Den foreliggende opfindelse angår hidtil ukendte substituerede 2-phenyl-4-quinolincarboxylsyrer med formlen I,

V^'^L\4)_R· „ * hvor er hydrogen, halogen eller Cj^-alkyl, R2 er hydrogen, halogen, trifluormethyl eller C1_3-alkyl, R3 er hydroxy eller C2-6“alkanoyloxy, R4 er halogen, hydroxy, C^.g-alkyl, 15 trifluormethyl, C3_6-cycloalkyl, phenyl, benzyl, phenoxy, phenylthio, 2,4-dichlorphenoxy eller mono- og di-substitueret phenyl, idet substituenterne er halogen eller C1_3-alkoxy, og R5 er hydrogen eller halogen, med det forbehold, at når Rx er hydrogen, methyl eller brom, R2 er forskellig fra 20 fluor, og R3 er hydroxy, så kan R4 ikke være chlor, brom, iod, methyl eller phenyl, samt en fremgangsmåde til fremstilling af en forbindelse med formlen I, hvilken fremgangsmåde er ejendommelig ved det i krav 4's kendetegnende del angivne.
Det er kendt fra US patentskrift nr. 2.776.290, at 25 forbindelsen 3-hydroxy-2-phenyl-cinchoninsyre (HPC) er godt virksom mod giftfeber, men ikke mod leddegigt. Det har overraskende vist sig, at forbindelserne ifølge opfindelsen udviser en immunundertrykkende virkning, som f.eks. er nyttig ved behandling af leddegigt.
30 Forbindelserne med formlen I kan let fremstilles ifølge det nedenstående reaktionsskema: 35 DK 169473 B1 2 (l) (*) , "XTXX+^-s-Q-r’ *ω R, Λ *«· idet Rlf R2, R4 og R5 er som ovenfor definerede, og R6 er -C2H5 eller -CH2OCO-C1_5-alkyl. Med henvisning til den oven-10 nævnte reaktionsskema blandes en passende substitueret 2,3--indolindion (1) i vandig opløsning, gjort basisk med et alkalimetalhydroxid og opvarmet, med en lavere alkanolopløs-ning af en hensigtsmæssigt substitueret acetophenon (2), og den fremkomne reaktionsblanding holdes ved tilbagesvalings-15 temperatur i adskillige timer. Under denne fremgangsmåde fjernes en portion af den lavere alkanol ved destillering, remanensen opvarmes yderligere ved tilbagesvaling, afkøles derpå og filtreres, og filtratet gøres surt, hvorefter det ønskede produkt I udfældes. Produktet I opsamles derpå ved 20 filtrering og omkrystalliseres om nødvendigt ved gængse fremgangsmåder.
De her omhandlede forbindelser er aktive immununder-trykkende midler, når de indgives til varmblodede dyr. Som sådanne er de effektive ved behandling af tilstande, hvor 25 forhøjede niveauer af antistofproduktion eller monocyt/lym-focyt-aktivitet som et resultat af hyperreaktivitet af det immunregulerende netværk er tæt forbundet med udviklingen af autoimmunitetssygdomme, herunder rheumatoid artritis [Mellbye, O.J. og Natvig, J.B., Clin. Exp. Immunol., 8, 889 30 (1971)], multipel sklerose [Tourtellotte, W.W. og Parker, J.A. Science 154, 1044 (1966)], systemisk lupus erythematosus [Abdu, N.I., et al., Clin. Immunol. Immunopath., 6, 192 (1976)], thyreoiditis [Witebsky, E., et al., J. Immunol., 103, 708 (1969)], blandede bindevævssygdomme [Sharp, G.C., 35 et al., Am. J. Med., 52, 148 (1972)], dermato-/polymyositis [Venables, P.J.W., et al., Ann. Rheum. Dis., 40, 217 (1981)], DK 169473 B1 3 insulin-afhængig diabetes [Charles, M.A., et al., J. Immunol., 130, 1189 (1983)] og hos patienter, der gennemgår organtransplantationer. Den immunundertrykkende aktivitet af forbindelserne ifølge opfindelsen konstateres ved den 5 følgende prøve. Akut qraft-vs.-host-reaktion♦
En akut transplantat-versus-vært-reaktion (graft-vs.-host, GvH) induceres i normale B6D2F1 hanmus ved intravenøs injektion af 30-50 x 106 parentale miltceller fra C57BL/6-forælderen. Ti dage efter GvH-induktion immununder-10 trykkes B6D2Fl-musene akut. På den 10. dag fjernes miltceller fra B6D2Fl-musene aseptisk, anbringes i vævskultur og stimuleres med T6-cellemitogen [Concanavalin-A (Con-A)] med en koncentration på 2 μg pr. ml. Miltcellernes evne til at dele sig som reaktion på mitogenet bestemmes ved impulsmærk-15 ning af celler under deling med 3H-thymidin i de sidste 24 timer af vævskulturperioden på 72 timer. De mærkede celler høstes på "milliporeH-filtre, og mængden af 3H-radioaktivitet kvantificeres med et væske-scintillations-spektrometer. Lægemiddeldosering begynder på den samme dag som GvH-induk-20 tion og fortsætter gennem den 10 dage lange in vivo procedure. Prøveforbindelserne indgives oralt i et phosphatpuffer-bæremedium med en pH-værdi på 7,4 indeholdende 0,025 M phos-phat, 0,075M natriumchlorid og 0,002% "polysorbat 20”. Data fra mus, doserede med lægemiddel, sammenlignes med GvH-mus, 25 doserede med bæremedium, og med normale mus. En forbindelse anses for at være aktiv, hvis den nedsætter graden af undertrykkelse, der ses ved Con-A-celledelingsresponsen for vehikelbehandlede GvH-mus, sammenlignet med normale mus.
Resultaterne af denne prøve med hensyn til typiske 30 forbindelser ifølge opfindelsen er angivet nedenfor.

Desuden er disse forbindelser effektive til behandling af betændelser og ledødelæggelse, knyttet til artritisk sygdom i varmblodede dyr, som det konstateres ved den følgende prøve.
5 Induktion af adiuvans-artritis
Udavlede han-Royalhart-Wistar-rotter (Royalhart Farms,
New Hampton, NY, USA) , der vejer ca. 165 g, får en intradermal injektion i den højre bagpote med dræbt og tørret Mycobacterium tuberculosis, emulgeret i mineralolie (adjuvans) 10 ved en dosis på 2 mg pr. kg legemsvægt. Denne procedure til induktion af artritis er beskrevet i detaljer af A.E. Sloboda and A.C. Osterberg, Inflammation, 1, 415 (1976).
Syv dage efter immunisering med adjuvansen inddeles rotterne i grupper og behandles dagligt ved til mavesækken 15 ved hjælp af en slange at føre forskellige doser af prøveforbindelserne. Kontrolgrupper af rotter immuniseres med adjuvans, men behandles derpå kun med stivelsesbærestof.
Efter 23 dages forløb efter adjuvansimmunisering måles diameterne af den venstre bagpote for alle rotter 20 rundt om ankelleddet med en skydelære.
Resultaterne af denne prøve vedrørende typiske forbindelser ifølge opfindelsen er vist nedenfor.
Den statistiske signifikans for forskelle mellem kontrol- og behandlede grupper beregnes ved anvendelse af 25 Students t-test.

Denne procedure er identisk med det forsøg, hvis resultater er beskrevet i tabel I. Efter 23 dages forløb dræbes rotterne, deres venstre bagpoter amputeres, og der 25 foretages en bedømmelse ved hjælp af røntgenbilleder som følger: røntgenbilleder af led i de venstre bagpoter tages på Polaroid røntgenfilm (type 55) ved anvendelse af en Faxi-tron Røntgenstråleenhed (Model 43805-N, Hewlett Packard, McMinnville, OR, USA). Afstanden mellem fokus og film er 45 30 cm, og eksponeringstiden for røntgenstrålekilden er 5 minutter ved 60KVP. Hvert røntgenbillede klassificeres (blindforsøg) for tilstedeværelsen og alvoren af de følgende parametre: a) juxtaarticulære erosioner af fodrodsknoglerne, og 35 b) bruskmellemrumsindsnævring.
En karakter på 0 til 4 (idet 0 er = normal og 4 er = svære forandringer) tildeles hver af parametrene.
Atter bestemmes den statistiske signifikans mellem artritiske kontrolrotter og artritiske behandlede rotter 40 ved anvendelse af Students t-test. Resultaterne af denne test på repræsentative forbindelser ifølge opfindelsen er angivet i nedenstående tabel.

De her omhandlede forbindelser kan indgives oralt til behandling af artritis eksempelvis med et indifferent fortyndingsmiddel, eller sammen med en assimilerbar spiselig 35 bærer, eller de kan være omsluttet i kapsler med hård eller blød skal, eller de kan være komprimerede til tabletter, eller de kan være inkorporeret direkte sammen med maden i kostplanen. Til oral terapeutisk indgift kan disse aktive forbindelser være inkorporerede sammen med strækkemidler og 40 anvendt eksempelvis i form af tabletter, kapsler, eliksirer, suspensioner og sirupper. Sådanne præparater og tilberedninger bør indeholde mindst 0,1% aktiv forbindelse. Procentdelen af aktiv forbindelse i disse præparater kan naturligvis varieres og kan bekvemt ligge mellem ca. 2% og ca. 60% af 45 vægten af enheden. Mængden af aktiv forbindelse i sådanne terapeutisk anvendelige præparater er sådan, at der opnås 12 UK lOW/B B l en passende dosering. Foretrukne præparater fremstilles således, at en oral dosisenhed indeholder mellem ca. 50 og ca. 250 mg aktiv forbindelse.
Tabletterne, kapslerne osv. kan ligeledes indeholde 5 et bindemiddel, såsom tragant gummi, acacie, majsstivelse eller gelatine, strækkemidler, såsom dicalciumphosphat, et disintegreringsmiddel, såsom majsstivelse, kartoffelstivelse eller alginsyre, et smøremiddel, såsom magnesiumstearat og et sødemiddel, såsom saccharose, lactose eller saccharin.
10 Når dosisenhedsformen er en kapsel, kan den foruden materialer af den ovennævnte type indeholde en flydende bærer, såsom en fed olie.
Forskellige andre materialer kan være tilstede som overtræk eller for på anden måde at modificere dosisenhedens 15 fysiske form. Eksempelvis kan man overtrække tabletter med shellak, sukker eller begge dele. En sirup eller eliksir kan foruden den aktive ingrediens indeholde saccharose som et sødemiddel, methyl- og propylparabener som konserverende stoffer, et farvestof og smags- og aromastof, såsom kirsebær-20 eller appelsinaromastof.
Disse aktive forbindelser kan ligeledes indgives parenteralt. Opløsninger eller suspensioner af disse aktive forbindelser kan fremstilles i vand, passende blandet med et overfladeaktivt middel, såsom hydroxypropylcellulose.
25 Dispersioner kan ligeledes fremstilles i glycerol, flydende polyethylenglycoler og blandinger deraf i olier. Under almindelige betingelser for opbevaring og anvendelse indeholder disse tilberedninger et konserverende middel for at hindre væksten af mikroorganismer.
30 De farmaceutiske former, som er egnede til injicerbar anvendelse, omfatter sterile vandige opløsninger eller dispersioner og sterile pulvere til fremstilling af tilberedninger af sterile injicerbare opløsninger eller dispersioner på bestilling. I alle tilfælde skal formen være steril og 35 være flydende i en sådan udstrækning, at væsken let kan indsprøjtes ved hjælp af en sprøjte. Eksempel 16 3- Hvdroxv-6-methvl-2- Γ 1.1' -biphenvl-4-vl-4-CTuinolincarboxyl- 30 svre
En blanding af 30 g 5-methyl-2,3-indolindion i 200 ml vand og 30,6 g natriumhydroxid i 100 ml vand omsættes med en opløsning af 47,4 g acetoxyacetylbiphenyl i 500 ml ethanol. Blandingen tilbagesvales i 3 timer, og derefter 35 fjernes der 250 ml ethanol ved destillering. En portion vand på 500 ml tilsættes, blandingen omrøres, afkøles til DK 169473 B1 17 stuetemperatur og filtreres gennem diatoméjord. Filtratet gøres surt med 50 ml koncentreret saltsyre og 20 ml iseddike.
Det fremkomne faste stof opsamles, tages op i fortyndet ammoniak, og den uopløselige del opsamles. Dette faste stof 5 opløses i 7,4N ammoniak, filtreres og udfældes med iseddike, hvorved man får 32,0 g af det ønskede produkt, smp. 236-238 °C.
Eksempel 17 10 6,8-Dichlor-3-hydroxv-2- Γ1.11 -biphenvn-4-vl-4-cruinolincar- boxvlsvre
En suspension af 21 g 5,7-dichlor-2,3-indolindion i 120 ml vand behandles med en tilstrækkelig mængde af en opløsning på 16,6 g natriumhydroxid i 55 ml vand til til-15 vejebringelse af en opløsning. En varm opløsning af 25,4 g acetoxyacetylbiphenyl i 350 ml ethanol tilsættes, fulgt af resten af den basiske opløsning. Blandingen tilbagesvales i 2,5 timer. I løbet af den sidste halve time bortdestilleres der 50 ml ethanol. Der tilsættes 300 ml vand, blandingen 20 omrøres, afkøles og filtreres gennem diatoméjord. En suspension af 16,1 g 7-methyl-2,3-indolindion i 120 ml vand behandles med en tilstrækkelig mængde af en opløsning på 16,6 g natriumhydroxid i 55 ml vand til tilvejebringelse af en opløsning. En varm opløsning af 25,4 g 35 acetoxyacetylbiphenyl i 350 ml ethanol tilsættes, fuldt af resten af den basiske opløsning. Blandingen tilbagesvales i DK 169473 B1 18 2,5 timer. En suspension af 6,6 g 5-fluor-2,3-indolindion i 48 ml vand behandles med en tilstrækkelig mængde af en opløsning på 6,62 g natriumhydroxid i 22 ml vand til tilvejebringelse 15 af en opløsning. En varm opløsning af 10,17 g acetoxyacetyl-biphenyl i 80 ml ethanol tilsættes, fulgt af resten af den basiske opløsning. Blandingen tilbagesvales i 3 timer. I løbet af den sidste halve time bortdestilleres der 100 ml ethanol. Der tilsættes en portion vand på 103 ml, blandingen 20 omrøres, afkøles og filtreres gennem diatoméjord. Claims (6)
1. Substituerede 2-phenyl-4-quinolincarboxylsyrer med formlen
5 CO* H R;i/yVrk i1» lo fS, Ra idet Rx er hydrogen, halogen eller C1_3-alkyl, R2 er hydrogen, halogen, trifluormethyl eller C1_3-alkyl, R3 er hydroxy 15 eller C2_6-alkanoyloxy, R4 er trifluormethyl, halogen, hydroxy, C^.g-alkyl, phenyl, benzyl, phenoxy, phenylthio, C3_ g-cycloalkyl, 2,4-dichlorphenoxy eller mono- og disubsti-tueret phenyl, idet substituenterne er halogen eller Cj_3--alkoxy, og R5 er hydrogen eller halogen, med det forbehold, 20 at når R3 er hydrogen, methyl eller brom, R2 er forskellig fra fluor, og R3 er hydroxy, så må R4 ikke være chlor, brom, iod, methyl eller phenyl.
2. Forbindelse ifølge krav 1, kendetegnet ved, at forbindelsen er 6-fluor-3-hydroxy-2-[l,1'-biphenyl]- 25 4-yl-4-quinolincarboxylsyre, 3-(acetyloxy)-2-[l,l'-biphenyl] --4-yl-4-quinolincarboxylsyre eller 6,8-dichlor-3-hydroxy-2-[1,1' -biphenyl]-4-quinolincarboxylsyre.
3. Forbindelse ifølge krav 1, kendetegnet ved, at forbindelsen er 6-fluor-2-(2'-fluor[1,1'-biphenyl]- 30 -3-hydroxy-4-quinolincarboxylsyre, 6,8-dichlor-2-(3,4-di-chlorphenyl) -3-hydroxy-4-guinolincarboxylsyre, 3-(acetyl-oxy) -2 — [ 1,1 · -biphenyl] -4-yl-6-brom-4-guinolincarboxylsyre eller 6-fluor-3-hydroxy-2-(4'-methoxy-[1,1'-biphenyl]-4--yl)-4-quinolincarboxylsyre.
4. Fremgangsmåde til fremstilling af en forbindelse med formlen DK 169473 B1 v^rV._ R, Ri· idet Rlr R2, R3, R4 og R5 er som definerede i krav l, k e n-10 detegnet ved, at man behandler en 2,3-indolindion med formlen -—T=0
