Description
Die Hauptanmeldung P 36 28 747.4 betrifft ein Resistenzgen
gegen Phosphinothricin (PTC), erhältlich aus der Gesamt-DNA
von auf Phosphinothricyl-alanyl-alanin (PTT)-Resistenz
selektiertem Streptomyces viridochromogenes DSM 40736 durch
Schneiden mit BamHI, Klonieren eines 4,0 kb großen
Fragments und Selektion auf PTT-Resistenz, sowie die
Verwendung dieses Gens zur Herstellung PTC-resistenter
Pflanzen, als PTT-Resistenz-Marker in Bakterien und als
PTC-Resistenz-Marker in Pflanzenzellen. Das Resistenzgen
ist durch eine Restriktionskarte (Fig. 1) charakterisiert,
die ein 4 kb großes BamHI-Fragment näher definiert.
Durch Klonieren von Teilbereichen dieses 4 kb-Fragmentes
wurde die Lage des Codierbereichs näher eingegrenzt.
Hierbei zeigte sich, daß das Resistenzgen auf dem 1,6 kb
SstII-SstI-Fragment (Positionen 0,55 bis 2,15 in Fig. 1 der
Hauptanmeldung) liegt. Durch Verdauung mit BglII wird das
0,8 kb große Fragment gewonnen, das nach Einbau in ein
Plasmid und Transformation von S. lividans PTT-Resistenz
vermittelt. Diese Resistenz ist durch die N-Acetylierung
von PCT bedingt (Nachweis wie in der Hauptanmeldung
angegeben).
Die Sequenzierung nach Maxam und Gilbert des 0,8 kb-Fragments
ergibt die DNA-Sequenz I (Anhang). Aus den
offenen Leserahmen dieser Sequenz läßt sich die Lage des
Resistenzgens ermitteln (ab Position 258). Es zeigt sich,
daß das Ende des Gens auf dem nichtsequenzierten Bereich
jenseits der BglII-Schnittstelle (Position 1,8 in Fig. 1
der Hauptanmeldung) liegt. Da jedoch das 0,8 kb große
BglII-Fragment PTT-Resistenz vermittelt, und zwar auch -
nach Einbau in einen E. coli-Vektor unter Vorschaltung
eines E. coli-Promotors wie lac - in E. coli, kann der
nichtsequenzierte Teil nur wenige Aminosäuren umfassen.
In der DNA-Sequenz I ist die "Shine-Dalgarno-Sequenz"
durch Unterstreichen hervorgehoben, ebenso das als
Startcodon wirkende GTG. Der maßgebliche Leserahmen ist
also in der obersten Zeile ausgedruckt.
Die DNA-Sequenz II zeigt die Restriktionsschnittstellen
innerhalb des sequenzierten Gens. Enzyme, die die Sequenz
mehr als sechsmal schneiden, sind nicht angegeben.
Claims (4)
1. Resistenzgen gegen Phosphinothricin (PTC), enthaltend
die DNA Sequenz I (Anhang) ab Position 258.
2. Verwendung des Gens nach Anspruch 1 zur Herstellung
PTC-resistenter Pflanzen.
3. Verwendung des Gens nach Anspruch 1 als
Phosphinothricyl-alanyl-alanin (PTT)-Resistenz-Marker
in Bakterien.
4. Verwendung des Gens nach Anspruch 1 als PTC-Resistenz-Marker
in Pflanzenzellen.
