- Technisches Gebiet
- Die vorliegende Erfindung betrifft die Verwendung eines Prostaglandinderivats als Wirkstoff bei der Herstellung einer Schlaf-induzierenden Präparation.
- Stand der Technik
- Da Prostaglandin (hiernach bezeichnet als PG) bereits in Spurenmengen verschiedene wichtige physiologische Wirkungen ausübt, wurden die Synthesen der Derivate von natürlichen PGs und ihre biologischen Aktivitäten mit der Absicht einer praktischen Verwendung als Medizin untersucht und darüber wurde in vielen Literaturstellen berichtet.
- Insbesondere wurde berichtet, dass PGs verschiedene zentralnervöse Aktionen aufweisen und es wurde klargestellt, wie ihr intracerebraler Gehalt, ihre Biosynthese, ihr Stoffwechselweg, ihre intracerebralen Lokalisierungen und Veränderungen im Hinblick auf Wachstum oder Alterung aussehen und man interessierte sich für die Beziehung von PGs zu Schlaf- und Wachzuständen. Unter diesen war bekannt, dass PGD2 ein intracerebraler humoraler Faktor ist, der das Auftreten oder den Erhalt des Schlafes kontrolliert und es wurde deutlich, dass der durch PGD2 bei Affen induzierte Schlaf von einem spontanen natürlichen Schlaf im Hinblick auf die Gehirnwellen oder das Verhalten nicht unterscheidbar war (Proc. Natl. Acad. Sci. USA, Band 85, Seiten 4082–4086 (1988)), weshalb erwartet wird, dass diese Verbindung eine Verbindung mit einer neuen, schlafinduzierenden Wirkung ist.
- PGD2-Derivate einschließlich PGD2 sind jedoch zur Zeit aufgrund von Problemen unpraktisch, die ihre Wirkung und Stabilität als Arzneimittel betreffen.
- Die
EP 0 737 676 A betrifft Prostaglandinderivate der folgenden Formel worin R1 Wasserstoff, Alkyl oder Cycloalkyl ist, R2 ist Cycloalkyl, Methylcycloalkyl, Cycloalkylalkyl, verzweigtes Alkyl oder verzweigtes Alkenyl, X ist Halogen, A ist eine Ethylen-, eine Vinylen- oder Ethinylengruppe. Dieses Dokument betrifft die Erniedrigung des intraokulären Drucks als pharmazeutische Wirkung. -

- Die
US 5,599,838 offenbart ein Prostaglandinderivat, repräsentiert durch die folgende Formel worin X ein Halogenatom ist, R1 ist ein Wasserstoffatom oder eine Alkylgruppe mit 1 bis 6 Kohlenstoffatomen und R2 ist eine Cycloalkylgruppe mit 3 bis 10 Kohlenstoffatomen, eine Cycloalkylmethylgruppe mit 4 bis 10 Kohlenstoffatomen oder eine Cycloalkylethylgruppe mit 5 bis 12 Kohlenstoffatomen oder ein Salz davon, das eine erniedrigende Wirkung auf den intraokulären Druck und verbessernde Wirkung auf renale Erkrankungen, ischämische Herzerkrankungen und Herzversagen ausübt. - Offenbarung der Erfindung
- Als Ergebnis intensiver Studien haben die gegenwärtigen Erfinder festgestellt, dass Prostaglandinderivate mit einer Dreifachbindung zwischen den 13- und 14-Positionen, repräsentiert durch die folgende Formel (I) eine charakteristische, den Schlaf induzierende Wirkung aufweisen und dadurch wurde die folgende Erfindung vervollständigt.
- Das heißt, die vorliegende Erfindung betrifft die Verwendung eines Prostaglandinderivats, dargestellt durch Formel (I) zur Herstellung eines Medikaments, das als schlafförderndes Präparat wirksam ist: worin X ein Halogenatom ist, Y eine durch (CH2)m dargestellte Gruppe, eine cis-Vinylengruppe oder eine Phenylengruppe ist, Z eine Ethylengruppe, eine trans-Vinylengruppe, OCH2 oder S(O)nCH2 ist, R1 eine C3-10-Cycloalkylgruppe, eine mit (einer) C1-4-Alkylgruppe(n) substituierte C3-10-Cycloalkylgruppe, eine C4-13-Cycloalkylalkylgruppe, eine C5-10-Alkylgruppe, eine C5-10-Alkenylgruppe, eine C5-10-Alkinylgruppe oder eine verbrückte cyclische Kohlenwasserstoffgruppe ist, R2 ein Wasserstoffatom, eine C1-10-Alkylgruppe oder eine C3-10-Cycloalkylgruppe ist, m eine ganze Zahl von 1 bis 3 ist und n 0, 1 oder 2 ist oder ein pharmazeutisch annehmbares Salz davon.
- Weiterhin betrifft die vorliegende Erfindung die obige Verwendung, worin in dem durch Formel (I) dargestellten Prostaglandinderivat oder einem pharmazeutisch akzeptablen Salz davon X ein Chlor- oder Bromatom ist, Y ist eine Gruppe, repräsentiert durch (CH2)m oder cis-Vinylengruppe, Z ist eine Ethylengruppe, eine trans-Vinylengruppe, OCH2 oder S(O)nCH2, R1 ist eine C3-10-Cycloalkylgruppe oder eine C4-13-Cycloalkylalkylgrupe, R2 ist ein Wasserstoffatom oder eine C1-10-Alkylgruppe, m ist eine ganze Zahl von 1 bis 3 und n ist 0, 1 oder 2.
- Weiterhin betrifft die vorliegende Erfindung die obige Verwendung, worin in dem durch Formel (I) dargestellten Prostaglandinderivat oder einem pharmazeutisch akzeptablen Salz davon X ein Chlor- oder Bromatom ist, Y ist eine Gruppe, dargestellt durch (CH2)m oder eine cis-Vinylengruppe, Z ist OCH2, R1 ist eine C3-10-Cycloalkylgruppe oder eine C4-13-Cycloalkylalkylgruppe, R2 ist ein Wasserstoffatom oder eine C1-10-Alkylgruppe und m ist eine ganze Zahl von 1 bis 3.
- Noch weiterhin betrifft die vorliegende Erfindung die obige Verwendung, worin in dem durch Formel (I) dargestellten Prostaglandinderivat oder einem pharmazeutisch akzeptablen Salz davon X ein Chlor- oder Bromatom ist, Y ist eine Gruppe, repräsentiert durch (CH2)m oder eine cis-Vinylengruppe, Z ist SCH2, R1 ist eine C3-10-Cycloalkylgruppe oder eine C4-13-Cycloalkylalkylgruppe, R2 ist ein Wasserstoffatom oder eine C1-10-Alkylgruppe und m ist eine ganze Zahl von 1 bis 3.
- Weiterhin betrifft die vorliegende Erfindung die obige Verwendung, worin in dem durch Formel (I) dargestellten Prostaglandinderivat oder einem pharmazeutisch akzeptablen Salz davon Y eine Gruppe ist, repräsentiert durch (CH2)m, Z ist SCH2, R1 ist eine C3-10-Cycloalkylgruppe, R2 ist ein Wasserstoffatom oder eine C1-10-Alkylgruppe und m ist eine ganze Zahl von 1 bis 3.
- In der vorliegenden Erfindung bezieht sich Halogenatom auf ein Fluor-, Chlor-, Brom- oder Iodatom.
- Beispiele für die C3-10-Cycloalkylgruppe sind eine Cyclopropylgruppe, eine Cyclobutylgruppe, eine Cyclopentylgruppe, eine Cyclohexylgruppe und eine Cycloheptylgruppe.
- Beispiele für die C3-10-Cycloalkylgruppe, substituiert mit einer oder mehreren C1-4-Alkylgruppen sind eine Methylcyclopropylgruppe, eine Methylcyclohexylgruppe und eine Ethylcyclohexylgruppe.
- Beispiele für die C4-13-Cycloalkylalkylgruppe sind eine Cyclopropylmethylgruppe, eine Cyclobutylmethylgruppe, eine Cyclopentylmethylgruppe, eine Cyclopentylethylgruppe, eine Cyclohexylmethylgruppe, eine Cyclohexylethylgruppe und eine Cycloheptylmethylgruppe.
- Die C5-10-Alkylgruppe bezieht sich auf eine lineare oder verzweigte Alkylgruppe und Beispiele hierfür sind eine Pentylgruppe, eine Hexylgruppe, eine Heptylgruppe, eine Octylgruppe, eine 1-Methylpentylgruppe, eine 2-Methylpentylgruppe, eine 1-Methylhexylgruppe, eine 2-Methylhexylgruppe, eine 2,4-Dimethylpentylgruppe, eine 2-Ethylpentylgruppe, eine 2-Methylheptylgruppe, eine 2-Ethylhexylgruppe, eine 2-Propylpentylgruppe, eine 2-Propylhexylgruppe und eine 2,6-Dimethylheptylgruppe.
- Die C5-10-Alkenylgruppe bezieht sich auf eine lineare oder verzweigte Alkenylgruppe und Beispiele hierfür sind eine 3-Pentenylgruppe, eine 4-Hexenylgruppe, eine 5-Heptenylgruppe, eine 4-Methyl-3-pentenylgruppe, eine 2,4-Dimethylpentenylgruppe, eine 6-Methyl-5-heptenylgruppe und eine 2,6-Dimethyl-5-heptenylgruppe.
- Die C5-10-Alkinylgruppe bezieht sich auf eine lineare oder verzweigte Alkinylgruppe und Beispiele hierfür sind eine 3-Pentinylgruppe, eine 3-Hexinylgruppe, eine 4-Hexinylgruppe, eine 1-Methylpent-3-inylgruppe, eine 2-Methylpent-3-inylgruppe, eine 1-Methylhex-3-inylgruppe und eine 2-Methylhex-3-inylgruppe.
- Beispiele für die verbrückte cyclische Kohlenwasserstoffgruppe sind eine Bornylgruppe, eine Norbornylgruppe, eine Adamantylgruppe, eine Pinanylgruppe, eine Thujylgruppe, eine Carylgruppe und eine Camphanylgruppe.
- Die C1-10-Alkylgruppe für R2 bezieht sich auf eine lineare oder verzweigte Alkylgruppe und Beispiele hierfür sind eine Methylgruppe, eine Ethylgruppe, eine Propylgruppe, eine Isopropylgruppe, eine Butylgruppe, eine Isobutylgruppe, eine tert-Butylgruppe, eine Pentylgruppe, eine Isopentylgruppe, eine 2-Ethylpropylgruppe, eine Hexylgruppe, eine Isohexylgruppe, eine 1-Ethylbutylgruppe, eine Heptylgruppe, eine Isoheptylgruppe, eine Octylgruppe, eine Nonylgruppe und eine Decylgruppe.
- Beispiele für das pharmazeutisch akzeptable Salz sind Salze mit einem Alkalimetall (z.B. Natrium oder Kalium), einem Erdalkalimetall (z.B. Calcium oder Magnesium), Ammoniak, Methylamin, Dimethylamin, Cyclopentylamin, Benzylamin, Piperidin, Monoethanolamin, Diethanolamin, Monomethylmonoethanolamin, Tromethamin, Lysin, ein Tetraalkylammonium und Tris(hydroxymethyl)aminomethan.
- Gemäß der vorliegenden Erfindung ist Y im Hinblick auf die Schlaf-induzierende Wirkung vorzugsweise eine Ethylen- oder eine cis-Vinylengruppe, Z ist vorzugsweise OCH2 oder SCH2 und R1 ist vorzugsweise eine Cycloalkylgruppe in Formel (I) der Prostaglandinderivate als Wirkstoff.
- Einige der Verbindungen der Formel (I) gemäß der vorliegenden Erfindung sind aus der WO 94/02457, WO 94/08959, dem japanischen Patent Kokai Hei-6-192218, dem japanischen Patent Kokai Hei-7-242622, dem japanischen Patent Kokai Hei-7-242623, dem japanischen Patent Kokai Hei-7-233144, dem japanischen Patent Kokai Hei-7-285929, dem japanischen Patent Kokai Hei-8-208599, dem japanischen Patent Kokai Hei-7-233143, dem japanischen Patent Kokai Hei-9-286775, dem japanischen Patent Kokai Sho-58-8059, dem japanischen Patent Kohyo Sho-60-501813 und dem japanischen Patent Kohyo Sho-60-500787 bekannt.
- Andererseits können die Verbindungen, worin Z S(O)CH2 und S(O)2CH2 ist, durch Reaktion der Verbindungen, worin Z SCH2 ist unter Verwendung eines Natriummetaperiodids in einem Lösungsmittel wie Methanol hergestellt werden.
- Gemäß der vorliegenden Erfindung werden repräsentative Verbindungen der Formel (I) wie folgt beschrieben:
- Die in der vorliegenden Erfindung verwendeten Verbindungen können oral oder parenteral, wie z.B. intravenös oder nasal verabreicht werden. Zum Beispiel können sie oral in Form von Tabletten, Streupuder, Körnern, Pulvern, Kapseln, Lösungen, Emulsionen oder Suspensionen verabreicht werden, die jeweils gemäß den konventionellen Verfahren hergestellt werden können. Als Dosierungsformen für die intravenöse Verabreichung werden wässrige oder nicht-wässrige Lösungen, Emulsionen, Suspensionen oder feste Präparationen verwendet, um in einem Lösungsmittel für die Injektion direkt vor der Verwendung gelöst zu werden. Weiterhin kann die nasale Verabreichung durch quantitatives Versprühen einer Lösung oder eines Pulvers (Hartkapseln), enthaltend das Arzneimittel, in die Nasenhöhle durch Verwendung eines dafür bestimmten Nasentropfers oder einer Sprühvorrichtung durchgeführt werden. Die Verbindungen der vorliegenden Erfindung können in Form von Einschlussverbindungen mit einem α-, β- oder γ-Cyclodextrin oder einem methylierten Cyclodextrin formuliert werden. Die Dosis variiert je nach Alter, Körpergewicht usw., liegt jedoch allgemein bei 1 ng bis 1 mg/Tag/Erwachsenem.
- Gewerbliche Anwendbarkeit
- Die vorliegende Erfindung ermöglicht es, eine Schlaf-induzierende Präparation bereitzustellen, die ausreichend effektiv und erstaunlich stabil ist.

- Die vorliegende Erfindung wird spezifischer durch die folgenden Beispiele und Experimente illustriert.
- Beispiel 1
- Mit 1 mg der Verbindung 4, synthetisiert gemäß dem in WO 94102457 beschriebenen Verfahren wurden 30 mg Calciumcarbonat (leicht) kombiniert, wodurch eine Hartkapselpräparation erhalten wurde.
- Beispiel 2
- In 1 ml eines isotonischen Phosphatpuffers (pH 7,4) wurden 10 mg der Verbindung 32 gelöst, synthetisiert gemäß dem in der WO 94/02267 beschriebenen Verfahren, wodurch eine Nasentropfenpräparation erhalten wurde.
- Beispiel 3
- In 1 ml eines isotonischen Phosphatpuffers (pH 7,4) wurden 10 mg der Verbindung 23 gelöst, synthetisiert gemäß dem in WO 94/08959 beschriebenen Verfahren, wodurch eine Nasentropfenpräparation erhalten wurde.
- Experiment 1: Schlaf-induzierender Test durch nasale Verabreichung
- Verfahren:
- Neun männliche Rhesusaffen, die ungefähr 6 kg wogen, wurden in Gruppen von 3 Affen unterteilt. Um die Arzneimittel homogen in die Nasenhöhle zu versprühen, wurden Calciumcarbonat und Sofalcon als Träger verwendet und Testarzneimittel 1 (eine Hartkapselpräparation, enthaltend 1 mg der Verbindung 4 und 30 mg Calciumcarbonat (leicht), Testarzneimittel 2 (eine Hartkapselpräparation, enthaltend 1 mg der Verbindung 4 und 30 mg Calciumcarbonat (schwer) und Testarzneimittel 3 (eine Hartkapselpräparation, enthaltend 1 mg Verbindung 4 und 30 mg Sofalcon) wurden nasal an 3 Rhesusaffen verabreicht, die jeweils auf dem Affenstuhl ohne Anästhesie fixiert waren, wobei eine Nasentropfvorrichtung verwendet wurde (Jetlizer: hergestellt von Unisiajex Co.) und die Schlaf induzierende Wirkung wurde für 1 Stunde nach der Verabreichung (Aufzeichnung durch ein Videoband) beobachtet.
- Eine Woche später wurde das Verhalten derselben Affen in der Kontrollperiode beobachtet.
- Ergebnisse:
- In jedem Fall des Testarzneimittels wurde die Schlaf induzierende Wirkung definitiv bei 2 von 3 Affen beobachtet (6 von 9 Affen), in einer Zeitspanne von 5 bis 15 Minuten nach der nasalen Verabreichung, gefolgt von schläfrigen Zuständen.
- Andererseits wurde keine besondere Veränderung in der Kontrollgruppe beobachtet.
- Experiment 2: Schlaf induzierender Test durch Verabreichung in die Cisterna
- Verfahren:
- Drei männliche Rhesusaffen, die 3,6 bis 4,4 kg wogen, wurden individuell in Käfige platziert und das Verhalten der Tiere wurde durch Videoband für 1 Stunde vor Verabreichung des Arzneimittels bis 5 Stunden nach Verabreichung des Arzneimittels aufgezeichnet. Verbindung 32 und Prostaglandin-D2-methylester (PGD2) wurden jeweils in Salzlösung gelöst und durch einen Milliporefilter sterilisiert. Die Arzneimittel wurden in die Cisterna der Affen, die mit Isohalothan-Inhalation anästhesiert worden waren, infundiert. Die Dosierungen betrugen 1 μg und 10 μg/0,1 ml/Affe. Dieselben Dosierungen des Vehikels wurden in die Cisterna zum Erhalt einer Kontrollgruppe infundiert. Der Test wurde gemäß dem folgenden Testvorgehen durchgeführt:
Woche 1: Gruppe, behandelt mit Vehikel
Woche 2: Gruppe, behandelt mit 1 μg der Verbindung 32/Affe
Woche 3: Gruppe, behandelt mit 10 μg der Verbindung 32/Affe
Woche 4: Gruppe, behandelt mit 10 μg PGD2/Affe. - Um den Schlaf zu bestimmen, wurde die Periode, während derer die Affen beide Augen schlossen, in Intervallen von 15 Minuten gemessen, indem das aufgezeichnete Videoband zurückgespielt wurde und die Ergebnisse wurden gemäß der folgenden Skala eingeteilt:
- Ergebnisse:
- 90 Minuten, 135 bis 150 Minuten und 210 bis 240 Minuten nach der Verabreichung wurde beobachtet, dass die mit Vehikel behandelte Gruppe in einen schwachen Schlaf verfiel. In der mit 1 μg von Verbindung 32 pro Affe behandelten Gruppe wurde eine signifikante Schlafwirkung 30 bis 75 Minuten nach der Verabreichung beobachtet, im Vergleich mit der mit Vehikel behandelten Gruppe (
1 ). In der mit 10 μg der Verbindung 32 pro Affe behandelten Gruppe wurde die signifikante Schlafwirkung 45 Minuten bis 5 Stunden nach der Verabreichung im Vergleich mit der mit Vehikel behandelten Gruppe beobachtet (1 ). - Eine transiente Schlafwirkung wurde in der Gruppe beobachtet, die mit 10 μg PGD2/Affe behandelt wurde und zwar 45 Minuten nach Verabreichung (
2 ). - Experiment 3: Schlaf induzierende Wirkung durch Verabreichung in die Cisterna
- Die Gehirnwellen wurden gemäß dem in der Literatur beschriebenen Verfahren (H. ONOE, Proc. Natl. Acad. Sci., 1988, Band 185, Seite 4082) aufgezeichnet.
- Verfahren:
- Vier männliche, Krabben essende Affen, die 3,0 bis 4,7 kg wogen, wurden verwendet. Für eine chronische Implantation von Elektroden wurde aseptisch eine chirurgische Operation unter Pentobarbitalanästhesie durchgeführt, wobei Schraubelektroden aus rostfreiem Stahl in den cerebralen Cortex und den Occipital-Lappen platziert wurden und Elektroden aus rostfreiem Stahl für ein Elektromyogramm wurden auf die Cervicalmuskeln platziert, gefolgt von einem Löten von Blei eines Telemetriesystem-Transmitters (TL10M3-D70-EEE, Data Sciences, Inc.). Der Transmitter wurde subkutan in den Cervex posterior implantiert. Nach der Operation verheilte die Operationswunde vollständig und die Gehirnwellen wurden stabil und dann dienten die Tiere als Testtiere.
- Die Cerebrospinalflüssigkeit wurde durch Insertion einer Spiralnadel in die Cisterna cerebello-medullaris von dem Occipitalbereich unter Sevofluran-Inhalationsanästhesie identifiziert, woraufhin das Testarzneimittel (isotonische Natriumchloridlösung der Verbindung 32, sterilisiert durch einen Milliporefilter) in die Cisterna cerebello-medullaris in einer Menge von 10 μg/0,1 ml/Affe verabreicht wurde.
- Die Gehirnwellen (EEG) wurden kontinuierlich durch einen Datenaufzeichner unter Verwendung eines Telemetriesystems (UA-10, Data Sciences, Inc.) für 4 Stunden nach Verabreichung der Testsubstanz aufgezeichnet und das Verhalten wurde durch ein Videoaufzeichnungssystem beobachtet. Die Veränderungen der Schlaf-/Wachzustände (Wachsein, Nicht-REM-Schlaf und REM-Schlaf) über die Zeit wurden durch Veränderungen der Gehirnwellen, des Elektromyogramms und des Verhaltens beobachtet. Die Wach-, Nicht-REM-Schlaf- und die REM-Schlafzeiten und die Frequenz des REM-Schlafes wurden in einer Periode von 20 Minuten bis 240 Minuten nach der Verabreichung der Testsubstanz bestimmt und die Ergebnisse sind in Tabelle 2 dargestellt. Claims (5)
- Verwendung eines Prostaglandinderivats der Formel (I) oder eines pharmazeutisch annehmbaren Salzes davon zur Herstellung eines Medikaments, das als schlafförderndes Präparat wirksam ist: worin X ein Halogenatom ist, Y eine durch (CH2)m dargestellte Gruppe, eine cis-Vinylengruppe oder eine Phenylengruppe ist, Z eine Ethylengruppe, eine trans-Vinylengruppe, OCH2 oder S(O)nCH2 ist, R1 eine C3-10-Cycloalkylgruppe, eine mit (einer) C1-4-Alkylgruppe(n) substituierte C3-10-Cycloalkylgruppe, eine C4-13-Cycloalkylalkylgruppe, eine C5-10-Alkylgruppe, eine C5-10-Alkenylgruppe, eine C5-10-Alkinylgruppe oder eine verbrückte cyclische Kohlenwasserstoffgruppe ist, R2 ein Wasserstoffatom, eine C1-10-Alkylgruppe oder eine C3-10-Cycloalkylgruppe ist, m eine ganze Zahl von 1 bis 3 ist und n 0, 1 oder 2 oder ein pharmazeutisch annehmbares Salz davon ist.
- Verwendung gemäß Anspruch 1, worin in der Formel (I) X ein Chlor- oder ein Bromatom ist, Y eine durch (CH2)m dargestellte Gruppe oder eine cis-Vinylengruppe ist, R1 eine C3-10-Cycloalkylgruppe oder eine C4-13- Cycloalkylalkylgruppe ist, und R2 ein Wasserstoffatom oder eine C1-10-Alkylgruppe ist.
- Verwendung gemäß Anspruch 2, worin in Formel (I) Z OCH2 ist.
- Verwendung gemäß Anspruch 2, worin in Formel (I) Z SCH2 ist.
- Verwendung gemäß Anspruch 4, worin in Formel (I) Y eine durch (CH2)m dargestellte Gruppe ist und R1 eine C3-10-Cycloalkylgruppe ist.
