Description
- Technisches Gebiet
- Die vorliegende Erfindung betrifft neue Prostaglandin-Derivate.
- Stand der Technik
- Da Prostaglandin (hiernach als "PG" bezeichnet) verschiedene wichtige physiologische Wirkungen in einer Spurenmenge ausübt, wurde die Synthese der Derivate von natürlichen PGs und die biologischen Aktivitäten im Hinblick auf ihre praktische Verwendung als Arzneimittel untersucht und es wurden viele Veröffentlichungen gemacht.
- Insbesondere wurde von PGs, im Hinblick auf ihre verschiedenen zentralnervösen Wirkungen berichtet und Klarstellungen wurden unternommen im Hinblick auf ihren Intrazerebralgehalt, die Biosynthese, den Stoffwechselweg, ihre Intrazerebrallokalisierungen und Veränderungen mit Wachstum oder Alterung und es wurde ein Interesse im Hinblick auf PGs bezüglich Schlaf und Wachzuständen genommen. Unter diesen ist von PGD2 bekannt, daß er ein intrazerebraler humoraler Faktor ist, der das Auftreten oder den Erhalt von Schlaf kontrolliert und es wurde klargestellt, daß der durch PGD2 bei Affen induzierte Schlaf von dem spontanen natürlichen Schlaf in Gehirnwellen oder Verhalten nicht unterscheidbar ist (Proc. Natl. Acad. Sci. USA, Bd. 85, S. 4082–4086 (1988)); daher wird von dieser Verbindung angenommen, daß sie eine neue, den Schlaf induzierende Wirkung aufweist.
- WO 9208697 und JP-A-1104040 offenbaren ebenfalls die Verwendung von Prostaglandin-Derivaten für die Induktion von Schlaf.
- PGD2-Derivate einschließlich PGD2 sind jedoch zur Zeit aufgrund von Problemen unpraktisch, die ihre Wirkung und die Stabilität als Arzneimittel betreffen.
- Offenbarung der Erfindung
- Als Ergebnis intensiver Untersuchungen haben die vorliegenden Erfinder festgestellt, daß die Prostaglandin-Derivate mit einer Dreifachbindung zwischen den 13- und 14-Stellungen, dargestellt durch die folgende Formel (I), eine charakteristische den Schlaf induzierende Wirkung aufweisen und daher wurde die vorliegende Erfindung bewirkt.
- D. h., die vorliegende Erfindung betrifft ein Prostaglandin-Derivat, dargestellt durch die Formel (I): worin X ein Halogenatom bedeutet, n ist eine ganze Zahl von 1 bis 5, R1 ist eine C3-10-Cycloalkyl-Gruppe, eine C3-10-Cycloalkyl-Gruppe, substituiert mit einer C1-4-Alkyl-Gruppe(n), eine C4-13-Cycloalkylalkyl-Gruppe, eine C5-10-Alkyl-Gruppe, eine C5-10-Alkenyl-Gruppe, eine C5-10-Alkinyl-Gruppe oder eine überbrückte cyclische Kohlenwasserstoff-Gruppe und R2 ist ein Wasserstoffatom, eine C1-10-Alkyl-Gruppe oder eine C3-10-Cycloalkyl-Gruppe oder ein pharmazeutisch akzeptables Salz davon.
- Weiterhin betrifft die vorliegende Erfindung ein Prostaglandin-Derivat, dargestellt durch Formel (I), worin R1 eine C3-10-Cycloalkyl-Gruppe, eine C3-10-Cycloalkyl-Gruppe, substituiert mit C1-4-Alkyl-Gruppen (einer Gruppe) oder eine C4-13-Cycloalkylalkyl-Gruppe ist, und X, n und R2 sind wie oben definiert oder ein pharmazeutisch akzeptables Salz davon.
- Weiterhin betrifft die vorliegende Erfindung eine pharmazeutische Zusammensetzung, die als einen effektiven Bestandteil das oben erwähnte Prostaglandin-Derivat, dargestellt durch Formel (2) oder ein pharmazeutisch akzeptables Salz davon, umfaßt.
- Weiterhin betrifft die vorliegende Erfindung eine schlafinduzierende Präparation, die als effektiven Bestandteil das oben erwähnte Prostaglandin-Derivat, dargestellt durch Formel (2) oder das pharmazeutisch akzeptable Salz davon umfaßt.
- Noch weiterhin betrifft die vorliegende Erfindung die Verwendung der oben erwähnten Prostaglandin-Derivate für die Herstellung eines Medikaments zur Induktion von Schlaf.
- Bei der vorliegenden Erfindung betrifft das Halogenatom ein Fluoratom, ein Chloratom, ein Bromatom oder ein Iodatom.
- Beispiele für die C3-10-Cycloalkyl-Gruppe sind eine Cyclopropyl-Gruppe, eine Cyclobutyl-Gruppe, eine Cyclopentyl-Gruppe, eine Cyclohexyl-Gruppe und eine Cycloheptyl-Gruppe.
- Beispiele für die C3-10-Cycloalkyl-Gruppe, substituiert mit einer C1-4-Alkyl-Gruppe (Gruppen) sind eine Methylcyclopropyl-Gruppe, eine Methylcyclohexyl-Gruppe und eine Ethylcyclohexyl-Gruppe.
- Beispiele für die C4-13-Cycloalkylalkyl-Gruppe sind eine Cyclopropylmethyl-Gruppe, eine Cyclobutylmethyl-Gruppe, eine Cyclopentylmethyl-Gruppe, eine Cyclopentylethyl-Gruppe, eine Cyclohexylmethyl-Gruppe, eine Cyclohexylethyl-Gruppe und eine Cycloheptylmethyl-Gruppe.
- Die C5-10-Alkyl-Gruppe bezieht sich auf eine lineare oder verzweigte Alkyl-Gruppe und Beispiele hierfür sind eine Pentyl-Gruppe, eine Hexyl-Gruppe, eine Heptyl-Gruppe, eine Octyl-Gruppe, eine 1-Methylpentyl-Gruppe, eine 2-Methylpentyl-Gruppe, eine 1-Methylhexyl-Gruppe, eine 2-Methylhexyl-Gruppe, eine 2,4-Dimethylpentyl-Gruppe, eine 2-Ethylpentyl-Gruppe, eine 2-Methylheptyl-Gruppe, eine 2-Ethylhexyl-Gruppe, eine 2-Propylpentyl-Gruppe, eine 2-Propylhexyl-Gruppe und eine 2,6-Dimethylheptyl-Gruppe.
- Die C5-10-Alkenyl-Gruppe bezieht sich auf eine lineare oder verzweigte Alkenyl-Gruppe und Beispiele hierfür sind eine 3-Pentenyl-Gruppe, eine 4-Hexenyl-Gruppe, eine 5-Heptenyl-Gruppe, eine 4-Methyl-3-pentenyl-Gruppe, eine 2,4-Dimethylpentenyl-Gruppe, eine 6-Methyl-5-heptenyl-Gruppe und eine 2,6-Dimethyl-5-heptenyl-Gruppe.
- Die C5-10-Alkinyl-Gruppe bezieht sich auf eine lineare oder verzweigte Alkinyl-Gruppe und Beispiele hierfür sind eine 3-Pentinyl-Gruppe, eine 3-Hexinyl-Gruppe, eine 4-Hexinyl-Gruppe, eine 1-Methylpent-3-inyl-Gruppe, eine 2-Methylpent-3-inyl-Gruppe, eine 1-Methylhex-3-inyl-Gruppe und eine 2-Methylhex-3-inyl-Gruppe.
- Beispiele für die überbrückte cyclische Kohlenwasserstoff-Gruppe sind eine Bornyl-Gruppe, eine Norbornyl-Gruppe, eine Adamantyl-Gruppe, eine Pinanyl-Gruppe, eine Thujyl-Gruppe, Caryl-Gruppe und eine Camphanyl-Gruppe.
- Die C1-10-Alkyl-Gruppe für R2 bezieht sich auf eine lineare oder verzweigte Alkyl-Gruppe und Beispiele hierfür sind eine Methyl-Gruppe, eine Ethyl-Gruppe, eine Propyl-Gruppe, eine Isopropyl-Gruppe, eine Butyl-Gruppe, eine Isobutyl-Gruppe, eine tert-Butyl-Gruppe, eine Pentyl-Gruppe, eine Isopentyl-Gruppe, eine 2-Ethylpropyl-Gruppe, eine Hexyl-Gruppe, eine Isohexyl-Gruppe, eine 1-Ethylbutyl-Gruppe, eine Heptyl-Gruppe, eine Isoheptyl-Gruppe, eine Octyl-Gruppe, eine Nonyl-Gruppe und eine Decyl-Gruppe.
- Beispiele für das pharmazeutisch akzeptable Salz sind Salze mit einem Alkalimetall (z. B. Natrium oder Kalium), einem Erdalkalimetall (z. B. Calcium oder Magnesium), Ammoniak, Methylamin, Dimethylamin, Cyclpentylamin, Benzylamin, Piperidin, Monoethanolamin, Diethanolamin, Monomethylmonoethanolamin, Tromethamin, Lysin, ein Tetraalkylammonium und Tris(hydroxymethyl)aminomethan.
- Die Verbindungen der Formel (2) der vorliegenden Erfindung können z. B. durch durch die folgende Reaktionsformeln zusammengefaßten Verfahren hergestellt werden.
- In der Reaktionsformel ist R3 eine C1-10-Alkyl-Gruppe oder eine C3-10-Cycloalkyl-Gruppe und X, R1 und n sind wie oben definiert.
- Die oben erwähnte Reaktion wird wie folgt illustriert:
- (1) Zunächst wird eine bekannte Verbindung der Formel (II) mit 0,8 bis 2,0 Äquivalenten einer organischen Aluminium-Verbindung, dargestellt durch Formel (III) in einem inerten Lösungsmittel (z. B. Benzol, Toluol, Tetrahydrofuran, Diethylether, Methylenchlorid oder n-Hexan) bei –10 bis 30°C, vorzugsweise 0 bis 10°C, gemäß dem Verfahren von Sato et al. (Journal of Organic Chemistry, Bd. 53, Seite 5590 (1988)) umgesetzt, um stereospezifisch eine Verbindung der Formel (IV) zu ergeben.
- (2) Die Verbindung der Formel (IV) wird mit 0,5 bis 4,0 Äquivalenten einer organischen Kupfer-Verbindung, dargestellt durch Formel (V) und 0,5 bis 4,0 Äquivalenten Trimethylchlorsilan in einem inerten Lösungsmittel (z. B. Benzol, Toluol, Tetrahydrofuran, Diethylether, Methylenchlorid, n-Hexan oder n-Pentan) bei –78 bis 40°C, gefolgt von einer Hydrolyse unter Verwendung einer anorganischen Säure (z. B. Salzsäure, Schwefelsäure oder Salpetersäure) oder eine organischen Säure (z. B. Essigsäure, oder p-Toluol-sulfonsäure) oder eines Aminsalzes davon (z. B. Pyridinium-p-toluolsulfonat) in einem organischen Lösungsmittel (z. B. Aceton, Methanol, Ethanol, Isopropanol, Diethylether oder einer Mischung davon) bei 0 bis 40°C umgesetzt, um stereospezifisch eine Verbindung der Formel (VI) zu ergeben.
- (3) Die Verbindung der Formel (VI) wird mit 0,5 bis 5 Äquivalenten eines Reduktionsmittels reduziert (z. B. Kaliumborhydrid, Natriumborhydrid, Natriumcyanoborhydrid oder Lithiumtri-sek-butylborhydrid) in einem organischen Lösungsmittel (z. B. Tetrahydrofuran, Diethylether, Ethylalkohol oder Methylalkohol) bei –78 bis 40°C um Verbindungen der Formel (VII) oder (VII') zu ergeben. Diese Verbindungen der Formel (VII) und (VII') können durch ein konventionelles Trennverfahren, wie z. B. einer Säulenchromatographie, gereinigt werden.
- (4) Die Verbindung der Formel (VII) oder (VII') wird mesyliert oder tosyliert, z. B, mit 1 bis 6 Äquivalenten Methansulfonylchlorid oder p-Toluolsulfonylchlorid in einem geeigneten Lösungsmittel, wie z. B. Pyridin (falls nötig in Gegenwart von 0,8 bis 6 Äquivalenten 4-Dimethylaminopyridin) bei –20 bis 40°C, gefolgt von einer Chlorierung mit 1 bis 16 Äquivalenten Tetra-n-butylammoniumchlorid um eine Verbindung der Formel (VIII) bzw. (VIII') zu ergeben, worin X ein Chloratom ist. Hier kann die Bromierung oder Fluorierung ebenfalls auf gewöhnliche Weise durchgeführt werden. Z. B. kann eine Bromierung durch eine Reaktion mit 1 bis 10 Äquivalenten Kohlenstofftetrabromid in Gegenwart von 1 bis 10 Äquivalenten Triphenylphosphin und 1 bis 10 Äquivalenten Pyridin in Acetonitril durchgeführt werden und die Fluorierung kann durch eine Umsetzung mit 5 bis 20 Äquivalenten Diethylaminoschwefeltrifluorid (DAST) in Methylenchlorid bewirkt werden.
- (5) Die Schutzgruppe der Hydroxyl-Gruppe der Verbindung der Formel (VIII) oder (VIII'), d. h. eine tert-Butyldimethylsilyl-Gruppe wird durch Verwendung von Flußsäure, Pyridiniumpoly(hydrogenfluorid) oder Salzsäure unter konventionellen Bedingungen in einem Lösungsmittel (z. B. Methanol, Ethanol, Acetonitril, einer Mischung davon oder einer Mischung dieser Lösungsmittel und Wasser) durchgeführt, um ein PG-Derivat der Formel (Ia) oder (Ia') zu ergeben.
- (6) Das PG-Derivat der Formel (Ia) oder (Ia') wird unter Verwendung von 1 bis 6 Äquivalenten einer Base in einem Lösungsmittel hydrolysiert, das gewöhnlicherweise für die Hydrolyse verwendet wird, um ein PG-Derivat der Formel (Ib) oder (Ib') der vorliegenden Erfindung zu ergeben. Beispiele für die hier verwendete Base sind Lithiumhydroxid und Kaliumcarbonat und Beispiele für das Lösungsmittel sind Acetonitril, Aceton, Methanol, Ethanol, Wasser und eine Mischung davon.
- Weiterhin wird die Verbindung der Formel (Ia) oder (Ia') durch eine Reaktion mit einem Enzym in einer Pufferlösung hydrolysiert, wie z. B. einem Phosphatpuffer oder Trishydrochldoridpuffer, falls nötig, durch Verwendung eines organischen Lösungsmittels (z. B. eines wassermischbaren Lösungsmittels wie z. B. Aceton, Methanol oder Ethanol), um das Prostaglandin-Derivat der vorliegenden Erfindung zu ergeben, d. h. die Verbindung der Formel (Ib) oder (Ib'). Beispiele für das hier zu verwendende Enzyme sind Enzyme, die von Mikroorganismen erzeugt werden (z. B. Enzyme, die von Mikroorganismen erzeugt werden, die zu Candida sp. oder Pseudomonas sp. gehören) und Enzyme, die aus tierischen Organen hergestellt werden (z. B. Schweineleber oder Schweinepankreas). Kommerziell erhältliche Enzyme sind z. B. Lipase VII (abgeleitet von dem Mikroorganismus von Candida sp., Sigma Co.), Lipase AY (abgeleitet von dem Mikroorganismus von Candida sp.; Amano Pharmaceutical Co.), Lipase AY (abgeleitet vom Mikroorganismus von Pseudomonas sp.; Amano Pharmaceutical Co.), Lipase PS (abgeleitet vom Mikroorganismus Pseudomonas sp.; Amano Pharmaceutical Co.), Lipase MF (abgeleitet vom Mikroorganismus von Pseudomonas sp.; Amano Pharmaceutical Co.), PLE (hergestellt aus Schweineleber; Sigma Co.), Lipase II (her gestellt aus Schweinepankreas; Sigma Co.) oder Lipoproteinlipase (hergestellt aus Schweinepankreas; Tokyo Kasei Kogyo Co.).
- Die Menge des zu verwendenden Enzyms beträgt, obwohl sie von der Potenz des Enzyms und der Menge des Substrats abhängt (die Verbindung der Formel (Ia)) in der Regel 0,1 bis 20 Gew.-Teile, basierend auf dem Substrat und die Reaktionstemperatur liegt bei 25 bis 50°C, vorzugsweise 30 bis 40°C.
- Die Verbindungen der Formel (I) der vorliegenden Erfindung sind z. B. die folgenden.
- Die Verbindungen der vorliegenden Erfindung können oral oder parenteral wie z. B. intravenös oder nasal verabreicht werden. Z. B. können sie oral in Form von Tabletten, Staubpulvern, Körnchen, Pulvern, Kapseln, Lösungen, Emulsionen oder Suspensionen verabreicht werden, die jeweils gemäß konventionellen Verfahren hergestellt werden können. Als Dosierungsformen für die intravenöse Verabreichung werden wäßrige oder nicht-wäßrige Lösungen, Emulsionen, Suspensionen oder feste Präparationen, die in einem Lösungsmittel für eine Injektion direkt vor der Verwendung gelöst werden müssen, verwendet. Weiterhin kann die nasale Verabreichung durch quantitatives Versprühen einer Lösung oder eines Pulvers (harte Kapseln) enthaltend das Arzneimittel, in die nasale Höhlung durch Verwendung einer dafür bestimmten nasalen Tropfvorrichtung oder Sprühvorrichtung durchgeführt werden. Weiterhin können die Verbindungen der vorliegenden Erfindung in Form von Einschlußverbindungen formuliert werden, mit α-, β- oder γ-Cyclodextrin, oder methyliertem Cyclodextrin. Die Dosis variiert je nach Alter, Körpergewicht usw., liegt jedoch allgemein bei 1 ng bis 1 mg/Tag pro Erwachsenem.
- Gewerbliche Anwendbarkeit
- Die Verbindungen der vorliegenden Erfindung weisen eine ausreichende schlafinduzierende Wirkung und eine ausgezeichnete Stabilität auf, und sind daher als schlafinduzierende Mittel geeignet.
- Beste Ausführungsform der Erfindung
- Die vorliegende Erfindung wird im Detail durch die folgenden Beispiele und Experimente illustriert.
- In der Nomenklatur der Verbindung bedeutet "nor", daß ein Kohlenstoffatom an der Position fehlt, (z. B. 16,17,18,19,20-Pentanor bedeutet das Fehlen von Kohlenstoffatomen an den 16- bis 20-Stellungen), und "homo" bedeutet den Anstieg eine Kohlenstoffatoms (z. B. 1a-Homo bedeutet die Gegenwart eines Kohlenstoffatoms an der 1a-Stellung zwischen den 1- und 2-Stellungen).
- Beispiel 1
- Herstellung von 9-Desoxy-9β-chlor-16,17,18,19,20-Pentanor-15-cyclohexyl-2,2,3,3,13,14-hexadehydro-PGF1α-tert-butylester (Verbindung 8)
-
- (1) Unter einem Argonstrom wurde (3S)-3-(tert-Butyldimethylsiloxy)-3-cyclohexylprop-1-yn (3,61 g) in Toluol (28,8 ml) gelöst und n-Butyllithium (1,95 M, Hexan-Lösung 6,4 ml) bei 0°C zugefügt, gefolgt von Rühren bei derselben Temperatur für 30 Minuten. Zu der Lösung wurde Diethylaluminiumchlorid (0,97 M, Hexan-Lösung, 14,8 ml) bei 0°C zugefügt, gefolgt von einem Rühren bei Raumtemperatur für 30 Minuten. Zu der Lösung wurde (4R)-2-(N,N-Diethylamino)methyl-4-(tert-butyldimethylsiloxy)cyclopent-2-en-1-on (0,25 M, Toluol-Lösung, 14,8 ml) bei Raumtemperatur zugefügt, gefolgt von einem Rühren für 15 Minuten. Die Reaktionslösung wurde in einer Mischung aus Hexan (100 ml) – eine gesättigte wäßrige Ammoniumchlorid-Lösung (100 ml) – eine wäßrige Salzsäure-Lösung (3 N, 30 ml) unter Rühren gegossen und die organische Schicht wurde abgetrennt und mit einer gesättigten wäßrigen Natriumbicarbonat-Lösung (50 ml) gewaschen. Die resultierende organische Schicht wurde über wasserfreiem Magnesiumsulfat getrocknet, gefiltert und konzentriert und der resultierende Rest wurde durch Silicagel-Säulenchromatographie gereinigt (Entwicklungslösungsmittel; Hexan : Ethylacetat = 10 : 1) um (3R,4R)-2-Methylen-3-[(3S)-3-(tert-Butyldimethylsiloxy)-3-cyclohexylprop-1'-inyl]-4-(tert-butyldimethylsiloxy)-cyclopentan-1-on (3,69 g) zu ergeben. 1H-NMR (CDCl3, 200 MHz) δppm; 0,07, 0,08 und 0,12 (3s, 12H), 0,88 (s, 18H), 0,92–1,92 (m, 11H), 2,32 (dd, J = 17,8, 7,4 Hz, 1H), 2,71 (dd, J = 17,8, 6,5 Hz, 1H), 3,48–3,58 (m, 1H), 4,11 (dd, J = 6,2, 1,4 Hz, 1H), 4,20–4,32 (m, 1H), 5,55 (d, J = 2,6 Hz, 1H), 6,13 (d, J = 3,0 Hz, 1H). IR (neat); 2930, 2850, 1375, 1640, 1470, 1380, 1255, 830, 770 cm–1.
- (2) Unter einem Argonstrom wurde Kupfer(I)-cyanid-Dilithiumdichlorid (1,0 M, Tetrahydrofuran-Lösung, 41,21 ml) zu 5-tert-Butoxycarbonyl-4-pentinylzink(II)-iodid (0,81 N, Tetrahydrofuran-Lösung, 40,69 ml) bei –70°C zugefügt, gefolgt von einem Rühren bei derselben Temperatur für 20 Minuten. Zu der Lösung wurde eine Diethylether-Lösung (66,0 ml) der Verbindung (7,86 g), wie oben bei (1) erhalten und Chlortrimethylsilan (3,77 ml) bei –70°C zugefügt und die Temperatur wurde auf 0°C unter Rühren über ungefähr eine Stunde angehoben. Die Reaktionslösung wurde nach Zugabe einer gesättigten wäßrigen Ammoniumchlorid-Lösung (250 ml) mit Hexan extrahiert. Die organische Schicht wurde mit einer gesättigten wäßrigen Natriumchlorid-Lösung gewaschen, getrocknet und konzentriert und der resultierende Rest wurde in Diethylether (16,5 ml) – Isopropylalkohol (66,0 ml) gelöst und Pyridinium-p-toluolsulfonat (208 mg) wurde zugefügt, gefolgt von einem Rühren bei Raumtemperatur für 12 Stunden. Die Reaktionslösung wurde nach Zugabe von Hexan (200 ml) mit einer gesättigten wäßrigen Natriumbicarbonat-Lösung und einer gesättigten wäßrigen Natriumchlorid-Lösung, gewaschen getrocknet und konzentriert und der resultierende Rest wurde durch eine Silicagel-Säulenchromatographie gereinigt (Entwicklungslösungsmittel; Hexan : Ethylacetat = 15 : 1) um 16,17,18,19,20-Pentanor-15- cyclohexyl-2,2,3,3,13,14-hexadehydro-PGE1-tert-butylester-11,15-bis(tertbutyldimethylsilyl)ether (5,73 g) zu ergeben. 1H-NMR (CDCl3, 200 MHz) δppm; 0,08 (s, 3H), 0,09 (s, 3H), 0,10 (s, 3H), 0,13 (s, 3H), 0,89 (s, 9H), 0,90 (s, 9H), 0,96–1,96 (m, 19H), 1,49 (s, 9H), 1,97–2,35 (m, 2H), 2,56–2,75 (m, 2H), 4,03–4,19 (m, 1H), 4,22–4,35 (m, 1H). IR (neat); 2932, 2857, 2238, 1747, 1708, 1452, 1393, 1370, 1278, 1258, 1162, 1078, 840, 779, 755, 670 cm–1.
- (3) Eine Methylalkohl-Lösung (94,7 ml) der Verbindung (5,73 g), erhalten in dem obigen Schritt (2) wurde auf 0°C abgekühlt und Kaliumborhydrid (1,02 g) wurde hinzugefügt, gefolgt von einem Rühren für 15 Minuten. Nach Zugabe von Wasser wurde die Extraktion mit Ether (200 ml) durchgeführt und der Extrakt wurde mit einer gesättigten, wäßrigen Ammoniumchlorid-Lösung und einer wäßrigen Natriumchlorid-Lösung gewaschen, über wasserfreien Magnesiumsulfat getrocknet und konzentriert. Der Rest wurde durch eine Silicagel-Säulenchromatographie gereinigt (Entwicklungslösungsmittel; Hexan : Ethylacetat = 5 : 1) um 16,17,18,19,20-Pentanor-15-cyclohexyl-2,2,3,3,13,14,hexadehydro-PGF1α-tert-butylester-1,15-bis(tertbutyldimethylsilyl)ether (2,17 g) und 16,17,18,19,20-Pentanor-15-cyclohexyl-2,2,3,3,13,14-hexadehydro-PGF1β-tert-butylester-11,15-bis(tertbutyldimethylsilyl)ether (2,75 g) zu ergeben. 16,17,18,19,20-Pentanor-15-cyclohexyl-2,2,3,3,13,14-hexahydro-PGF1α-tert-butylester-11,15-bis(tert-butyldimethylsilyl)ether 1H-NMR (CDCl3, 200 MHz) δppm; 0,09 (s, 3H), 0,10 (s, 3H), 0,11 (s, 3H), 0,12 (s, 3H), 0,90 (s, 9H), 0,91 (s, 9H), 0,94–2,07 (m, 21H), 1,50 (s, 9H), 2,26–2,38 (m, 1H), 2,42–2,51 (m, 1H), 2,55 (d, J = 9,5 Hz, 1H), 4,02–4,20 (m, 1H), 4,09 (dd, J = 6,4, 1,7 Hz, 1H), 4,24–4,33 (m, 1H). - (4) Zu einer Pyridin-Lösung (4,95 ml) von 16,17,18,19,20-Pentanor-15-cyclohexyl-2,2,3,3,13,14-hexadehydro-PGF1α-tert-butylester-11,15- bis(tert-butyldimethylsilyl)ether (641 mg), erhalten im obigen Schritt (3) wurde Methansulfonylchlorid (0,153 ml) bei 0°C zugefügt, gefolgt von einem Rühren bei Raumtemperatur für 2 Stunden. Dieses wurde zu einer Toluol-Suspension (4,95 ml) von n-Tetrabutylammoniumchlorid (4,40 g) zugefügt, gefolgt von einem Rühren bei 40°C über Nacht. Nach Zugabe einer gesättigten wäßrigen Natriumchlorid-Lösung (50 ml) und Ethylacetat (50 ml) wurde die organische Schicht abgetrennt und die wäßrige Schicht wurde mit Ethylacetat (20 ml) extrahiert. Die resultierenden organischen Schichten wurden kombiniert, mit gesättigter wäßriger Natriumchlorid-Lösung gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und gefiltert. - (5) Zu einer Acetonitril-Lösung (30,2 ml) der Verbindung (604 mg), erhalten im obigen Schritt (4) wurde eine wäßrig Flußsäure-Lösung (46%, 8,80 ml) bei 0°C zugefügt, gefolgt von einem Rühren bei derselben Temperatur für 2 Stunden. Die Reaktionslösung wurde in eine Mischung aus Ethylacetat (100 ml) und einer gesättigten wäßrigen Natriumbicarbonat-Lösung (155 ml) unter Rühren gegossen, die organische Schicht wurde abgetrennt und die wäßrige Schicht wurde mit Ethylacetat (20 ml) extrahiert. Die resultierenden organischen Schichten wurde kombiniert, mit gesättigter wäßriger Natriumchlorid-Lösung gewaschen, über wasserfreien Magnesiumsulfat getrocknet und filtriert. - Herstellung von 9-Desoxy-9β-chlor-16,17,18,19,20-pentanor-15-cyclohexyl-2,2,3,3,13,14-hexadehydro-PGF1α (Verbindung 14)
- Eine Aceton-Lösung (22,4 ml) der Verbindung (410 mg) erhalten in Beispiel 2 wurde zu einer Suspension von Lipase PS (11,7 g) in Wasser (66 ml) zugefügt und Phosphatpuffer (pH 7,0) (11,3 ml) und Wasser (161 ml) wurden zugefügt, gefolgt von einem Rühren bei 38°C für 12 Stunden. Die Reaktionslösung wurde nach Filtration mit Ethylacetat extrahiert. Die organische Schicht wurde mit einer gesättigten wäßrigen Natriumchlorid-Lösung gewaschen, über wasserfreien Magnesiumsulfat getrocknet und dann filtriert. Das Filtrat wurde unter reduziertem Druck konzentriert und das resultierende Rohprodukt wurde durch Silicagel-Säulenchromatographie gereinigt (Entwicklungslösungsmittel; Ethylacetat) um die Titelverbindung (400 mg) zu ergeben.

- Beispiel 9
- Herstellung von 9-Desoxy-9β-chlor-1a-homo-16,17,18,19,20-pentanor-15-cyclohexyl-1a,1a,2,2,13,14-hexadehydro-PGF1α (Verbindung 20)
- Im wesentlichen auf dieselbe Weise wie in Beispiel 3 unter Verwendung der in Beispiel 8 erhaltenen Verbindung wurde dadurch die Titelverbindung erhalten.
1H-NMR (CDCl3, 300 MHz) δppm; 0,85–1,89 (m, 19H), 2,09–2,42 (m, 4h), 2,38 (t, J = 6,9 Hz, 2H), 2,70–3,40 (br, 3H), 3,91–4,00 (m, 1H), 4,19 (dd, J = 6,1, 1,9 Hz, 1H), 4,32–4,42 (m, 1H).
IR (neat); 3368, 2929, 2854, 2624, 2236, 1691, 1450, 1262, 1081, 1006, 893, 757, 595 cm–1. - Experiment [Schlafinduktionstest durch cisternale Verabreichung]
- Verfahren:
- Vier männliche krabbenfressende Affen, die 2,0 bis 3,5 kg wogen, wurden einzeln in Käfige plaziert und das Verhalten der Tiere wurde durch Videoaufnahmen für eine Stunde vor der Verabreichung des Arzneimittels und drei Stunden nach Verabreichung des Arzneimittels aufgezeichnet. Die Verbindung 14 wurde in Salzlösung gelöst und durch einen Millipore-Filter sterilisiert. Das Arzneimittel wurde cisteral, durch mit Isofluran-Inhalation anästhesierte Affen, infundiert. Die Dosierungen betrugen 1 μg und 10 μg/0,1 ml/Affe. Dieselben Dosierungen des Vehikels wurden cisternal einer Kontrollgruppe infundiert. Der Test wurde gemäß dem folgenden Testaufbau durchgeführt.
- Woche 1: Gruppe mit Vehikel behandelt
- Woche 2: Gruppe mit 1 μg der Verbindung 14/Affe behandelt
- Woche 3: Gruppe mit 10 μg der Verbindung 14/Affe behandelt
- Um den Schlaf zu bestimmen wurde die Zeit gemessen, für die der Affe entspannt beide Augen schloß, indem das aufgezeichnete Videoband zurückgespielt wurde. Die Schlafzeit (Sekunden) pro Stunde wurde bestimmt und ist in Tabelle 2 angegeben.
Claims (6)
- Prostaglandinderivat, dargestellt durch Formel (I): worin X ein Halogenatom bedeutet, n ist eine ganze Zahl von 1 bis 5, R1 ist eine C3-10-Cycloalkylgruppe, eine C3-10-Cycloalkylgruppe, substituiert mit einer C1-4-Alkylgruppe(n), eine C4-13-Cycloalkylalkylgruppe, eine C5-10-Alkylgruppe, eine C5-10-Alkenylgruppe, eine C5-10-Alkinylgruppe oder eine überbrückte cyclische Kohlenwasserstoffgruppe und R2 ist ein Wasserstoffatom, eine C1-10-Alkylgruppe oder eine C3-10-Cycloalkylgruppe oder ein pharmazeutisch akzeptables Salz davon.
- Prostaglandinderivat, dargestellt durch Formel (I) gemäss Anspruch 1, worin R1 eine C3-10-Cycloalkylgruppe, eine C3-10-Cycloalkylgruppe, substituiert mit einer C1-4-Alkylgruppe(n) oder eine C4-13-Cycloalkylalkylgruppe ist oder ein pharmazeutisch akzeptables Salz davon.
- Pharmazeutische Zusammensetzung, die als effektiven Bestandteil das Prostaglandinderivat gemäss Anspruch 1 oder 2 umfasst oder das pharmazeutisch akzeptable Salz davon.
- Prostaglandinderivat oder das pharmazeutisch akzeptable Salz davon gemäss Anspruch 1 oder 2 zur Verwendung als pharmazeutisch effektiver Bestandteil.
- Schlafinduzierende Präparation, die als effektiven Bestandteil das Prostaglandinderivat oder das pharmazeutisch akzeptable Salz davon gemäss Anspruch 1 oder 2 umfasst.
- Verwendung einer pharmakologisch effektiven Menge des Prostaglandinderivats oder des pharmazeutisch akzeptablen Salzes gemäss Anspruch 1 oder 2 für die Herstellung eines Medikaments zur Induktion von Schlaf.
