Claims (10)
- FORBEDRET FREMGANGSMÅDE TIL DYRKNING AF CELLER Patentkrav1. Fremgangsmåde til dyrkning af eukaryote celler i en reaktor i suspension i et celledyrkningsmedium, hvori cellerne producerer et ønsket biologisk stof udvalgt fra proteiner og vacciner, som kan anvendes som et aktivt stof i et farmaceutisk præparat, hvori mindst en celledyrkningsmediekomponent tilføres cellekulturen, og hvori cellekulturen, der omfatter cellerne, det ønskede biologiske stof og celledyrkningsmediet, cirkuleres over et filter ved anvendelse af et tangentialflow, og hvori filteret har en porestørrelse kendetegnet ved et molekylvægt-cutoff på mindst 30 kDa og højst 100 kDa, der er egnet til separering af det ønskede biologiske stof fra stoffer med en lavere molekylvægt end det ønskede biologiske stof, hvori udgangsflowet af væske fra filteret i det væsentlige kun indeholder komponenter med en molekylvægt, der er lavere end det ønskede biologiske stofs, og hvori det ønskede biologiske stof tilbageholdes i eller føres tilbage til reaktoren.
- 2. Fremgangsmåde ifølge krav 1, hvori det ønskede biologiske stof kodes for af mindst ét gen, der er transficeret ind i cellerne.
- 3. Fremgangsmåde ifølge et hvilket som helst af kravene 1-2, hvori det ønskede biologiske stof er et IgG.
- 4. Fremgangsmåde ifølge et hvilket som helst af kravene 1-3, hvori de eukaryote celler er pattedyrsceller.
- 5. Fremgangsmåde ifølge krav 4, hvori pattedyrscelleme er CHO-celler, hybridomer, BHK-celler, myelomceller, humane celler eller museceller.
- 6. Fremgangsmåde ifølge krav 5, hvori de humane celler er HEK-293-celler, humane lymfoblastoide celler eller El-immortaliserede HER-celler
- 7. Fremgangsmåde ifølge krav 5, hvori musecellerne er NSO-celler.
- 8. Fremgangsmåde ifølge krav 5, hvori pattedyrscelleme er CHO-celler.
- 9. Fremgangsmåde ifølge et hvilket som helst af kravene 1-8, hvori cellerne har en cellelevedygtighed på mindst 98 %.
- 10. Fremgangsmåde ifølge et hvilket som helst af kravene 1-9, hvori det ønskede biologiske stof høstes fra cellerne og/eller fra cellekulturen.
