MéTODO PARA A PRODUçãO DE áCIDOS GRAXOS POLIINSATURADOS COM QUATRO OU MAIS LIGAçõES DUPLAS, Saccharomyces cerevisiae GENETICAMENTE MODIFICADO QUE TEM CAPACIDADE DE PRODUZIR áCIDOS GRAXOS POLIINSATURADOS COM QUATRO OU MAIS LIGAçõES DUPLAS QUANDO DESENVOLVIDO EM UM SUBSTRATO QUE NãO DE áCIDO GRAXO, COMPOSIçãO, USO DA COMPOSIçãO E USO DE UM Saccharomyces cerevisiae GENETICAMENTE MODIFICADO A presente invenção refere-se à construção e à engenharia de células, mais particularmente de microorganismos para produzir PUFAs com quatro ou mais ligações duplas a partir de substratos que não de ácido graxo através da expressão heteróloga de uma passagem que requer oxigênio. A invenção envolve especialmente o aumento do teor de PUFA no organismo hospedeiro através da otimização da fermentação, por exemplo, ao diminuir a temperatura e/ou ao projetar um meio mais favorável ou através de aumento do fluxo em direção a ácidos graxos pela engenharia metabólica, por exemplo, através da superexpressão de sintases de ácido graxo, da superexpressão de outras enzimas envolvidas na biossintese do precursor para PUFAs ou da otimização de códons dos genes heterólogos ou da a expressão das enzimas heterólogas envolvidas na biossíntese do precursor para PUFAs.
