項1または 2に記載のレトロウイルスベクター作製システム。 4.機能性補助遺伝子vpr,vif,tat,nefおよびvpuのうち3つ若しくは4つま たは5つすべてがシステムに存在しない、HIVを基にしたベクター粒子を作製す るための請求項1〜3のうちいずれか1項に記載のレトロウイルスベクター作製 システム。 5.ベクターのRNAゲノムをコードする核酸配列が、1以上の治療上活性を有す る遺伝子を含んでなる、請求項1〜4のうちいずれか1項に記載のレトロウイル スベクター作製システム。 6.ベクターの成分をコードした一組の核酸配列が、ベクターのRNAゲノムをコ ードする3つのDNA構築物、gagおよびpolタンパク質並びにenv タンパク質または各々の代替物を包含する、請求項1〜5のうちいずれか1項に 記載のレトロウイルスベクター作製システム。 7.ベクターのRNAゲノムをコードする核酸配列がrevおよびRRE配列または機能 的にそれと同等なものを含んでなり、ゲノムの転写物を核から細胞質へ輸送する ことを可能にする、請求項1〜6のうちいずれか1項に記載のレトロウイルスベ クター作製システム。 8.宿主細胞中の、請求項1〜7のうちいずれか1項に記載のレトロウイルスベ クター作製システム。 9.ベクター粒子の基になったレンチウイルスに通常存在する機能性補助遺伝子 のうち、場合によって存在するrev以外のすべてがシステムに存在しない、請求 項1〜8のうちいずれか1項に記載のレトロウイルスベクター作製システム。 10.パッケージ可能なRNAベクターゲノムをコードし、かつ機能し得るようにプ ロモーターに結合した、請求項9に記載のシステムにおいて使用するためのDNA 構築物。 11.プロモーターが非レトロウイルス性の効率の高いプロモーターである、請求 項10に記載のDNA構築物。 12.請求項10または11に記載のDNA構築物並びにgagおよびpolタンパク質をコー ドするDNA構築物を含んでなる、請求項9に記載のシステムにおいて使用するた めの一組のDNA構築物。 13.envタンパク質またはその代替物をコードするDNA構築物をさらに含んでなる 、請求項12に記載の一組のDNA構築物。 14.1以上の発現ベクター中に、請求項10〜14のうちいずれか1項に記載のDNA 構築物を含んでなる、請求項9に記載のシステムにおいて使用するためのDNA構 築物。 15.請求項1〜9のうちいずれか1項に記載のシステムによって作製されるレト ロウイルスベクター粒子。 16.遺伝子治療のために、標的細胞に感染および形質導入するための請求項15に 記載のレトロウイルスベクター粒子の使用。 17.遺伝子治療に使用するための薬剤の調製における、請求項15に記載のレトロ ウイルスベクター粒子の使用。 18.請求項15に記載のレトロウイルスベクターを用いて標的細胞に感染及び形質 導入することを含んでなる、標的細胞に遺伝子治療を実施するための方法。 19.請求項16〜18のうちいずれか1項に記載の使用および方法の結果得られる標 的細胞。
