Claims (11)

  1. FREMSTILLING AF POLYUMÆTTEDE FEDTSYRER I OLIEHOLDIG GÆR PATENTKRAV
    1. Fremgangmåde til fremstilling af omega-3-fedtsyrer, hvilken fremgangsmåde omfatter: a) tilvejebringelse af en olieholdig Yarrowia lipolytica, der akkumulerer mindst 25 % af sin tørvægt som olie, og som omfatter gener, der koder for en funktionel, biosyntetisk og heterolog co-3-fedtsyrebane, hvor heterologe gener, der koder for A17-desaturase-, A5-desaturase- og elongaseenzymer af den biosyntetiske co-3-fedtsyrebane, er blevet indført i den olieholdige Yarrowia lipolytica og hvor den olieholdige Yarrowia lipolytica fremstiller substrater til enzymerne; b) dyrkning af gæren fra trin (a) i nærvær af en fermenterbar carbonkilde under forhold, der er egnede til oliefremstilling, hvorved en eller flere ω-3-fedtsyrer fremstilles; og c) eventuel genvinding af en eller flere co-3-fedtsyrer.
  2. 2. Fremgangsmåde ifølge krav 1, hvor de heterologe gener, der koder for en funktionel, biosyntetisk og heterolog ω-3-fedtsyrebane endvidere indbefatter ét eller flere gener, der er udvalgt fra gruppen bestående af: A12-desaturase, Δό-desaturase, A15-desaturase, A9-desaturase og A4-desaturase.
  3. 3. Fremgangsmåde ifølge krav 1, hvor den fermenterbare carbonkilde er udvalgt fra gruppen bestående af monosaccharider, oligosaccharider, polysaccharider, monoglycerider, diglycerider, triglycerider, methanol og carbon-holdige aminer.
  4. 4. Fremgangsmåde ifølge krav 2, hvor det heterologe gen, der koder for A6-desaturasen, er udvalgt fra gruppen bestående af: (a) et isoleret nukleinsyremolekyle, der koder for amino syre sekvensen som vist i SEQ ID NO: 2; og (b) et isoleret nukleinsyremolekyle, der koder for et polypeptid med A6-desaturaseaktivitet, og som hybridiseres med (a) under følgende hybridiseringsforhold: 0,1X SSC, 0,1 % SDS, 65 °C og vasket med 2X SSC, 0,1 % SDS efterfiilgt af 0,1X SSC, 0,1 % SDS.
  5. 5. Fremgangsmåde ifølge krav 1, hvor det heterologe gen, der koder for et elongasepolypeptid, er udvalgt fra gruppen bestående af: (a) et isoleret nukleinsyremolekyle, der koder for amino syre sekvensen som vist i SEQ ID NO: 8; og (b) et isoleret nukleinsyremolekyle, der koder for et polypeptid med elongaseaktivitet, og som hybridiseres med (a) under følgende hybridiseringsforhold: 0,1X SSC, 0,1 % SDS, 65 °C og vasket med 2X SSC, 0,1 % SDS efterfiilgt af 0,1X SSC, 0,1 % SDS.
  6. 6. Fremgangsmåde ifølge krav 1, hvor det heterologe gen, der koder for et A5-desaturasepolypeptid, er udvalgt fra gruppen bestående af: (a) et isoleret nukleinsyremolekyle, der koder for aminosyresekvensen udvalgt fra gruppen bestående af SEQ ID NO: 4, SEQ ID NO: 115, SEQ ID NO: 119 og SEQ ID NO: 123; og (b) et isoleret nukleinsyremolekyle, der koder for et polypeptid med A5-desaturaseaktivitet, og som hybridiseres med (a) under følgende hybridiseringsforhold: 0,1X SSC, 0,1 % SDS, 65 °C og vasket med 2X SSC, 0,1 % SDS efterfulgt af 0,1X SSC, 0,1 % SDS.
  7. 7. Fremgangsmåde ifølge krav 1, hvor det heterologe gen, der koder for et A17-desaturasepolypeptid, er udvalgt fra gruppen bestående af: (a) et isoleret nuklcinsyrcmolckylc, der koder for amino syre sekvensen som vist i SEQ ID NO: 6; og (b) et isoleret nukleinsyremolekyle, der koder for et polypeptid med A17-desaturaseaktivitet, og som hybridiseres med (a) under følgende hybridiseringsforhold: 0,1X SSC, 0,1 % SDS, 65 °C og vasket med 2X SSC, 0,1 % SDS efterfulgt af 0,1X SSC, 0,1 % SDS.
  8. 8. Fremgangsmåde ifølge krav 1, hvor den olieholdige Yarrowia lipolytica fremstiller eicosapentaensyre.
  9. 9. Fremgangsmåde ifølge krav 1, hvor den olieholdige Yarrowia lipolytica fremstiller docosahexaensyre.
  10. 10. Fremgangsmåde ifølge krav 9, hvor den olieholdige Yarrowia lipolytica endvidere omfatter et heterologt gen, der koder for et A4-desaturaseenzym.
  11. 11. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 10, hvor mindst ét af de heterologe gener omfatter et translationsinitieringssted som vist i SEQ ID NO: 126.
