Claims (10)
1. Förfarande för framställning av rekombinant-DNA, som innehäller en amylaskodande gen, i vilket förfarande
5 DNA frän en amylasproducerande donormikroorganism av bakteriellt ursprung samt vektor-DNA spjälks och kombineras, de introduceras i en lämplig värd och de sälunda bildade klonerna sovras och väljs pä basis av närvaror av en amylaskodande gen, kanne-t e c k n a t därav, att vektorn är en plasmid eller DNA:n 10 av ett lambda-fagderivat.
2. Förfarande enligt patentkravet 1, kanne- t e c k n a t därav, att det omfattar ett ytterligare steg, i vilket den valda klonens DNA extraheras och spjälks, subklonas i en annan vektor och utväljs igen pä basis av 15 närvaror av en amylaskodande gen, varvid den andra vektorn är en plasmid eller DNA:n av ett annat lambda-fagderivat; och varvid denna subkloning eventuellt följs av en eller flera senare subkloningar.
3. Förfarande enligt patentkravet 1 eller 2, 20 kännetecknat därav, att valet utförs genom odling av klonerna i ett odlingsmedium innehällande stärkelse och genom färgning av mediet med jod genom att bringa det i kontakt med jodänga.
4. Förfarande enligt patentkravet 1 eller 2, 25 kännetecknat därav, att den amylaskodande genen är en pullulanaskodande gen och valet utförs genom odling av klonerna i ett BBL-tryptikasmedium innehällande pullulan, varvid positiva kloner identifieras medelst ogenomskinliga ringar, som omger dem. 30 5. Förfarande enligt patentkravet 1 eller 2, känne tecknat därav, att donormikroorganismen är en stam av släktet Bacillus.
6. Förfarande enligt patentkravet 1 eller 2, kännetecknat därav, att donormikroorganismen är Bacillus 35 megaterium, Bacillus coagulans, Bacillus cereus eller Klebsiella pneumoniae. il
